Abstract
【解決手段】多孔質体は、孔径が1−100μmである連続孔を有し、空隙率が50%以上であり、表面にタンパク質と結合可能な官能基を有する高分子樹脂から成る。官能基にリガンドとしてプロテインAを結合させた多孔質体は、通水性、耐圧性に優れるので、高流速下で、特定物質を吸着できる。高分子基材をエチレン−グリシジルメタクリレート共重合体とすることで、官能基としてのエポキシ基を表面に導入するのが容易になり、したがって、リガンドとしてプロテインAを結合させるのも容易になる。リガンドを結合させた多孔質体を担体としてカラムに充填した分離装置は、IgGなどの抗体の精製を、高速、低コストで行うことができる。

Description
この発明は、多孔質体の形成後に官能基を導入する態様と、官能基を予め有する高分子樹脂を用いて多孔質体を形成する態様の両方を含む。
この発明は、官能基を予め有する高分子樹脂で多孔質体を形成する態様に対応するものである。官能基を有するモノマーを重合させる態様の外、基体の形成後に官能基をグラフト重合によって導入する態様や、予め官能基がグラフト重合された高分子成分を用いて重合体を形成する態様を含んでいる。
この発明における官能基を含む成分とは、たとえばグリシジルメタクリレート(GMA)であり、その他の成分とはたとえばオレフィンである。
得られた成形物の少なくとも一部を、前記高分子樹脂の融点よりも高い温度の液体に浸漬する工程、および
水系溶媒で前記形孔剤を溶出して連続孔を有する多孔質体を得る工程
を含むことを特徴とする多孔質体の製造方法である。
また、予め所定割合で官能基を導入した高分子から形成するため、細孔構造形成後に官能基を導入するのと異なり、均一な分布の官能基の導入が可能である。
基体は、弾性変形可能な高分子樹脂で形成されることが望ましい。また、高分子樹脂として熱可塑性樹脂を用いると、連続孔の孔径や空隙率の調整が容易になる。このような高分子樹脂としては、例えば、エチレン−酢酸ビニル共重合体(PEVA)や、エチレンーグリシジルメタクリレート共重合体(PEGM)が好適である。特にPEGMは、PEVAと比較して、官能基としてのエポキシ基を表面に導入するのが容易であるので、より好ましい。
官能基としては、タンパク質と結合可能な官能基であれば、特に制限されないが、エポキシ基、アミノ基、アルデヒド基、ヒドロキシ基、カルボキシル基、スルホ基、N−ヒドロキシサクシンイミド基などが好適に用いられる。
(1)タンパク質と結合可能な官能基を有する高分子樹脂に、1〜100μmの粒子径を有する水系溶媒に溶解可能な形孔剤を混練して成形する工程、
(2)得られた成形物の少なくとも一部を、前記高分子樹脂の融点よりも高い温度の液体に浸漬する工程、および
(3)水系溶媒で前記形孔剤を溶出して連続孔を有する多孔質体を得る工程
を含む。
モノリス型樹脂ビーズとして用いる場合は、例えば通常の高速液体クロマトグラフィ用のカラム容器に樹脂ビーズを充填させて使用することができる。
以上の特性より、本発明の多孔質体は医薬品精製、特に抗体医薬などのバイオ医薬品の精製の高速化と低コスト化をもたらすものと期待できる。
多孔質体
多孔質体の材料には、エポキシ基を含む高分子であるPEGMを用いる。
多孔質体の形成は、熱可塑性のPEGMポリマー(住友化学社製、ボンドファーストCG5001、GMA比率19重量%)100重量部と、形孔剤として水溶性樹脂である粒子径20〜50μmペンタエリスリトール240重量部とを熱混練によって均一化した後、これを円柱状に押し出し成形する。その後、水溶性のペンタエリスルトールを水洗浄によって成形体から取り除くことによって、多孔性の多孔質体を得る。得られた多孔質体は、孔径が平均10μmで空隙率が75%のスポンジ状基材である(スポンジモノリスとも言う)。
図2に示すように、円柱状に成形した多孔質体(スポンジモノリス)は、のり、グリセリンなどの潤滑剤を用いることにより、または熱収縮チューブを用いることにより、極めて簡便にカラム化できる。そして、このカラム化されたPEGM基材の多孔質体を、HPLC用カラム(例えば、内径4.6mm長さ100mm)に装填し、図示のように不要部分を切除する。これをHPLC装置に適用することにより、特定物質の分離装置とすることが可能である。
