Abstract
Description
これらのCMVワクチンストラテジーは、糖タンパク質B(gB)、pp65およびIE−1を潜在標的として評価し、これらは弱毒CMV Towne株(Jacobson,Sinclair et al.2006)、全長抗原およびエピトープをコードする組換えウイルスベクター(Bernstein,Reap et al.2009;Zhong and Khanna 2009)、DNAワクチン(Wloch,Smith et al.2008)、濃染顆粒ワクチン(Frankenberg,Pepperl−Klindworth et al.2002)、およびサブユニットワクチン(Drulak,Malinoski et al.2000)を含む多くの送達プラットフォームによって送達されてきた。しかしながら、これらのアプローチで確信的な臨床有効性を示したものはなく、臨床実施に取り入れられていない。

別の実施形態では、ヘルペスウイルスはEBVである。
また、単離されたタンパク質は同じまたは異なるヘルペスウイルス(例えば、CMVおよび/またはEBV)由来のCTLエピトープを含んでよいと考えられる。
好ましい実施形態では、介在アミノ酸またはアミノ酸配列は、プロテアソーム遊離アミノ酸またはアミノ酸配列である。
第2の態様において、本発明は、第1の態様の単離されたタンパク質をコードする単離された核酸を提供する。
好ましくは、遺伝子構築物は、前記第2の態様の単離された核酸が発現ベクター中に存在する1つ以上の調節配列に作動可能に連結されている発現構築物である。
第4の態様において、本発明は、第3の態様の発現構築物を含む宿主細胞を提供する。
第7の態様において、本発明は、第1もしくは第6の態様の単離されたタンパク質または第3の態様の遺伝子構築物と、薬学上許容される担体、希釈剤または賦形剤とを含む医薬組成物を提供する。
好適には、免疫刺激性分子またはアジュバントは、1つ以上のToll様受容体(TLR:toll−like receptor)アゴニストを含む。
好ましいアジュバントとしては、限定されるものではないが、モノホスホリル脂質(MPL)および/または免疫刺激性DNA、例えば、CpG ODN1826、CpG ODN2006、CpG ODN2216および/またはCpG ODN2336が含まれる。
第9の態様において、本発明は、動物において細胞傷害性Tリンパ球(CTL)免疫応答を誘導する方法であって、前記動物に第1もしくは第6の態様の単離されたタンパク質、または第7の態様の医薬組成物を投与し、それにより、前記動物において細胞傷害性Tリンパ球(CTL)免疫応答を誘導または惹起する工程を含む方法を提供する。
(i)動物から単離された1つ以上の細胞を第1または第6の態様の単離されたタンパク質と接触させる工程、および
(ii)前記1つ以上の細胞を培養し、それにより、前記1つ以上の細胞由来のヘルペスウイルス特異的CTLを増殖させる工程
を含む方法を提供する。
第11の態様において、本発明は、第10態様の工程(ii)で生産された前記CMV特異的CTLを動物に投与し、それにより、前記動物のCMV感染を予防的にまたは治療的に処置する工程を含む養子免疫療法を提供する。
第12の態様において、本発明は、動物におけるヘルペスウイルス感染の予防的処置または治療的処置に使用するための、第1もしくは第6の態様の単離されたタンパク質、または第3の態様の遺伝子構築物を提供する。
好ましくは、上述の態様によれば、動物は哺乳類である。
より好ましくは、動物はヒトである。
「ペプチド」は、50個以下のアミノ酸を有するタンパク質である。
本明細書で使用する場合、単離されたタンパク質は、単離されたポリエピトープまたはポリトープタンパク質、例えば、単離された「CMVポリエピトープ」、「EBVポリエピトープ」または単離された「CMVポリエピトープタンパク質」または「EBVポリエピトープタンパク質」と呼ばれる場合がある。
特定の実施形態では、CMVポリエピトープタンパク質は、表1(配列番号1〜21)または表3(配列番号22〜41)から選択される複数のHLAクラスI拘束CTLエピトープを含む。
また、本発明が同じまたは異なるヘルペスウイルス(例えば、CMVおよび/またはEBV)に由来するCTLエピトープの包含を企図することも認識されるであろう。従って、単離されたタンパク質の一実施形態は、CMV抗原およびEBV抗原の両方に由来するCTLエピトープを含む。
特定の実施形態では、前記複数のエピトープは、合計10〜15個のエピトープを含む。
また、国際公開第03/000720号に記載されているものなどの他のCMV CTLエピトープを使用してもよいことも認識されるであろう。
当業者には、選択されたエピトープが任意の集団、人種または他の個体群に適合するように調整可能であることが認識されるであろう。
CTLエピトープに加え、単離されたタンパク質は、介在アミノ酸またはアミノ酸配列をさらに含み得る。介在アミノ酸またはアミノ酸配列は、前記CTLエピトープアミノ酸配列の少なくとも2つの間に、または各隣接CTLエピトープアミノ酸配列の間に存在し得る。
プロテアソーム遊離アミノ酸またはアミノ酸配列の限定されない例は、AD、KまたはRであるか、またはそれらを含む。
好ましい形態では、CMVおよび/またはEBV CTLエピトープは、各エピトープのカルボキシル末端で、プロテアソーム遊離アミノ酸配列および任意選択によりTAP認識モチーフにより連結または結合されている。
また、本明細書に記載の単離されたタンパク質は、本発明の概念から逸脱することなく、さらなる修飾、変更および/または誘導体化を施すことができることも認識されるであろう。
いくつかのアミノ酸は、単離されたタンパク質の活性の性質を変化させずに、広範囲に類似する特性を有する他のアミノ酸に変更可能であること(保存的置換)は、当技術分野において十分理解されている。

