Abstract
Description
エンドトキシン、例えば、リポ多糖(LPS)は、グラム陰性菌、例えば、大腸菌(Escherichia coli)の外側の細胞壁に見出される共有結合した脂質と多糖サブユニットを含む。
a.候補抗原ポリペプチドが密に会合した、貪食可能な粒子を与え、会合したポリペプチドを含む粒子に対して変性洗浄を行い、エンドトキシンのレベルを、後の工程を妨害しないほど十分に低くする工程と;
b.可変抗原提示細胞を与える工程と;
c.抗原提示細胞によって粒子の貪食作用が可能になる条件で、洗浄された粒子と、抗原提示細胞とをin vitroで接触させる工程と;
e.抗原提示細胞によって提示される抗原に反応して特異的なT細胞活性化が可能になる条件で、T細胞サンプルと、粒子と接触した抗原提示細胞とをin vitroで接触させる工程と;
f.T細胞サンプルにおけるT細胞活性化度を決定する工程と
を含む、方法。

に由来する、項目1〜32のいずれかに記載の方法。
a.候補ペプチド自己抗原が密に会合した、貪食可能な粒子を与え、会合したポリペプチドを含む粒子に対して変性洗浄を行い、エンドトキシンのレベルを、後の工程を妨害しないほど十分に低くする工程と;
b.試験被験体から、生存能力のあるT細胞と生存能力のある抗原提示細胞とを含む末梢血単核細胞(PBMC)サンプルを与える工程と;
c.抗原提示細胞によって粒子の貪食作用が可能になり、抗原提示細胞によって提示される抗原に反応して特異的なT細胞活性化が可能になる条件で、PBMCサンプルと、粒子とをin vitroで接触させる工程と;
d.接触したPBMC細胞において、活性化したT細胞の割合を定量する工程と;
e.定量された割合と、健康な被験体に由来する相当する定量された割合とを比較する工程であって、それによって、試験被験体において活性化したT細胞の割合が大きいことは、この試験被験体において候補自己抗原に対する自己反応性の指標となる工程と
を含む、方法。
第1の態様において、本発明は、抗原特異性T細胞活性化をアッセイするためのin vitro方法であって、
a.候補抗原ポリペプチドが密に会合した、貪食可能な粒子を与え、会合したポリペプチドを含む粒子に対して変性洗浄を行い、エンドトキシンのレベルを、本方法の後のいずれかの工程(b〜f)を妨害しないほど十分に低くする工程と;
b.可変抗原提示細胞(APC)を与える工程と;
c.抗原提示細胞によって粒子の貪食作用が可能になる条件で、洗浄された粒子と、粒子と接触した抗原提示細胞とをin vitroで接触させる工程と;
d.可変T細胞を含む、アッセイされるT細胞サンプルを与える工程と;
e.抗原提示細胞によって提示される抗原に反応して特異的なT細胞活性化が可能になる条件で、T細胞サンプルと、抗原提示細胞とをin vitroで接触させる工程と;
f.T細胞サンプルにおけるT細胞活性化度を決定する工程と
を含む、方法を提供する。
この方法は、(a’)貪食可能な粒子に候補ポリペプチドを密に会合させる工程、および/または(a”)粒子に会合した候補抗原に対して変性洗浄を行う工程をさらに含んでいてもよい。したがって、第1の態様の方法は、
a.貪食可能な粒子を与える工程と;
b.粒子に候補抗原ポリペプチドを密に会合させる工程と;
c.会合したポリペプチドを含む粒子に対して変性洗浄を行い、エンドトキシンのレベルを、本方法の後の任意の工程(d〜h)を妨害しないほど十分に低くする工程と;
d.可変抗原提示細胞を与える工程と;
e.抗原提示細胞によって粒子の貪食作用が可能になる条件で、洗浄された粒子と、抗原提示細胞とをin vitroで接触させる工程と;
f.可変T細胞を含む、アッセイされるT細胞サンプルを与える工程と;
g.抗原提示細胞によって提示される抗原に反応して特異的なT細胞活性化が可能になる条件で、T細胞サンプルと、粒子と接触した抗原提示細胞とをin vitroで接触させる工程と;
h.T細胞サンプルにおけるT細胞活性化度を決定する工程と
を含んでいてもよい。
