Abstract
Description
化粧品組成物におけるペパーミント抽出物の使用は、長い間知られている。
したがって、本発明の主要な目的は、ペパーミント抽出物の新規使用を提供することである。
本発明は、ケラチノサイト前駆細胞におけるNOTCHタンパク質の発現不足に関連する、皮膚の表皮細胞の再生を維持または制御するための、ペパーミント抽出物の化粧品組成物における使用に関し、前記組成物は、美容的に許容され得る賦形剤も含む。
−CHIMEXがMEXORYL(登録商標)SXの名称で販売しているテレフタリリデンジカンフルスルホン酸(INCI名:テレフタリリデンジカンフルスルホン酸)
−ビス−ベンゾアゾリル誘導体、例えば、欧州特許第669323号および米国特許第2,463,264号に記載されているもの、より詳細には、Haarmann & Reimerが商品名NEO HELIOPAN(登録商標)APとして販売している化合物フェニルジベンゾイミダゾールテトラスルホン酸二ナトリウム
−p−アミノ安息香酸(INCI名:PABA)およびその誘導体、例えば、1−(4−アミノベンゾエート)−1,2,3−プロパントリオール(INCI名:グリセリルPABA)およびBASFがUVINUL(登録商標)P25の名称で販売しているPEG−25PABA、
−特に、MERCKが商品名EUSOLEX(登録商標)232として販売している、2−フェニルベンズイミダゾール−5−スルホン酸(INCI名:フェニルベンズイミダゾールスルホン酸)
−サリチル酸トリエタノールアミン、
−CHIMEXがMEXORYL(登録商標)SLの名称で販売している3−(4’−スルホベンジリデン)カンフル(INCI名:ベンジリデンカンフルスルホン酸)、
−参照名Tinosorb(登録商標)MとしてまたはFAIRMOUNT CHEMICALがMIXXIM(登録商標)BB/100として販売しているメチレンビス−ベンゾトリアゾリルテトラメチルブチルフェノール(USAN名:ビスオクトリゾール);
−CHIMEXが「MEXORYL SO」の名称で販売している3−(4’−トリメチルアンモニウムベンジリデン)−1−ボルナン−2−オンメチルスルフェート(INCI名:カンファーベンザルコニウムメトスルフェート)、
−商品名UVINUL(登録商標)MS40として販売されているベンゾフェノン−4。
本研究は、クローンを形成する能力が高いケラチノサイト(「ケラチノサイト幹細胞」または「幹細胞」または「前駆細胞」とも称される)またはその能力が中間もしくは低いケラチノサイト(一時的増幅細胞と称される娘細胞)を解析および識別することができる、BarrandonおよびGreenが開発した方法(Barrandon Y and Green H.;Proc Natl Acad Sci USA 1987,84:2302−2306)に従ってケラチノサイトクローンを用いて行った。
26歳の女性ドナーの腹壁形成術を起源とする正常ヒトケラチノサイト(NHK)を使用する。
II.1 MicroLabSTAR(HAMILTON)を用いた全RNAの入手
細胞培養液を除去し、250μlの溶解緩衝液RLT(Nucleospin RNAキット,Ref.740709.4,Macherey−Nagelに供給されているもの)を加える。細胞スクレイパーを用いて細胞を掻き取り、次いで、細胞溶解産物を1.2mlの深ウェルプレート(Nucleospin RNAキットに供給されているもの)に回収する。次いで、全RNAを抽出する。得られた全RNAの溶液を、MicrolabSTARに連結されたマイクロプレートリーダーであるspectrostarNANO(BMG Labtech)を用いてアッセイする。
使用される逆転写(RT)キットは、供給されるプロトコルに従って使用されるHigh Capacity Reverse Transcription Kit(Ref.4368813,Life Technologies−Applied)である。500ngの全RNAを水に希釈して、最終体積を25μlにする。次いで、それらを、下記に示されるように前もって調製された25μlのHigh Capacity Reverse Transcription Kit 2X反応混合物の存在下において、25℃で10分間、次いで37℃で2時間インキュベートする。異なる操作をMicrolabSTARプラットフォームによって行い、異なるインキュベーションをTRobot(Biometra)において行う。

