強力な免疫刺激性活性を有するポリヌクレオチドおよびキメラ化合物が依然として必要とされている。その上、再現性よく製造することができる強力なキメラ化合物が依然として必要とされている。許容される毒性プロファイルを有するポリヌクレオチドおよびキメラ化合物が特に望ましい。

ポリヌクレオチド(PN)およびキメラ化合物(CC)の合成。
ポリヌクレオチドは、製造者のプロトコールに従って酸化的硫化を用いてホスホラミジット化学反応を使用して固相合成によって製造し、精製および単離した(Molecules、2013年、18巻、14268〜14284頁)。使用したヌクレオシドモノマーは、5’−ジメトキシトリチル−保護−2’−デオキシヌクレオシド、3’−((2−シアノエチル)−(N,N−ジイソプロピル))−ホスホラミジットであった。CCについては、HEGスペーサーを、18−O−ジメトキシトリチルヘキサエチレングリコール,1−((2−シアノエチル)−(N,N−イソプロピル))−ホスホラミジット(例えば、Glen Research、Sterling、VA製Space Phorphoramidite 18)を使用して組み込んだ。D56−11については、5’−C6−ジスルフィドリンカーを、1−O−ジメトキシトリチル−ヘキシル−ジスルフィド−1’−((2−シアノエチル)−(N,N−ジイソプロピル))−ホスホラミジット(例えば、Glen Research、Sterling、VA製Thiol−Modifier C6 S−S)を使用して組み込んだ。D56−07については、3’−C3−ジスルフィドリンカーを、1−O−ジメトキシトリチル−プロピル−ジスルフィド,1’−スクシニル−固体支持体(例えば、Glen Research、Sterling、VA製3’−Thiol−Modifier C3 S−S CPG)を使用して組み込んだ。D56−02については、3’−C6−ジスルフィドリンカーを、1−O−ジメトキシトリチル−ヘキシル−ジスルフィド,1’−スクシニル−固体支持体(例えば、Glen Research、Sterling、VA製3’−Thiol−Modifier C6 S−S CPG、またはPrime Synthesisからの特注として)を使用して組み込んだ。

D56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)製造スキームは、図1に示した通り、3段階で構成される。FICOLLへの他のPNまたはCCコンジュゲートは、PNもしくはCC配列、PNもしくはCCへのチオールリンカー、および/またはアミン架橋剤へのチオールを変更することによって、同じ製造経路により調製することができる。
FICOLL含有中間体および生成物中のFICOLL濃度を判定する手順。
FICOLL含有中間体および生成物のFICOLL濃度は、FICOLL PM400を、アッセイの検量線を作成するのに使用したことを除いて、製造者のプロトコールに従ってPierce Glycoprotein Carbohydrate Estimation Kit(製品#23260、Thermo Scientific、Rockford、IL)を使用して判定した。
[AECM]z−FICOLL溶液中のアミン濃度およびアミン:FICOLLモル比(z)を判定する手順。
[AECM]z−FICOLLのアミン濃度は、製造者のプロトコールに従ってPierce Fluoraldehyde OPA Reagent Solution(製品#26025、Thermo Scientific、Rockford、IL)を使用して判定した。グリシンを使用してアッセイの検量線を作成した。アミン:FICOLLモル比(z)は、アミン濃度をFICOLL濃度で除すことによって計算し、ここでFICOLL濃度は、実施例S4に記載した通りに判定し、濃度は、モル濃度の単位でのものであった。
[マレイミド]y−FICOLL溶液中のマレイミド濃度およびマレイミド:FICOLLモル比(y)を判定する手順。
