Abstract
Description
即ち、本発明は、
項1.網膜組織を含む細胞凝集体であり、前記網膜組織におけるChx10陽性細胞の存在割合が20%以上100%以下である細胞凝集体を、Wntシグナル経路作用物質及びFGFシグナル経路阻害物質を含む無血清培地又は血清培地中で、RPE65遺伝子を発現する細胞が出現するに至るまでの期間中に限り、培養した後、得られた「RPE65遺伝子を発現する細胞が出現していない細胞凝集体」を、Wntシグナル経路作用物質を含まない無血清培地又は血清培地中で培養する工程を含む、毛様体周縁部様構造体を含む細胞凝集体の製造方法(以下、本発明製造方法と記すこともある。);
項2.RPE65遺伝子を発現する細胞が出現していない細胞凝集体が、前記網膜組織におけるChx10陽性細胞の存在割合が30%から0%の範囲内である細胞凝集体である前項1記載の製造方法;
項3.得られた「RPE65遺伝子を発現する細胞が出現していない細胞凝集体」を、前記網膜組織におけるChx10陽性細胞の存在割合が30%以上になるまで、Wntシグナル経路作用物質を含まない無血清培地又は血清培地中で培養する前項1または2記載の製造方法;
項4.前記網膜組織がヒト多能性幹細胞由来である前項1から3のいずれか一項に記載の製造方法;
項5.前項1から4のいずれか一項に記載の製造方法により製造される毛様体周縁部様構造体を含む細胞凝集体の毒性・薬効評価用試薬としての使用;
項6.前項1から4のいずれか一項に記載の製造方法により製造される毛様体周縁部様構造体を含む細胞凝集体に被検物質を接触させ、該物質が該構造体又は該凝集体に及ぼす影響を検定することを含む、該物質の毒性・薬効評価方法;
項7.前項1から4のいずれか一項に記載の製造方法により製造される毛様体周縁部様構造体を含む細胞凝集体の移植用生体材料としての使用;
項8.前項1から4のいずれか一項に記載の製造方法により製造される、有効量の毛様体周縁部様構造体を含む細胞凝集体を、移植を必要とする対象に移植することを含む、網膜組織の障害に基づく疾患の治療方法;及び
項9.毛様体周縁部様構造体の障害に基づく疾患の治療における使用のための前項1から4のいずれか一項に記載の製造方法により製造される毛様体周縁部様構造体を含む細胞凝集体;
等を提供する。
多能性幹細胞は、自体公知の方法により維持培養できる。例えば、ヒト幹細胞は、KnockoutTM Serum Replacement(KSR)を用いて培養することにより維持できる。例えば、マウス幹細胞は、牛胎児血清(fetal bovine serum, FBS)、白血病阻止因子(leukemia inhibitory factor, LIF)を添加し無フィーダー下に培養することにより維持できる。
「凝集体を形成させる」とは、細胞を集合させて細胞の凝集体を形成させて浮遊培養させる際に、「一定数の分散した幹細胞を迅速に凝集」させることで質的に均一な細胞の凝集体を形成させることをいう。例えば、多能性幹細胞を迅速に集合させて多能性幹細胞の凝集体を形成させると、形成された凝集体から分化誘導される細胞において上皮様構造を再現性よく形成させることができる。
凝集体を形成させる実験的な操作としては、例えば、ウェルの小さなプレート(96穴プレート)やマイクロポアなどを用いて小さいスペースに細胞を閉じ込める方法、小さな遠心チューブを用いて短時間遠心することにより細胞を凝集させる方法などが挙げられる。
本発明における「網膜組織」としては、例えば、網膜への分化に適した条件下で多能性幹細胞の凝集体を浮遊培養することにより当該細胞凝集体の表面に形成され得る、網膜前駆細胞又は神経網膜前駆細胞を含む上皮組織を挙げることもできる。
「神経網膜前駆細胞(neural retinal progenitor)」としては、眼杯(optic cup)の内層となる運命の細胞であって、神経網膜を構成するいずれの成熟な細胞にも分化しうる前駆細胞を挙げることができる。
