ポリクローナル抗体の合成
抗体は、Genosphere Biotechnologies(Paris,France)によって合成した。2羽のニュージーランドウサギを使用し、免疫原としてKLH−ペプチドを用い、70日間にわたって4回免疫化した(「Pep4」:T14−ハプテンCAEFHRWSSYMVHWK−配列番号7)。動物から4回採血し、血液をプールした。次いで抗血清を、共有結合したペプチド支持体を有する重力カラムを通過させ、洗浄した後、抗体を酸性緩衝液中に溶出し、溶液を中和した。PBS緩衝液に対するさらなる透析及び凍結乾燥でプロセスを完了した。
抗体に関する製造元の報告を使用して、最適条件でELISAの実験を行った。この報告において、製造元は、抗体の濃度に関する最適密度(下記の表を参照されたい)、及びこの手順に使用されるELISAプロトコル(下記のプロトコルを参照されたい)を特定している。
抗原を、EIAストリップ上に1ウェル当たり10μgでコーティングした。ウェルを、200μL PBS緩衝液で洗浄した。
本明細書に記載されるすべての実験に対して、選択される抗体希釈は、1:1000であった。
ELISAアッセイの手順は以下のとおりであった。
AChE活性は、AChE活性の結果としてのチオール基の存在を測定するエルマン試薬を使用して測定した。G4実験の場合、AChE(G4)を単独で、またはNBP−14と組み合わせて、またはガランタミンと組み合わせてアッセイした。細胞生存性アッセイに関する実験の前日に細胞を播種した。細胞を、異なる濃度のNBP−14(0.1〜100μM)及びT30、T14、ならびにAβ10μM単独で、またはNBP−14(0.1及び0.7μM)と組み合わせて処理した。処理後、各処理の上澄(灌流液)を収集し、各条件の25μLを、新たな平底96ウェルプレートに付加し、続いて175μLのエルマン試薬を付加した(溶液A:KH2PO4 139mM、及びK2HPO4 79.66mM、pH7.0;溶液B(基質):ヨウ化アセチルチオコリン 11.5mM;溶液C(試薬):5,5’−ジチオビス(2−ニトロ安息香酸)8mM、及びNaHCO3 15mM)。エルマン試薬は、3つの溶液の混合物として、33(A):3(B):4(C)の比率で調製した。吸光度測定値は、405nmにおいて、実験を通して規則的な間隔(3、10、30、及び60分)で記録した。
試料は、以下のように調製した。脳試料を計量した後、ダウンス内に配置し、脳物質1mg当たり1.5μLのPBSを付加した。ダウンスの底部にある脳試料を、「ルースな」プランジャーを使用して少なくとも10回、ダウンスの底部に至るまでプランジすることによって均質化した。「タイトな」プランジャーを使用して材料をさらに均質化し、最低10回の完全プランジを確実にした。均質化した試料を、2mLのエッペンドルフに収集し、4℃で冷蔵した遠心分離器内で、13,000gで15分間遠心分離した。一旦遠心分離が終了すると、懸濁液を、調製した0.5mLの30KDa MWCOフィルターに収集した。これらの試料を、13,000gで30分間遠心分離し、プロテアーゼ阻害剤カクテル(RocheコンプリートPIC04693116001)を濾過物に付加した。これを、T14ペプチドに関するELISAに使用した。未濾過の試料残余分(>30kDa)を、フィルターを反転させることによって、別個の微量遠心分離管に収集し、これを使用し、ピアースアッセイを使用して、初期試料のタンパク質濃度を決定した(下記のとおり)。
Thermo Scientific Pierce 660nmタンパク質アッセイは、ウシ血清アルブミンのタンパク質標準物と比較して、(A660nm)総タンパク質濃度を速やかに測定するための既製の洗剤及び還元剤適合性アッセイ試薬である。このアッセイの場合、PBS中に1:10で希釈した10μLの各ヒト脳ホモジネート試料を、マイクロタイター96ウェルプレートに付加し、続いて150μLのピアースアッセイを付加した。5分のインキュベーション後、吸光度を、Vmaxプレートリーダー(Molecular devices,Wokingham,UK)内で、660nmで測定し、光学密度の結果を、BSAの標準曲線に挿入して、1μL当たりのミリグラムを得た。
