本発明は、概して、炭酸脱水酵素IX(CAIX)に特異的なキメラ抗原受容体細胞、および腎細胞癌などのCAIX発現癌の処置のために該細胞を使用する方法に関する。

炭酸脱水酵素は、生体内のpHバランスを維持するために二酸化炭素の可逆的な水和を触媒する亜鉛金属酵素のファミリーである。炭酸脱水酵素IX(CAIX)は、分子量が54/58 kDaの膜貫通糖タンパク質である。構造的に、CAIXは、N末端プロテオグリカン様ドメイン(PG)(aa53〜111)、CA触媒ドメイン(CA)(aa135〜391)、膜貫通らせんセグメント(aa415〜434)および短い細胞質内尾部(aa434〜459)の4つのドメインからなる。低酸素条件下で、CAIX遺伝子は、低酸素誘導転写因子HIF-1αによって転写レベルで直接的に活性化され、細胞外培地へのプロトンの輸送およびpHの低下をもたらす。したがって、CAIXの発現は、様々な腫瘍における低酸素の代理マーカーであるとみなすことができる。CA活性により生じた腫瘍微小環境の酸性化およびCAIXのPGドメインにおけるO結合型グリカン構造内のケラチン硫酸単位は、細胞接着および腫瘍進行のプロセスにおいて重要な役割を果たしていると考えられている。

本発明にしたがう細胞は、それを必要とする患者において癌を処置するために使用され得る。別の態様において、本発明にしたがう単離された細胞は、それを必要とする患者において癌を処置するための医薬の製造において使用され得る。
本発明は、典型的にはドナーから得られたT細胞を非同種反応性細胞に形質転換することができる限り、同種免疫療法に特に適している。これは、標準的なプロトコルの下で行われ得、必要な回数複製され得る。得られた改変T細胞は、プールされ得、そして1または複数の患者に投与され得、したがって「在庫あり」の治療製品として利用可能となる。
本発明のCARは、発現ベクターから発現され得る。そのような発現ベクターを作製する組み換え技術は、当技術分野で周知である。
「ベクター」という用語は、核酸配列を、それを複製することができる細胞内に導入するために挿入することができるキャリア核酸分子を表すために使用される。核酸配列は、「外因性」であり得、これはそれがベクターを導入する細胞に対して外来性であることまたはその配列がその細胞内の配列に対して相同であるがその配列が通常見出されない宿主核酸内の位置にあることを意味する。ベクターは、プラスミド、コスミド、ウイルス(バクテリオファージ、動物ウイルスおよび植物ウイルス)ならびに人工染色体(例えば、YAC)を含む。当業者は、標準的な組み換え技術を通じてベクターを構築する技術を十分に有している(例えば、両方とも参照により本明細書に組み入れられる、Maniatis et al., 1988およびAusubel et al., 1994を参照のこと)。
「プロモーター」は、転写の開始および割合を制御する核酸配列の一領域である制御配列である。それは、核酸配列の個々の転写を開始させるための、調節タンパク質および分子、例えばRNAポリメラーゼおよび他の転写因子が結合し得る遺伝子要素を含み得る。「機能的に配置」、「機能的に連結」、「制御下」および「転写制御下」というフレーズは、プロモーターが、核酸配列との関係で、その配列の転写開始および/または発現を制御するよう正確な機能的位置および/または向きにあることを意味する。
特定の態様において、プラスミドベクターは、宿主細胞を形質転換するために使用されることが想定されている。一般に、レプリコンおよび宿主細胞と適合する種由来の制御配列を含むプラスミドベクターが、これらの宿主に関連して使用される。ベクターは通常、複製部位および形質転換細胞において表現型選択を提供することができるマーキング配列を有する。非限定的な例において、大腸菌(E.coli)はしばしば、大腸菌種由来のプラスミドであるpBR322の誘導体を用いて形質転換される。pBR322は、アンピシリンおよびテトラサイクリン耐性のための遺伝子を含み、したがって形質転換細胞を同定するための簡易な手段を提供する。pBRプラスミドまたは他の微生物プラスミドまたはファージはまた、例えば、それ自信のタンパク質の発現のために微生物によって使用され得るプロモーターを含まなければならないまたは含むよう改変されなければならない。