例えば、「Wntシグナル経路作用物質を含む培地」とは、外来性のWntシグナル経路作用物質が添加された培地または外来性のWntシグナル経路作用物質を含む培地である。「FGFシグナル経路阻害物質を含まない培地」とは、外来性のFGFシグナル経路阻害物質が添加されていない培地または外来性のFGFシグナル経路阻害物質を含まない培地である。
細胞非接着性の培養器としては、培養器の表面が細胞との接着性を向上させる目的で人工的に処理(例えば、細胞外マトリクス等によるコーティング処理)されていないもの等を使用することがよい。細胞非接着性の培養器としては、培養器の表面が細胞との接着性を低下させる目的で人工的に処理(例えば、超親水処理)されたもの等を使用してもよい。
(A)多能性幹細胞を無血清培地中で浮遊培養することにより多能性幹細胞の凝集体を形成させる工程;及び
(B)工程(A)で形成された凝集体を、ソニック・ヘッジホッグシグナル伝達経路作用物質を含まずBMPシグナル伝達経路作用物質を含む無血清培地又は血清培地中で浮遊培養し、網膜前駆細胞を含む細胞凝集体を得る工程。
工程(A)における培養温度、CO2濃度等の培養条件は適宜設定できる。培養温度は、例えば約30℃から約40℃、好ましくは約37℃である。CO2濃度は、例えば約1%から約10%、好ましくは約5%である。
多能性幹細胞の凝集体が形成されたことは、細胞凝集体のサイズおよび細胞数、巨視的形態、組織染色解析による微視的形態およびその均一性、分化および未分化マーカーの発現およびその均一性、分化マーカーの発現制御およびその同期性、分化効率の細胞凝集体間の再現性などに基づき判断することが可能である。
かかる培地に用いられる無血清培地又は血清培地は、上述したようなものである限り特に限定されない。例えば、市販のKSR等の血清代替物を適量添加した無血清培地(例えば、IMDMとF−12の1:1の混合液に10% KSR、450μM 1−モノチオグリセロール及び1x Chemically Defined Lipid Concentrateが添加された培地)を挙げることができる。無血清培地へのKSRの添加量としては、例えばヒトES細胞の場合は、通常約1%から約20%であり、好ましくは約2%から約20%である。
工程(B)で用いられる無血清培地は、工程(A)で用いた無血清培地をそのまま用いることもできるし、新たな無血清培地に置き換えることもできる。工程(A)で用いた無血清培地をそのまま工程(B)に用いる場合、BMPシグナル伝達経路作用物質を培地中に添加すればよい。
BMPシグナル伝達経路作用物質の濃度は、多能性幹細胞の凝集体を形成する細胞の網膜細胞への分化を誘導可能な濃度であればよい。例えばBMP4の場合は、約0.01nMから約1μM、好ましくは約0.1nMから約100nM、より好ましくは約1.5nMの濃度となるように培地に添加する。
BMPシグナル伝達経路作用物質が培地に添加され、多能性幹細胞の凝集体を形成する細胞の網膜細胞への分化誘導が開始された後は、BMPシグナル伝達経路作用物質を培地に添加する必要は無く、BMPシグナル伝達経路作用物質を含まない無血清培地又は血清培地を用いて培地交換を行ってよい。これにより、培地に掛かる経費を抑えることができる。網膜細胞への分化誘導が開始された細胞は、例えば、当該細胞におけるRax遺伝子の発現を検出することにより確認することができる。GFP等の蛍光レポータータンパク質遺伝子がRax遺伝子座へノックインされた多能性幹細胞を工程(A)で用いて形成された細胞凝集体を、網膜細胞への分化誘導に必要な濃度のBMPシグナル伝達経路作用物質の存在下に浮遊培養し、発現した蛍光レポータータンパク質から発せられる蛍光を検出することにより、網膜細胞への分化誘導が開始された時期を確認することもできる。工程(B)の実施態様の一つとして、工程(A)で形成された細胞凝集体を、Rax遺伝子を発現する細胞が出現し始めるまでの間、網膜細胞への分化誘導に必要な濃度のBMPシグナル伝達経路作用物質を含みソニック・ヘッジホッグシグナル伝達経路作用物質を含まない無血清培地又は血清培地中で浮遊培養し、網膜前駆細胞を含む細胞凝集体を得る工程、を挙げることができる。
