Abstract
Description
本出願は米国仮特許出願第61/763,630号(2013年2月12日出願)および同第61/837,743号(2013年6月21日出願)の優先権を主張する(それらのそれぞれの開示は参照によりそれらの全体が本明細書中に組み込まれる)。
テトラヒドロカンナビノール(THC)はマリファナの主要な精神活性成分である。気分を変化させる影響に加えて、THCは、鎮痛性、抗炎症性および制吐性の性質を含めて、他の様々な活性(そのいくつかが治療的価値を有する場合がある)を示すことが報告されている。THCの潜在的な治療的価値は、精神活性影響を最小限に抑え、一方で、潜在的に医療に有益である活性を保持する関連化合物についての探索を引き起こしている。
本発明は、精製された形態のアジュレム酸を調製するプロセスを提供する。本発明のプロセスは下記の工程を含む場合がある:(a)パラ−メンタ−2,8−ジエン1−オール(PMD)および5−(1,1−ジメチルヘプチル)レゾルシノール(DMHR)を反応させて、(6aR,10aR)−3−(1',1'−ジメチルヘプチル)−Δ8−テトラヒドロカンナビノール(化合物8)を形成する工程;(b)化合物8をアセチル化して、(6aR,10aR)−3−(1',1'−ジメチルヘプチル)−Δ8−テトラヒドロカンナビノールアセタート(化合物9)を形成する工程;(c)化合物9を酸化して、11−オキソ−(6aR,10aR)−3−(1',1'−ジメチルヘプチル)−Δ8−テトラヒドロカンナビノールアセタート(化合物10)を形成する工程;(d)化合物10を、過酸化水素を使用して酸化して、(6aR,10aR)−3−(1',1'−ジメチルヘプチル)−Δ8−テトラヒドロカンナビノール−9−カルボン酸アセタート(化合物11)を形成する工程(ただし、過酸化水素対化合物9のモル比は約2:1から約7:1にまで及ぶ);(e)化合物11を加水分解して、粗アジュレム酸を形成する工程;(f)粗アジュレム酸をアセチル化して、化合物11を形成する工程;および(g)化合物11を加水分解して、精製された形態のアジュレム酸を形成する工程。
A.アジュレム酸の合成に適用されるアリル酸化の最適化
メチルを11位に含む化合物9のアリル酸化を、二酸化セレン、その後、過酸化水素を用いて行うことにより、不完全に酸化された中間体、例えば、CB1高活性なHU−210を与える11位でのアルコールなどを生じさせることなく完了に至る、アジュレム酸を合成する経路が提供される。化合物9に対して8当量の過酸化水素を使用する初期の実験室実験では、十分な転換が4時間〜6時間で達成されることが示された4。図2を参照のこと。
超高純度なDMHR(5−(1,1−ジメチルヘプチル)レゾルシノール))を、Norac Pharma(Azusa、カリフォルニア州)から購入してもよい。
工程1 (6aR,10aR)−3−(1',1'−ジメチルヘプチル)−Δ8−テトラヒドロカンナビノールの調製
最初に、反応で使用されるPMD(図3における7a)の量を1.25当量から1.1当量に減らした。これは、0.1当量過剰のPMDが、DMHR(図3における6)のすべてと反応するために十分であったからであった。
以前の手順では、3当量の過酸化水素が酸化反応のために使用された。このことは、別の以前の手順で使用される8当量よりもはるかに安全であったが、依然として熱暴走の可能性があった。したがって、2当量の過酸化水素を評価した。これは、以前の安全性研究では、2当量の過酸化水素が熱暴走の危険性を劇的に低下させることが示されたからである。48時間後、反応液は、未反応アルデヒドは10%(AUC)未満であるという規格値を満たしていなかった。それにもかかわらず、反応液は粗アジュレム酸にまで至っていた(6.2%の収率、96.8%(AUC)の純度)。純度が同程度であったとしても、この収率は、3当量の過酸化水素による反応よりも著しく低かった。3当量の過酸化水素を使用したが、このように、製造期間中のこの工程の綿密な技術的管理により、良好な温度制御を保証することができる。また、400gの非GMP回分処理物のために以前に行われたように、冷水が、制御されなかった発熱反応の場合における停止選択肢として存在した。