細胞に感染しまたは受容体を介したエンドサイトーシスを通じて細胞に侵入し、宿主細胞のゲノムに組み入りウイルス遺伝子を安定的かつ効率的に発現させる特定のウイルスの能力は、それらを、細胞(例えば、哺乳動物細胞)への外来核酸の移入における魅力的な候補にしている。本発明の要素は、本発明の1つまたは複数のCARをコードするウイルスベクターであり得る。本発明の核酸を送達するために使用され得るウイルスベクターの非限定的な例は以下に記載されている。
特定の核酸送達方法は、アデノウイルス発現ベクターを使用する。アデノウイルスベクターはゲノムDNAへの組み込みのための積載能力が低いことが知られているが、この特徴は、これらのベクターによりもたらされる高い遺伝子移入効率によって相殺される。「アデノウイルス発現ベクター」は、(a)コンストラクトのパッケージングを支えるおよび(b)究極的にはその中にクローニングされた組織または細胞特異的なコンストラクトを発現させるのに十分なアデノウイルス配列を含むコンストラクトを含むことを意味する。遺伝子編成およびアデノウイルスの知識により、36 kb、直鎖状の二本鎖DNAウイルスは、アデノウイルスDNAの大きな部分を最大7 kbの外来配列で置換させることができる(Grunhaus and Horwitz, 1992)。
核酸は、アデノウイルスを介したトランスフェクションを用いて細胞に導入され得る。高いトランスフェクション効率が、アデノウイルス利用システムを用いた細胞システムで報告されている(Kelleher and Vos, 1994; Cotten et al., 1992; Curiel, 1994)。アデノ随伴ウイルス(AAV)は、高い組み込み度数を有し、非分裂細胞に感染することができ、したがって例えば組織培養下での(Muzyczka, 1992)またはインビボでの哺乳動物細胞への遺伝子の送達に有用であることから、本発明の細胞で使用される魅力的なベクターシステムである。AAVは、感染性に関して幅広い宿主範囲を有する(Tratschin et al., 1984; Laughlin et al., 1986; Lebkowski et al., 1988; McLaughlin et al., 1988)。rAAVベクターの作製および使用に関する詳細は、各々参照により本明細書に組み入れられる米国特許第5,139,941号および同第4,797,368号に記載されている。
レトロウイルスは、それらの遺伝子を宿主ゲノムに組み込み、多量の外来遺伝物質を移入させ、広範囲の種および細胞型に感染し、特別な細胞株においてパッケージングされる能力から、送達ベクターとして有用である(Miller, 1992)。
他のウイルスベクターも、本発明においてワクチンコンストラクトとして利用され得る。ワクシニアウイルス(Ridgeway, 1988; Baichwal and Sugden, 1986; Coupar et al., 1988)、シンドビスウイルス、サイトメガロウイルスおよび単純ヘルペスウイルス等のウイルス由来のベクターが利用され得る。それらは、様々な哺乳動物細胞に対して様々な魅力的な特徴を提供する(Friedmann, 1989; Ridgeway, 1988; Baichwal and Sugden, 1986; Coupar et al., 1988; Horwich et al., 1990)。
送達される核酸は、特定の結合リガンドを発現するように操作された感染性ウイルス内に収容され得る。ウイルス粒子は、したがって、標的細胞の同種受容体に特異的に結合し、その内容物を細胞に送達するであろう。レトロウイルスベクターの特異的標的化を可能にするよう設計された新規のアプローチは、ウイルスエンベロープへのラクトース残基の化学的付加によるレトロウイルスの化学的改変に基づき開発された。この改変は、シアロ糖タンパク質受容体を通じた肝細胞の特異的感染を実現し得る。
細胞のトランスフェクションまたは形質転換のための適当な核酸送達方法は、当業者に公知である。そのような方法は、例えばエクスビボトランスフェクションによる、注射による等のDNAの直接送達を含むがこれらに限定されない。当技術分野で公知の技術の適用を通じて、細胞は、安定的または一過的に形質転換され得る。
