Description
キヌレニナーゼ(KYNU)は、アスパラギン酸アミノトランスフェラーゼスーパーファミリーとして知られているピリドキサール5’−リン酸(PLP)依存性酵素のファミリーのメンバーである。真核生物の構成的キヌレニナーゼ類は3−ヒドロキシ−l−キヌレニンの加水分解的切断を優先的に触媒して3−ヒドロキシアントラニル酸およびl−アラニンを生成する。ヒトのKYNUのクローニング、発現、精製、特性付けおよび結晶化が報告されている(Lima et al, Biochemistry 46(10):2735-2744 (2007)。下記の実施例3で記述するように、KYNUはその保存されたH3ドメイン中にコア2F5エピトープを有する。
本発明における使用に適したHIV−1抗原には、膜近位外部領域(MPER)抗原(Armbruster et al, J. Antimicrob. Chemother. 54:915-920 (2004), Stiegler and Katinger, J. Antimicrob. Chemother. 512:757-759 (2003), Zwick et al, Journal of Virology 79:1252-1261 (2005), Purtscher et al, AIDS 10:587 (1996))ならびにその変種、例えばMPER Mab 2F5および4E10に対する、または他の広く中和するEnvに対するより高い中和感受性を与える変種、例えばMPER中にL669S変異を含有するMPER変異体Envペプチド脂質複合体が含まれる(Shen et al, J. Virology 83:3617-25 (2009))。好ましい免疫原には、図25および26、ならびに図16B、16C、図17、図18および図20において示した免疫原が含まれる。別の好ましい態様において、その変種はMPER mAb 2F5および4E10に対するより高い中和感受性を与えるL669S変異を有するMPERエピトープペプチドである(Shen et al, J. Virology 83: 3617-25 (2009))。
実験の詳細
急性のHIV−1に感染した患者。研究のために選択された患者は、患者の履歴およびFiebigの分類(Fiebig et al, AIDS 17:1871-1879 (2003))から見積もられた伝染の日により、伝染後17から30日までであった。患者065−0およびFIKEはFiebig第1期であり、一方で患者068−9、684−6およびMCERはFiebig第2期であった。
インフルエンザワクチン接種。インフルエンザワクチン接種された対象からの、単一細胞選別された形質細胞/形質芽球に由来する抗体のクローンを試験し、その応答が高度にクローン的であることが分かった。そのクローンメンバーはほぼ全て試験したインフルエンザ抗原と反応した。図1は、H1ソロモン諸島(Soloman Islands)ヘマグルチニンに対する代表的なインフルエンザ抗体クローンを示す。合計450の抗体を3人のインフルエンザワクチン接種された対象の形質細胞/形質芽球から分離し、これらの内で57.7%がインフルエンザ特異的であった。インフルエンザに感染した対象から分離された全部で265の抗体のうちで、クローン的に関連した抗体の20種類の独立したクローンが同定され、その中で115の抗体(92%)がインフルエンザ抗原と反応した。
・HIVに対する初期の抗体応答は、非中和性Env gp41エピトープに焦点が合わせられている。
・AHIまたは未感染の対象から分離されたgp41に結合するIgM抗体は腸細菌叢にも結合するが、gp41陰性IgMは腸細菌叢抗原に結合しない。
mAb HV00276と反応する腸細菌溶解物中のタンパク質のバンドの濃縮および同定を図52に示す。Native PAGEゲル泳動後のウェスタンブロット分析は、mAb HV00276が嫌気性および好気性腸細菌溶解物中の約520kDaのタンパク質のバンドに結合することを示している。サイズ排除クロマトグラフィー(SEC)の後に、約500kDaの分子量を有する細菌溶解物からのタンパク質画分を集めた。SEC画分は、クーマシーブルー(1)、銀染色(2)およびmAb HV00276によるウェスタンブロッティング(3、矢印)により520kDaのタンパク質の濃縮を示す。zoom等電点分画法は、mAb反応性タンパク質のpH6.2〜7を有するゲル区画A4への移動を示す。
自己寛容がどのようにHIV−1に対する防御的体液性応答に影響を及ぼす可能性があるかを理解するためには、どの自己抗原がHIV−1エピトープにより模倣されるのか、およびどこで/いつこれらの自己抗原がTおよびBリンパ球に晒されるのかを決定することが重要である。図30において、HIV−1 gp41 MPERのエピトープに特異的なモノクローナルヒト抗体はアセトン固定されたマウス3T3細胞中に存在する自己抗原とも反応することが示されている。図31において示されているように、少なくとも4種類の別個の分子がマウス3T3細胞からビオチン化2F5抗体により免疫沈降され得る。沈降した主要な種は、おおよそ50〜54kDaの見た目の分子量を有する。
2F5の固定された3T3細胞への反応性が、2F5 MPERコアエピトープ(ELDKWA)を含有するタンパク質/ポリペプチドにより阻害され得るかどうかを決定するため、2F5モノクローナル抗体(10μg/ml)を増大するモル濃度の同種(JRFLおよびDP178)または異種(R4A)阻害剤と反応させた(1時間、25℃)。続いてこれらの混合物を、メタノール/アセトンで固定した3T3細胞で覆われた水和された/ブロッキングされたスライドに添加した(2時間、25℃)。スライドをすすぎ、次いで250mlの(0.1%Tween−20および0.5%BSAを含むPBS)中で一夜洗浄した。洗浄されたスライドを、ヤギ抗ヒトIgG−FITC(0.1%Tween−20および0.5%BSAを含むPBS中1:400)で覆った。1時間後、スライドを洗浄し、Fluoromount−G中でカバーガラスを乗せた(coversliped)。24時間後、Zeiss Axiovert 200M共焦点顕微鏡を200倍の倍率および固定された300msecの露光時間で用いて蛍光画像を得た。
上記で記述したように、本発明は、HIVエンベロープタンパク質(ペプチドまたはポリペプチド)を投与して、第1に広く中和する成熟した抗体を生じることができる未変異祖先抗体を発現するB細胞を標的とし、次いでその体細胞成熟を経ているB細胞を選択されたHIVエンベロープタンパク質(ペプチドまたはポリペプチド)により追加免疫することにより、B細胞クローンの成熟を望まれる中和の広さに向けて駆動することを含むワクチン戦略に関する。その戦略の開発には、この成熟経路の再構築が含まれていた。望まれる最終的な(成熟した)抗体を広く中和する抗体を産生する患者から分離し、その抗体を特性付けした。そのそれぞれの推定上の祖先抗体を推定し、実際の抗体として発現させ、何にそれらが結合するかに関する決定を行った。未変異の“祖先”および中間抗体を発現するB細胞は、適切なタンパク質(ペプチドまたはポリペプチド)により引き金を引くと親和性が成熟してその患者において観察される広く中和する抗体を分泌するB細胞をもたらすであろうと考えられる。
対象707−01−021−9の凍結されたPBMCから得られたおおよそ30,000個の記憶B細胞をスクリーニングし、CAP45の感染性を50%より大きく中和する28の培養物を見つけた(図56)。モノクローナル抗体CH01、CH02、CH03、CH04およびCH05(CH01〜CH05)をこれらの培養ウェルの4つから分離した(1−27−G2、1−27−G11、1−19−Fl0および1−19−B7)(図56)。
CH01〜CH05抗体は、以下の要因に基づいて全て同じクローンファミリーのメンバーであることが決定された:(1)V(D)Jファミリー;(2)HCDR3の長さ;(3)HCDR3領域およびn−挿入のヌクレオチド配列。
B細胞に結合させてCH01〜CH05様抗体の産生を誘発するためにどの単量体性エンベロープをワクチン配合物中で用いることができるかを決定するために、CH01〜CH05モノクローナル抗体およびRUAを32種類の単量体性エンベロープの集団への結合に関して試験した。表6は、μMで表したEC50を示す。
1.gp120のa4b7への結合のためのモチーフはLDVおよびLDIである
HSV gD LPVおよびLDQ
これは、gDに対する抗体はHIV gp120のa4b7への結合を妨害することができるかどうかという疑問を生じさせる。
これは、gDに対する抗体はHIV Envのヘパラン硫酸への結合を妨害することができるかどうかという疑問を生じさせる。
ほとんどの単量体性gp120/gp140エンベロープへの結合の欠如は、CH01〜CH05が三量体性エンベロープ上で優先的に発現している立体構造感受性の4次抗体に結合することを示している。同様の発見がPG9およびPG16抗体に関して報告されている(Walker et al, Science 326(5950):285-9 (2009))。逆に、A244gD+ gp120エンベロープへの強い結合は、HSV−1糖タンパク質Dの同時発現が機能性エピトープを回復したことを示唆していた。
表8は、CH03はRNP、ヒストンおよびセントロメアB自己抗原と自己反応性であることを示している。CH03におけるセントロメアに結合する抗体の存在は、HEp−2細胞における間接蛍光抗体染色を用いても見出された(図67)。表12は、CH01〜CH05の4種類の非HIV抗原への結合(Luminexアッセイにより測定した)を報告する。そのデータは、CH01〜CH03が強く多反応性であることを示している。CH04およびCH05の多反応性は、さらに低いレベルにおいてさえもなお検出可能である。逆に、PG9およびPG16は多反応性能力を示さなかった。これらのデータは、CH01〜CH05ならびにPG9およびPG16抗体の間のそれぞれの発生の生物学における関連する可能性のある違いを指摘する。
CH01〜CH05抗体は、Walkerらにより最近記述された(PLoS Pathog. 6(8).pii:el 001028 (2010年8月5日))4次の広く中和するPG9およびPG16抗体と独特の特徴を共有している。特に、CH01〜CH05 bNabは、以下の4つの基準に基づいて“PG様”抗体として特性付けられた:(1)gp120タンパク質の位置160におけるアスパラギンのリシンへの点変異(N160K)が、そうでなければ中和感受性である分離株の中和を抑止する(abrogates)、(2)そうでなければ中和感受性である分離株の中和が、そのウイルスがマンノシダーゼI−阻害剤キフネンシンで処理された細胞中でのそれの産生により部分的に糖鎖除去された際に抑止される、(3)そのエピトープはエンベロープ三量体の状況において優先的に提示されるが、単量体性gp120またはgp140エンベロープ上には見付からない、そして(4)スレッディング(threading)がPG9またはPG16 bNabとの高い類似性を示す。bNabのCH01〜CH05クローンファミリーの代表として、CH01をそれがその4つの基準を全て満たすかどうか決定するために試験した。
要約すると、上記で示したデータは以下のことを実証している:(1)ファージディスプレイライブラリーを生成する必要のない自然抗体の迅速な同定および分離を可能にする戦略を開発した;(2)5種類のクローン的に関連する広く中和する抗体のファミリーを記述し、それらの発達を追跡した;(3)ヒトにおける末梢での受容体の編集の予備的な証拠を提供した;(4)以前に記述されたPG9およびPG16の広く中和する抗体に遺伝的に関連しない、PG様ファミリーの広く中和する抗体の新規のメンバーを記述した;そして(5)1個より多くのモノクローナル抗体が入手可能である場合に推定上の復帰未変異祖先の予想の正確さを増大するための方法を開発した。
Claims (24)
- 対象においてHIV−1に対する広く中和する抗体の産生を誘導する方法であって、以下の:
i)前記の対象に生殖細胞系列B細胞受容体に結合する非HIV−1抗原を投与し、前記の非HIV−1抗原はHIV−1 Envと交差反応する抗体を分泌するB細胞の中間クローンが生成されるような量で、およびそのような条件の下で投与され、そして
ii)前記の対象にHIV−1抗原を、前記の広く中和する抗HIV−1抗体を分泌するナイーブB細胞または前記のB細胞の中間クローンが生成されるような量で、およびそのような条件の下で投与する
ことを含む、前記方法。 - 前記の対象がヒトである、請求項1に記載の方法。
- 前記の非HIV−1抗原が脂質である、請求項1に記載の方法。
- 前記の脂質がカルジオリピン、ホスファチジルセリン、ホスファチジルエタノールアミン、ホスファチジルコリン、ホスファチジルイノシトール、スフィンゴミエリン、またはそれらの誘導体である、請求項3に記載の方法。
- 前記の脂質が1−パルミトイル−2−オレオイル−sn−グリセロ−3−[ホスホ−L−セリン](POPS)、1−パルミトイル−2−オレオイル−ホスファチジルエタノールアミン(POPE)、またはジオレオイルホスファチジルエタノールアミン(DOPE)である、請求項4に記載の方法。
- 前記の脂質がリン脂質の六方II相である、請求項3に記載の方法。
- 前記の非HIV−1抗原がフィコエリトリン(PE)、C−フィコシアニン(C−PC)、アポフェリチン、または嫌気性もしくは好気性腸細菌叢もしくはその構成要素である、請求項1に記載の方法。
- 前記の非HIV−1抗原が細菌または真核生物のRNAポリメラーゼコアタンパク質のαサブユニットを含む、請求項1に記載の方法。
- 前記の非HIV−1抗原がキヌレニナーゼ(KYNU)またはその抗原性断片である、請求項1に記載の方法。
- 前記のKYNUがCHOまたは293T細胞で発現させた組換えKYNU、またはその抗原性断片である、請求項9に記載の方法。
- さらにアジュバントを投与することを含む、請求項1に記載の方法。
- 前記のアジュバントがスクアレンに基づくアジュバント、TRL作動薬、オリゴヌクレオチド(oligonucletides)(oCpG)またはR848である、請求項11に記載の方法。
- 前記のHIV−1抗原が膜近位外部領域(MPER)抗原、またはその変種である、請求項1に記載の方法。
- 前記のHIV−1抗原が図16B、16C、17、18、20、25または26において示した免疫原である、請求項13に記載の方法。
- 前記の変種がL669S変異を有するMPERエピトープペプチドである、請求項13に記載の方法。
- 前記のHIV−1抗原が伝染始祖HIV−1 Env、またはその抗原性断片である、請求項1に記載の方法。
- 前記の断片がgp120のCD4結合部位の一部、MPER配列、またはgp120のV2もしくはV3領域を含むgp120の一部を含む、請求項16に記載の方法。
- その方法が、前記の対象に前記の非HIV−1抗原を含む予備刺激免疫処置、続いて前記のHIV−1抗原を含む1回以上の追加免疫を施すことにより達成される、請求項1に記載の方法。
- 前記の非HIV−1抗原が脂質、嫌気性または好気性腸細菌叢の細菌の構成要素、フィコビリタンパク質、またはKYNUもしくはその断片を含み、前記のHIV−1抗原がマラウイからの伝染始祖Env 1086.C、マラウイからの089.C、米国からの040_C9およびクレードB急性HIV−1に感染した米国の患者からの63521からなるグループから選択されるHIV−1 Env抗原を含む、請求項1に記載の方法。
- 前記の非HIV抗原または前記のHIV抗原がタンパク質を含み、前記のタンパク質をコードするDNA配列が、前記のDNA配列が発現し、それにより前記のタンパク質が生成されるような条件の下で前記の対象に投与される、請求項1に記載の方法。
- A244gD+エンベロープが予備刺激として投与され、CHO1、CHO2、CHO3、CHO4またはCHO5により結合されるエンベロープが追加免疫として投与される、請求項1に記載の方法。
