Abstract
Description
本発明は、生物医薬の分野に関し、具体的に、抗TNFモノクローナル抗体および炎症の治療におけるその使用に関する。
腫瘍壊死因子α(TNF)は、様々な細胞、主にマクロファージやT細胞によって分泌される多機能サイトカインである。TNFはTNF受容体1および2 (TNFR1およびTNFR2)によって多くの生物学的機能を発揮し、ここで、TNFR2は免疫細胞だけでTNFR1の作用(1-3)を果たす。
多くのTNF機能は、主に、1) 転写因子の核内因子カッパ B(NF-κB)を刺激することによって、細胞の活性化およびサイトカインの生成につながること、2) 細胞アポトーシスの外部経路の誘導、および3) 壊死の誘導といった3つの細胞内イベントである。これらの活性の細胞内シグナル伝達経路は一部の成分を共有するが、異なる結果につながる。NF-κBの活性化によって、炎症促進因子の分泌および細胞の生存と活性化につながる。アポトーシスは細胞死の状態で、カスパーゼ-3の活性化、核分断、早期で完全な細胞膜を含有する、炎症性反応が少ないかないを特徴とする。しかしながら、壊死は細胞死のもう一つの機序で、ここで、カスパーゼ-3の活性化がなく、細胞膜の完全性が損なわれる。壊死は、強烈な免疫と炎症性反応を刺激する細胞内物質の放出につながる(4、5)。アポトーシスの条件において、カスパーゼの抑制はプログラムされた細胞死の形態につながることがあり、発生するとき壊死の特徴を有し、「プログラムされた壊死」と呼ばれる(6-11)。このようなプログラムされた壊死は炎症性の過程であるため、臨床でたとえば関節リウマチ、クローン病や乾癬などの疾患に関連する。しかしながら、今まで、これらの疾患におけるプログラムされた壊死を発生させる因子はまだ同定されていない。
敗血症患者において、TNFが顕著に向上し、かつ病状の重篤度に正相関する。しかしながら、従来のTNF遮断型抗体および遊離の受容体はいずれも有効に敗血症を治療することができず、ほかの要素または因子が発病の過程でTNFと協同して重要な作用を果たすことが示唆された。
そのため、本分野で炎症性疾患が治療できるように細胞の壊死の調節、ひいては逆転の物質的手段が切望されている。
本発明の目的は、炎症性疾患が治療できるように細胞壊死の調節、ひいては逆転の物質的手段を提供することにある。
第一の側面では、本発明は、
配列番号1で示されるCDR1、
配列番号3で示されるCDR2、および
配列番号5で示されるCDR3
からなる群から選ばれる相補性決定領域CDRを有する抗体の軽鎖可変領域を提供する。
好適な実施形態において、前記の軽鎖可変領域は配列番号7で示されるアミノ配列を有する。
好適な実施形態において、前記軽鎖の定常領域は配列番号9で示される。
好適な実施形態において、前記軽鎖のアミノ酸配列は配列番号10で示される。
好適な実施形態において、前記軽鎖をコードするポリヌクレオチド配列は配列番号11で示される。
第三の側面では、本発明は、
配列番号12で示されるCDR1、
配列番号14で示されるCDR2、および
配列番号16で示されるCDR3
といった3つの相補性決定領域CDRを含む抗体の重鎖可変領域を提供する。
好適な実施形態において、前記重鎖可変領域は配列番号18で示されるアミノ配列を有する。
好適な実施形態において、前記の重鎖定常領域は配列番号20で示される。
好適な実施形態において、前記重鎖のアミノ酸配列は配列番号21で示される。
好適な実施形態において、前記重鎖をコードするポリヌクレオチド配列は配列番号22で示される。
第五の側面では、本発明は、
(1) 本発明の第一の側面に記載の軽鎖可変領域、および/または
(2) 本発明の第三の側面に記載の重鎖可変領域、
を有する抗体を提供する。
好適な実施形態において、前記抗体は、本発明の第二の側面に記載の軽鎖、および/または本発明の第四の側面に記載の重鎖を有する。
