Abstract
Description
− DNAアデニンメチラーゼをコードする遺伝子(本明細書ではdam)中の第1の機能損失型突然変異ならびに
− sifA、spvBおよびmgtCからなる群から選択される遺伝子中の第2の機能損失型突然変異
を含む腸内細菌を提供する。
− 上記の腸内細菌;および
− 担体、希釈剤、賦形剤またはアジュバント
を含むワクチンが提供される。
− dam中の機能損失型突然変異を有する腸内細菌を提供すること、
− 機能損失型突然変異を、sifA、spvBおよびmgtCからなる群から選択される細菌の遺伝子中に導入すること
を含む方法が提供される。
− sifA、spvBおよびmgtCからなる群から選択される機能損失型突然変異を有する腸内細菌を提供すること、
− 機能損失型突然変異を、細菌のdam遺伝子中に導入すること
を含む方法が提供される。
「機能損失型突然変異」は、一般に、所与の生物学的プロセスにおける遺伝子の関連機能を完全にまたは部分的に不活性化させる遺伝子の突然変異を指す。目的の特定の機能損失型突然変異は、宿主中の腸内細菌の生活環を遮断する一方、細菌の免疫学的プロファイルを破壊しないものである。
− それが必要とされる対象に:
− ある量の生弱毒化サルモネラ属(Salmonella)微生物であって、dam遺伝子中の機能損失型突然変異ならびにsifA、spvBおよびmgtCからなる群から選択される遺伝子中の少なくとも1つのさらなる機能損失型突然変異を含む微生物または
− 生弱毒化サルモネラ属(Salmonella)微生物および好適な担体または希釈剤を含むワクチン組成物であって、前記生弱毒化微生物は、dam遺伝子中の機能損失型突然変異ならびにsifA、spvBおよびmgtCからなる群から選択される遺伝子中の少なくとも1つのさらなる機能損失型突然変異を含むワクチン組成物
を投与することを含み、
− 投与される微生物またはワクチンの量は、対象における免疫応答を誘発するために十分である
方法を提供する。
1.1.細菌株および増殖条件
サルモネラ属(Salmonella)動物単離株は、異なる発生、個々の症例、または臨床検査室へのサーベイランス提出に由来した[31]。ビルレントネズミチフス菌(S.Typhimurium)UK−1を比較のために全ての試験において使用した[17]。特に記載のない限り、細菌は、ルリア・ベルターニ(LB)培地を含有する37℃において通気させた静止期培養物に由来した[18]。抗生物質は、以下の濃度において使用した:カナマイシン(Kn)、50μg/ml、アンピシリン(Ap)、50μg/ml。
ネズミチフス菌(S.Typhimurium)UK−1Δdamは、標準的な遺伝子プロトコル[20]を使用して、定義されたdam配列のインフレーム300bp欠失を導入することにより構築した(damΔ232と称する[19])。得られたネズミチフス菌(S.Typhimurium)UK−1 damΔ232株(MT3134)は、非機能的DNAアデニンメチラーゼを欠く株に毒性であるプリン類似体、2−アミノプリン(2−AP)に感受性であることが示され[21、22]、それを親サルモネラ属(Salmonella)damワクチン株として全ての試験に使用した。記載[20]の自殺ベクターpCVD442を利用して、第2のビルレンス弱毒化欠失突然変異を親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232株中に導入し、以下の標的化遺伝子中の定義されたコード配列のインフレーム欠失の構築をもたらした:dam aroA(MT3138;1056bp欠失);dam htrA(MT3142;1341bp欠失);dam mgtC(MT3146;606bp欠失);dam sifA(MT3150;807bp欠失);dam spiC(MT3154;306bp欠失);dam spvB(MT3158;1563bp欠失);およびdam ssaV(MT3162;1959bp欠失)。得られた遺伝子構築物は、欠失配列をフランキングするプライマーを使用するPCRにより確認した。
経口および腹腔内致死用量50(LD50):感染動物の50%を殺傷するために要求される用量を、経口(胃腸挿管を介して)および腹腔内(i.p.)経路を介して、少なくとも10匹のマウスを感染させることにより決定した[30、19]。サルモネラ属(Salmonella)試験株および野生型ネズミチフス菌(S.Typhimurium)参照株14028を、LB培地中で一晩増殖させた。0.2mlの0.2MのNa2HPO4 pH8.1または0.1mlの0.15MのNaCl(それぞれ経口および腹腔内投与用)中で再懸濁された細菌細胞を使用してマウスを感染させ、それを罹患率および死亡率について感染から3週間後まで毎日試験した。ネズミチフス菌(S.Typhimurium)UK−1についての経口および腹腔内LD50は、それぞれ105および<10匹の生物である[30]。