Claims (11)
- ヒトの臨床および治療適用のためのヒト脂肪間質細胞の臨床的成長のための細胞培養培地であって、前記培地は、
哺乳細胞培養に適した基礎培地と、
抗凝血剤なしで収集され、かつ血清処理前に凝固することができるヒト血清と、
(i)成長促進量のヒトインスリン、(ii)ヒトトランスフェリン、(iii)ヒト組み換え型表皮成長因子、(iv)ヒト組み換え型血漿由来成長因子−BB、(v)塩基性線維芽細胞成長因子(FGF2)、および(vii)フェノールレッドのうちの少なくとも1つと、
を有し、前記培地はリノール酸を含まない、細胞培養培地。 - 請求項1記載の培地において、前記培地は40以上、かつ50倍加できる培地。
- 請求項1記載の培地であって、18〜20時間につき1倍加の成長率を達成し、かつ維持する培地。
- 請求項1記載の培地であって、少なくとも35〜37に持続される多能性である培地。
- 請求項1記載の培地であって、分化が生じる前に2ヶ月以上成長を持続する培地。
- ヒトの臨床および治療適用のためのヒト脂肪間質細胞の臨床的成長のための細胞培養培地を作成する方法であって、前記方法は、
哺乳細胞培養に適した基礎培地、
抗凝血剤なしで収集され、かつ血清処理前に凝固することができるヒト血清、及び
(i)成長促進量のヒトインスリン、(ii)ヒトトランスフェリン、(iii)ヒト組み換え型表皮成長因子(iv)ヒト組み換え型血漿由来成長因子−BB、(v)塩基性線維芽細胞成長因子(FGF2)、および(vii)フェノールレッドのうちの少なくとも1つ、
の構成要素を組み合わせる工程からなる、方法。 - 多能性の間充織幹細胞の成長と存在を拡大する方法であって、
a.請求項1記載の成長培地を用意する工程と、
b.工程aの培地中で間充織幹細胞を増殖させる工程と、
を有する方法。 - 請求項7記載の方法によって作成された間充織幹細胞であって、前記細胞は40+倍加まで増殖可能であり、自発的な分化が生じる前に最大2ヶ月までの間、成長が拡大され得る間充織幹細胞。
- 請求項7記載の方法によって作成された間充織幹細胞であって、多能性は35〜50倍加で存在する間充織幹細胞。
- 請求項7記載の方法によって作成された間充織幹細胞であって、間充織幹細胞は脂肪由来の幹細胞であり、未分化細胞は以下のフェノタイプ:CD14−、CD19−、CD29+、CD31−、CD34−、CD44+、CD45−、CD49d+、CD73+、CD90+、CD105+、およびCD146+を有する間充織幹細胞。
- 請求項7記載の方法によって作成された間充織幹細胞であって、前記細胞はオイルレッド+脂肪細胞、アルシアンブルー+軟骨細胞、およびアリザリンレッド+骨細胞に容易に分化する間充織幹細胞。
