ウシ血清アルブミン(BSA)及び12KDaのデキストランは、Sigma−Aldrich社(Rehovot、イスラエル)から購入した。エキセナチドは、Teva Pharmaceuticals社(Jerusalem、イスラエル)の好意で供与された。水中8%のグルタルアルデヒドは、Sigma−Aldrich社(Rehovot、イスラエル)から購入した。トリメタリン酸ナトリウム(STMP)は、Alfa Aesar社(Haverther chemicals and hill、マサチューセッツ州、米国)から購入した。ポリ(メタクリル酸、エチルアクリレート1:1(オイドラギット(登録商標)L100〜55)は、Rohm社(Dramstadt.GmbH、ドイツ)から得た。ヒドロキシプロピルメチルセルロース(Methocel E4M Premium)は、Dow Chemical社(Midland、ミシガン州、米国)から購入した。一塩基性リン酸ナトリウム・一水和物はMallinckrodt chemicals社(Phillipsburg、ニュージャージー州、米国)から購入した。全ての有機溶媒はHPLC規格であり、J.T.Baker社(Deventer、オランダ)から購入した。
繊細な生体高分子であるエキセナチドの保護に最も重要なことは、該ペプチドを一次BSA NPに搭載することにより達成した。2つの異なるタイプのNPを調製した:8%グルタルアルデヒドで架橋されたBSA NP、及びSTMPで架橋された12KDaのデキストランNPと組み合わされたBSA。
グルタルアルデヒドで架橋されたBSA NPを、Weberら[ref−Desolvation process and surface characterisation of protein nanoparticles]によって前に記載された確立された脱溶媒方法により調製した。200mgのBSA及び4mg又は8mgのエキセナチドを20mlの再蒸留水(DDW)に溶解した。0.5時間後に、溶液のpHを0.1MのNaOHにより8.5に調節した。次に、40mlのアセトンを、ゆっくり水性相に加えた。分散媒体における乳光の急速な形成により明らかなように、o/wエマルションが形成された。次にBSA NPを、12.5μ1のグルタルアルデヒドの8%溶液を使用して2時間かけて架橋した。架橋反応が完結した後、アセトンを、空気の層流下で蒸発させた。この剤形をGlut−1と命名した。
BSA/デキストランNPを、20mlのDDW中に、以下の化合物を溶解することにより同様に調製した:200mgのBSA、50mgの12KDaのデキストラン及び必要なときには4mg又は8mgのエキセナチド。0.5時間後に、該溶液のpHを0.1MのNaOHにより8.5に調節して、デキストラン上で隣接するヒドロキシル基がSTMP架橋剤との反応に使用できるようにした。次に、20mlのアセトンを、該水性相にゆっくり加えた。次にBSA/デキストランNPを、50mgのSTMPを使用して3時間かけて架橋させ、アセトンを上で記載したように蒸発させた。予備剤形を調製してプロセスパラメーターを変化させることにより評価した。デキストラン量が異なる2通りの剤形(50mg及び150mg)をさらなる動物実験のために選択した。50mgの剤形をDX−50と、及び150mgの剤形をDX−150と名付けた。
エキセナチドを含有するNPを噴霧乾燥技法を使用してマイクロカプセル化することにより、ミクロスフェア(MP)を形成させた。マイクロカプセル化の目的で、100mlのNaH2PO4緩衝液を調製した。緩衝液のpHは、1MのNaOH溶液により6.5に調節した。pHを6.5に保っているその溶液に、750mgの量のオイドラギットを溶解した。それに加えて、100mlの予備加熱した(約80℃)DDWに、1000mgのヒドロキシプロピルメチルセルロース(HPMC)を添加することにより、1%w/vのHPMC溶液を調製した。次に、上記オイドラギット溶液を、gazaバンド付きの漏斗を用いて(溶解しなかったオイドラギット粒子を濾過するために)HPMC溶液に加えた。