(C)多能性幹細胞を、Wntシグナル経路阻害物質を含む無血清培地中で浮遊培養することにより多能性幹細胞の凝集体を形成させる工程;
(D)工程(C)で形成された細胞凝集体を、基底膜標品を含む無血清培地中で浮遊培養する工程;及び
(E)工程(D)で培養された細胞凝集体を、血清培地中で浮遊培養する工程。
工程(C)で用いた無血清培地をそのまま本工程に用いる場合、「基底膜標品」を培地中に添加すればよい。
浮遊培養終了後、細胞凝集体をパラホルムアルデヒド溶液等の固定液を用いて固定し、凍結切片を作製する。得られた凍結切片を免疫染色し、網膜組織の層構造が形成されていることを確認する。網膜組織は、各層を構成する網膜前駆細胞(視細胞、水平細胞、双極細胞、アマクリン細胞、網膜節細胞)がそれぞれ異なるため、これらの細胞に発現している上述のマーカーに対する抗体を用いて免疫染色することにより、層構造が形成されていることを確認することができる。
(1)まず、「網膜組織を含む細胞凝集体」の凍結切片を作製する。
(2)次いで、Raxタンパク質の免疫染色を行う。Rax遺伝子を発現する細胞でGFP等の蛍光タンパク質が発現するように改変された遺伝子組換え細胞を用いた場合には、前記蛍光タンパク質の発現を、蛍光顕微鏡等を用いて観察する。得られた免疫染色像または蛍光顕微鏡像において、Rax遺伝子を発現する網膜組織領域を特定する。
(3)Rax遺伝子を発現する網膜組織領域が特定された凍結切片と同じ切片又は隣接する切片を試料として、DAPI等の核染色試薬を用いて核を染色する。そして、上記で特定されたRax遺伝子を発現する網膜組織領域の中において、染色された核の数を計測することにより、網膜組織領域の細胞数を測定する。
(4)Rax遺伝子を発現する網膜組織領域が特定された凍結切片と同じ切片又は隣接する切片を試料として、Chx10タンパク質の免疫染色を行う。上記で特定された網膜組織領域におけるChx10陽性細胞中の核の数を計測する。
(5)上記(3)及び(4)で計測された各々の核の数に基づいて、Chx10陽性細胞中の核の数を、上記で特定されたRax遺伝子を発現する網膜組織領域の核の数で除することにより、「Chx10陽性細胞の存在割合」を算出する。
ここで、好ましい培養としては、浮遊培養を挙げることができる。
無血清培地としては、基礎培地にN2またはKSRが添加された無血清培地を挙げることができ、より具体的には、DMEM/F−12培地にN2 supplement(N2,Invitrogen社)が添加された無血清培地を挙げることができる。血清培地としては、基礎培地に牛胎児血清が添加された血清培地を挙げることができる。
このような特定な期間を設定するには、前記「網膜組織を含む細胞凝集体」を試料として、当該試料中に含まれるRPE65遺伝子の発現有無又はその程度を、通常の遺伝子工学的手法又は生化学的手法を用いて測定すればよい。具体的には例えば、前記「網膜組織を含む細胞凝集体」の凍結切片をRPE65タンパク質に対する抗体を用いて免疫染色する方法を用いてRPE65遺伝子の発現有無又はその程度を調べることができる。
ここで、好ましい培養としては、例えば、浮遊培養を挙げることができる。
無血清培地としては、基礎培地にN2またはKSRが添加された培地を挙げることができる。血清培地としては、基礎培地に牛胎児血清が添加された培地を挙げることができ、より具体的には、DMEM/F−12培地に牛胎児血清が添加された血清培地を挙げることができる。
