Abstract

方法
化学物質および酵素。ハイグロマイシンBをCalbiochem(San Diego,CA)から購入した。すべての他の化学物質を入手可能な最高純度でMerck(Whitehouse Station,NJ)またはSigma(St.Louis,MO)から購入した。DNA制限および修飾用の酵素をNew England Biolabs(Ipswich,MA)およびInvitrogen(Carlsbad,CA)から入手した。オリゴヌクレオチドをIDT(Coralville,Iowa)から入手した。
低分子量の親水性化合物の取り込みに関与するMtbの外膜タンパク質を特定する以前の試みにおいて、ウシ型結核菌BCGのトランスポゾンライブラリを構築し、アンピシリンに耐性を示す変異体についてスクリーニングした(Danilchanka et al.,Antimicrob.Agents Chemother.52:2503−11(2008))。マイコバクテリウム外膜タンパク質の予測のために以前に制定された基準(Song et al.,Tuberculosis 88:526−44(2008))に基づいて特定されたこのライブラリ中に可能性のある外膜タンパク質が存在しなかった。しかしながら、OmpATbは、グラム陰性細菌のポリン(de Keyzer et al.,Cell Mol.Life Sci.60:2034−52(2003))において観察されるように、古典的なSecシグナル配列(Alahari et al.,J.Bacteriol.189:6351−8(2007))を有しないMtbの外膜タンパク質である。したがって、ライブラリ中のアンピシリン耐性変異体を、シグナル配列を除いて既知のポリンの特性(Song et al.,Tuberculosis 88:526−44(2008))を有する仮説上のタンパク質について再度分析した。スクリーニングした変異体のうちの1つ、ML1012は、仮説上のアラニン−プロリン豊富なタンパク質をコードするbcg3960cにおける挿入(図1A)を含む。ウシ型結核菌BCGのBcg3960cは、MtbのRv3903cと同一であり、したがって、簡潔化のために、bcg3960cウシ型結核菌BCGトランスポゾン変異体は、Δrv3903c変異体またはΔMtpA変異体と称される。MtpAの不活性化は、アンピシリンに対するウシ型結核菌BCGの32倍に増加した耐性をもたらす(表4)、(Danilchanka et al.,Antimicrob.Agents Chemother.52:2503−11(2008))。このタンパク質は古典的なSecシグナル配列(シグナルP可能性=0.1未満)(Bendtsen et al.,J.Mol.Biol.340:783−95(2004))を有しないが、さらなる分析は、MtpAが、V型分泌系(Jacob−Dubuisson et al.,Mol.Micriobiol.40:306−13(2001),Szabady et al.,Proc.Natl.Acad.Sci.USA 102:221−6(2005))のいくつかのタンパク質で見つけられた「伸長した」シグナル配列を有することを示した。MtpAの予測されたシグナル配列は、N末端伸長(アミノ酸(aa)1〜20)、続いて、Secシグナル配列に関連する正準のドメイン:帯電ドメイン(aa21〜24)、疎水性ドメイン(aa25〜36)、および切断ドメイン(aa37〜45)からなる。
MtpAが表面会合タンパク質であるかを試験するために、ニトリル誘導性プロモーターの制御下でC末端にHAおよびHisをタグ付けした全長タンパク質およびそのN末端ドメイン(aa1〜444)(ドメイン1)をクローニングし、それらをスメグマ菌mc2155で発現させた。細胞内分画を用いてそれらが膜で局在化されたかを決定するためにこれら両方のタンパク質を試験した。細胞エンベロープ画分(P100)およびその画分を含有する水溶性細胞質タンパク質およびペリプラズムタンパク質(SN100)を全細胞溶解物の超遠心分離(100,000×g)(Song et al.,Tuberculosis 88:526−44(2008))によって分離した。N末端ドメインも全長MtpAもいずれも膜画分P100に会合し、可溶性画分SN100には会合しなかった(図2A)。同様の分画プロファイルが、MspA、OmpATb、およびRv1698等の他の既知のマイコバクテリウム外膜タンパク質において以前に見出されている(Song et al.,Tuberculosis 88:526−44(2008))。