NPキャラクタリゼーション
種々のNPの平均直径及びゼータ電位を、MalvernのZetasizer(ナノシリーズ、Nanos−ZS社、英国)を25℃で使用して、水を希釈剤として使用して特徴づけた。形態学的評価を、冷凍透過電子顕微鏡(冷凍TEM)を使用して実施した。冷凍TEM方法では、1滴の該溶液を、300メッシュのCu格子(TedPella社、Redding、カリフォルニア州、米国)に支持された炭素でコートされた穴のあるポリマーフィルム上に置いて、該検体をVitrobot(FEI)を使用して液体エタン中で−170℃に急冷することにより自動的にガラス化する。該試料をFEI Tecnai 12G2 TEMを使用して、−180℃に保ったGatan冷凍ホルダーを用いて120kVで検査し、像を低速走査の冷却した電荷結合素子カメラで記録した。
噴霧乾燥されたMPの形態及びサイズの評価を、超高分解走査電子顕微鏡(モデル:Magellan 400L、FEI、ドイツ)を使用して実施した。試料をSEM用試料台上に両面粘着テープを使用して固定して、次に真空で金スパッタリング(Pilaron E5100)手順による標準的コーティングによって電気的に導電性にした。
散剤中のエキセナチドの合計量を、試料を2mlの水に溶解して終夜置くことにより分析した。その後、該混合物を14000rpmで2分間遠心分離した。上清からの1mlをHPLCに以下の条件で注入した:カラム Restek Viva C4(5μm)、250/4.6mm。カラム温度は45℃に保った。移動相Aはアセトニトリル(ACN)、及び移動相Bはリン酸によりpH2.5に調節されたリン酸二水素カリウム(KH2PO4、20mmol/L)であった。KH2PO4緩衝液を使用前に0.2μm膜フィルターを通して濾過した。以下の勾配条件をエキセナチドのために使用した:移動相をA15分で30%から45%に、及び移動相Aを30%で3分間再平衡させた。流速は1.5mL/分であった。注入体積は20μlであった。UVシグナルは215nmで検出した。エキセナチド含有率は、0から20μg/mlの間の範囲のエキセナチド濃度で構成したて相関関係を生じた(r2=0.999)較正曲線を使用して計算した。
全ての動物実験は、エルサレムのヘブライ大学における学内のEthical Committee of Laboratory Animal Careにより承認された(MD−13575〜4)。Sprague Dawley雄ラット(300〜350g)をこの検討に使用した。動物はSPF条件で収容して絶食させ、飲み水には自由に利用させた。無作為に分割した3匹のラットの7群について、経口吸収によりエキセナチドの血漿レベルを経時的に評価した。エキセナチドを溶液として又はバイエッタ(登録商標)に剤形化して65g/kgの用量(20μg/ラット)で皮下に注射した。第3及び第4群のラットに、エキセナチドを外部で加えた31mgのブランクMP又はエキセナチド溶液のいずれかで、165μg/kg(50μg/ラット)の用量で経口的に投与してブランク剤形が効果を有するか否かを決定した。最終的に、35、33及び32mgのGlut−1、DX−50及びDX−150を、それぞれ、経口的に165μg/kg(50μg/ラット)の用量で投与した。全ての経口懸濁液は、2mLのDDWに分散させたが、それに対して皮下注射の体積は200μ1であった。
薬物動態学パラメーターを、AUCの計算のための台形ルール適用するWinNonlinソフトウェアを使用して計算した。AUC値はサイズ用量補正に従って調節した。
エキセナチド剤形
剤形F−l:BSA及び架橋されデキストランによるエキセナチドの一次ナノカプセル化