前記無血清培地又は血清培地に、既知の増殖因子、増殖を促進する添加剤や化学物質等を添加してもよい。既知の増殖因子としては、EGF、FGF、IGF、insulin等を挙げることができる。増殖を促進する添加剤として、N2 supplement(N2,Invitrogen社)、B27 supplement(Invitrogen社)、KSR等を挙げることができる。増殖を促進する化学物質としては、レチノイド類(例えば、レチノイン酸)、タウリンを挙げることができる。
このようにして製造された「毛様体周縁部様構造体を含む細胞凝集体」においては、毛様体周縁部様構造体が、2つの神経網膜組織の間の領域に形成される、すなわち、神経網膜組織と毛様体周縁部様構造体ともう一つの神経網膜組織とが連続した組織が形成されることもある。この場合、毛様体周縁部様構造体の部分が隣接する神経網膜組織に比べて薄いという特徴から、顕微鏡観察等で毛様体周縁部様構造体の存在を確認することが可能である。
本発明製造方法により製造された毛様体周縁部様構造体を含む細胞凝集体は、網膜細胞の障害に基づく疾患の治療薬のスクリーニング、疾患研究材料、創薬材料として利用可能である。本発明製造方法により製造された毛様体周縁部様構造体を含む細胞凝集体は、化学物質等の毒性や薬効の評価においても、光毒性、神経毒性等の毒性研究、毒性試験等に活用可能である。例えば、本発明製造方法により製造された毛様体周縁部様構造体を含む細胞凝集体に被検物質を接触させ、該物質が該細胞又は該組織に及ぼす影響を検定することにより、該物質の毒性または薬効を評価する。
本発明製造方法により製造された毛様体周縁部様構造体を含む細胞凝集体は、細胞損傷状態において、障害を受けた組織自体を補充する(例えば、移植手術に用いる)ため等に用いる移植用生体材料として用いることができる。例えば、本発明製造方法により製造された、有効量の毛様体周縁部様構造体を含む細胞凝集体を、移植を必要とする対象に移植することにより、網膜組織の障害に基づく疾患を治療する。
RAX::GFPノックインヒトES細胞(KhES-1由来;Nakano, T. et al. Cell Stem Cell 2012, 10(6), 771-785)を「Ueno, M. et al. PNAS 2006, 103(25), 9554-9559」、「Watanabe, K. et al. Nat Biotech 2007, 25, 681-686」に記載の方法に準じて培養した。培地にはDMEM/F−12培地(Sigma)に20% KSR(KnockoutTM Serum Replacement;Invitrogen)、0.1mM 2−メルカプトエタノール、2mM L−グルタミン、1x 非必須アミノ酸、8ng/ml bFGFを添加した培地を用いた。培養された前記ES細胞を、TrypLE Express(Invitrogen)を用いて単一分散した後、単一分散されたES細胞を非細胞接着性の96穴培養プレート(スミロン スフェロイド プレート,住友ベークライト社)の1ウェルあたり1.2×104細胞になるように100μlの無血清培地に浮遊させ、凝集体を速やかに形成させた後、37℃、5% CO2で培養した。その際の無血清培地には、F−12培地とIMDM培地の1:1混合液に10% KSR、450μM 1−モノチオグリセロール、1x Chemically Defined Lipid Concentrate、20μM Y27632を添加した無血清培地を用いた。浮遊培養開始後6日目に終濃度1.5nMのBMP4を添加して浮遊培養を継続した。ウェル内の培養液の半量を3日おきに、BMPシグナル伝達経路作用物質を添加していない上記培地に交換した。
