本発明では、予期せぬことに、CMCとデキストリンDEXの組み合わせを用いて生体適合性マイクロニードルアレイ(MN)を製作することが見出される。したがって、本発明は、侵襲性および経皮性デリバリーシステムとして使用することができる、多糖類ベースの溶解可能なマイクロニードルベースのアレイの製作方法を提供する。本発明方法から得られるマイクロニードルアレイは、安全で生体適合性があり、インスリンなどの薬物の経皮投与に使用されうる。
カルボキシメチルセルロース(CMC)とデキストリン(DEX)とを混合して、多糖類ベースの組み合わせを提供すること;および

を含む方法を提供する。
MNアレイのための鋳型の設計
図1に示すように、高速カッティングマシンとコンピュータ支援設計(CAD)システム(AutoCAD 2012)を使用して、アルミニウム(Al)プレート上に鋳型を製作した。アルミ鋳型は、予備的な結果に基づいて設計変更された。1つのアルミニウム鋳型は、キャスティングプロセスにおいて2つのマイクロニードルアレイを製作するための2つのアレイユニットを含む。アルミ鋳型には4つの機能領域があった:(1)ニードル領域(NA);および(2)ブランク領域(BA);(3)リッジ領域(RA);および(4)接続トレンチ(CT)。鋳型の中央のNAは、10×10の円錐形のニードルアレイの2つのグループを有する。NAは、靭性を確保し、金型から開放されたときの応力破壊を回避するためにモデルに厚さを提供するBAに囲まれた。RAは、各シングルアレイユニットを分離するために使用され、CTは、粘性のある溶液の流れを遅くし、迅速な注入中に気泡が発生するのを避けるのを補助した。
それらの生体適合性および水溶性に基づいて、我々は、溶解可能なMNを製作するのに適した分子を同定するために、最初にそれらの物理的特性を分析するために4種の異なる多糖類を選択した。セルロース誘導体であるCMCは、さまざまな食品(Chilloら、2007)および医療用途で広く使用されている水溶性天然多糖類である。DEXは、デンプンの加水分解によって生成されるさまざまなサイズの水溶性グルコースポリマーを含む。DEXは、食品添加物として食品産業で、および多くの医薬品において使用されている。高分岐アミロペクチン(AMP)は、デンプンの2つの主要成分の1つである水溶性多糖類である。AMPは、食品増粘などのさまざまな用途において使用される。トレハロース(TRE)は、α,α-1,1-グルコシド結合によって連結された2つのグルコース単位を含む天然の二糖類である(Higashiyama、2002)。TREは、天然に広く分布しており、産業用途にとって魅力的な物質である。この糖類は、食品業界で甘味料として使用される(Higashiyama、2002)。
すべての多糖類の含水率を、赤外線水分計MA150、Sartoriusを用いて測定した。その粉末形態の約0.1gの各糖を加熱し、水分のパーセンテージについて分析した。キャスティングプロセスにおける多糖類物質の加熱温度範囲を得るために、Setaram Labsys-TGDSC、DSC131装置を使用してTGAを実施した。その粉末形態の各糖0.008gのサンプルを、室温から500℃まで加熱速度5℃/分にてTGA分析のために加熱した。粉末形態の材料を、Rigaku(著作権)TTRAXIII粉末X線回折装置を用いるXRD分析にも用いて、材料の結晶化および分子配列に関する情報を得た。
MNキャスティングのために、65℃で真空蒸着を行った。多糖類溶液(5%w/v)を、それらをリン酸緩衝液に溶解し、完全分散および溶解のために60分間超音波処理することによって調製した。気泡を除去するために、溶液を室温にて10分間3500rpmで遠心分離した。次に、溶液をアルミニウム鋳型の縁に沿ってゆっくりと注入した。溶液を入れた鋳型を室温で15〜20分間真空中に保持して気泡を持ち上げ、目に見える気泡が観察されなくなるまで繰り返した。鋳型を室温から65℃に加熱し、定常温度65℃で24時間まで加熱した。昇温速度は1℃/分であった。
キャスティング後、多糖類フィルムまたは製作されたマイクロニードルの物理的性質を、TGAおよびXRD分析(セクション2.3参照)および硬度試験によって特徴決定した。
3-(4,5-ジメチルチアゾール-2-イル)-2,5-ジフェニルテトラゾリウムブロミド(MTT)アッセイを用いて、正常なNIH 3T3(マウス胚細胞)細胞株でインビトロ細毒性を実施した。NIH 3T3細胞を、10%ウシ胎児血清(FBS)を含有するダルベッコ変法イーグル培地(DMEM)中で増殖させた。毒性研究のために、細胞を、10%FBSを含有するDMEM中の24ウェルプレートに5,000細胞/ウェルの播種密度で播種し、試験化合物の添加前に、5%CO2で37℃にて24時間インキュベートした。ポリマーをPBSに溶解し、0.22μMフィルターで濾過した。24時間後、0.05%(w/w)濃度の試験化合物を含む培地10μlを添加し、5%CO2で37℃にて48時間インキュベートした。実験を3回実施し、試験化合物を含まない細胞をコントロールとして供した。48時間後、PBS中のMTT溶液(5mg/ml)200μlを各ウェルに加え、37℃で4時間インキュベートした。MTTを含む培地を除去した後、形成されたホルマザン結晶を150μLのジメチルスルホキシドに可溶化し、540nmにおける吸光度をマイクロプレートリーダーを用いて測定した。細胞阻害%=100-Abs(薬物)/Abs(コントロール)×100(ここで、Absは540nmでの吸光度を意味する)を用いて細胞阻害のパーセンテージを決定した。
研究で使用されたヌードマウス、BALB/cAnN.Cg-Foxn1nu/Cr1Nar1は、台湾の国立実験動物センターから入手した。MNを、包帯バンドルを用いてマウスに取り付けた。処置後、皮膚を外科的に剥がし、組織切片用に準備した。
本研究では、経皮ドラッグラリーデリバリーのための生分解性MNを製造する方法を提示する。適切な材料を選択するために、多糖類であるCMC、DEX、TRE、およびAMPをその粉末形態で分析して、それらの含水率、結晶性および熱安定性を調べた。多糖類をさまざまな比率で混合して、水性緩衝液中のそれらの溶解度を試験した。続いて、個々のまたは組み合わせた可溶性多糖類を鋳型表面上に注ぎ、それらのフィルム形成をチェックした。個々の多糖類の細胞毒性も試験した。