候補抗原ポリペプチドは、好ましくは少なくとも50個のアミノ酸、さらにより好ましくは少なくとも75個のアミノ酸、最も好ましくは少なくとも100個のアミノ酸を含む。候補抗原ポリペプチドは、Protein Epitope Signature Tag(PrEST)の形態であってもよい。PrESTからなる大きなタンパク質ライブラリー、例えば、Human Atlas Projectの中で作製されたライブラリーが既に存在する(Uhlen M、Fagerberg L、Hallstrom BM、Lindskog C、Oksvold P、Mardinoglu Aら、Proteomics.Tissue−based map of the human proteome.Science.2015;347(6220):1260419)。PrESTは、対応する全長タンパク質に固有のアミノ酸配列を含み、ほとんどのヒトタンパク質に対する抗体を作成するための大規模Human Atlas Projectで使用されてきた。同じヒトタンパク質のためにいくつかのPrESTを使用すると、関連するT細胞エピトープを発見する機会が増える。APCによる抗原の分解は均一なプロセスではないため、APCによって提示されるフラグメントより大きな抗原ポリペプチドは、各抗原の多種多様なエピトープが提示されることも確実にする。言い換えると、特定の大きさを有するポリペプチド抗原の使用は、APCの貪食経路を利用することによって可能になり、細胞処理を行わずに直接的にMCH受容体に結合することができる短いペプチド中の抗原を提示することと比較すると、より少ない抗原ポリペプチドを使用し、優れた抗原の検出が可能になる。
a.自己免疫疾患に罹患した組織または細胞の中で高度に発現することが知られているポリペプチド。例は、プロインスリン、ミエリン関連タンパク質である。
b.新生物疾患に関わりがあることが知られているポリペプチド。例は、エストロゲン受容体、上皮成長因子受容体、サイクリン依存性キナーゼ1である。
c.自己免疫疾患に関わりがあることが知られているポリペプチド。例:ミエリンオリゴデンドロサイトタンパク質、ミエリン塩基性タンパク質、トランスアルドラーゼ。
d.感染症に関わりがあることが知られているポリペプチド。例:ウイルスキャプシド抗原、細菌エンテロトキシン。
e.アレルギーまたは同様の過敏症に関わりがあることが知られているポリペプチド。例:Can f 1、Equ c 1、Fel d 1。
f.既知の腫瘍抗原;例えば、p53、ERBB2(Erb−B2受容体チロシンキナーゼ2)、PIK3CA(ホスファチジルイノシトール−4,5−ビスホスフェート 3−キナーゼ)における共通の変異によって作られるネオアンチゲン。
g.既知の改変タンパク質、例えば、シトルリン化されたバリアントまたはリン酸化タンパク質。
粒子は、抗原提示細胞(APC、以下に論じる)によって貪食可能である。APCは、多くの異なる材料および形状の粒子を貪食することができる。これとは対照的に、粒子の大きさは、貪食作用のために限定される。大きさが小さすぎると、抗原提示細胞は、粒子を貪食するように誘発されないだろう。大きさが大きすぎると、抗原提示細胞に適合しないため、粒子を貪食することができないだろう。
抗原ポリペプチドは、粒子が抗原から解離することなく、以下に記載する変性洗浄を行うことができる様式で、粒子に会合する。
変性洗浄は、候補抗原ポリペプチド中のエンドトキシンのレベルを、抗原特異性T細胞活性化度を決定する後の工程を妨害しないレベルまで下げるために行われる。
本発明の文脈において、APCは、専門的な抗原提示細胞、例えば、単球/マクロファージまたは樹状細胞である。APCは、初代細胞であってもよく、または不死化細胞であってもよい。
本方法は、T細胞活性化度を、関連するリファレンスと比較する工程をさらに含んでいてもよく、リファレンスと比較して、サンプルにおけるT細胞活性化度が高いことが、その候補抗原が、そのサンプルにおける抗原特異性T細胞活性化を引き起こすという結果の指標である。リファレンスは、「活性化」を定義するために重要であるため、活性化度は、好ましくは、リファレンスサンプルと比較して定義される。リファレンスサンプルは、例えば、患者サンプルを分析するときは正常な健康な個体に由来するサンプル、腫瘍または腫瘍を排出するリンパ節サンプルを分析するときは正常な組織由来のサンプル、治療後の効果をフォローアップするときには治療前のサンプルであってもよい。リファレンスサンプルを使用し、診断/予後判定のための閾値を設定し、抗原特異的な陽性(すなわち、活性化)または陰性(すなわち、下方制御)を決定する。
