miR−29ミミックとは、天然に存在するmiR−29と比較して、構造または配列に1つまたは複数の改変を有するが、miR−29によって調節されるmRNAのmiR−29結合部位とハイブリダイズする能力およびこのようなmRNAの翻訳を阻害するか、その分解を促進して、例えば、そのmRNAによってコードされるタンパク質の産生を阻害する能力を保持している、オリゴヌクレオチドのことである。miR−29によって調節されるmRNAとしては3型コラーゲン(Col3a1)が挙げられる。

担体分子はほかにも、標的細胞の表面に結合することが可能な標的化剤を運搬し得る。例えば、標的化剤は、標的腱細胞の表面に発現する分子と特異的に結合することが可能な特異的結合パートナーであり得る。適切な結合パートナーとしては、細胞表面分子またはこのような細胞表面分子のリガンドもしくは受容体に対する抗体などが挙げられる。腱細胞への標的化を補助し得る表面マーカーとしては、テネイシンC、CD55およびテノモジュリンが挙げられる。

miR−29オリゴヌクレオチド、ミミックおよび前駆体を標的細胞に直接送達する代わりに、miR−29オリゴヌクレオチド、そのミミックまたはいずれかの前駆体をコードする核酸を標的細胞に送達して、標的細胞内でmiR−29オリゴヌクレオチド、ミミックまたは前駆体を発現させることが可能である。このような方法は「遺伝子治療」と見なされ得る。
miR−29、ミミックおよび前駆体をコードする核酸は、任意の好都合な経路によって送達され得る。
腱は筋肉と骨とを結びつける結合組織である。筋肉の収縮により生じた力を筋肉そのものから少し離れて付着している骨格構造に伝達する1。
本発明者らは、腱細胞のmiR−29活性を増大させることにより、コラーゲンバランスを1型コラーゲン合成に傾き3型コラーゲン合成から離れるよう変化させることが可能であることを明らかにした。
治療の対象として最も一般的なのはヒトであるが、本発明の方法は、ヒト以外の霊長類(特に、ゴリラ、チンパンジーおよびオランウータンなどの大型類人猿のほかにも、旧世界ザルおよび新世界ザル)およびげっ歯類(マウスおよびラットを含む)をはじめとする一般的な実験動物、飼育動物および農業動物(特に限定されないが、ウサギ、イヌ、ネコ、ウマ、ウシ、ヒツジ、ヤギなどを含む)を含めた他のあらゆる動物に及び得る。
本明細書に記載される分子は医薬組成物に製剤化することができる。これらの組成物は、上記物質のうちの1つに加えて、薬学的に許容される補形剤、担体、緩衝剤、安定剤をはじめとする当業者に周知の材料を含み得る。このような材料は、無毒性でなければならず、また有効成分の効果に干渉するものであってはならない。担体をはじめとする材料の正確な性質は、投与経路、例えば、経口、静脈内、皮内または皮下、経鼻、筋肉内および腹腔内の各経路によって左右され得る。投与に適した組成物および方法の例については、EssekuおよびAdeyeye(2011)ならびにVan den Mooter G.(2006)に記載されている。
材料および方法
腱障害のヒトモデル
手順およびプロトコルはいずれも、ACEC番号99/101の下、倫理委員会の承認を受けたものである。肩の外科手術を受けている回旋筋腱板断裂の患者から棘上筋腱の試料15例を採取した(表1)。回旋筋腱板断裂患者の平均年齢は54歳(範囲35〜70歳)、断裂の平均の大きさは2.5cmであった。同患者からほかにも、肩甲下筋腱を採取した。術前MRIスキャン時の肩甲下筋腱障害、関節鏡検査時の肩甲下筋腱の肉眼的損傷のいずれについても臨床的に検出可能な証拠が認められない患者のみを組み入れ、この基準により、患者は厳密に前臨床コホートとなった。回旋筋腱板断裂がなく肩の安定化のための関節鏡下手術を受けている患者から採取した肩甲下筋腱の試料10例を含む独立した対照群を得た。関節鏡検査により回旋筋腱板断裂がないことを確認した。対照群の平均年齢は35歳(範囲20〜41歳)であった。
