Abstract
Description

MTDは典型的にはもともと疎水性であり、このようなMTDを含むキメラタンパク質は、大腸菌などの産生宿主で発現させた場合、不溶性となり得る。特性の予期せぬ組み合わせを示す構築物を作製することができる。実施例に示されるように、産生細胞宿主内で可溶性のままであるよう十分に親水性であり、産生宿主細胞を横断できないように、構築物を設計することができる。改変された構築物は驚くべきことに、細菌の外側細胞壁を横断するMTD機能を保持して、標的細菌死滅をもたらす。これは、細菌の細胞膜特性(および構造)が細菌の外膜と十分に異なるために、達成され得る。
ポリペプチドまたは他の分子に対するポリアルキレングリコール(PAG)成分の付加は、PAG化と称される。PAG化(典型的にはPEG化)は、ポリペプチドの薬物動態学的特性および薬力学的特性を改善すること、例えば、インビトロおよびインビボの活性および安定性を高める、半減期を延長する、タンパク質分解を減少させる、免疫原性を減少させる、ならびに生物学的利用能および生体内分布を改善することができる。PAG化のために通常標的化されるアミノ酸は、アルギニン、アスパラギン酸、グルタミン酸、チロシン、セリン、スレオニン、ヒスチジン、システイン、およびリジンである。PAG化のために、通常はリジンの側鎖上のε-アミノ基が標的化される。

さらに、細胞壁分解ドメインまたは膜移行ドメインをコードする核酸を提供する。このようなポリヌクレオチドは、CHAPドメイン(特にC末端CHAPドメイン)を有するタンパク質および細胞壁分解活性を有する他のタンパク質を含む、本明細書に記載されるムラリティックタンパク質をコードする。

実施例1. GP36酵素試験構築物
A. ムラリティックドメイン構築物
本発明者らは、シュードモナスファージP134からORF36配列を単離した(SEQ ID NO: 1)。翻訳産物は、SEQ ID NO:2として示される。配列の解析に基づき、ムレイン分解触媒ドメイン断片は、SEQ ID NO:2のアミノ酸737〜875にまたがると同定された。核酸構築物がアミノ酸683〜898をコードするように、アミノ酸683にプロモーターおよび隣接する開始コドンが連結した発現ベクターを構築した。SEQ ID NO: 5を参照されたく、これはSEQ ID NO: 6として翻訳される。このセグメントをGP36 CD断片と命名した。
上記のように、グラム陰性菌の細菌外膜は、細胞の外部環境からのペプチドグリカンへの接近を妨げるが、外膜は、膜の完全性を損なう薬剤の影響を受けやすい。緑膿菌細胞をクロロホルム(外膜を損なう)で処理し、次に精製GP36 CD断片で処理した。GP36 CD断片は、ODの迅速な濃度依存的減少(細菌の溶解)をもたらした。さらなる試験および対照を実施して、死滅の時間経過およびGP36 CDの量の力価測定を用いて活性を確認した。細菌溶解はクロロホルム処理に依存し、ムラリティックドメインが、ひとたびペプチドグリカン層に到達した場合にグラム陰性標的を死滅させ得ることが確認された。
上記のように、大部分のグラム陰性菌のペプチドグリカン層は比較的薄い。GP36 CD構築物を、グラム陰性標的細菌の臨床的に関連した様々な種のペプチドグリカンに対して試験した。同様の実験を、アシネトバクター・バウマニ、肺炎桿菌、大腸菌、ネズミチフス菌、家禽チフス菌(Salmonella gallinarum)、腸炎菌、およびサルモネラ・ダブリン(Salmonella dublin)に対して実施した。GP36 CDは同様に、試験したすべての種のクロロホルム処理細菌の迅速な濃度依存的溶解をもたらした。
A. ムラリティックドメインおよび膜移行ドメインを含むキメラ
通常のグラム陰性細菌標的のペプチドグリカン層に接近させるため、細菌外膜障壁に侵入させるための生理的に適合性のある手段を考慮した。具体的には、侵入活性は、真核細胞に対して顕著な負の影響を及ぼすことなく、細菌外膜に特異的でなければならない。