200mgのBSA(Sigma−A7906)及び50mgの12KDaのデキストラン(Sigma−31418)を10mlのDDWに溶解した。4mgのエキセナチドを10mlのDDWに別に溶解した。アルブミン/デキストラン溶液をエキセナチド溶液に加えてペプチド溶解を完了させた(溶液A)。6.8のpHを0.1MのNaOHでpH約8.5になるまで調節した。20mlのアセトンを溶液−Aに強く攪拌しながら注入してエキセナチドの大部分を含むBSΑ−デキストランNPの形成を誘発した。デキストラン架橋は5%(1ml)のトリメタリン酸ナトリウム(STMP)の添加により得て、該溶液を、900rpm、室温で3時間攪拌した。剤形F−lの合成の模式的描写を図1Α〜1Bに示す。
200mgのBSA(Sigma−A7906)を、10mlのDDWに溶解した。4mgのエキセナチドを10mlのDDWに別に溶解した。アルブミン溶液をエキセナチド溶液に加えてペプチドの溶解を完了させた。pHを0.1MのNaOHでpH8.5の間になるように調節した。15mlのアセトンを、溶液−Aにボルテックスで強く攪拌しながら注入して、エキセナチドの大部分を含むBSΑ−エキセナチドNPの形成を誘発した。BSA架橋は25μ1の4%グルタルアルデヒドの添加により得て、該溶液を900rpm、室温で3時間攪拌した。剤形F−2の架橋の模式的描写を図2に示す。
エキセナチド活性化を、4mgのエキセナチドを、スペーサーのスルホ−SMCC(スルホスクシンイミジル−4−(N−マレイミドメチル)シクロヘキサン−1−カルボキシレート)と2時間室温で反応させることにより実施した。
100mgのBSA(Sigma−A7906)を2mlのDDWに溶解した。5mgのエキセナチドを3mlのDDWに別に溶解した。アルブミン溶液をエキセナチド溶液に加えてペプチド溶解を完了させた。pHを1MのNaOHで7.4〜8に調節した。10mlのアセトンを該溶液に強く攪拌しながら注入してエキセナチドの大部分を含むBSAナノ粒子の形成を誘発した。
500mgのHPMCを100mlの予め加熱されてDDWに溶解して、500mgのオイドラギット(アニオン性)を100mlのPBSに溶解した。
マウスの順化を、2〜3日の間グルコースを与えてグルコースレベルを6時間にわたってモニターすることにより実施した。その後、全てのマウスにバイエッタ(市販のエキセナチド注射用剤形)を20μg/kgの用量で注射して血中グルコースレベルを6時間にわたってモニターした。

v)エキセナチド(F−3)
vi)エキセナチド(F−4)
各マウスについて体重測定を同時に実施した。エキセナチド(F1〜F5)及びバイエッタの異なる剤形の結果を図3に示す。
血液試料を1日目から10日目まで収集した。15〜19週齢の動物を、血中グルコース測定手順前に、マウスを、清浄な巣の材料の底面にそれらの古いケージからの少量の汚れた敷きわら及び環境を加えたクリーンケージに移すことにより、終夜絶食させた(16時間まで)。ケージ及び敷きわらを変えることにより、マウスがこぼれた食物を利用する可能性を未然に防いだ。水は、全絶食期間を通して自由に利用できるままにした。食物は、最終の血液試料の収集後に元に戻した。1滴の血液を、抑制されていないマウスから尾の先端をカミソリで切開することにより取った。測定は、携帯式の.血中グルコース計(Accu−chek Aviva、Roche Diagnostics社、英国)を使用して行った。バイエッタを用いた予備実験からの血中グルコースレベルを図4に示し、一方、エキセナチド(F1〜F5)及びバイエッタの異なる剤形についての血中グルコースレベルを図5に示す。
