Abstract
Description
一方、慢性炎症は、動脈硬化症、アトピー性皮膚炎(Atopic dermatitis:AD)及び肥満など先進国で増加しつつある疾患に於いて潜在し治療の標的となり得る。基礎となる炎症は、異常な脂肪組織のリモデリングに関与することで、肥満と脂肪異栄養症(非特許文献3:先行技術文献については、末尾に纏めて示す。)の両方の病因に寄与する。炎症性サイトカインのIL-1を阻害する治療法に関する臨床試験が、アテローム性動脈硬化症(非特許文献4)とADに関する臨床試験で検討中である(http://clinicaltrials.gov/ct2/show/NCT01122914)。
表皮角化細胞は、サイトカインカスケードを活性化し、炎症反応の引き金となるマスターサイトカインであるIL-1α、及びIL-1βの前駆体を提供する貯蔵槽となっている。IL-1αは皮膚炎と引っ掻きによって生産・分泌され、傷害を受けた表皮角化細胞に対して、CTL/NKプロテアーゼ・グランチームB(非特許文献5)及びカルシウム活性化プロテアーゼ・カルパイン(非特許文献6)を生じさせる。IL-1βは不活性な前駆体として貯蔵されており、カスパーゼ1/IL-1β変換酵素などの特異的な酵素によって活性化され、放出される。表皮角化細胞から放出されたIL-1は、局所的なメディエータとして作用する。しかし、重症型ADでは、ケラチノサイト由来のIL-1は血流によって循環され、潜在的に遠隔器官に影響を及ぼす高IL-1血症を誘導すると考えられている。このような全身性病態は、臨床的に大きな関連があるが、今のところ未解明のままである。
また、アミロイドーシスは、アミロイド蛋白が全身の臓器の細胞外に沈着する疾患であり、日本では特定疾患(難病)に指定されている。アミロイドーシスの病態については、不明な点が多く、モデル動物が望まれていた。
こうして、上記課題を解決するための第一の発明に係る非ヒト哺乳動物は、内因性IL-1α及び/又はIL-1βを誘導することにより、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患を生じさせることを特徴とする。
本発明者は、皮膚炎等により皮膚が損傷すると発生する現象であって、IL-1α、IL-1βが浸み出す形態で誘発され、これらが白血球を活性化させることで、皮膚炎が増悪する。これにより高用量IL-1投与等と異なり、非ヒト哺乳動物(例えば、マウス)を短期間で衰弱させることなく、長期間の過剰IL-1α、IL-1βを原因とする疾患、例えば動脈硬化症、るい痩、全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患を生じさせ、疾患予防、治療用物質の検証に耐える実験用動物を効率よく育成できる結果となった。
本発明の要旨の一つは、皮膚炎と動脈硬化他の疾患の関係、すなわち皮膚炎という形でIL-1が長期間誘発されることで、長期間に渡ってIL-1α、IL-1βが血管を含む内臓に影響を与え、種々の疾患を発症させる実験動物を育成することを可能にしたことである。
上記発明において、非ヒト哺乳動物は、(1)皮膚特異的に発現する遺伝子プロモータの下流に前駆型カスパーゼ1遺伝子を結合した組み換えDNAを有し、持続的に皮膚炎を生じるトランスジェニック非ヒト哺乳動物、又は(2)皮膚特異的に発現する遺伝子プロモータの下流に副甲状腺ホルモン遺伝子リーダーシークエンス及びIL-18遺伝子を結合した組み換えDNAを有し、血中に持続的に成熟型IL-18を分泌し、持続的に皮膚炎を生じるトランスジェニック非ヒト哺乳動物であることが好ましい。
第三の発明に係る閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患の予防又は治療用物質のスクリーニング方法は、上記いずれかに記載の非ヒト哺乳動物に被験物質を投与し、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患の改善効果を検定することを特徴とする。
