Abstract
Description

上式の例として、[Na+]=[0.368]および50%ホルムアミドを用い、GC含有量42%および平均プローブサイズ200塩基とすると、Tmは57℃となる。DNA二本鎖のTmは、相同性が1%減少する毎に1〜1.5℃下がる。したがって、配列同一性が約75%を超える標的であれば、42℃のハイブリダイゼーション温度で検出される。
「試料」、「患者試料」、または「生体試料」とは、一般に、特定の分子、好ましくはT2D特異的なマーカー分子について検査することができる試料を意味する。試料としては、これに限定されるものではないが、血液、血清、血漿、尿、唾液、涙、胸膜液などを含む細胞や体液などが挙げられる。
本明細書で同定されたタンパク質DNA結合複合体は、T2Dの病因に関連しているため、核酸とそれと相互作用するタンパク質とを含むTCF7L2のSNPの活性を調節する薬剤を同定する方法により、この疾患に伴う様々な障害を治療するための有効な治療薬が生成されるはずである。
本明細書に記載のT2D関連SNPが細胞代謝において果たす役割を解明することは、T2Dの治療および診断に有用な医薬組成物の開発を容易にする。これらの組成物は、上記の物質の1つに加えて、薬学的に許容可能な賦形剤、担体、バッファー、安定剤、または当業者によく知られている他の物質を含んでいてもよい。そのような物質は、非毒性であるべきであり、有効成分の有効性に干渉するべきではない。それが、個人に投与されるのが、ポリペプチド、抗体、ペプチド、核酸分子、小分子、または本発明に係る他の薬学的に有用な化合物のいずれであっても、「予防的有効量」または「治療的有効量」(場合によっては、予防が治療と見なされてもよいが)で投与されることが好ましく、これは個人に有効であることを示すのに十分な量である。
上記製品はいずれも、キットに組み込むことができ、キットは、T2D関連SNP特異的マーカーTCF7L2ポリヌクレオチド、または遺伝子チップ(Gene Chip)上に固定された1つ若しくは複数の当該マーカー、オリゴヌクレオチド、本明細書に記載の核酸を含むSNPに結合するポリペプチド、当該結合を分断するように設計されたペプチド、siRNA、小分子、抗体、ラベル、マーカー、またはレポーター、薬学的に許容可能なキャリア、生理学的に許容可能なキャリア、取り扱い説明書、容器、投与のための容器、アッセイ用基板、またはこれらの任意の組み合わせを含むことができる。
ヒトHCT116細胞をDMEM(4.5g/lグルコース、10%FCS、100U/mLペニシリン、および100ig/mLストレプトマイシン)中で培養した。インスリン刺激実験のため、細胞を、1uMインスリンで1時間処理した。SILAC実験のため、培養物を、標準的な細胞培養手順に従い、10%透析ウシ胎児血清(fetal bovine serum:FBS)、13C6−リジン、および13C6−アルギニンを加えたSILACライトおよびSILACヘビーでラベルしたダルベッコ改変イーグル培地(Dulbecco‘s Modified Eagle Medium:DMEM、サーモフィッシャー社(ThermoFisher))中で増殖させた。細胞を、冷リン酸緩衝生理食塩水(phosphate−buffered saline:PBS)で洗浄し、こそげとって回収し、5000rpm、4℃で5分間遠心分離した。108個の細胞を、5分ごとに10秒間ボルテックスしながら氷上で20分間インキュベートすることにより、250ulの低塩緩衝液(10mM HEPES、1.5mM MgCl2、10mM KCl、0.5%NP−40、プロテアソーム阻害剤、ホスファターゼ阻害剤、10%グリセロール、pH7.9)中に溶解させた。核を、10,000rpm、4℃で1分間遠心分離して、細胞質画分から分離した。核ペレットを、冷低塩緩衝液で1回洗浄し、遠心分離して回収した。高塩緩衝液(20mM HEPES、1.5mM MgCl2、420mM NaCl、0.2mM EDTA、プロテアソーム阻害剤、ホスファターゼ阻害剤、25%グリセロール、pH7.9)により核タンパク質を放出させ、12,000rpm、4℃で10分間遠心分離して、上清を回収した。
SNPであるrs7903146 5‘ビオチン標識二本鎖オリゴヌクレオチドを、インテグレイテッドDNAテクノロジーズ社(Integrated DNA Technologies,Inc.)にて合成した。オリゴヌクレオチドの配列は、以下の通りである。
サンプルをトリプシン消化し、ペンシルベニア大学のPenn Proteomics CoreにてナノLC/MS/MSで分析した(グラントP30CA016520(アブラムソンがんセンター(Abramson Cancer Center))およびグラントES013508−04(CEET)による支援を受けている)。データを、Sequest(サーモフィニガン社(ThermoFinnigan)、San Jose、カリフォルニア州、バージョン:SRF v.5)で分析した。Scaffold(バージョン:Scaffold_3.6.0、Proteome Software Inc.、Portland、オレゴン州)を用いて、MS/MSに基づいたペプチドおよびタンパク質同定を検証した。SILAC実験のため、質量分析法にズームスキャンを加え、標識および非標識ペプチドをモニターする。ズームスキャンしたSLICAピークを定量に使用できるようにカスタマイズ改変したProteoIQソフトウェアで、SILACピークの定量を行った。
