Abstract
Description
本発明の目的は、FGF−18タンパク質を含む新規製剤を提供することである。さらに詳しくは、前記製剤はFGF−18を含有するヒドロゲルであり、このヒドロゲルは好ましくは感熱性ヒドロゲル、より好ましくはキシログルカンゲルである、本発明はまた、液体配合物から出発して、本発明のヒドロゲルを調製するための方法を提供する。本明細書に記載のFGF−18を含有するヒドロゲルは、骨関節炎又は軟骨損傷などの軟骨障害の治療での投与に有用であり得る。特に関係するものは、FGF−18タンパク質をさらに含むキシログルカンヒドロゲルである。注入前、又はゲル化温度に供される前に、本発明のヒドロゲルは液体形態であることに留意されたい。
1)キシログルカン及び緩衝液と一緒にFGF−18を含むか又はそれから成る溶液を調製する工程と、
2)ゲルを37℃又は約37℃の温度に曝露してゲルを形成する工程と、

− 本明細書において用語「FGF−18タンパク質」又は「FGF−18」は、ヒトFGF−18タンパク質の少なくとも1つの生物活性を維持しているタンパク質であることが意図される。FGF−18は、未変性でも、成熟型でも、切断型でもよい。ヒトFGF−18タンパク質の生物活性は、特に骨芽細胞活性(国際公開第98/16644号を参照)又は軟骨形成(国際公開第2008/023063号を参照)の上昇を含む。未変性の又は野生型のヒトFGF−18は、関節軟骨の軟骨細胞により発現されるタンパク質である。ヒトFGF−18は最初zFGF−5と呼ばれ、国際公開第98/16644号に詳細に記載されている。配列番号1は未変性のヒトFGF−18のアミノ酸配列に対応し、アミノ酸残基1(Met)〜27(Ala)からなるシグナルペプチドを有する。ヒトFGF−18の成熟型は、配列番号1の残基28(Glu)〜残基207(Ala)からのアミノ酸配列(180アミノ酸)に対応する。

本発明の主要な目的は、キシログルカン、FGF−18タンパク質、及び緩衝液を含むか又はそれらから成るキシログルカンゲル製剤(又はヒドロゲル)である。前記ヒドロゲルは、イン・サイチュで注射前に又はゲル化温度に曝露される前に液体の形態であるため、本発明の別の主要な目的は、キシログルカン、FGF−18タンパク質、及び緩衝液を含むか又はそれらから成るキシログルカンゲル液体製剤である。好適な実施態様において、キシログルカンは脱ガラクトシル化キシログルカンであり、さらにより好ましくは、脱ガラクトシル化度が約44又は約45%であるDeg−キシログルカンであり、緩衝液は、PBSなどのリン酸緩衝液である。
− FGF−18: 0.00001〜0.6重量%、例えば0.0054重量%;
− キシログルカン: 1〜5重量%、例えば3又は4重量%;
− 緩衝液: 95〜99重量%、例えば96又は97重量%。

本実施例の組換え切断型FGF−18(trFGF−18又はスプリフェルミン)は、国際公開第2006/063362号に記載された技術に従って、大腸菌(E. coli)での発現により自家調製された。以下の実施例では、スプリフェルミンとFGF−18は交換可能に使用される。
− タマリンド(Tamarind)種子からキシログルカンは、Megazyme International (Ireland) から得た。それは、Rilton et al., 2011 により報告されたプロトコールに従って脱ガラクトシル化された。
− BSA、HSA、及びポロキサマーF68は、Sigma Aldrich から得た。
− ペニシリンストレプトマイシン(Pen-Strep)及びダルベッコーPBS 10×は Gibco から得た。
− キトサン75% DD HMW、Sigma Aldrich 419419、