第四の発明に係る抗IL-1抗体は、閉塞性動脈硬化、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患の予防又は治療用であることを特徴とする。
また、別の発明に係る実験用非ヒト哺乳動物は、内因性IL-1α及び/又はIL-1βを誘導する非ヒト哺乳動物を、生後一定期間育成し、皮膚炎発症を観察し、皮膚炎発症後にCTスキャン観察、体重観察、血圧観察、体外における体脂肪培養観察、血中アミロイドAタンパク質濃度観察の少なくともいずれか一つの観察結果を用いて、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患の実験用として選別されたことを特徴とする。このとき、実験用非ヒト哺乳動物の育成期間中、定期的に外的刺激を与えて育成された事が好ましい。また、外的刺激がテープストリーピングまたは、薬品塗布のうちの少なくともいずれか一つであることが好ましい。
「IL-1」(インターロイキン-1:Interleukin-1)は、サイトカインと称される生理活性物質の一種であり、最初に同定されたインターロイキン分子である。IL-1は、炎症反応に深く関与することから、炎症性サイトカインに含まれる。現在のところ、IL-1には、IL-1αとIL-1βの2種類が同定されており、これら2種類の分子が同一のインターロイキン-1受容体に結合して生理作用を発現することが分かっている。本発明において、IL-1と言うときは、IL-1α若しくはIL-1βのいずれか、又は両方を意味する。
「閉塞性動脈硬化症」とは、主に下肢の大血管が慢性に閉塞することによって、軽い場合には冷感、重症の場合には下肢の壊死にまで至ることがある病気を意味し、特に50歳以降の男性に多いことが知られている。
「るい痩」とは、脂肪組織が病的に減少した症候を意味し、痩せの程度が著しい状態をいう。病的であることを強調するために、るい痩症、症候性やせということがある。脂肪組織が過剰に蓄積した肥満症、又は症候性肥満と反対にある概念である。
「皮膚特異的に発現する遺伝子プロモータ」としては、ケラチン1、ケラチン5、ケラチン10、ケラチン14などの遺伝子プロモータを利用できる。このうち、ケラチン14プロモータを用いることが好ましい。
上記「(1)皮膚特異的に発現する遺伝子プロモータの下流に前駆型カスパーゼ1遺伝子を結合した組み換えDNAを有し、持続的に皮膚炎を生じるトランスジェニック非ヒト哺乳動物」としては、例えば、外来性のヒト前駆型カスパーゼ1遺伝子とケラチン14プロモータとを含む組み換えDNAを有し、持続的にアトピー性皮膚炎を生じるトランスジェニック非ヒト哺乳動物であるトランスジェニックマウス(特許文献1に開示のもの)を用いることができる。このトランスジェニックマウスでは、少なくとも3ヶ月の飼育により、皮膚炎を起こし、閉塞性動脈硬化等の疾患を生じる。
また、「(2)皮膚特異的に発現する遺伝子プロモータの下流に副甲状腺ホルモン遺伝子リーダーシークエンス及びIL-18遺伝子を結合した組み換えDNAを有し、血中に持続的に成熟型IL-18を分泌し、生後1年以降に持続的に皮膚炎を生じるトランスジェニック非ヒト哺乳動物」としては、例えば、ケラチン14プロモータの下流に副甲状腺ホルモン遺伝子リーダーシークエンス及び外来性のIL-18遺伝子を結合した組み換えDNAを有し、血中に持続的に成熟型IL-18を分泌し、生後1年以降に持続的にアトピー性皮膚炎を生じるトランスジェニック非ヒト哺乳動物であるトランスジェニックマウス(特許文献2に開示のもの)を用いることができる。
「テープストリーピング」とは、皮膚にテープを貼り、はがす行為を一定回数定期的に実施する刺激を意味する。また、「薬品塗布」に用いられる薬品としては、直接に実験用動物の生命を脅かすことなく、皮膚炎症を起こすものであればよい。このような薬品としては、特に限定されないが、例えば、界面活性剤(SDSなど)、クロトンオイル、漆、生体毒(昆虫、海洋生物などから得られるものなど)などが例示される。
図1から図21は、本願発明実施およびその検証に用いたものであり、下記にまとめて、その構成、効果を説明する。