6から8週齢のBALB/cマウスを全てのビルレンス試験において使用した。保護アッセイ。マウスを、109CFUの用量におけるネズミチフス菌(S.Typhimurium)damワクチン株により経口免疫化した[30、19]。宿主組織内のワクチン株の生残性に起因する一過的な非特異的交差保護免疫応答を回避するため[23〜25]、免疫化マウスは、ワクチン株が免疫化動物の粘膜(パイエル板;腸間膜リンパ節)および全身組織(肝臓;脾臓)からクリアランスされてから4から5週間後までビルレントサルモネラ属(Salmonella)によりチャレンジしなかった。免疫化から11週間後、マウスを、100から200倍LD50と等しい感染用量におけるビルレントサルモネラ・エンテリカ(Salmonella enterica)血清型により経口チャレンジした。マウスは、罹患率および死亡率についてチャレンジから3週間後までチャレンジ後毎日試験した。
ネズミチフス菌(S.Typhimurium)UK−1 damΔ232二重突然変異ワクチン候補のカナマイシン耐性(Knr)誘導体を構築してワクチン糞便排出を評価した。ネズミチフス菌(S.Typhimurium)株MT2057は、野生型参照株14028のKnr誘導体であり、それを本来Lac−である他のサルモネラ属(Salmonella)から区別するために使用されるKnrをコードするLac+MudJ転写融合物を含有する[19、26]。ドナー株MT2057上で増殖させたファージP22を使用してカナマイシン耐性のためにレシピエントサルモネラ属(Salmonella)damワクチン候補を形質導入し[18]、Knrネズミチフス菌(S.Typhimurium)UK−1 damΔ232二重突然変異ワクチン候補、dam mgtC(MT3183)、dam sifA(MT3184)、dam spvB(MT3186)、およびdam UK−1親株(MT3180)を生成した。ワクチン株排出。BALB/cマウスに、Knrネズミチフス菌(S.Typhimurium)UK−1 dam二重突然変異ワクチン候補を経口経路によりワクチン接種した(109CFU)。免疫化後2、4、7、11、14、および21日目に糞便を個々のマウスから回収し、カナマイシン50μg/mlのLB培地上のCFU/gのためにプレーティングした。チャレンジ株排出。BALB/cマウスに、Knrネズミチフス菌(S.Typhimurium)UK−1 dam二重突然変異ワクチン候補を経口経路によりワクチン接種した(109CFU)。免疫化から4週間後までにワクチン株糞便クリアランスが生じた。免疫化から11週間後、ワクチン接種マウスを100LD50の用量のネズミチフス菌(S.Typhimurium)UK−1のKnr誘導体(MT2315;107CFU)によりチャレンジした。免疫化後2、4、7、11、14、および21日目に糞便を個々のマウスから回収し、カナマイシン50μg/mlのLB培地上のCFU/gのためにプレーティングした。脱イオン水およびヒツジ糞便内の生残性。20mlの脱イオン水を5gの乾燥ヒツジ糞便(Barbara Byrne,University of California,Davisから寄贈;[27、28])に添加することにより、20パーセントの糞便乾燥物を調製した。脱イオン水(20ml)および20%のヒツジ糞便に、ネズミチフス菌(S.Typhimurium)UK−1 damΔ232二重突然変異ワクチン候補、dam mgtC(MT3183)、dam sifA(MT3184)、dam spvB(MT3186)、またはdam UK−1親株(MT3180)のKnr誘導体(2×105CFU)を接種した。緩いキャップを有する50mlのコニカルチューブ中で3つ組アッセイを室温において実施した。試料をボルテックスにかけ、2週間の期間にわたりCFUのためにプレーティングした。
残差(または制限付き)最尤法(REML)分析(Genstat,15th Edition,VSN International,UK,[34])を使用して連続反復測定データを分析した。単変量、反復測定モデルを、変数CFUについての時間または処理の因子にフィットさせた。ワルドカイ二乗検定を使用して有意な個々の効果およびまたは因子間の有意な相互作用を決定した。任意の非有意項目をモデルから引き、分析を反復した。以下の分析データは、予測モデルに基づく平均として提示する。予測平均は、未処理試料平均ではなくフィットモデルから得られたものである。これは、重要である。それというのも、予測平均は、共通の変数集合に調整された手段を表し、したがって、平均間の妥当な比較を可能とするためである。0.05未満のP値を、統計的に有意であるとみなした。解剖における組織中に存在するCFUの数は、分散分析(ANOVA,Genstat,15th Edition,VSN International,UK)を使用して分析した。REMLおよびANOVAを使用して計算された個々の平均間の差は、近似最小有意差(LSD)を計算することにより決定した。計算LSDを超過した平均の差は、有意とみなした。