既に記載されている標準的な3点法を用いて、回旋筋腱板の関節鏡下修復術を施行した。既に記載されている通りに、回旋筋腱板断裂の横断面の大きさを推定し記録した39。関節鏡下、関節唇の側方1cmの位置にある腱の上方境界から肩甲下筋腱を採取した。外科手術による修復を実施する前、断裂部の端1.5cm以内のところから棘上筋腱を採取した。免疫組織化学染色には、組織試料を直ちに10%(v/v)ホルマリンで4〜6時間固定した後、パラフィンに包埋した。Leica−LMミクロトーム(Leica Microsystems社、ドイツ)を用いて厚さ5μmの切片を薄切し、Superfrost Ultra Plusガラススライド(Gerhard Menzel社、ドイツ)に載せた。既に確立されている方法論に従って、キシレンで組織切片からパラフィンを除去し、段階的に濃度を変化させたアルコールで脱水し、組織染色および免疫組織化学染色に用いた40。
ヒト切片をヘマトキシリン/エオシンおよびトルイジンブルーで染色し、Bonarスコアの改変版による評価で腱障害の程度を判定した41(グレード4=著明な腱障害、グレード3=進行した腱障害、2=中等度の変性、1=軽度の変性、0=正常な腱)。これには、浮腫および変性の有無とともに線維芽細胞の細胞充実性および類軟骨性化生の程度を含めた。次いで、切片を以下のマーカー:IL−33(Alexis社、マウスモノクローナル)、ST2(Sigma Aldrich社、ウサギポリクローナル)、IL−1RaCP(ProSci社、ウサギポリクローナル)、CD68(汎マクロファージ)、CD3(T細胞)、CD4(Tヘルパー細胞)、CD206(M2マクロファージ)および肥満細胞トリプターゼ(肥満細胞)(Vector Labs社)に対する抗体で染色した。
テノサイトをコラーゲン1型およびコラーゲン3型の免疫細胞化学染色について評価して、テノサイトのマトリックス産生を評価した(Abcam社)。Sircolアッセイキット(Biocolor社、キャリクファーガス、北アイルランド)を製造業者のプロトコル通りに用いて、総可溶性コラーゲン量を測定した。被験試料100μlにSircol色素試薬1mlを加え、室温で30分間かき混ぜた。10,000×gで10分間遠心分離することによりコラーゲン−色素複合体を沈殿させた、次いで、エタノール500μlで2回洗浄した。ペレットをアルカリ試薬500μlに溶かした。マイクロプレートリーダーで540nmにおける吸光度を測定した。製造業者によって提供されたコラーゲン標準品に基づき検量曲線を作成した。さらに、ELISAを用いて、ヒトおよびマウスのコラーゲン1型および3型の濃度を評価し、製造業者(USCNK Life Science社)によって供給された標準品とともにマイクロプレートリーダーで450nmにおける呈色変化を測定した。
ヒトホスホ−MAPK Array(R & D Systems Europe社、英国)を製造業者の指示通りに用いて、マイトジェン活性化プロテインキナーゼ(MAPK)、細胞外シグナル制御キナーゼ(ERK1/2)、c−JunN末端キナーゼ(JNK)およびp38アイソフォームのリン酸化状態を評価した。CalbioChem社(Merck KGaA社、ドイツ)からERK阻害剤(FR180204)を購入し、既に本発明者らの研究室で用いてオフターゲット効果に比べ最適な特異的阻害をもたらすことが既に明らかになっている濃度43、IC50=10μMで使用した。

miRNeasyキット(Qiagen社)により全RNAを単離した。cDNA調製にmiScript Reverse Transcription Kit(Qiagen社)を用いた。ヒトmiR−29a(MS00001701)、miR−29b(MS00006566)およびmiR−c(MS00009303)ならびにマウス29a(MS00003262)、29b(MS00005936)およびc(MS00001379)発現の半定量的測定にTaqMan mRNAアッセイ(Applied Biosystems社)またはmiScriptプライマーアッセイ(Qiagen社)を用いた。内在性対照としてU6B小分子核RNAまたはベータ−アクチンの発現を用いた。