生物から取り出された真核生物細胞および組織をエクスビボ状況でトランスフェクトする方法は、当業者に公知である。したがって、細胞または組織は、取り出され、そして本発明の核酸を用いてエクスビボでトランスフェクトされ得ることが想定されている。特定の局面において、移植された細胞または組織は、生物中に導入され得る。好ましい局面において、核酸は、移植された細胞において発現される。
本明細書に記載される任意の組成物は、キットに含まれ得る。非限定的な例において、細胞治療に使用するための1つもしくは複数の細胞および/または組み換え発現ベクターを有する細胞治療に使用するための1つもしくは複数の細胞を生成するための試薬が、キットに含まれ得る。キットの構成要素は、適当な収容手段によって提供され得る。
本発明の特定の態様において、臨床的局面のための本発明の方法は、過剰増殖性疾患の処置において有効な他の薬剤、例えば抗癌剤と併用される。「抗癌」剤は、例えば癌細胞を殺す、癌細胞においてアポトーシスを誘導する、癌細胞の成長速度を低下させる、転移の発生率もしくは回数を減少させる、腫瘍のサイズを減少させる、腫瘍の成長を阻害する、腫瘍もしくは癌細胞への血液供給を減少させる、癌細胞もしくは腫瘍に対する免疫応答を促進する、癌の進行を防ぐもしくは阻害する、または癌を有する対象の寿命を延ばすことによって、対象において癌に負の影響を与えることができる。より一般的に、これらの他の組成物は、細胞を殺すまたは細胞の増殖を阻害するのに効果的な併用量で提供され得る。このプロセスは、発現コンストラクトと薬剤または複数の要素を同時に癌細胞と接触させることを含み得る。これは、両方の薬剤を含む単一の組成物もしくは薬学的配合物を細胞と接触させることによって、または1つの組成物が発現コンストラクトを含み、他が第2の薬剤を含む、2つの別個の組成物もしくは配合物を同時に細胞と接触させることによって達成され得る。
癌治療はまた、化合物ベースおよび放射線ベースの両方の処置との様々な併用療法を含む。併用化学療法は、例えば、アブラキサン、アルトレタミン、ドセタキセル、ハーセプチン、メトトレキサート、ノバントロン、ゾラデックス、シスプラチン(CDDP)、カルボプラチン、プロカルバジン、メクロレタミン、シクロホスファミド、カンプトテシン、イホスファミド、メルファラン、クロラムブシル、ブスルファン、ニトロソ尿素(nitrosurea)、ダクチノマイシン、ダウノルビシン、ドキソルビシン、ブレオマイシン、プリカマイシン(plicomycin)、マイトマイシン、エトポシド(VP16)、タモキシフェン、ラロキシフェン、エストロゲン受容体結合剤、タキソール、ゲムシタビン、ナベルビン、ファルネシルタンパク質トランスフェラーゼ阻害剤、トランス白金、5-フルオロウラシル、ビンクリスチン、ビンブラスチンおよびメトトレキサートまたは上記の任意のアナログもしくは誘導体変種ならびにそれらの組み合わせを含む。
DNA損傷を与える、広く使用されている他の要素は、ガンマ線、X線および/または腫瘍細胞への放射性同位体の直接送達として広く知られているものを含む。他の形態のDNA損傷要素、例えばマイクロ波およびUV照射もまた想定されている。これらの要素のすべてが、DNAに対して、DNAの前駆体に対して、DNAの複製および修復に対して、ならびに染色体の構築および維持に対して広範囲の損傷を与えると考えられる。X線の線量範囲は、長期間(3〜4週間)にわたる50〜200レントゲンの1日線量から2000〜6000レントゲンの単回線量に及ぶ。放射性同位体の線量範囲は様々であり、同位体の半減期、発生する放射線の強度およびタイプならびに新生物細胞による取り込みに依存する。
免疫療法は、通常、癌細胞を標的化し癌細胞を破壊する免疫エフェクター細胞および分子の使用に基づく。免疫エフェクターは、例えば、腫瘍細胞の表面上のいくつかのマーカーに特異的な抗体であり得る。抗体は単独で、治療剤のエフェクターとして機能し得、またはそれは実際に細胞死滅がもたらされるよう他の細胞を誘導し得る。抗体はまた、薬物または毒素(化学療法剤、放射性核種、リシンA鎖、コレラ毒素、百日咳毒素等)に連結され、標的化剤としてのみ利用され得る。あるいは、エフェクターは、腫瘍細胞標的と直接的または間接的のいずれかで相互作用する表面分子を有するリンパ球であり得る。様々なエフェクター細胞は、細胞傷害性T細胞およびNK細胞を含む。
さらに別の態様において、二次的処置は、本発明の臨床態様の前、後またはそれと同時に治療用ポリヌクレオチドを投与する遺伝子療法である。細胞増殖の誘導因子、細胞増殖の阻害因子またはプログラムされた細胞死の調節因子を含む様々な発現産物が本発明に含まれる。
癌患者のおよそ60%は、予防、診断または病期判定、治癒および緩和手術を含むいくつかのタイプの手術を受けるであろう。治癒手術は、他の治療、例えば本発明の処置、化学療法、放射線療法、ホルモン療法、遺伝子療法、免疫療法および/または代替療法と組み合わせて使用され得る癌処置である。
他の薬剤も、処置の治療効果を改善するために、本発明と組み合わせて使用され得ることが想定される。これらのさらなる薬剤は、免疫調節剤、細胞表面受容体の上方調節およびGAP結合に影響を及ぼす薬剤、細胞増殖抑制性および分化剤、細胞接着の阻害剤、またはアポトーシス誘導剤に対する過剰増殖細胞の感度を高める薬剤を含む。免疫調節剤は、腫瘍壊死因子、インターフェロンアルファ、ベータおよびガンマ、IL-2および他のサイトカイン、F42Kおよび他のサイトカインアナログ、またはMIP-1、MIP-1ベータ、MCP-1、RANTES、ならびに他のケモカインを含む。細胞表面受容体またはそれらのリガンド、例えばFas/Fasリガンド、DR4またはDR5/TRAILの上方調節は、過剰増殖細胞に対するオートクリンまたはパラクリン効果の確立により本発明のアポトーシス誘導能力を高めることもさらに想定される。GAP結合の数を増加させることによる細胞内シグナルの増加は、隣接する過剰増殖細胞集団に対する抗過剰増殖効果を高めるであろう。他の態様において、細胞増殖抑制性または分化剤が、処置の抗過剰増殖効果を改善するために本発明と組み合わせて使用され得る。細胞接着の阻害剤は、本発明の効果を改善すると考えられる。細胞接着阻害剤の例は、接着斑キナーゼ(FAK)阻害剤およびロバスタチンである。アポトーシスに対する過剰増殖細胞の感度を高める他の薬剤、例えば抗体c225が、処置の効果を改善するために本発明と組み合わせて使用され得ることもさらに想定される。
細胞、培養培地および試薬。ヒトCAIX+腎細胞癌細胞株sk-rc-52(本明細書においてSkrc52とも称される)、sk-rc-09およびCAIX- sk-rc-59(本明細書においてSkrc59とも称される)は、Memorial Sloan-Kettering Cancer Center, New YorkのGerd Ritter博士から入手した。それらを、10% FCS、2 mmol/L L-グルタミン、100 U/mlペニシリンおよび100μg/mlストレプトマイシン(Sigma)を補充したRPMI 1640培地(Life Technologies)を含むR-10完全培地中、5% CO2下、37℃で培養した。初代ヒトT細胞は、10%ヒト血清および100 IU/ml組み換えヒトインターロイキン2(IL-2)(Chiron)を含むR-10中で維持した。ヒト胚腎臓細胞株293T(ATCC)およびマウス線維芽細胞NIH3T3細胞(ATCC)は、10% FCS、100 U/mlペニシリンおよび100μg/mlストレプトマイシン(Sigma)を含むDMEM培地を含むD-10完全培地(Life Technologies)中で成長させた。Children's Hospital Bostonの血液バンクから入手したLeukopackは、書面上のインフォームドコンセントを得た健常ボランティアから収集されたものであった。

(CD28コンストラクトの順方向プライマー)および両コンストラクトの逆方向プライマー
、イタリック体はPacI部位、である。これらのDNAフラグメントは、以下の配列にしたがって配置された機能的特徴をコードする:NotI-C9タグ(TETSQVQPQ)-GGGGS-CD8またはCD28-TCRζ-PacI。配列TETSQVQPQはSEQ ID NO:78を有する。配列GGGGSはSEQ ID NO:79を有する。内部C9ペプチドによりタグ付加されたキメラTCRコンストラクトを、NotIおよびPacI制限部位を用いて、抗CXCR4 scFv-Fc、クローン48を含むpcDNA3.1-F105L-ヒンジ-スタッファーベクターにクローニングした。この設計により、我々は、キメラTCR受容体コンストラクトをFc部分フラグメントと置き換わるよう挿入した。次に、抗CAIX scFv(クローンG36)および抗CCR5 scFv(無関係のscFv対照としての、クローンA8)抗体フラグメントを、SfiI/NotI部位で抗CXCR4 scFvと置き換わるようクローニングし、CAIX特異的キメラTCRコンストラクトを作製した。
