プラスミドのバルク生産は、大腸菌(E. coli)培養から行い、続いてプラスミドの単離、精製および特性決定を行った。精製したバルクプラスミドを、DNAの完全性、OD 260/280比、アガロースゲル分析、制限分析、DNA配列、混入タンパク質およびエンドトキシンについて、当業者の既知の標準法に従い試験した。
続いて抗血清は、各群6匹の2群のウサギを免疫感作することにより生成した。各ウサギに、0.5 mlのDNA物質(すなわちプラスミド)を、試験の0、28および56日に接種した。各用量は1.0 mgの全DNAからなった。1つの群のウサギに、一価の HA DNAを免疫感作し、そしてもう1群のウサギに二価の1:1のHA および NA DNA混合物を免疫感作した。免疫感作前および後の血液サンプルをすべての動物から、0、28、35、42 および56日に採取した。すべての動物は70日目に瀉血により殺した。すべてのウサギは健康で、活動的で、しかも観察期間を通して投薬または試験品に関する問題を示唆する臨床的兆候はなかった。すべてのウサギは健康で、そして試験期間を通して生存し、そして抗原に対するいかなる副作用も示さなかった。

細胞培養物の50%に病理学的変化を生じる病原性物質の量)のヒト パラインフルエンザ3ウイルスの接種に相当した。1つの陽性対応は0日に接種され、そして陽性対照として14日の血球吸着および/または血球凝集試験に使用することができる。
標的細胞に、約1000TCID50のヒトパラインフルエンザ3ウイルスで事前にスパイクした被験サンプルを接種することができる。
陰性対照には、フラスコ毎に各細胞株に特異的な希釈培地を接種できる。この対照は被験サンプルと同じ条件下で処理することができる。
反応した、すなわち中和された可能性があるWSV希釈物は、次いで3種の検出細胞株(Vero細胞 (アメリカンタイプカルチャーコレクション (ATTC)から得た CCL-81;Yasumunra Y. et al., 1962)、MRC-5 細胞 (ATCCから得た CCL-171;Jacobs J. P. et al., 1970) およびMDCK 細胞 (ECACC 84121903; S. H. Darby, 1958))に接種することができる。3 mlの各溶液(希釈培地、被験サンプル)は、各フラスコ中で中和される可能性があるWSVおよび対照に接種することができる。37℃+/−2℃で70分+/−10分後、接種物を取り出すことができる。次いで生存培地(survival medium)を加えて20mlの最終容量を得た。培養フラスコは5+/−0.5%CO2の存在下で37℃+/−2℃に配置することができる。

血球吸着試験は、試験期間の終わりに行うことができる(14日)。単層細胞をPBSバッファーで1、2回洗浄することができる。次いでPBS中に調製した0.4%の3種赤血球(ヒト、モルモット Hartleyおよび雄鶏)を含有する溶液を各ウェルに加えることができる。血
球吸着はウイルス感染に特異的であり、そして1つの種類の赤血球が細胞上に固定された時に陽性となる。5℃+/−3℃で約30分間インキュベーションした後、細胞を顕微鏡検査にかけることができる。次いでプレートを37℃+/−2℃でさらに約30分間インキュベーションした後、2度目の観察を行うことができる。
血球凝集試験は、14日に血球吸着試験で使用したフラスコからの細胞培養物の上清について行うことができる。上清を回収し、そして低速遠心で清澄化することができる。次いで清澄化した上清を6ウェルプレートに入れることができる。次にPBS中に調製した0.25%の3種赤血球(ヒト、モルモット Hartleyおよび雄鶏)を含有する溶液を各ウェルに加えることができる。5℃+/−3℃で約30分後、上清を顕微鏡検査にかけることができる。次いで6ウェルプレートを37℃+/−2℃でさらに約30分間インキュベーションした後、2度目の観察を行うことができる。
欧州薬局方第6版、2.6.16章に従い、血球凝集試験を孵化鶏卵を使用して行うことができる。
