Abstract
Description
ある種の態様は、細胞におけるpHの操作のための光活性化化合物を対象とする。ある種の局面において、光活性化化合物は2-ニトロベンズアルデヒド(NBA)である。NBAを用いて、標的細胞におけるアポトーシス、つまり標的化アポトーシスに基づく治療法を誘導することができる。標的化アポトーシス治療法は、(i) 生体系における光活性化化合物(例えば、NBA)の拡散および毒性、(ii) 局所アシドーシスの誘導、(iii) 突発性アシドーシスに応じた細胞の損傷および死の誘導、ならびに(iv) 生体系の細胞または組織の標的化を提供する。
大部分のがんは、ワールブルク効果として知られる好気的解糖を介した乳酸の過剰な生成および排出により特徴付けることができる。過剰生成の細胞内酸は、がん細胞により効果的に排出され、酸性の細胞外環境を引き起こし、これは転移能に直接関連付けられている(McCarty and Whitaker, Alternative Medicine Review, 2010, 15(3):264-72)。細胞外アシドーシスは、腫瘍の部位で、血管形成、または血管新生の速度増大、および本来アルカリ性である大部分の化学療法薬の効果減少を引き起こす。この酸性環境において生存するために、がん細胞は、プロトンを排出し、弱アルカリ性の細胞内pH (pHi)を維持する、プロトン交換輸送体、最も注目すべきは、ナトリウム/水素交換体1 (NHE1)を発現または上方制御する。
光線力学的治療(PDT)は、多くの種類のがんを処置するための最も急速に成長している様式のうちの1つである。この技術はその初期の段階にあり、現在、外科手術または化学療法のような、より効果的な処置の必要性をなくすのに十分なほど効果的ではない。PDTは、手術後の内膜過形成(Lamuraglia et al., Journal of Vascular Surgery, 1995, 21(6):882-90)、乾癬(Almutawa et al., Photodermatol Photoimmunol Photomed. 2013)、およびポリープ状脈絡膜血管症(Sliwinska et al., Prog Retin Eye Res. 2013, 37: 182-99)を抑えるために、静脈撮影装置(vein graphs)のような、他の疾患のための処置として利用されてきた。しかしながら、PDTに対しての最も有望な用途は、腫瘍学におけるその使用である。インビトロでおよびインビボでがん細胞に適用されたPDTは、60%もの高さの細胞死を示した(Idris et al., Nature Medicine, 2012, 18(10): 1580-85)。現在、最も効果的なPDT使用のうちの1つでは、がん細胞治療に向けて活性酸素種(ROS)の細胞内放出を引き起こすためにナノ粒子の光活性化を採用している(Idris et al., Nature Medicine, 2012, 18(10): 1580-85)。
局所的な形で特定の細胞を効果的に標的化する能力は、有望な治療送達機構である。がん細胞は、治療用物質の局所送達のために標的化されうる種々の固有の特徴を示しうる。特定のがん細胞を標的化するためにデザインされた送達機構は、通常は化学療法処置に関連している中毒性副作用を低減するための能力を与える(Gabizon et al., Cancer Chemother Pharmacol. 2010, 66(1):43-52)。考えられる多様ながん標的には、ヒト上皮増殖因子受容体2 (Emde et al., Critical reviews in oncology/hematology 2012, 84:49-57; Colombo et al., Pharmacol Res 2010, 62(2): 150-65)、葉酸受容体(Salazar et al., Cancer metastasis reviews, 2007, 26: 141-52)、およびエストロゲン受容体(Kleinsmith et al., Principles of Cancer Biology. ナトリウム水素交換体1 (NHE1)は、遺伝的に高度に保存されている遍在的に発現されるイオン輸送体である。NHE1は、細胞外Na+の細胞内H+への交換を容易にし、緊張低下を促進し、細胞容積を増加させる。NHE1は高度に可変性の細胞内C末端により特徴付けられ、これを調節して、接着、形態、遊走および増殖を含む細胞挙動を媒介することができる(Putney et al., Annual Reviews. 2002, (42):527-52)。NHE1は細胞生存に極めて重要な輸送体である。
ホスファチジルセリン(PS)は、細胞のリン脂質二重層の内部小葉に通常は隔離されている膜リン脂質である。細胞膜の外表面上のPSの発現には2つの原因、つまり、アデノシン三リン酸依存性アミノリン脂質トランスロカーゼによる膜非対称性維持の欠如、およびいずれかの膜表面へPSを素早くはじく脂質スクランブラーゼの活性化がある(Verhoven et al., Journal of Experimental Medicine, 1995, 182(5): 1597-601)。