− キトサン95% DD LMW、Faravelli 43000、
− キトサン95% DD HMW、Heppe medical 24711。
FGF−18溶液の調製
5.41mg/mlのFGF−18を、個々の3mlのバイアル中で−80℃にて保存し、室温で溶解後、それを直接ポリマー溶液(ポリマー溶液とFGF−18との比は9:1)に添加して目標の最終濃度540mcg/mlを達成するか、又は「タンパク質バルク」で希釈して54mcg/mlの目標最終濃度を達成した後、ポリマー溶液に加える。「タンパク質バルク」は、Na2HPO4(7mM)、KH2PO4(1mM)、及びKCl(2.7mM)から調製したpH7.3のPBS溶液である。この緩衝液のイオン強度は約25mMである。
タイプI(またD−PBSとも呼ぶ):ダルベッコーPBS 10×、ミリポア水で10倍希釈、0.1重量% Pen−Strep;pH5.5。D−PBS 1Xイオン強度は約166mMであり、その組成は以下の通りである:CaCl2(0.9mM)、MgCl2(0.49mM)、KCl(2.66mM)、KH2PO4(1.47mM)、NaCl(137.9mM)、のNa2HPO4(8.06mM)。
タイプII:0.1重量% BSAを含むタイプI。
タイプIII:0.25g/Lポロキサマー、1重量%Pen−Strep、及び1重量% HSAを含む「タンパク質バルク」として;pH7.3。
タイプI溶媒(D−PBS)又はPBS pH7.3(「タンパク質バルク」として調製された)中で調製されたDeg−XG溶液について、システムにFGF−18溶液を重量比9:1で充填した時と同じポリマー最終濃度を得るために、ポリマー濃度を10%で増分させた。
Deg−XG溶液は、ミリポア水とタイプI溶媒(pH5.5)の両方で調製した。4.4重量%のDeg−XG溶液も、pH7.3の「タンパク質バルク」中で調製した。
・固体ポリマーを冷水又は所望濃度の冷PBSに添加、
・5℃及び13500rpmで5時間ホモジナイズ、
・120℃で20分間オートクレーブ、
・5℃で保存。
シリンジ可能性:シリンジ可能性は、G25針を有するシリンジにより1mlを注入することにより室温で試験した。注入時間とシリンジ中の残量を評価した。
剪断粘度:剪断粘度測定は、25℃でレオメーター Ar 1000(TA Instruments)を用いて行った。
傾き挙動試験:2〜3mlの溶液を透明な円筒管中で37℃にてインキュベートし、異なる時間後に観察した。管を傾けて、物質が液体様(「流れる(flow)」)であるか又はゲル状(「流れない(no flow)」)であるかを評価した。
Deg−XGゲルの動的機械的特性 (dynamic-mechanical properties)は、(応力制御された (stress controlled))小振幅の剪断実験により評価した。試験は、アクリル板形状(直径4cm)と500μmのギャップを有する応力制御されたレオメーター Ar 1000(TA Instruments)を使用して行なった。
・37℃でのゲル化速度は、4.10-3 の繰り返し周波数掃引試験により検討した。
・25℃でのゲル化速度は、1Hzの固定周波数と4.10-3 の歪みの時間掃引試験 (time sweep test)で検討した。
表面形態は、10kVの加速電圧で電界放射型走査電子顕微鏡(FESEM)システム(JEOL)により画像化した。FESEM用試料は、走査前に30mAで50秒間、JFC-1300金コーター(JEOL)を用いて金層で被覆した。凍結乾燥された試料は、黒鉛接着剤層によりSEMアルミニウムスタブ上に取り付けた。