IP緩衝液(50mM tris、150mM NaCl、プロテアソーム阻害剤、ホスファターゼ阻害剤、10%グリセロール、pH8.0)を用いて、4℃で一晩、ローテーター上で免疫共沈降を行った。標準的手順に従い、ウェスタンブロッティングを行った。抗TCF7L2抗体(ミリポア社(Millipore)、05−511)、抗Parp−1抗体(セルシグナリング社(Cell Signaling)、46D11)、抗RNAヘリカーゼA抗体(アブカム社(Abcam)、Ab26271)、抗THRAP3(ノーバス社(Novus)、NB100−40848)を用いた。
SNPであるrs7903146のTCF7L2−T2D関連作用における役割を解明するため、我々は、トランス作用タンパク質因子がこのゲノム領域に結合して、TCF7L2機能を調節するという仮説を立てた。この仮説を試験するために、我々は、質量分析計を用いて、SNP rs7903146を含むエレメントと相互作用するいくつかのタンパク質を同定した。表1を参照。HCT116細胞からの核溶解物を、SNP rs7903146全体に渡るビオチン標識二本鎖オリゴヌクレオチドと共にインキュベートした(図1A)。DNA−タンパク質複合体をストレプトアビジンビーズで沈殿させ、結合したタンパク質を変性SDS−PAGEにより分離し、クマシーブルーR−250で染色した。図1Bに示されるように、プルダウンサンプルにおいて、いくつかのバンドが見られた。1つの主要なバンドが、SNP rs7903146サンプルにおいては見られたが、混合サンプルでは見られなかった。この特定のバンドをゲルから切り出し、トリプシン消化した後、LC−MS/MS分析を行った。同定したペプチド数に関して、N=10でカットオフした。カットオフ以上の同定したタンパク質を、上から下へペプチドの多い順に表1に列挙する。ここで、我々は3種の多量に存在するタンパク質:顕著に結合したポリ(ADP−リボース)ポリメラーゼ(Parp−1)と、インスリン応答性であったATP依存性RNAヘリカーゼAおよび甲状腺ホルモン受容体関連タンパク質3(Thrap3)とに焦点を当てた。「サンプル4 C PARPバンド110kD ins」を参照。
Parp−1、ATP依存性RNAヘリカーゼA、およびThrap3のSNP rs7903146領域に対する動的結合を調べるため、我々は、細胞培養(SILAC)においてアミノ酸による安定同位体標識を用い(Ong,Blagoev et al.2002)、インスリン処理の有無による結合親和性の変化を定量化した。
Parp−1、RNAヘリカーゼA、およびThrap3は、同じオリゴプルダウンサンプルから同定されたため、我々は、これらの因子がTCF7L2ゲノム機構においてどのように関与しているのかを理解するために、これらの間の相互作用を特徴付けようと試みた。我々は、これらはSNP rs7903146領域に結合する複合体を形成するように互いに相互作用している可能性があると提案した。この仮説を試験するために、免疫共沈降実験を行った。図3Aに示されるように、Parp−1は、RNAヘリカーゼAおよびThrap3と強い相互作用を示した。興味深いことに、我々はまた、TCF7L2がこれらのタンパク質と相互作用することを発見した。我々が以前行ったTCF7L2のchip−seq結果により、TCF7L2ゲノム内の4つのサイトが特徴付けられている(Zhao,Schug et al.2010)。総括して、我々は、TCF7L2が、Parp−1、RNAヘリカーゼA、およびThrap3と相互作用して、複合体を形成し、TCF7L2それ自身を調節するというメカニズムの可能性を提案した(図3B)。これらの相互作用の機能的効果をさらに特徴付けるために、これらのタンパク質のより詳細な研究を行っている。
いくつかの独立した研究により、転写因子7様2遺伝子(TCF7L2)多型rs7903146のT対立遺伝子が、2型糖尿病(T2D)に関与していることが示されている(Grant,Thorleifsson et al.2006,Saxena,Gianniny et al.2006;Scott,Bonnycastle et al.2006,Helgason,Palsson et al.2007;Palmer,Hester et al.2011)。多数の民族での研究により、イントロン3における単一の変異体rs7903146への関与が特定された。これにより、SNP rs7903146は、全体的な共通性により糖尿病の原因である感受性変異体と考えられている。しかしながら、SNPがTCF7L2の機能に作用するメカニズムは分かっていない。我々は、SNP全体に渡って結合し、それによりTCF7L2の機能を調節する対立遺伝子選択的な結合タンパク質があることを提案する。我々の仮説を試験するために、我々は、「双方向の」オリゴプルダウン実験を行った。フォーワード実験において、SILAC培地内で同位体標識したライト細胞からの核抽出物を、C対立遺伝子オリゴによるプルダウン実験に用いた一方、SILAC培地内で同位体標識したヘビー細胞からの核抽出物を、T対立遺伝子オリゴによるプルダウン実験に用いた。当該SILAC標識抽出物を、リバース実験に切り替えた。このデザインは、アプローチの信頼性および再現性を改善することができる。