本出願中の各図において、正常な比較対照群をNormal(またはNorと表記)、ケラチノサイトIL-1α、IL-1β及びIL-18を全身に大量に供給することで皮膚炎のADを発症する二種類のモデルマウス、すなわちケラチン14プロモーターにより発現制御されるカスパーゼ1トランスジェニックマウスをKCASP1Tg(またはKCASPと表記)と称する。さらにケラチン14プロモーターにより発現制御されるマウスIL-18のトランスジェニック・マウスをKIL-18Tgとし、その中でも皮膚炎を伴うKIL-18TgをKIL-18Tg(+)(またはIL-18Tg(+)と表記)と、皮膚炎を伴わないKIL-18TgをKIL-18Tg(-)(またはIL-18Tg(-)と表記)」と示した。
図1は対照群(Normal)と、KCASP1Tg、KIL-18Tg(-)及びKIL-18Tg(+)の各群における体重(BW)と皮膚炎の状態(dermatitis)を示すグラフである。(A)は、対照群、KCASP1Tg及びKIL-18Tg(-)の三群についてのグラフ、(B)は、対照群とKIL-18Tg(+)の二群についてのグラフである。
図2は、CTスキャンによって、内臓脂肪の様子を調べた写真図である。左より、対照群(Normal)、KCASP1Tg、KIL-18Tg(-)及びKIL-18Tg(+)の様子を示す。KCASP1TgとKIL-18Tg(+)では、正常対照マウス又はKIL-18Tg(-)に比べ、内臓脂肪が劇的に減少することが分かった。KCASP1TgとKIL-18Tg(+)では、皮下脂肪も減少した。このように、1週毎等の定期的なCTスキャンによって、内臓脂肪の様子を調べることによる観察は、実験動物の選別に有用なファクターとなる。
図3は、対照群(Normal)、KIL-18Tg(-)及びKCASP1Tgにおける体脂肪率を測定した結果を示すグラフである。6ヶ月齢の3群について、体脂肪率を比較したところ、他の2群に比べると、KCASP1Tgでは、有意に減少することがわかった。
図5は、対照群(Normal)、KCASP1Tg、KIL-18Tg(-)(IL18(-))及びKIL-18Tg(+)(IL18(+))の各群において、腹部脂肪細胞をH&E染色したときの顕微鏡写真図である。対照群とKIL-18Tg(-)では、腹部脂肪細胞は大きく、ふっくらした形状であった。一方、KCASP1TgとKIL-18Tg(+)では、対照群とKIL-18Tg(-)との比較において、脂肪細胞は小さく、丸かった。単核細胞の浸潤数は4群間で同様であった。
このように、定期的な体脂肪率を測定、腹部脂肪細胞をH&E染色したときの顕微鏡写真観察によっても、実験用に適するかどうかのマウスの育成選別を有効に行うことができる。これらは、るい痩は外観で判断できるが、閉塞性動脈硬化症、又は全身性アミロイドーシスを直接観察することなく、実験用動物を効率的に育成、精度よく対象疾病の予防又は治療用物質の研究に使える動物を提供できることとなる。
図6は、KCASP1Tgに対して、無処置(KCASP1Tg)、抗IL-1α中和抗体(anti-IL-1α)、抗IL-1β中和抗体(anti-IL-1β)、及び抗IL-1α中和抗体と抗IL-1β中和抗体(anti-IL-1α+β)を4週齢から24週齢に渡って腹腔内投与したときの体重変化を示すグラフである。体重減少は、抗IL-1α中和抗体又は抗IL-1β中和抗体の投与によって改善され、両抗体を投与することによって、相加的な効果が認められた。
図8は、培養脂肪細胞に各種の皮膚上清を添加して培養し、14日目にオイルレッドOで染色したときの様子を示す顕微鏡写真図である。図中の記号は、それぞれ、添加なし(Medium)、正常マウス皮膚上清(Normal skin)、KCASP1Tg皮膚上清(KCASP1Tg)、KCASP1Tg皮膚上清を抗IL-1α中和抗体で前処理(KCASP1+anti-IL-1α)、KCASP1Tg皮膚上清を抗IL-1β中和抗体で前処理(KCASP1+anti-IL-1β)、KCASP1Tg皮膚上清を抗IL-1α中和抗体及び抗IL-1β中和抗体で前処理(KCASP1+anti-IL-1α+β)を意味する。