P100画分が内膜および外膜タンパク質ならびに膜会合タンパク質を含有するため、MtpAの表面到達性が決定された。この目的を達成するために、MtpAのプロテイナーゼKへの到達性を試験し(Song et al.,Tuberculosis 88:526−44(2008))、これは、プロテイナーゼKがマイコバクテリウム細胞エンベロープに浸透することができないことに基づいている。したがって、細胞外ループを有するタンパク質のみが全細胞に消化される。アッセイの対照として、表面タンパク質PE_PGRS33HA(Cascioferro et al.,Mol.Microbiol.66:1536−47(2007))を過剰発現させ、これはプロテイナーゼKの存在下で著しく消化し、ペリプラズムタンパク質PhoAHA(Siroy et al.,J.Biol.Chem.283:17827−37(2008))を過剰発現させ、これはプロテイナーゼK処理の影響を受けなかった(図2B)。PE_PGRS33HAと同様に、全長MtpAは、プロテイナーゼKで処理したときに大部分が分解し、MtpAが表面到達可能であることを示した。膜会合実験および表面到達性実験の組み合わせは、MtpAが外膜タンパク質であることを示す。対照的に、MtpAのN末端ドメイン(ドメイン1)は、全細胞においてプロテイナーゼKによって分解されなかったが、細胞溶解物をプロテイナーゼKで処理したときに完全に分解された(図2B)。MtpAのN末端ドメインの膜会合に基づいて、タンパク質が外膜に対して標的化されるが、全細胞におけるプロテイナーゼK分解に到達可能な細胞外ループを欠くことを示唆する。
グラム陰性細菌およびスメグマ菌のポリンにおける変異は、唯一の炭素源として使用される際のグルコースおよびグリセロール等の低分子または親水性の溶質の取り込みの減少により、増殖速度の低下をもたらし得る(Liu and Ferenci,J.Bacteriol.180:3917−22(1998)、Stephan et al.,Mol.Microbiol.58:714−30(2005))。増殖の遅いマイコバクテリウムにおける栄養素の取り込みがMtpAに依存的であるかを決定するために、野生型ウシ型結核菌BCGの増殖をΔMtpA変異体と比較して分析した。オレイン酸を唯一の炭素源として使用したときに増殖に差異は観察されず、ΔMtpA変異体における栄養素としての疎水性溶質の利用がウシ型結核菌BCGの野生型株と同一であり、ΔMtpA変異体が固有の増殖欠陥を有しないことを示唆した。炭素源としてグリセロール、グルコース、およびオレイン酸を含有するMiddlebrook 7H9富栄養培地を用いて、わずかの増殖欠陥がΔMtpA変異体において観察された。この培地において損なわれた化合物のΔMtpA変異体における利用を特定するために、様々な炭素源を有する最小Hartmans−de Bond(HdB)培地を用いた。ΔMtpA変異体の増殖は、0.1%グリセロールを唯一の炭素源として添加したときに最小限にとどまった(図3A)。次に、ΔMtpA変異体の観察された増殖遅延がグリセロール輸送の欠如に起因するかを試験した。取り込み実験は、ΔMtpA変異体における[14C]グリセロール蓄積が野生型株と比較して大幅に減少したことを明らかにし、MtpAがウシ型結核菌BCGにおけるグリセロールの取り込みに大きな役割を果たすことを示唆した(図3B)。
MtpAのN末端ドメインもMspAもいずれもΔMtpA変異体の増殖欠陥を補完するという観察に基づいて、このドメインがポリン活性を有すると推測した。ポリンは、小分子の拡散を可能にする水を充填したチャネルタンパク質、および直接脂質膜を通って拡散することができないイオンである(Schulz,Curr.Opin.Struct.Biol.6:485−90(1996))。ポリンを通るイオンの通過を平面脂質二重層アッセイを用いて電気生理学的に測定することができる。この方法は、チャネルタンパク質の平面脂質膜への取り込みを検出し、継時的に測定される膜電流の離散的増加に相当する(Benz et al.,Biochim.Biophys.Acta 511:305−19(1978))。この目的を達成するために、MtpAのN末端ドメイン(aa1〜444)をニトリル誘導性プロモーターの制御下でスメグマ菌で発現させた。MtpAのN末端ドメインを37℃の0.8%のSDSを有するペレット画分から抽出し、Ni+−NTA親和性カラム上で精製し、その後、アニオン交換精製した。