Abstract
Description

本出願は、創傷を治療又は予防するための方法に関する。
創傷治癒は、凝固段階、炎症段階、肉芽組織形成段階、及び組織再構築を含む複雑なプロセスである。これらの事象は、損傷部位で放出されるサイトカインや増殖因子により開始される。多くの因子が、正常で適切な創傷治癒を悪化させるか又は妨害する。例えばそのような因子には、例えば年齢、感染、栄養不良、免疫抑制、薬物療法、放射線照射、糖尿病、末梢血管疾患、全身性疾患、喫煙、ストレスなどがある。

創傷は、他の欠陥、例えば代謝欠陥(例えば、本明細書に記載されているように、又はストレプトゾトシンの投与により)に患う動物で産生することができる。
VEGF−Bシグナル伝達を阻害する化合物(活性成分)は、予防的又は治療的処置のための、非経口、局所、経口、又は局所投与、エアロゾル投与、又は経皮投与のために有用である。ある例において本化合物は、非経口的に、例えば皮下又は静脈内に投与される。

中和抗VEGF−B抗体は糖尿病マウスの創傷を治療する
VEGF−Bの抗体介在阻害は糖尿病性db/db//BKSマウスの血中グルコース値に適度に影響を与える
自家飼育した雄のC57BKS/Leprdb(db/db/BKS)マウスは、Jackson Laboratoryから購入した。創傷前及び実験中、1つのケージに1匹のマウスを収容した。動物施設は12時間の明/暗サイクルを有し、食餌と水は自由に与えた。強化材料は、ストレスを軽減するために、ラットのトンネルとネスティング材料を含んでいた。db/db/BKS雄マウス(8週齢)を400μgのVEGF−B抗体(2H10)又はアイソタイプ一致対照抗体(BM4)を用いて、創傷形成前4週間、週に2回(腹腔内)前処理した。創傷形成後、マウスを屠殺するまで、同じ治療計画を継続した。実験前及び実験中に、体重及び血中グルコースを追跡した。血中グルコース値は、血中グルコース計(Countour stick, Bayer)を用いて尾から分析し、体重を実験室規模で測定した。
15mM超の非空腹時血中グルコース値に達した後、2H10又はBM4(対照)処置されたdb/dbの又はdb/+ は、創傷形成前にさらに2週間飼育された。これは、創傷形成時に重症の糖尿病動物を得るために行なった。創傷形成は、イソフルラン吸入による全身麻酔下で行なった。首の後ろの毛を、電気バリカンと脱毛クリーム(Veet)の塗布により除去した。皮膚を70%エタノールで消毒し、次に6mmの生検パンチを使用して、皮膚と皮筋層を貫通する2つの対をなす円形全層創傷形成を行なった。創傷形成後の最初の2日間、マウスに1日2回、0.05〜0.1mg/kgのブプレノルフィンを皮下麻酔注射した。創傷の大きさと治癒の進行は、創傷形成後直接デジタル写真で追跡し、次に実験中は1日おきに追跡した。創傷面積は、Image J ソフトウェアを使用して各創傷について2回測定した。カメラと創傷との距離は、創傷の大きさを、すべての画像に含まれる基準物の大きさと比較して、補正した。1つの傷(動物当たり2個)は、単一の実験データ点として処理した。
雄のdb/db及びdb/+マウス(上記のように処置し、16日目に屠殺した)を発現分析のために使用した。創傷を切開し、ドライアイス上で急速凍結した。製造業者の指示に従ってRNeasyミニキット(Qiagen)を用いて創傷から総RNAを抽出し、精製した。0.5〜1μgの総RNAから iScript cDNA synthesis kit (Bio-Rad) を用いて、第1鎖cDNAを合成した。Rotor-Gene Q (Qiagen) リアルタイムPCRサーマルサイクラー中の KAPA SYBR FAST qPCR Kit Master Mix (2x) Universal (KAPA Biosystems) を使用して、製造業者の指示に従ってリアルタイム定量PCRを行なった。発現レベルは、L19の発現に対して標準化した。
免疫学的分析のために、雄のdb/dbマウス(上記のように処置し、16日目に屠殺した)を使用した。創傷を切開し、4℃で24時間パラホルムアルデヒド中で固定し、70%エタノール中で水和し、パラフィンに包埋し、断面の厚さ3μmの切片にした。抗原回収溶液をpH6(DAKO)を用い、切片を+98℃に10分間加熱して、抗原回収を行なった。適切な1次抗体(BrDU、CD45、CIV及びCD31、1〜5μg/mlに希釈)を用いて、切片を+4℃で12時間インキュベートした。一致する蛍光標識した2次抗体(Invitrogen, Alexa fluor、1:1000に希釈)を適用し、切片を室温(RT)で1時間インキュベートし、次に、これらを顕微鏡観察のために調製した。AxioVision 顕微鏡 (Carl Zeiss) を20×倍率で使用して、切片を写真撮影した。次に、AxioVision Run wizardプログラム(pixels2)を用いて、切片を評価し定量した。BrdU染色強度は、DAPIの対比染色強度に対して定量し、他のすべての染色は全画素領域について定量した。
対照抗体(BM4)で処置したdb/db又はdb/+を基本的に上記したように創傷形成させ、創傷形成後16日目に屠殺した。発現と組織学的分析のために、6ミリメートル生検パンチを用いて創傷を取った。組織を液体窒素中で凍結させ、さらなる処理まで−80℃で保存した。製造業者の説明書に従って TRIzol 試薬(Invitrogen)及び QIAGEN RNeasy (Qiagen) を用いてRNAを単離した。総RNA(500ng)を製造業者の説明(iScript cDNA synthesis kit, Bio-Rad)に従って逆転写した。Platinum SYBR green SuperMix (Invitrogen) と反応あたり25ngのcDNAを用いて、qPCRを行なった。発現レベルはTUBB5(チューブリン、ベータ5クラスI)の発現に対して標準化した。