このようにして調製された浮遊培養開始後18日目の細胞凝集体を4% パラホルムアルデヒド固定し凍結切片を調製した。調製された凍結切片につき、GFP蛍光像の蛍光顕微鏡観察(図1A)及び神経網膜前駆細胞のマーカーの1つであるChx10の免疫染色(図1B)、又は、細胞の核を染色するDAPI染色(図1C)を行った。網膜組織細胞の全体の存在を示す図1Aと、Chx10陽性細胞の存在を示す図1Bとの比較から、上記のようにして製造された細胞凝集体に含まれる網膜組織には、Chx10陽性細胞が80%程度存在していることが確認された(図1)。
実施例1に記載された方法により製造された浮遊培養開始後18日目の網膜組織を含む細胞凝集体を、Wntシグナル経路作用物質及びFGFシグナル経路阻害物質を含まない血清培地(DMEM/F−12培地に、10% 牛胎児血清、1% N2 supplement、0.5μM レチノイン酸、及び100μM タウリンが添加された培地)で6日間すなわち浮遊培養開始後24日目まで(図2A,B)、又は、17日間すなわち浮遊培養開始後35日目まで(図2C,D)浮遊培養した。
得られた細胞凝集体を4% パラホルムアルデヒド固定し凍結切片を調製した。調製された凍結切片につき、神経網膜前駆細胞のマーカーの1つであるChx10の免疫染色(図2A,C)、又は、網膜色素上皮細胞のマーカーの1つであるMITFの免疫染色(図2B,D)を行った。浮遊培養開始後24日目でも浮遊培養開始後35日目でも、細胞凝集体に含まれる網膜組織は80%程度がChx10陽性細胞であり(図2A,C)、MITF陽性細胞の存在割合は3%以下である(図2B,D)ことがわかった。
実施例1に記載された方法により製造された浮遊培養開始後18日目の網膜組織を含む細胞凝集体を、Wntシグナル経路作用物質(3μM CHIR99021)及びFGFシグナル経路阻害物質(5μM SU5402)を含む無血清培地(DMEM/F−12培地に、1% N2 supplementが添加された培地)で6日間すなわち浮遊培養開始後24日目まで(図3A,B)浮遊培養し、さらにWntシグナル経路作用物質(3μM CHIR99021)を含みFGFシグナル経路阻害物質を含まない血清培地(DMEM/F−12培地に、1% 牛胎児血清及び1% N2 supplementが添加された培地)で11日間すなわち浮遊培養開始後35日目まで(図3C,D)浮遊培養した。
得られた細胞凝集体を4% パラホルムアルデヒド固定し凍結切片を調製した。調製された凍結切片につき、神経網膜前駆細胞のマーカーの1つであるChx10の免疫染色(図3A,C)、網膜色素上皮細胞のマーカーの1つであるMITFの免疫染色(図3B,D)を行った。浮遊培養開始後24日目でも浮遊培養開始後35日目でも、細胞凝集体に含まれる網膜組織は80%程度がMITF陽性細胞であり(図3B,D)、Chx10陽性細胞の存在割合は3%以下であることわかった(図3A,C)。浮遊培養開始後24日目の細胞凝集体には、RPE65遺伝子を発現する細胞は出現していない。
図2と図3の比較から、Wntシグナル経路作用物質及びFGFシグナル経路阻害物質を含む無血清培地で培養する操作により、細胞凝集体に含まれる網膜組織において、Chx10陽性の神経網膜から、MITF陽性の網膜色素上皮(RPE)様組織への運命転換が起きたことが示唆された。
実施例2に記載された方法により調製された浮遊培養開始後24日目の細胞凝集体を、Wntシグナル経路作用物質及びFGFシグナル経路阻害物質を含まない血清培地(DMEM/F−12培地に、10% 牛胎児血清、1% N2 supplement、0.5μM レチノイン酸、及び100μM タウリンが添加された培地)で、40% O2条件下で更に11日間すなわち浮遊培養開始後35日目までまたは39日間すなわち浮遊培養開始後63日目まで浮遊培養し、得られた細胞凝集体を4% パラホルムアルデヒド固定し凍結切片を調製した。