WSVはウサギまたはヒツジ抗血清で中和することができる。卵からのバクテリアの混入を回避するために1%抗生物質水溶液を加えることができる。次いでサンプルは適切なシリンジ針を使用して卵に注入することができる。
卵を受け取った時、およびプレインキュベーションの前に、卵を大ざっぱに観察して傷がある卵は捨てることができる。次いで卵は37℃で9〜11日間、プレインキュベーションすることができる。プレインキュベーション期間の終わりに、卵は加熱を回避するために冷光下で観察することができ、そして不受精卵または生きている胚が無い卵を捨てた。
被験サンプル(中和した可能性があるWSV)の分析には、9〜11日のインキュベーション後に30個の卵に尿膜経路を介して接種することができ、そして対照の試験には25個の卵に接種することができる。
(例えば5個の卵)、および2種類のヒツジ抗血清のみ(例えば1種類あたり5個の卵)を卵に接種することができる。
卵殻を消毒した後、殻に穴を空けることができ、そして0.5 mlの被験サンプルを55個の卵に尿膜経路で接種することができる。卵殻の穴を塞ぐことができ、そして卵は37℃+/−2℃で7日間インキュベーションすることができる。
インキュベーション期間の終わりに、血球凝集試験を以下のように行うことができる:流体は遠心(2500 g、10分)により清澄化し、そして血球凝集試験を行うために、200μlの流体を4枚の96ウェルプレート(各ウェルについて50μl)で処理することができる。簡単に説明すると、2枚の96ウェルプレートのウェルに50μlの0.5%赤血球含有溶液 (モルモット; フランスのCharles River Laboratoryから購入)を加えることができ、そして別の2枚の96ウェルプレートに50μlの0.5%赤血球含有溶液(雌鳥)を加えることができる。プレートの半分を5℃+/−3℃でインキュベーションすることができ、他の半分を室温でインキュベーションすることができる。2時間インキュベーションした後、プレートは血球凝集活性について検査することができる。
次いで血清の中和は、付随物質試験に使用することができる。これらの試験は公定要件(欧州薬局方 2.6.16章)に従い行うことができる。これらの試験は、成体マウス、哺乳マウスおよびモルモットを対象として、公定要件、例えば欧州薬局方 2005 2.6.16章(ウイルスシードロット)の要件に従い行うことができる。哺乳マウスで行うことができる試験に関しては、哺乳マウスは被験組成物で1〜2日齢に接種できることに留意されたい。鳥類組織で伝染するウイルスについて、鳥類ウイルスの試験は、欧州薬局方 2005 2.6.16章(ウイルスシードロットおよびウイルス回収物)に記載されているように行うことができる。
3群の例えば10匹の成体CD1マウス(15-20 g、例えばCharles River Laboratoryから購入)を、少なくとも48時間順化することができる。1群は中和血清(30μlを脳内に注射(Ic)、500μlを腹腔内に注射(IP))を受容することができ、1群は観察期間に死亡が発生する場合に備えて維持することができ、そして1群は陰性対照として使用することができる。
1〜2日齢の30匹のマウス(例えば Charles River Laboratoryから購入)に、中和血清を接種することができ、10匹のマウスは陰性対照として使用することができる。各哺乳マウスは、被験血清10μlのIc.および100μlのIPを受容する。マウスは試験期間中、定期的に観察されることができる(1日に1、2回)。
9匹のモルモット(350-450 g、例えばフランスのCharles River Laboratoryから購入)から、1群5匹の動物に被験サンプルを5000μl、IP注射した。4匹の動物は、陰性対照として42日の試験期間観察することができる。試験期間中、ウイルス感染の兆候が検出されない場合(すなわち死亡または肉眼による損傷)、被験サンプル中にウイルスは存在しない。