TP53は、複雑な、しかし一般的な、経路に関与する多面的遺伝子である。P53の調節ネットワークは、神経変性、アテローム性動脈硬化症およびがんのような、病的状態におけるその最も一般的な発生について研究されている(Amaral et al., Discovery Medicine, 2010, 9(45): 145-52)。最も一般的に見られるがん原因は、TP53遺伝子の損傷または欠失によるものである(Olivier et al., Cold Spring Harbor Perspectives in Biology, 2010, 2: 1-17)。この遺伝子に対する変異は、今日見られているがんの50%超の原因である(Kleinsmith, 「Cancer screening, diagnosis, and treatment」, Principles of Cancer Biology. Pearson Education, Inc., 2006, 218-24中)。
MCF-7細胞が本発明者らの固有の処置に導入される場合、早ければ処置から3分で細胞容積の喪失が観察される。細胞容積の減少がアポトーシスの最初の段階で観察されており(Orlov et al., Am J Physiol Cell Physiol. 2013, 305(4): C361-372)、透水性チャネルの活性化の増強に起因していた(Remillard et al., Am J Physiol Lung Cell Mol Physiol. 2004, 286(1):L49-67)。加えて、本発明者らの新規の処置の間にH+がアンケージされる場合、NBAは2-ニトロソ安息香酸への光化学的転位を起こす(Diaspro et al., Q Rev Biophys. 2005, 38(2):97-166)。細胞内H+緩衝成分の減少と組み合わせてH+の増加も、細胞容積の減少に寄与するであろう(Fraser et al., Journal of Physiology, 2005, 563(3):745-64)。この細胞内撥水は、多くの細胞内調節タンパク質を応答させ、水分喪失に対抗するための機構を活性化させる。水分喪失とその後の細胞容積の減少により急速に活性化される調節経路の1つは、JAK/STAT経路、具体的にはヤヌス(Janus)キナーゼII (Jak2)である。
細菌の抗生物質耐性は、健康管理に対しての進行中の課題を提示する。耐性の大部分は、細菌が、その標的に影響を与える前に抗生物質を分解することを可能にする酵素的適応によるものである(Benveniste et al., Annual Reviews, 1973, 42:471-506)。この酵素作用が発現されると、それは形質転換を通じある生物から別の生物へ素早く伝えられ、薬物の有効性を無効化しうる。他の耐性方法には、テトラサイクリンおよびスルホンアミドで見られる、抗生物質の取込みを妨害する細菌の能力、またはリボソームを適応してエリスロマイシンに対する耐性を与える黄色ブドウ球菌(Staphylococcus aureus)で見られる標的の変化が含まれる(Davies and Davies, Microbiology and Molecular Biology Reviews, 2010, 74(3):417-33)。抗生物質における単純な、単一標的かつ単一構造の進歩では、耐性を示すこれらの生物において同様に単純な防御の適応が実施される必要がある。
代謝障害は何世紀にもわたって社会を悩ませてきた。最近の科学の進歩は、過度に活性な酵素がこれらの疾患の多くの原因の1つであることを明らかにしている; しかし、代謝障害の病態生理はほとんど理解されていない(Mairet-Coello et al., Neuron, 2013, 78(1):94-108)。アルツハイマー病は、米国で5百万を超える人々に影響を与えている、最も多く見られる認知症の形態の1つであり、細胞酵素の活性亢進によって引き起こされる(Mairet-Coello et al., Neuron, 2013, 78(1):94-108)。インビボでの研究から、AMP活性化キナーゼ(AMPK)による微小管結合タンパク質Tauの過剰リン酸化は、興奮性シナプス伝達に用いられる神経スパインの喪失につながることが明らかにされている(Mairet-Coello et al., Neuron, 2013, 78(1):94-108)。このキナーゼの活性を、薬理学的にまたは遺伝子欠失により、低減させることで、神経スパインがシナプス毒性のあるTau過剰リン酸化に供されず、海馬ニューロンに防護効果を及ぼすことが明らかにされた(Mairet-Coello et al., Neuron, 2013, 78(1):94-108)。
幹細胞は高度に研究された生物学の分野である; しかしながら、幹細胞を作出するためのプロセスは、脱分化した生細胞をほとんど生じさせない。現行の技法は、最終分化した細胞を胚の状態へ戻すか、または脱分化させることにより再プログラム化することを目標としている。体内の多くの細胞は、制御された分裂状態にあり、皮膚細胞および骨髄細胞のような、細胞置換または細胞分裂の一定した供給源を必要とする身体の領域に一般的に位置している。Gaoら(Cellular Physiology and Biochemistry, 2014, 33(1): 185-94)は、ヒト由来間葉臍帯細胞(hUC-MSC)がpHiだけの持続的低下に応答して骨芽細胞へ分化することを最近になって報告した。Gaoら(Cellular Physiology and Biochemistry, 2014, 33(1): 185-94)は、NHE1をカリポリドで同時に遮断しながら、hUC-MSCを塩化アンモニウムプレパルス技法に曝露することによってpHiの低下を誘導しながらpHiを光学的に測定した。本明細書において記述される方法は、より長い、持続的間隔の間にpHiのみの正確な、漸進的減少の誘導を可能にして特定の幹細胞株からの分化を達成する(Gao et al., Cellular Physiology and Biochemistry, 2014, 33(1): 185-94)。