AC:完全に被覆、C:部分的に被覆、LC:管を被覆することはできない、ゆっくりと流れる。
DEXと組み合わせたCMCの粘度試験を行った(図6)。3つの異なる水性緩衝液(脱イオン水、リン酸緩衝液, pH7.4およびPBS, pH7.4)を使用して、CMCおよびDEXをさまざまな比率で溶解させた(100% CMC、50% CMC+50% DEX、34% CMC+66% DEX、66% CMC+34% DEX、および60% CMC+40% DEX)。材料を混合することにより、流体の性質が大きく変化した。CMC比が高くなると、粘度が上昇した(図5)。緩衝液中に塩が存在するため、溶媒は粘度をわずかに増加させた。緩衝液中に塩が存在するので、溶媒は、粘度をわずかに増加させた。溶媒を置換しても粘度を低下させることはできなかった。しかしながら、溶媒PBSは、純粋なCMCの粘度を有意に増加させた。最後に、MNキャスティングのPBを選択した。
アルミ鋳型は、モデルキャスティングのためのいくつかの利点を提供する。それは、従来のMNキャスティング方法と互換性がある(Leeら、2008;Migalskaら、2011;Parkら、2016)。Al ASTM 6061(FDAにより食品容器としての使用が許可されている)は、重い構造体のための耐食性の高い材料であり、本研究では再使用可能な鋳型を準備するために使用された。MNの製作のために、多糖類混合物をAl鋳型上に注ぎ、65℃で24時間乾燥させた。最終製品が、多糖類と共に生体分子および薬物で製造されるので、キャスティングには低温を使用した。標的分子の熱安定性は、乾燥プロセス中に考慮された(図3Bおよび表2)。図5は、約480μmの長さ、370μmの基板、および600μmの針中心間隔の多糖類ベースのMNのSEMおよびOM観察を示す。
鋳型の分離後の最終製品の硬さを保証するために鉛筆硬度試験を行った。表4に鉛筆硬度試験の詳細を示す。DEXと組み合わせたCMCは、CMC単独よりも優れた硬度を示した。多糖類の組み合わせは、脱イオン水と比較して、リン酸緩衝液中で硬度の増加を示した。

インビボ皮膚浸透試験のために、BSAと組み合わせたCMC/DEX混合物をMN製作に使用した。MNをマウスに24時間付着させた。H&Eで染色されたマウス皮膚切片の光学顕微鏡観察は、その表面におけるマイクロニードルの貫通を明らかに示した(図9A、9B)IHC+H染色は、皮膚に貫通した後のBSAの放出(ダークブラウンとして強調された)の追跡を示した(図9C、9D)。H&EおよびIHC+H染色を組み合わせて、さらなる確認を行い、ドラッグデリバリー法の完全性を評価した。製造されたMNアレイをヌードマウスの皮膚に使用することができ、その結果、BSAの浸透および放出が成功したことが示された。製作したMNアレイを、ヌードマウスの皮膚に使用することができ、その結果、BSAの浸透および放出が成功したことが示された。
MNの溶解を評価するために、ヌードマウスの皮膚に挿入する前と挿入した後のCMC針の外観を確認した(図11)。4時間(図11A)および8時間(図11B)の処置後、針の外観において、有意な視覚的変化は観察されなかった。24時間のMN治療後、針の外観は、明らかに変化した(図11C)。体液により溶解し、吸収されるので、針はより緩く柔らかになった。この結果は、24時間までの処置後にMNが部分的に溶解することも示した。したがって、MNは、ドラッグデリバリーにおける長期間の放出に適している可能性がある。

- 治療薬の経皮デリバリーためのマイクロニードルベースのアレイを製作するための方法であって、
カルボキシメチルセルロース(CMC)とデキストリン(DEX)とを混合して、多糖類ベースの組み合わせを提供すること;および
該治療薬を該多糖類ベースの組み合わせに加えること;
を含む方法。 - 多糖類ベースの組み合わせが、30〜70%のCMCおよび30〜70%のDEXを含む、請求項1に記載の方法。
- 多糖類ベースの組み合わせが、50%のCMCおよび50%のDEXを含む、請求項2に記載の方法。
- 治療薬が、薬物である、請求項2に記載の方法。
- 治療薬が、インスリンである、請求項2に記載の方法。
- カルボキシメチルセルロース(CMC)とデキストリン(DEX)との多糖類ベースの組み合わせからできている、治療薬の経皮デリバリー用マイクロニードルアレイ。
- 請求項1〜4のいずれか1つに記載の方法によって製造されるマイクロニードルアレイ。
- 多糖類ベースの組み合わせが、30〜70%のCMCおよび30〜70%のDEXを含む、請求項6に記載のマイクロニードルアレイ。
- 多糖類ベースの組み合わせが、50%のCMCおよび50%のDEXを含む、請求項8に記載のマイクロニードルアレイ。
- 治療薬が、インスリンである、請求項6に記載のマイクロニードルアレイ。