トルエンからヘプタンへの困難な溶媒切り換えを避けるために、アセチル化反応を45℃〜55℃でヘプタンにおいて完了させた。この変更は小規模では成功し、しかし、反応混合物が、約0.5当量のピリジンが14.5gの規模で反応器に装入された後では固化した。アジュレム酸を回収することを試みるために、MTBEを加えて、沈殿物を可溶化し、HClを使用して、ピリジンを除いた。この試みは不成功であった。これは、固形物が、溶媒をロータリーエバポレーターで蒸発させている間に析出したからであった。NMR分析により、沈殿物がアジュレム酸−ピリジンの部分塩であることが示された。いくつかの実験を、塩形成の原因を明らかにするために完了した。最初に、同じ反応をより小規模(約2.5g)で行った。ピリジン(0.5当量)を混合物にゆっくり加えた。またもや、反応液が固化した。しかしながら、これは、完全な2.1当量のピリジンを加えたときに改善された。その後、反応を25℃および75℃で行い、再び、0.5当量のピリジンを加え、その後、完全な量のピリジンを加えた。両方の場合において、塩が沈殿し、しかし、ピリジンのすべてが添加されると溶解した。より大きな回分処理サイズでは、より遅い添加が要求されることになるので、この手順は、拡大可能でないと判断された。
類似する変更を、同じ反応がその都度、行われるように工程2での処理の場合のようにこの工程において行った。反応混合物の酸性化を周囲温度で行って、MTBEの加水分解を回避し、また、水による追加の洗浄を加えて、オーブンの腐食を回避した。
工程1
200ガロン(約757リットル)の反応器に、超高純度DMHR(20.0kg、1当量)、PTSA(3.40kg、0.2当量)、およびトルエン(102.3kg、5体積当量)を装入した。これに、PMD(14.18kg、1.1当量)を、回分処理物の温度を15℃〜30℃で維持しながら38分かけて加え、その後、トルエン洗浄(17.4kg、1体積当量)を行った。回分処理物を不完全な真空のもとで70℃〜80℃に加熱し、そして、トルエンが満たされるディーン・スターク・トラップを使用して、水を、一定体積のトルエンを維持しながらトルエンとの共沸蒸留によって除いた。反応が、HPLCによって2時間後には完了していると決定された。この場合、HPLCにより、カンナビジオールが全く検出されず、106:1のΔ8:Δ9比が得られた(規格値は、2.0%(AUC)以下のカンナビジオール、および、Δ8:Δ9比が4:1以上であることであった)。回分処理物を25℃および大気圧において一晩保った。
200ガロン(約757リットル)の反応器に、化合物2(25.76kg、1当量)、二酸化セレン(8.66kg、1.25当量)、テトラヒドロフラン(98.5kg、4.3体積当量)、および水(5.2kg、0.2当量)を装入した。反応器を加熱し、55℃〜65℃で維持した。20.5時間後、反応は、HPLCによって完了していると判断され、1.8%(AUC)の残留化合物2により、2.0%(AUC)以下の化合物2の規格値に合格した。この回分処理物を約3時間にわたって0℃〜10℃に冷却した。回分処理物の温度を25℃未満で維持しながら、35wt%過酸化水素(18.21kg、3当量)を加えた。その後、回分処理物の温度を10℃〜25℃に調節し、この温度で保った。これを、反応液が、化合物10はHPLCによって10.0%(AUC)以下であるという規格値を満たすまで行った。約16時間後、反応は完了していると判断され(1.7%(AUC)の残留化合物10)、反応を、回分処理物の温度を35℃未満で維持しながら20wt%チオ硫酸ナトリウム溶液(98.8kg、2当量)によりゆっくり停止させた。2時間13分後、過酸化物の痕跡が全く認められなかった。回分処理物をセライト(登録商標)のパッドでろ過し、セライト(登録商標)ケークを22.9kgのTHF(1体積当量)により洗浄した。相を分離させ、水層を捨てた。その後、反応混合物を10wt%塩化ナトリウム(51.6kg、2当量)により洗浄し、清浄化された200ガロン(約757リットル)の反応器に水(128.8kg、5当量)と一緒に戻した。これに、50wt%水酸化ナトリウム溶液(18.8kg、3.8当量)を、回分処理物の温度を55℃未満で維持しながら加えた。回分処理物を45℃〜55℃で約1時間保ち、その後、サンプルを採取した。この回分処理物は、0.1%(AUC)の残留化合物11が検出されて、化合物11はHPLCによって2.0%(AUC)以下であるという規格値を満たしていた。