タンパク質構造および機能は、遊離H+によるイオン化で改変されうる。いくつかの研究から、中枢神経系シグナル伝達に関与するタンパク質に及ぼすpHの重要な役割が支持されている。Wemmieら(Neuron, 2002, 34(3):463-77)により海馬ニューロンにおいて記述されている酸活性化電流は、ヌルマウスでのこれらの酸感受性イオンチャネル(ASIC)の喪失が海馬長期増強の障害ならびに欠陥のある瞬目反射条件付けおよび空間学習を生じたことから、長期増強に必要であった。ASIC 1aは、海馬ニューロンの樹状突起棘上のタンパク質受容体として働き、細胞内カルシウムシグナル伝達に影響を与える(Zha et al., Proc Natl Acad Sci USA, 2006, 103(44): 16556-61)。細胞外遊離H+の濃度の増加は、多くのリガンド開口型イオンチャネルに影響を及ぼす; アセチルコリンおよびNMDA受容体は酸性の細胞外pHにより阻害され、アルカリ性の細胞外pHにより阻害される(Del Castillo et al., 1962, J. Cell Comp Physiol 59:35-44; Giffard et al., Brain Res, 1990, 506:339-42; Palma et al., 1991, J Membr Biol, 120:67-73; Traynelis and Cull-Candy, 1990, Nature, 345:347-50)。GABA受容体活性は酸性の細胞外pHにより増強され、アルカリ性の細胞外pHにより阻害される(Kaila and Ransom, 1994, pH and brain function (New York: Wiley-Liss); Smart and Constanti, 1982, Proc R Soc Lond B Biol Sci,215:327-41; Takeuchi and Takeuchi, 1967, J Physiol, 1967, 191 :575-90。上記のリガンド開口型イオンチャネル、およびプロトン化によって影響を受ける実質的に全てのリガンド開口型イオンチャネルに隣接した細胞外空間において局所アシドーシスを引き起こす本発明者らの技法の能力は、神経入力および神経回路の機能的出力を変化させる固有の機構を提供するであろう。
標的化剤を、本明細書において記述される化合物または粒子に付着させて、結合体をインビボで標的領域へガイドまたは標的化することができる。インビボでの標的送達は、これらの化合物または粒子の細胞取込みを増強させて、治療効果を増強させる。ある種の局面において、抗体または細胞透過ペプチドは、腫瘍部位への標的化送達のためでありうる。
本発明との関連で用いられるリンカーは、二官能性リンカーを含むことができる。二官能性リンカーは永続性または脱離性リンカーであることができる。二官能性リンカーはアミノ酸、アミノ酸誘導体または化学的リンカーを含む。アミノ酸は天然アミノ酸および非天然アミノ酸の中の1つであることができる。天然アミノ酸の誘導体および類似体、ならびにさまざまな当技術分野で公知の非天然アミノ酸(DまたはL)、疎水性または非疎水性アミノ酸も本発明の範囲内であることが企図される。非天然アミノ酸の適当な非限定的リストには、2-アミノアジピン酸、3-アミノアジピン酸、β-アラニン、β-アミノプロピオン酸、2-アミノ酪酸、4-アミノ酪酸、ピペリジン酸、6-アミノカプロン酸、2-アミノヘプタン酸、2-アミノイソ酪酸、3-アミノイソ酪酸、2-アミノピメリン酸、2,4-ジアミノ酪酸、デスモシン、2,2-ジアミノピメリン酸、2,3-ジアミノプロピオン酸、N-エチルグリシン、N-エチルアスパラギン、3-ヒドロキシプロリン、4-ヒドロキシプロリン、イソデスモシン、アロ-イソロイシン、N-メチルグリシン、サルコシン、N-メチル-イソロイシン、6-N-メチル-リジン、N-メチルバリン、ノルバリン、ノルロイシンおよびオルニチンが含まれる。いくつかの好ましいアミノ酸残基は、グリシン、アラニン、メチオニンおよびサルコシン、より好ましくは、グリシンから選択される。
ある種の局面において、本明細書において記述される化合物または粒子は、脱離性リンカーに付着された光活性化部分を含む。光活性化部分は制御された形で放出されることができる。
ある種の局面において、SCAのような標的化部分が、永続性リンカーを通じて多官能性リンカーに付着される。永続性リンカーは標的化部分および多官能性リンカーを結合することができる。1つの好ましい永続性リンカーは、チオエーテル結合を提供しうる、マレイミジル含有分子のような分子でありうる。
ある種の態様において、組成物は以下: 薬学的に許容される希釈剤; 担体; 可溶化剤; 乳化剤; 保存剤; および/または補助剤の1つまたは複数とともに1つ、2つ、3つまたはそれ以上の治療剤を含む。そのような組成物は少なくとも1つの抗がん剤の有効量を含有しうる。したがって、医薬の薬学的組成物の調製で本明細書において提供される1つまたは複数の抗がん剤の使用も含まれる。そのような組成物は、種々のがんまたは他の疾患もしくは状態の処置において用いることができる。