(i) 本発明の第一の側面に記載の軽鎖可変領域の配列、本発明の第二の側面に記載の軽鎖の配列、本発明の第三の側面に記載の重鎖可変領域の配列、本発明の第四の側面に記載の重鎖の配列、あるいは本発明の第五の側面に記載の抗体の配列と、
(ii) 任意の発現および/または精製を補助するタグ配列と、

好適な実施様態において、前記のタグ配列は6Hisタグを含む。
第七の側面では、本発明は、配列QLVVPSEと特異的に結合する抗体を提供する。
第八の側面では、本発明は、

第十の側面では、本発明は、遺伝子工学化された宿主細胞であって、本発明の第九に記載のベクターを含むか、あるいはゲノムに本発明の第八に記載のポリヌクレオチドが組み込まれた宿主細胞を提供する。

好適な実施形態において、前記細胞壊死促進抗体阻害剤は突然変異型TNFで、野生型TNFと比べ、前記突然変異型TNFは本発明の第五または第七の側面に記載の抗体と結合することができるが、TNF受容体と結合しない。
好適な実施形態において、前記突然変異型TNFは配列番号23で示される。
第十三の側面では、本発明は、炎症性疾患を治療する薬物の製造における本発明の第十一の側面に記載の細胞壊死促進抗体阻害剤の使用を提供する。
好適な実施形態において、前記炎症性疾患は関節リウマチ、クローン病、乾癬、敗血症を含む。
好適な実施形態において、前記使用は患者を分類することである。
第十五の側面では、本発明は、炎症性疾患を診断する検出キットまたは炎症性疾患患者を分類する検出キットであって、
a.標準品としてのQLVVPSEで示される断片あるいは本発明の第五または第七の側面に記載の抗体と、
b.QLVVPSEで示される断片あるいは本発明の第五または第七の側面に記載の抗体を標準品として患者からのサンプルにQLVVPSEと特異的に結合する抗TNF自己抗体あるいは本発明の第五または第七の側面に記載の抗体と競合する抗TNF自己抗体が存在するか検出することに関する使用説明書と、
が入れてあるキットを提供する。

もちろん、本発明の範囲内において、本発明の上記の各技術特徴および下記(たとえば実施例)の具体的に記述された各技術特徴は互いに組合せ、新しい、または好適な技術方案を構成できることが理解される。紙数に限りがあるため、ここで逐一説明しない。
発明者は、幅広く深く研究したところ、意外に、抗体がプログラムされた壊死を引き起こすことができること、具体的に、抗体244-12がTNFの存在下でTNFに誘導されたアポトーシスをプログラムされた壊死に転換させることを見出した。そして、一部の炎症の患者、たとえば関節炎、敗血症の患者の体内に類似のプログラムされた壊死を引き起こす自己抗体が存在し、かつこれらの患者の病状は顕著に重篤になっていることを見出した。そのため、TNFR1と結合しない突然変異型TNFで患者の自己抗体を遮断することによって炎症を治療することができるが、重篤な副作用が生じない。これに基づき、本発明を完成させた。
本明細書に記載の壊死または細胞壊死とは細胞が外力によって破壊して死亡することをいい、その特徴は細胞が破砕し、細胞膜が破壊して不完全になることにある。壊死時細胞が多くの炎症性物質、たとえば核酸、尿酸、HMGB1などを放出するため、炎症反応を引き起こす。
本明細書に記載のプログラムされた壊死または細胞のプログラムされた壊死とは細胞のシグナル伝達による死亡をいう。その特徴は細胞が破砕し、細胞膜が破壊して不完全になることにある。上記細胞壊死と同様に、プログラムされた壊死の過程において、壊死細胞が多くの炎症性物質、たとえば核酸、尿酸、HMGB1などを放出するため、炎症反応を引き起こす。
TNFは主にマクロファージやT細胞によって分泌される多機能サイトカインである。TNFはTNF受容体1と2(TNFR1とTNFR2)によって多くの生物学的機能を発揮し、転写因子の核内因子カッパ B(NF-κB)の刺激、細胞アポトーシスの外部経路の誘導、および壊死の誘導を含む。
