Abstract

Description

一実施形態において、エジェクタ面とフレキシブルプリント回路をともに接着すること(例えば、ジェネレータプレートに連結されたエジェクタプレート)は、例えば、表面処理(プラズマエッチング、ウェットエッチング、機械的研磨などによる粗面化処理)、及び、高圧縮におけるプラスチック(一般値750F/350 psiのポリイミド、350F/350 psiのPEEKなど)のガラス遷移温度を超える加熱加圧によって、接着剤に特異的な加熱及び加圧下で硬化する接着剤の薄いシートを適用すること、又はその他の接着によって、達成することができる。
実施例1
本実施例は、ヒト対象における本開示のエジェクタ装置を用いた臨床拡張試験に関する。本実施例は、本開示のエジェクタ装置により、指向性液滴流での低投与量の医薬組成物を送達することによって、点眼器の投与量の1/4を用いるだけで、標準的な点眼器と同等の拡張が示されることを実証する。
本試験の第1の治療群では、35人の対象の片方の眼に、エジェクタ装置から、3.0 μlの用量の2.5%フェニレフリンを2回、3.0 μlの用量の1%トロピカミドを2回投与し、また、他方の眼に、標準的な点眼器から、2.5%フェニレフリンを1回(およそ26 μl)、1%トロピカミドを1回(およそ26 μl)投与する。本試験の第2の治療群では、35人の対象の片方の眼に、エジェクタ装置から、6 μlの用量の2.5%フェニレフリンを1回、6 μlの用量の1%トロピカミドを1回投与し、また、他方の眼に、標準的な点眼器から、2.5%フェニレフリンを1回、1%トロピカミドを1回投与する。本試験の第3の治療群では、35人の対象の片方の眼に、エジェクタ装置から、1.5 μlの用量の2.5%フェニレフリンを1回、1.5 μlの用量の1%トロピカミドを1回投与し、また、他方の眼に、標準的な点眼器から、2.5%フェニレフリンを1回、1%トロピカミドを1回投与する。
図15Aは、従来の点眼器の投与と比較した、本開示の噴霧エジェクタ装置による送達における処置前基準線からの測定された平均拡張変化率を示している。図15Bは、点眼器と比較した、噴霧エジェクタ装置における投与前拡張基準線に対する平均拡張相違率を示している。相違は、対象に特定して算出し、そして、平均される。
図15Aは、エジェクタ装置(例えば、低投与量の医薬組成物の指向性液滴流)と、標準的な点眼器の双方によって送達された拡張薬剤によって、実質的に対象の眼を拡張させ、また、投与後、時間が経過するほど、60分の最大投与後測定点まで、単調に平均拡張度が増加することを示している。
本実施例は、眼圧(IOP)に対するラタノプロストの効果を評価するための、緑内障のビーグル犬を用いた臨床試験に関する。具体的には、本試験は、未処置の眼と比較した、本開示の噴霧エジェクタ装置による1.5 μlの0.005%ラタノプロストの1日1回の滴下後におけるIOP及び瞳孔径(PD)の減少について評価する。
フロリダ大学の緑内障ビーグル犬のマッケイコロニー(MacKay Colony)から、1匹の緑内障ビーグル犬(雌、3歳)を、本試験に選択した。試験を開始する前に、動物を最低1週間休ませた。試験開始前に、動物の片方の眼を、1.5 μlの0.005%ラタノプロストを投与するように割り当て、反対側の眼を、対照として未処置のままにするように割り当てた。基本的な測定プロトコルに常に従い、同じ操作者によって行なった。PDを、ジェイムソンキャリパー(Jameson Caliper)(mm水平)によって測定した。IOPを、清浄なプローブ(iCare)を備えたTonoVet機器を用いて測定した。IOP及びPDを、0、1、2、4、7及び18時間の時点において、2日間、毎日測定した。0時間の時点での測定直後に、噴霧装置を用いて、動物の割り当てられた眼に1.5 μlの0.005%ラタノプロストを投与した。反対側の眼は、未処置のままにした。噴霧装置の精度をすべての使用前後に確認し、処置前に10%の送達用量の精度が必要であった。標準測定を、1、2、4、7及び18時間の時点において、毎日、継続した。考えられ得るエンドポイントには、過度の眼刺激、眼の損傷、及びその他の疾患又は創傷が含まれていたが、これらに限定されなかった。統計交差は、本試験に利用しなかった。