緑膿菌細胞を精製P225に曝露したところ、付加的な透過剤(例えば、クロロホルム)の非存在下で、迅速な濃度依存的なOD減少(すなわち、細菌溶解)が生じた。さらなる試験および対照を実施して、1時間の時点でタンパク質の量を力価測定して、活性を確認した。異なるCFU低下アッセイを用いて、細菌死滅が観察された。生存グラム陰性菌の死滅により、BPI TMDが、細菌外膜の内側にあるペプチドグリカン層へのGP36酵素ドメインの接近をうまく提供し得ることが実証される。
P225構築物を、グラム陰性標的細菌の臨床的に関連した様々な種のペプチドグリカンに対する活性について試験した。同様の実験を、緑膿菌のさらなる株、大腸菌、クレブシエラ属(多くの抗生物質に対して高度に耐性である)、およびアシネトバクター・バウマニに対して実施したが、そのすべてが効率的に死滅した。

Claims (32)
- 以下を含むキメラポリペプチド:
(i) ビリオン関連ムラリティック(mralytic)酵素由来のムラリティックドメイン(MD)、またはクロロホルム処理した緑膿菌(Pseudomonas aeruginosa)細菌を溶解し得るその変種;および
(ii) SEQ ID NO:4のアミノ酸16〜39、またはDASプロファイルが1.2〜2.6であるその変種の配列を含む膜横断ドメイン(MTD)。 - 1〜100 nmolのMDが、CFU低下アッセイにおいて、クロロホルム処理した107個の緑膿菌細菌のうちの少なくとも50%を溶解する、請求項1記載のキメラポリペプチド。
- 以下を含むキメラポリペプチド:
(i) SEQ ID NO:2のアミノ酸737〜875、またはクロロホルム処理したグラム陰性菌を溶解し得るその変種の配列を含むムラリティックドメイン(MD);および
(ii) 1.5〜3のDASプロファイルを有し、かつ緑膿菌細菌の外膜を横断し得る膜横断ドメイン(MTD)。 - 1〜100 nmolのMTDが107個の緑膿菌細菌のうちの少なくとも50%の膜を横断する、請求項3記載のキメラポリペプチド。
- MDが、SEQ ID NO:2のアミノ酸737〜875、またはメチオニンアミノ酸のうちの1、2、もしくは3個が非メチオニンアミノ酸で置換されている変種の配列を含む、前記請求項のいずれか一項記載のキメラポリペプチド。
- MDが、SEQ ID NO:2のアミノ酸683〜889、またはメチオニンアミノ酸のうちの1〜7個が非メチオニンアミノ酸で置換されている変種の配列を含む、前記請求項のいずれか一項記載のキメラポリペプチド。
- MTDが、SEQ ID NO:4のアミノ酸16〜39、または1〜6個の疎水性アミノ酸が-2もしくはそれよりも低いハイドロパシースコアを有するアミノ酸で置換されている変種の配列を含む、前記請求項のいずれか一項記載のキメラポリペプチド。
- SEQ ID NO:4のV20、V22、およびI24が、-2またはそれよりも低いハイドロパシースコアを有するアミノ酸で置換されている、請求項7記載のキメラポリペプチド。
- MTDのDASプロファイルが2.5未満である、前記請求項のいずれか一項記載のキメラポリペプチド。
- キメラポリペプチドのDASプロファイルが2.5未満である、前記請求項のいずれか一項記載のキメラポリペプチド。
- MTDとMDが、3〜5個の正荷電アミノ酸を含むリンカーによって連結されている、前記請求項のいずれか一項記載のキメラポリペプチド。
- MDがSEQ ID NO:2のアミノ酸737〜875と少なくとも90%同一である配列を含み、かつMTDがSEQ ID NO:4のアミノ酸16〜39と少なくとも80%同一である配列を含む、前記請求項のいずれか一項記載のキメラポリペプチド。
- SEQ ID NO:9、SEQ ID NO:11、およびSEQ ID NO:13より選択される配列と少なくとも90%の同一性を有する配列を含む、前記請求項のいずれか一項記載のキメラポリペプチド。
- SEQ ID NO:11の配列を含む、前記請求項のいずれか一項記載のキメラポリペプチド。
- SEQ ID NO:13の配列を含む、前記請求項のいずれか一項記載のキメラポリペプチド。
- ポリエチレングリコールに付着している、前記請求項のいずれか一項記載のキメラポリペプチド。
- 前記請求項のいずれか一項記載のキメラポリペプチドおよび薬学的に許容される賦形剤を含む薬学的組成物。
- 請求項17記載の薬学的組成物を個体に投与する段階を含む、個体における細菌細胞増殖を阻害する方法。
- 細菌が、緑膿菌、肺炎桿菌(Klebsiella pneumoniae)、大腸菌(Escherichia coli)、肺炎桿菌、アシネトバクター・バウマニ(Acinetobacter baumanii)、ネズミチフス菌(Salmonella typhimurium)、サルモネラ・インファンティス(Salmonella infantis)、赤痢菌属(Shigella)、プロテウス・ミラビリス(Proteus mirabilis)、およびバークホルデリア・タイランデンシス(Burkholderia thailandensis)からなる群より選択される、請求項18記載の方法。
- 前記請求項のいずれか一項記載のキメラポリペプチドおよび酸化を減少させる剤を含む抗菌製剤。
- 請求項1〜16のいずれか一項記載のキメラポリペプチドを環境に適用する段階を含む、環境における細菌細胞増殖を阻害する方法。
- 細菌が、緑膿菌、肺炎桿菌、大腸菌、肺炎桿菌、アシネトバクター・バウマニ、ネズミチフス菌、サルモネラ・インファンティス、赤痢菌属、プロテウス・ミラビリス、およびバークホルデリア・タイランデンシスからなる群より選択される、請求項21記載の方法。
- 以下:
(i) 表Aに収載されるムラリティックドメイン(MD)供給源より選択されるMD、またはその変種;および
(ii)表Bに収載される膜移動ドメイン(MTD)供給源より選択されるMTD、またはDASプロファイルが1.2〜2.6であるその変種
を含むキメラポリペプチドであって、
未処理対照培養物と比較して、グラム陰性菌の培養物のCFUを減少させる、キメラポリペプチド。 - 1〜100 nmolのキメラポリペプチドが、CFU低下アッセイにおいて、107個のグラム陰性菌のうちの少なくとも50%を溶解する、請求項23記載のキメラポリペプチド。
- 細菌が、緑膿菌、肺炎桿菌、大腸菌、肺炎桿菌、アシネトバクター・バウマニ、ネズミチフス菌、サルモネラ・インファンティス、赤痢菌属、プロテウス・ミラビリス、およびバークホルデリア・タイランデンシスからなる群より選択される、請求項23または24記載のキメラポリペプチド。
- MTDが、SEQ ID NO:4のアミノ酸16〜39;SEQ ID NO:15のアミノ酸242〜264;SEQ ID NO:17のアミノ酸242〜271;SEQ ID NO:19のアミノ酸220〜406;SEQ ID NO:21のアミノ酸220〜400;SEQ ID NO:23のアミノ酸220〜885;およびDASプロファイルが1.2〜2.6であるそれらの変種からなる群より選択される配列を含む、請求項23〜25のいずれか一項記載のキメラポリペプチド。
- SEQ ID NO:9、11、13、15、17、19、21、および23からなる群より選択される配列と少なくとも90%の同一性を有する配列を含む、請求項23〜26のいずれか一項記載のキメラポリペプチド。
- ポリエチレングリコールに付着している、請求項23〜27のいずれか一項記載のキメラポリペプチド。
- 請求項23〜28のいずれか一項記載のキメラポリペプチドおよび薬学的に許容される賦形剤を含む薬学的組成物。
- 請求項29記載の薬学的組成物を個体に投与する段階を含む、個体における細菌細胞増殖を阻害する方法。
- 請求項23〜28のいずれか一項記載のキメラポリペプチドおよび酸化を減少させる剤を含む抗菌製剤。
- 請求項23〜28のいずれか一項記載のキメラポリペプチドを環境に適用する段階を含む、環境における細菌細胞増殖を阻害する方法。