標準品と比較したq−PCRにより、長い3’UTR型および短い3’UTR型の1型および3型転写産物の絶対レベルを測定した。AffinityScript(Agilent社)にランダムヘキサマーおよびオリゴ−dTプライマーの両方を用いてcDNAを作製した。以下のプライマーを用いてSYBRグリーン定量的PCRを実施し、試料はGAPDH内在性対照に対して正規化した。

Dharmacon(登録商標)DharmaFECT(登録商標)3 siRNAトランスフェクション試薬(Thermo Scientific社)を用いて、細胞に最終濃度20nMのmiR−29aおよびmiR−29bに対する合成成熟miRNAまたはネガティブ対照(Dy547で標識した線虫(C.elegans)miR−67ミミック、Thermo Scientific社)をトランスフェクトした。48時間後、トランスフェクション細胞溶解物を収集して、目的遺伝子の発現を解析した。
COL1および3ならびに可溶性ST2 3’UTRに対してオリゴをアニーリングすることによりヒト2 miRNA標的部位を作製し、pMIR−REPORTルシフェラーゼベクター(Ambion社)のルシフェラーゼ遺伝子下流にセンス方向およびアンチセンス方向にクローニングした。これらの構築物の配列を決定してインサートを確認し、pMIR−COL I/COL III/sST2−miR29a/b/cおよびpMIR(A/S)−COL I/COL III/sST2−miR29a/b/cを命名し、HEK293細胞のトランスフェクションに用いた。HEK293細胞を96ウェルプレートで培養し、pMIR−COL I/COL III/sST2−miR29a/b/c、pMIR(A/S)−COL I/COL III/sST2−miR29a/b/cまたはpMIR−REPORTのいずれか0.1μgを、ウミシイタケルシフェラーゼを含むpRL−TKベクター(Promega社)0.01μgおよび40nMのmiR−155またはスクランブルmiRNA(Thermo Scientific Dharmacon(登録商標))とともにトランスフェクトした。Effectene(Qiagen)を製造業者の指示通りに用いてトランスフェクションを実施した。トランスフェクションから24時間後、Dual−Luciferase Reporter Assay(Promega社)を用いてルシフェラーゼ活性を測定した。以下のプライマー、sST2fw 5’AGTTTAAACTGGCTTGAGAAGGCACACCGT3’およびsST2rev 5’AGTCGACGGGCCAAGAAAGGCTCCCTGG3’を用いて、ゲノムDNAからヒトsST2の3’UTRを増幅し、それぞれPmeI部位およびSalI部位を作製した。これらの部位を用いて、PCR増幅産物をpmiRGLO(Promega社)の同じ部位にクローニングした。QuickChange部位特異的変異誘発キット(Agilent社)を用いて、Targetscanで予測された2つのmiR29a MRE部位のシード領域である29−1および29−2を変異させた。Attactene(Qiagen社)を製造業者の指示通りに用いて、各ベクターをmiR29aまたはスクランブル対照ミミックをともにHEK293細胞にトランスフェクトした。24時間後、Dual−Gloルシフェラーゼアッセイ(Promega社)を用いてルシフェラーゼ活性を測定し、ウミシイタケ(Renilla)に対して正規化した。正規化したルシフェラーゼ活性を同じ構築物のスクランブル対照に対する百分率で表した。