水の中で調製されたDeg−XGゲルを用いる予備試験のために、ゲル試料(各システムで4〜6個)を、多孔質底部(空隙率G0/G1の焼結ガラス)を有する予め秤量した円筒形ガラスバイアルに入れ、大過剰量の放出媒体タイプIに浸漬し、37℃に設定した恒温槽に入れた。FGF−18を含むDeg−XGゲルの場合は、ゲル試料は、底部に多孔質膜(0.4μm)を有するマルチウェルプレートの予め秤量したインサートの上に置き、放出媒体タイプIIに浸漬し、37℃に設定した恒温槽に入れた。100rpmで回転振盪を行なった。放出媒体は2〜3日毎に交換した。ゲル試料は、インキュベーション前と37℃で異なる時間インキュベートしたに秤量した。Ws(t)は、時刻tにおける膨潤試料の重量であり、Ws(0)は時間=0における試料の重量である。
pH7.3のPBS(「タンパク質のバルク」として)で調製され、FGF−18を含む4重量%のDeg−XG上で行われた放出試験について、ゲル試料は、底部に多孔質膜(0.4μm)を有するマルチウェルプレートの予め秤量したインサートの上に置き、放出媒体のタイプIIIに浸漬し、37℃に設定した回転振盪機に入れた。この試験(各々について4試料)に供されたシステムは、
・Deg−XG 4重量%、
・540μg/mlのFGF−18を含むDeg−XG 4重量%、
・54μg/mlのFGF−18を含むDeg−XG 4重量%。
膨潤試験に使用したものと同じ試料を、インビトロ放出試験についても分析した。具体的には、受容相をHPLCにより分析した。選択された試料はまた、ビアコアでも分析した(データは示していない)。
Deg−XG系の温度応答性ゲル化システム
Deg−XG溶液を水で、1、2、3、4、5重量%で調製した。Deg−XGはD−PBS中で、3.3重量%及び4.4重量%で調製し、それらにはまた54μg/mlのFGF−18を充填した。すべてのシステムは、5℃で一晩保存した後(時間=0)及びさらに37℃で1、2、及び3時間インキュベーション後、シリンジ可能性試験 (syringability test)に供した。
Deg−XG系の温度応答性ゲル化システムの動的機械的挙動 (dynamic mechanical behaviour)
37℃で異なる時間インキュベートしたゲルの動的機械的挙動を、歪み掃引試験 (strain sweep test)及び周波数掃引試験 (frequency sweep test)により調べた。
ゲル化速度試験 (gelation kinetics study)
Deg−XG/水システムについて37℃で所定の時間間隔で繰り返し周波数掃引により、ゲル化速度の試験を行なった。1Hzでの貯蔵弾性率と損失弾性率の値は、時間の関数としてプロットされる(図11)。
凍結融解 (freezing-thawing)及び凍結乾燥の影響
D−PBS中で調製した4.4重量%のシステムで、Deg−XGゲルの可能な貯蔵条件に関するいくつかの情報を集めるために、動的機械スペクトル (dynamic mechanical spectrum)に対する1回の凍結融解サイクルの影響を調べた。同じ目的のために、凍結乾燥及び再構成の影響を評価した。この最後の場合に、ゾル状態(5℃の保存温度で)及びゲル状態(37℃でコンディショニング後)の両方からの材料を凍結乾燥した(図12及び13)。
凍結乾燥したゲルの走査電子顕微鏡試験
Deg−XGゲルを液体窒素(バイアル内)に浸漬し凍結乾燥することにより迅速に凍結した、水中で調製したDeg−XGゲルを、走査電子顕微鏡試験により分析した(データは示していない)。すべてのシステムは、数十ミクロンの大きな孔とわずか数ミクロンの小さな孔のある不規則な空隙率を示す。ポリマーの濃度を上昇させると、一般的な作用は、より大きな空洞の寸法を低減させる。しかし、本来の不均一性にもかかわらず、試料の形態は非常に均一であった。
膨潤−浸食試験