すべての実験に関して、一元配置分散分析(ANOVA)及びGraphPAD Instat(GraphPAD software,San Diego,CA)を使用するターキー事後検定によって、複数治療群と同じ対照との間の比較を行った。これらの試験は、あらゆる処置手段を、あらゆる他の処置手段と比較し、すなわち、すべての対合比較の組に同時に適用し、2つの手段の差が、許容することが予想される標準誤差より大きい場合を特定する。統計的有意差は、P値<0.05において記録した。グラフを、Microsoft Excelを使用してプロットした。
ラット:Charles Riverからの35日齢の雄ウィスターラットからの新鮮な均質化した全脳。
脳試料の調製
10名の死後AD患者及び同年齢対照からの皮質外皮、青斑核、海馬、及びCSF部分は、寛大にもProf.Margaret Esiri及びDr Gabriele DeLucaによってJohn Radcliffe Hospital Brain Bank,Oxfordから寄付された。各脳試料を計量し、デュースに配置した。1μgの脳物質当たり、ミニEDTA遊離プロテアーゼ阻害剤カクテル(RocheコンプリートPIC04693116001)を含有する2μLのPBS(1倍)を、デュースに付加し、最初に「ルースな」プランジャーを使用し、続いて「タイトな」プランジャーを使用して均質化を行った。続いて、試料を13,000gで30分間、4℃で回転させ、上澄を採取した。CSF試料は調製を必要としなかった。全試料を−80℃で貯蔵した。
上記試料からのタンパク質濃度を、Pierce(商標)660nmタンパク質アッセイ(Thermo Scientific)を使用して測定した。手短に言えば、連続希釈(0〜2mg/mL)を、ウシ血清アルブミン(BSA)の10mg/mLストックから作製した。10μLのタンパク質を透明な96ウェルプレート(Greiner)に移すことによって、各BSA濃度の3つの複製を調製した。次いで、試料を、3つの濃度(1:1、1:2、1:10)で希釈し、各濃度の3つの複製を、10μLの試料を含有する各複製と共に、同じ96ウェルプレートに配置した。続いて、150μLのピアース試薬を標準物質及び全試料に付加し、混合物を放置して5分間、優しく振りながらインキュベートした。最後に、プレートを、分光光度計(Molecular Devices)上で、660nmで読み取った。これらの試料のタンパク質濃度は、BSA標準曲線からの傾斜及びy切片を使用して決定され、両者はMicrosoft Excelを介して計算される。
ポリアクリルアミドゲル(ミニPROTEAN(登録商標)TGX stain free(商標)ゲル、BIO−RAD)を、電気泳動タンク(BIO−RAD、ミニPROTEANテトラシステム)内に入れ、走行緩衝液(25mM TRISベース、pH8.6、192mMグリシン、0.1% SDS)を、ゲル及びタンク貯蔵器(BioRad)に付加した。タンパク質試料を、蒸留水及び4×Laemmli試料緩衝液(最終濃度:69.5mM TRIS−HCl pH6.8、1.1% LDS、11.1%(w/v)グリセロール、0.005%ブロモフェノールブルー、BIO−RAD)、及び2.5%メルカプトエタノール(BIO−RAD)を混合することによって調製した。また、試料相当濃度の外因性T14も調製し、これが、外因性T14ペプチドを測定するための正の対照として作用した。これらの混合物を、95℃で5分間加熱した後、氷上で冷却した。試料及び正の対照をゲル中に負荷し、分子量マーカー(Precision Plus Protein(商標)Dual Xtra Standards、BIO−RAD)と共に、35mVで90分間電気泳動させた。氷ブロックを、走行タンクの内側に配置して、いかなる過熱も防止した。
濃縮ゲルを切り落とし、分離するゲルをMini Transblot Cell(BIO−RAD)内のPVDF移動膜(Thermo Scientific)の上に移動させた。つまり、PVDF移動膜を、メタノールに1分間浸漬し、続いて蒸留水に2分間浸漬することによって活性化した。続いて、すべての層を移動緩衝液で飽和させた(20mM TRISベースpH8.6、154mMグリシン、0.8% w/v SDS、及び20%メタノール)。底部から上部の順に、移動スポンジ、ブロッティングペーパー、ゲル、PVDF移動膜、ブロッティングペーパー、移動スポンジからなる移動サンドイッチを、移動カセットの中に配置し、これを移動緩衝液で充填されたMini Transblot Cellに挿入した。最後に、電気泳動移動は、200mAで90分間行った。氷ブロックを、移動タンクの内側に配置して、いかなる過熱も防止した。