25−Plexヒトサイトカインアッセイにより、Luminex技術を用いた25の別個のヒトサイトカインのin vitro定量測定を評価した。上清(n=3)
外科的処置の準備には、マウスにイソフラン(3%)と酸素(1%)の混合物による麻酔を実施し、両後肢を剪毛した。外科的処置の間、酸素を用いてイソフランのレベルを1%に下げ、ノーズコーンから送達して麻酔を実施した。皮膚切開後、支帯の両側に腱と平行になるよう2つの切れ目を入れ、次いで、膝蓋腱の下に直剪刀を置いた。鋏の刃を支持体として、直径0.75mmの生検パンチ(World Precision Instruments社)を用いて、右膝蓋腱の全層にわたって部分断裂を生じさせた。左膝蓋腱には偽処置を実施し、この処置は、腱の下にプラスチック製の支持体を置くだけで、傷害を一切生じさせないものであった。皮膚創傷を皮膚ステープルで閉じ、術後1日目、3日目、7日目および21日にマウスを屠殺した。マウスをCO2吸入により屠殺し、直ちに体重を測定した。BALB/c対照(CTL)およびST2−/−BALB/cの2つのグループのマウスを用いた。1時点当たりのマウスは各グループとも16匹(ST2−/−BALB/c、BALB/cそれぞれn=8)とした。上記の実験を別々に4回繰り返した。
生体力学的解析には、各グループのマウスの膝蓋腱を傷害し、3つの時点のうち1つの時点でマウス8匹を屠殺し、Linら10により既に記載されている通りに力学的試験を実施した。簡潔に述べれば、膝蓋腱を解剖して付着物を取り除き、膝蓋、膝蓋腱および脛骨のみを1つの単位として残した。腱の幅と厚さを定量化し、この2つの数値の積として断面積を計算した。特別設計の備品に入れたIsopon p38(High Build Cellulose Filler)に脛骨を包埋し、金属鉗子で適切な位置に固定した。BOSE ElectroForce(登録商標)3200試験機器に万力を用いて膝蓋を適切な位置に固定した。各腱標本を370Cの生理食塩水浴に浸漬して以下のプロトコルを実施した−0.02Nに再負荷し、0.1%/秒(0.003mm/秒s)の速度で0.02〜0.04に10サイクル前準備し、10秒間保持した。直後、腱を25%(0.75mm/秒)の速度で5%(0.015mm)の歪みまでの伸長させた後、600秒間弛緩させることにより応力緩和実験を実施した。最後に、破断するまで0.1%/秒(0.003mm/秒)の速度で力を増大させながら加えた。これらの試験から、最大応力を求め、応力−歪み曲線のほぼ直線の領域から得られる線形回帰を用いて弾性率を計算した。
150ng/ml miR−29aミミック、9μg/mlポリエチレンイミン(PEI)および5%グルコースを含有するトランスフェクション複合体を調製した。この複合体50μlを外科手術の直後にマウスの膝蓋腱に注射した。1日後および3日後にマウスを屠殺し、col1a1およびcol3a1のmRNAおよびタンパク質のレベルを測定した。蛍光標識したmiR−29aミミックを用いて、ファロイジン(細胞骨格構造を示す)および核(DAPI)に対する対比染色を用いた免疫蛍光法により、腱内のmiR−29aミミックのin vivo分布を評価した。

図の説明に示される通り、結果はいずれも平均+/−標準誤差平均(SEM)で表し、統計解析はいずれも、Graph Pad Prism 5ソフトウェアを用いてスチューデントのT検定、ANOVA検定またはマン・ホイトニーの検定のいずれかにより実施した。p値が0.05未満であれば統計的に有意であると見なした。
ヒト腱障害におけるIL−33およびST2の発現