統計学的有意性は、両側スチューデントt検定を用いて決定した。
我々は以前に、それらのエピトープマッピング、発現レベルおよびCAIXをインターナライズする能力に関して相違する高親和性ヒト抗CAIX抗体のパネルを報告した23。我々の最初の目的は、CAR構築の候補としてこれらの抗CAIX単鎖抗体のうちの5つの抗腫瘍活性を調査することであった。抗CAIX mAbを通じたADCCを試験するため、scFvをscFv-Fc(hIgG1)ミニボディに変換した23。我々は、すべてのscFv-Fcが抗原特異的腫瘍溶解を示すことを見出した。CAIX-腫瘍細胞株sk-rc-59に対する溶解が<5%というバックグラウンドの下で、高CAIX+発現を示す腫瘍細胞株sk-rc-09に対して、特異的溶解は40〜57%の範囲であり、中程度のCAIX+発現を示すsk-rc-52に対して、特異的溶解は46〜60%の範囲であった。陰性対照scFv-Fc、例えば抗CXCR4 48-Fc23および抗SARS 11A-Fc24に対しては、バックグラウンドレベルの細胞溶解しか観察されなかった(図1)。ADCC死滅および他の報告されている分析に基づき、scFvG36を、CAR標的化部分としてさらなる評価のために選択した。
MHC提示を回避し、T細胞エフェクター機能を向上させるために共刺激分子CD28のシグナル要素を組み込んだ第2世代G36-CD28z CART細胞の報告された優れた効果を第1世代G36-CD8z CART細胞と比較するための研究を行った。図3Aに示されるように、CAIX+ sk-rc-52細胞との一晩のインキュベート後、対照A8 CD28z CART細胞またはLAK細胞単独では、低レベルのI型サイトカインIL-2、IFNγおよびIL-17分泌のみが観察された。対照的に、第1世代および第2世代の両方のG36発現CART細胞は高レベルのサイトカイン分泌を示し、第2世代G36-CD28z CART細胞は第1世代G36-CD8z CART細胞との比較で、それらの高い活性化状態を反映する多量のI型サイトカインを分泌した。具体的に、G36-CD28z CART細胞は、G36-CD8z CART細胞よりそれぞれ6.5倍、2.3倍および4倍多いIL-2、IFNγおよびIL-17を分泌した。この2つのG36 CART細胞によるサイトカイン分泌の誘導の特異性は、CAIX- sk-rc-59細胞を用いた場合のそれらの最小の刺激により観察される。
異なるG36 CART細胞の死滅活性をさらに評価するため、インビトロ細胞傷害性アッセイを構築した。異なるエフェクター対標的比を用いて、G36-CD28z CART細胞およびその2回インビボ継代サブクローン4-1は、CAIX+腫瘍sk-rc-52の最も多量の細胞溶解を示した(図3C)。25:1を超える高い比を用いた場合、G36-CD28z CART細胞は、G36-CD8z CART細胞よりも2〜3倍高い細胞傷害性を示し、5:1の低い比を用いた場合、G36-CD28z CART細胞は、G36-CD8z CART細胞よりも8〜9倍高い溶解性を示した。しかし、G36-CD28z CART細胞は100:1のE:T比を用いた場合でもなおも60%超の腫瘍溶解を示す良好な細胞傷害性を示した。無関係のA8-CD28z CART細胞および対照T細胞LAKは、最大100:1のE:T比を用いた場合に、約20%の溶解を示すバックグラウンドの非特異的腫瘍溶解を示した。CAIX-腫瘍sk-rc-59を用いたすべての例で、形質導入および非形質導入T細胞は、バックグラウンド溶解を示した。
CAIX+腫瘍細胞と短期間接触させたときの向上したサイトカイン分泌および細胞傷害性とは別に、CARコンストラクトへのCD28共刺激分子の組み込みは、抗原特異的腫瘍細胞と長期間接触させたときに改善された増殖を示した。非形質導入および形質導入(約20%)T細胞を、100単位/mlのヒトIL-2の存在下で毎週新たに照射した腫瘍細胞と混合した。一定量のT細胞に対する異なるレベルの抗原刺激を試験するため、我々は1:8、1:4および1:2の腫瘍細胞対T細胞比を使用した。T細胞数は、トリパン除去により計数し、CART細胞画分をフローサイトメトリーにより試験した。