結晶形は、限定されるものではないが、例えば、適切な溶媒混合物からの結晶化または再結晶化、昇華、溶融物からの成長、別の相からの固体状態変換、超臨界流体からの結晶化、およびジェット噴霧を含む種々の方法によって調製され得る。溶媒混合物中での結晶形の結晶化または再結晶のための技術は、限定されるものではないが、例えば、溶媒の蒸発、溶媒混合物の温度の低下、化合物および/またはその塩の過飽和溶媒混合物の結晶播種、溶媒混合物の凍結乾燥、および溶媒混合物に逆溶媒(カウンター溶媒)を添加することを含む。高スループット結晶化技術は、多形を含む結晶を調製するために用いられ得る。
出発物質であるN−(3−フルオロ−4−((7−(2−ヒドロキシ−2−メチルプロポキシ)キノリン−4−イル)オキシ)フェニル)−1,5−ジメチル−3−オキソ−2−フェニル−2,3−ジヒドロ−1H−ピラゾール−4−カルボキサミドはWO2012118632A1に従って調製することができ、その内容は、参照によりその全体が本明細書に援用される。
N−(3−フルオロ−4−((7−(2−ヒドロキシ−2−メチルプロポキシ)キノリン−4−イル)オキシ)フェニル)−1,5−ジメチル−3−オキソ−2−フェニル−2,3−ジヒドロ−1H−ピラゾール−4−カルボキサミド(1.0g、1.80mmol)をメタノール(30mL)およびジクロロメタン(15mL)に溶解した。メタノール(2mL)中の4−メチルベンゼンスルホン酸水和物(360mg、1.88mmol)の溶液をゆっくり加えた。周囲温度で1時間撹拌した後、溶媒を除去して淡黄色の油を得、これを20℃でメタノール(20mL)中で結晶化させて、標題の化合物を白色の固体として得た(1.26g、96%)。
1.化合物(I)の結晶形CのX線粉末回折研究
X線粉末回折(XRPD)パターンを、自動透過反射型サンプルホルダー(3*15)を備えるX線粉末回折計(Empyrean、PANalytical、オランダ)上に収集した。X線管(Cu、kα、Kα1(Å):1.540598;Kα2(Å):1.544426;Kα2/Kα1=0.50)を45kVの電圧および40mAの電流、照射長=10.0mmに設定した。走査パラメータは、連続スキャン、3°から40°まで(2θ±0.2°)の範囲、ステップサイズ0.0168°、ステップごとの時間10秒であった。データを周囲温度(約18℃から約30℃まで)で収集した。試料(通常、1〜2mg)をガラススライド上でわずかにプレスして平らな表面を得ることによって平板標本として作製した。データはDataCollectorソフトウェアで収集し、DataViewerおよびHighScorePlusソフトウェアで分析した。化合物(I)の形CのXRPDパターンを図1に示し、XRPDパターンにおけるピークおよびその相対強度を以下の表1に示す。
全てのDSC測定をTAInstruments(登録商標)モデルQ2000示差走査熱量計上で行った。試料(2〜6mg)をアルミニウムパンに秤量し、100分の1ミリグラムまで記録し、DSC装置に移した。装置を50mL/分の窒素ガスでパージした。データを10℃/分の加熱速度で室温から300℃までにおいて収集した。データをTa UniversalAnalysisソフトウェアによって分析した。
全てのTGA解析をTGATAQ500熱重量分析計上で行った。試料(約10〜30mg)を予め秤量した白金パンに入れた。試料の重量を正確に測定し、装置によって千分の1ミリグラムまで記録した。炉を60mL/分の窒素ガスでパージした。データを10℃/分の加熱速度で室温から300℃までにおいて収集した。データをTa UniversalAnalysisソフトウェアによって分析した。
化合物(I)の結晶形Cの試料(100〜200mg)を薄膜の形状(厚さ≦5mm)で時計皿上に置いた。試料を以下の条件下にそれぞれ曝露した:10日間の高温(60±2℃)、10日間の高湿(25±2℃、90%±5%相対湿度)、照明条件(0.7W・h/m2以上の紫外光を伴う、10日間の可視光4500lx±500lx、25±2℃、60%±5%相対湿度)、および10日間の室温(30±2℃、65%±5%相対湿度)。高速液体クロマトグラフィー(HPLC)によって、試料中の不純物含有量を異なる時点(0、5および10日後)で測定し、100%として設定された最高ピーク(化合物Iに対応する)に対して吸収ピークを正規化した。HPLCの機器および条件を表2に示し、データを表3に示す。