[マレイミド−PEG6]y−FICOLLおよび[マレイミド−MC]y−FICOLLのマレイミド濃度は、エルマン試薬(5,5’−ジチオ−ビス−(2−ニトロ安息香酸)、製品番号22582、Thermo Scientific、Rockford、IL)を使用して判定した。[マレイミド]y−FICOLLを製造者のプロトコールに従って過剰のシステインと反応させ、残っているシステインをシステイン検量線を使用して定量化した。マレイミド濃度は、初期システイン濃度から残っているシステイン濃度を減じることによって判定した。マレイミド:FICOLLモル比(y)は、マレイミド濃度をFICOLL濃度で除すことによって計算し、ここでFICOLL濃度は、実施例S4に記載した通りに判定し、濃度は、モル濃度の単位でのものであった。
FICOLLコンジュゲート(例えば、D56−05、[(D56−01)−PEG6]x−FICOLLとも呼ばれる)中のポリヌクレオチド(PN)またはキメラ化合物(CC)濃度およびPN:FICOLLまたはCC:FICOLLモル比(x)を判定する手順。
D56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)のD56−01(CC)濃度は、紫外分光光度法およびベールの法則式を使用して判定した。(慣習により、FICOLLに付着したキメラ化合物は、この段階で、リンカーを有するキメラ化合物、D56−03がこの化合物を形成するのに使用されたが、配列名称D56−01によって参照されることに留意されたい)。260nmの吸光度を判定し、D56−01について22.65mg/ml−1×cm−1の吸光係数を使用した。FICOLLおよびリンカーは、260nmで吸収せず、したがって吸光度は、CC、D56−01の吸光度に単に起因する。mg/mLでのD56−01濃度をD56−01の遊離酸の分子量を使用してモル濃度に変換した。CC:FICOLLモル比(x)は、CC濃度をFICOLL濃度で除すことによって判定し、ここでFICOLL濃度は、実施例S4に記載した通りに判定し、濃度は、モル濃度の単位でのものであった。他のPN−FICOLLまたはCC−FICOLL溶液の濃度は、必要に応じて、使用したPNまたはCCの吸光係数および遊離酸分子量を使用して判定する。
粒径を判定する手順。
FICOLL試料(例えば、D56−05)の粒径(Z−アベレージ)および標準偏差(SD)は、Malvern Zetasizer機器を使用して動的光散乱(DLS)によって測定した。試料を10mMリン酸ナトリウム、142mM塩化ナトリウム、pH7.2 緩衝液で0.5mg/mLのFICOLL濃度に希釈し、規定された機器設定下で測定した。較正された50nmポリスチレンナノスフェア試料(製品#3050A、Thermo Scientific、Rockford、IL)をシステム適性対照として分析に含め、これは、49±6nmの粒径を有していた。
精製した、[(D56−01)−PEG6]x−FICOLLとも呼ばれるD56−05の物理化学的特徴付け。
D56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)の5つのパイロットロットを実施例S3に概説した手順を使用して製造した。これらのロットを特徴付け、結果を表S9−1に要約する。純度は、215nmで検出して表S3−2に概説した手順を使用してSEC−HPLCによって判定したところ、99%超〜100%の範囲であった。FICOLL濃度は、実施例S4に記載の手順によって判定した。FICOLL濃度については、ダイアフィルトレーションにおける保持液の最終濃度を制御することによって目標を定めることができる。D56−01濃度およびD56−01:FICOLL比(x)は、実施例S7に記載の手順によって判定した。D56−01:FICOLLモル比(x)は、117〜140の範囲であり、実施例S3に記載の製造手順がFICOLLに対するキメラ化合物の担持量について高レベルの制御をもたらすことを実証した。このプロセスの標的D56−01:FICOLLモル比(x)は、120±30である。D56−05の粒径は、実施例S8に記載した通りに判定した。
D56−05凍結乾燥製剤。