多くの場合、TNFによって刺激された細胞は主にNF-κBを活性化することによって細胞を生存させる。アポトーシスおよび壊死はNF-κB経路が抑制される場合だけ触発される(24)。TNFが膜結合複合体Iを刺激し(25)、NF-κBの活性化を開始させるが、アポトーシス/壊死を開始させないことが提出された。しかしながら、NF-κBの活性化が阻止されると、TNFがその標的細胞を刺激して細胞質で第二の複合体(複合体II)を形成させ、それがシグナル経路を細胞死に導く。
現在のすべての研究では、TNF-TNFR1結合後の下流の結果が強調されている。しかしながら、異なる細胞機能が観察されうるTNF-TNFR1の相互作用のレベルで微妙な分子上の基礎が研究されていない。通常の概念は、一般的に、TNF-TNFR1結合は、NF-κBの刺激および細胞死の誘導を含むすべてのTNF機能を開始させるには十分である。
本明細書において、「本発明の抗体」、「本発明のモノクローナル抗体」、「244-12」は同じ意味を有し、TNF、特に特異的に配列QLVVPSEと結合することができることをいう。
本発明のモノクローナル抗体はプログラムされた壊死を引き起こすモノクローナル抗体である。当該モノクローナル抗体はTNFと結合することができるだけでなく、形成される抗原抗体複合体はさらにTNFの細胞膜表面の受容体と結合して細胞のプログラムされた壊死を促進することもできる。このような壊死はネクロスタチン−1によって抑制される。
また、本発明のモノクローナル抗体は特異的にQLVVPSEと結合する抗TNF抗体、または244-12と競合することができる抗体を含み、これらの抗体はTNFの存在下で、たとえば増加の場合、炎症反応を重篤にさせる。
本明細書で用いられるように、用語「重鎖可変領域」と「VH」は入れ替えて使用することができる。
本明細書で用いられるように、用語「可変領域」と「相補性決定領域(complementarity determining region、CDR)」は入れ替えて使用することができる。
本明細書で用いられるように、用語「軽鎖可変領域」と「VL」は入れ替えて使用することができる。
具体的な実施形態において、本発明の抗体の軽鎖可変領域は、コードヌクレオチド配列が配列番号2で示される、配列番号1で示されるCDR1、コードヌクレオチド配列が配列番号4で示される、配列番号3で示されるCDR2、およびコードヌクレオチド配列が配列番号6で示される、配列番号5で示されるCDR3からなる群から選ばれる相補性決定領域CDRを有する。
一つの好適な例において、前記の軽鎖可変領域のアミノ酸配列は配列番号7で示され、そのコードヌクレオチド配列は配列番号8で示される。
本発明の好適な実施形態において、前記抗体の軽鎖は上記軽鎖可変領域および軽鎖定常領域を含む。
一つの好適な例において、前記軽鎖の定常領域は配列番号9で示される。

(ii) 任意の発現および/または精製を補助するタグ配列と、
を有する組み換えタンパク質およびそのコードポリヌクレオチドを提供する。
一つの好適な例において、前記のタグ配列は6Hisタグを含む。
もう一つの好適な例において、前記のベクターは、細菌プラスミド、ファージ、酵母プラスミド、植物細胞ウイルス、哺乳動物細胞ウイルス、たとえばアデノウイルス、レトロウイルス、またはほかのベクターを含む。
更なる実施形態において、本発明は、遺伝子工学化された宿主細胞であって、上記ベクターを含むか、あるいはゲノムに前記組み換えタンパク質のコードポリヌクレオチドが組み込まれた宿主細胞を提供する。
そのため、もう一つの側面では、本発明はプログラムされた壊死を引き起こす抗TNF自己抗体を検出する方法を提供する。当該方法は患者の体内に特異的にQLVVPSEと結合する抗TNFの自己抗体が存在するか検出すること、あるいは患者の体内に本発明のモノクローナル抗体244-12と競合する抗TNFの自己抗体が存在するか検出することを含む。特異的にQLVVPSEと結合する抗TNFの自己抗体が存在するか、あるいは検出される自己抗体が244-12と競合することができると、これらの自己抗体はTNFの存在下で、たとえば増加の場合、炎症反応を重篤にさせる可能性がある。患者の体内に特異的にQLVVPSEと結合する抗TNFの自己抗体が存在するか、あるいは本発明のモノクローナル抗体244-12と競合する抗TNFの自己抗体が存在するか検出した後、患者をたとえば予後の良い患者または予後の良くない患者に分類してもよい。