浮遊培養開始後24日目の細胞凝集体(図4A,B)、35日目の細胞凝集体(図4C,D)から調製された凍結切片につき、Chx10の免疫染色(図4A,C)、MITFの免疫染色(図4B,D)を行った。浮遊培養開始後24日目では、細胞凝集体に含まれる網膜組織は80%程度がMITF陽性細胞であり(図4B)、Chx10陽性細胞の存在割合は3%以下であった(図4A)ことから、浮遊培養開始後24日目の細胞凝集体に含まれる網膜組織は網膜色素上皮様の性質をもつことがわかった。浮遊培養開始後24日目の細胞凝集体には、RPE65遺伝子を発現する細胞は出現していない。浮遊培養開始後35日目では、細胞凝集体に含まれる網膜組織は80%程度がChx10陽性細胞であり(図4C)、MITF陽性細胞の存在割合は3%以下であった(図4D)ことから、浮遊培養開始後35日目の細胞凝集体に含まれる網膜組織は、Chx10陽性の神経網膜の性質を持つことがわかった。図3と図4の比較から、浮遊培養開始後24日目から35日目の間に、細胞凝集体に含まれる網膜組織において、網膜色素上皮様組織から神経網膜様組織への運命転換が起きたことが示唆された。
浮遊培養開始後63日目の細胞凝集体から調製された凍結切片(図5A,B,C)につき、Rax::GFP蛍光像の蛍光顕微鏡観察(図5A)、毛様体周縁部のマーカーの1つであるOtx1の免疫染色(図5B)、又は、毛様体周縁部のマーカーの1つであるRdh10の免疫染色(図5C)を行った。図5A,Bの比較から、Rax陽性の神経網膜の一部がOtx1共陽性であることがわかり、図5Cから、さらにRdh10陽性細胞があることがわかる。上記のようにして、Wntシグナル経路作用物質を含まない血清培地で39日間浮遊培養した後の細胞凝集体において、網膜組織(具体的には、神経網膜)と網膜色素上皮との境界領域に存在する組織の中には、Otx1及びRdh10のいずれか又は両方の染色が陽性である細胞(即ち、毛様体周縁部様構造体のマーカー遺伝子であるOtx1及びRdh10遺伝子のいずれか又は両方を発現する細胞)が略均一な群領域として存在しており(図5B,C)、毛様体周縁部様構造体を含む細胞凝集体が高効率で製造されたことが確認された。
網膜組織を含む細胞凝集体を以下の3条件でそれぞれ浮遊培養し、浮遊培養開始後35日目の細胞凝集体を調製した。
(培養条件A)実施例1に記載された方法により調製された浮遊培養開始後18日目の網膜組織を含む細胞凝集体を、Wntシグナル経路作用物質及びFGFシグナル経路阻害物質を含まない血清培地(DMEM/F−12培地に、10% 牛胎児血清、1% N2 supplement、0.5μM レチノイン酸、及び100μM タウリンが添加された培地)で17日間すなわち浮遊培養開始後35日目まで浮遊培養した(図6A)。
(培養条件B) 実施例1に記載された方法により調製された浮遊培養開始後18日目の網膜組織を含む細胞凝集体を、Wntシグナル経路作用物質(3μM CHIR99021)及びFGFシグナル経路阻害物質(5μM SU5402)を含む無血清培地(DMEM/F12培地に、1% N2 supplementが添加された培地)で3日間すなわち浮遊培養開始後21日目まで培養した。この浮遊培養開始後21日目の細胞凝集体には、RPE65遺伝子を発現する細胞は出現していない。さらに、当該浮遊培養開始後21日目の細胞凝集体をWntシグナル経路作用物質及びFGFシグナル経路阻害物質を含まない血清培地(DMEM/F−12培地に、10% 牛胎児血清、1% N2 supplement、0.5μM レチノイン酸、及び100μM タウリンが添加された培地)で14日間すなわち浮遊培養開始後35日目まで浮遊培養した(図6B)。