材料
ダルベッコ改変イーグル培地(DMEM)、RPMI 1640 (Roswell Park Memorial Institute 1640)、ウシ胎仔血清(FBS)、ウマ血清、ゲンタマイシン、トリプシン-EDTA、カルボキシ-DCFDA (5-(および-6)-カルボキシ-2',7'-ジクロロフルオレセインジアセテート)ならびにアネキシンV Alexa Fluor 568は、Life Technologiesから購入した。NaCl、KCl、MgCl2、CaCl2、HEPESおよびグルコースは全て、Fisher Scientificから購入した。2-ニトロベンズアルデヒド(NBA)、アミロライド、ニゲリシン、NGF-7S、空気抜きキャップ付のCorning(登録商標) 75 cm2 Rectangular Canted Neck Cell Culture Flaskは全て、Sigma Aldrichから購入した。ラット褐色細胞腫(PC12)細胞およびヒト乳腺腺がん(MCF-7)細胞は、アメリカン・タイプ・カルチャー・コレクション(American Type Culture Collection)から購入した。35 mmの培養皿は、Santa Cruz Biotechnologyから購入した。ナノ粒子は、テキサス大学サンアントニオ校のBrian Yust博士およびFrancisco Pedraza博士によって合成された。細胞および細胞培養PC12細胞を、5% FBS、10%ウマ血清およびゲンタマイシン500μlを補充したRPMI 1640 (1X)中にて5% CO2および95%空気の雰囲気下、37℃で培養した。細胞を適切な範囲で35 mm培養皿にプレーティングし、80〜90%の集密まで培養した。MCF-7細胞は、5% FBSおよびゲンタマイシン500μlを補充したDMEM (1X)中にて5% CO2および95%空気の雰囲気下、37℃で培養した。細胞を75 cm2 Corning flask中にプレーティングし、適切な密度までの細胞増殖を可能とし、35 mm培養皿に継代し、その後、実験用に80〜90%の集密まで増殖させた。PC12またはMCF-7細胞を継代した際に、0.25%トリプシン-EDTA (1X) 1 mLを用いて継代用の細胞を懸濁させた。DCFDAストック溶液カルボキシ-DCFDA (5-(および-6)-カルボキシ-2',7'-ジクロロフルオレセインジアセテート; 1 mg)をジメチルスルホキシド(DMSO) 201μLに溶解して、9.4 mMのストック溶液を作出した。ストック溶液を暗所の生物学的安全キャビネット中に貯蔵して、感光性化合物を防護した。
レシオメトリックpHi測定の場合、PC12細胞にpH感受性色素カルボキシ-DCFDA (ラットaCSF中10μM)を負荷した。ニゲリシンを2.0、4.0、5.0、6.0、7.0または8.0のpHに滴定した。DCFDAの放出蛍光を、滴定したpH溶液の各々で細胞から記録した。レシオメトリック放出蛍光強度を、放出蛍光(504/530 nm)が定常状態に達するまで30秒間隔にて495 nm/440 nmの励起で観察した。個別の細胞(n = 421)からの各比率をpHごとに記録した。最良適合の曲線を作出し(R2 = 93.02)、蛍光のpHiへの変換に用いた。
NBAをメタノール500 mLに(3.0224グラム)溶解して、40 mMのストック溶液を作出した。実験ごとに、ストック75μLをかん流液中投与向けにラットaCSF 3 mLと組み合わせた。1 mM NBA-ラットaCSFの最終希釈液は、許容可能な5 mMの細胞毒性限界をはるかに下回っている(Sweitach et al., Biophys J, 2007, 92(2):641-53)。PC12細胞のNBA負荷手順は、MCF-7細胞の手順と同じものであった。
アポトーシス指標アネキシンV Alexa Fluor 568は、25 mM HEPES、140 mM NaCl、1 mM EDTA、pH 7.4に加えて0.1%ウシ血清アルブミン(BSA)を含有する溶液中で貯蔵された。ストックのアネキシンV Alexa Fluor 568 10μLを結合用緩衝液2 mL中で希釈した。結合用緩衝液は、pH 7.4で、10 mM HEPES, 140 mM NaCl, および2.5 mM CaCl2からなっていた。
アミロライド(N-アミジノ-3,5-ジアミノ-6-クロロピラジンカルボキサミド塩酸塩水和物) (266.09 mg)をDMSO 20 mLに添加して、50 mMの最終ストック溶液を作出した。このストックを、培養細胞へのかん流液中投与の前に、1 mMまでさらに希釈した。
PC12およびMCF-7細胞を実験手順中、ラットaCSF溶液(150 mM NaCl, 5 mM KCl, 1 mM MgCl2, 5 mM HEPES, 5 mMグルコース, pH 7.4) 2 mL中でかん流させた。
DCFDAストック溶液をラットaCSF溶液3 mL中で10μMの濃度まで希釈した。PC12細胞に倍率40×で、それぞれ、900/700 msの間495/440 nmにて30秒ごとにおよび倍率10×の下で各波長にて2秒ごとに光を当てた。10分後におよび蛍光を記録して定常状態をモニターする前に細胞をラットaCSFで3回洗浄した。