5℃超で貯蔵されるD56−05溶液製剤の安定性に限界があったため、制御された室温で良好な安定性を実現することを目標としたD56−05凍結乾燥製剤の開発を行った(実施例S11を参照)。凍結乾燥される製剤の組成の選択は、熱安定性に影響し得るpHおよびイオン強度を制御するのに一般的に安全と認められる(GRAS)賦形剤を使用したプレフォーミュレーション研究に基づいた。D56−05霊長類投与量に基づいた一連の試験製剤を冷凍/解凍安定性について試験し、溶液透明度およびHPLCサイジングクロマトグラフィーに基づいて評価した。1mg/ml D56−05(パイロットロット2)、10mMリン酸カリウム(pH=7.5)、および300mMトレハロースを含有する製剤は、10サイクル実験を通じて同一のクロマトグラフィー挙動および動的光散乱プロファイルを示したので、さらなる開発のために選択した。上述した製剤を、およそ−35℃での棚冷凍、その後の−35℃(約60μbar真空)での36時間の一次乾燥、30℃の棚温度への2時間の移行、および追加の24時間にわたる二次乾燥からなる凍結乾燥サイクルに付した。凍結乾燥した生成物(40バイアル)は、1〜1.4%の残留水分を有すると示された許容されるケーキであった。製剤を水1mL中に復元し、生成物は、製剤直後または凍結乾燥および復元プロセス後に分析したとき、SEC−HPLCによってまったく同様に挙動することが示された。
D56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)溶液および凍結乾燥製剤の安定性。
本実施例は、液体および凍結乾燥した物質の安定性を記述する。
異なるマレイミド:FICOLLモル比(y)を用いた[マレイミド−PEG6]y−FICOLLの調製、および精製したD56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)におけるD56−01:FICOLLモル比(x)に対するインパクト。
異なるマレイミド:FICOLLモル比(y)を有する[マレイミド−PEG6]y−FICOLLロットを、より小さいスケールであることおよび異なる量のSM−PEG6(アミン当たり0.25、0.5、0.75、1.0、& 2.0当量)を使用したことを除いて、実施例S3、セクションEに記載の手順を使用して(AECM)z−FICOLL(アミン:FICOLLモル比(z)=224)から生成した。マレイミド:FICOLLモル比(y)は、実施例S4およびS6に記載の手順によって判定した。表S12−1の結果は、異なる量のSM−PEG6を添加すると、8〜185のマレイミド:FICOLLモル比(y)を有する[マレイミド−PEG6]y−FICOLLが生じたことを示す。次いで[マレイミド−PEG6]y−FICOLLロットをD56−03(マレイミド当たり1.1当量)と反応させ、得られたD56−05ロットを実施例S3、セクションGに記載した通りに精製した。D56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)ロットのD56−01:FICOLLモル比(x)を実施例S4およびS7に記載した通りに判定したところ、24〜154の範囲であった(表S12−1)。これらの結果は、D56−05中のD56−01担持量を、[マレイミド−PEG6]y−FICOLLの調製に使用されるSM−PEG6の量によって制御することができることを示す。これらの化合物のin vitro効力について実施例B9を参照。
それぞれn=24、45、および70であるSM−PEGnリンカーを使用するD56−25、D56−26、およびD56−27([(D56−01)−PEGn]x−FICOLLとも呼ばれる)の調製。