通常の培地で培養したマウス脂肪細胞は、培養14日目には、豊富な脂肪滴が含まれていた。正常皮膚の培養上清を補充すると、オイルレッドOで染色された脂質を含むふっくらした脂肪細胞が減少を示した。この脂肪細胞は、KCASP1Tg 皮膚培養上清によって消滅した。抗IL-1α中和抗体又は抗IL-1β中和抗体でKCASP1Tg皮膚上清を前処理すると、脂肪細胞の抑制効果が改善され、両中和抗体で前処理すると、抑制効果は殆ど見られなかった。
図10は、造影CTスキャンによる結果を示す写真図である。左より、対照群(Normal)、KCASP1Tg、KIL-18Tg(-)(IL-18Tg(-))及びKIL-18Tg(+)(IL-18Tg(+))を示す。KCASP1TgとKIL-18Tg(+)では、正常対照群とKIL-18Tg(-)に比べ、大動脈狭窄が認められた。

図17は、肝臓(Liver)、腎臓(Kidney)及び脾臓(Spleen)を比較する写真図である。左より、対照群(Normal)、KCASP1Tg、KIL-18Tg(-)(IL-18Tg(-))及びKIL-18Tg(+)(IL-18Tg(+))から採取した臓器を示す。KCASP1TgとKIL-18Tg(+)では、臓器は大きくなっており、肉眼観察では”異常”な色を示した。
図18は、肝臓(Liver)、腎臓(Kidney)及び脾臓(Spleen)の重量を比較するグラフである。各グラフについて、左より、対照群(Normal)、KCASP1Tg、KIL-18Tg(-)(IL-18Tg(-))及びKIL-18Tg(+)(IL-18Tg(+))から採取した臓器の平均値を示す。KCASP1TgとKIL-18Tg(+)では、対照群に比べると、各臓器の重量は大きくなった。
図20は、肝臓(Liver)、腎臓(Kidney)及び膵臓(Spleen)の組織学的所見を示す顕微鏡写真図である。各臓器について、HE染色(HE)又はコンゴレッド染色(Congo Red)の結果を示す。また、左より、対照群(Normal)、KCASP1Tg、KIL-18Tg(-)(IL-18Tg(-))及びKIL-18Tg(+)(IL-18Tg(+))から採取した臓器のものを示す。組織学的所見では、組織構造が崩壊しており、肝細胞では肝臓は単調非構造性沈着によって置き換えられ、腎臓では糸球体と尿細管が壊滅的な損傷を受け、脾臓ではリンパ濾胞は明確でなかった。KCASP1Tg とKIL-18Tg(+)では、コンゴレッド染色によって、濃いアミロイド沈着が認められた。
図21は、血中GOT、GPT、BUN及びCreatinine濃度を纏めたグラフである。各グラフにおいて、左より、対照群(Normal)、KCASP1Tg、KIL-18Tg(-)(IL-18Tg(-))及びKIL-18Tg(+)(IL-18Tg(+))の平均値を示す。KCASP1TgとKIL-18Tg(+)では、若干の肝障害を示し、また腎機能障害が認められた。
上記の各図説明と重複する部分もあるが、以下に本発明の実施及び検証する実験について説明する。
1.トランスジェニックマウス
K14プロモーターを含有し、ケラチノサイト特異的にヒトカスパーゼ1遺伝子を過剰に発現するトランスジェニックマウス(KCASP1Tg)を本研究に使用した(非特許文献1)。また、ケラチノサイト特異的にマウスIL-18を発現するトランスジェニックマウス(KIL-18Tg)を使用した(非特許文献2)。C57BL/6同腹子を対照マウスとした。KIL-18Tgを除く動物については、6ヶ月齢まで観察した。KIL-18Tgについては、慢性皮膚炎は1年齢以降に発症するため、長期間に渡って観察した。動物は、倫理ガイドラインに沿って飼育され、発明者の属する組織における動物飼育と使用のための審査委員会によって承認された。
皮膚変化と体重測定は、2週間毎に実施した。皮膚病変及び全身表面積は、ルーセント・プラスチックフィルム上にマークすることによって評価し、全身面積に対する百分率として示した(各群についてn=10)。皮膚変化と体重測定は図1に示す。
3.CT及び3D解析
イソフルレン(アボット社製)吸入による全身麻酔下にて、マイクロX線CT(理学社製)を用いてCT撮影を行った。データはi-VIEW-Rソフトウエア(理学社製)にて解析した。体脂肪率を計算し、3Dグラフを得た。CT撮影結果は図2、図10に、3Dグラフは図12に示す。