処置された眼のIOPは、2時間で、11 mmHgまで減少し、初期値から最大16 mmHg減少した(図16A)。PDは、1時間で極小まで(0)減少し、7時間まで維持された(図16B)。
Whisper(商標)MDD装置によって送達される1.5 μlの用量の0.005%ラタノプロストは、従来の治療法と同様に、IOPの低下及び瞳孔の収縮において有効であることを示した。本試験は、噴霧エジェクタ装置が、1.5 μlの用量のラタノプロストを常に送達できることを確認する。

処置された眼のIOPは、2時間で、19 mmHgまで減少し、初期値から12 mmHg減少した(図17A)。最大の減少は、第2日目の7時間においてみられた。PDは、1時間で極小まで(0)減少し、7時間まで維持された(図17B)。
結果は、1.5 μlの用量の0.005%ラタノプロストとともに、3.0 μlの用量が、従来の治療法と同様に、IOPの低下及び瞳孔の収縮において有効であることを示す。本試験は、噴霧エジェクタ装置が、3.0 μlの用量のラタノプロストを常に送達できることを確認する。

フロリダ大学の緑内障ビーグル犬のマッケイコロニーから、6匹の緑内障ビーグル犬(4匹の雄及び2匹の雌、3〜8歳)を、本試験に選択した。包含される条件には、専門動物眼科医(boarded veterinary ophthalmologist)の肉眼による検査によって判断される、IOPの上昇及び緑内障の症状の文書化が含まれていた。
噴霧エジェクタ装置及び一般的な点眼器の処置の結果は同様であった。9 μlの用量は、従来の治療法と同様に、IOPの低下及び瞳孔の収縮において有効であった。点眼器投与の作用持続性は大きかったが、噴霧エジェクタ装置による治療は、IOPの基準線よりも有意に低い値を維持していた(図18A及び18B)。噴霧エジェクタ装置を用いて処置された眼の刺激は、点眼器によって処置された眼で観察されたものよりも少なかった。
噴霧エジェクタ装置を介した1日1回午前中投与による、9 μlの0.005%ラタノプロスト投与は、7時間の試験期間で5日間にわたって、一般的な点眼器の用量のおよそ1/3で、従来の点眼器による治療法と同様に、IOPの制御において有効であった。薬力学(PD)試験は、実施例12の薬物動態(PK)試験のコンパニオン試験(companion study)であり、実施例12は、噴霧エジェクタ装置による9 μlのラタノプロストの送達後における眼房水中のラタノプロストの酸のバイオアベイラビリティを、点眼器によって送達される26 μlのラタノプロストと比較している。
本実施例は、眼圧(IOP)に対するラタノプロストの効果を評価するための、緑内障のビーグル犬を用いた臨床試験に関する。具体的には、本試験は、6匹の緑内障ビーグル犬での並行無作為化交差試験のIOP低下における噴霧エジェクタ装置による低用量ラタノプロストの効果について評価する。試験時に、標準的な点眼器から1滴(およそ24 μl)として送達する場合に対する、本開示のエジェクタ装置を用いてラタノプロストを(例えば、低投与量の指向性液滴流として)6 μlの用量で2回送達する場合の緑内障ビーグル犬のIPOを測定する。
フロリダ大学(UF)のマッケイ緑内障ビーグル犬コロニーから、6匹の緑内障動物(4匹の雄及び2匹の雌、3〜8歳)を、本試験に選択した。包含される条件には、専門動物眼科医の肉眼による検査によって判断される、眼圧(IOP)の上昇及び緑内障の症状の文書化が含まれていた。
2日の休息後、片方の眼を、噴霧エジェクタ装置の送達によって薬剤を投与するように無作為に割り当て、反対側の眼に、点眼容器から従来のラタノプロストを投与する。