さらに、本発明は、炎症性疾患を診断する検出キットまたは炎症性疾患患者を分類する検出キットであって、
a.標準品としてのQLVVPSEで示される断片あるいは本発明の抗体と、
b.QLVVPSEで示される断片あるいは本発明を標準品として患者からのサンプルにQLVVPSEと特異的に結合する抗TNF自己抗体あるいは本発明の抗体と競合する抗TNF自己抗体が存在するか検出することに関する使用説明書と、
が入れてあるキットを提供する。
以上の発見に基づき、本発明の抗体の拮抗剤、阻害剤または中和物質は炎症性疾患、たとえば関節リウマチ、クローン病や乾癬を治療、軽減、緩和することができる。たとえば、具体的な実施形態において、244-12またはその類似抗体、たとえば炎症性疾患患者の体内の類似抗体と結合することができるが、TNF受容体の突然変異型TNFと結合しないか、あるいは本発明の抗体244-12またはその類似抗体の特異的な抗体は244-12による壊死を逆転させることができる。
一つの好適な例において、前記細胞壊死促進抗体阻害剤は突然変異型TNFで、野生型TNFと比べ、前記突然変異型TNFは細胞壊死を促進する抗体と結合することができるが、TNF受容体と結合しない。
一つの好適な例において、前記突然変異型TNFのアミノ酸配列は配列番号23で示される(突然変異:S162F、Y163H)。
これに基づき、本発明は、
(i) 以上に記載の軽鎖可変領域、以上に記載の軽鎖、以上に記載の重鎖可変領域、以上に記載の重鎖、以上に記載の抗体、以上に記載の組み換えタンパク質と、
(ii) 任意の薬学的に許容される担体と、
を含む薬物組成物も提供する。
1.本発明者は、初めて、抗体が細胞のプログラムされた壊死を引き起こすことができることを見出した。
2.本発明者の成果は顕著な臨床意義を有する。
3.本発明の抗体はそれと抗原結合部位を競争する抗体を検出することによって、これらの自己抗体を除去することで、炎症反応を治療、緩和または軽減することができる
4.本発明の抗体はTNFと細胞のプログラムされた壊死を引き起こすことができ、そしてこれらの壊死は炎症を引き起こして免疫細胞による特異的な免疫反応をもたらすため、腫瘍またはウイルス・細菌感染の治療に使用することができる。
患者
患者は、Nuffield Orthopaedic Centre Oxfordのリウマチ診療所によって募集され、すべてのサンプルは分析まで-80℃で保存された。治療性関節穿刺術の一部として、滑液を炎症性疾患患者の膝関節から吸い取った。1987米国リウマチ学院または2010 ACR/EULAR分類基準によって関節リウマチを定義する。ほかの炎症性関節炎は臨床およびX線撮影の基準によって診断した。すべてのリウマチ患者はリウマチ因子の血清反応で陽性を示し、中等の疾患活性を有する。ドナーのインフォームドコンセントの上、完全に国家および制度の倫理的要求(COREC number COREC06/Q1606/139)に従って得られたサンプルおよび/またはデータを収集した。
マウス線維肉腫L929細胞およびヒトリンパ球Jurkat A3細胞は米国タイプカルチャーコレクション(ATCC, Manassas, VA)から購入された。ヒトC28I2軟骨細胞はMary B. Goldring博士によって提供された。ヒトSaOs-2骨芽細胞はSigmaから購入された。TNFはImmunotools(ドイツ)から購入された。
アポトーシスおよび壊死の試験
