- 第1のボツリヌス神経毒血清型F(BoNT/F)亜型由来のLHN骨格および異なるBoNT/F亜型由来の活性化ループ領域を含むキメラポリペプチド。
- 当該LHN骨格が、配列番号17〜32のうちの少なくとも1種と少なくとも60%の配列同一性を有する、請求項1に記載のポリペプチド。
- 当該LHN骨格が、
(a)配列番号33〜48のうちの少なくとも1種と少なくとも40%の配列同一性を有するポリペプチド配列および/または
(b)配列番号49〜64のうちの少なくとも1種と少なくとも70%の配列同一性を有するポリペプチド配列
を含む、請求項1または2に記載のポリペプチド。 - 当該LHN骨格が、BoNT/F5亜型またはBoNT/F7亜型に由来する、請求項1から3のいずれか一項に記載のポリペプチド。
- 当該活性化ループが、コンセンサス配列KSIIPRKGTKAPPRL(配列番号65)に適合するアミノ酸配列またはコンセンサス配列において5個以下のアミノ酸が付加、欠失もしくは置換されているアミノ酸配列を含む、請求項1から4のいずれか一項に記載のポリペプチド。
- 当該活性化ループが、BoNT/F1亜型、BoNT/F2亜型、BoNT/F3亜型、BoNT/F4またはBoNT/F6亜型に由来する、請求項5に記載のポリペプチド。
- 当該活性化ループが、
(a)KSVIPRKGTKAPPRL(配列番号66)、
(b)KSIIPRKGTKQSPSL(配列番号67)または
(c)KSIIPRKGTKAPPRL(配列番号65)
から選択されるアミノ酸配列を含む、請求項5または6に記載のポリペプチド。 - 当該LHN骨格が、BoNT/F7亜型に由来し、BoNT/F活性化ループが、KSVIPRKGTKAPPRL(配列番号66)のアミノ酸配列を含む、前記の請求項のいずれか一項に記載のポリペプチド。
- BoNT/F活性化ループ内の切断部位が、エンドペプチダーゼによって特異的に認識され切断される、前記の請求項のいずれか一項に記載のポリペプチド。
- エンドペプチダーゼによる切断が、活性ホロ毒素の形成をもたらす、請求項9に記載のポリペプチド。
- キメラ二本鎖BoNT/Fポリペプチドであって、
(a)配列番号33〜48のうちの少なくとも1種と少なくとも40%の配列同一性を有するアミノ酸配列を含む軽鎖ポリペプチドおよび
(b)配列番号49〜64のうちの少なくとも1種と少なくとも70%の配列同一性を有するアミノ酸配列を含むHNポリペプチドと
を含み、
HNポリペプチドのN末端が、HNポリペプチドの残部とは異なるBoNT/F亜型由来のアミノ酸残基を含む、キメラ二本鎖BoNT/Fポリペプチド。 - HNポリペプチド配列のN末端アミノ酸残基が、AまたはQである、請求項11に記載の二本鎖ポリペプチド。
- HNポリペプチド配列のN末端が、APPRLCIK(配列番号70)、APPRLCIT(配列番号71)、QSPSLCIK(配列番号72)およびQSPSLCIT(配列番号73)からなる群から選択されるアミノ酸モチーフで始まる、請求項11または12に記載の二本鎖ポリペプチド。
- 軽鎖ポリペプチド配列および/またはHNポリペプチド配列が、BoNT/F5亜型またはBoNT/F7亜型由来である、請求項11から13のいずれか一項に記載の二本鎖ポリペプチド。
- 請求項1から10のいずれか一項に記載のポリペプチドから得られる、請求項11から14のいずれか一項に記載の二本鎖ポリペプチド。
- ターゲティング部分の細胞との直接的な結合による、またはターゲティング部分の、細胞と結合するリガンド(単数または複数)との結合による、ポリペプチドの細胞との結合に適しているターゲティング部分をさらに含む、請求項1から10のいずれか一項に記載のポリペプチドまたは請求項11から15のいずれか一項に記載の二本鎖ポリペプチド。

- 細胞において請求項19に記載のポリヌクレオチドを発現させるステップおよび発現されたポリペプチドを回収するステップを含む、請求項1から10のいずれか一項に定義するポリペプチドを製造する方法。
- 請求項19に記載のポリヌクレオチドを細胞中に導入するステップをさらに含む、請求項21に記載の方法。