測定されたチャネル活性がクマシー染色を用いて検出されなかった他のタンパク質夾雑物によって生じるという可能性を排除するために、MtpAのN末端ドメインに対応するタンパク質バンドをSDS−PAGEゲルから切除し、電気溶出を用いて精製した(図4Aおよび4B)。洗剤を含有する貯蔵緩衝液を脂質二重層に添加したときに挿入事象は検出されず、緩衝液/洗剤は単独でチャネルを形成する固有の能力を有さず、かつチャネル活性を有するいずれの夾雑タンパク質も含有しないことを示した。100ng未満の精製されたMtpAのN末端ドメインの同一の膜への添加は、電流の段階的増加をもたらした(図4C)。記録されたいくつかの異なるタンパク質調製物の孔は100個を超えた。最も高い周波数の単一チャネルコンダクタンスは、4.0±0.2nSであった(図4Dおよび4E)。
スメグマ菌中のポリンの欠失は、ファゴソームに存在する殺菌成分の取り込みの減少によるマクロファージの生存率の改善をもたらす(Fabrino et al.,Microbes Infect.11:868−75(2009)、Purdy et al.,Mol.Microbiol.73:844−57(2009))。マクロファージのMtbの生存にMtpAが果たす役割を定義するために、ヒト急性単球性白血病細胞(THP−1)を野生型ウシ型結核菌BCG、ΔMtpA変異体、または全長MtpAもしくはmspAのいずれかで補完された変異体に感染させた。以前に報告されたように、THP−1細胞は、生きた野生型ウシ型結核菌BCG細菌を感染後7日間にわたって低いレベルで一定に保った(図7A)(Jordao et al.,Cell Microbiol.10:529−48(2008))。対照的に、ΔMtpA変異体は、感染後最初の3日間に急速に死滅した。MtpAで補完したときに、観察された表現型は完全に逆になり、MtpAが栄養素の取り込みおよびマクロファージにおけるウシ型結核菌BCGの生存に必要であることを示した。ΔMtpA変異体におけるMspAの発現は、特に感染の最初の段階中に株の生存率を大幅に低下させ(図7A)、マクロファージにおけるMspAおよびMtpAの機能が大きく異なることを示唆した。
反応性窒素および酸素中間体の生成は、結核感染の制御の役割を果たす(MacMicking et al.,Annu.Rev.Immunol.15:323−50(1997))。以前に、mspポリン遺伝子の不在が一酸化窒素へのスメグマ菌の耐性を与えることが示されている(Fabrino et al.,Microbes Infect.11:868−75(2009))。MtpAが一酸化窒素への増殖の遅いマイコバクテリウムの耐性に重要であるかを調べるために、野生型ウシ型結核菌BCG、ΔMtpA変異体、または全長MtpAもしくはmspAのいずれかで補完された変異体を、一酸化窒素ドナー無水ニトロフェリシアン化ナトリウム(III)(SNP)で処理した。液体Middlebrook 7H9培地中ですべての株をSNPで処理することによって、用量および時間依存的様式で回収されたコロニーの数を減少させた(図8Aおよび8B)。回収された野生型株の細菌の数は、ΔMtpA株と比較して少なくとも100倍減少した(図8B)。ΔMtpA株におけるMtpAの発現は、ウシ型結核菌BCGの一酸化窒素感受性を完全に回復させ、ウシ型結核菌BCGがMtpAの不在下で一酸化窒素に高度に耐性を示すことを示唆した(図8A)。ΔMtpA変異体におけるmspAの過剰発現は、処理の24時間後に株をほぼ完全に除去し(図8A)、したがって、毒性化合物のMspA媒介取り込みが一酸化窒素応力下で生存率の低下につがなるという以前の観察を裏付けた。
グラム陰性細菌におけるポリンの欠如が低レベルの耐性のみを低分子量の親水性薬物に与える(Nikaido,Microbiol.Mol.Biol.Rev.67:593−656(2003))一方で、スメグマ菌中のポリンの喪失は、より高い耐性レベルをもたらす(Danilchanka et al.,Antimicrob.Agents Chemother.52:3127−34(2008))。増殖の遅いマイコバクテリウムにおける薬物の取り込み経路はこれまで特定されていない。しかしながら、ポリンがβ−ラクタム系抗生物質、エタンブトール、イソニアジド、およびストレプトマイシンの取り込みを媒介することが示唆されている(Stephan et al.,Antimicrob.Agents Chemother.48:4163−70(2004))。Mtbの薬物耐性がMtpAによって媒介されるかを決定するために、マイクロプレートアラマーブルーアッセイ(MABA)を用いた。