(培養条件C)実施例1に記載された方法により調製された浮遊培養開始後18日目の網膜組織を含む細胞凝集体を、Wntシグナル経路作用物質(3μM CHIR99021)及びFGFシグナル経路阻害物質(5μM SU5402)を含む無血清培地(DMEM/F−12培地に、1% N2 supplementが添加された培地)で5日間すなわち浮遊培養開始後23日目まで浮遊培養した。この浮遊培養開始後23日目の細胞凝集体には、RPE65遺伝子を発現する細胞は出現していない。さらに、当該浮遊培養開始後23日目の細胞凝集体をWntシグナル経路作用物質及びFGFシグナル経路阻害物質を含まない血清培地(DMEM/F−12培地に、10% 牛胎児血清、1% N2 supplement、0.5μM レチノイン酸、及び100μM タウリンが添加された培地)で12日間すなわち浮遊培養開始後35日目まで浮遊培養した(図6C)。
得られた浮遊培養開始後35日目の細胞凝集体をそれぞれ、蛍光実体顕微鏡(オリンパス社製SZX16)にて観察した。RAX::GFP強陽性部分である神経網膜、透過光にて色素化が観察される上皮組織である網膜色素上皮、神経網膜と網膜色素上皮の境界領域に位置する特徴的な構造を有する毛様体周縁部様構造体の有無を観察し、神経網膜のみを含む細胞凝集体の割合(図6、灰色棒グラフ)、毛様体周縁部様構造体と連続した神経網膜を含む細胞凝集体の割合(図6、黒棒グラフ)、神経網膜を含まず網膜色素上皮その他の組織を含む細胞凝集体の割合(図6、白棒グラフ)を調べた。
その結果、毛様体周縁部様構造体と連続した神経網膜を含む凝集塊の割合が、培養条件Aでは3%以下であったのに対して、本発明製造方法である培養条件Bでは30%、培養条件Cでは39%と増大することがわかった。
実施例1に記載された方法により製造された浮遊培養開始後18日目の網膜組織を含む細胞凝集体を、Wntシグナル経路作用物質(3μM CHIR99021)及びFGFシグナル経路阻害物質(5μM SU5402)を含む無血清培地(DMEM/F−12培地に、1% N2 supplementが添加された培地)で6日間すなわち浮遊培養開始後24日目まで浮遊培養した。
得られた細胞凝集体を4% パラホルムアルデヒド固定し凍結切片を調製した。調製された凍結切片につき、細胞核を染めるDAPI染色(図7A)、成熟網膜色素上皮細胞のマーカーの1つであるRPE65の免疫染色(図7B)、及び網膜色素上皮細胞マーカーの1つであるMitfの免疫染色を行った。浮遊培養開始後24日目では、細胞凝集体に含まれる網膜組織におけるRPE65陽性細胞の存在割合は1%以下であることわかった(図7A,B)。このとき、当該細胞凝集体に含まれる網膜組織は80%程度がMITF陽性細胞であった。
ヒトiPS細胞株201B7(独立行政法人理化学研究所バイオリソースセンター又はiPSアカデミアジャパン株式会社から入手可能)を「Ueno, M. et al. PNAS 2006, 103(25), 9554-9559」、「Watanabe, K. et al. Nat Biotech 2007, 25, 681-686」に記載の方法に準じて培養する。培地にはDMEM/F−12培地(Sigma)に20% KSR(KnockoutTM Serum Replacement;Invitrogen)、0.1mM 2−メルカプトエタノール、2mM L−グルタミン、1x 非必須アミノ酸、5ng/ml bFGFを添加した培地を用いる。培養された前記iPS細胞を、TrypLE Express(Invitrogen)を用いて単一分散した後、非細胞接着性の96穴培養プレート(スミロン スフェロイド プレート,住友ベークライト社)の1ウェルあたり1.2×104細胞になるように100μlの無血清培地に浮遊させ、37℃、5% CO2で浮遊培養する。