処理の効果を、7900HTの高速96ウェルブロックおよびLife Technologies−Applied製の試薬を用いるリアルタイム定量的PCRによって評価する。使用されるTaqMan遺伝子発現アッセイプライマーは、以下である:

−NOTCH1標的遺伝子:Hs00413187
−NOTCH2標的遺伝子:Hs00225747

リアルタイム定量的PCRは、その効率が90%〜110%である場合に利用され得る。この効率は、適切な細胞から抽出されたcDNAの連続希釈物から標準的な範囲を設定することによって評価される。得られた傾きの値は、増幅効率を反映し;2nの法則(n=サイクル数、すなわち40)が満たされ、傾きの値が−3.32であるとき、PCRは100%の効率である。各サンプルについて、シグナルが現れるサイクル数を、SDS2.3ソフトウェアによって測定し;標準的な範囲を用いて確立された較正直線によって、転写物のコピー数に関する濃度が算出され得る。同じアッセイでは、目的の転写物の発現レベルを、ハウスキーピング遺伝子のベータ−2−ミクログロブリンについて得られた値に対して標準化する。この遺伝子は、その発現が構成的かつ不変であり、特に逆転写工程の効率のばらつきに起因する、アッセイ間の量の任意の変動を克服することを可能にする。転写活性の定量は、標的遺伝子およびハウスキーピング遺伝子のCt(サイクル閾値)の差によって測定される。これは、相対的定量であり、以下の計算によって得られ、ここで、無処理が1に等しい。

ペパーミント抽出物は、NOTCH1およびNOTCH2の遺伝子発現を刺激する。
ペパーミント抽出物を、以下の化粧品組成物において、表皮細胞の再生を維持または制御するための活性物質として使用する。

Claims (10)
- ケラチノサイト前駆細胞におけるNOTCHタンパク質の発現不足に関連する皮膚の表皮細胞の再生を増加させるために使用するための有効量のMentha piperitaとも称されるペパーミントの抽出物を含む組成物。
- 前記NOTCHタンパク質が、NOTCH1およびNOTCH2タンパク質であることを特徴とする、請求項1記載の使用。
- 無処理コントロールと比較して表現される、定量的リアルタイムPCR(qRT−PCR)によって測定されるNOTCH1またはNOTCH2タンパク質をコードする遺伝子の遺伝子発現の定量(RQ)が、1を超えることを特徴とする、請求項2記載の使用。
- 前記抽出物が、水性媒質中の酵素的加水分解によって得られる、気生部分の抽出物であることを特徴とする、請求項1〜3のいずれか1項記載の使用。
- 前記組成物が、該組成物の重量に対して0.005〜0.5重量%の乾物の該抽出物を含むことを特徴とする、請求項1〜4のいずれか1項記載の使用。
- 前記組成物が、顔面、より詳細には、日射に曝露される範囲である、眼の周囲、胸部、口唇および手に適用されることを特徴とする、請求項1〜5のいずれか1項記載の使用。
- 前記皮膚が、乾燥した皮膚または成熟した皮膚などの再生プロセスが乱れた皮膚であることを特徴とする、請求項1〜6のいずれか1項記載の使用。
- 前記組成物が、ブナ(Fagus Sylvatica)の芽の水性抽出物またはBellis perennisの花の水性抽出物から選択される、NOTCHタンパク質の発現に対して作用する活性物質を含むことを特徴とする、請求項1〜7のいずれか1項記載の使用。
- 前記組成物が、UVスクリーニング剤および/または太陽光を反射することができる粒子を含むことを特徴とする、請求項1〜8のいずれか1項記載の使用。
- ケラチノサイト前駆細胞におけるNOTCHタンパク質の発現不足に関連する皮膚の表皮細胞の再生を維持または制御するための皮膚の美容的ケアのための方法であって、該方法は、皮膚の老化、光老化、皮膚の再生プロセスの乱れによって引き起こされるたるみまたは乾燥の徴候を示す顔面または身体の少なくとも一部への有効量のペパーミント抽出物の適用を含む、方法。
