Abstract

関節軟骨の完全性の維持は、一般に、任意の治療の効果を評価する上で測定するためのOAの重要な側面とみなし、OAの疾患進行を評価する重要な測定とみなされる。単純X線写真における骨内の距離は、関節軟骨の厚さの利用可能な最善の代用測定である。膝の範囲内の関節空間の喪失は、関節軟骨の喪失と等しく扱われている。したがって、関節の異なる構成物間の空間の量を測定することによってこの変質を定量化することは医師によく知られている。関節空間の狭小化は、変形性関節症の悪化を示す。関節空間狭小化(JSN)はしばしば、連続的な放射線学的実験における関節の放射解剖学的見方を標準化するために評価項目として利用される。

対立遺伝子の検出

Claims (9)
- IL1RN rs9005 G>Aの遺伝子型G/Gを、対象における変形性関節症の重症疾患進行および関節空間狭小化のリスクが増加する素因を検出するための指標として使用する方法であって、該対象から得た核酸試料において前記IL1RN rs9005 G>Aの遺伝子型G/Gを検出する工程を含み、遺伝子型G/Gが存在することは、該対象に前記重症疾患進行および関節空間狭小化の素因があることを示し、この遺伝子型が存在しないことは、該対象にその素因がないことを示す方法。
- IL1RN rs9005 G>Aの遺伝子型A/AまたはG/Aを、対象における変形性関節症の重症疾患進行および関節空間狭小化のリスクが減少する素因を検出するための指標として使用する方法であって、該対象から得た核酸試料において前記IL1RN rs9005 G>Aの遺伝子型A/AまたはG/Aを検出する工程を含み、遺伝子型A/AまたはG/Aが存在することは、該対象が、重症疾患への進行および関節空間狭小化から保護されていることを示し、これらの遺伝子型が存在しないことは、該対象がこれらから保護されていないことを示す方法。

- 前記検出工程が、a)対立遺伝子特異的オリゴヌクレオチドハイブリッド形成;b)サイズ分析;c)配列決定;d)ハイブリッド形成;e)5'ヌクレアーゼ分解;f)一本鎖の高次構造多型;g)対立遺伝子特異的ハイブリッド形成;h)プライマー特異的伸長;およびi)オリゴヌクレオチドライゲーションアッセイからなる群から選択される、請求項1から7のいずれか一項に記載の方法。
- 検出前に、または検出と併せて、前記核酸試料を増幅工程に供する、請求項1から8のいずれか一項に記載の方法。