BLOT−Faststain(商標)(G−Biosciences,USA)を使用して、負荷対照として作用する総タンパク質を染色した(Colinsら、2015)。電気泳動移動の直後に、PVDF移動膜を、希釈したBLOT−Faststain(商標)固定剤溶液(10倍)で2分間、優しく振りながら染色した。次いでこの膜を、希釈したBLOT−Faststain(商標)現像液(4倍)で1分間、優しく振りながらインキュベートした。続いて、この膜を4℃の暗所において現像液中で30分間貯蔵して、タンパク質バンドを最大強度に到達させた。最後に、この膜を冷水で洗浄し、地汚れを排除し、G box(Syngene)を使用して撮像した。次いでこの膜を、温かい脱イオン水(40〜45℃)を使用して脱染し、ブロッキング段階のために準備することができる。
PVDF移動膜を、5%脱脂粉乳を含有するTBS(TRIS緩衝生理食塩水、20mM TRISベースpH7.5、0.5mM NaCl)中で1時間ブロックし、次いで各々7分間、TTBS(0.05% v/v Tween−20で補充したTBS)中で2回洗浄した。この膜を、1%脱脂粉乳を含有するTTBS中に希釈した一次抗体を用いて、4℃で一晩インキュベートした(表1)。翌日、一次抗体を除去した。この膜を、各々5分間、TTBS中で3回洗浄し、次いで二次抗体を用いて、室温で1時間インキュベートした。選択される二次抗体は、使用される一次抗体の種類に依存する。それは、1%脱脂粉乳を含有するTTBS中に希釈されたHRP(a9309、Sigma)に複合されたヤギ抗マウス二次抗体(作用濃度:1:1000)、または1%脱脂粉乳を含有するTTBS中に希釈されたHRP(ab6721、abcam)に複合された抗ウサギ二次抗体(作用濃度:1:5000)のいずれかであり得る。二次抗体インキュベーション後、膜を5分間、TTBS中で3回洗浄した後、最後に10分間、TBS中で洗浄した。タンパク質バンドを、G box(Syngene)を使用して検出した。
PVDF膜を、G box(Syngene)内に配置した。焦点及びズーム設定は、確実に膜が画面の中心で最大になるように調整した。Clarity(商標)Western ECL基質(BIO−Rad)からのルミノール及びペルオキシド溶液を等量部で混合し、膜に適用した。画像を、暗所において1分間隔で5分間にわたって撮影して、タンパク質バンドのための最適シグナルを得た。それに続いて、この膜を、分子ラダーの画像を得るために、自動設定を使用して白色光に暴露した。次いでブロット画像を、画像Jを使用して分析した。等しいサイズのボックスを、各レーンのタンパク質バンドの周りに配置し、タンパク質バンドの強度の測定を可能にした。次いで、バックグラウンドをバンド強度から差し引いて、結果をMicrosoft Excel及びGraphpadソフトウェアにおいて分析した。
PVDF移動膜からのタンパク質シグナルを剥離し、異なるタンパク質に関して再検出することができる。つまり、この膜をマイルドな剥離緩衝液(200mMグリシン、3.5mM SDS、1% v/v Tween−20、pH2.2)で2回、各々10分間にわたって洗浄した。続いてこの膜を、PBSで2回、各々10分間洗浄した後、TTBSで2回、各々5分間洗浄した。Clarity(商標)Western ECL基質(BIO−Rad)を膜に付加し、残留タンパク質シグナルをチェックするために、G Box(Syngene)を使用して撮像した。残留シグナルが強すぎる場合、次いで剥離プロセス全体を繰り返した。次いでこの膜を、続くブロッキング段階及び一次抗体検出のために準備した(上記を参照されたい)。
HSを、リーズ大学分子細胞生物学研究所のProfessor Nigel Hooperによって提供された対照及びAD患者の群から採取した。これらの試料は、2008年にオックスフォード大学薬理学部のSusan Greenfiledチームの一員として勤務していた前博士研究員のAmy Hallidayにより、分子量カットオフフィ(MWCO)フィルターで濾過された。血清を濾過し、30kDa超、30〜10kDa、及び10kDa未満の分留に分離した。この研究の場合、30〜10kDa、及び10kDa未満の部分を等量で組み換えて、30kDa未満のHS中に存在するT14のスペクトルを付与した。