全ての動物実験は、実験動物の飼育およびケアのためのNational Institutes of Health指針に従って行い、University of California,Santa BarbaraにおけるInstitutional Animal Care and Use Committeeによる関連調査および承認後に協会の規程に従って実施した。
2.1.第2のビルレンス弱毒化突然変異を含有するサルモネラ属(Salmonella)damワクチン候補の構築
改変生ワクチンの商業的成功は、治療指数、安全性/毒性の比に依存的であり、したがって、第2のビルレンス弱毒化突然変異をネズミチフス菌(S.enterica serovar Typhimurium)damワクチン中に導入してワクチン安全性を改善した。親株UK−1の抗生物質感受性のdam欠失誘導体を構築して他の微生物株への抗生物質耐性の潜在的な伝達を排除した(材料および方法)。得られたネズミチフス菌(S.Typhimurium)UK−1 damΔ232(MT3134)を、全ての試験に親ワクチンバックグラウンドとして使用した。続いて、第2のビルレンス弱毒化突然変異をネズミチフス菌(S.Typhimurium)UK−1 damΔ232中に導入してワクチン安全性を改善した(材料および方法)。これらの突然変異は、細胞内および/または全身生存に関与する遺伝子、例として、aroA(アミノ酸生合成);htrA(ストレス応答);mgtC(マグネシウム輸送);sifA、spiC、ssaV(サルモネラ属病原性遺伝子島(Salmonella Pathogenicity Island)−2(SPI−2);およびspvB(細胞毒素産生)に標的化させた。続いて、得られたサルモネラ属(Salmonella)dam二重突然変異ワクチン候補、dam aroA、dam htrA、dam mgtC、dam sifA、dam spiC、dam spvB、dam ssaVを、親サルモネラ属(Salmonella)damワクチン株と比較して改善された安全性/効力について評価した。
第2のビルレンス弱毒化突然変異を改変生ワクチン中に導入することの主な懸念は、ワクチンクリアランスの加速の結果としての抗原曝露の低減に起因する効力の損失の潜在性である。したがって、ネズミチフス菌(S.Typhimurium)UK−1 damΔ232二重突然変異ワクチン候補、dam aroA、dam htrA、dam mgtC、dam sifA、dam spiC、dam spvB、dam ssaVを、親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232ワクチン株において見出されたものと類似するコロニー化および生残性パラメータを維持したものについて試験した。BALB/cマウスにサルモネラ属(Salmonella)dam二重突然変異ワクチン候補を経口感染させ(109CFU)、ワクチン株のコロニー化/生残性を粘膜(パイエル板;腸間膜リンパ節)および全身組織(肝臓および脾臓)中で感染から2および4週間後において評価した(図1)。サルモネラ属(Salmonella)dam二重突然変異ワクチン候補を2つの群に分類した、クラスI:親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232単一突然変異ワクチン株に対して類似するコロニー化および生残性を示したもの(dam mgtC;dam sifA;dam spvB);ならびにクラスII:親サルモネラ属(Salmonella)damワクチンにより示されるものに対するコロニー化および生残性を示したもの(dam aroA、dam htrA、dam spiC、dam ssaV)。これらのデータは、クラスIワクチン候補ワクチンが宿主組織中で軽度の生残性を持続した一方、クラスIIワクチンはワクチン接種動物中で急速なクリアランスを示したことを示す。
サルモネラ属(Salmonella)dam二重突然変異ワクチン候補(クラスIおよびクラスII)を試験して親サルモネラ属(Salmonella)damワクチンにより誘発されるものと類似する保護免疫応答を付与するために軽度の生残性が必要であるか否かを区別した。BALB/cマウスを、7つのサルモネラ属(Salmonella)dam二重突然変異ワクチン候補のそれぞれにより経口免疫化した(109CFU)。宿主組織内のワクチン株の生残性に起因する一過的な非特異的交差保護免疫応答を回避するため[23〜25]、免疫化マウスは、ワクチン株が免疫化動物の粘膜(パイエル板;腸間膜リンパ節)および全身組織(肝臓;脾臓)からクリアランスされてから4から5週間後までビルレントサルモネラ属(Salmonella)によりチャレンジしなかった。免疫化から11週間後、マウスを、200倍LD50感染用量のビルレント親株ネズミチフス菌(S.Typhimurium)UK−1により経口チャレンジした。