インスリン決定のために、150mlの血液を尾部静脈から採取した(血液は、3000サイクル/分で10分間及び5分間遠心分離機で処理した)。血液パラメーター及びインスリン測定のために、血漿を2本のヘパリンコートされたチューブに分けて入れた(各々30mL)。血漿インスリンレベルを、マウスインスリン酵素結合免疫吸着剤アッセイキット(Mercodia社、Sylveniusgatan、Sweden)を使用して測定した。異なる日における異なる群の血漿インスリンレベルを図6に示す。
EDTAを抗凝血剤として使用して血漿を収集する。試料を、収集の30分以内に1000×gで15分間遠心分離した。血漿を取り出して直ちにアッセイするか又は試料を分割して−20℃又は−80℃で貯蔵する。グリコシレートヘモグロビンレベルをマウスインスリン酵素結合免疫吸着剤アッセイキット(Life Science社、フロリダ州、米国)を使用して測定した。異なる日における異なる群のグリコシレートヘモグロビンレベルを図7に示す。
ウシ血清アルブミン(BSA)は周知であり、経口薬剤送達のための豊富なタンパク質担体である(ペプチド及びタンパク質は除く)。その主要な利点は、生分解性、生適合性、安全性、非抗原性、望ましい許容性及び入手し易さである。さらに、ペプチド及びタンパク質の一次NPへの組み込みは、コーティングポリマーの大部分が水に可溶であるから難題であり、充分な量の溶解媒体を使用して試験される条件下におけるペプチドのin−vitroの長期放出を誘発するために架橋される必要がある。ナノキャリアとして広く許容されているBSA NPの場合には、問題はさらにより複雑である。グルタルアルデヒドによる架橋、熱による変性又は有機溶媒の使用を含むアルブミンの如何なる変性プロセスも、本研究においても観察されるように、ペプチド又はタンパク質の化学的完全性に明らかに影響する。
各治療剤について、3匹のラットからのエキセナチドの血漿レベルを図10に示す。
この検討における動物の取り扱い、世話、及び処置に関する全ての手順は、Association for Assessment and Accreditation of Laboratory Animal Care(AAALAC)の指針に従うInstitutional Animal Care and Use Committee(IACUC)により承認された指針に従って実施した。
エキセナチドの異なる剤形(Fl〜F5)の治療の効力及び安全性を、OB/OBマウスで評価した。特に、エキセナチド(F1〜F5)及びバイエッタを投与後の体重において有意の差はなかった(図3)。
h−インスリンを含む二重ナノカプセル化された試料を、in vivoにおけるペプチドの長期放出を誘発するための注射用乾燥散剤を得るために調製した。
200mgのヒト血清アルブミン(HSA)を5mlのDDWに攪拌下で溶解した。それとは別に、20mgのヒト又はウシインスリンを5mlのDDWに溶解して、30秒間ボルテックスで攪拌した。次にh−インスリン溶液をHSA溶液に加えて30分間攪拌し、ペプチド溶解を完了させた。生じた溶液のpHを、0.1MのNaOHで7.4〜8に調節した。次に、20mlのアセトンを速やかに(20秒以内)激しく攪拌しながら(900rpm)注入してh−インスリンを搭載したHSAナノカプセルの形成を誘発した(溶液A)。溶液Aをアセトンの蒸発を避けるためにアルミニウム箔で覆って1時間900rpmで攪拌した。試料(50〜100μ1)を、ゼータ電位及びサイズ測定のために、1時間攪拌後に抜き出した。別に、PLGA100Kの溶液(50:50)を80mlのアセトニトリルに溶解して溶液Aに加えた。
ナノカプセルを閉じたループ様式で作動する、それ故、空気の代わりにN2(g)及びCO2(g)をシステム中に流したNSDB−90で、噴霧乾燥により調製した。全ての実験において、ガス流は約120l/分であった。空気に揮発性蒸気及び湿気を加えて、乾燥及び凝縮するために除湿器に移し、次に円形通路中のシステムに乾燥して戻した。噴霧乾燥は、低温(T;n=30℃〜60℃)で4μmメッシュのサイズの膜を用いて実施した。異なるHSA/h−インスリン/PLGA比の種々の剤形を調製した。乱流で作動する従来の噴霧乾燥機と異なって、NSDB−90は層流で作動し、それ故穏やかな加熱を達成することができて、したがって該システムを熱に敏感な生物薬剤生成物に対して適合性にする。