実施例4に示すように、いくつかの候補抗原を同じT細胞サンプルに対してアッセイしてもよい。好ましくは、少なくとも10の候補抗原を同じT細胞サンプルに対してアッセイする。本発明の方法を用いると、追加のサンプルが必ずしも実験室の作業を大幅に増加させるとは限らない。本発明の方法の強みは、多くの労力を要することなく多数の抗原をスクリーニングすることが容易なことである。このことの利点は、例えば、可能性がある自己抗原の大きなスクリーニングを行うことができ、真の自己抗原を発見する機会が増えることである。
本方法の第1の態様は、試験被験体において自己反応性を診断するための方法であって、
a.候補ペプチド自己抗原が密に会合した、貪食可能な粒子を与え、会合したポリペプチドを含む粒子に対して変性洗浄を行い、エンドトキシンのレベルを、後の工程を妨害しないほど十分に低くする工程と;
b.試験被験体から、可変T細胞と可変抗原提示細胞とを含む末梢血単核細胞(PBMC)サンプルを与える工程と;
c.抗原提示細胞によって粒子の貪食作用が可能になり、抗原提示細胞によって提示される抗原に反応して特異的なT細胞活性化が可能になる条件で、PBMCサンプルと、粒子とをin vitroで接触させる工程と;
d.接触したPBMC細胞において、活性化したT細胞の割合を定量する工程と;
e.定量された割合と、健康な被験体に由来する相当する定量された割合とを比較することによって、試験被験体において活性化したT細胞の割合が大きいことは、この試験被験体において候補自己抗原に対する自己反応性の指標となる工程と
を含む、方法であってもよい。
ビーズへのポリペプチド接続
遊離カルボン酸基を含むビーズに共有結合したポリペプチド。この実施例では、Dynabeads(登録商標)MyOne(商標)Carboxylic Acid(ThermoFischer Scientific)を使用した(直径が1μmの球)。ビシンコニン酸、BCA、タンパク質アッセイを用いて、首尾良い接続を確保するために、接続の前後でタンパク質濃度を測定した。数種類のポリペプチドを試験し、1mgのビーズあたり平均で48.7μg(平均:48.7、SD:20.5、N=10)が接続した。製造者の指示に従うと、ビーズ1mgあたり50μgのポリペプチドを接続させることができ、この実施例で行われた接続の効率が高かったことを示している。
変性洗浄によるエンドトキシンの除去
異なる種類の変性洗浄を用いたエンドトキシン除去を試験するために、4種類の様々な変性洗浄条件を使用した。(a)高pH(2M NaOH pH14.3)、(b)加熱(95℃)および変性剤((c)8M尿素、(d)6M塩酸グアニジン)。単球の活性は、エンドトキシンによって強く増加し、したがって、単球の活性化を残留LPSをモニタリングするために使用することができるため、単球活性化アッセイ(IL−1β/IL−6 FluoroSpot)を用い、残留するエンドトキシンのレベルを試験した。
T細胞活性化アッセイ
FluoroSpot
16人の多発性硬化症患者および9人の健康なコントロールに由来するサンプルを用い、FluoroSpotアッセイを行った。患者およびコントロールに由来するPBMCを、FluoroSpotプレート中、疑わしい自己抗原と共にインキュベートした。図3に見られるように、MS患者の何人かは、有意に多い数の活性化された細胞を示した(パネルa)。MS患者は、平均数が37.4(95%CIで13.0〜52.9)のIL−22を分泌する活性化された細胞を有し、一方、健康なコントロールは、平均数が3.3(95%CIで0.97〜5.6)の活性化された細胞を有していた。差のP値=0.0062。IL−17を分泌する活性化された細胞について、0.28(CI95%が−1.4〜2.0)に対し、患者における数は14.7(CI95%で5.3〜24.1)であり、差のP値は<0.0007である(パネルb)。しかし、IFNγを分泌する細胞について、その差は、統計的に有意ではなく、コントロールでは33(CI95%で7.1〜59.0)であるのに対し、患者では平均数が80.0(CI95%で22.3〜138)であり、P=0.495である(パネルc)。有意性は、Mann−Whitney−U検定を用いて計算した。