4.血中サイトカイン濃度
KIL-18Tgの長期間観察については、18ヶ月齢の血液サンプルを採取し、その他のマウスについては、6ヶ月齢の血液サンプルを採取した(各群についてn=10)。血中サイトカイン濃度は、特異的ELISAキット(IL-1α、IL-1βについてはR&Dシステムズ社製、IL-18についてはMBL社製)を使用し、添付されたマニュアルに従って測定した。

6.中和抗体による処理
1ヶ月齢のKCASP1Tgに対して1週間毎に10μgの抗IL-1α中和抗体及び/又は抗IL-1β中和抗体(バイオレジェンド社製)を腹腔内投与し、6週齢から2週間毎に身体的変化を観察した。PBSにて処理したCASP1Tg同腹子を対照群(各群についてn=7)として用いた。
7.組換えタンパク質の腹腔内投与
正常マウスには、6週齢から10週間に渡って毎週3回ずつ、1μgの組換えIL-1α又は組換えIL-1β(バイオレジェンド社製)のいずれかを腹腔内投与し、身体的変化を正常PBS処理マウスと比較した(各群についてn=6)。
マウス胚線維芽脂肪細胞様の3T3-L1細胞株(ATCCより入手)を既報(非特許文献16)に従って10日間培養し、成熟脂肪細胞に分化させた。培地には、通常マウスの皮膚上清、KCASP1Tgの皮膚上清、抗IL-1α中和抗体及び/又は抗IL-1β中和抗体で処理したKCASP1Tgの皮膚上清を添加した。皮膚上清の作成は6ヶ月齢の正常マウス又はKCASP1Tgマウスから1cm2の皮膚を採取し、ハサミで細分化した後、10% FCS、2mM L-グルタミン、100U/mLペニシリン及び100μg/mLストレプトマイシンを含有するRPMI1640培地(2mL)を注いだ24穴培養プレート(コスター社製)にて皮膚を培養した。皮膚培養上清に1μgの抗IL-1α中和抗体及び/又は抗IL-1β中和抗体(バイオレジェンド社製)を添加し、2時間培養し前処置をした。培養14日目にPBSで培養細胞を洗浄し、オイルレッドO(シグマ・アルドリッチ社製)とヘマトキシリンで染色した後、顕微鏡下で観察した(各群についてn=6)(図8)。
既報(非特許文献17)に示すように、ペントバルビタール(50mg/kg体重、腹腔内投与)にてマウスを麻酔し、腹部動脈を採取後、脂肪と結合組織を取り除き、1mm長さのリング状に切断した。20mL容の容器に115mM NaCl、4.7mM KCl、2.5mM塩化カルシウム、1.2mM MgCl2、25mM炭酸水素ナトリウム、1.2mM KH2PO4及び10mMデキストロースを含む改良クレブス・ヘンゼライト溶液(室温)を注ぎ、リングを2個のステンレス製フックによって容器内に垂直に支持し、アイソメトリック・フォース・トランスデューサ(TB-651T;日本光電社製)によって張力を測定した。等尺性張力の変化を張力・変位トランスデューサ(TB-651T;日本光電社製)とキャリア増幅器(EF601G;日本光電社製)で測定し、ペンレコーダ(WT-645G;日本光電社製)で記録した。容器中の溶液は37℃に維持し、5%二酸化炭素を含む空気を連続的に供給した。大動脈リングは洗浄後に30分間平衡化した。張力を測定するため、大動脈リングをスナップし、等尺性張力を徐々に増加した(n=12)(図14)。
既報に示すように(非特許文献18)、意識下の6ヶ月齢マウスの血圧をBP-98A尾部カフ・システム(ソフトロン社製)にて測定し、末梢循環をサーモグラフィ(FLIRシステムズ社製)にて測定した。血圧と末梢循環は、18ヶ月齢のKIL-18Tg(+)でも測定した(各群について、n=8)血圧は図15、サーモグラフィ結果は図16に示す。
11.統計解析
統計解析は、フリードマン検定を用いて行った。KIL-18Tg(+)の長期的な観測の解析と張力試験スナップの解析には、t検定を用いた。5%未満の危険率(p<0.05)を有意とした。グラフの作成、及び統計的解析については、Graphpad prism 6.0を用いた。