図19A〜19Cは、エジェクタ装置が、従来の点眼器を用いて得られたものと概して一致することを示している。試験時のIOP圧を図19Aに示し、試験時のIOPの変化を図19Bに示し、そして、試験時の瞳孔径を図19Cに示している。
要約すると、1日1回滴下した場合に、低投与量の0.005%ラタノプロストは、標準的な点眼器に相応して、有意に緑内障ビーグルのIOPを低下させる。IOPの低下は、約20 mmHg(45%)から27 mmHg(60%)まで変動した。結果は、たとえ、本開示のエジェクタ装置によって送達される2回の6 μlの用量が、標準的な点眼器によって送達される量のおよそ1/2であっても、(例えば、低投与量の指向性液滴流として)本開示のエジェクタ装置によって達成されたイヌのIOPの低下が、低投与量の標準的な点眼器によるものと統計的に区別できないことを示す。

噴霧エジェクタ装置及び点眼器によって処置された動物のIOPの低下は、同様であった(図20)。噴霧エジェクタ装置による送達される30 μlの用量は、従来の点眼器による治療法と同様に、IOPの低下及び瞳孔の収縮において有効であった。
結果は、噴霧エジェクタ装置が、30 μlの用量の0.005%ラタノプロストを送達できることを確認する。本パイロット試験において、噴霧エジェクタ装置による薬剤送達は、従来の点眼器による治療法と同様に、IOPを制御するのに有効であり、作用持続性が増加する傾向にあることが実証された。本試験は、この一連の試験において1.5 μl〜30 μlである、プロスタグランジンプロドラッグ送達の有効投与量範囲を遂行する。
本実施例は、眼圧(IOP)に対するトラボプロストの効果を評価するための、緑内障のビーグル犬を用いた無作為化交差臨床試験に関する。具体的には、本試験は、本開示の噴霧エジェクタ装置によって送達される9 μlの用量の0.004%トラボプロスト(Travatan Z(登録商標)、Alcon Laboratories社(Fort Worth, TX USA)製)の有効性について、24時間眼圧(IOP)用量/応答曲線の抑制及び平低化、並びに午前中の眼圧(IOP)スパイクの制御において、マイクロピペットによる送達と比較する。また、本試験は、噴霧エジェクタ装置の有効性について、IOP及び眼の刺激をモニタリングすることによっても、標準的なマイクロピペットと比較する。

噴霧エジェクタ装置によって、試験第1日目の12時間の時点において、基準線からIOPが有意に変化した。最初の24時間におけるIOPの最大変化は、−20.0mmHg(59%)であった。第1日目の初期平均IOPレベルは33.75mmHgであり、5日間の処置後、観察された最も高い日平均ピークは18.83mmHgであり、これは、同様の基準線レベルの30.83mmHgを下回っていた(図21A)。試験第1日目の12時間の時点において、噴霧エジェクタ装置処置によって有意なPD変化がみられた。虹彩は、12時間の時点において最大の縮瞳(1.17mm)に達し、モニタリングの残りの時間にわたって、基準線レベルに上昇し始めた(図21B)。本試験において、HRの変化はなく、その平均値は、25拍/15秒(100bpm)であった。過度の眼刺激又はその他の眼の問題についての報告はなかった。
トラボプロストは、毎日24時間、IOPを低下させるために検討したプロスタグランジンアゴニストの中で最も有効であり、プロスタグランジンプロドラッグによる処置で多くみられた午前中のIOPスパイクの優れた制御を含む。
本実施例は、眼圧(IOP)に対するトラボプロストの効果を評価するための、緑内障のビーグル犬を用いた無作為化交差臨床試験に関する。具体的には、本試験は、本開示の噴霧エジェクタ装置による18μlの0.004%トラボプロストの1日1回午前中又は午後の滴下後の眼圧(IOP)に対する効果について、従来の点眼器による1日1回平均26μlの送達と比較して、評価する。