上記で得られた多孔質体カラムを、メタノール/水を溶媒として、送液ポンプを用いて洗浄する。次に、プロテインA溶液(1mg/mL、リン酸塩緩衝液pH7.0)でカラムを完全に満たした後、40℃で24時間反応させた。反応後のカラムを水、メタノールで洗浄し、プロテインA固定化カラムを得た。タンパク質の固定量は4nmol/g程度である。
以前までの研究において得られていたEVAを基材とする多孔質体では、比較的疎水的な溶質に対する保持が見られていたため、基本的に水を溶媒として用いるアフィニティ分離への適用に対しては、親水化が必須であると予想されていた。EVAの表面処理では、アセチル基の加水分解にともなう親水性の付与が確認されたが、他の基材による多孔質体の作製を検討した結果、グリシジル基を含むPEGMが最適であることが分かった。
実施例1と同様に、4mm内径30mm長のカラムを用いて、市販ProteinAカラム(MAbPac ProteinA:サーモフィッシャーサイエンティフィック株式会社製)と、本発明多孔質体にProteinAを結合させたものを用いて作成したカラムとを準備し、両者について、流速に対する圧損の変化を比較した。結果を表2に示す。表2から、本実施例の多孔質体は、従来型の粒子型カラム(MAbPac ProteinA)に比べ、流速に対して示す圧損が低いことが分かる。

検出波長:280nm
本実施例の試験に使用した液体クロマトグラフ装置は,LC−20シリーズ(島津製作所製)である。
本発明の多孔質体は、微粒子との複合化も可能である(Kubo,T.et al.Anal.Sci. 29,2013,417−421参照)。図11に、微粒子を複合化した多孔質体のSEM写真を示す。微粒子にエポキシ基等の官能基を含ませることにより、多孔質体に、特異的アフィニティを付与することが可能である。
Claims (12)
- 基体が高分子樹脂からなり、孔径が1−100μmである連続孔を有し、空隙率が50%以上である多孔質体であって、少なくとも基体の表面にタンパク質と結合可能な官能基を有することを特徴とする多孔質体。
- 前記基体が、タンパク質と結合可能な前記官能基を構成要素とする高分子樹脂からなる請求項1に記載の多孔質体。
- 前記高分子樹脂が、タンパク質と結合可能な前記官能基を含む成分とその他の成分との共重合体である請求項2に記載の多孔質体。
- 前記官能基がエポキシ基である請求項1〜3のいずれか1つに記載の多孔質体。
- 前記その他の成分がオレフィン類である請求項3または4に記載の多孔質体。
- 前記高分子樹脂が、エチレン−グリシジルメタクリレート共重合体である請求項2〜5のいずれか1に記載の多孔質体。
- 前記基体が弾性変形可能である請求項1〜6のいずれか1に記載の多孔質体。
- タンパク質と結合可能な官能基を介してリガンドが結合された請求項1〜7のいずれか1に記載の多孔質体からなることを特徴とするモノリス型アフィニティクロマトグラフィ用担体。
- 前記リガンドが、プロテインA、G、Lからなる群より選択される1または2以上のタンパク質よりなる請求項8に記載のモノリス型アフィニティクロマトグラフィ用担体。
- タンパク質と結合可能な官能基を介してリガンドが結合された請求項1〜7のいずれか1に記載の多孔質体からなることを特徴とするモノリス型樹脂ビーズ。
- タンパク質と結合可能な官能基を介して酵素が結合された請求項1〜7のいずれか1に記載の多孔質体からなることを特徴とするモノリス型リアクター担体。
- タンパク質と結合可能な官能基を有する高分子樹脂に、1〜100μmの粒子径を有する水系溶媒に溶解可能な形孔剤を混練して成形する工程、
得られた成形物の少なくとも一部を、前記高分子樹脂の融点よりも高い温度の液体に浸漬する工程、および
水系溶媒で前記形孔剤を溶出して連続孔を有する多孔質体を得る工程
を含むことを特徴とする多孔質体の製造方法。