40リットルの反応器に、粗アジュレム酸(5.28kg、1当量)およびヘプタン(21.8kg、6体積当量)を装入した。250リットルの反応器に、ピリジン(2.18kg、2.1当量)およびヘプタン(7.2kg、2体積当量)を装入した。その後、両方の反応器を50℃〜60℃に加熱した。40L反応器の内容物が溶解した後、溶液を250L反応器に移し、さらなるヘプタン(7.2kg、2体積当量)を使用して、40L反応器を洗浄し、これを250L反応器に移した。回分処理物の温度を50℃〜60℃で維持しながら、無水酢酸(2.50kg、1.8当量)を加え、反応混合物を2時間撹拌した。反応混合物からサンプルを採取した。この反応混合物は0.8%(AUC)の粗アジュレム酸を示し、これは、0.5%(AUC)以下の規格値を満たしていなかった。第2のサンプルを3時間後に得た。このサンプルは、0.2%(AUC)の粗アジュレム酸が検出されたことにより規格値に合格した。
40リットルの反応器に、アセチル化アジュレム酸(5.02kg、1当量)、MTBE(15.37kg、4.13体積当量)、および2NのNaOH(14.54kg、2.400当量)を、回分処理物の温度を50℃以下で維持しながら装入した。回分処理物を45℃〜55℃で4時間2分にわたって維持した。その後では、回分処理物は未反応アセチル化アジュレム酸を全く有しておらず、HPLCによる0.5%(AUC)以下のアセチル化アジュレム酸の規格値を満たしていた。反応混合物を25℃に冷却した。回分処理物の温度を25±5℃で維持しながら、回分処理物を37wt%のHCl(3.62kg、0.60体積当量)により酸性化した。30分間撹拌した後、下側の水層を分離し、有機層を水(5.53kg、1.1当量)により洗浄した。pHが2であった。さらに2回の水洗浄を行い、反応混合物はpH3に達した。これはpH>3の規格値を満たしていなかったにもかかわらず、反応を続けた。
親和性、すなわち、結合親和性は、平衡結合定数または速度論的結合速度パラメーターによって定義することができる2つ以上の別個の分子実体の間(例えば、化合物と受容体間)における結合相互作用の強さの尺度である。好適な定数またはパラメーターおよびそれらの測定単位の様々な例がこの技術分野では広く知られており、これらには、会合定数(KA);解離定数(KD)または阻害定数(Ki);会合速度定数(Kon)および解離速度定数(Koff)が含まれるが、これらに限定されない。1つの実施形態において、Kiは、[受容体]・[阻害剤]/[阻害剤が結合する受容体]に等しく、したがって、Kiは、受容体に結合する阻害剤についての平衡定数である。KAの場合には、より大きい値により、より強い、またはより大きい結合親和性が意味される。Ki(またはKD)の場合には、より小さい値により、より強い、またはより大きい結合親和性が意味される。
本発明はまた、本明細書中に記載される状態を、本発明の化合物または組成物を対象に投与することによって処置または防止する方法を提供する。
本明細書中に記載される化合物および組成物は、上記で議論されるような、また、下記においてさらに記載されるような様々な状態/疾患の処置、防止、および/または診断を行うために、培養中の細胞に、例えば、インビトロまたはエクスビボにおいて、あるいは、対象に、例えば、インビボにおいて投与することができる。
対象の処置は、当該対象の状態を求めることによって最適化することができ、その結果、例えば、処置がそれぞれの化合物の次善の用量または無効の用量により開始され、そして増大されて、当該対象における線維性疾患または炎症性疾患の処置および/または防止のための超高純度アジュレム酸の最適な用量を決定することができるようにすることができる。