態様1:少なくとも1種の活性物質を含んでなる組成物中の外来物質の試験方法において、a)活性物質の少なくとも一部をコードする配列を含んでなるポリヌクレオチド構築物の発現産物に対して生じた抗体と、少なくとも1種の活性物質を含んでなる組成物とを接触させる工程であって、抗体が活性物質に結合する工程、およびb)工程a)の後に組成物中の外来物質の存在または不存在を決定する工程、を含んでなる、上記方法。
態様2:工程a)が:a−1)活性物質の少なくとも一部をコードする配列を含んでなるポリヌクレオチド構築物の発現産物に対して生じた抗体を準備する工程、およびa−2)該抗体を、少なくとも1種の活性物質を含んでなる組成物と接触させる工程、を含んでなる態様1に記載の外来物質の試験方法。
態様3:工程a)が:a−1)個体にポリヌクレオチド構築物を免疫感作することにより生成された抗体を準備する工程、およびa−2)該抗体を、少なくとも1種の活性物質を含んでなる組成物と接触させる工程、を含んでなる態様1に記載の外来物質の試験方法。態様4:工程b)の前に活性物質が抗体に結合することにより中和または不活性化される態様1ないし3のいずれかに記載の方法。
態様5: 組成物中の外来物質の存在または不存在を決定する工程が:a)活性物質の少なくとも一部をコードする配列を含んでなるポリヌクレオチド構築物を接種したヒト以外の動物を使用し、そして該動物に被験組成物を接種し、b)特定期間の後、生きている動物の割合を評価し、ここで接種した動物の少なくとも80%が生存し、かつ該期間中に感染の証拠を示さなかった場合、その組成物は外来物質を含まないと見なされ、そして接種した動物の80%未満が生存し、かつ/または該期間中に少なくとも1匹の動物が感染した証拠を示した場合、その組成物は外来物質を含有すると見なされる、ことを含んでなる態様1ないし3のいずれかに記載の方法。
態様6:組成物中の外来物質の存在または不存在を決定する工程が:a)ヒト以外の動物に、中和または不活性化活性物質を含有する被験組成物を接種し、b)特定期間の後、生きている動物の割合を評価し、ここで接種した動物の少なくとも80%が生存し、かつ該期間中に感染の証拠を示さなかった場合、その組成物は外来物質を含まないと見なされ、そして接種した動物の80%未満が生存し、かつ/または該期間中に少なくとも1匹の動物が感染した証拠を示した場合、その組成物は外来物質を含有すると見なされる、ことを含んでなる態様1ないし4のいずれかに記載の方法。
態様7:ヒト以外の動物が、成体マウス、哺乳マウスおよびモルモットからなる群から選択され、そして生きている動物の割合および感染の証拠の発生が、接種動物が成体マウスの場合には被験組成物の接種から少なくとも7〜10日の期間後、場合により21日の期間後に評価され、接種動物が哺乳マウスの場合には被験組成物の接種から14日の期間後
に評価され、そして接種動物がモルモットの場合には被験組成物の接種から少なくとも42日の期間後に評価される、態様5または6に記載の方法。
態様8:被験組成物の接種が脳内および/または腹腔内に行われる態様5ないし7のいずれかに記載の方法。
態様9:組成物中の外来物質の存在または不存在を決定する工程が、規定の要件、好ましくは欧州薬局方、2005、2.6.16章の要件にしたがって行われる前記態様のいずれかに記載の方法。
態様10:被験組成物が、それぞれ活性物質が生産される細胞培養物のサンプル、または該細胞培養物から誘導される生産物、またはシードウイルスまたはシードウイルスを含有する組成物である前記態様のいずれかに記載の方法。
態様11:組成物が製薬学的組成物、好ましくはワクチン調製物またはその中間産物である、前記態様のいずれかに記載の方法。

Claims (1)
- 少なくとも1種の活性物質を含む組成物中の外来物質の試験方法において使用するための試験キットであって、必須成分として
a)活性物質の少なくとも一部を形成するポリヌクレオチドコードする配列を含むポリヌクレオチド構築物を含んでなり、かつ、
b)該組成物中の外来物質の存在または不存在を決定する前に、該ポリヌクレオチド構築物が、それを用いてヒト以外の個体の免疫感作により、該ポリヌクレオチドの発現産物に対して抗体を生じさせるために使用される、
試験キット。