その際の無血清培地には、F−12培地とIMDM培地の1:1混合液に10% KSR、450μM 1−モノチオグリセロール、1x Chemically Defined Lipid Concentrate、20μM Y27632を添加した無血清培地を用いる。浮遊培養開始1日目から15日目の何れかの時点に、終濃度1.5nMのヒト組み換えBMP4(R&D)、終濃度100ng/mlのBMP2(R&D)、終濃度100ng/mlのBMP7(R&D)、終濃度100ng/mlのGDF7(R&D)のうちの何れかを添加して浮遊培養を行う。ウェル内の培養液の半量を、BMPシグナル伝達経路作用物質をいずれも添加していない上記培地に3日おきに交換する。浮遊培養開始18日目に凝集体を一部回収し、4% パラホルムアルデヒドで固定処理を実施し凍結切片を作製する。作製された凍結切片につき、神経網膜前駆細胞マーカーの一つであるChx10の免疫染色を行うことにより、上記のようにして製造された細胞凝集体に含まれる網膜組織におけるChx10の発現を確認する。
このようにして製造された「網膜前駆細胞を含む細胞凝集体」を、スタート原料である「網膜組織を含む細胞凝集体」として用いて、本発明製造方法により、毛様体周縁部様構造体を含む細胞凝集体を製造できる。
Claims (6)
- 網膜組織を含む細胞凝集体であり、前記網膜組織におけるChx10陽性細胞の存在割合が20%以上100%以下である細胞凝集体を、Wntシグナル経路作用物質及びFGFシグナル経路阻害物質を含む無血清培地又は血清培地中で、3日間〜14日間培養し、網膜組織におけるChx10陽性細胞の存在割合が減少し、MITF陽性細胞を含む細胞凝集体を得る工程を含む、網膜色素上皮細胞を含む細胞凝集体の製造方法、
ここで、該Wntシグナル経路作用物質は、Wntにより媒介されるシグナル伝達を増強し得るものである。 - 網膜組織におけるChx10陽性細胞の存在割合が減少し、MITF陽性細胞を含む細胞凝集体が、前記網膜組織におけるChx10陽性細胞の存在割合が30%から0%の範囲内である細胞凝集体である請求項2記載の製造方法。
- 網膜組織におけるChx10陽性細胞の存在割合が減少し、MITF陽性細胞を含む細胞凝集体が、MITF陽性細胞の存在割合が80%程度である細胞凝集体である、請求項1または2に記載の製造方法。
- 前記網膜組織がヒト多能性幹細胞由来である請求項1〜3のいずれか一項に記載の製造方法。
- 前記網膜組織におけるChx10陽性細胞の存在割合が20%以上100%以下である細胞凝集体が、下記(A)及び(B)の工程を含む方法により調製され得る細胞凝集体である、請求項1〜3のいずれか一項に記載の製造方法
(A)多能性幹細胞を無血清培地中で浮遊培養することにより多能性幹細胞の凝集体を形成させる工程;及び
(B)工程(A)で形成された凝集体を、ソニック・ヘッジホッグシグナル伝達経路作用物質を含まずBMPシグナル伝達経路作用物質を含む無血清培地又は血清培地中で浮遊培養し、網膜前駆細胞を含む細胞凝集体を得る工程、
ここで、該ソニック・ヘッジホッグシグナル伝達経路作用物質は、ソニック・ヘッジホッグにより媒介されるシグナル伝達を増強し得る物質であり、該BMPシグナル伝達経路作用物質はBMPにより媒介されるシグナル伝達経路を増強し得る物質である。 - 前記網膜組織におけるChx10陽性細胞の存在割合が20%以上100%以下である細胞凝集体が、下記(C)、(D)及び(E)の工程を含む方法により調製され得る細胞凝集体である、請求項1〜3のいずれか一項に記載の製造方法
(C)多能性幹細胞を、Wnt シグナル経路阻害物質を含む無血清培地中で浮遊培養することにより多能性幹細胞の凝集体を形成させる工程;
(D)工程(C)で形成された細胞凝集体を、基底膜標品を含む無血清培地中で浮遊培養する工程;及び
(E)工程(D)で培養された細胞凝集体を、血清培地中で浮遊培養する工程。