本発明の単離された核酸は、動物においてin vivoでの、またはその後の組換えタンパク質精製のために宿主細胞での、組換えタンパク質発現に有用であり得る。
特定の例では、ヌクレオチド配列は、生物または細胞種のコドン選好または利用頻度に従って操作し、それにより、その生物または細胞種においてコードされるタンパク質の翻訳および発現を最適化し得る。
核酸は、限定されるものではないが、アデニン、グアニン、シトシン、チミンおよびウラシルなどの遺伝的にコードされている塩基、またはイノシン、メチルイノシンおよびメチルアデノシン、チオウリジンおよびメチルシトシンなどの修飾塩基を含み得る。

単離された核酸の一実施形態は、配列番号50〜57のいずれか1つで示され、かつ、図15に示されるようなヌクレオチド配列を含む。
いくつかの実施形態では、核酸変異体は、上記のような単離されたタンパク質の変異体をコードする。
高ストリンジェンシー条件は、
(i)42℃でのハイブリダイゼーションのためには、少なくとも約31%v/v〜少なくとも約50%v/vのホルムアミドおよび少なくとも約0.01M〜少なくとも約0.15Mの塩、42℃での洗浄のためには、少なくとも約0.01M〜少なくとも約0.15Mの塩;
(ii)65℃でのハイブリダイゼーションのためには、1%BSA、1mM EDTA、0.5M NaHPO4(pH7.2)、7%SDS、65℃を超える温度で約1時間の洗浄のためには、(a)0.1×SSC、0.1%SDS;または(b)0.5%BSA、1mM EDTA、40mM NaHPO4(pH7.2)、1%SDS;および

遺伝子構築物は、単離された核酸の増幅、クローニングおよび/または発現を容易にすることができる。
「発現ベクター」は、プラスミドなどの自己複製する染色体外ベクターか、または宿主ゲノムに組み込まれるベクターのいずれかであり得る。好適には、発現ベクターは、前記1つ以上の調節ヌクレオチド配列を提供する。「作動可能に連結される」とは、前記1つまたは複数の調節ヌクレオチド配列が本発明の組換え核酸に対して、転写を開始する、調節する、またはその他の方法で制御するように配置されることを意味する。

特定の形態において、本発明の発現構築物は、ポックスウイルスおよびアデノウイルスまたはDNAプラスミドベクターなどのウイルス起源の発現および送達ベクターを使用する構築物であり得る。
1つの特定の実施形態では、この実施形態による発現構築物は、プラスミドベクター中に存在するp7.5プロモーターなどのワクシニアウイルスプロモーターを含む。例えば、Khanna et al.,1992.J Exp Med.176 169で提供されるようなマーカーレスキュー組換えを用いたTK組換えワクシニアウイルスの生産がある。
アデノウイルスに基づく発現構築物の一形態は、少なくともE1遺伝子を欠く複製不能A5アデノウイルスに由来する。
「薬学上許容される担体、希釈剤または賦形剤」とは、全身投与に安全に使用され得る固体もしくは液体の増量剤、希釈剤またはカプセル化物質を意味する。特定の投与経路に応じて、当技術分野で周知の多様な担体が使用可能である。これらの担体は、糖類、デンプン、セルロースおよびその誘導体、麦芽、ゼラチン、タルク、硫酸カルシウム、植物油、合成油、ポリオール、アルギン酸、リン酸緩衝溶液、乳化剤、等張生理食塩水、および塩(例えば、塩酸塩、臭化物および硫酸塩を含む無機酸塩、酢酸塩、プロピオン酸塩およびマロン酸塩などの有機酸)および発熱性物質除去水を含む群から選択され得る。
ヘルペスウイルス(例えば、CMVまたはEBV)CTLを増殖させるための現行法は非常に難しく、CMV溶解液かまたは個々のペプチドエピトープのいずれかの使用に基づく場合が多い。
よって、養子免疫療法用のヘルペスウイルス特異的CTLを増殖させる方法は、
(a)動物から単離された1つ以上の細胞を本明細書で開示される単離されたタンパク質と接触させる工程、および
(b)前記1つ以上の細胞を培養し、それにより、前記1つ以上の細胞由来のヘルペスウイルス特異的CTLを増殖させる工程
を含む。
一実施形態では、本発明は、
(A)ヒトから単離された1つ以上の細胞を本明細書で開示される単離されたタンパク質と接触させる工程、
(B)前記1つ以上の細胞を培養し、それにより、前記1つ以上の細胞由来のヘルペスウイルス特異的CTLを増殖させる工程、および
(C)前記ヘルペスウイルス特異的CTLを前記ヒトに投与し、それにより、前記動物のヘルペスウイルス感染を予防的にまたは治療的に処置する工程
を含む、ヒトにおける自己養子免疫療法を提供する。
本発明がより容易に理解できるよう、また、実効下に置けるように、以下、特定の実施形態を下記の限定されない実施例により説明する。
実施例1
CMVポリエピトープタンパク質の精製および免疫原性
材料および方法
CMVポリエピトープベクターの構築
一連のCMVポリエピトープインサートを、5つの異なる抗原(pp65、IE−1、pp50、pp150およびgB)由来の複数のHLAクラスI拘束T細胞エピトープをコードするように設計した。これらのポリエピトープ配列は、13、14、15または20個の異なるHLAクラスI拘束CD8+エピトープをコードしていた(表1参照)。
化学的にコンピテントな大腸菌BL21(DE3)pLysS(Invitrogen、カリフォルニア州、USA)をCMVポリエピトープ発現ベクターで形質転換した。形質転換細胞を100μg/mLのアンピシリン(LB−Amp)を添加したルリア−ベルターニ(LB)寒天上に播種し、プレートを37℃で一晩インキュベートした。単離されたコロニーを採取し、10mlのLB−Ampブロスに植え込み、37℃、200rpmのシェーカーで一晩増殖させた。少量の一晩培養物を50mLのLB−Ampブロスに植え込み、12時間増殖させた後、培養物の1%を2LのLB−Ampブロスに移植し、その後、600nmでのO.D.が0.6となるまで増殖させた。CMVポリエピトープタンパク質誘導は、1mM/mLのIPTGを加えることにより行った。これらの細胞をさらに4時間増殖させ、12〜15%ドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動(SDS−PAGE)で非誘導サンプルと誘導サンプルを分析することにより、タンパク質発現レベルを決定した。
誘導期の終了時に、大腸菌培養物を10,000rpmで15分の遠心分離により採取し、細胞ペレットを、プロテアーゼ阻害剤カクテル(Roche、マンハイム、ドイツ)を添加した80mLの溶解バッファー(25mM Tris pH7.4、0.5%Triton(登録商標)X100、150mM NaCl、0.5mg/mLリゾチーム)に再懸濁させ、氷上で30分間インキュベートした。細胞溶解は、氷上で4×5分サイクルの音波処理により行い、各サイクル間に10分の中断を設けた。この溶解液を13,000rpmで30分間遠心分離し、上清およびペレット画分を、SDS−PAGEを用いて分析した。タンパク質の大部分はペレット画分中に封入体(IB)の形態で見られたので、IBを、撹拌下、室温で2時間、溶解バッファー(リゾチーム不含)で1回洗浄し、150mLの可溶化バッファー(100mM NaH2PO4、10mM Tris、8M尿素 pH8.0)中、4℃で一晩、可溶化した。可溶性タンパク質を13,000rpmで30分の遠心分離により清澄化し、上清をポリエピトープタンパク質の精製に用いた。
健康なウイルス保有者由来の末梢血単核細胞(PBMC)を25μgの精製ポリエピトープタンパク質とともに、37℃、6.5%CO2にて2時間インキュベートした。インキュベーション後、これらのPBMCを非パルス刺激PBMCと混合し、10%FCSを添加したRPMI 1640培地(増殖培地と称する)に再懸濁させた。これらの細胞を24ウェルプレートで10日間、37℃、6.5%CO2で培養した。3日目と6日目に、培養物に100Uの組換えIL−2を含有する1mLの増殖培地を添加した。これらのin vitro増殖細胞のT細胞特異性を、標準的なICSアッセイを用いて評価した。さらに、マルチパラメータフローサイトメトリーを用い、これらの培養物中のT細胞の多機能的能力も評価した。
エプスタイン・バーウイルス(EBV)で形質転換したLCLおよびHEK 293細胞を、これらのアッセイにおける抗原提示細胞として使用した。これらの細胞を37℃、6.5%CO2にて2時間、25〜100μgのCMVポリエピトープタンパク質でパルス刺激した後、RPMI 1640培地で2回洗浄し、増殖培地に再懸濁させ、37℃、6.5%CO2で一晩インキュベートした。一晩のインキュベーション後、抗原提示細胞を、37℃、6.5%CO2で4時間、応答細胞と刺激細胞の比を4:1としてCMV特異的T細胞に曝し、ICSアッセイを用いてT細胞のサイトカイン発現を評価した。
CMVポリエピトープタンパク質のプロセシングに関与する種々のプロテアーゼの役割を評価するために、LCLを様々な阻害剤で前処理した後に抗原提示細胞として使用した。これらの阻害剤は、リソソーム/エンドソーム酸性化の阻害(80μMクロロキンおよび10mMバフィロマイシンA1)、リサイクル経路の阻害(200μMプリマキン)、システインプロテアーゼの阻害(100μMロイペプチンおよび100μM E64)、酸性プロテアーゼの阻害(ペプスタチンA)、自己貪食メディエーターの阻害(10mM 3−メチルアデニン(3−MA))、プロテアソーム複合体の阻害(10μMラクタシスチン、1μMエポキソミシンおよびMG132)、ゴルジ輸送の阻害(1μg/mLブレフェルジンAおよび0.7μg/mLモネンシン)またはアミノペプチダーゼ酵素の阻害(0.5mMジチオトレイトール(DTT)を含む30μMロイシンチオール)を特異的に標的とするものであった。これらの阻害剤で前処理した後、細胞を25μgのCMVポリエピトープタンパク質とともに、37℃、6.5%CO2で2時間インキュベートし、RPMI 1640培地で2回洗浄し、増殖培地に再懸濁させ、37℃、6.5%CO2で一晩インキュベートした。一晩のインキュベーション後、細胞を37℃、6.5%CO2で4時間、応答細胞と刺激細胞の比を4:1としてCMV特異的T細胞に曝し、ICSアッセイを用いてT細胞のサイトカイン発現を評価した。
ATG12 shRNA(クローンID NM_004707.2−485s1c1、CCGGTGTTGCAGCTTCCTACTTCAACTCGAGTTGAAGTAGGAAGCTGCAACATTTTT;配列番号59)またはSec61 shRNA(クローンID NM_006808.2−410s1c1、CCGGCCCAACATTTCTTGGACCAAACTCGAGTTTGGTCCAAGAAATGTTGGGTTTTTTG;配列番号60)をコードするレンチウイルスに基づくベクターを大腸菌宿主においてSigma−Aldrichから入手した。shRNAをコードするプラスミドを、ラージスケールプラスミド精製キット(Qiagen、ヒルデン、ドイツ)を用いて精製した。shRNAベクターまたは対照ベクター(pLKO.1puro)をパッケージングベクターpHR8.2ΔRおよびエンベロープベクターpCMV−VSV−G(水疱性口内炎ウイルス糖タンパク質G)とともにコトランスフェクションすることにより、HEK293T細胞内でレンチウイルスを産生させた。トランスフェクション48時間後および72時間後に、レンチウイルス含有上清を採取し、0.45μmで濾過し、−80℃で保存した。形質導入は、3×105個のCEM.T1、CEM.T2細胞またはLCLを1mLのレンチウイルス含有上清に再懸濁させ、32℃にて800gで30分間遠心分離することによって行った。形質導入の48時間後にピューロマイシン(1μg/mL)を加えた。完全なノックダウンを生じさせるために、10日目に細胞に同じレンチウイルスベクターを再感染させ、形質導入の5〜7日後に細胞を以降のアッセイに用いた。
ウエスタンブロット解析は従前に記載されている通りに行った(Ausubel 1995)。簡単に述べれば、レンチウイルスshRNA感染細胞をPBSで洗浄し、氷上にて製造者の指示に従い、RIPAバッファー(Thermo Scientific、ロックフォード、IL、USA)で溶解させた。DCタンパク質アッセイキット(Bio−Rad laboratories、ハーキュリーズ、CA、USA)を用いてタンパク質を定量した。溶解液をSDS−PAGEローディングバッファーと混合し、12〜15%のSDS−PAGEゲル上で分離した後、Mini Trans−Blot装置(Bio−Rad、CA、USA)を用い、予め冷却した転写バッファー(20%メタノールを含有する1×Tris−グリシンバッファー)中、100Vで1時間、ニトロセルロース膜に転写した。転写後、ニトロセルロース膜を洗浄バッファー(0.05%V/V Tween(登録商標)−20を含有するPBS)で3回洗浄し、次いで、シェーカー上で、ブロッキングバッファー(5%スキムミルクを含有するPBS)中、室温で1時間インキュベートした。この膜をシェーカー上、4℃で一晩、ウサギ抗Sec61(Thermo Scientific、オーストラリア)またはウサギ抗ATG12(Cell Signaling Technology、ダンバース、MA)一次抗体溶液(ブロッキングバッファーで希釈)中でインキュベートした。膜を洗浄バッファーで各回10分間6回洗浄した後、セイヨウワサビペルオキシダーゼに結合させたヒツジ抗ウサギ(Chemicon、オーストラリア)二次抗体(ブロッキングバッファーで希釈)とともに室温で1時間インキュベートした。このニトロセルロース膜を洗浄バッファーで洗浄し、ECL試薬(Merck、ダルムシュタット、ドイツ)とともにインキュベートし、タンパク質をX線フィルム上に可視化した。
統計分析は、グラフパッドソフトウエアまたはマイクロソフト(登録商標)・オフィス・エクセル2007を用いて行った。CD8+T細胞応答について、平均±SDを算出し、スチューデントのt検定を用いてp値を決定した。エラーバーはS.E.M.を表す。*、**および***で示される場合は、対照と比較した際のそれぞれp<0.05、p<0.01およびp<0.001での統計学的有意性を表す。
CMVポリエピトープタンパク質の精製および特性評価
13、14、15または20個の最小CD8+T細胞エピトープをコードするCMVポリエピトープインサートを、図1に概略を示すように設計した。これらの各ポリエピトープ配列に含まれるCMVエピトープの総覧を表1に示す。これらのCMVポリエピトープ構築物を大腸菌に形質転換し、タンパク質発現条件を最適化し、SDS−PAGEで分析した。これらの実験から得られた結果は、CMVポリエピトープタンパク質(13、14、15および20マー)が37℃、IPTG誘導プロモーター下で細菌発現系を用いて上手く発現できることを示した(図2AおよびB)。直鎖CD8+T細胞エピトープの疎水性のため、CMVポリエピトープタンパク質は封入体(IB、データは示されていない)の形態で凝集されていた。これらのIBを可溶化し、13、14または15個のエピトープをコードする構築物由来のCMVポリエピトープタンパク質を、Ni−NTAマトリックスを用いて精製した。この一段階精製プロセスは、本発明者らにこれらのCMVポリエピトープタンパク質を均質となるまで精製することを可能とした(図3A〜C)。しかしながら、CMVポリエピトープ20マーの精製は、8M尿素および6M塩酸グアニジンの2種類の異なる変性剤を使用したにもかかわらず、IBを上手く可溶化できなかった。CMVポリエピトープの可溶化の後、この20マータンパク質はペレット画分に残り、溶出画分にはタンパク質は検出されなかった(図3D)。維持に適合するバッファー系を特定するための溶解度試験から得られたデータは、CMVポリエピトープタンパク質が可溶性を維持するために酸性pHのMESバッファーまたはグリシンバッファーを必要とすることを示した(図4A)。CMVポリエピトープ精製の後、種々の濃度のタンパク質をSDS−PAGEで分析して完全性を確認した。図4B〜Dに示されるデータは、最少の不純物を示し、組換えポリエピトープタンパク質の分子量はおよそ19、21および25kDaであり、これらはそれぞれ13、14および15マーポリエピトープの理論的に計算された分子量と一致していた。この一段階精製工程は、本発明者らに2Lの培養物から80mgの13マー、4mgの14マーおよび15mgの15マータンパク質を得ることを可能とした。
CMVポリエピトープタンパク質の免疫原性を評価するために、本発明者らは、種々のHLA型のCMV血清陽性ドナーのPBMCを用いてCMV特異的CD8+T細胞を増殖させるいくつかのin vitro実験を行った。健康なドナー由来のPBMCをex vivoにて精製CMVポリエピトープタンパク質で刺激した後、ICSアッセイにより抗原特異性を評価し、ex vivo応答と比較した。これらの実験から得られたデータは、13、14および15マーのCMVポリエピトープタンパク質は、そのポリエピトープ中に含まれるエピトープに特異的なCMV特異的CD8+T細胞の迅速な増殖を誘導したことを示した(図5A)。ほとんどの場合で、優勢なCD8+T細胞応答は、CMVポリエピトープ中に含まれる複数のエピトープに対するものであった。例えば、図5Aに示されるデータは、個々のドナーPBMC由来の複数のエピトープに対するCMV特異的CD8+T細胞のパーセンテージに著しい増加を示す。さらに、本発明者らはまた、CMVポリエピトープ内の全てのエピトープがPBMCからCMV特異的CD8+T細胞を増殖させる能力があり、これらの応答は全CD8+T細胞の2〜40%の範囲であったことも示した(図5Bおよび5C)。特に注目されるものはQIKエピトープであり、これは本発明者らのin vitro増殖試験が、ポリエピトープタンパク質で刺激した後にこのエピトープに特異的なT細胞が増殖され得ることを示したからである(図5B)。これに対して、QEF特異的T細胞では最少の増殖が見られ、このエピトープはヒト細胞により効率的にプロセシングされない可能性があることが示唆される。これらの結果は、ポリエピトープタンパク質中に含まれるCD8+T細胞エピトープがヒト細胞により効率的にプロセシングおよび提示され得ること、およびヒトPBMCのポリエピトープタンパク質による感作がCMV特異的T細胞の迅速な増殖を誘導することを明らかに示す。
報告されている多くの証拠が、多機能的CD4+およびCD8+T細胞応答がある範囲のウイルスおよび微生物病原体に対する防御を提供する上で重要であることを示唆している(Betts,Gray et al.2006;Darrah,Patel et al.2007;Millington,Innes et al.2007)。さらに、CMVに関して、多機能的CD8+T細胞は肝臓移植後の高レベルのウイルス複製に対して防御する(Nebbia,Mattes et al.2008)。これらの所見は、多機能的CD8+T細胞応答が有力なCMVワクチンの開発の必要条件であることを明らかに強調する。本発明者らの続いての実験で、本発明者らは、ポリエピトープタンパク質により増殖したCMV特異的CD8+T細胞のエフェクター機能を分析した。これらの分析は、これらのエフェクター細胞が細胞溶解機能を遂行し(CD107a動員)、複数のサイトカイン(IFN−γ、TNFおよびMIP−1β)を発現する能力を評価するように設計された。これらの分析の1つからの代表的データを図6に示す。ポリエピトープによって増殖したCMV特異的CD8+T細胞の大部分は強い細胞溶解機能(CD107a動員により示される)を示し、複数のサイトカイン(IFNγ+、TNF+およびMIP1β+)を発現した。
ポリエピトープタンパク質のプロセシングおよび提示におけるスペーサー配列の厳密な役割を示すために、本発明者らは、プロテアソームリンカーを用いずに(CMVポリ)、また用いて(CMVポリ−PL)、13個の最小のCD8+T細胞エピトープをコードするように設計した(図7Aおよび7B)。CMVポリエピトープ構築物を大腸菌に形質転換し、タンパク質発現条件を最適化し、ポリエピトープタンパク質を、Ni−NTAクロマトグラフィーを用いて精製した。これらの実験から得られた結果は、CMVポリおよびCMVポリ−PLの両方が細菌発現系を用いて上手く発現され、均質となるまで精製できたことを示した。
CMVポリ、CMVポリ−PLおよびCMVポリ−PTLタンパク質のプロセシングおよび提示を検討するために、本発明者らは、ヒトリンパ芽球様細胞株(LCLs:lymphoblastoid cell lines)を一晩CMVポリ、CMVポリ−PLおよびCMVポリ−PTLとともにインキュベートした後、細胞内IFN−γ分析を用い、CMV特異的T細胞パネルの活性化を評価した。図8Aに示される代表的なFACSプロットは、CMVポリ−PLまたはPTL由来のHLA A2拘束NLV(pp65)、HLA A1拘束VTE(pp50)、HLA B7拘束RPHおよびTPR(pp65)エピトープは、CMVポリでパルス刺激されたLCLに比べ、効率的にプロセシングされ、CMV特異的T細胞に提示されたことを示す。より重要なことに、CMV特異的T細胞の活性化は、CMVポリに比べてCMVポリ−PLまたはCMVポリ−PTLでパルス刺激したLCLで刺激した後に有意に高かった(図8B)。これらのデータを考え合わせると、外因的に送達されたポリエピトープタンパク質のプロセシングおよび抗原特異的CD8+T細胞への提示を増強させるためには、エピトープ間にプロテアソームリンカーおよび/またはTAPリンカーを必要とすることが示される。
次の一連の実験で、外因的に付加されたポリエピトープタンパク質由来のCD8+T細胞エピトープのプロセシングおよび提示の厳密な経路を示すために、本発明者らはポリエピトープタンパク質をTAP+(CEM.T1)およびTAP−(CEM.T2およびCEM.T2−HLA B7)LCL中でパルス刺激した後、これらの細胞をCMV特異的T細胞に曝した。図9に示されるデータは、TAP+B細胞およびTAP−B細胞の両方がポリエピトープタンパク質由来のCD8+T細胞エピトープを効率的に提示することができることを示す。ポリエピトープ提示の機序を示すために、本発明者らは、抗原提示細胞としてCEM.T1およびCEM.T2細胞を用いてHLA A2拘束NLV特異的CD8+T細胞を刺激した。これらの抗原提示細胞をまずリソソーム/エンドソーム酸性化の阻害剤(クロロキンおよびバフィロマイシンA1)、リサイクル経路の阻害剤(プリマキン)、システインプロテアーゼの阻害剤(ロイペプチンおよびE64)、および酸性プロテアーゼの阻害剤(ペプスタチンA)で前処理し、次に、ポリエピトープタンパク質でパルス刺激した。図10Aに示されるデータは、ポリエピトープタンパク質の提示を遮断するというよりもむしろ、リソソーム、リサイクル経路およびシステインプロテアーゼ阻害剤が、ポリエピトープタンパク質でパルス刺激されたCEM.T1細胞および/またはCEM.T2細胞のT細胞認識を有意に高めたことを示す。これらの所見は、ロイペプチン、E64またはペプスタチンAによる前処理が、ポリエピトープタンパク質内のCD8+T細胞エピトープをシステインプロテアーゼおよび酸性プロテアーゼによる分解から保護し得ることを示唆する。予期しないことに、クロロキンとバフィロマイシンA1は、ポリエピトープタンパク質の交差提示に対して反対の作用を示した。クロロキンはCEM.T2細胞における抗原提示を高めたが、バフィロマイシンA1による前処理は、ポリエピトープでパルス刺激した抗原提示細胞のT細胞認識を有意に低下させた(図10A)。従前の研究では、バフィロマイシンA1は空胞型H+ATPアーゼの強力かつ特異的な阻害剤でもあり、オートファゴソームとリソソームの間の融合を阻害することによって自己貪食空胞の成熟を妨げることが示されている。ポリエピトープタンパク質プロセシングが自己貪食経路を含み得るかどうかを調べるために、本発明者らは、抗原提示細胞をPI3K阻害剤である3−メチルアデニン(3−MA)で前処理した後、CMV特異的T細胞に曝した。図10Aに示されるデータは、3−MA処理がポリエピトープタンパク質由来のCD8+T細胞エピトープの提示も果たしたことを示す。これらの所見は、ポリエピトープタンパク質の交差提示が自己貪食依存的経路を介する可能性があることを示唆する。
