Abstract
Description
(1)小さいサイズの粒子は、製造および貯蔵の間に粒子自体に静電荷を発生させることがある。これにより、粒子がそれ自体もしくは肺グレードのラクトースなどの製剤添加剤に対して塊状化また凝集し、結果として、5ミクロン超の実効サイズを有する粒子の塊が生じる可能性がある。その場合、こうした大きい塊が肺の奥まで到達する確率は低くなる。このため、パッケージ化された薬物のうち、患者が吸収に利用できる割合が低くなる。
(2)患者に送達する必要がある活性薬物の量は、数十マイクログラム程度、またはそれより少ない場合がある。たとえば、喘息に使用される薬物であるアルブテロールの場合、これは通常25〜50マイクログラムである。現在の製造装置は、ミリグラムの用量範囲内の一定分量の薬物を、許容可能な精度で効果的に提供することができる。したがって、標準的な実施は、活性薬物をラクトース等の賦形剤または充填剤と混合することである。またこの添加物は、薬物を「流れやすく」する。この賦形剤は、これらの粒子に薬物粒子が静電的または化学的な結合によって固着するため、キャリアとも呼ばれる。こうしたキャリア粒子のサイズは、薬物粒子よりもずっと大きい。乾燥粉末吸入器がキャリアから薬物を分離する能力は、設計の有効性における重要な性能パラメータである。
(3)製薬業界では一般的に、5ミクロン超のサイズを有する活性薬物粒子は、口または咽頭の中に堆積すると考えられている。こうした場所における薬物の生物学的利用能および吸収は肺とは異なるため、これによって別のレベルの不確実性がもたらされる。乾燥粉末吸入器は、こうした場所に堆積する薬物を最小限に抑え、薬物の生物学的利用能に関連付けられる不確実性を低減する必要がある。
・ 先行技術の吸入器の性能は、使用者が生じさせる流量に依存する。流量が小さいと粉末が完全に分解されず、したがって患者に送達される用量に悪影響を及ぼす。
・ 分解プロセスに一貫性が欠如しているため、用量間の薬物の生物学的利用能に一貫性がない。
・ 電気機械ベースの吸入器を駆動するには大きいエネルギーが必要であるため、デバイスはサイズが大きくなり、携帯使用に不向きになる。
・ 開放されたまたは先端が切り取られたカプセルからの薬剤の損失。
・ 酸素または湿気に曝されることによる、開放されたまたは先端が切り取られたカプセル内の薬剤の劣化。

Claims (17)
- 薬を保持するための投薬チャンバと、前記投薬チャンバと相互作用して前記薬をエアロゾル化するためのバイブレータとを含む吸入デバイスであって、
バイブレータの面と前記投薬チャンバの壁との間のスペーサであって、前記バイブレータの前記面と前記投薬チャンバの前記壁との間に空気隙間が作られるようなスペーサを含むデバイス。 - 前記バイブレータは圧電トランスデューサ組立体を含む、請求項1に記載のデバイス。
- 前記バイブレータの前記面の上に隔離碍子のスリーブをさらに含む、請求項1に記載のデバイス。