20 μM z-VAD-FMK (R & D Systems、ミネアポリス、ミネソタ州)または20 μM ネクロスタチン-1(PeproTech、Rocky Hill、ニュージャージー州)を含有するか含有しないTNF(Immunotools、Friesoythe、ドイツ)またはTNFペプチドの存在下において、細胞を一晩インキュベートした(あるいは図で示される時間でインキュベートした)。一部の実験において、TNFは細胞および示される濃度のモノクローナル抗体または滑液と共培養した。メーカーのパンフレットの指導に従い、生存/死亡細胞染色キット(Invitrogen、Paisley、イギリス)によって細胞を染色し、cytofix/cytoperm固定/透過化溶液キット(BD Pharmingen、オックスフォード、イギリス)によって固定化した。その後、FITC-カップリングの抗-カスパーゼ-3抗体(Cell Signaling Technology, Danvers、マサチューセッツ州、米国)によって細胞内染色を行った。CyAnフローサイトメーター(Beckman Coulter、Fullerton、カルフォルニア州)によって細胞を得、Flowjo(Tree Star Inc. Ashland、オレゴン州)によってデータを分析した。
アクチノマイシンD(2-10μg/ml) (Sigma)の存在下か存在しない条件で、壁付着L929細胞(100 μl、4x105/ml)をTNFと37℃、5%CO2で一晩インキュベートした。すべての上清液を吸い取った後、50 μlの0.05%クリスタルバイオレットを各ウェルに入れて生存細胞を染色した。クリスタルバイオレットを洗い流した後、壁付着細胞の活力を測定した。
NF-κB試験
示された時間でTNFまたはそのペプチドによって処理されたJurkat A3細胞(図1Bおよび2A)を収集し、プロテアーゼ混合物阻害剤(Sigma)を含有するRIPA分解緩衝液(Cell Signaling Technology、Danvers、マサチューセッツ州、米国)で分解させた。SDS-PAGEによって細胞抽出物を分析した後、ニトロセルロース膜(Amersham Life Science、イギリス)にブロットし、さらに化学発光法によって、IκB抗体(Cell Signaling Technology、Danvers、マサチューセッツ州、米国)で検出した。
一部の実験において、細胞をペプチドとインキュベートした後、細胞を加工して透過電子顕微鏡法による検出に供した(Oxford Brooks Universityによってサービスを提供された)。すなわち、PBSで調製された2.5%グルタルアルデヒドによって細胞を固定化し、1%四酸化オスミウムでポスト固定化した。エタノール勾配で脱水した後、標本をTAAB 'Premix'エポキシ樹脂に包埋した(中等硬度)。RMC PT-PC超薄切片機によって、ダイヤモンドブレードで厚さ約60 nmの切片にカットし、200メッシュの未被覆(クリーンな)銅ネットで収集し、酢酸ウラニュウムおよびクエン酸鉛で染色し、かつHitachi H-7650 TEMによって120 Kvで検出した。
核DNA分断
あるいは、生存/死亡細胞染色キットおよび抗-カスパーゼ-3抗体(BD)以外、DAPI (Vector Laboratories、Burlingame、カルフォルニア州、米国)によって一部の細胞を染色した。その後、蛍光顕微鏡でこれらを観察した。
THP-1-XブルーNF-κB試験
THP-1-XブルーNF-κB表示細胞はinvivoGen(San Diego、CA92121)から得られた。細胞を示された濃度のTNF-αペプチドをインキュベートした。LPSおよび可溶性TNF-αは陽性対照である。細胞を収集して上清液を培養し、メーカーの使用説明書に従ってNF-κBを検出した。
モノクローナル抗体の選別/抑制試験