DCFDAストック溶液をラットaCSF溶液3 mL中で10μMの濃度まで希釈した。MCF-7細胞に倍率40×で、それぞれ、900/700 msの間495/440 nmにて30秒ごとにおよび倍率10×の下で各波長にて2秒ごとに光を当てた。蛍光の定常状態が観察された後で細胞をラットaCSFで3回洗浄したところ、がん細胞は、アルカリ性がより強いので、蛍光がその最大蛍光に達したことが示唆された。定常状態を表すために最低でも10分間、pHiの光学的記録をモニターした。

NBAストック溶液75μLをラットaCSF 3 mL中で混合し、10分間PC12細胞へかん流液中投与した。かん流液中投与の後、細胞をラットaCSFで最低でも3回洗浄した。さまざまなフラッシュパラダイムを用いて、PC12細胞をUV光に曝露した。各曝露後、pHを2分間モニターして、pHi酸性化を定量した。
NBAストック溶液75μLをラットaCSF 3 mL中で混合し、10分間MCF-7細胞へかん流液中投与した。かん流液中投与の後、細胞をラットaCSFで最低でも3回洗浄した。MCF-7細胞を60-30-60 UV光パラダイムに曝露した。各曝露後、pHを2分間モニターして、pHi酸性化を定量した。
NBAストック溶液75μLをラットaCSF 3 mL中で混合し、10分間MDA-MB-231細胞へかん流液中投与した。かん流液中投与の後、細胞をラットaCSFで最低でも3回洗浄した。MDA-MB-231細胞を60-30-60 UV光パラダイムに曝露した。各曝露後、pHを2分間モニターして、pHi酸性化を定量した。
細胞内アシドーシスを誘導するためのUVフラッシュパラダイムから1時間後に、細胞を軽く洗浄し、培地をホスファチジルセリン指標アネキシンV Alexa Fluor 568と交換した。この溶液は、ストック溶液10μLを用いて作出し、ラットaCSF 2 ml中で希釈した。15分、膜結合をさせ、その後、それから1時間または全ての細胞がアポトーシスを発現するまで蛍光をモニターした。
UVフラッシュパラダイムの後、小疱の存在が観察され、拡張時のDCFDA蛍光を用いて小疱内の膨張空間(intrablebular inflationary space)中のpH (pHb)の記録を取得した。DCFDA蛍光で見られた小疱は、持続的拡張を示し、サイズが低減することは決してなかった。本発明者らは、不可逆的な死プロセスとしての小疱形成について記述した既刊の所見に基づき、小疱を細胞死として定量した(Andrade et al., Biology of the Cell, 2010, 102(1):25-35)。
細胞内アシドーシスを誘導するためのUVフラッシュパラダイムから1時間後に、細胞を軽く洗浄し、培地をホスファチジルセリン指標アネキシンV Alexa Fluor 568と交換した。この溶液は、ストック溶液10μLを用いて作出し、ラットaCSF 2 ml中で希釈した。15分、膜結合をさせ、その後、それから1時間または全ての細胞がアポトーシスを発現するまで蛍光をモニターした。MCF-7細胞と同様に、本発明者らは、不可逆的な死プロセスとしての小疱形成について記述した既刊の所見に基づき、小疱を細胞死として定量した(Andrade et al., Biology of the Cell, 2010, 102(1):25-35)。
NHE1遮断薬アミロライドを、1μMから1 mMに及ぶ濃度でMCF-7細胞に、pH 7.4にてかん流液中投与した。細胞にDCFDA、NBAを負荷し、それぞれ洗浄し、UVパラダイムに曝露した後に、pHbを記録した。
さまざまな親水性生体適合性重合体、例えばポリエチレングリコール(PEG)、細胞膜介在重合体でコーティングされた、さまざまな希土類をドープしたアップコンバーティング(upconverting)コアのナノ粒子を合成し、MCF-7がん細胞に導入した。試験した2つの粒子は、KYb2F7またはNaYF4からなっていた。これらのがん細胞を、10分から1週にわたってナノ粒子により増殖培地中でインキュベートした。実験用に調製する場合、がん細胞をラットaCSFで5回洗浄し、顕微鏡下に置き、その後、先に述べたのと同じ手順の下でDCFDAを負荷した。980 nmのダイオード・レーザーの連続波を、pHiを記録している細胞の領域に合わせ、400または700 mWに設定した。10分または20分の曝露のためにレーザーシャッターを手動で制御した。
各実験における対照は、UVまたは980 nm曝露領域から10 mmまたはそれ以上離すことによって行った。pHiの記録を行って、NBAまたはナノ粒子が細胞に影響を与えたか、またはプレートの周辺部で水素を偶然にアンケージしたかを検出した。アポトーシスおよび/または小疱の検出は、アネキシンV Alexa Fluor 568およびDCFDA光学的記録を用いて行った。対照実験は以下の条件: UVのみ、時間のみ、およびNBAのみで行った。対照実験の各1つを最低でも3時間行った。
DCFDA較正
pH特異的なニゲリシン溶液に曝露された細胞の放出蛍光をDCFDA較正曲線の構築のために用いた。個々の細胞のレシオメトリック蛍光をpH 2.0、4.0、5.0、6.0、7.0または8.0で記録した。各pH単位は、pH特異的なニゲリシン溶液への曝露時にいくつかの細胞を光学的に記録し、定常状態についてモニターした、別個の実験を表す(図3a)。データをグラフに描き、R二乗値で最良適合の直線を明らかにし(y = -0.1356x2 + 2.62x - 3.6204) (R2 = 0.93)、pHiへの放出蛍光の先行き変換(future conversion)を可能にした。較正が完了した後に、各実験に適用された式から、平均の出発pHiが7.7であることが明らかにされた。