チミジン取り込みを用いた増殖アッセイ。オボアルブミン(OVA)免疫化マウスに由来する脾細胞を、OVAまたはBSAが接続したビーズと共にインキュベートし、抗原特異性増殖を測定した。ビーズと共にインキュベートした細胞の増殖は、刺激指数(SI)として表された。(図2に見られるように、OVA−ビーズと共にインキュベートした細胞は、増殖が増加し、SIは2.69であった(95%CIで1.6〜3.78、P<0.005))。BSAと共にインキュベートした細胞は、増殖が増加せず、SIは0.9であった(95%CIで0.7〜1.1、P=0.37)。

多発性硬化症における自己抗原の同定:T細胞活性化のアッセイとして、IFNγ/IL−22/IL−17A FluoroSpotを用いる、ビーズに接続した多数のPrESTに対するT細胞活性化の測定。このようなスクリーニングは、MS患者由来のPBMCにおいて、健康なコントロールと比較して高いT細胞応答を刺激する抗原を検出することによって、可能性のある自己抗原を同定し、どの抗原をさらに分析するかを決定するのに役立つ。
貪食可能なビーズの適切なビーズの大きさの特定
チミジン取り込みを用いた増殖アッセイを使用し、抗原特異性T細胞活性化に対するビーズの大きさの影響を試験した。オボアルブミン(OVA)免疫化マウスに由来する脾細胞を、異なる大きさのOVAが接続したビーズを用いて刺激し、抗原特異性増殖を測定した。図5に見られるように、直径が0.2μmのOVA−ビーズと共にインキュベートした細胞は、増殖が増加し、平均SIは4.1であった(95%CIで2.4〜5.8、P=0.007)。直径が1μmのOVA−ビーズと共にインキュベートした細胞は、増殖が増加し、平均SIは8.4であった(95%CIで6.1〜10.6、P<0.005)。直径が5.6μmのOVA−ビーズと共にインキュベートした細胞は、増殖を増加させることができず、平均SIは1.1であった(95%CIで0.4〜2.7、P=0.876)。
材料および方法
遊離カルボン酸基を含む常磁性ビーズへのタンパク質の共有結合。
この実施例では、直径が1μmのDynabeads(登録商標)MyOne(商標)Carboxylic Acid(ThermoFischer Scientific)を使用し、製造業者のプロトコルに従ってカップリング手順を実施した(NHS(N−ヒドロキシコハク酸イミド)およびEDC(エチルカルボジイミド)を用いた2工程の手順))。
変性洗浄によるエンドトキシンの除去。
ビーズを、E.coliで産生された組換えタンパク質と接続させた。タンパク質が接続したビーズを、いくつかの異なる変性条件で洗浄し、エンドトキシンの除去を確実にした。残留するエンドトキシンは、単球反応性アッセイ(IL1B/IL6 FluoroSpot−assay、MABTECH,Sweden)を用いて測定した。
FluoroSpot
MS中の患者対コントロールのT細胞活性化対懸濁した自己抗原を分析するためのFluoroSpotアッセイを、IFNγ/IL17/IL22 FluoroSpot(MABTECH)を用いて行った。このプロトコルは、実際には実施例4に記載した通りであったが、125種類のタンパク質の全ライブラリーの代わりに1個の抗原に対する反応だけを分析した。
オボアルブミン感作マウス由来の脾細胞を用いたチミジン取り込みによる増殖アッセイ。刺激として、ビーズ(Dynabeads(登録商標)MyOne(商標)Carboxylic Acid)に接続したオボアルブミン(SigmaAldrich)およびBSA(SigmaAldrich)を使用した。
タンパク質に接続したビーズ(Dynabeads(登録商標)MyOne(商標)Carboxylic Acid(ThermoFischer Scientific))を刺激として用い、T細胞活性化を測定するためのアッセイとして刺激FluoroSpot(IFNγ/IL17/IL22 FluoroSpot、MABTECH)を用いた、多発性硬化症の125種類のタンパク質のライブラリーに対する自己抗原スクリーニング。
オボアルブミン感作マウス由来の脾細胞およびオボアルブミンに接続したビーズを用い、異なる大きさのビーズの有効性を評価した。表面にカルボン酸を含み、直径が5.6μm、1μm、0.2μmの常磁性ビーズを、実施例1のプロトコルに従って、オボアルブミンまたはウシ血清アルブミンと接続した。
Claims (34)
- 抗原特異性T細胞活性化をアッセイするための方法であって、