アジュバントと組み合わせたCMVポリエピトープタンパク質の免疫原性
材料および方法
MPLおよびCpG ODN1826を含むCMVポリエピトープワクチン製剤
CMVポリエピトープワクチンを、100μL容量中、一用量当たり20μgのCMVポリ、CMVポリ−PLまたはCMVポリ−PTLと25μgのMPL(TLR4アゴニスト)および50μgのCpG ODN1826(TLR9アゴニスト)を混合することにより調剤した。TLRアゴニストはInvivoGen(サンディエゴ、CA、USA)から購入した。
マウスMHCクラスIが破壊されたヒトHLA−A*0201を含有するHHD Iマウスを育成し、QIMRにて特定病原体不在条件下で維持した。全てのプロトコールはQIMR動物倫理委員会に準拠した。各群、少なくとも5個体(M1〜5)の6〜8週齢マウスに、上記に明示したアジュバントの組合せで調剤したCMVポリエピトープワクチンで、尾の基部から皮下(s.c.)免疫を施した。21日目に同じワクチン製剤でマウスに追加免疫を施し、35日目にマウスを屠殺し、細胞内サイトカイン染色(ICS:intracellular cytokine staining)アッセイを用いてポリエピトープ特異的CD8+T細胞応答を測定した。
マウスをCO2窒息により屠殺し、3mLのマウスT細胞培養培地(10%FBS、100IU/mLペニシリン、200μg/mL硫酸ストレプトマイシン、β−メルカプトエタノール、非必須アミノ酸およびピルビン酸ナトリウムを添加したDMEM)中に脾臓を採取した。シリンジのプランジャーで脾臓を穏やかにすりつぶすことによって単細胞懸濁液を調製した。細胞を1200rpmで5分間遠心分離し、3mLの塩化アンモニウムおよびTrisバッファー(0.89%塩化アンモニウム中0.017MのTris塩基、pH7.4)に再懸濁させた後、室温で5分間インキュベートして赤血球を除去した。細胞を遠心分離し、2%FBSを含有するPBSで2回洗浄し、5mLのマウスT細胞培養培地に再懸濁させた。余分な組織および細胞残渣を除去するために、最終細胞懸濁液を70μMのセルストレーナー(Becton Dickinson、サンディエゴ、USA)で濾過した。その後、トリパンブルー排除法を用いて細胞生存率を求めた。
ワクチン接種マウス由来のおよそ5×106個の脾細胞を、100μlのマウスT細胞培養培地中、37℃、6.5%CO2で2時間、1μgのHLA A2拘束NLVおよびVLEペプチドで刺激した。インキュベーション後、1mLのマウスT細胞培養培地を加え、細胞を24ウェルプレートに移し、37℃、6.5%CO2で10日間培養した。3日目と6日目に、培養物に、100Uの組換えIL−2を含有する1mLのT細胞培養培地を補給した。これらのin vitro増殖細胞のT細胞特異性を、標準的なIFN−γ ICSアッセイを用いて評価した。さらに、これらの培養物中のT細胞の多機能的能力も、マルチパラメータフローサイトメトリーを用いて評価した。
NLVおよびVLEによるin vitro刺激の後、50μLのマウスT細胞培養培地中のおよそ2×105個のマウス脾細胞を、必要なウェルに加えた。これらの細胞を刺激するために、各ウェルに0.2μgのNLVおよびVLEペプチドを加え、次いで、0.3μLのブレフェルジンA(BD Pharmingen、サンディエゴ、CA)を含有する150μLのDMEMを加え、37℃、10%CO2で4時間インキュベートした。2%FCSを含有するPBS(洗浄バッファー)で細胞を2回洗浄し、洗浄バッファーに再懸濁させたAPC結合抗CD3、FITC結合抗CD4およびPerCP−Cy5.5結合抗CD8モノクローナル抗体で表面染色し、4℃で30分間インキュベートした。細胞を洗浄バッファーで2回洗浄し、100μL/ウェルのCytofix/Cytopermで固定し、Perm/洗浄バッファーで2回洗浄した。次いで、細胞を4℃で30分間、PE結合抗IFN−γモノクローナル抗体で細胞内染色し、細胞をPerm/洗浄バッファーで2回洗浄し、BD FACSCanto IIに採取した。
ワクチン接種後、前述のようにex vivoにて脾細胞を刺激した。細胞を4℃で30分間、FITC結合抗CD4およびPerCP−Cy5.5結合抗CD8で表面染色した。洗浄、固定および透過処理の後、細胞をPE結合抗IFN−γ、PE−Cy7結合抗TNFおよびAPC結合抗IL2抗体で細胞内染色した。細胞をBD FACSCanto IIに採取し、FlowJoソフトウエアおよびBooleanゲート分析を用いてデータを解析した。
最初の研究で、CMVによりコードされている糖タンパク質B(gB)とポリエピトープタンパク質に基づくサブユニットワクチン製剤を、ヒト適合性TLRアゴニストと組み合わせて試験した。ポリエピトープタンパク質は、CMVの異なる抗原に由来する複数の最小HLAクラスI拘束CD8+T細胞エピトープを含んでいた。このサブユニットワクチンは、持続的抗ウイルス抗体、Th1 CD4+およびCD8+T細胞応答を生じさせた。このワクチンにより誘導された体液性免疫応答は強い中和能を示し、抗原特異的T細胞は、長期記憶維持を伴う複数のサイトカインを発現した。さらに、このサブユニットCMVワクチンは、TLR4およびTLR9の活性化を介して、抗原特異的T細胞の活性化に重要な役割を果たすIL12p70、IFN−α、IL−6およびTNF−αを発現する異なる樹状細胞(DC)サブセットを活性化した。
CMVポリ、CMVポリ−PLおよびCMVポリ−PTLの免疫原性を決定するために、本発明者らは、次に、TLR4およびTLR9アゴニストと組み合わせた場合のポリエピトープタンパク質の免疫原性を評価した。ヒトHLA A2 MHCクラスI対立遺伝子を発現するHHD−IトランスジェニックマウスをCMVポリ、CMVポリ−PLまたはCMVポリ−PTLで免疫した。ワクチン接種後、脾細胞をin vitroにてHLA A2拘束NLVおよびVLEペプチドで刺激した。in vitroで刺激した脾細胞におけるCMVポリエピトープ特異的応答の確立を分析するため、細胞内IFN−γアッセイを用い、CMVポリ特異的(HLA A2拘束エピトープNLVおよびVLE)CD8+T細胞の存在を評価した。興味深いことに、in vitroデータと一致して、CMVポリ−PLまたはCMVポリ−PTLワクチン製剤で免疫したマウスは、CMVポリワクチン製剤で免疫したマウスに比べて、有意に高い頻度のCMVポリエピトープ特異的CD8+T細胞を誘導した(図12A)。さらに、T細胞に基づくワクチンの防御有効性は多機能エフェクターの頻度に相関するという実質的な証拠がある。従って、続いての実験で、本発明者らは、CMV特異的CD8+T細胞応答の機能の質を評価した。免疫マウス由来の脾細胞のin vitro増殖後、マルチパラメトリックフローサイトメトリーを用いてIL−2、TNFおよびIFN−γ産生のパターンを決定した。図12Bに示されるデータは、CD8+T細胞がより高度な多機能性を示し、最も重要なこととしては、より高い頻度のCD8+T細胞は、CMVポリワクチンよりもCMVポリ−PLまたはCMVポリ−PTLで免疫されたマウスのIFN−γおよびTNF産生細胞であったことを明らかに示す。これらの所見を考え合わせると、TLR4アゴニストおよびTLR9アゴニストの両方をアジュバントとしたCMVポリ−PLまたはCMVポリ−PTLに基づくCMVワクチン製剤が、多機能的能力を有するCMV特異的CD8+T細胞を誘導する上で最も効果的であったことが明らかに示される。
新たに出現した証拠は、健康なCMV血清陽性個体におけるCMV特異的CD8+T細胞の応答が、複数のCMV抗原、主としてpp65およびIE1に向けられるだけでなく、他の構造的初期/後期抗原および免疫調節剤(pp28、pp50、pp150、IE2 gH、gB、US2、US3、US6およびUL18)にも向けられるということを示唆している(Elkington,Walker et al.2003;Elkington,Shoukry et al.2004;Manley,Luy et al.2004;Khan,Bruton et al.2005;Sylwester,Mitchell et al.2005)。これらのCD8+T細胞の応答は、動物モデルならびにヒトの両方において、CMVの免疫性、ウイルス複製の制御および進行性感染の臨床症状の予防に重要な役割を果たす(Quinnan,Kirmani et al.1982;Rook,Quinnan et al.1984;Reddehase,Weiland et al.1985)。これらの所見は、複数の抗原に対してT細胞応答を誘導し得るCMVに対するワクチンがCMV関連疾患に対する防御を強化する可能性があることを示す。従って、複数の抗原を標的とするために、特に、CD8+T細胞応答を誘導するために、この研究で本発明者らは、新規な組換えに基づくポリエピトープワクチン技術を提案した。ポリエピトープに基づくワクチンは、未知の特性または病原性を含む可能性のある全長抗原を使用せずに、いくつかの抗原に由来する保存されている多様なエピトープに対して免疫応答を誘導するための有力なアプローチを提供する。
アジュバントと組み合わせたEBVポリエピトープタンパク質の免疫原性
材料および方法
EBVポリエピトープ構築物の構築
EBVポリエピトープを、9種の異なる抗原(BMLF1、BRLF1、BZLF1、LMP2、LMP2a、EBNA1、EBNA3A、EBNA3BおよびEBNA3C)由来の複数のHLAクラスI拘束T細胞エピトープをコードするように設計した。エピトープHLA拘束、アミノ酸配列およびこれらのエピトープのアミノ酸位置を表3に示し、および図13Aに模式的に示す。
化学的にコンピテントな大腸菌BL21(DE3)pLysS(Invitrogen、カリフォルニア州、USA)をEBVポリエピトープ発現ベクターで形質転換した。形質転換細胞を100μg/mLのアンピシリン(LB−Amp)を添加したLuria Bertani(LB)寒天上に播種し、プレートを37℃で一晩インキュベートした。単離されたコロニーを採取し、10mlのLB−Ampブロスに植え込み、37℃、200rpmのシェーカーで一晩増殖させた。少量の一晩培養物を50mLのLB−Ampブロスに植え込み、12時間増殖させた後、培養物の1%を2LのLB−Ampブロスに移植し、その後、600nmでのO.D.が0.6となるまで増殖させた。EBVポリエピトープタンパク質誘導は、1mM/mLのIPTGを加えることにより行った。これらの細胞をさらに4時間増殖させ、15%SDS−PAGEで非誘導サンプルと誘導サンプルを分析することにより、タンパク質発現レベルを決定した。
誘導期の終了時に、大腸菌培養物を10,000rpmで15分の遠心分離により採取し、細胞ペレットを、プロテアーゼ阻害剤カクテル(Roche、マンハイム、ドイツ)を添加した80mLの溶解バッファー(25mM Tris pH7.4、0.5%Triton(登録商標)X100、150mM NaCl、0.5mg/mLリゾチーム)に再懸濁させ、氷上で30分間インキュベートした。細胞溶解は、氷上で4×5分サイクルの音波処理により行い、各サイクル間に10分の中断を設けた。この溶解液を13,000rpmで30分間遠心分離し、上清およびペレット画分を、SDS−PAGEを用いて分析した。タンパク質の大部分はペレット画分中に封入体(IB)の形態で見られたので、IBを、撹拌下、室温で2時間、溶解バッファー(リゾチーム不含)で1回洗浄し、150mLの可溶化バッファー(100mM NaH2PO4、10mM Tris、8M尿素、0.5%TritonX100 pH8.0)中、4℃で一晩、可溶化した。可溶性タンパク質を13,000rpmで30分の遠心分離により清澄化し、上清をポリエピトープタンパク質の精製に用いた。
健康なウイルス保有者由来の末梢血単核細胞(PBMC)を25μgの精製EBVポリエピトープタンパク質とともに、37℃、6.5%CO2にて2時間インキュベートした。インキュベーション後、これらのPBMCを非パルス刺激PBMCと混合し、10%FCSを添加したRPMI 1640培地(増殖培地と称する)に再懸濁させた。これらの細胞を24ウェルプレートで14日間、37℃、6.5%CO2で培養した。3日目、6日目および9日目に、培養物に100Uの組換えIL−2を含有する1mLの増殖培地を添加した。これらのin vitro増殖細胞のT細胞特異性を、標準的なICSアッセイを用いて評価した。
プロテアソームリンカーを有するEBVポリエピトープタンパク質の設計および精製
CMVに対する免疫療法のための組換えポリエピトープタンパク質の設計、発現および精製に十分確立されたプロトコールが開発されたので、続いての研究で、本発明者らは、このようなアプローチを、EBV関連悪性腫瘍に対抗するための免疫療法用の別の組換えポリエピトープタンパク質の設計に拡張した。特に、EBV関連再発性ホジキン病および鼻咽頭癌の治療のために、EBV特異的CD8+T細胞はより有効であると考えられる。しかしながら、EBV特異的CD8+T細胞の生成は、複合体作製法などのいくつかの限定要因により制限され、ほとんどの場合、このようなプロセスは抗原を抗原提示細胞に送達するために、例えば組換えアデノウイルスベクターなどの感染力のある臨床級のウイルス材料を必要とする。従って、このような問題を克服するために、本発明者らは、細菌発現系を用いて発現させることができる新規なEBVポリエピトープを設計した。9種の異なる抗原(BMLF1、LMP2a、BRLF1、LMP2、EBNA3A、BZLF1、EBNA3C、EBNA1およびEBNA3B)に由来する20個の最小CD8+T細胞エピトープをコードするEBVポリエピトープを、図13Aに概略を示すように設計し、ポリエピトープ配列内の各エピトープをプロテアソームリンカーによって分離した。これらの各ポリエピトープ配列に含まれるEBVエピトープの総覧を表3に示す。このEBVポリエピトープ構築物を大腸菌に形質転換した。タンパク質発現条件を最適化し、発現したタンパク質をNi−NTAマトリックスを用いて精製した。CMVポリエピトープタンパク質の結果に一致して、これらの実験から得られた結果は、EBVポリエピトープが細菌発現系を用いて上手く発現でき、タンパク質が均質となるまで精製可能であることを示した(図13B)。
EBV特異的CD8+T細胞を増殖させるためのEBVポリエピトープの潜在的有効性を決定するため、複数の異なるドナー由来のPBMCを精製された組換えEBVポリエピトープタンパク質で刺激した。刺激後に増殖したEBV特異的CD8+T細胞を、細胞内サイトカインアッセイを用いて評価した。図14に示されるデータは、かなりの割合のドナーが、刺激されなかったPBMCに比べ、EBVポリエピトープで刺激された後にEBV特異的CD8+T細胞の増殖を示したことを示す。興味深いことに、各ドナー由来の細胞は、いくつかのHLAクラスI対立遺伝子により拘束される複数のエピトープを認識し、ドナーの大多数で、少なくとも3つの異なるエピトープに対して増殖したCD8+T細胞の頻度が高くなった。
Claims (31)
- 2つ以上の異なるヘルペスウイルス抗原に由来する複数のCTLエピトープのそれぞれの対応アミノ酸配列を含み、かつ、前記複数のCTLエピトープのうちの少なくとも2つの間に、プロテアソーム遊離アミノ酸またはアミノ酸配列および任意選択により抗原プロセシング関連輸送体(TAP)認識モチーフを含む介在アミノ酸またはアミノ酸配列をさらに含み、外因性タンパク質として動物に投与されると細胞傷害性Tリンパ球免疫応答を惹起可能である、単離されたタンパク質。
- 前記エピトープが、HLAクラスI特異性HLA−Al、−A2、−A3、−All、−A23、−A24、−A26、−A29、−A30、−B7、−B8、−B27、−B35、−B38、−B40、−B41、−B44、−B51、−B57および−B58のうちの少なくともいずれかに拘束される、請求項1に記載の単離されたタンパク質。
- 前記ヘルペスウイルスがサイトメガロウイルス(CMV)またはエプスタイン・バーウイルス(EBV)である、請求項1に記載の単離されたタンパク質。
- 前記CMV CTLエピトープがpp50、pp65、pp150およびIE−lのうちの少なくともいずれかに由来する、請求項3に記載の単離されたタンパク質。
- 前記CTLエピトープが配列番号1〜21で示されるアミノ酸配列のうちから選択されるアミノ酸配列を有する、請求項4に記載の単離されたタンパク質。
- 配列番号1〜13で示されるCTLエピトープアミノ酸配列のそれぞれを含む、請求項5に記載の単離されたタンパク質。

(b)前記1つ以上の細胞を培養することにより、前記1つ以上の細胞由来のヘルペスウイルス特異的CTLを増殖させる工程
を含む、方法。 - 請求項28の工程(b)で産生された前記CMV特異的CTLを動物に投与することにより、前記動物のCMV感染を予防的にまたは治療的に処置する工程を含む、養子免疫療法。