標準曲線及び試料を、3回実施した。ヒト脳ホモジネート試料を、1:160、ヒト血清で希釈した。組織試料中のT14ペプチドの決定のための標準曲線を、PBS緩衝液中で希釈した。標準曲線は、8〜100nMのT14の範囲であった。つまり、96ウェルイムノプレート(NUNC)を、100μL/ウェルの試料または標準T14でコーティングし、パラフィルムで被覆して、4℃で一晩インキュベートした。翌日、この試料を、水を流したシンク上でプレートをはじくことによって除去し、Tris緩衝生理食塩水、及びTween20(TBS−T)中に2%ウシ血清アルブミン(BSA)を含有する200μLのブロッキング溶液を付加し、室温で4時間インキュベートした。次いでブロッキング溶液を除去し、ブロッキング溶液中で1μg/mLに希釈した100μLの抗体を付加し、4℃で一晩インキュベートした。翌日に一次抗体を除去し、ウェルを200μLのTBS−Tで3回洗浄した。ブロッキング溶液中で0.1μg/mLに希釈した100μLの二次酵素複合抗体を付加し、室温で2時間インキュベートした後、全インキュベーション中にプレートをパラフィルムで被覆した。2時間後、プレートをTBS−Tで4回洗浄した。3,3,5,5−テトラメチルベンジジンは、発色反応を開始した。この反応を、30分後に2M H2SO4を含有する停止溶液で停止させ、吸光度を、脳ホモジネートの場合はVmaxプレートリーダー(Molecular Devices,Wokingham,UK)において、ヒト血清の場合はVersaMaxプレートリーダー(Molecular Devices,Wokingham,UK)において、450nmで測定した。

実施例1−ペプチド認識
本発明者らは、ポリクローナル抗体をウサギから単離した。図1〜4は、1:1000抗体の用量反応、及び異なるペプチド(すなわち、PEP4、T14、T30、及びT15)の指定ナノモル濃度を示す。
本発明者らは、一連の異なる線状ペプチドとの結合実験を行うことによって、本発明の抗体の免疫特異性を調査する。抗体の免疫特異性を決定するために使用される線状ペプチドの配列は、表2及び3に示される。
図11に示されるように、抗体は、アセチルコリンエステラーゼ(AChE、すなわち、「G4」)を検出し、図12〜14に示されるように、またAChE活性が高濃度の抗体に対して低い場合にのみ著しく、酵素の最終活性に影響を及ぼすことなく、アセチルコリンの加水分解の速度を変更する。脳及び体内のAChE活性が非常に高く、治療法としてのいずれの抗体も、より低い用量範囲内であることを考慮して、酵素活性を低下させることから生じるいかなる副作用も最小限に抑えられ得る。さらに、抗体は、臨床用途の薬物であるガランタミンとは対照的に、およそ15%、AChEを最大限にブロックし、これが活性をはるかに多く、すなわち50%阻害する(WO2015/004430を参照されたい)。
ラット及びヒト試料で検出を行う場合、試料を30KDa MWCOフィルターで濾過し、酵素の検出を妨げるAChE、及び試料中の偽陽性を除去した。結果は、抗体が、両方の試料においてどのようにペプチドを検出するかを示す。図15は、ラット脳ホモジネートにおける結果を示し、図16は、対照及びアルツハイマー病(AD)ヒト脳ホモジネートからの結果を示す。
抗体の特異性
ポリクローナル抗体は、T14ペプチド(決定的に、C末端配列、−VHWK)に高度に特異的であることが証明された。完全AChEタンパク質のその認識は、この配列が暴露され、三次構造で入手可能であることを示唆する。抗体が、線状T30ペプチドフラグメント(その中でT14が起こる)を認識しなかったという事実は、獲得された三次構造の暴露されないT30配列の最後におけるリジン(K)の不在に起因すると考えられる。