全て(3つのうち3つ)のクラスIワクチン候補(dam mgtC、dam sifA、dam spvB)により免疫化されたマウスは、ビルレント同種チャレンジに対するロバストな保護を示し、これは親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232株により示されたものと類似する(図2)。逆に、クリアランスの加速を示したクラスIIワクチン候補(dam aroA、dam htrA、dam spiC、dam ssaV)は1つも(4つのうち0)、親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232株と比較してビルレント同種チャレンジに対する有意な保護を付与しなかった(**P<0.01、***P<0.001)。
増大したビルレンスへの復帰は、全ての改変生ワクチンについての懸念事項である。サルモネラ属(Salmonella)dam突然変異ワクチンは、メチル指向ミスマッチ修復遺伝子中の突然変異の獲得を介する腹腔内(経口でない)感染後のより高いビルレント状態への復帰を受ける能力を有する[29]。このような復帰は、Dam機能を欠く細菌に毒性であるプリン類似体2−アミノプリン(2−AP)を使用して評価することができる[21]。すなわち、親dam株(2−APS)を、全身組織中の2−APr(増大したビルレンスの潜在的指標として)への復帰について評価することができる[29]。BALB/cマウスに、サルモネラ属(Salmonella)dam二重突然変異ワクチン候補、dam mgtC、dam sifA、dam spvB.または親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232株を腹腔内感染させた(103CFU)。感染から5日後、細菌を肝臓および脾臓から回収し、2−APs(生残性)および2−APr表現型への復帰について評価した(図4)。3つ全てのワクチン候補(dam mgtC、dam sifA、dam spvB)が、脾臓/肝臓において親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232株のものに対して有意に低減したコロニー化および生残性(2−APs)ならびに低減した2−AP耐性への復帰を示した(*P<0.05)。
ワクチン接種動物における低減したワクチンおよびチャレンジ株排出は、ワクチン安全性に望ましい形質である。ネズミチフス菌(S.Typhimurium)UK−1 damΔ232二重突然変異ワクチン候補のカナマイシン耐性誘導体を構築し、使用して免疫化動物の糞便中のワクチン株およびチャレンジ株排出を評価した。BALB/cマウスを、サルモネラ属(Salmonella)dam二重突然変異ワクチン候補(dam mgtC[MT3183];dam sifA[MT3184];dam spvB[MT3186])またはdam UK−1親株(MT3180)のいずれかにより経口経路により免疫化した(109CFU)。ワクチン株排出。感染後2、4、7、11、14、および21日目に糞便ペレットを得、Knr細菌について評価した。全てのサルモネラ属(Salmonella)dam二重欠失ワクチン候補は、親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232株のものと比較して有意に低減したワクチン株糞便排出を示した(図5;P<0.05)。チャレンジ株排出。免疫化から11週間後、ワクチン接種マウスを、100LD50の用量のネズミチフス菌(S.Typhimurium)UK−1のKnr誘導体(MT2315;107CFU)によりチャレンジした。感染後2、4、7、11、14、および21日目に糞便ペレットを得、Knr細菌について評価した。サルモネラ属(Salmonella)dam mgtC、dam sifAおよびdam spvB株は、3週間の期間にわたり親ネズミチフス菌(S.Typhimurium)UK−1 damΔ232ワクチンのものに対してチャレンジ株排出の有意な低減を示し、dam sifAおよびdam spvB株は、4から21日目に低減した排出を示した(図6;P<0.05)。これらのデータは、サルモネラ属(Salmonella)dam二重突然変異ワクチンによるワクチン接種が、親サルモネラ属(Salmonella)damワクチンのものに対して少ないワクチン糞便排出をもたらし、二重欠失ワクチン接種がdam親ワクチンよりもロバストな、ビルレントチャレンジ後の野生型サルモネラ菌排出の弱毒化を提供することを示す。