異なるPEGリンカー長を有する[マレイミド−PEGn]y−FICOLLロットを、より小さいスケールであることおよびそれぞれn=24、45、および70のSM−PEGnリンカーを使用したことを除いて、実施例S3、セクションEに記載の手順を使用して(AECM)z−FICOLL(アミン:FICOLLモル比(z)=224)から生成した(SM−PEGnの化学構造については図5を参照)。使用したSM−PEG24試薬は、Thermo Fisher(Rockford、IL)から得た。これは、nが24である図5Aに示した構造を有する。使用したSM−PEG45およびSM−PEG70試薬は、Nanocs Inc.(New York、NY)から得た。構造は、図5−Bに示した通りである(nは、それぞれ45および70である)。得られた[マレイミド−PEGn]y−FICOLLロットのマレイミド:FICOLLモル比(y)は、実施例S4およびS6に記載した通りに判定し、結果を表S13−1に示す。マレイミド:FICOLLモル比(y)は、199〜227の範囲であり、PEGリンカー長は、得られるマレイミド:FICOLLモル比に有意に影響しなかったことを示した。
[マレイミド−MC]y−FICOLLの調製。
[マレイミド−MC]y−FICOLLを、米国特許第8,597,665号に記載のスルホ−SMCCリンカーを使用して[AECM]z−FICOLLから製造した。[マレイミド−MC]y−FICOLLは、オイルアウトおよび/または沈殿として観察される水性緩衝液中の溶解性問題を示した。マレイミド−MC−FICOLLの取り扱いが困難だったため、チオール活性化ポリヌクレオチド(PN)またはキメラ化合物(CC)とのコンジュゲーション反応は一貫性がなかった。
Alexa Fluor(登録商標)555−(D56−05)(Alexa Fluor(登録商標)555/[(D56−01)−PEG6]x−FICOLLとも呼ばれる)の調製。
Alexa Fluor(登録商標)555のアミン反応性誘導体(Alexa Fluor(登録商標)555−NHSエステル)は、Life Technologies(Foster City、CA)から購入した。実施例S3に記載した通りに調製したAECM−FICOLLをAlexa Fluor(登録商標)555−NHSエステルおよびSM−PEG6の混合物と反応させることによって活性化してAlexa Fluor(登録商標)555/[マレイミド−PEG6]y−FICOLLを形成し、これを実施例S3に記載した通りにD56−03((D56−01)−3’−SHとも呼ばれる)と反応させてAlexa Fluor(登録商標)555−(D56−05)(Alexa Fluor(登録商標)555/[(D56−01)−PEG6]x−FICOLLとも呼ばれる)を得た。
D56−08、D56−09、D56−12、およびD56−13の調製。
D56−08、D56−09、D56−12、およびD56−13を米国特許第8,597,665号に記載されている通りに調製する。

(実施例B1)
ヒト白血球の単離および刺激。
ポリヌクレオチド(PN)およびキメラ化合物(CC)の活性を、ヒト末梢血単核細胞(PBMC)および単離B細胞によるサイトカイン分泌の測定、およびB細胞増殖の測定によってin vitroで査定した。細胞培養培地中に分泌されたサイトカインレベルは、酵素結合免疫吸着測定法(ELISA)によって測定した。
マウス脾細胞の単離および刺激
ポリヌクレオチド(PN)およびキメラ化合物(CC)の活性を、マウス脾細胞によるサイトカイン分泌を測定することによってin vitroで査定した。細胞培養培地中に分泌されたサイトカインレベルは、酵素結合免疫吸着測定法(ELISA)によって測定した。
線状キメラ化合物(CC)配列最適化。
2つの別個の実験を行った。以下の表において、平均は、幾何平均を指す。
D56−05は、D56−01より強力なin vitro応答を誘導する。
ヒトおよびマウスサイトカインの両方の誘導の効力に基づいて、D56−01をナノ粒子製剤を開発するためのリード候補として選択した。D56−01配列を実施例S3に記載した通りにFICOLLにコンジュゲートさせて、D56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)を生成した。次いでD56−01およびD56−05を、ヒトPBMC IFN−αおよびヒトB細胞IL−6の誘導についての相対的なin vitro効力を比較した。ヒトPBMC IFN−α活性およびヒトB細胞IL−6活性の結果を、それぞれ表B4−1および表B4−2に示す。配列D56−10およびD56−14を本実験における歴史的陽性対照として使用した。