本発明の超高純度な化合物(例えば、超高純度なアジュレム酸)の様々な投薬形態物を、様々な状態を先行技術のアジュレム酸よりも良好な安全性および許容性のプロフィルにより処置および/または防止するために本発明の方法において使用することができる。ある特定の実施形態において、投薬形態物は経口用投薬形態物であり、例えば、錠剤、またはカプセル、または腸溶性被覆錠剤、または浸透圧放出カプセル、または賦形剤の独特の組合せなどである。他の実施形態において、投薬形態物は、液体剤、局所用パッチ、ゲル、軟膏、クリーム、エアロゾル、または吸入配合物である。
本明細書中に記載される投薬形態物はキットにおいて提供される場合がある。キットは、(a)本明細書中に記載される方法において使用される化合物と、必要な場合には、(b)情報提供物とを含む。情報提供物は、本明細書中に記載される方法および/または本明細書中に記載される方法のための投薬形態物の使用に関する記述物、説明物、販売用資料、または他のものであることが可能である。
超高純度なアジュレム酸(JBT−101)を合成し、CB1受容体およびCB2受容体に対するその結合を以前の調製物の結合と比較した。
1.結合−CB1/CB2* CB2結合は、向精神性作用を伴うことなく抗炎症性影響および抗線維性影響を媒介するようであるので非常に望ましい性質である。
超高純度なアジュレム酸(JBT−101)を、マウスにおけるその薬理学的影響について、同様にまた、CB1およびCB2のカンナビノイド受容体の機能的なインビトロ活性化について評価した。
抗侵害受容のためのホットプレート試験
ホットプレート試験を使用して、超高純度AJAおよび他の薬理学的薬剤の鎮痛活性を、マウスがその前足を舐める反応時間、および/または、54℃〜56℃に加熱され、かつ、その温度で維持されるアルミニウム製ホットプレートに置かれた後で飛び跳ねる反応時間に基づいて測定した。Kitchen IおよびGreen PG、Differential Effects of DFP Poisoning and Its Treatment on Opioid Antinociception in the Mouse、Life Sci.、33:669−672(1983)。この試験は、CB1アゴニスト活性を測定することが示されている。
カタレプシー応答を、Pertweeによって記載されるリング試験を使用して測定した(Pertwee RG、The Ring Test.A Quantitative Method of Assessing the Cataleptic Effect of Cannabis in Mice、Br.J.Pharmacol.、46:753−763(1972))。マウスを、16cmの垂直の棒に取り付けられる直径が5.5cmの水平のワイヤリングに載せた。後足および前足をリングの反対側に置いた。周囲温度を30℃で維持し、環境は聴覚刺激および明るい光が除かれていた。応答を、マウスが5分の試験期間にわたって不動であった時間の割合として計算した。
CB1受容体またはCB2受容体がアゴニストによって活性化されるとき、Gタンパク質アルファサブユニットの親和性がGDPに対してGTPに関して増大する。結果として、GDPがGタンパク質から追い出され、GTPまたはGTPγSが結合する。放射性標識(例えば、[35S]など)がGTPγS分子に結合するならば、Gタンパク質/[35S]GTPγS複合体の形成を、液体シンチレーション分光光度法を使用して直接に測定することができる。Weiland他(1994)、Methods Enzymol、237:3−13。Griffin他、PET、285:553−560、1998。
インビトロ機能的アッセイのためのストック溶液の調製
インビトロ機能的アッセイのために、JBT−101のストック溶液をエタノールまたはDMSOにおいて調製した。
Δ9−THC[National Institute on Drug Abuse(NIDA)、メリーランド州Rockville]、インドメタシン(Sigma−Aldrich、ミズーリ州St.Louis)、およびJBT−101をピーナッツ油またはベニバナ油(食品等級)のビヒクルに溶解した。化合物を経口胃管法により20μl/kgの体積で投与した。
材料および方法