本発明者らはまず、本発明者らが既に開発したモデル22を用いて、ヒト腱障害におけるIL−33発現を検討した。初期腱障害では、対照または断裂腱の生検に比して、IL−33、可溶性の膜結合ST2の転写産物が有意にアップレギュレートされていた(図1A〜1C)。初期腱障害の組織には、断裂腱または対象の生検に比して、IL−33およびST2の有意に高い染色がみられた(図1D)。内皮細胞、特に線維芽細胞様細胞、すなわち、初期腱障害の調節に極めて重要であると考えられるテノサイトに顕著な染色がみられた(データ不掲載)。同時に、in vitroで培養したテノサイトをTNFおよびIL−1βで刺激したところ、核IL−33の発現がmRNAレベルおよびタンパク質レベル(図1Eおよびデータ不掲載)の両方でアップレギュレートされた。これに対して、休止テノサイトおよび未刺激テノサイトにはともに、ST2の恒常的発現がみられた(データ不掲載)。
コラーゲン3型発現に傾いたマトリックス調節異常は、腱障害において修復促進の鍵となる初期表現型変化であるが、コラーゲン3型は生体力学的に劣っている。IL−33は、総コラーゲンタンパク質量の用量依存的および時間依存的上方制御を誘導した(データ不掲載)が、これは1型、特に3型コラーゲンのmRNAおよびタンパク質の発現増大によって説明される(図1F、1G)。アレイ解析を実施したところ(データ不掲載)、IL−33の下流シグナル伝達に関する報告12、16と同じく、この作用はERK阻害によって打ち消された(データ不掲載)。ほかにも、rhIL−33によってIL−6、IL−8およびMCP−1の産生が有意に増大し(データ不掲載)、この増大がNF−κB阻害によって調節されたことから、IL−33はテノサイトにおいてその標準的なIL−1Rシグナル伝達経路を介して作用することが示唆された(データ不掲載)。これに対し、他のサイトカインの産生に対しては、線維芽細胞で既に報告されているIL−33誘導性サイトカイン産生プロファイル20〜23と一致する効果は全く見られなかった。
本発明者らは、上記の観察を十分に確立されたin vivoの腱傷害モデルまで広げた。WTマウスでは、腱傷害後第1日および第3日にIL−33 mRNAの増大がみられた(図2A)。受傷ST2−/−マウスではこれが有意に減少したことから、オートクリン調節が示唆された。WTマウスでは、可溶性ST2が受傷後のいずれの時点でも未受傷対照に比して有意にアップレギュレートされた(図2B)のに対し、膜ST2 mRNAの増大がみられたのは、受傷後第3日までに限られた(データ不掲載)。受傷後第7日、第21日ともに、WTマウスにはIL−33およびST2の転写産物およびタンパク質の発現に有意な変化はみられず、ST2−/−マウスではIL−33発現に有意な変化はみられず、IL−33発現の影響が現れるのは、腱の傷害/修復における「アラーミン」型活性と同じく初期であることが示唆された。
この可能性を裏付けるため、本発明者らは、IL−33エフェクターの生物学的性質をin vivoで直接修正することを試みた。rhIL−33の投与は、コラーゲン1型の合成には影響を及ぼさなかった(図3A、3B)が、特に受傷腱におけるコラーゲン3型の合成が有意に増大した(図3D、3Eおよびデータ不掲載)。さらにWTマウスでは、rhIL−33投与により、受傷後のいずれの時点でも最終腱強度の有意な低下がみられ(図3Eおよびデータ不掲載)、このような変化が機能的影響を及ぼすことが示唆された。治癒しつつあるST2−/−マウスの腱では、IL−33の投与はコラーゲンマトリックス合成にも最終腱強度にも影響を及ぼさず、IL−33がST2依存性経路を介して作用することが裏付けられた(データ不掲載)。