a.候補抗原ポリペプチドが密に会合した、貪食可能な粒子を与え、会合したポリペプチドを含む粒子に対して変性洗浄を行い、エンドトキシンのレベルを、後の工程を妨害しないほど十分に低くする工程と;
b.可変抗原提示細胞を与える工程と;
c.前記抗原提示細胞によって前記粒子の貪食作用が可能になる条件で、前記洗浄された粒子と、前記抗原提示細胞とをin vitroで接触させる工程と;
d.可変T細胞を含む、アッセイされるT細胞サンプルを与える工程と;
e.抗原提示細胞によって提示される抗原に反応して特異的なT細胞活性化が可能になる条件で、前記T細胞サンプルと、前記粒子と接触した前記抗原提示細胞とをin vitroで接触させる工程と;
f.前記T細胞サンプルにおけるT細胞活性化度を決定する工程と
を含む、方法。 - 前記方法が、(a’)貪食可能な粒子に候補ポリペプチドを密に会合させる工程、および/または(a”)粒子に会合した候補抗原に対して変性洗浄を行う工程をさらに含む、請求項1に記載の方法。
- 抗原特異性T細胞活性化をアッセイするための方法であって、
a.貪食可能な粒子を与える工程と;
b.前記粒子に候補抗原ポリペプチドを密に会合させる工程と;
c.前記会合したポリペプチドを含む前記粒子に対して変性洗浄を行い、エンドトキシンのレベルを、後の工程を妨害しないほど十分に低くする工程と;
d.可変抗原提示細胞を与える工程と;
e.前記抗原提示細胞によって前記粒子の貪食作用が可能になる条件で、前記洗浄された粒子と、前記抗原提示細胞とをin vitroで接触させる工程と;
f.可変T細胞を含む、アッセイされるT細胞サンプルを与える工程と;
g.抗原提示細胞によって提示される抗原に反応して特異的なT細胞活性化が可能になる条件で、前記T細胞サンプルと、前記粒子と接触した前記抗原提示細胞とをin vitroで接触させる工程と;
h.前記T細胞サンプルにおけるT細胞活性化度を決定する工程と
を含む、方法。 - 前記T細胞活性化度を、関連するリファレンスと比較する工程をさらに含み、リファレンスと比較して、前記サンプルにおけるT細胞活性化度が高いことが、前記候補抗原が、前記サンプルにおける抗原特異性T細胞活性化を引き起こすという結果の指標である、請求項1〜3のいずれかに記載の方法。
- 前記T細胞サンプルにおけるT細胞活性化度を決定することが、前記サンプル中の活性化されたT細胞の割合を決定することを含む、請求項1〜4のいずれかに記載の方法。
- いくつかの候補抗原を同じT細胞サンプルに対してアッセイする、請求項1〜5のいずれかに記載の方法。
- 少なくとも10の候補抗原を同じT細胞サンプルに対してアッセイする、請求項1〜6のいずれかに記載の方法。
- 前記粒子は、最大寸法が5.6μm未満、好ましくは4μm未満、より好ましくは3μm未満、さらにより好ましくは0.5〜2μmの区間、または最も好ましくは約1μmである、請求項1〜7のいずれかに記載の方法。
- 前記粒子は、実質的に球状である、請求項8に記載の方法。
- 前記変性洗浄が、前記会合したポリペプチドを含む粒子を高いpH、例えば、少なくともpH13、より好ましくは少なくともpH14、最も好ましくは少なくともpH14.3にすることを含む、請求項1〜9のいずれかに記載の方法。
- 前記変性洗浄が、前記会合したポリペプチドを含む粒子を低いpHにすることを含む、請求項1〜10のいずれかに記載の方法。
- 前記変性洗浄が、前記会合したポリペプチドを含む粒子を高温、例えば、少なくとも90℃、より好ましくは少なくとも92℃、最も好ましくは少なくとも95℃にすることを含む、請求項1〜11のいずれかに記載の方法。
- 前記変性洗浄が、前記会合したポリペプチドを含む粒子を、十分な濃度、例えば、少なくとも5M、6M、7Mまたは8Mで、変性剤、例えば、尿素または塩酸グアニジンにさらすことを含む、請求項1〜12のいずれかに記載の方法。
- 前記変性洗浄によって、抗原中のエンドトキシンの量が、T細胞活性化アッセイにおいて、エンドトキシンの最終的な濃度が100pg/ml未満、好ましくは50pg/ml未満、より好ましくは25pg/ml未満、最も好ましくは10pg/ml未満になるような量になる、請求項1〜13のいずれかに記載の方法。
- 前記粒子が、常磁性特性を有する、請求項1〜14のいずれかに記載の方法。
- 前記候補抗原ポリペプチドが、前記粒子に共有結合している、請求項1〜15のいずれかに記載の方法。
