Description
本発明は、インビトロ及びインビボで哺乳動物細胞、特に胚性幹細胞の心筋形成を誘導するためのニューレグリンの使用に関する。
心(心室)肥大は、心仕事量に対するストレス又は需要の増加に応答しての重要な適応生理反応である。肥大のための刺激後に発生する初期の細胞変化の1つは、個々の細胞のサイズの比例増加を伴うミトコンドリアの合成及び筋原線維質量の増加(壁肥厚化)であるが、細胞数の増加はない(又は最小限である)。
本発明は、ES細胞の分化を心筋系列の細胞へと誘導し、方向付けるためのニューレグリンの使用を提供する。
(5.1定義)

本発明は、哺乳動物細胞において心筋形成を誘導する方法を提供する。ある種の実施態様において、本発明の方法は、哺乳動物細胞をニューレグリンと接触させる工程であって、該哺乳動物細胞が心筋系列の細胞へと分化する工程を含む。哺乳動物細胞は、インビボで又はインビトロで、ニューレグリン又はその組成物と接触させることができる。例えば、ニューレグリンは、手術の間に、静脈内投与で、又は直接投与によって、損傷した心筋又は機能不全の心筋に直接投与できる。
ニューレグリンは、インビボで心筋形成を誘導するために使用することができる。ニューレグリンは、哺乳動物細胞の心筋系列の細胞への分化を誘導するのに有効な量で、対象(例えば、ヒトなどの哺乳動物)に投与できる。心筋形成を誘導するその能力に鑑み、ニューレグリンは、急性心疾患の損傷心筋の修復に、及び心筋症などの障害を治療するために有用であると考えられている。いくつかの実施態様において、ニューレグリンを使用して、心筋組織の発生を研究するための心筋細胞を生成することができる。別の実施態様において、ニューレグリンは、損傷し、又は弱った心筋組織の修復又は増大を必要とする対象の治療の間、使用できる。別の実施態様において、ニューレグリンは、損傷していない、又は弱っていない心筋組織の増大又は強化を要求する対象を治療するために使用することができる。当該対象には、例えば、心臓病又は心臓障害の危険がある対象を含み得る。
ニューレグリンは、インビトロで心筋形成を誘導するために都合よく使用できる。ある種の実施態様において、哺乳動物細胞をニューレグリンと接触させ、そこにおいて該哺乳動物細胞は心筋系列の細胞へと分化する。
インビボで又はインビトロでのニューレグリンの投与後における心筋形成の誘導は、以下を含むがこれらに限定されない多くの異なる方法により検出できる:心筋細胞特異的タンパク質の発現の検出、心臓筋細胞特異的転写因子の発現の検出、心臓筋機能に必須なタンパク質の発現の検出、及び心臓筋細胞の拍動の検出。
分化した心筋細胞は、当業者に公知の任意の手段により、対象に投与できる。本発明の一実施態様において、完全な固体支持体(例えば、三次元マトリクス又は平坦面)上の分化した心筋細胞は、例えば外科移植を介して対象に投与できる。あるいは、分化した心筋細胞は、対象への例えば静脈内投与、皮下投与又は腹腔内投与前に、マトリクスからはがす、すなわちプロテアーゼで処理できる。
任意のニューレグリン(例えば、NRG-1、NRG-2、NRG-3及びNRG-4及びこれらのアイソフォーム)タンパク質、ペプチド又は断片を本発明に使用できる。
ニューレグリンは、当業者に明らかな任意の技術に従って調製できる。ニューレグリンの調製の例示的技術は、例えば、米国特許第7,226,907号、米国特許第5,367,060号、WO 94/026298、WO 03/099300に記載され、当該内容は引用によりその全てが本明細書に組み込まれる。ある種の実施態様において、ニューレグリンは、例えば液相又は固相ペプチド合成により、合成的に調製できる。Merrifieldの文献, 1963, J. - 心筋形成を誘導する方法であって、哺乳動物細胞をニューレグリンと接触させることを含み、これにより該哺乳動物細胞が心筋系列の細胞へと分化する、前記方法。
- 前記ニューレグリンが、ニューレグリン 1、ニューレグリン 2、ニューレグリン 3又はニューレグリン 4である、請求項1記載の方法。
- 前記ニューレグリンが、EGF様ドメインを含む、請求項1記載の方法。
- 前記ニューレグリンが、配列番号1のアミノ酸配列を含む、請求項1記載の方法。
- 前記ニューレグリンが、医薬として許容し得る担体にある、請求項1記載の方法。
- 前記哺乳動物細胞が、哺乳動物にある、請求項1記載の方法。
- 前記接触工程が、前記哺乳動物へのニューレグリンの静脈内投与による、請求項6記載の方法。