IL−33がテノサイトにおけるコラーゲン1型および3型の差次的調節を駆動することが確認されたため、本発明者らは、この過程にmiRNAネットワークが何らかの機構的役割を果たしていると仮定した。これまでの研究で、miR−29ファミリーが1型および3型コラーゲンを含めた多数の細胞外トリックス遺伝子を直接標的とし24〜25、自然免疫および適応免疫の調節に関与していることが明らかにされている26。コンピュータアルゴリズムでは、miR−29がほかにもsST2を標的とし得ることが予測される。本発明者らは、ヒト腱生検および外植テノサイトにmiR−29ファミリーの全メンバーが発現し(図4A)、miR−29aが最も発現の変化が大きいことを明らかにした。テノサイト培養物では、IL−33がNFκB依存性シグナル伝達を介して作用することにより、6時間後、12時間後および24時間後にmiR−29aの発現が有意に低下する(図4B)のに対し、miR−29bおよびmiR−cに対する効果には一致がみられなかった(データ不掲載)。IL−33が仲介するコラーゲン3型マトリックスの変化がmiR−29aによって調節されたことから、本発明者らは、miR−29aの操作がin vitroでコラーゲンマトリックス合成に及ぼす機能的効果を分析した。まず、ルシフェラーゼアッセイを用いて、既に示されているように27、miR−29aがcol 1a1およびcol 3a1を直接標的とすることを確認した(図7B)。ほかにも、それまで確認されていなかったcol 1a2サブユニット転写産物との相互作用を観察した(図7)。miR−29aが実際に疾患関連細胞の標的mRNA候補のレベルを調節するかどうかを試験するため、テノサイトにmiR−29aのミミックおよびアンタゴマーをトランスフェクトした。初代テノサイトでは、miR−29a操作によりコラーゲン3型のmRNAおよびタンパク質の発現が調節されたが、コラーゲン1型のmRNAおよびタンパク質の発現は調節されなかった(図4C、4D)。さらに、miR−29aの過剰発現により、IL−33誘導によるコラーゲン3型のmRNAおよびタンパク質の合成が有意に阻害された(図4E)。さらに、miR−29aの阻害によりcol 3a1発現が有意に増大し、ヒトテノサイトにおいてmiR−29aがこれらの転写産物を能動的に調節するだけでなく、その減少が腱障害にみられる3型コラーゲン産生の増大に重要な因子であること示された。これに対し、col 1a1転写産物のレベルには変化がみられなかった(図4I)。
コンピュータ解析では、可溶性ST2がmiR−29aの標的になり得ることが明らかになり、IL−33エフェクター機能に何らかの調節的役割を果たしている可能性が示唆された。2つの有望なmiR−29abc結合部位を有することが予測されるヒトsST2の3’UTRを含むルシフェラーゼレポーター遺伝子を作製した。sST2−ルシフェラーゼレポータープラスミドとmiR−29ミミックとの共トランスフェクションにより、スクランブル対照に比してルシフェラーゼ活性の有意な低下がみられた(図7B)。さらに、sST2の両miR−29 MREのシード領域を変異させたところ、ルシフェラーゼ活性が完全に回復したことから、sST2がmiR−29aの直接の標的であることが確証された(図5A)。miR−29aが実際にテノサイトにおいて標的mRNA候補のレベルを調節するかどうかを検討するため、再びヒトテノサイトにmiR−29aのミミックおよびアンタゴマーをトランスフェクトした。miR−29aミミック/アンタゴマーのトランスフェクションによって可溶性ST2のメッセージが有意に(p<0.01)約5倍変化し(図5B)、これに対応して可溶性ST2タンパク質が有意に変化したことから、miR29aが可溶性ST2の標的であることが裏付けられた(図5C)。
最後に、本発明者らは、in vivoの腱障害モデルにおけるmiR−29a発現を検討した。WTマウスの腱傷害では、miR29aが第1日に(ベースラインに対して)22分の1に減少し、第3日までに6分の1の減少まで戻り(図5Dおよびデータ不掲載)、第7日には有意差が認められなかった。この効果はST2−/−マウスでは有意に阻害された(データ不掲載)。さらに、未受傷腱では、外来rh−IL−33の投与により、いずれの時点でもPBS注射対照に比してmiR−29a発現の減少がみられた(データ不掲載)。この効果は受傷WTマウスに最も著明にみられ、rhIL−33を加えると、miR−29aのさらに10倍の減少が仲介された(図5E)。ST2−/−マウスにrhIL−33を加えたところ、同じく受傷腱にも未受傷腱にもmiR−29a発現に対する有意な効果は認められず、miR−29aのダウンレギュレーションの一部はIL−33/ST2依存性シグナル伝達によって直接仲介されていることが示唆された。IL−33に対する中和抗体を加えたところ、受傷後第1日および第3日にmiR−29a遺伝子発現に対する傷害の効果の有意な低下がみられた(図5F)。