4重量%と5重量%のDeg−XG/水ゲルの膨潤−浸食挙動の予備試験を、60日間にわたって行った。37℃でのインキュベーション前及びインキュベーション中にゲル試料を秤量した。Ws(t)は時間tにおける膨潤試料の重量であり、Ws(0)は、時間t=0における試料の重量である(表3)。60日後、カビの増殖が原因で試験を中止した。両方のシステムについて、Ws(t)/Ws(0)[%]は、経時的にゆっくり低下する。浸食されたゲルは、液体窒素中で速やかに凍結し凍結乾燥した後、SEM顕微鏡試験に供した(データは示していない)。浸食後、ゲル構造は崩壊せず、空隙率はより均一になる。また、空隙率は、5重量%のシステムの場合よりも4重量%のシステムについて、より開いていて相互接続されているように見えた。
ゲル浸食/タンパク質放出の試験
PBS pH7.3中で調製した4重量% Deg−XGについて試験を行った。24時間、48時間、及び7日後に集めた受容相を、ビアコア及びRP−HPLC分析に供した(データは示していない)。RP−HPLC分析は、FGF−18のバースト放出に起因する明確なクロマトグラフピークを示さなかった。同じ受容相にビアコア分析も行った。同様に、FGF−18は検出されなかった。これは、観察期間中、このタンパク質がゲルマトリックス中に捕捉されたことを示唆している。
細胞増殖アッセイ
8.1. 細胞増殖のためのアラマーブルー(Alamar Blue)アッセイ
FGF−18を含まないか、又は3つの異なる濃度(54mcg/ml、6,7mcg/ml、6,7ng/ml)のFGF−18を含む、Deg−XGヒドロゲル(3.3重量%で)上で培養した軟骨細胞を用いて、細胞増殖に関するアラマーブルーアッセイ(5000個の軟骨細胞/ウェル)を行った。対照システム(FGF−18を含まないDeg−XGゲル)を含むすべてのシステムは、経時的に顕著な細胞増殖を示した(図15を参照)。増殖因子の存在は、最初の8日間、増殖速度に影響を与えなかったが、延長されたインキュベーション時間のためにわずかに上昇した。増殖因子の試験した濃度範囲内で、FGF−18濃度の影響は観察されなかった。
FGF−18を含むか又は含まないDeg−XGヒドロゲルで12日間培養した細胞について、アクリジンオレンジで染色した後、共焦点顕微鏡分析を行った。アクリジンオレンジは、細胞浸透性の核酸結合色素であり、これは2本鎖DNAに結合すると緑色の蛍光を発し、1本鎖DNA又はRNAに結合すると赤色蛍光を発する。この染色は、生きている(緑の核)細胞とアポトーシス(赤の核)細胞とを区別する。図16に示されるように、緑色の丸形の核は、細胞がFGF−18の存在下と非存在下の両方でDNA損傷を受けなかったことを示す。
FGF−18を含むか又は含まないDeg−XGゲル上の軟骨細胞の増殖を、光学顕微鏡下で7日間にわたって追跡した。成長因子の含有量は、6.7ng/ml〜54μg/mlの範囲であった。すべてのシステムで、軟骨細胞はまずクラスターを形成することが観察された。低濃度(6.7ng/ml及び6.7mcg/ml)のFGF−18をゲルに充填すると、48時間インキュベーション後、軟骨細胞はクラスターから移動して離れ、足場の他の部分にコロニー形成した(データは示していない)。
全体厚さが約2mmのヒドロゲル試料の210μm厚の層から採取した異なる切片の共焦点顕微鏡分析を、4日間インキュベーション後のDeg−XG+FGF 6.7mcgのシステムについて行った。細胞を臭化エチジウムで染色した(赤)。結果は、材料の厚さにわたる軟骨細胞の存在を示す。ヒドロゲル表面に細胞接種を行ったため、厚さにわたる異なる位置における細胞の存在は、増殖細胞がゲルの内層にアクセスすることができることを示唆している(データは示していない)。
キトサンゲルの調製