フロリダ大学の緑内障ビーグル犬のマッケイコロニーから、6匹の緑内障ビーグル犬(4匹の雄及び2匹の雌、3〜8歳)を、本試験に選択した。包含される条件には、専門動物眼科医の肉眼による検査によって判断される、眼圧(IOP)の上昇及び緑内障の症状の文書化が含まれていた。
噴霧エジェクタ装置及び従来の点眼器の結果は同様であった。噴霧エジェクタ装置よって送達される18μlの投与量は、従来の治療法と同様に、IOPの低下及び瞳孔の収縮において有効であった。点眼器と比較して、噴霧エジェクタ装置による薬剤送達後のIOPの低下の持続性に有意差はなかった。両方の処置は、試験期間において、全24時間の基準線よりもはるかに低いIOP値を維持していた(図22A〜22D)。噴霧エジェクタ装置を用いて処置された眼の刺激は、点眼器によって処置された眼で観察されたものよりも少なかった。
噴霧エジェクタ装置による18μlの午前中又は午後用量のトラボプロストの送達は、点眼器平均送達用量(26μl)のおよそ2/3であっても、従来の治療法と同様に、IOPの制御に有効である。処置後のIOPは、常に20mmHg未満を維持していた。IOP及びこの一連の先行試験における日変化の効果を含む数学モデルに基づき、トラボプロストの1日1回午後投与によって、最大24時間のIOP低下が提供される必要がある。噴霧エジェクタ装置による眼の刺激が認められないことは、プロスタグランジンプロドラッグ感受性患者、随伴性眼疾患患者、及び最適な制御を必要とする重度の緑内障患者におけるその用途を示すものである。
本実施例は、眼圧(IOP)に対するビマトプロストの効果を評価するための、緑内障のビーグル犬を用いた臨床試験に関する。具体的には、本試験は、本開示の噴霧エジェクタ装置による6 μlの0.03%ビマトプロスト(Lumigan(登録商標);Allergan社(Irvine, CA USA)製)の1日1回午前中滴下の眼圧(IOP)及び瞳孔径(PD)に対する効果について、従来の点眼器による1日1回平均26μlの送達と比較して、評価する。
フロリダ大学の緑内障ビーグル犬のマッケイコロニーから、6匹の緑内障ビーグル犬(4匹の雄及び2匹の雌、3〜8歳)を、本試験に選択した。包含される条件には、専門動物眼科医の肉眼による検査によって判断される、IOPの上昇及び緑内障の症状の文書化が含まれていた。
噴霧エジェクタ装置及び従来の点眼器の結果は同様であった。6μlの用量は、従来の治療法と同様に、IOPの低下及び瞳孔の収縮において有効であった。点眼器によって投与される用量の持続性は大きかったが、噴霧エジェクタ装置による治療は、基準線よりも低いIOP値を維持していた(図23A〜23B)。噴霧エジェクタ装置を用いて処置された眼でみられた刺激は、点眼器によって処置された眼よりも顕著に少なかった。
噴霧エジェクタ装置による6μlの0.3%ビマトプロストの1日1回午前中の送達は、26μlの点眼器平均用量のおよそ1/4未満であっても、従来の治療法と同様に、IOPの制御に有効である。試験時に眼の刺激のないことが認められた。
本実施例は、標準的な点眼器と比較した、濃度を変更させた有効成分を含む低投与量の医薬組成物によって達成されるIOPの減少を評価するための、緑内障のビーグル犬を用いた臨床試験に関する。具体的には、本試験は、6匹の緑内障ビーグル犬での並行無作為化交差試験のIOP低下における噴霧エジェクタ装置によって送達される低用量高濃度ラタノプロストについての有効性を評価する。
種々のレベルの遺伝性緑内障に罹患した6匹のビーグル犬を用いた。すべての動物を、フロリダ大学で飼育されている緑内障ビーグル犬の既存コロニーから入手した。3〜9歳の4匹の雄及び2匹の雌を本試験に選択した。すべてのイヌの体重は少なくとも5kgであった。馴化及び隔離は必要ではなかった。動物を、刺青、マイクロチップ及びマークによって特定した。
噴霧エジェクタ機構(0.025%で6.0μl)と一般的なラタノプロスト(0.005%)点眼器はともに、同様の結果であった。本試験において、いずれかの時点に、眼圧(IOP)(p=0.823)間にも、瞳孔径(PD)(p=0.943)間にも有意差はなかった。IOPは、試験の初日におよそ45.4 mmHg減少し、その後の日には、24.7mmHg減少した。PDは、毎日、1時間の時点までに極小サイズ(0mm)になると予測されるように、減少した。異常又は予期しない結果はなかった。プロスタグランジン類似体によって生じる一般的な刺激があった。
要約すると、噴霧エジェクタ装置による6.0±0.6μlの0.025%ラタノプロスト送達におけるIOP及びPDの結果は、1日1回1滴(およそ26.0±10.0μl)のラタノプロストによる従来の送達と統計的に同等である。両方の処置は、毎日滴下する場合、緑内障ビーグル犬において有意にIOPを低下させる。点眼器によって送達されるラタノプロストの濃度の5倍(5X)の、噴霧エジェクタ装置によって送達されるラタノプロストは、点眼器による投与後よりも刺激を生じなかった。噴霧エジェクタ装置による等価用量の低用量高濃度処方の使用は、用量当たり低い防腐剤量が可能になり、また、咽頭への少ない流出によって全身毒性を低減することにより、治療効果を向上させることができる。
本実施例は、従来の点眼器による平均26μlの1日1回滴下と比較した、噴霧エジェクタ装置による12μlのラタノプロストの12時間間隔(q12h)滴下(BID)の眼圧(IOP)に対する効果を評価する、緑内障のビーグル犬を用いた臨床試験に関する。
種々のレベルの遺伝性緑内障に罹患した4匹のビーグル犬を用いた。すべての動物を、フロリダ大学で飼育されている緑内障ビーグル犬の既存コロニーから入手した。3〜9歳の2匹の雄及び2匹の雌を本試験に選択した。すべてのイヌの体重は少なくとも5kgであった。馴化及び隔離は必要ではなかった。動物を、刺青、マイクロチップ及びマークによって特定した。
5日間測定したところ、基準(baseline)IOPは、日平均48.4±1.16mmHgであった。基準PDは、平均6.4±0.15mmであった。午前中と午後の測定値との間に有意差はなかった。IOPの一部の傾向は、午後に低い圧力が測定され、わずかに昼行性の効果を示した。試験第7日目の0時間の時点の測定値は、基準データを反映していた。
要約すると、噴霧エジェクタ装置による0.005%ラタノプロストの送達は、BIDを滴下した場合、緑内障ビーグル犬のIOPを有意に低下させた。ラタノプロストの1日2回の滴下では、標準用量の50%未満の使用でも、日IOPの変動が少なかった。点眼器による送達と比較して、噴霧エジェクタ装置による投与は、予測される午前中IOPスパイクを防止した。
本実施例は、本開示の噴霧エジェクタ装置による9μlの0.005%ラタノプロストの送達後における眼房水(AH)中のラタノプロストの酸レベルを、標準的な点眼器による26μlの0.005%ラタノプロストの送達と比較した、緑内障のビーグル犬を用いた薬物動態臨床試験に関する。
8匹の健康な実験用ビーグル犬を本試験に選択した。包含される条件には、専門動物眼科医の肉眼による検査によって判断される、一般に全体的な健康及び正常な眼の状態が含まれていた。噴霧装置の投与前及び投与後の質量堆積較正は、9μlの意図された用量の常に10%以内であった。9.0μl用量の市販プロスタグランジンプロドラッグ、ラタノプロストを、噴霧エジェクタ装置を用いて、8匹の正常血圧のビーグル犬それぞれの2つの眼に局所的に滴下した。ラタノプロストの酸のレベルを、7時間の期間で5日間にわたって、毎日、前眼房水除去(aqueocentesis)によって得られた眼房水(AH)中で測定した。このプロトコルを、点眼器(平均用量26μl)によって送達される0.005%ラタノプロストにおいて繰り返した。
すべてのイヌに2週間の休薬期間及び馴化期間を設けた。第1週目において、すべての動物を群に割り当て、それぞれの群に2匹の動物を群1、2、4及び7に割り当てた。試験第1日目、0時間(±15分)において、噴霧エジェクタ装置により、それぞれの動物の眼に9μlの0.005%ラタノプロストを投与した。第1時間において、群1の2匹のイヌに鎮静剤を与え(Torbugesic(登録商標)0.1〜1mg/kg)、局所眼科用麻酔薬(プロパラカイン塩酸塩点眼液USP、0.5%)を与えた。局所麻酔薬の効果が現われたら、IOP及びPDを測定した。不安及び動作が示される場合、優れた結果を有する1mcg/kgデクスメデトミジンを用いて、鎮静プロトコルを変更し、研究者が試験において4つのアリコートのAHを得るのを妨げた。次いで、それぞれの眼を、0.1%ベタジン溶液で穏やかに洗浄した。それぞれの眼に、27g又は30gの針を慎重に用いて、前房から最低50〜75μlのAHを採取した。それぞれの試料を1.5mlエッペンドルフチューブに入れ、標識し、ドライアイス上でまず保存し、その後、−80℃の急速冷凍装置(ultra-freezer)に保存した。滅菌綿棒から直接圧力を加えることによって針穿刺を行なった。局所用抗生物質を毎日、最終的注意として眼に適用した。これは、第2、4及び7時間において、グループ化されたイヌのそれぞれの対で繰り返した(5日間、1日当たり16試料)。5日間で全76のAH試料を採取した。第2週目は、動物の休薬及び治癒の週として利用した。
第3週目は、0時間の時点において、点眼器による平均用量26μlの0.005%ラタノプロスト送達の代わりに、第1週のプロトコルに従った。5日間で全74のAH試料を、80回の前眼房水除去による試みで採取した。AH試料をすべて−80℃に凍結し、ドライアイス上で保存し、その後、ラタノプロストに特異的なHPLC法を用いて、ラタノプロストの酸について分析した。試験終了時に、動物を物理的に検査し、選定、又は適切であると考えられる他の利用のために準備した。
第1部
噴霧エジェクタ装置によって送達される9 μlの0.005%ラタノプロストの投与後、1、2,4及び7時間に採取したAH試料はそれぞれ、0.43±0.11μg/ml、0.54±0.10μg/ml、0.28±0.08μg/ml及び0.30±0.06μg/mlのレベルのラタノプロストの酸を有することが分かった(表2)。
点眼器によって送達される平均投与量26μlの0.005%ラタノプロストの投与後、1、2,4及び7時間に採取したAH試料はそれぞれ、0.50±0.14μg/ml、0.15±0.03μg/ml、0.28±0.09μg/ml及び0.27±0.04μg/mlのレベルのラタノプロストの酸を有することが分かった(表3)。
1時間後では、AHにおけるラタノプロストの酸レベルは、本開示の噴霧エジェクタ装置による9μlのラタノプロスト送達後よりも、点眼器による26μlのラタノプロスト送達後において高い。第2時間では、噴霧エジェクタ装置送達によってラタノプロストが投与された眼中のラタノプロストの酸レベルは、25%を超えて上昇したが、点眼器によって処置された眼のレベルは、80%を超えて減少した。第4時間及び7時間では、噴霧エジェクタ装置により送達されるラタノプロストの酸の量は、その初期レベルの50%で安定し、点眼器によって送達される酸形態のラタノプロストは、およそ40%で安定していた。IOPの減少及びPDの減少は、実施例4のコンパニオン試験での7時間の試験期間において、両方の送達方法(噴霧エジェクタ装置によって送達される9μl、及び点眼器によって送達される26μl)で同等であった。
本実施例は、エジェクタ面の帯電、及び本開示の電荷分離噴霧エジェクタ装置の流体負荷について評価する。
本実施例は、本開示の電荷分離エジェクタ機構によって達成される質量堆積について評価する。エジェクタ装置の質量堆積を測定するために、エジェクタ装置を試験装置に固定し、接地線及び陽極ワイヤ(positive wire)を演算増幅器に接続し、そして、電流プローブ及び電圧プローブをオシロスコープに接続する。周波数及び電圧は、例えば、50キロヘルツ(kHz)の周波数において、ピーク(90Vpp)正弦波に対して90Vピークに設定し、エジェクタ装置からの噴霧は、1ミリグラム(mg)の感度を有するスケールを用いて、24 mm×60 mmのNo.1カバーガラス上で5回測定し、追跡可能な証明書を用いて、1 mgクラス1重量によって較正する。それぞれの測定において、カバーガラスをスケール上に置き、スケールをゼロにする。スライドをエジェクタ装置の噴霧経路に置き、所定時間、電圧を印加する。スライドをスケールに戻し、質量を測定し、記録する。カバーガラスを清浄化し、それぞれの測定前に、スケールを再度ゼロにする。全5つの測定を周波数ごとに記録する。過程を、所定のステップサイズ(通常1 kHz)に基づいて周波数を漸増させて繰り返す。
本実施例は、本開示の電荷分離エジェクタ機構表面の液滴駆動及び腐食について評価する。振動電場によって電荷分離エジェクタ機構の液滴駆動を検討するために、エジェクタプレート又は接地された圧電アクチュエータのいずれかを備えた図14Cに従って、エジェクタ装置を設定する。試験において、流体は、蒸留水(非導電性)、又は導電性にさせるように塩を加えた水のいずれかである。液滴形成を生じる吐出から、液滴駆動による効果を切り離すため、エジェクタ装置を、非吐出周波数である1 kHzの周波数で駆動させる。
本実施例は、噴霧エジェクタ機構によって吐出される液滴の帯電について評価する。
電位計と塩水又はその他の試験流体を吐出する装置に連結されたファラデーケージに収容されているファラデーカップを用いて、すべての測定を行なった。吐出面をすべて、ファラデーカップの上1〜2 cmに配置した。吐出方法による質量の差を考慮して、電荷測定を吐出質量に標準化した。必要に応じて、接地面をエジェクタ機構の上下に配置した。
図示されているように、本開示の態様によれば、液滴の誘導帯電は、制御可能かつ反復可能であり、帯電極性及び振幅は制御可能である。この点に関して、本開示のエジェクタ機構の複数の構成について評価した。
本試験の目的は、正に帯電したスライドガラスを用いて、本開示の噴霧エジェクタ装置によって吐出された液滴の表面相互作用を分析することである。本試験は、眼表面と噴霧エジェクタ装置との間の電荷差を模倣することを目的とする。
本試験において、眼の表面をシミュレートするのにスライドガラスを用いる。スライドガラスに正電荷を発生させる。DCオフセット電荷によって、噴霧エジェクタ装置に電荷を発生させる。噴霧中の電荷をファラデーケージによって測定する。
正電荷噴霧によって、液滴は、スライドガラス表面に玉のようになる傾向がある。負電荷噴霧によって、液滴は、スライドガラス表面を湿らせる傾向がある。
液滴とスライドガラス表面との間の電荷差によって、表面相互作用における挙動に顕著な相違がある。この点に関して、眼の表面は、正味の負電荷を有し、このため、正電荷噴霧は、眼表面への液体分散及び付着を促進することができる。同様に、負電荷は、電荷と同等の極性により、眼表面からの流出低減を促進することができる。
本実施例は、処置に対する液滴電荷の効果を評価するための、緑内障ビーグル犬を用いた臨床試験に関する。
緑内障ビーグル犬のインビボ薬力学(PD)試験、及び健康なイヌの薬物動態(PK)試験は、本開示の噴霧エジェクタ装置を用いて角膜上に噴霧した市販プロスタグランジンアゴニストの優れた眼圧(IOP)低下効果、及びバイオアベイラビリティの有意な増加を示している(実施例2〜15を参照)。これらの試験に用いた装置は、摩擦帯電及び誘導正電荷を有することが分かった。
本開示の噴霧エジェクタ装置による制御可能な液滴帯電によって、薬剤を改良することなく、IOP低下効果が向上し、効果の持続性が延長し、PF2aプロスタグランジンアゴニストプロドラッグのバイオアベイラビリティが増加する。

誘電体層と、を備え
第1の導電層が、前記誘電体層から前記圧電アクチュエータを分離し、前記誘電体層が、第2の導電層から前記第1の導電層を分離し、前記第2の導電層が、前記ジェネレータプレートから前記誘電体層を分離し、それによって、前記ジェネレータプレートを電荷分離し、接地する、