本発明者は、ケラチノサイトIL-1α、IL-1β及びIL-18を全身に大量に供給することで皮膚炎のADを発症する二種類のモデルマウス(すなわち、ケラチン14ドリブン・カスパーゼ1(KCASP1Tg)(非特許文献1)及びマウスIL-18トランスジェニック・マウス(KIL-18Tg)(非特許文献2))を用いた。KCASP1Tgは、遺伝子導入されたカスパーゼ1、あるいはカスパーゼ1様酵素によって、表皮角化細胞IL-1βを分泌する(非特許文献7−9)。また皮膚炎が生じ表皮角化細胞が損傷することにより、角化細胞中のIL-1αが放出される。KCASP1Tg は、生後8週目より眼窩周囲病変から、びらん性皮膚炎を発症する。皮膚炎は、徐々に顔全体及び胴体に広がり、5ヶ月で体表面の約15%にまで及ぶ。KCASP1Tg では、10週齢から皮膚炎の進展に伴って、体重減少が始まるのに対し、同時期のKIL-18Tg では皮膚炎を伴わず体重減少は見られなかった(図1)。生後1年目には、KIL-18Tgは皮膚炎を発症し、その後に体重減少を起こした。CTスキャンによれば、KCASP1Tg と皮膚炎を伴うKIL-18Tg (KIL-18Tg(+))では、皮膚炎と共に体細胞が劇的に減少していたが、皮膚炎を伴わないKIL-18Tg (KIL-18Tg(-))では、そのような減少は見られなかった(図2,図3)。血中IL-1α濃度及び血中IL-1β濃度は、KCASP1Tg とKIL-18Tg(+)では、対照群及びKIL-18Tg(-)に比べて有意に増加した。血中IL-18濃度は、KCASP1Tg では増加し、若齢のKIL-18Tg においても既に増加していた(図4)。腹部脂肪組織の病理組織学的研究によれば、単核細胞の浸潤数は4群間で差違が認められなかったものの、正常コントロールマウスとKIL-18Tg(-)では、脂肪細胞が大きくかつ膨らんでおり、KCASP1TgとKIL-18Tg(+)では、脂肪細胞は小さく丸かった、(図5)。
データには示さないが、生後6ヶ月齢の動物についての超音波エコー検査によれば、明らかな左心室機能不全は検出されなかった。左心室機能不全は、大動脈狭窄症と大動脈壁の硬さとしなやかさの減少に起因する変化である可能性が高い(図14)。大動脈狭窄症は、末梢灌流にも影響し、尾部での血圧を測定したところ、KCASP1TgとKIL-18Tg(+)では、収縮期及び拡張期の両方で低かった(図15)。
閉塞性動脈硬化、特にアテローム性動脈硬化症におけるIL-1及びIL-18の寄与については、既報がある(非特許文献13)。アテローム性動脈硬化におけるIL-1αとIL-1βの起源として、アテローム病変における単球が疑われている。一般的には、アテローム性動脈硬化症には、アテローム変化と動脈壁の硬化性変化の両方が認められる。また、アテローム性動脈硬化症の殆どの動物モデルは、高脂血症誘発血管アテローム変性に基づいている。興味深いことに、我々のモデルでは、アテローム・プラークは認められなかった。本研究によって、アテローム変性を伴わない動脈硬化症の発症について、IL-1αとIL-1βの直接的な効果が確認された。自己免疫疾患の一つで断続的にIL-1に曝される家族性地中海熱(FMF)において、動脈硬化血管の変化が報告されているが(非特許文献14)、FMFにおける血管変化では、アテローム斑は殆ど認められていない。本研究では、血中IL-18濃度は、KIL-18Tgの表現型の発症前には既に高値となったため、KIL-18Tgの硬化症の発症はIL-18ではなくIL-1αとIL-1βの影響によることが強く推測される。自己免疫疾患における血管変化に関する更なる研究が望まれる。
このことにより、上記した非ヒト哺乳動物に被験物質を投与し、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスの改善効果を検定することを特徴とする閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスの予防又は治療用物質のスクリーニングを効率よく実施することを可能とし、一例として、閉塞性動脈硬化、るい痩、又は全身性アミロイドーシスの予防又は治療用に抗IL-1抗体が有効であることも開示した。