一次ナノカプセル中のインスリン含有率
HPLCによる適当な分析方法を以下の条件で開発した。
c4カラムを、分離のために及びh−インスリンを分析するために使用した(RESTEKviva 内径4.6mm×250mm、5μmの粒子、Bellefonte、ペンシルバニア州、米国)。カラム温度は45℃に保った。移動相Aはアセトニトリル(ACN)であり、移動相Bはリン酸二水素カリウム(KH2PO4、20mmol/L)であり、リン酸でpH2.5に調節した。移動相は0.45μmの膜フィルターを通して濾過して、使用前に真空により脱ガスした。
試料を超純水+vortexで懸濁して、次に少なくとも30分間震盪した後凍結した。1.5μmの体積の懸濁液を、間にスペーサーとして200メッシュのTEM格子を有する2枚の小さいアルミニウム平板の間にサンドイッチした。次に試料をHPM010高圧凍結機(Bal−Tec、Liechtenstein)で高圧凍結した。凍結した試料を支持台に載せ、VCT 100 Vacuum Cryo Transfer device(Bal−Tec)を使用してBAF60凍結破砕デバイス(Bal−Tec)に移した。−120℃の温度で破砕した後、試料を−110℃で5分間エッチングして二軸回転シャドーイングにより3nmのPt/Cでコートした。VCT100を使用して試料をUltra 55 SEM(Zeiss、ドイツ)に移し、二次電子インレンズ検出器を1.5kV、−120℃の温度で使用して観察した。そのSEM像を図11Α〜11Cに示す。
0.05Mクエン酸塩緩衝液で希釈した(50mg/kg体重)ストレプトゾトシン(STZ)の静脈注射により、ラットにDMを誘発した。2週間後に、糖尿病性として選択された動物は、250mg/dLを超える空腹時血糖を示したものであった。血糖は、グルコースオキシダーゼ法(Bergmeyer及びBernt、1974)により臨床用グルコース計(Contour(商標)、BAYER社)を使用して測定した。
Claims (31)
- 水溶性タンパク質、グルカン及び親水性の活性作用物質を含むナノ粒子であって、前記グルカンはメタリン酸塩により少なくとも部分的に架橋されているナノ粒子。
- 前記水溶性タンパク質がアルブミンである、請求項1に記載のナノ粒子。
- 前記アルブミンがヒト血清アルブミン(HSA)及びウシ血清アルブミン(BSA)から選択される、請求項2に記載のナノ粒子。
- 前記グルカンがα−グルカンである、請求項1から3のいずれか一項に記載のナノ粒子。
- 前記α−グルカンが、デキストラン、グリコーゲン、プルラン、デンプン、グリコーゲン、リケニン、マンナン、ガラクトマンナン、アラビノキシラン、及びガラクトンから選択される、請求項4に記載のナノ粒子。
- 前記グルカンが約5KDaと2,000KDaの間の分子量を有する、請求項1から4のいずれか一項に記載のナノ粒子。
- 前記メタリン酸塩が、トリメタリン酸ナトリウム(STMP)、五塩基性トリリン酸ナトリウム(STPP)及びオキシ塩化リン(POCl3)から選択される、請求項1から6のいずれか一項に記載のナノ粒子。
- 前記メタリン酸塩がトリメタリン酸ナトリウムである、請求項7に記載のナノ粒子。
- 前記親水性の活性作用物質が、−NH2末端部分又は−NH2側鎖部分を含む、請求項1から8のいずれか一項に記載のナノ粒子。

- 水溶性球状タンパク質、グルカン及び親水性の活性作用物質を含むナノ粒子を調製するプロセスであって、前記グルカンは、メタリン酸塩により少なくとも部分的に架橋されており:
