Description
NK1、NK2およびNK3のタイプのNK受容体(物質P(Substance P)およびタキキニンのニューロペプチド受容体)は、体細胞において広く分布される。それらの存在は、哺乳動物の中枢神経系および末梢神経系において、消化系、循環系、造血細胞および炎症細胞及び/又は免疫反応細胞において、また軟組織、特に血管内皮において示された。NK1、NK2およびNK3の受容体がそれらの調節に関係する、複数の生物学的プロセスが現在知られている。
1. NK1、NK2およびNK3の受容体は、人体に広く拡散される。
2. タキキニンおよび特に物質Pは、NK1受容体に作用する。
3. 非ペプチドNK1受容体アンタゴニストは、うつ病、精神病および不安症などの、いくつかの中枢神経系障害の処置のための薬剤の製造に使用することができ、これは、いくつかの特許出願(WO 95/16679、WO 95/18124、WO 95/23798、およびWO 01/77100)における請求項の対象であった。

下の表1は、NK1、NK2及び/又はNK3の受容体抗体が作用する、特異的な商用腫瘍細胞株の選択を詳述する。これらの商用細胞株を、異なるマーケティング会社、DSMZ(Deutsche Sammlung von Mikrorganismen und Zellkulturen)、ICLC(Interlab Cell line collection −CBA− Genoa)、およびECACC(European Collection of Cell Cultures)によって与えられた説明書に従って培養した。簡潔に言うと、商用腫瘍細胞株を、熱による処置によって不活性化された10%のウシ胎児血清(FBS)を補足された、DMEM培地(イーグル基礎培地を単純に変更したものである、ダルベッコ変法イーグル培地)またはRPMI 1640(Roswell Park Memorial Institute)(Gibco, Barcelona, Spain)における培養中で維持した。さらに、1%の抗生物質−抗真菌剤(Invitrogen)を培地に加えた。細胞株を、75−cm2の表面積を有する培養瓶において播種した(Falcon, Heidelberg, Germany)。培地を、2日ごとに更新した。細胞を、37℃の温度で培養チャンバーにおいて、および95%の空気および5%のCO2を有する湿潤雰囲気においてインキュベートした。細胞播種の6日後に、これらを、トリプシンによる処置によって集めた(カルシウムまたはマグネシウムのない0.05および0.02%のEDTA;Sigma−Aldrich, Madrid, Spain)。他に明記のない限り、試験を行う前夜に、すべての細胞を血清から取り除いた。すべてのこれらの細胞株において、NK1、NK2およびNK3の受容体の存在を確認し、これによって、抗NK1、抗NK2および抗NK3の抗体の抗腫瘍作用が証明される。
ウェスタンブロットアッセイに関して、各商用細胞株のサンプルを、表1に示すように試験した。商用の及びヒトサンプル(一次培養)から直接得たすべての細胞株が、NK1、NK2およびNK3の受容体の存在を証明したと確証するために、この技術を使用した。簡潔に言うと、総タンパク質の抽出は、腫瘍培養細胞から得られたサンプルから実行された。細胞を、先行技術に一般に知られている方法によって溶解し、それらの濃度を、製造業者の説明書に従って、Bio−Rad (Bio−Rad Laboratories, S.A. Madrid)からの市販キット「Protein Assay」を使用して測定した。各サンプルに対して、50mgのタンパク質を、10%のゲル電気泳動法のSDSポリアクリルアミドによって分離し、ポリフッ化ビニリデン(PVDF)細胞膜に電気泳動的に移した。これらの細胞膜を、ブロッキング溶液(0.1%のTween−PBSTを有するリン酸緩衝液(PBS)中の5%のスキムミルク)中で一晩インキュベートし、その後、PBST緩衝液中で1/4000に希釈された一次抗体によって一晩インキュベーションし:
抗NK1である抗体は、製品番号S8305下で、会社Sigma−Aldrichから得られた、アミノ酸393−407(配列番号1)におけるNK1受容体のカルボキシル末端領域に相当するドメインを認識する。
抗NK2は、製品番号HPA012084下で、Sigma−Aldrichから得られたNK2受容体の配列番号2の配列を認識する。
抗NK3は、製品番号HPA009418下で、Sigma−Aldrichから得られたNK3受容体の配列番号3の配列を認識する。
免疫組織化学手法は、一次抗体のエピトープを認識し、蛍光または発色であり得る(発色性の視認方法(chromogenic view method)が本発明において使用された)視認装置を含む二次抗体の適用によって証明される、これらの分子のエピトープに特異的な、一次抗体の適用による分子検出を可能にする。それ故、免疫組織化学的方法は、これらの分子を視認し、細胞または細胞外の間質のそれらの正確な位置をアッセイすることを可能にし、なぜなら、その研究が組織切片上で実行され得るからである。免疫組織化学手法を、一次細胞および商用細胞の両方である、すべての細胞株が、NK1、NK2およびNK3の受容体の存在を証明したと確証するために使用した。
細胞増殖を、製造業者によって与えられた説明書に従ってテトラゾリウム化合物3−(4,5−ジメチルチアゾル−2−イル)−5−(3−カルボキシメトキシフェニル)2−(4−スルホフェニル)−2H−テトラゾリウム(MTS)を使用して評価した(Kit "CellTiter 96 Aqueous One−Solution Cell Proliferation Assay" Promega, USA;url:http://www.promega.com/resources/protocols/technical−bulletins/0/celltiter−96−aqueous−one−solution−cell−proliferation−assay−system−protocol/で利用可能な使用の十分なプロトコル)。細胞の数を、コールターメーターを使用して測定した。腫瘍細胞培養のプラークは、(細胞がない)空サンプルおよび(104個の細胞/mLを含む)対照サンプルを含んだ。培養のプラークを、増加する濃度の、抗NK1、抗NK2または抗NK3のモノクローナル抗体の存在下で培養した。増殖アッセイにおいて染色を得るために、20μLのMTSを、492nm(アッセイの波長)および690nm(参照波長)でマルチスキャナー分光光度計(TECAN Spectra classic, Barcelona, Spain)においてサンプルの読み取りを実行する90分前に、細胞培養のプラークのウェルの各々に加えた。異なる用量を2回繰り返し試験し、各試験を、3回繰り返して実行した。
アポトーシスの形態を、DAPIでの染色後に、蛍光顕微鏡法によってアッセイした。NK1、NK2およびNK3の受容体抗体が腫瘍細胞培養においてアポトーシスを誘導することができるか否かを決定するために、染色をDAPIによって実行した。簡潔に言うと、NK1、NK2およびNK3の受容体抗体での処置後、細胞を、4%のパラホルムアルデヒド中に固着した。PBS中での2回目の洗浄後、細胞を、暗闇で30分間1/1000の濃度でDAPI溶液(Sigma−Aldrich)中でインキュベートした。続いて、細胞を、蛍光顕微鏡下で検査した(Zeiss, Oberkochen, Germany)。アポトーシスの細胞を、アポトーシスの典型的な特性(例えば、核濃縮、核空胞化、細胞膜の完全性の損失およびアポトーシスの抗体の形成)によって識別した。アポトーシスの細胞の数を数え、3つの異なるスライドにおいて計算を繰り返した。
本発明で議論されるように、腫瘍細胞の分子機構が、癌の形成および進行に関係するだけでなく、間質細胞、炎症、および腫瘍細胞且つ間質細胞と炎症との間の相互作用は、非常に重要である(McAllister SS, et al. 2010; Ikushima H, Miyazono K, 2010)。実施例1は、NK1、NK2およびNK3の受容体抗体が、いくつかの商用腫瘍細胞株においてアポトーシスを誘導することを示す。本実施例は、さらに、抗体での処置が、例えば、線維芽細胞、炎症細胞(単核および多形核球)、および血管内皮において、癌の処置に有用である、腫瘍微小環境を形成する細胞の生理機能の変化および変更を誘導することを示す。科学文献において十分に立証された重要性、およびNK1、NK2またはNK3の受容体のアゴニスト(例えば、物質P)の存在下で、およびこれらの受容体に対する抗体の存在下で変更される発現を有する因子は、この生理機能に関係する。
PHFを、De Bari et al.(De Bari et al., 2001)に記載されるような、皮膚の真皮から得られたサンプルから得、精製した。簡潔に言うと、小さな塊の皮膚の真皮を、37℃で15分間、1mg/mLの濃度でヒアルロニダーゼ溶液(Sigma−Aldrich, St. Louis, MO, US)中で消化し、続いて、37℃で2時間、6mg/mLのIV型コラゲナーゼ(Invitrogen)によって処置した。その後、細胞を、洗浄し、1%の抗生物質−抗真菌剤(Invitrogen)、および1%のナトリウムピルベート(Invitrogen)で補足した、高濃度のグルコース(ダルベッコ変法イーグル培地、Invitrogen)によってDMEM培地中で再懸濁し、1平方センチメートル当たり10,000個の細胞の濃度で6ウェルの培養のプラークにおいて播種した。細胞がコンフルエンスに達した時、付着細胞を、0.5%の無菌のトリプシン(Invitrogen)を使用して分離し、試験(run)3および9の間で使用した。物質Pの刺激および抗NK1、抗NK2および抗NK3の抗体への曝露のために、PHFを、1ウェル当たり25,000個の細胞の濃度で24ウェルのプラークにおいて播種した。
健康なドナーからのヘパリン処理した血液を、遠心分離にかけ、その異なる成分を分離した。赤血球は、チューブの底に残存する。それらの上に、白血球によって形成された小さな白っぽい層があり、その上のチューブの上部に血漿がある。白血球層を、24ウェルの培養プレートで分散し、それらの各々に、1%の抗生物質−抗真菌剤(Invitrogen)で補足した、同じ濃度の血漿を加えた。上に記載されるように得られた、皮膚の真皮からの約1000個の細胞(PHF)を、各ウェルに加えた。
若年期の包皮からの微小血管内皮細胞によって形成された商用の株からの細胞培養物も使用した(PromoCell GmbH, Sickingenstrase 63/65, D−69126 Heidelberg, Germany; reference C−12210)。増殖アッセイを、NK1、NK2およびNK3の受容体抗体の存在下で、実施例1と同様に実行した(セクション「増殖アッセイ」を参照)。

Claims (34)
- 癌の処置のための薬剤または医薬組成物の製造に関する、NK1、NK2及び/又はNK3の細胞受容体またはそれらの組み合わせに対して特異的な、少なくとも1つの抗体またはそのフラグメントの使用。
- 腫瘍細胞におけるアポトーシスの誘導のための薬剤または医薬組成物の製造に関する、NK1、NK2及び/又はNK3の細胞受容体またはそれらの組み合わせに対して特異的な、少なくとも1つの抗体またはそのフラグメントの使用。
- 腫瘍の大きさを縮小するための薬剤または医薬組成物の製造に関する、NK1、NK2及び/又はNK3の細胞受容体またはそれらの組み合わせに対して特異的な、少なくとも1つの抗体またはそのフラグメントの使用。
- 腫瘍の大きさの縮小は、腫瘍周囲微小環境を変更することによって実行されることを特徴とする、請求項3に記載の使用。
- 腫瘍周囲微小環境の変更は、間質のマトリックスおよび間質細胞の生理機能の変更、及び/又は、炎症反応ならびに/もしくは免疫反応に関係する細胞の生理機能の変更、及び/又は、血管化の原因である細胞の生理機能の変更を含むことを特徴とする、請求項4に記載の使用。
- 間質のマトリックスおよび間質細胞の生理機能及び/又は炎症反応ならびに免疫反応の細胞の生理機能の変更は、腫瘍マーカーの阻害を含むことを特徴とする、請求項5に記載の使用。
- 腫瘍マーカーは、好ましくは、NF−kB、TGFアルファ、TGFベータ1、TGFベータ2、TGFベータ3、SPARC、MMP−3、MMP−7、MMP−9、MMP−11、MMP−13、MMP−14及び/又はそれらの任意の組み合わせのリストのいずれかから選択されることを特徴とする、請求項6に記載の使用。
- 血管化の原因である細胞の生理機能の変更は、好ましくは、血管内皮細胞の増殖を阻害することによる、血管新生の阻害を含むことを特徴とする、請求項5に記載の使用。
- 癌は、黒色腫、神経膠腫、癌腫、腺癌、白血病、リンパ腫、骨髄腫、胚芽腫または神経外胚葉腫のタイプの新生細胞及び/又は肉腫を含む群のいずれかから選択されることを特徴とする、請求項1乃至8のいずれかに記載の使用。
- 癌腫は、以下の、乳管癌を含む乳癌、小細胞肺癌および非小細胞肺癌の両方を含む肺癌、濾胞性甲状腺癌および甲状腺乳頭癌の両方を含む甲状腺癌、前立腺癌、子宮頚癌、子宮内膜癌、卵巣癌、内皮および移行性の膀胱癌の両方を含む膀胱癌及び/又は絨毛癌のいずれかから選択されることを特徴とする、請求項9に記載の使用。

- 肉腫は、好ましくは、骨肉腫、横紋筋肉腫、子宮内膜間質肉腫、線維肉腫、悪性線維性組織球腫またはユーイング肉腫であることを特徴とする、請求項9に記載の使用。
- 新生細胞は、好ましくは、神経芽腫または網膜芽細胞腫であることを特徴とする、請求項9に記載の使用。
- 抗体は、配列番号1、配列番号2または配列番号3から選択される、NK1、NK2またはNK3の受容体の少なくとも1つの配列を認識することを特徴とする、請求項1乃至15のいずれかに記載の使用。
- NK1、NK2及び/又はNK3の受容体、またはそれらの組み合わせに対して特異的な、少なくとも1つの抗体またはそのフラグメントを含む医薬組成物。
- 抗体は、配列番号1、配列番号2または配列番号3から選択される、NK1、NK2またはNK3の受容体の少なくとも1つの配列を認識することを特徴とする、請求項17に記載の医薬組成物。

- 間質のマトリックスおよび間質細胞の生理機能の変更、及び/又は、炎症反応ならびに免疫反応に関係する細胞の生理機能の変更は、腫瘍マーカーの阻害を含むことを特徴とする、請求項31に記載の使用。