これらのデータは、抗体が、驚くべきことに、深温凍結されていたものでも生物組織内で生存可能であること、及びT14様ペプチドが、間接的な証拠のみによって以前に示唆されているように、独立して生化学的実体として存在することを示す(Garcia−Ayllon,M.S.et al.Altered levels of acetylcholinesterase in Alzheimer plasma.PloS one 5,e8701,doi:10.1371/journal.pone.0008701(2010)、Arendt,T.,Bruckner,M.K.,Lange,M.&Bigl,V.Changes in acetylcholinesterase and butyrylcholinesterase in Alzheimer’s disease resemble embryonic development−−a study of molecular forms.Neurochemistry international 21,381−396(1992)。中でも最も特筆すべきことは、対照脳と比較して、ADにおいて有意差が検出され得ることである。
これらの発見は、検出可能な配列(すなわち、−VHWKエピトープ)、及び特にC末端における暴露されたK残基が、診断ツールとして、ならびに細胞毒性部分に結合される場合、抗体自体の可能な使用を含めて、治療介入のための標的として使用され得ることを示唆する。後者の適用は、抗体が、AChE酵素反応の速度を低減し得るが、AChEに対する触媒速度は非常に高いため、閾値を超え得るという発見によって制限され得、なぜガランタミン及びアリセプトなどの薬物が、それにもかかわらず依然として臨床用途にあるかを実際に説明する。
本発明の抗体を使用して、本発明者らは次に、脳脊髄液(CSF)中のT14フィブリル及びAβを比較した。本発明者らは、T14が、フィブリルを形成するため、ウェスタンブロッティングによって50KDaバンドとして検出されることを発見した。これは、T14ウェスタンブロットシグナルの増加によって示されるように、経時的に集合する外因性T14によるさらなる証拠である(図21Aを参照されたい)。
実施例5から引き続き、本発明者らは、次にT14のC末端に特異的なポリクローナル抗体を使用して、追加の脳試料中のT14モノマーを検出した。本発明者らは、脳皮質及び海馬内のT14レベルを比較した。これらの試料を、30kDa MWCOフィルターを使用して濾過し、AChE及びアルブミンなどのより大きなタンパク質を排除し、それによりバックグラウンドシグナルを低減する。
実施例5及び6から明らかであるが、T14は、本発明の抗体を使用して脳及びCSFにおいて容易に検出することができ、生きている患者から脳及びCSF試料を得ることは、高度に侵襲的かつ危険な行為である。したがって、本発明者らは、長期間にわたって−80℃で貯蔵されたヒト血清(HS)試料中のT14を検出するために、抗体を使用する可能性について調査した。この研究のねらいは、これらの試料を使用して、処理され得るより新鮮なHS試料と共に使用するために、ELISAを最適化することである。これらの試料は、30kDa MWCOフィルターを使用して濾過され、AChE及びアルブミンなどのより大きなタンパク質が排除されており、それによりバックグラウンドシグナルを低減する。
図24を参照すると、免疫組織化学による進行したアルツハイマー病におけるグローバルニューロンに存在するT14の第1の可視化が示される。示されるこの特定領域は、パーキンソン病において典型的に脆弱であるため、2つの変性疾患が、毒性T14に基づいて共通の機序を共有するという理論を支持する。
図25を参照すると、対照及び重度のアルツハイマー病(AD)試料における中脳の黒質のT14免疫組織が、40倍及び200倍の拡大で示される。重度のADにおける細胞外T14免疫標識された沈着物(矢印)は、対照及びADにおける典型的なドーパミン作動性ニューロン細胞質T14免疫染色パターン(矢頭)と同様に示される。一連のT14ニューロン細胞質染色パターンが、視野内で捕捉される。図に見られるように、T14は、AD試料の細胞から放出されるが(矢印)、対照では放出されない。AD細胞からのT14放出または分泌は、瀕死のニューロンからの漏出に起因すると考えられる。T14の細胞外沈着物は、暗色メラニン、すなわちドーパミンと癒合すると思われる。

- 前記神経変性障害が、アルツハイマー病である、請求項24または25に記載の使用のための抗体またはその抗原結合フラグメント。
- 各々が必要に応じて誘導体化される、請求項1〜18のいずれか1項に記載の抗体もしくはその抗原結合フラグメント、または請求項21に記載の抗体薬物複合体、及び必要に応じて薬学的に許容されるビヒクルを含む、薬剤組成物。
- 請求項27に記載の薬剤組成物の作製プロセスであって、各々が必要に応じて誘導体化される、治療上有効な量の請求項1〜18のいずれか1項に記載の抗体もしくはその抗原結合フラグメント、または請求項21に記載の抗体薬物複合体を、薬学的に許容されるビヒクルと複合することを含む、プロセス。