要約すると、持続炎症を起こしている皮膚から供給されるIL-1が、硬化血管の変化とアディポサイトカインのシステムを変更し、全身性アミロイドーシスと全身るい痩を引き起こす。今回の研究では、AD、メタボリックシンドローム、及び自己炎症性疾患におけるIL-1の生物学的役割に新たな知見を提供している。また、本研究のデータは、抗IL-1療法の有効性を強く支持している。
このように本実施形態によれば、閉塞性動脈硬化症モデル動物、抗肥満モデル動物、及び全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患モデル動物などを提供できた。これらの動物は、IL-1の増加によって特異的な症状を発症することが分かったので、この作用に対抗する作用を示す物質を検索することで、上記疾患の予防又は治療用物質をスクリーニングできる。
Claims (11)
- 内因性IL-1α及び/又はIL-1βを誘導することにより、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患を生じさせることを特徴とする非ヒト哺乳動物。
- (1)皮膚特異的に発現する遺伝子プロモータの下流に前駆型カスパーゼ1遺伝子を結合した組み換えDNAを有し、持続的に皮膚炎を生じるトランスジェニック非ヒト哺乳動物、又は(2)皮膚特異的に発現する遺伝子プロモータの下流に副甲状腺ホルモン遺伝子リーダーシークエンス及びIL-18遺伝子を結合した組み換えDNAを有し、血中に持続的に成熟型IL-18を分泌し、持続的に皮膚炎を生じるトランスジェニック非ヒト哺乳動物であることを特徴とする請求項1に記載の非ヒト哺乳動物。
- 非ヒト哺乳動物に対して、IL-1α及び/又はIL-1βを投与することにより、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患を生じさせることを特徴とする実験用非ヒト哺乳動物の作製方法。
- 請求項1〜請求項3のいずれかに記載の非ヒト哺乳動物に被験物質を投与し、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患の改善効果を検定することを特徴とする閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患の予防又は治療用物質のスクリーニング方法。
- 閉塞性動脈硬化、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患の予防又は治療用の抗IL-1抗体。
- 内因性IL-1α及び/又はIL-1βを誘導する非ヒト哺乳動物を、生後一定期間育成し、皮膚炎発症を観察し、
皮膚炎発症後にCTスキャン観察、体重観察、血圧観察、体外における体脂肪培養観察、血中アミロイドAタンパク質濃度観察の少なくともいずれか一つの観察結果を用いて、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患の実験用として選別された実験用非ヒト哺乳動物。 - 前記実験用非ヒト哺乳動物の育成期間中、定期的に外的刺激を与えて育成された事を特徴とする請求項6記載の実験用非ヒト哺乳動物。
- 前記外的刺激がテープストリーピングまたは、薬品塗布のうちの少なくともいずれか一つであることを特徴とする請求項7記載の実験用非ヒト哺乳動物。
- 内因性IL-1α及び/又はIL-1βを誘導する非ヒト哺乳動物を、生後一定期間育成し、皮膚炎発症を観察し、皮膚炎発症後に
CTスキャン観察、体重観察、血圧観察、体外における体脂肪培養観察、又は血中アミロイドAタンパク質濃度観察からなる群から選択される少なくともいずれか一つの観察結果により、閉塞性動脈硬化症、るい痩、又は全身性アミロイドーシスからなる疾患群のうちの少なくとも一つの疾患を発症した被検動物を選別する、実験用非ヒト哺乳動物の育成方法。 - 前記実験用非ヒト哺乳動物の育成期間中、定期的に外的刺激を与えて育成する事を特徴とする請求項9記載の実験用非ヒト哺乳動物の育成方法。
- 前記外的刺激がテープストリーピングまたは、薬品塗布のうちの少なくともいずれか一つであることを特徴とする請求項10の実験用非ヒト哺乳動物の育成方法。