CAIX- sk-rc-59腫瘍細胞との培養下で、形質導入および非形質導入T細胞の数は維持された(図4A下)。対照T細胞の基準レベルの増殖の欠如は、腫瘍細胞株によって分泌される多量の抑制性サイトカインに起因するものと考えられる。対照的に、CAIX+ sk-rc-52腫瘍細胞との2週間の培養の後、1:8の比で、G36-CD28z CART細胞の集団は30倍に増加し、G36-CD8z CART細胞は17倍に増殖し、1:4の比で、G36-CD28z CART細胞の数は19倍に増加し、G36-CD8z CART細胞は4倍に増加した。多量の腫瘍細胞の下で、G36-CD28zまたはG36-CD8zのいずれのCART細胞も増殖することができなかった。無関係のA8-CD28z CART細胞および対照T細胞LAKは、腫瘍細胞の下で増殖を示さなかった(図4A上)。
照射した腫瘍細胞と1または2週間接触させた形質導入T細胞を、新たな非照射腫瘍細胞との24時間の接触後のサイトカイン分泌についても試験した。CAIX+腫瘍(sk-rc-52)との1または2週間の接触により、G36-CD28z CARを通じた共刺激シグナルはG36-CD8z CARで見られたよりも2倍〜2.5倍多いIFN-γ分泌を示したものの、G36-CD28zおよびG36-CD8z CART細胞は同様のIFN-γ分泌レベルを示した(表1)。IL-2分泌に関して、G36-CD28zおよびG36-CD8z CART細胞の2週間の腫瘍接触は、1週間の接触よりも多いIL-2分泌を示した。G36-CD28z CART細胞は、1週間の接触ではG36-CD8z CART細胞よりも5倍多い、2週間の接触では2.5倍多いIL2を生じた。加えて、腫瘍細胞と2週間接触させたG36-CD28z CART細胞は、1回の腫瘍接触よりも3.3倍多いIL-2を分泌し、G36-CD8z CARTは、1週間の腫瘍接触と比較して2週間の後に6.8倍多いIL-2分泌を示した。これらの結果は、形質導入CART細胞が、2回目の腫瘍刺激後に疲弊せず機能的活性を維持していたことを示している。CAIX- sk-rc59細胞と接触させたA8-CD28z、LAKおよびG36 CART細胞の処置では、バックグラウンドレベルのIFN-γおよびIL-2分泌しか観察されなかった。
我々は次に、同様の腫瘍曲線を生じるよう形成されたsk-rc-52腫瘍細胞を左脇腹に、sk-rc-59腫瘍細胞を右脇腹に接種したヌードマウスにおいて、CART細胞が形成された腫瘍細胞の成長を阻害することを試験した。典型的な腫瘍サイズが約6x6 mmとなった腫瘍移植後7日目に、5000万個のG36-CD28z CART細胞、A8-CD28z CART細胞または非形質導入T細胞(LAK)を静脈内注射した。養子T細胞療法を、第1の試験ではn=7、第2の試験ではn=8のグループサイズの2つの別個の実験で、腹腔内注射を通じて高用量IL-2(2x105 IU)の存在下で実施した。腫瘍の成長および細胞療法の効果を比較するために、T細胞処置を含めなかった。
インビボ研究で使用した形質導入T細胞の総集団のサンプルを、Far red色素で事前染色し、その集団内でZsGreenタンパク質を発現するCART細胞を共焦点顕微鏡により分析した。これらの結果は、我々のFACS分析と同様、約30%の形質導入効率を示した(図6A)。
* - 形質導入されたT細胞を照射した腫瘍細胞と共に1または2週間インキュベートし、次いで収集し、洗浄し、新鮮な非照射腫瘍細胞と共に一晩インキュベートし、24時間後に上清をサイトカイン分析のために回収した。腫瘍細胞と相互作用しなかったT細胞培養物では、<50 pg/ml IFN-γおよび<10 pg/ml IL-2のレベルのバックグランドレベルのサイトカインしか検出されなかった。
図5に報告されている実験(実験1、n = 7および実験2、n = 8)由来のマウスを10日目における応答について採点した。部分応答は、対照腫瘍(左脇腹にsk-rc-52および右脇腹に対照腫瘍sk-rc-59を有する同じマウスにおける同じT細胞処置)の容積の30%未満への腫瘍の退行と定義される。
フィッシャー検定の結果 - *G36-CD28z対LAK;**G36-CD28z対A8-CD28z;N.S. - 腫瘍の数とLAKを形質導入したT細胞とA8-CD28zを形質導入したT細胞の間の部分応答の間に統計学的に有意な関係がない。
