実施直後の生検採取物(内視鏡または経皮的な方法によって行ったTAC)を、本発明の組成物(50%のアルコール混合物+50%の2-エチルヘキシルベンゾエート)中に1時間(試料が既にホルマリン中に固定されているか否かにかかわらず30分間)直ちに浸漬する。全プロセシングは、30分の固定、本発明の組成物対象中でそれぞれ約7分の3工程、58℃で融解したパラフィン中でそれぞれ約5分の2工程を含む。したがって、試料をパラフィン中に包埋し、ミクロトームによって切断する。得られた切片を60℃で約20分間ストーブ上に置く。後に、ヘマトキシリンおよびエオシンで予備染色を実施する(スライドのアセンブリングを含んだ染色段階は、約1時間の標準継続時間を有する)。したがって、スライドを顕微鏡下で眺める。
手術室で回収した直後の組織試料を、上記で述べたサイズまで小さくし、約60分の3工程で、本発明の組成物(50%のアルコール混合物+2-エチルヘキシルベンゾエート)中に3時間浸漬する。
Claims (11)
- a)2-エチルヘキシルベンゾエート、2-エチルヘキシルパルミテート、2-エチルヘキシルココエート、2-エチルヘキシルステアレート、2-エチルヘキシルアセテートからなる群から選択される少なくとも1種の2-エチルヘキシルエステル;b)エチルアルコールおよび/またはイソプロピルアルコールを含み、またはこれらからなる組成物。
- 2-エチルヘキシルエステルの濃度が30〜70%の範囲であり、エチルアルコールの濃度が20〜60%の範囲であり、イソプロピルアルコールの濃度が10〜30%の範囲であり、前記百分率が組成物の全体積に対する体積百分率である、請求項1に記載の組成物。
- 2-エチルヘキシルエステルの濃度が50%であり、35%のエチルアルコールおよび15%のイソプロピルアルコールであり、前記百分率は、組成物の全体積に対する体積百分率である、請求項1から2のいずれか一項に記載の組成物。
- 前記少なくとも1種の2-エチルヘキシルエステルが、55〜70%の濃度のグルタル酸ジイソブチルまたはグルタル酸ジメチル、10〜30%の濃度のアジピン酸ジイソブチルまたはアジピン酸ジメチル、および10〜25%の濃度のコハク酸ジイソブチルまたはコハク酸ジメチルの混合物によって置き換えられ、前記百分率は、ジイソブチルまたはジメチルのグルタル酸エステル、アジピン酸エステル、およびコハク酸エステルの混合物の全体積に対する体積百分率である、請求項1から3のいずれか一項に記載の組成物。
- 生体試料プロセシングのための請求項1から3または4のいずれか一項に記載の組成物の使用。
- 生体試料が、組織学的試料、細胞学的試料、検死試料からなる群から選択される、請求項5に記載の使用。
- 生体試料が、顕微分析、免疫組織化学的分析、ISH、FISH、CISH、PCRの分子生物学技法に基づく分析にかけられる試料である、請求項5から6のいずれか一項に記載の使用。
- 請求項1から3または4のいずれか一項に記載の組成物での生体試料処理の段階を含み、またはそれからなる、生体試料プロセシングのための方法。
- 前記処理が、30℃〜80℃の範囲の温度、または室温で行われる、請求項8に記載の方法。
- 前記生体試料が、組織学的試料、細胞学的試料、検死試料からなる群から選択される、請求項8から9のいずれか一項に記載の方法。
- 生体試料のダイアファニゼーション剤としての、2-エチルヘキシルベンゾエート、2-エチルヘキシルパルミテート、2-エチルヘキシルココエート、2-エチルヘキシルステアレート、2-エチルヘキシルアセテート、2-エチルヘキシルベンゾエートからなる群から選択される少なくとも1種の2-エチルヘキシルエステルの使用。