本発明者らは、NBAからのプロトン放出の前後にインビトロで個々のPC12細胞(n = 423)からのpHiを光学的に記録することにより、顕著な細胞内アシドーシスを局部的に誘導する光活性化NBAの能力を評価した。DCFDAを負荷した後に、放出蛍光比を、波長350 nmのUV光のさまざまなフラッシュ時間パラダイムに細胞を曝露する前に記録した。NBAの適用後および光分解の前のPC12細胞の平均pHi (7.55 ± 0.025)は、UV曝露によるNBAの光分解後の平均pHi (6.37 ± 0.03; P < 0.001)とは有意差があった。処置によるpHiの変化(ΔpHi)は、処置前のpHiからの変化(ΔpHi = 1.18)として規準化された。
局部酸性化がPC12細胞において達成された後で、アネキシンV Alexa Fluor 568をかん流液中投与して、アポトーシスから生じかつアポトーシスを示すホスファチジルセリン膜反転にマーキングを行った。アポトーシスを示唆しうる蛍光について、細胞をNBAの存在下でフラッシュ光分解後に少なくとも1時間モニターした。本発明者らは、平均84.0±1.33%のアポトーシスで、76〜100%に及ぶアポトーシスの範囲(n = 362; R2 = 0.98)を観察した。経時的なこれらのデータの線形回帰分析により、NBAのフラッシュ光分解からのアシドーシスに応答して2時間以内に顕著なアポトーシスが達成されることが示唆された(P < 0.01)。NBA処置による平均アポトーシス率(84.0±1.33%)は、PC12細胞をNBAの非存在下においてUV光に曝露した対照実験より有意に高かった(n = 76; R2 = -0.54; P < 0.001)。UV曝露のみに応答してアポトーシスを示す細胞の割合は、2.3〜10.3%に及び、平均6.4±1.0%の割合であった。NBAかつUV処置によるアポトーシスの割合も、細胞をNBAにもUV光にも曝露しなかった対照実験(すなわち時間のみの効果; n = 71; R2 = 0.92)よりも有意に高かった(P < 0.01)。時間のみに応答してアポトーシスを示す細胞の割合は、3.1〜4.5%に及び、平均3.8±0.4%の割合であった。NBAのみに応答してアポトーシスを示す細胞の割合は、2.1% (n = 47)であった。線形回帰分析により、UV曝露のみ、時間のみ、またはNBAのみに応答したアポトーシスの割合がNBAかつUV処置に応じたアポトーシスの割合よりも有意に低いことが示唆された(P < 0.01)。対照実験は互いに有意差がなかった(P < 0.01)。
PC12細胞におけるpHiを局部的に減少させるNBAの能力を評価した後に、同じ実験手順をMCF-7乳がん細胞に対して行った。放出蛍光比をNBAフラッシュ光分解の前後にとって、pHiをモニターした。NBA負荷後および60-30-60 UVフラッシュパラダイム前のMCF-7細胞の平均pHi (7.38±0.13; n = 76)は、NBAの存在下におけるフラッシュパラダイム後の細胞の平均pHi (6.22±0.15)と有意差があった。処置によるpHiの変化は、処置前のpHiからの変化として規準化された。処置後の平均ΔpHi (1.16 pH単位)は、処置前の平均pHiと有意差があった(P < 0.001)。
局部酸性化がMCF-7細胞において達成された後で、アネキシンV Alexa Fluor 568をかん流液中投与して、アポトーシスから生じかつアポトーシスを示すホスファチジルセリン膜反転にマーキングを行った。アポトーシスを示唆しうる蛍光について、細胞をNBAの存在下でフラッシュ光分解後に少なくとも1時間モニターした。MCF-7細胞はまた、局部細胞内酸性化により細胞小疱形成を示すことが観察され、小疱が細胞から完全に分離されるまで、または処置後の数時間(1〜6時間)後までアネキシンV Alexa Fluor 568の存在下で蛍光を示さなかった。これまでに論じられたように、研究によって、小疱形成がアポトーシスを示すことが示唆されている。小疱の数を形成時に細胞死の別指標として時間の関数として数えた。これまでの研究では、NCX1の活性化により、多くの場合には小疱のサイズの低減が認められることが示唆されている(Yi et al., 2012)が、本発明者らは、本発明者らのNBA-UV処置に応じた経時的な小疱の発生またはサイズのいずれの低減も観察しなかった。NBAのフラッシュ光分解に応答して、本発明者らは、細胞小疱形成および/またはアネキシンVの蛍光を介したアポトーシスから示されるように、94.9〜100.0% (n = 262; R2 = 0.92)に及び、経時的アポトーシスが平均98.3±0.3%の、ある範囲のアポトーシスを観察した。線形回帰分析により、2時間以上にわたってUV光のみに曝露されたMCF-7細胞に対するアポトーシスの割合(7.1%)で有意な減少(P < 0.01)が明らかにされた。NBAにのみ曝露されたMCF-7細胞において観察されたアポトーシスの割合(2.3%, n = 236, R2 = 0.87)は、NBAかつUV光で処置されたMCF-7細胞におけるアポトーシスの割合よりも有意に低かった(P < 0.0001)。時間のみに応答したアポトーシスの割合を評価するための本発明者らの対照実験(n = 20)により、2時間以上にわたってアポトーシスのないことが明らかにされた。本発明者らの結果は細胞死を全く生じなかったので、線形回帰は行われなかった。