グルカゴン様ペプチド−1(GLP−1)は、グルコース刺激によるインスリン分泌の促進、β細胞抗アポトーシスの刺激およびグルカゴン分泌の促進と阻害、食物摂取、および胃内容排出を含め、多数の生理学的機能を有する。GLP−1のこれらの抗糖尿病特性は、2型糖尿病に罹患した患者の治療のための、この短いペプチドおよびそのアゴニストの使用に対する強い関心を生んでいる。GLP−1分泌の基本的メカニズムをより良く理解することにより、2型糖尿病の治療に使用される新規のアプローチに繋がる可能性がある。TCF7L2は、2型糖尿病に最も強く関連した遺伝子座であり、プログルカゴン遺伝子のプロモーター領域に結合することが長年にわたり知られている。また、オリゴプルダウンを使用し、続いて質量分析することにより、我々は、これまでの実施例において、原因と考えられる変異体であるTCF7L2内のrs7903146に結合する特定のタンパク質因子(そのうち最も多量なものはPARP−1である)を同定した。興味深いことに、TCF7L2およびPARP−1は共に作用し、PARP−1ノックアウトマウスは、糖尿病の誘発から保護されることを示唆する文献が既にいくつかある。そのため、我々は、腫瘍研究で以前試験した既存のPARP−1阻害剤が、GLP−1アゴニスト作用を有するかどうかを調査した。この関連の可能性を調べるためのGLP−1分泌評価のため、標準ヒトL細胞株であるNCI−H716を使用した。細胞を、3種の異なるPARP−1阻害剤、オラパリブ、ルカパリブ、およびイニパリブを用いて、グルコース刺激(16.8mM)有りと無しとで、48時間前処理した。得られたGLP−1の濃度を、ELISAキット(ミリポア社)を用いて測定した。データにより、GLP−1の基礎レベルがグルコースにより上昇したこと分かる。しかしながら、グルコースは無いが阻害剤が有る場合には、同等またはそれ以上のGLP−1分泌レベルが認められた。また、この分泌の増加は、グルコースの添加によりさらに上昇した。図5を参照。この結果を踏まえると、PARP−1阻害剤が、GLP−1分泌を増加させるため、2型糖尿病の治療のための新規の治療戦略が存在するように思われる。
Claims (10)
- 転写因子7様2(TCF7L2)をコードする核酸を含むSNPと、表Iに列挙されたタンパク質の少なくとも1つとを有する単離結合複合体であって、前記SNPは、rs7903146である単離結合複合体。
- 請求項1記載の単離結合複合体において、前記複合体は、表Iに列挙されたタンパク質の1つ、2つ、3つ、または4つを有する単離結合複合体。
- 請求項2記載の単離結合複合体において、前記1つのタンパク質は、PARP−1である単離結合複合体。
- 請求項2記載の単離結合複合体において、前記1つのタンパク質は、Thrap3である単離結合複合体。
- 請求項2記載の単離結合複合体において、前記1つのタンパク質は、RNAヘリカーゼAである単離結合複合体。
- 上記請求項のいずれか一項に記載の結合複合体を破壊し、それによりTCF7L2の機能を調整する薬剤を同定する方法であって、
a)有効量の前記薬剤の存在下および非存在下で前記複合体をインキュベートする工程であって、前記複合体は、少なくとも1つの検出可能に標識されたタンパク質または核酸を有する、前記インキュベートする工程と、
b)前記薬剤の非存在下でみられる前記結合複合体の破壊に対する、前記薬剤の存在下における前記結合複合体の破壊を測定する工程であって、前記複合体を破壊する薬剤は、TCF7L2の機能の調節因子として同定される、前記測定する工程と
を有する方法。 - 請求項6記載の方法において、前記方法は細胞内で行われ、前記TCF7L2の機能は、Wntシグナリング、クロマチンリモデリング、標的遺伝子の発現の活性化、並びにDNA損傷の検出および修復からなる群から選択される方法。
- 請求項7記載の方法において、前記薬剤は、siRNA、アンチセンスオリゴヌクレオチド、小分子、およびペプチドからなる群から選択される方法。
- 請求項7記載の方法において、前記細胞は、INS細胞、PC12細胞、MIN6細胞、膵島β細胞、およびαTC6細胞からなる群から選択される方法。
- 請求項8記載の方法において、インスリン分泌、グルカゴン分泌、およびグルコサミン誘導β細胞アポトーシスからなる群から選択されるパラメータに対する前記siRNAの調節効果が測定される方法。
