Abstract
Description
miR−29模倣物のin vitro活性
機能的有効性を試験するために、第1の鎖として配列番号2を含有し、第2の鎖として配列番号1を含有するmiR−29b模倣物をマウス線維芽細胞株(NIH 3T3)にトランスフェクトし、miR−29の公知の直接標的遺伝子(van Rooijら、2008年)であるコラーゲン1a1(Col1a1)発現に対する効果を、qPCRを使用して測定した。miR−29b模倣物の量を増加させることにより、無処理の細胞、偽トランスフェクト(いかなるオリゴも用いない)細胞または非標的対照(NTC)オリゴを用いて処理した細胞と比較してCol1a1の用量依存的な減少が示された。これにより、miR−29b模倣物が機能的であることが示される。Col1a1を直接標的とするsiRNAを陽性対照として使用した(図1B)。
miR−29模倣物のin vivo分布、安定性およびクリアランス
miR−29模倣物のin vivoにおける適用性および分布を探究するために、マウスに、第1の鎖として配列番号2を含有し、第2の鎖として配列番号1を含有するmiR−29b模倣物を10mg/kg、50mg/kg、100mg/kg、または125mg/kgで静脈内注射し、4日後に屠殺した。

- miR−29アゴニストまたはmiR−29アンタゴニストを用いた処置の有効性を評価するための方法であって、前記miR−29アゴニストまたはmiR−29アンタゴニストを用いた処置の前に対象の細胞または線維性組織における1種または複数種の遺伝子の発現のレベルを決定するステップであって、前記1種または複数種の遺伝子が、miR−29によって調節される遺伝子のセットから選択されるステップと、前記miR−29アゴニストまたはmiR−29アンタゴニストを用いた処置後に前記対象の細胞/線維性組織における同じ1種または複数種の遺伝子の発現レベルを決定するステップと、前記処置の前後の前記発現レベルに基づいて、前記処置に効果があるか、効果が少ないか、または効果がないかを決定するステップとを含む方法。
- miR−29によって調節される前記1種または複数種の遺伝子が、表5から選択される、請求項36に記載の方法。