4℃で、2 μg/ml TNFでELISAプレートを一晩、あるいは37℃で2時間被覆させた後、37℃でモノクローナル抗体と1時間インキュベートした。HRPとカップリングされた二次抗体を反応の検出に使用した。抑制試験に関し、TNFR1または滑液をモノクローナル抗体と共に入れた。
TNFとTNFR1の結合
以下のようにELISAによってTNFR1とTNFの結合を測定した。4℃で、1.5 μg/ml TNFまたはmTNF-HAでELISAプレートを一晩、あるいは37℃で2時間被覆させた。37℃でTNFR1(1 μg/ml)と2時間インキュベートした後、抗TNFR1または抗HA抗体とさらに2時間インキュベートし、HRPとカップリングされた二次抗体-マウスIgG抗体を入れ、30分間で、さらにその基質を入れて検出に供した。呈色させ、Wallace Victor2 1420マルチラベルカウンター(PerkinElmer、Massachusettsマサチューセッツ州、米国)によってOD 450 nmで検出した。
室温において、20 ng/mlのTNFを2 μg/mlのmAb M26または244-12と1時間インキュベートした。TNF/M26またはTNF/244-12の混合物を氷上においてカバーガラスで生長したL929細胞と15分間インキュベートした。4%パラアルデヒド(PBSで調製)で細胞を10分間固定化し、0.5% BSAおよび0.1%冷水魚ゼラチンを含有するリン酸塩緩衝液で15分間ブロッキングした後、さらにウサギ抗TNFR1抗体(Abcam)と1時間インキュベートした。PBSで細胞を洗浄した後、Alexa Fluor 488またはAlexa Fluor 568(Invitrogen)にカップリングされた関連二次抗体とインキュベートした。その後、Gelvatol/DABCO(Sigma-Aldrich)でPBS-洗浄のサンプルをセットした。DAPI(Sigma-Aldrich)でDNAを対比染色した。Nikon Eclipse 80iを使用し、60xで蛍光顕微法によってすべてのサンプルを分析した。Hamamatsuカメラを使用し、NIS-Elements AR3.0ソフトで画像を得た。
実施例1. モノクローナル抗体244-12の製造およびその結合エピトープの同定
発明者は通常の方法によって33株のモノクローナル抗体を製造し、かつ製造されたモノクローナル抗体とTNFの結合状態を検出したが、中では、ELISA試験によってモノクローナル抗体244-12とTNFの結合が最も強かったことが証明された。
その後、発明者はモノクローナル抗体244-12の結合エピトープを同定したところ、QLVVPSEであった。
発明者はモノクローナル抗体(mAb)244-12のTNFの機能に対する影響を研究した。
まず、図2における共焦点顕微鏡の写真でモノクローナル抗体244-12とTNFの結合がTNFとTNF受容体の結合に影響しないことが示された。共焦点顕微鏡において、TNF受容体は赤色に、244-12は緑色に染色された。ここで、上の図ではTNFが存在する場合、緑色と赤色が重なった(黄色)ことが示され、244-12がTNFおよびTNF受容体と重なったことが説明された。下の図ではTNFが存在しない場合、赤色だけであったことが示され、244-12が直接細胞の表面に結合しないことが説明された。
その後、発明者はTNFがL929細胞を刺激して細胞アポトーシス(活性カスパーゼ-3の発現)を引き起こしたが、モノクローナル抗体244-12を入れた場合、細胞アポトーシスが抑制された(活性カスパーゼ-3が抑制された)ことを観察した。図3ではモノクローナル抗体244-12がL929細胞(マウス線維芽細胞腫瘍細胞、上の図)とC28I2細胞(軟骨細胞、下の図)の二つの細胞株のいずれにおいてもTNFによる細胞アポトーシス(すなわち、活性カスパーゼ-3の発現)を遮断したことが示された。