CB1受容体アッセイおよびCB2受容体アッセイは、HEK−293発現系から単離される、Perkin Elmer(マサチューセッツ州Waltham)から購入される膜調製物を伴う。試験化合物のGタンパク質共役シグナル伝達(GTP−γ−[35S])アッセイを、0.4mLのアッセイ緩衝液(50mM TRIS−HCl(pH7.4)、1mM EDTA、100mM NaCl、5mM MgCl2、0.5%(w/v)BSA)における試験化合物(250nM〜1mM)、GDP(20μM)、GTP−γ−[35S](100pM)、ならびに、hCB1膜調製物およびhCB2膜調製物(0.4pM)から成るインキュベーション混合物において行った。非特異的結合を100μMの非標識GTP−γ−Sの存在下で求め、基礎的結合を薬物の非存在下で求めた。二連のサンプルを30℃で1時間インキュベーションし、結合した複合体を前記のような反応混合物からろ過し、液体シンチレーションカウンターで計数した。特異的結合を、非特異的結合を総結合から引き、総基礎的結合−非特異的結合によって除することによって計算した。データをプロットし、GraphPad Prism(GraphPad Software,Inc.、カリフォルニア州San Diego)を用いて分析した。
CP55940(陽性コントロール)は、GTP−γ−35Sターンオーバーを、hCB1受容体およびhCB2受容体の両方を介してnM濃度で刺激した(表2;CB1についてはEC50=9.99±2.5nM、CB2についてはEC50=3.96±1.3nM)。これらの結果は、CP55,940がこれらのGタンパク質共役受容体部位においてアゴニストとして作用することを示している(図13、上段パネル)。JBT−101もまた、GTP−γ−35Sターンオーバーを、両方のカンナビノイド受容体を介して刺激し、しかし、はるかにより小さい効力により刺激した(図13、下段パネル)。CB1受容体におけるJBT−101についてのEC50が9209±2042nMであり、これに対して、CB2受容体におけるEC50が1020±92nMであった(表2)。CB1受容体に対してCB2受容体の活性化についての効力における9倍の差は、CB2受容体と結合することについてのこの化合物の12倍の選択性と一致しており、このことは、JBT−101が、CB1受容体において活性でない用量でCB2受容体を活性化するであろうことを示唆している。
被験体
Charles River(ノースカロライナ州Raleigh)から得られるメスのCD−1マウス(20g〜25g)を、ホットプレート侵害受容アッセイ、直腸温度アッセイ、およびリング不動性アッセイにおける評価のために使用した。別々のマウスを、それぞれの化合物のそれぞれの用量をこの一連の手順で試験するために使用した。マウスは、そのホームケージに居るときには餌および水を自由に取ることができた。すべての動物を、12時間の明暗サイクル(午前7時に照明開始)を伴う温度制御された(20℃〜22℃)環境で飼育した。
それぞれのマウスを一連の3つの試験で試験した。この場合、試験では、カンナビノイドCB1アゴニストにより、マウスにおけるインビボ影響(Martin他、1991)、抗侵害受容(ホットプレートアッセイ)、低下した直腸温度およびリング不動性がもたらされる。試験化合物の投与に先立って、直腸温度およびホットプレート試験でのベースライン潜伏時間をマウスにおいて測定した。後者の手順では、マウスを55℃の設定での加熱された表面(マウス低温/高温プレート痛覚消失装置、Stoelting、イリノイ州Wood Dale)に置くことが伴った。マウスが足を上げるか、または舐めるまでの時間を測定し、測定後、マウスを装置から除いた。マウスが30秒以内に足を上げなかったか、または舐めなかったならば、マウスを装置から除き、30秒の潜伏時間を記録した。ベースラインにおける温度およびホットプレート潜伏時間の測定の後、マウスにはビヒクルまたは薬物を経口胃管法により投与した。ホットプレート潜伏時間および温度を、ピーナッツ油ビヒクルまたはJBT−101の(経口胃管法による)投与の90分後に、あるいは、Δ9−THCまたはインドメタシンの(経口胃管法による)投与の60分後に再び測定した。続いて、マウスを、16cmの高さでリングスタンドに取り付けられる5.5cmのリングに載せ、動物が5分の期間の期間中に動かないままであった時間の量を記録した。加えて、マウスがリングから落ちたか、または飛び降りた回数を記録した。動物マウスが5回を超えてリングから落ちたならば、試験を終了した。