オイルレッドO (Oil red O)(ORO)分析のために、雄のdb/db及びdb/+マウス(上記のように処理された)を使用した。創傷を切開し、ドライアイス上で急速凍結し、クライオスタット (cryostat)の金型上で Tissue-Tek(登録商標)(Sakura)に直接包埋した。凍結切片(12μm)を、オイルレッドO作業溶液(400mlの99%イソプロパノールに溶解した2.5gオイルレッドO(Sigma-Aldrich)を、水でさらに6:10希釈し、22μmフィルター(Corning)で濾過した)中に12分間浸漬し、さらに水道水で20分間すすいだ後、これらをマウントした。染色した切片を40×倍率で明視野顕微鏡(Axio Vision 顕微鏡 Carl Zeiss)で観察し、ウス1匹あたり少なくとも4フレームをキャプチャーした。画像は、創傷の中央領域からキャプチャーした。脂肪滴の定量のために、各フレーム内の赤色の画素の量を、Axio Vision Run ウィザードプログラムを用いて総ORO染色(ピクセル、a.u)について 定量した。
Claims (19)
- 創傷を患う被験体の、創傷を治療するか、創傷治癒を増強するか、又は創傷の進行を防止若しくは遅延させるための方法であって、VEGF−Bシグナル伝達を阻害する化合物を被験体に投与することを含む、方法。
- 創傷が急性又は通常の創傷である、請求項1に記載の方法。
- 創傷が慢性である、請求項1に記載の方法。
- 創傷が皮膚創傷である、請求項1〜3のいずれか1項に記載の方法。
- 被験体が肥満及び/又は糖尿病を患っている、請求項1〜4のいずれか1項に記載の方法。
- 創傷を患う糖尿病被験体の、創傷治癒を増強若しくは誘導するための、又は糖尿病被験体の創傷の進行を低下させるか若しくは防止するための方法であって、VEGF−Bシグナル伝達を阻害する化合物を前記被験体に投与することを含む、方法。
- 前記化合物が、以下の効果:
(a)前記化合物が投与されていない被験体(例えば、糖尿病と創傷とを患っている被験体)における創傷閉鎖の速度と比較して、創傷閉鎖の速度を上昇させること、及び/又は
(b)前記化合物が投与されていない被験体(例えば、糖尿病と創傷とを患っている被験体)の割合と比較して、ある特定の時点(例えば治療開始後)における創傷閉鎖の量を増加させること、及び/又は
(c)前記化合物が投与されていない被験体(例えば、糖尿病と創傷とを患っている被験体)と比較して、被験体の血管成熟を向上させること、
の1つ以上を有するために有効な量で投与される、請求項1〜6のいずれか1項に記載の方法。 - VEGF−Bシグナル伝達を阻害する前記化合物がVEGF−Bに結合する、請求項1〜7のいずれか1項に記載の方法。
- 前記化合物が抗体模倣物である、請求項8に記載の方法。
- 前記化合物が、VEGF−Bに結合するか又は特異的に結合し、VEGF−Bシグナル伝達を中和する、抗体の可変領域を含むタンパク質である、請求項8に記載の方法。
- 前記化合物がFvを含むタンパク質である、請求項10に記載の方法。
- 前記タンパク質が、以下の(i)〜(x):
(i)1本鎖Fv断片(scFv);
(ii)二量体scFv(di−scFv);又は
(iv)二重特異性抗体;
(v)三重特異性抗体;
(vi)四重特異性抗体;
(vii)Fab:
(viii)F(ab')2;
(ix)Fv;又は
(x)抗体の定常領域であるFc、又は重鎖定常ドメイン(CH)2及び/又はCH3に連結した(i)〜(ix)の1つ
からなる群から選択される、請求項11に記載の方法。 - 前記タンパク質が抗体である、請求項11に記載の方法。
- 前記化合物が、(配列番号3で示される配列を含む重鎖可変領域(VH)と、配列番号4で示される配列を含む軽鎖可変領域(VL)とを含む)抗体2H10のVEGF−Bへの結合を競合的に阻害する、請求項1〜13のいずれか1項に記載の方法。
- 前記化合物が、抗体2H10のヒト化可変領域を含むタンパク質である、請求項1〜14のいずれか1項に記載の方法。
- 前記タンパク質が、抗体2H10のVH及び/又はVLの相補性決定領域(CDR)を含む可変領域を含む、請求項15に記載の方法。
- 前記タンパク質が、
(i)VHであって、
(a)配列番号3のアミノ酸25〜34で示される配列を含むCDR1;
(b)配列番号3のアミノ酸49〜65で示される配列を含むCDR2;及び
(c)配列番号3のアミノ酸98〜108で示される配列を含むCDR3、を含むVH、及び/又は
(ii)VLであって、
(a)配列番号4のアミノ酸23〜33で示される配列を含むCDR1;
(b)配列番号4のアミノ酸49〜55で示される配列を含むCDR2;及び
(c)配列番号4のアミノ酸88〜96で示される配列を含むCDR3、を含むVL
を含む、請求項16に記載の方法。 - 前記タンパク質が、配列番号5で示される配列を含むVHと、配列番号6で示される配列を含むVLとを含む抗体である、請求項17に記載の方法。
- 前記化合物がVEGF−Bの発現を阻害するか又は防止する、請求項1〜7のいずれか1項に記載の方法。