サルモネラ属(Salmonella)dam二重突然変異ワクチン候補を、脱イオン水中およびヒツジ糞便中の環境生残性について評価した。脱イオン水。脱イオン水に、サルモネラ属(Salmonella)dam二重突然変異ワクチン候補(dam mgtC[MT3183];dam sifA[MT3184];dam spvB[MT3186])またはdam UK−1親株(MT3180)のいずれかのKnr誘導体を接種した(104CFU/ml)(図7)。水試料を2週間の期間にわたりCFU/gのためにプレーティングした。全て(3つのうちの3つ)のサルモネラ属(Salmonella)dam二重突然変異ワクチン候補および親dam株は、野生型UK−1株のものと比較して有意に低減した2週間のインキュベーションにわたる脱イオン水中の生存能を示した(図7;P<0.05)。さらに、水中の低レベルワクチン生残性は、容器水ワクチン投与に適合し得る。ヒツジ糞便。20パーセントの乾燥物ヒツジ糞便に、サルモネラ属(Salmonella)dam二重突然変異ワクチン候補(dam mgtC[MT3183];dam sifA[MT3184];dam spvB[MT3186])またはdam UK−1親株(MT3180)のいずれかのKnr誘導体を接種した(104CFU/ml)。糞便試料を2週間の期間にわたりCFU/gのためにプレーティングした。全て(3つのうちの3つ)のサルモネラ属(Salmonella)dam二重突然変異ワクチン候補および親dam株は、2週間のインキュベーション期間にわたる野生型UK−1株のものと比較して有意に低減した2週間のインキュベーションにわたるヒツジ糞便中の生存能を示した(図8;P<0.05)。さらに、サルモネラ属(Salmonella)dam sifAは、他の3つのワクチン株試験のものに対して有意に低減したヒツジ糞便中の生存能力を示した(P<0.05)。これらのデータは、サルモネラ属(Salmonella)ワクチン候補が、野生型UK−1株のものと比較して脱イオン水およびヒツジ糞便中の両方で低減した環境生残性を示すことを示す。
良好な畜産実務にかかわらず、サルモネラ症は、糞口伝達を支持する集約的な生産系において重要な問題であり続ける。疾患は、主として、増加した病原体曝露および疾患感受性により引き起こされる。環境条件の変動は、環境病原体負荷および続いて宿主チャレンジの変化を引き起こす。妊娠および出産に伴う生理学的変化は、非ワクチン接種免疫状態の新生児と同様に疾患に対する感受性を増加させる。管理実務も、農場で(寄せ集め、囲い入れ、輸送前の食料および水制限)、輸送の間(食料および水制限、環境ストレス)および販売場(混じり合い、病原体曝露)家畜が受ける蓄積的なストレス因子により宿主免疫に悪影響を与え得る。

Claims (17)
- サルモネラ属(Salmonella)微生物であって、dam遺伝子中の機能損失型突然変異ならびにsifA、spvBおよびmgtCからなる群から選択される遺伝子中の少なくとも1つのさらなる機能損失型突然変異を含む微生物。
- 前記機能損失型突然変異が、前記遺伝子中の1つ以上のヌクレオチドの挿入、欠失および/または置換からなる群から選択される、請求項1に記載のサルモネラ属(Salmonella)微生物。
- ネズミチフス菌(S.Typhimurium)、ゲルトネル菌(S.Enteritidis)、サルモネラ・ダブリン(S.Dublin)、サルモネラ・ニューポート(S.Newport)、ブタコレラ菌(S.Choleraesuis)、およびサルモネラ・ボビスモルビフィカンス(S.Bovismorbificans)からなる群から選択されるサルモネラ・エンテリカ(Salmonella enterica)亜種エンテリカ(Enterica)血清型である、請求項1または2に記載のサルモネラ属(Salmonella)微生物。
- ネズミチフス菌(S.Typhimurium)である、請求項1〜3のいずれか一項に記載のサルモネラ属(Salmonella)微生物。
- 前記少なくとも1つのさらなる機能損失型突然変異が、sifA遺伝子中に存在する、請求項1〜4のいずれか一項に記載のサルモネラ属(Salmonella)微生物。
- サルモネラ属(Salmonella)微生物に対する対象における免疫応答を誘導するための組成物であって、前記対象における免疫応答を誘発するために十分な量の請求項1〜5のいずれか一項に記載の微生物およびアジュバント、希釈剤、担体または賦形剤を含む組成物。
- 請求項1〜5のいずれか一項に記載のサルモネラ属(Salmonella)微生物の精製培養物。
- 凍結乾燥、凍結、または再構成される、請求項7に記載の精製培養物。
- ビルレントサルモネラ属(Salmonella)微生物による対象の感染の可能性を予防または低減させる方法であって、
− それが必要とされる対象に、ある量の請求項1〜5のいずれか一項に記載の微生物、請求項6に記載の組成物または請求項7もしくは8に記載の精製培養物を投与すること;

− 投与される微生物、組成物または精製培養物の前記量は、前記対象における免疫応答を誘発するために十分である

- 前記医薬品が、ワクチンである、請求項14または15に記載の使用。
- ビルレントサルモネラ属(Salmonella)微生物による対象の感染の可能性の予防または低減において使用される、請求項1〜5のいずれか一項に記載のサルモネラ属(Salmonella)微生物。