D56−05は、D56−01より強力なin vivo応答を誘導する。
D56−05およびD56−01をマウスおよびカニクイザルに投与した後の自然免疫応答の誘導を評価した。サルにおいて、インターフェロン経路関連遺伝子発現を、D56−05およびD56−01を投与する前、および投与して24時間後に収集した血液試料中で測定した。マウスにおいて、インターフェロン経路およびケモカイン関連遺伝子発現を、化合物投与の18時間後に回収した注射部位流入領域リンパ節中で測定した。D56−05およびD56−01を獲得する抗原提示細胞集団の相対能力を、化合物注射の24時間後にマウスから回収した注射部位流入領域リンパ節中で評価した。その上、成熟マーカー発現(CD69、CD86)を、化合物注射の20時間後に回収したリンパ節由来の形質細胞様樹状細胞(pDC)に対して測定した。
D56−05アジュバント急速高力価毒素中和抗体は、rPAに応答し、生きたBacillus Anthrax胞子での致死的チャレンジからサルを保護する。
D56−05の哺乳動物被験体における防御抗原特異的抗体応答を誘導する能力(すなわち、免疫原性)を、D56−05+rPAで免疫したサルにおいて評価した。比較のために、追加の群のサルをD56−01+rPAまたはrPA単独で免疫した。D56−05で1回または2回の免疫した後の保護を試験するために、サルに致死量の炭疽胞子でチャレンジし、生存時間を監視した。
等価な用量で、ナノ粒子製剤D56−05は、遊離線状キメラ化合物D56−01と比較して、マウスにおいて安全性利点を実証する。
in vivoでCC配列の安全性/耐容性に対するナノ粒子製剤の効果を試験するために、マウスに、筋肉内経路によってD56−05およびD56−01の繰り返しの高用量注射を投与した。毒性を査定する種として、マウスは、マウスにおけるTLR9発現のより広範な細胞分布に起因して、霊長類と比較して、CpG ODN含有ヌクレオチドに対して誇張された薬理学的応答を実証する。本発明者らは、遊離およびナノ粒子バージョンのCCの相対的安全性を査定するために、合計4回の注射で2週間毎にD56−05またはD56−01のいずれか100μg(D56−01の重量に基づいて)を受けているマウスにおいて血清サイトカイン応答を測定し、体重の変化を監視した。
D56−05の腫瘍内投与は、リンパ腫細胞株EG7−OVA腫瘍を有するマウスにおける腫瘍増殖を抑制する。
がん免疫療法のモデル(Mooreら、Cell、54巻:777頁、1998年)におけるD56−05の適用を試験するために、リンパ腫細胞株EG7 l OVAを使用して、確立された腫瘍の増殖に対するD56−05または対照非CpG含有オリゴヌクレオチド(D56−30)の腫瘍内注射の効果を査定した。
異なるD56−01:FICOLLモル比(x)を有するD56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)コンジュゲートのin vitro効力評価。
生物学的応答の効力に対するD56−05([(D56−01)−PEG6]x−FICOLLとも呼ばれる)コンジュゲート中の様々なD56−01:FICOLLモル比(x)の効果をin vitro分析によって査定した。ヒトB細胞内のin vitro効力を、異なるD56−01:FICOLLモル比(x)を有するD56−05コンジュゲート:D56−05 x=24、D56−05 x=53、D56−05 x=82、D56−05 x=124、およびD56−05 x=154について実施例B1に記載した通りに判定した。ヒトB細胞IL−6アッセイの結果を表B9−1に示す。異なるD56−01:FICOLLモル比(x)を有するD56−05の合成については実施例S12を参照。

- 前記連結のすべてが、ホスホロチオエートエステル連結である、請求項12から16のいずれか一項に記載のポリヌクレオチド。
- 5’−TCGGCGC−3’−HEG−5’−AACGTTC−3’(配列番号4)として2つの核酸部分および1つのヘキサエチレングリコール(HEG)スペーサーを含む線状キメラ化合物であって、50未満のヌクレオチドを含有し、前記ヌクレオチド同士間および前記ヌクレオチドと前記HEGスペーサーとの間の1個または複数個の連結は、ホスホロチオエートエステル連結である、線状キメラ化合物。
- 5’−TCGGCGC−3’−HEG−5’−AACGTTC−3’−HEG−5’−TCGGCGC−3’(配列番号2)として3つの核酸部分および2つのヘキサエチレングリコール(HEG)スペーサーを含む線状キメラ化合物であって、50未満のヌクレオチドを含有し、前記ヌクレオチド同士間および前記ヌクレオチドと前記HEGスペーサーとの間の1個または複数個の連結は、ホスホロチオエートエステル連結である、線状キメラ化合物。
- 5’−TCGGCGC−3’−HEG−5’−AACGTTC−3’−HEG−5’−TCGGCGC−3’(配列番号2)からなる、請求項19に記載の線状キメラ化合物。
- 前記核酸部分が、2’−デオキシリボポリヌクレオチドである、請求項18から20のいずれか一項に記載の線状キメラ化合物。
- 前記連結のすべてが、ホスホロチオエートエステル連結である、請求項18から21のいずれか一項に記載の線状キメラ化合物。
- (i)薬学的に許容される賦形剤、ならびに(ii)請求項1から11のいずれか一項に記載の分枝状キメラ化合物、請求項12から17のいずれか一項に記載のポリヌクレオチド、および請求項18から22のいずれか一項に記載の線状キメラ化合物からなる群のうちの1つを含む医薬組成物。
- 前記分枝状キメラ化合物、前記ポリヌクレオチド、および前記線状キメラ化合物がそれぞれ、
ヒト末梢血単核細胞によるIFN−アルファの産生を刺激すること;
ヒトBリンパ球によるIL−6の産生を刺激すること;ならびに
マウス脾細胞によるIL−12p40およびIL−6の一方または両方の産生を刺激すること
からなる群のうちの1つまたは複数を含む、哺乳動物白血球によるサイトカイン産生を刺激することができる、請求項23に記載の医薬組成物。 - 前記分枝状キメラ化合物、前記ポリヌクレオチド、および前記線状キメラ化合物がそれぞれ、哺乳動物Bリンパ球の増殖を刺激することができる、請求項23または請求項24に記載の医薬組成物。
- 滅菌溶液である、請求項23から25のいずれか一項に記載の医薬組成物。
- 前記分枝状キメラ化合物、前記ポリヌクレオチド、および前記線状キメラ化合物に共有結合的に連結されていない抗原をさらに含む、請求項23から26のいずれか一項に記載の医薬組成物。
- 前記抗原が、微生物抗原、アレルゲン、または腫瘍抗原である、請求項27に記載の医薬組成物。
- 本質的にエンドトキシンフリーである、請求項23から28のいずれか一項に記載の医薬組成物。
- 哺乳動物被験体における免疫応答を刺激する方法であって、哺乳動物被験体における免疫応答を刺激するのに十分な量で請求項23から29のいずれか一項に記載の医薬組成物を前記哺乳動物被験体に投与することを含む方法。
- 免疫応答を刺激することが、
IFN−アルファ産生を刺激すること;
IL−6産生を刺激すること;
Bリンパ球増殖を刺激すること;
インターフェロン経路関連遺伝子発現を刺激すること;
化学誘引物質関連遺伝子発現を刺激すること;および
形質細胞様樹状細胞(pDC)成熟を刺激すること
からなる群のうちの1つまたは複数を含む、請求項30に記載の方法。 - 前記医薬組成物が抗原をさらに含む場合、免疫応答を刺激することは、抗原特異的抗体応答を誘導することを含み、前記抗原特異的抗体応答の力価は、前記抗原が前記分枝状キメラ化合物、前記ポリヌクレオチド、または前記線状キメラ化合物と組み合わせて投与されるときには、前記抗原が前記分枝状キメラ化合物も前記ポリヌクレオチドも前記線状キメラ化合物も無しで投与されるときより高い、請求項30または31に記載の方法。
- 哺乳動物被験体における抗原特異的抗体応答を誘導する方法であって、哺乳動物被験体における抗原特異的抗体応答を誘導するのに十分な量で請求項27から29のいずれか一項に記載の医薬組成物を前記哺乳動物被験体に投与することを含む方法。