- 前記組成物が、神経変性障害の治療組成物である、請求項27に記載の組成物、または請求項28に記載のプロセス。
- 神経変性障害の検出または診断用のバイオマーカーとして使用するための配列番号3またはその変異体もしくはフラグメント。
- バイオマーカーとして使用される前記配列番号3の変異体またはフラグメントが、配列番号6を含むかまたはそれからなる、請求項30に記載の配列番号3またはその変異体もしくはフラグメント。
- 神経変性障害に罹患している被検体、もしくはそれに対する素因の診断、または前記被検体の病態の予後判定の提供のためのキットであって、前記キットが、試験被検体からの試料中に存在する抗原の濃度の検出のための検出手段を含み、前記検出手段が、必要に応じて誘導体化される、請求項1〜18のいずれか1項に記載の抗体またはその抗原結合フラグメントを含み、前記試料中の抗原の存在が、前記被検体が神経変性障害に罹患していることを示唆する、キット。
- 神経変性障害に罹患している被検体、もしくはそれに対する素因の診断方法、または前記被検体の病態の予後判定の提供方法であって、前記方法が、被検体から得られた試料中に存在する前記抗原の濃度を検出することを含み、前記検出が、必要に応じて誘導体化される、請求項1〜18のいずれか1項に記載の抗体またはその抗原結合フラグメントを使用して達成され、前記試料中の抗原の存在が、前記被検体が神経変性障害に罹患していることを示唆する、方法。
- 前記試料が、脳脊髄液(CSF)を含む、請求項32に記載のキット、または請求項33に記載の方法。
- 前記試料が、血液、尿、または組織を含む、請求項32に記載のキット、または請求項33に記載の方法。
- 前記試料が、血液を含む、請求項35に記載のキットまたは方法。
- 前記神経変性障害が、アルツハイマー病、パーキンソン病、ハンチントン病、運動ニューロン病、脊髄小脳1型、2型、及び3型、筋萎縮性側索硬化症(ALS)、ならびに前頭側頭型認知症からなる群から選択され、好ましくは、アルツハイマー病である、請求項30または請求項31に記載の使用のための、配列番号3またはその変異体もしくはフラグメント、あるいは請求項32〜36のいずれか1項に記載のキットまたは方法。
- 神経変性障害の治療、予防、または寛解における使用のための治療化合物の特定のための薬物発見スクリーニングにおける、請求項1〜18のいずれか1項に記載の抗体またはその抗原結合フラグメントの使用。
- T14ペプチド(配列番号3)の合成または活性を阻害する薬剤の特定のための比色標識または蛍光標識T14ペプチド(配列番号3)のエクスビボ使用。
- 神経変性障害の治療、予防、または寛解における使用のための候補薬剤の特定方法であって、
(i)インビトロまたはエクスビボで、請求項1〜18のいずれか1項に記載の抗体またはその抗原結合フラグメントの存在下、細胞を試験薬剤と接触させるステップと、
(ii)前記抗体またはその抗原結合フラグメントを使用して、T14ペプチド(配列番号3)の存在、濃度、または活性を検出するステップと、を含み、T14ペプチドの不在、または対照と比較したT14ペプチドの合成、濃度、もしくは活性の低減が、前記薬剤が、不適切な補体活性化を特徴とする疾患の治療、寛解の予防のための候補であることの指標である、方法。 - T14ペプチド(配列番号3)の合成または活性を阻害する薬剤の特定のためのアッセイであって、
(i)細胞ベースの発現系と、
(ii)請求項1〜18のいずれか1項に記載の抗体またはその抗原結合フラグメントと、
(iii)少なくとも1つの試験薬剤の、前記発現系との接触を可能にするように構成された容器と、を含む、アッセイ。 - T14ペプチド(配列番号3)の合成または活性を調節する薬剤の特定方法であって、
(i)インビトロまたはエクスビボで、請求項1〜18のいずれか1項に記載の抗体またはその抗原結合フラグメントの存在下、細胞を試験薬剤と接触させることと、
(ii)前記抗体またはその抗原結合フラグメントを使用して、T14ペプチド(配列番号3)の存在、濃度、または活性を検出することと、を含み、対照と比較したT14の合成、濃度、または活性における変化が、前記試験薬剤がT14ペプチド(配列番号3)の合成または活性を調節することの指標である、方法。