栓を備えるガラスの秤量瓶を秤量し、重量をm1として記録した。結晶形Cまたは非晶質形の化合物(I)(約1.0g)を秤量したガラス秤量瓶の中に入れ、栓で蓋をした。その総量をm2として記録した。秤量瓶(栓を除く)を、25±1℃で塩化アンモニウムの飽和溶液を含むデシケーター(80%±2%のRH(相対湿度))に入れた。秤量瓶を栓で蓋をし、5日後および10日後に秤量し、その重量をm3として記録した。吸湿容量を以下の式に従って計算し、結果を表4に掲げる。
化合物(I)の非晶質または本明細書に開示した化合物(I)の結晶形Cの薬物動態学的特性をビーグル犬で評価した。LC/MS/MS系をアジレント1200シリーズ真空脱気装置、バイナリポンプ、ウェルプレートオートサンプラー、恒温カラムコンパートメント、エレクトロスプレーイオン化(ESI)源を備えるアジレントG6430TripleQuadrupoleMassSpectrometerからなる分析に用いた。MRMモードを用いて定量分析を行った。MRM遷移のパラメータは、表Aにある。

- 請求項1〜4の何れか1項に記載の結晶形または請求項5〜7の何れか1項に記載の医薬組成物を罹患体に投与することによって罹患体における増殖性疾患を予防し、管理し、治療し、または重症度を軽減する方法。
- 前記増殖性疾患は、結腸癌、直腸癌、胃癌、胃腺癌、膵臓癌、膀胱癌、胆嚢癌、乳癌、腎臓癌、腎細胞癌、肝臓癌、肝細胞癌、肺癌、皮膚癌、メラノーマ、甲状腺癌、骨肉腫、軟部組織の肉腫、頭頸部癌、中枢神経系の癌、神経膠腫、神経膠芽腫、卵巣癌、子宮癌、子宮内膜癌、前立腺癌、急性骨髄性白血病または急性リンパ芽球性白血病、或いはその転移癌である請求項8に記載の方法。
- 請求項1〜4の何れか1項に記載の結晶形または請求項5〜7の何れか1項に記載の医薬組成物により受容体チロシンキナーゼを阻害するかまたは調節する方法であって、前記受容体チロシンキナーゼは、VEGFR、c−Met、Axl、Merまたはその組み合わせである方法。
- 罹患体における増殖性疾患を予防し、治療し、または重症度を軽減することにおける使用のための請求項1〜4の何れか1項に記載の結晶形または請求項5〜7の何れか1項に記載の医薬組成物。
- 前記増殖性疾患は、結腸癌、直腸癌、胃癌、胃腺癌、膵臓癌、膀胱癌、胆嚢癌、乳癌、腎臓癌、腎細胞癌、肝臓癌、肝細胞癌、肺癌、皮膚癌、メラノーマ、甲状腺癌、骨肉腫、軟部組織の肉腫、頭頸部癌、中枢神経系の癌、神経膠腫、神経膠芽腫、卵巣癌、子宮癌、子宮内膜癌、前立腺癌、急性骨髄性白血病または急性リンパ芽球性白血病、或いはその転移癌である請求項11に記載の結晶形または医薬組成物。
- 受容体チロシンキナーゼを阻害しまたは調節することにおける使用のための請求項1〜4の何れか1項に記載の結晶形または請求項5〜7の何れか1項に記載の医薬組成物であって、前記受容体チロシンキナーゼは、VEGFR、Flt3、c−Met、Axl、Merまたはその組み合わせである結晶形または医薬組成物。
- 罹患体における増殖性疾患を予防し、治療し、または重症度を軽減することにおける請求項1〜4の何れか1項に記載の結晶形または請求項5〜7の何れか1項に記載の医薬組成物の使用。
- 前記増殖性疾患は、結腸癌、直腸癌、胃癌、胃腺癌、膵臓癌、膀胱癌、胆嚢癌、乳癌、腎臓癌、腎細胞癌、肝臓癌、肝細胞癌、肺癌、皮膚癌、メラノーマ、甲状腺癌、骨肉腫、軟部組織の肉腫、頭頸部癌、中枢神経系の癌、神経膠腫、神経膠芽腫、卵巣癌、子宮癌、子宮内膜癌、前立腺癌、急性骨髄性白血病または急性リンパ芽球性白血病、或いはその転移癌である請求項14に記載の使用。
- 受容体チロシンキナーゼを阻害しまたは調節することにおける請求項1〜4の何れか1項に記載の結晶形または請求項5〜7の何れか1項に記載の医薬組成物の使用であって、前記受容体チロシンキナーゼは、VEGFR、Flt3、c−Met、Axl、Merまたはその組み合わせである使用。