- 前記候補抗原ポリペプチドが、金属キレートを介し、前記粒子に結合している、請求項1〜16のいずれかに記載の方法。
- 前記抗原提示細胞と前記T細胞サンプルが、同じ個体に由来する、請求項1〜17のいずれかに記載の方法。
- 前記抗原提示細胞と前記T細胞サンプルが、同じ血液サンプルに由来する、請求項1〜18のいずれかに記載の方法。
- 前記抗原提示細胞と前記T細胞サンプルが、同じ個体由来のPBMCサンプルに由来する、請求項1〜19のいずれかに記載の方法。
- 前記PBMCサンプルが、新鮮であるか、または凍結されている、請求項20に記載の方法。
- 前記T細胞サンプルが、腫瘍、好ましくは、腫瘍中のリンパ管に由来する、請求項1〜18のいずれかに記載の方法。
- 前記T細胞サンプルが、腹水に由来する、請求項1〜18のいずれかに記載の方法。
- 前記T細胞サンプルが、CD4+細胞および/またはCD8+T細胞を含む、請求項1〜23のいずれかに記載の方法。
- 前記洗浄された粒子、前記抗原提示細胞および前記T細胞サンプルを同時に接触させる、請求項1〜24のいずれかに記載の方法。
- T細胞サンプルにおけるT細胞活性化度を決定することは、IFN−γ、IL−17、IL−22、またはこれらの組合せの分泌によって反応する抗原提示細胞と接触したT細胞の割合を決定することを含む、請求項1〜25のいずれかに記載の方法。
- 前記方法が、多発性硬化症を診断するためのものであるか、または多発性硬化症の経過を追跡するためのものであり、T細胞サンプルにおけるT細胞活性化度を決定することを含み、T細胞サンプルにおけるT細胞活性化度を決定することは、IL−17および/またはIL−22の分泌によって反応する抗原提示細胞と接触したT細胞の割合を決定することを含む、請求項1〜26のいずれかに記載の方法。
- 前記T細胞サンプルにおけるT細胞活性化度を決定することが、ELISpotまたはFluoroSpot技術を用いて行われる、請求項1〜27のいずれかに記載の方法。
- 前記候補抗原ポリペプチドが、少なくとも50個のアミノ酸、好ましくは少なくとも75個のアミノ酸、最も好ましくは少なくとも100個のアミノ酸を含む、請求項1〜28のいずれかに記載の方法。
- 前記候補抗原ポリペプチドが、タンパク質エピトープシグネチャタグ(PrEST)の形態である、請求項1〜29のいずれかに記載の方法。
- 前記T細胞サンプルがヒト由来であり、前記候補抗原ポリペプチド配列がヒト由来である、請求項1〜30のいずれかに記載の方法。
- 前記候補抗原ポリペプチドが、疾患に関わりがあることが知られている抗原、または疾患に関わりがあることが疑われる抗原に由来する、請求項1〜31のいずれかに記載の方法。
- 前記候補抗原ポリペプチドが、以下のポリペプチド:
a.自己免疫疾患に罹患した組織または細胞の中で高度に発現することが知られているポリペプチド;
b.新生物疾患に関わりがあることが知られているポリペプチド;
c.自己免疫疾患に関わりがあることが知られているポリペプチド;
d.感染症に関わりがあることが知られているポリペプチド;または
e.アレルギーまたは同様の過敏症に関わりがあることが知られているポリペプチド
に由来する、請求項1〜32のいずれかに記載の方法。 - 試験被験体において自己反応性を診断するための請求項1〜33のいずれかに記載の方法であって、
a.候補ペプチド自己抗原が密に会合した、貪食可能な粒子を与え、前記会合したポリペプチドを含む粒子に対して変性洗浄を行い、エンドトキシンのレベルを、後の工程を妨害しないほど十分に低くする工程と;
b.前記試験被験体から、生存能力のあるT細胞と生存能力のある抗原提示細胞とを含む末梢血単核細胞(PBMC)サンプルを与える工程と;
c.抗原提示細胞によって前記粒子の貪食作用が可能になり、抗原提示細胞によって提示される抗原に反応して特異的なT細胞活性化が可能になる条件で、前記PBMCサンプルと、前記粒子とをin vitroで接触させる工程と;
d.前記接触したPBMC細胞において、活性化したT細胞の割合を定量する工程と;
e.定量された前記割合と、健康な被験体に由来する相当する定量された割合とを比較する工程であって、それによって、試験被験体において活性化したT細胞の割合が大きいことは、前記試験被験体において前記候補自己抗原に対する自己反応性の指標となる工程と
を含む、方法。