直腸温度の値を、(注入前の)コントロール温度と薬物投与後の温度との差(Δ℃)として表した。抗侵害受容を、下記のように30秒最大試験潜伏時間を使用してパーセント最大可能影響(MPE)として表した:[(試験−コントロール)/(30−コントロール)]×100。カタレプシーについての評価の期間中、マウスがリング装置の上で動かないままであった時間(秒単位)の総量(呼吸およびひげ運動については除く)を測定し、カタレプシー様行動の目安として使用した。この値を300sによって除し、100を乗じて、パーセント不動性を得た。マウスが5回を超えてリングから落ちたならば、試験を終了し、当該マウスについてのリング不動性データは分析において含まれなかった。別々の被験体間ANOVAsを使用して、それぞれの測定を分析した。コントロール(ビヒクル)からの有意な差を、必要に応じてチューキー事後検定(α=0.05)によりさらに分析した。
コントロール試験により、ピーナッツ油ビヒクル(陰性コントロール)はこれら3つの試験(ホットプレート、直腸温度、およびカタレプシー)のいずれにおいても活性でなかったことが明らかにされた[図14、図15、および図16、それぞれのパネルの左側]。インドメタシンの30mg/kgの用量もまた、有意な影響をこれら3つの測定のいずれにも及ぼさなかった。対照的に、30mg/kgおよび/または100mg/kgの経口用量において、Δ9−THC(陽性コントロール)は、それぞれの測定をビヒクル条件と比較した場合、有意な抗侵害受容[100mg/kgのみ;F(9,50)=5.71、p<0.05、図15]、リング不動性[両方の用量;F(9,48)=21.18、p<0.05、図14]、および低体温[両方の用量;F(9,50)=15.08、p<0.05、図16]をもたらした。JBT−101を、0.05mg/kgから56mg/kgにまで及ぶ経口用量でこれら3つのインビボ試験で評価した。試験された用量のどれも、ホットプレート試験における有意な抗侵害受容をもたらさず(図15、右側)、また、直腸温度における有意な変化をもたらさなかった(図16、右側)。30mg/kgおよび56mg/kgの経口用量において、JBT−101は、ビヒクル条件と比較した場合、カタレプシー試験におけるパーセントリング不動性を有意に増大させた(図14、右側。F(9,48)=21.18、p<0.05]。増大の大きさは30mg/kgおよび56mg/kgの用量において類似しており、Δ9−THCの30mg/kgの用量で認められる増大に近かった。JBT−101のより低い用量(最大で10mg/kg)はこの測定に影響を及ぼさなかった。
JBT−101(超高純度アジュレム酸)によってもたらされる薬理学的影響のプロフィルは、アジュレム酸の以前に報告された影響と著しく異なる。以前に合成された(精製されていない)アジュレム酸は効力を(Wiley(2005)において総説される)痛みおよび炎症のいくつかの前臨床モデルにおいて示している;しかしながら、これはまた、Δ9−THCおよび他の精神活性なCB1受容体アゴニストに特徴的であるインビボでの薬理学的影響のプロフィルをもたらした。これらの影響には、マウスにおける自発的活動の抑制、抗侵害受容、低体温、およびカタレプシー、ならびにラットにおけるΔ9−THC様の判別可能な刺激影響(Vann他、2007)が含まれた。これらの影響は、これらのより初期のアジュレム酸合成産物によって示される良好なCB1受容体結合親和性と一致している:Novartis化合物についてはKi=5.7nM(Dyson他、2005)、HU−239についてはKi=32.3nM(Pertwee他、2010)。さらに、これらの化合物についてのCB1/CB2結合の比率が低かった(0.10および0.19、それぞれ)。対照的に、JBT−101は、CB1受容体(Ki=628±6nM)と比較した場合、CB2受容体における12倍を超える選択的結合親和性(Ki=51±11nM)を示した。本明細書中に示されるように、JBT−101はまた、CB1受容体と比較した場合、CB2受容体の活性化についての類似する選択性を示した。