9.1. 総論
キトサン製剤のスクリーニングのために、ポリマー液体溶液の調製は、3種類の異なるキトサンを利用した:高分子量(HMW)を有する95%の脱アセチル化度(DD)、低MW(LMW)を有する95%DD、及び高MW(HMW)を有する75%DD。ポリマーの液体溶液は、5℃で又は25℃で激しく撹拌しながら、酢酸0.1 N溶液に徐々にキトサンを添加することにより調製した。ポリマーの量は、ポリマー液体溶液中で1重量%、1.5重量%、又は2%重量の最終ポリマー濃度を有するように計算された。いったんキトサンが完全に可溶化された後、ミリQ水中の10mM、100mM、又は500mMの濃度のKH2PO4の溶液を撹拌しながら添加して、ポリマー液体溶液中の1mM、10mM、又は50mMのいずれかの最終濃度を得た。最後に、ミリQ水中20重量%濃度のβ−グリセロリン酸(β−GP)の溶液を加えて、最終的な液体溶液のpHを6.0、6.5、又は7.0のいずれかに調整した。これは、多すぎる量のβ−GPを必要として、350mOsm/kgの目標オスモル濃度値を超えるか、又は既に室温でゲルを得られてしまうため、絶えず所望のpH値に到達することは必ずしも可能ではなかった。ポリマーの液体溶液は適宜、ゲル形成まで37℃でインキュベートした。すべてのスクリーニングした製剤のオスモル濃度を測定し、350mOsm/kgより高いオスモル濃度を有する製剤、すなわち最終β−GP濃度が2.5重量%超の製剤を捨てた。
キトサンは、温度変化に伴うゾル−ゲル遷移を受け得ることが報告されているが、このプロセスは、ポリマーの分子量(MW)、その脱アセチル化度(DD)、溶液中のポリマー濃度、温度、時間、ポリマーの可溶化中の混合速度、溶液の最終pH、及び他の賦形剤の存在により大きく影響される。
この製剤を改善するために、β−GPの濃度を1.65〜1.7重量%の値でほぼ一定に維持し、キトサン濃度を1.5重量%〜1.8重量%に変化させ、及びHECの量を徐々に0.1重量%まで低下させて、以下の試験を行った。

Claims (12)
- FGF−18、キシログルカン、及び緩衝液を含むか又はそれらから成る液体製剤。
- 前記キシログルカンが部分的に脱ガラクトシル化されている、請求項1に記載の液体製剤。
- 前記キシログルカンが44若しくは45%、又は約44若しくは約45%の脱ガラクトシル化度を有する、請求項2に記載の液体製剤。
- 前記緩衝液がリン酸緩衝液である、請求項1〜3のいずれか1項に記載の液体製剤。
- FGF−18が、
a.配列番号1のアミノ酸残基28〜207を含むか又はそれから成るポリペプチド、
b.配列番号1のアミノ酸残基28〜196を含むか又はそれから成るポリペプチド、及び
c.配列番号2を含むか又はそれから成るポリペプチド
から成る群から選択される、請求項1〜4のいずれか1項に記載の液体製剤。 - 前記キシログルカンが3〜4重量%の濃度であり、前記緩衝液が96又は97重量%の濃度である、請求項1〜5のいずれか1項に記載の液体製剤。
- ポリマー:FGF−18の比率が9:1である、請求項1〜6のいずれか1項に記載の液体製剤。
- 前記製剤が、37℃又は約37℃の温度に曝露された時にヒドロゲルを形成する、前記請求項のいずれか1項に記載の液体製剤。
- 請求項8に記載のヒドロゲルを製造する方法であって、
a.請求項1〜7のいずれか1項に記載の液体製剤を調製する工程と、
b.前記液体製剤を37℃又は約37℃の温度に曝露してゲルを形成する工程と、
を含む、方法。 - 前記ゲルがイン・サイチュで形成される、請求項9に記載の方法。
- 請求項9又は10に記載の方法に従って得られる、ヒドロゲル。
- 請求項1〜8のいずれか1項に記載の液体製剤を含むか又はそれから成る容器を含む、製品。