発明者はさらに研究したところ、244-12 + TNFによる壊死はシグナル伝達によるものであることを証明した。図5は本発明の抗体244-12+TNFによる壊死がNec-1によって抑制されることを示す。ここで、244-12 + TNFはNec-1を入れた後、細胞壊死はまた細胞アポトーシスに転換した(上の図の右)。細胞壊死はシグナル伝達によるもの、すなわちプログラムされた壊死であることが説明された。
発明者は、意外に、一部の未治療の関節リウマチまたは骨関節疾患の患者の関節液に大量の抗体が存在することを見出した(図7A)。これらの抗体は244-12と抗原結合部位を競合することができる(図7B)。これらの抗体を含有する関節液は細胞のプログラムされた壊死を引き起こすことができる(図7C)。
ここで、図7Aでは、一部の関節炎患者の関節液に抗TNFの自己抗体が存在したことが示された。患者SF8の関節液に大量の自己抗体が含まれていた。7Bでは、自己抗体は244-12とTNF結合部位を競合したことが示された。244-12とTNFの結合試験(ELISA)において、SF8関節液を入れると、244-12とTNFの結合が抑制された。7Cでは、SF8関節液がTNFの存在下でプログラムされた壊死を引き起こしたことが示された。TNFはL929細胞のアポトーシスを引き起こした(上の図の右)が、SF8関節液を入れると、細胞はプログラムされた壊死に転換した(中の図の右)。一方、対照関節液SF4は壊死を引き起こさなかった(下の図の右)。
発明者はさらに24名の敗血症患者を観察したところ、14名の患者で244-12と抗原結合部位を競合する抗体が検出されたが、ほかの10名の患者で類似する抗体が含まれていなかった。14名の244-12と抗原結合部位を競合する抗体を含む患者は全力を尽くして看護と治療を行われたが、全員死亡した。一方、ほかの10名の患者のうち、5名が死亡し、ほかの5名は2〜3週間後完治して退院した(結果は表1を参照する)。死亡患者では抗体レベルは非常に顕著に完治群よりも高かった(p<0.01)(図8)。
1.TNFa突然変異体S86F/Y87Hのクローン
アミノ酸配列に合わせて突然変異体のコードDNA配列(配列番号24)を設計し、クローニングのため、TNF突然変異体のDNA5’末端にTACTTCCAATCCATGを、3’末端にTATCCACCTTTACTGTTA配列を付加した。以上のDNA分子はGeneArt社によって人工合成された後、T4 DNAポリメラーゼおよびdCTPで30分間処理した。pNIC28Bsa4ベクターはオクスフォード大学構造生物学実験室によって寄贈され、ベクターにおけるクローニングと発現に関連する要素を図9に示す。pNIC28Bsa4はBsaIで1時間酵素切断された後1%アガロースゲル電気泳動によって線状化したベクターを単離し、かつT4 DNAポリメラーゼおよびdCTPで30分間処理した。2種類のT4 DNAポリメラーゼ処理産物を混合した後、感受性DN5αに形質転換させ、プレートを作って単一クローンを選んで培養した。菌液はPCRによって陽性集落を同定した。
陽性集落のプラスミドを抽出して大腸菌BL21(DE3)を形質転換し、単一集落を取ってLB培養液に接種して37℃で一晩培養した後、LB培養液で1:100の比率で培養物を一晩希釈してOD600 = 1.0になるまで37℃で振とう培養した後、18℃に下げて0.2 mMのIPTGを入れてタンパク質の誘導発現を行い、続いて18℃で16時間振とう培養した後、4000rpmで遠心、菌体を収集し、かつTis-HCL、250mM NaCl、5 mMイミダゾール、pH 8.0で再懸濁させた(湿菌1 gあたりに5 mLの緩衝液)。
3.TNFa突然変異体S86F/Y87Hの精製
超音波法で細胞を分解させ、15000 rpmで遠心して目標タンパク質を含む上清を収集し、Ni-NTAカラム(1 Lあたりの細菌培養液で得られたタンパク質を1 mLのNi-NTAカラムで精製した)を通させ、ヒスチジン標識を含有する一本鎖オーバーラップペプチドがカラムに結合し、十分な洗浄で(50倍のNi-NTAカラムの体積に相当する緩衝液で、洗浄液= Tis-HCL、250 mM NaCl、pH 8.0、15 mMイミダゾール、pH 8.0)不純物のタンパク質を除去した後、TNFa突然変異体S86F/Y87Hを5倍的Ni-NTAカラムの体積の溶離液(Tis-HCL、100 mM NaCl、300 mMイミダゾール、pH 8.0のイミダゾール含有溶離緩衝液)で溶離させた。