PC12細胞およびMCF-7細胞の両方におけるpHiを局部的に減少させるNBAの能力を評価した後に、同じ実験手順を非常に侵襲性の強い、三重陰性MDA-MB-231乳がん細胞に対して行った。放出蛍光比をNBAフラッシュ光分解の前後にとって、pHiをモニターした。NBA負荷後および60-30-60 UVフラッシュパラダイム前のMDA-MB-231細胞の平均pHi (6.47±0.06; n = 38)は、NBAの存在下におけるフラッシュパラダイム後の細胞の平均pHi (2.78±0.23)と有意差があった(P < 0.001)。処置後の平均ΔpHi (3.68±0.18 pH単位)は、処置前の平均pHiと有意差があった(P < 0.001)。
局部酸性化がMDA-MB-231細胞において達成された後で、アネキシンV Alexa Fluor 568をかん流液中投与して、アポトーシスから生じかつアポトーシスを示すホスファチジルセリン膜反転にマーキングを行った。アポトーシスを示唆しうる蛍光について、細胞をNBAの存在下でフラッシュ光分解後に少なくとも1時間モニターした。MDA-MB-231細胞はまた、局部細胞内酸性化により細胞小疱形成を示すことが観察され、小疱が細胞から完全に分離されるまで、または処置後の数時間(1〜6時間)後までアネキシンV Alexa Fluor 568の存在下で蛍光を示さなかった。これまでに論じられたように、研究によって、小疱形成がアポトーシスを示すことが示唆されている。本発明者らのMCF-7の結果と同様に、MBA-MB-231細胞は、NBAのフラッシュ光分解に応答して、細胞小疱形成および/またはアネキシンVの蛍光を介したアポトーシスから示されるように、ある範囲のアポトーシスを示した(図18)。本発明者らは、NBAフラッシュ光分解後3時間未満で55.3%のMDA-MB-231細胞死を観察した。
いくつかのアップコンバージョンナノ粒子を合成し、MCF-7がん細胞へ送達されるようこれにNBAを負荷した。選択したナノ粒子は、NBA負荷用にPEGでコーティングされたKYb2F7コアからなった(図14A)。MCF-7細胞にDCFDAを負荷した後に、ナノ粒子を超音波処理し、細胞(n = 618)へ10分間かん流液中投与した。980 nmのダイオード・レーザーを次いで、顕微鏡の視野内の細胞に合わせた。pHiの記録を30分間集めて、ナノ粒子がpHに影響を与えたか、または細胞毒性であったかモニターした; 結果から、時間枠内で細胞毒性はもたらされなかった。細胞を次に、NBAへの蛍光共鳴エネルギー転移のため、ヒト細胞に対して耐容される強度をはるかに下回る、400 mWで10分間980 nmの波長に曝露した(Idris et al., Nature Medicine, 2012, 18(10): 1580-85)。MCF-7細胞を次に、それから172分にわたりpHiの変化についておよび細胞死を示す細胞小疱形成についてモニターした。記録およびアネキシンV Alexa Fluor 568適用の後、細胞死は95.1%で記録された。KYb2F7ナノ粒子の光アップコンバージョンに応答したMCF-7乳がん細胞死の割合の定量化を、図17に示す。
本発明のこの態様において、2-ニトロベンズアルデヒドを用い、系においてアシドーシスを局部的に誘導する。系、例えば細胞内空間の酸性化のためにプロトンを放出するNBAの能力を証明することが必要である。コンセプト実験の証明を行い、これによって以下が明らかにされた。
本発明のこの態様において、本発明者らは、フラッシュ光分解に曝露された細胞内NBAが局部の細胞損傷および死を引き起こすことを証明する。NBAをPC12細胞へ受動的に拡散させ、フラッシュ光分解パラダイムに曝露させ、これによって細胞内でのプロトンの局部放出を引き起こした。DCFDAを用いて、細胞内での誘導NBAプロトン放出に応答したレシオメトリック蛍光を光学的に記録した。フラッシュ光分解前からフラッシュ光分解後までの平均pHiは、1.18 pH単位だけ顕著に低減された(図8)。pHiの顕著な減少から生じる細胞死を、アネキシンV Alexa Fluor 568およびエチジウムホモ二量体III、つまりそれぞれ、アポトーシスおよびネクローシスに応答して目に見えて蛍光を発する2種の色素による蛍光標識によって定量化した。アポトーシスによる蛍光の量をアシドーシス攻撃後の時間の関数として記録した(図5)。同様に、ネクローシスによる蛍光をアシドーシス攻撃後の時間の関数として記録した(図6)。NBAのフラッシュ光分解後のアポトーシスによる平均細胞死率は、細胞をDCFDAのみ、NBAのみ、または時間のみに曝露した対照実験でのアポトーシスによる平均細胞死率と有意差があった(図7)。
NBAを用いた本発明者らの発明にかかる処置は、細胞内pH (pHi)の低下を引き起こし(図12)、これがMCF-7乳がん細胞において局部的かつ顕著な細胞死を引き起こす。このpHi変化は、細胞がNBAで処置されたらそれらがUV光に曝露される時間の量により判定され、生物学的pH範囲内の軽度アシドーシスを生じうる。また、細胞内pHが通常の生物学的範囲の中にもはやないようにアシドーシスが誘導されうる(Ravindran, Journal of Health Care for the Poor and Underserved, 2011. 22(4): 174-186)。MCF-7細胞におけるNBAの毒性および拡散を試験するために、対照実験を行った。DCFDAのみ、NBAのみ、UVフラッシュのみの毒性を試験する対照実験とともに、細胞のpHiを蛍光色素DCFDAでモニターした。その他の対照において見られた死は、培養細胞がインキュベーター中になかった時間によるものであったかどうかを判定するために、時間対照実験も行った。細胞を実験条件に曝露し、およそ2時間記録をとった後に、アポトーシスを観察するためにアネキシンV Alexa Fluor 568および光学的な小疱モニタリングを用いた。条件の1つのみに曝露された細胞において、観察されたアポトーシスは、NBAおよびUVの両方で処置された細胞において死んでいるか、またはアポトーシスを起こしている細胞の割合と比べて最低限であった(図13)。
本発明者らの発明にかかるNBA処置は、細胞内pH (pHi)の低下を引き起こし、これが非常に侵襲性の強い、三重陰性MDA-MB-231乳がん細胞において局部的かつ顕著な細胞死を引き起こす。このpHi変化は、細胞がNBAで処置されたらそれらがUV光に曝露される時間の量により判定され、生物学的pH範囲内の軽度アシドーシス、または生理学的範囲外の重度アシドーシスを生じうる。本発明者らは、3.68±0.18 pH単位のpHiの有意な減少(P < 0.001)を引き起こした、NBAとフラッシュ光分解パラダイムとに曝露された侵襲性の三重陰性乳がん55.3%においてアポトーシスおよび細胞死を誘導する能力を実証するものである。
本発明者らの記述した発明にかかる細胞処置は、細胞死を促進する新規の陽性フィードバックシステムをもたらし、しかも、この陽性フィードバック経路は好都合な機構としての機能を果たし、多剤耐性がんおよび抗生物質耐性菌などの、一般的な治療法に対して耐性を示しているいずれの細胞においても細胞死を誘導しうる。
本発明者らの記述した発明にかかる処理によって、本発明者らは、pHiのみのわずかな、漸進性の、かつ的確な減少をもたらすことが可能とされ、多能性状態へのマウス脾臓CD45+細胞の再プログラミングの百分収率とpHiの変化の大きさを関連付けることが可能とされよう。
本発明者らの発明にかかる技法は、インビボでがん性腫瘍の増殖を顕著に減少させるうえで有効である。本発明者らは、5週齢の雌性ヌードマウスの乳腺脂肪体へ侵襲性の三重陰性乳がんMDA-MB-231-GFP (細胞2×106個)を注射した。注射に続いておよそ1週後に腫瘍が出現した。腫瘍が5 mmの長さに達し、それを2日連続で維持したら、処理を開始した。腫瘍に対してaCSF 0.1 mlまたはNBA (1 mM) 0.1 ml、引き続いて光活性化の場合、マウスに対照注射のいずれかの一度限りの処理を受けさせた。NBA/光活性化処理を次のように行った: NBA注射から1時間後、腫瘍塊への200μmの光ファイバーカニューレの挿入前に動物に麻酔をかけた(1.5〜4%のイソフルラン)。腫瘍の内部を、インビトロにおいて有効であった同一の照明パラダイム、具体的には60秒照明、60秒照明なし、30秒照明、60秒照明なし、および60秒照明を用いてセラミックカニューレにより405 nmの光(80〜85 mW)で照らした。このパラダイムは「60-30-60」照明パラダイムといわれる。
記述した発明にかかる技法は、インビボでがんを有する動物において生存期間を増加させるのに有効である。本発明のこの態様において、本発明者らは、がん腫瘍においてNBAを光活性化させる技法を採用することで動物生存の顕著な増加につながることを証明する。樹立された6週齢ヌードマウス三重陰性乳がんMDA-MB-231腫瘍へNBAを注射した。がん細胞の細胞内空間へのNBAの十分な拡散を1時間可能にさせた。この1時間の拡散期間の後、腫瘍を60-30-60の励起パラダイム(既述)のために405 nmの光活性化光(85 mW)に曝露した。
Claims (13)
- (i) ニトロベンズアルデヒドを患者に投与する段階、および(ii) がん組織を光源に曝露する段階を含み、該組織の細胞により取り込まれたニトロベンズアルデヒドが水素イオンを放出し、該細胞のpHの低下およびアポトーシスの誘導を引き起こす、がんを処置するための方法。
- 光源が350nmの波長を有する、請求項1に記載の方法。
- 光源は、各フラッシュ間が2分間隔の、60秒、30秒、および60秒間の一連の3回のフラッシュに曝露されるものである、請求項1に記載の方法。
- ニトロベンズアルデヒドがナノ粒子にカップリングされる、請求項1に記載の方法。
- ナノ粒子が、がん標的化剤にカップリングされる、請求項4に記載の方法。
- ニトロベンズアルデヒドが、がん標的化剤にカップリングされる、請求項5に記載の方法。
- 第2のがん治療法を投与する段階をさらに含む、請求項1に記載の方法。
- (i) ニトロベンズアルデヒドを細胞に投与し、そこでニトロベンズアルデヒドが細胞により内在化される段階、および(ii) 細胞を光源に曝露する段階を含み、ここで細胞により内在化されたニトロベンズアルデヒドが水素イオンを放出し、該細胞のpHの低下およびアポトーシスの誘導を引き起こす、細胞のpHを低下させるための方法。