WTマウスの膝蓋腱にmiR−29a/PEI複合体を直接注射することにより、miR−29aミミックをテノサイトに送達した。ファロイジン(緑、細胞骨格構造に対する染色)およびDAPI(核を示す染色)で対比染色するミミック(赤)の免疫蛍光染色を用いて、miR−29aミミック注射の24時間後におけるテノサイト周辺へのミミックの局在を可視化した(不掲載)。図8に示されるように、miR−29aミミックを注射した腱では、対照群に比してコラーゲン3型のmRNAおよびタンパク質レベルの有意な低下がみられた。これに対し、コラーゲン1型のレベルには変化がみられなかった。
マイクロRNは、疾患および組織リモデリングに重要な役割を果たす多様な細胞プロセスの強力な調節因子として登場した。腱障害をモデル系として用いた上記の研究は、単一のマイクロRNA(miR−29)が複雑な初期の生物学的過程において炎症性サイトカインエフェクター機能および細胞外マトリックス調節を交差調節し、組織修復をもたらすことができることを初めて明らかにするものである。

Claims (32)
- 対象の腱治癒を調節する方法であって、腱細胞のmiR−29の発現または活性を増大させることを含む、方法。
- 腱細胞にmiR−29、そのミミックまたはいずれかの前駆体を送達する段階を含む、請求項1に記載の方法。
- 前記miR−29ミミックまたは前駆体が、1つまたは複数の改変糖残基を含む、請求項2に記載の方法。
- 前記miR−29ミミックまたは前駆体が、1つまたは複数の改変ヌクレオシド間結合を含む、請求項2または3に記載の方法。
- 前記miR−29ミミックまたは前駆体が、1つまたは複数の改変塩基を含む、請求項2〜4のいずれか1項に記載の方法。
- 前記miR−29ミミックまたは前駆体が膜通過部分を含む、請求項2〜5のいずれか1項に記載の方法。
- 前記miR−29、ミミックまたは前駆体が担体と結合している(例えば、担体と複合体を形成しているか、担体に封入されている)、請求項2〜6のいずれか1項に記載の方法。
- 前記担体が、薬学的に許容される脂質またはポリマーを含む、請求項7に記載の方法。
- 前記担体分子が、標的細胞の表面に結合することが可能な標的化剤を含む、請求項7または8に記載の方法。
- 腱細胞にmiR−29、そのミミックまたはいずれかの前駆体が発現するように、前記腱細胞に前記miR−29、ミミックまたは前駆体をコードする核酸を送達する段階を含む、請求項1に記載の方法。
- 前記核酸が担体と結合している(例えば、担体と複合体を形成しているか、担体に封入されている)、請求項10に記載の方法。
- 前記担体が、薬学的に許容される脂質またはポリマーを含む、請求項11に記載の方法。
- 前記担体分子が、前記標的細胞の表面に結合することが可能な標的化剤を含む、請求項11または12に記載の方法。
- ウイルスベクターを介して前記核酸を送達する、請求項10に記載の方法。
- 前記ウイルスベクターが、アデノウイルスベクター、アデノ随伴ウイルス(AAV)ベクター、レトロウイルスベクターまたはヘルペスウイルスベクターである、請求項14に記載の方法。
- 前記レトロウイルスベクターがレンチウイルスベクターである、請求項15に記載の方法。
- 前記miR−29が、miR−29a、miR−29b1、miR29b2もしくはmiR−29cまたはその組合せである、請求項2〜16のいずれか1項に記載の方法。