4.Ni-NTAによって精製されたTNFa突然変異体S86F/Y87Hポスト緩衝液をPBSに置換した。
PBSでPD10カラムを平衡化し、2.5 mLのNi-NTAによって精製されたタンパク質を入れ、タンパク質溶液が全部PD10カラムに入った後、3.5 mLのPBSを入れ、溶離してPBSにおけるTNFa突然変異体を得た。
発明者は、突然変異型TNF(TNF-mu)が244-12および関節液における抗体と結合することができること(図6Aで示される)、突然変異型TNF(TNF-mu)がプログラムされた壊死を逆転させることができること(図6Bで示される)を見出した。
本発明者はさらに特異的にほかのTNF分子および断片と結合する抗体を研究したところ、これらの抗体がTNF関連アポトーシスをプログラムされた壊死に逆転させる効果を有することが確認されなかった。
また、本発明者は全長TNFを選別したところ、QLVVPSEと特異的に結合する抗体がTNF関連アポトーシスをプログラムされた壊死に逆転させる効果を有することを見出した。
発明者の発見に基づき、炎症の新たな治療策略を提出した。プログラムされた壊死の機序は自己抗体およびTNFに関する。プログラムされた壊死は炎症の原因の一つである。現在、炎症性疾患の治療策略の一つはTNFを抑制することによってTNFを遮断することである。当該策略は炎症の抑制に有効であるが、命を脅かす副作用、たとえばTBまたはリンパ腫につながる。本発明者は突然変異型TNF(TNFR1と結合しない)によって自己抗体を遮断する代替方法を提出した。当該策略の結果は炎症性負荷を低下させると同時にプログラムされた液を抑制するが、TNFを元のままに維持することによって、腫瘍および重篤な感染、たとえばTBを阻止する可能性がある。

- (i) 請求項1に記載の軽鎖可変領域の配列、請求項2に記載の軽鎖の配列、請求項3に記載の重鎖可変領域の配列、請求項4に記載の重鎖の配列、または請求項5もしくは7に記載の抗体の配列、あるいは
(2) 請求項6に記載の組み換えタンパク質、
からなる群から選ばれるポリペプチドをコードするポリヌクレオチド。 - 請求項8に記載のポリヌクレオチドを含むベクター。
- 遺伝子工学化された宿主細胞であって、請求項9に記載のベクターを含むか、あるいはゲノムに請求項8に記載のポリヌクレオチドが組み込まれている、前記宿主細胞。
- (i) 請求項1に記載の軽鎖可変領域、請求項2に記載の軽鎖、請求項3に記載の重鎖可変領域、請求項4に記載の重鎖、請求項5または7に記載の抗体、または請求項6に記載の組み換えタンパク質と、
(ii) 任意の薬学的に許容される担体と、
を含む薬物組成物。 - 細胞壊死促進抗体阻害剤であって、請求項5または7に記載の抗体と結合することができることを特徴とする前記阻害剤。
- 炎症性疾患を治療する薬物の製造における請求項11に記載の細胞壊死促進抗体阻害剤の使用。
- 炎症性疾患を診断する検出試薬または炎症性疾患患者を分類する検出試薬の製造におけるQLVVPSEで示される断片あるいは請求項5または7に記載の抗体の使用。
- 炎症性疾患を診断する検出キットまたは炎症性疾患患者を分類する検出キットであって、
a.標準品としてのQLVVPSEで示される断片あるいは請求項5または7に記載の抗体と、
b.QLVVPSEで示される断片あるいは請求項5または7に記載の抗体を標準品として患者からのサンプルにQLVVPSEと特異的に結合する抗TNF自己抗体あるいは請求項5または7に記載の抗体と競合する抗TNF自己抗体が存在するか検出することに関する使用説明書と、
が入れてある前記キット。