そのうえ、56mg/kg(p.o.)までの用量において、Δ9−THCおよび他の精神活性カンナビノイドの投与の後で特徴的に認められる行動影響が、超高純度アジュレム酸に関しては最小限に認められた。JBT−101はリング不動性を増大させたが、より大きい用量(30mg/kgおよび56mg/kg)においてのみであった。まとめると、これらの結果は、JBT−101の影響が(非精製の)アジュレム酸のより初期の合成物の影響とは異なることを明らかにする。結論として、JBT−101の薬理学的プロフィルは、JBT−101が、CB1受容体活性をほとんど有しないCB2選択的化合物であることと一致している。
図12は、超高純度AJA、ならびに、CB2およびCB1に対して選択的な参照カンナビノイドアンタゴニストについての結合曲線を示す。
膜の調製−HEK−293T細胞をATCC(バージニア州Manassas)の指針に従って培養し、Polyfect(Qiagen、カリフォルニア州Valencia)またはFugene(Roche、ニュージャージー州Nutley)を製造者の説明書に従って使用して、SV40プロモーターに機能的に連結されるヒトCB1 cDNA(Genbank X54937)またはCB2 cDNA(Genbank X74328)によりトランスフェクションした。トランスフェクション後48時間で、細胞を、細胞スクレーパーを使用して、氷冷の膜緩衝液(20mM HEPES、6mM MgCl2、1mM EDTA、pH7.2)に集めた。細胞を窒素キャビテーションチャンバーに移し、900barの圧力を30分間加えた。圧力を解放し、細胞破片を回収し、4℃で10分間、1000gで遠心分離した。上清を回収し、遠心分離を、上清が沈殿物を含まなくなるまで繰り返した。その後、膜を、4℃で20分間の12,000gでの遠心分離によってペレット化した。膜を適量の膜緩衝液に再懸濁した。膜濃度を、BioRad(カリフォルニア州Hercules)タンパク質アッセイ色素試薬を製造者の説明書に従って使用して求めた。膜を1mg/mlに希釈し、小分け物を液体窒素において急速凍結し、−80℃で貯蔵した。
線維症動物モデル−ブレオマイシン誘導皮膚線維症
8匹のマウス(6週齢〜12週齢のC57B1)から成る群にブレオマイシン(20ug/マウス)またはビヒクルのs.c.注射を14日間与え、その後、1週間の回復を与えた。並行して、1日目に開始して、2%メチルセルロース(MC)に懸濁される超高純度AJA、またはビヒクルを、0mg/kg、2.5mg/kg、5.0mg/kg、および10mg/kgで経口胃管法によって投与した。21日目に、マウスを屠殺した。その皮膚を集め、常法による組織学アッセイ(H&E染色、トリクローム染色、およびピクロシリウスレッド染色)、免疫組織化学アッセイ(パラフィン包埋サンプルまたは凍結サンプル)、インサイチューハイブリダイゼーション・アッセイ、コラーゲンアッセイ(SIRCOL)、およびリアルタイムqRCRまたはマイクロアレイハイブリダイゼーションのためのRNA単離のために処理した。皮膚切片を注意深く特徴づけし、損傷皮膚における皮膚炎症、皮膚肥厚、コラーゲン蓄積(トリクローム)、およびコラーゲン架橋(シリウスレッド)について定量化した。
CD−1マウス(メス)を実験群に無作為に割り当て、1週間にわたって順応させた。試験化合物の投与に先立って、ベースラインでの右側後足の体積を、水置換デバイス(プレチスモメーター、Stoelting)を使用してガス(イソフルラン)麻酔下で測定した。0日目に、2%のMCに懸濁される超高純度AJA、またはビヒクルを、経口胃管法によって、0ug/kg、5ug/kg、50ug/kg、および500ug/kgで投与した。処置を施した90分後、動物には、5%エタノールにおける100mg/mlのアラキドン酸溶液の10マイクロリットルを皮下注射によって右側後足の足裏面に与えた。コントロール動物群(群1)には等体積の5%エタノール溶液を投与した。左側後足には注射しなかった。足底内注射をガス麻酔下で行った。足裏内注射の45分後、右側後足の体積を、プレチスモメーターを使用してガス麻酔下で測定した。