ウシ型結核菌BCGのいくつかの低分子量の親水性抗生物質への耐性(表4、A群)は、MtpAの不在下で大幅に増加した。例えば、32倍および16倍増加した最小阻害濃度(MIC)が、それぞれ、ΔMtpA株中のアンピシリンおよびクロラムフェニコールにおいて観察された一方で、抗結核薬エタンブトール、イソニアジド、およびp−アミノサリチル酸(PAS)への耐性は8倍超増加した。驚いたことに、エリスロマイシンおよびリファンピシン等の高分子量で疎水性の抗生物質のMICもΔMtpA変異体において著しく増加した(表4、C群)。低分子量および/または親水性抗生物質へのMspAまたはMtpAを発現するΔMtpA株の耐性は、野生型ウシ型結核菌BCGの耐性と同一であり(表4、A群、D群)、MtpAがウシ型結核菌BCGをこれらの薬物に対してより感受性の高いものにし、かつMspAおよびMtpAが同様の機構によって機能することを示唆する。
スメグマ菌、大腸菌、および出芽酵母においてMtpAのCTDを生体内で発現させる試みは失敗したが、生体外(細胞を含まない)発現は、微量のタンパク質の回収を可能にした。MtpAのCTDが哺乳類細胞において毒性であるかを試験するために、サイトメガロウイルス(CMV)プロモーターの制御下でHAタグと融合したMtpAのCTDを含有するpRK7系ベクター(Graycar et al.,Mol.Endocrinol.3:1977−86(1989))をヒト293T細胞系に一時的にトランスフェクトした。トランスフェクトしてから24時間後の細胞の顕微鏡検査は、gfp含有ベクターでトランスフェクトした細胞と比較して(図9A)、細胞の大半が死んだ(図9B)ことを明らかにした。細胞死がCTDの活性に関連するものであり、過剰発現のせいではないことを示すために、CTDの非毒性変異体のスクリーニングを実行した。大腸菌におけるCTDの発現が有害であるとして、CTD−Gfp翻訳融合がCTDの点変異またはインフレーム欠失を含むクローンにおいてのみ蛍光性であると仮定した。予想通り、CTDの野生型配列を有する蛍光クローンは得られなかった。しかしながら、点変異を含む4つの異なる変異体(CTDM1〜4)がGfp−蛍光に基づいて特定された:CTDM1(G818E)、CTDM2(A811E)、CTDM3(G752V)、およびCTDM4(G662D/R788L)。変異体CTDM1は、ヒト293T細胞において完全に非毒性であった(図9C)。これらの結果は、MtpAが以前に特徴付けられていない細菌性毒素ドメインを含むことを示し、マイコバクテリウムOMタンパク質が新規の機構によって機能するという仮定を強調する。多くの細菌性毒素の活性は、細胞表面での分泌(I型およびIII型)または切断(V型)を必要とする。これらの機構のどちらがMtpAに適用されるかを試験するために、ウシ型結核菌BCGの全細胞および培養濾液ならびにC末端でHAおよびHisタグ付けしたMtpAを含有するMtb ΔmtpA株を分析した。抗HA抗体を用いて、全長MtpAが全細胞において検出され、約24kDaの切断されたタンパク質が濃縮培養濾液において検出され(図9D)、CTDが全長タンパク質としてOMに移動し、その後、切断され、CFに解放されることを示した。さらに、CTDの切断部位を質量分析によって特定した。OMチャネルドメインおよび毒素ドメインでのMtpAの組織化は、ピロリ菌のVacA等の自己輸送体(Va型分泌)の組織化に類似している(Cover&Blanke,Nat.Rev.Microbiol.3:320−32(2005))が、これらのドメインは、グラム陰性細菌とは異なって組織化される。自己輸送体の類似性がそれらのOMチャネルドメインに限定されるとして、マイコバクテリウムOMの構造がグラム陰性細菌の構造とは大きく異なるため、マイコバクテリウム自己輸送体が今まで見出されていないことは驚くべきことではない。

- ヒト型結核菌ポリンA(MtpA)のカルボキシ末端ドメインを含む単離されたポリペプチドであって、毒素である、単離されたポリペプチド。
- MtpAのカルボキシ末端ドメインが配列番号1のアミノ酸650〜846を含む、請求項4に記載の単離されたポリペプチド。

- ヒト型結核菌(Mtb)感染を有するか、またはMtb感染を発症する危険性のある対象におけるMtb感染を治療または予防する方法であって、前記対象に、ヒト型結核菌によって生成された毒素を中和する第1の作用物質と、前記Mtb感染を治療または予防する第2の作用物質と、を投与することを含む、方法。
