整粒〜混合顆粒工程
整粒機を用いて、前記造粒物を整粒し、D-マンニトール、クロスポビドン、軽質無水ケイ酸及びスクラロースを加え、混合機を用いて混合した。次に、タルク及びステアリン酸カルシウムを加え混合機を用いて混合し、打錠用顆粒とした。
打錠工程
打錠機を用いて、前記打錠用顆粒を1錠中のミチグリニドカルシウム水和物の量が10mg及びボグリボースの量が0.2mgとなるように打錠し、錠剤を得た。
得られた錠剤の初期硬度の平均は64N、錠厚は2.6mm、崩壊試験法(日局一般試験法)に従い、精製水を用いて試験を行った(補助盤なし)場合、崩壊時間は22秒であった。
(試験例1:ミチグリニドカルシウム水和物の溶出試験)
前記実施例1及び比較例1で得られた錠剤につき、試験液として、日本薬局方の溶出試験液第1液(pH1.2)を用い、日本薬局方一般試験法溶出試験第2法(パドル法)50rpmにより、溶出試験を行った。結果を図1に示す。
図1の結果から、結晶セルロースを16%添加した実施例1の方が、6%添加した比較例1よりも、優れた溶出挙動を示した。

For the tablets obtained in Example 1 and Comparative Example 1, the first pharmacopeia dissolution test solution (pH 1.2) was used as a test solution, and the second pharmacopeia dissolution test method 2 (paddle). Method) The dissolution test was performed at 50 rpm. The results are shown in FIG.
From the results of FIG. 1, Example 1 to which 16% of crystalline cellulose was added showed better elution behavior than Comparative Example 1 to which 6% was added.
