オリゴマーに沿った側鎖の配列は、病状に対して所望の生理活性を有した原型αペプチド(標的)に基づいていることが好ましい。所望の化合物の特性を最適化するために、側鎖の配列は、α/βペプチドの主鎖上への翻訳後に変えることもできる。

操作されたらせん内部の塩橋及びXxx→Ala置換によってらせん傾向を強めるよう意図された側鎖変異を多数含む。以前の研究において、3は、細胞培養において高められた抗ウイルス効力を示し、野生型CHR配列に基づくペプチドに対する半減期を増やした。3における変異は、gp41 NHR領域とのその結合相互作用の構造的性質を改変するよう意図しなかったけれども、6ヘリックスバンドル構造が不変であったというさらなる実験的証拠を求めた。αペプチド1と3との共結晶を、ハンギングドロップ蒸気拡散法によって得て、その構造を2.0Åの分解能まで解析した(表1及び図4Aを参照)。結果として生じる6ヘリックスバンドルは、天然のNHR+CHRペプチド複合体によって形成されるものと本質的に同一である。図4A(1+3共結晶)と図4B(1+2共結晶)とを比較されたい。
X線結晶学を利用して、NHRαペプチド1によって形成されたヘテロマーの6−ヘリックスバンドルをCHRαペプチド3、キメラCHRα/βペプチド8、又は、CHRα/βペプチド10と比較した(表3を参照)。
生物学的状況におけるαペプチド3並びにα/βペプチド4、5、及び10の活性を評価するために、二組の実験を行った。第一の実験は、gp41により媒介されるHIV−細胞間融合のモデルを作るために一般的に使用される、HIV−1クローンHxB2のenv遺伝子の実験に基づく細胞間融合アッセイにおいてオリゴマーを比較した。(Deng YQ,Zheng Q,Ketas TJ,Moore JP,&Lu M(2007)Protein design of a bacterially expressed HIV−1 gp41 fusion inhibitor.Biochemistry 46(14):4360−4369)細胞間融合アッセイの結果(表4)は、α/βペプチド5及び10がαペプチド3と区別がつかないIC50値を有し、一方、α/βペプチド4ははるかに効果的ではないことを示した。化合物3、4、5、及び10を、次に、株化細胞TZM−b1のHIV感染を防ぐ能力について評価した。(Wei XP,et al.(2002)Emergence of resistant human immunodeficiency virus type 1 in patients receiving fusion inhibitor(T−20)monotherapy.Antimicrob Agents Chemother 46(6):1896−1905)これらの研究は、1つのT細胞系統適応株(T−cell line adapted strain)及び3つの一次分離株(primary isolate)を利用し、株のうち2つはX4向性であり、その他2つはR5向性である。
蛋白質間相互作用のフォルダマー拮抗薬を開発するための重要な動機づけは、蛋白質分解に対する感受性を減少することの見込みである。蛋白質分解酵素による急速な破壊は、αペプチド薬物の臨床的使用に対する重要な欠点を表している。乱交雑のセリンプロテアーゼであるプロテイナーゼKによる分解に対するαペプチド3並びにα/βペプチド4及び10の感受性を比較した。アッセイ条件の下、αペプチド3を数分以内に完全に分解し(図9A)、質量分析によって、配列内の少なくとも10個の異なるアミド結合の加水分解が明らかになった(図9D、一番上の配列)。排他的にα→β3置換を有したα/βペプチド4は、原型αペプチド3と比較して、安定性において20倍の改善を示した。図9B及び図9Dの真ん中の配列を参照されたい。堅くされたα/βペプチド10は、αペプチド3と比較して、安定性においてさらに優れた改善(280倍)を示した。図9C及び図9Dの一番下の配列を参照されたい。α/βペプチド4と比較した場合のα/βペプチド10のより優れた安定性はおそらく、CDによって検出される10のより高いらせん傾向から生じる。質量分析によってα/βペプチド10に対して観察した少数の蛋白質分解産物(図9D)は、混ぜ合わされたα/β主鎖におけるβ−残基が隣接するアミドを蛋白質切断から保護する傾向がある、以前の観察を支持している。
ペプチド合成において使用される保護されたα−アミノ酸及び樹脂を、Novabiochem社(EMD Chemicals Inc.社及びMerck KGaA社、Darmstadt,Germanyの完全所有子会社)から購入した。保護されたβ3−アミノ酸を、PepTech社(Burlington,Massachusetts,USA)から購入した。環状にされたβ−残基、Fmoc−ACPC及びFmoc−APC(Boc)を、すでに記載されたように調製した。Lee,LePlae,Proter,and Gellman,J.Org.Chem.2001,66,3597−3599;LePlae,Umezawa,Lee,and Gellman,J.Org.Chem.2001,66,5629−5632.2−(1H−ベンゾトリアゾール−1−イル)−1,1,3,3−テトラメチルアミニウムヘキサフルオロホスフェート(HBTU)を、AnaSpec社(San Jose,California,USA)から購入した。5−カルボキシフルオレスセインを、Invitrogen社(Carlsbad,California,USA)から購入した。1−メチル−2−ピロリジノン(NMP)を、Advanced Chemtech社(Louisville,Kentucky,USA)から購入した。他の試薬全てを、Sigma−Aldrich Corp.社(St.Louis,Missouri,USA)又はFisher Scientific社(Pittsburgh,Pennsylvania,USA)から購入し、受け入れたままの状態で使用した。
全てのペプチドを、“NovaSyn TGR”ブランドの樹脂(Novabiochem社)で調製した。αペプチドを、Symphony Multiple Peptide Synthesizer(Protein Technologies Inc.社、Tucson,Arizona,USA)で標準的なFmoc固体相ペプチド合成方法によって調製した。α/βペプチドを、Synergy432A automated synthesizer(Applied Biosystems社、Foster City,California,USA)で自動化されたFmoc固体相ペプチド合成によって調製した。α/βペプチドを、マイクロ波補助Fmoc固体相ペプチド合成によって手動でも調製した。Erdelyi and Gogoll(2002)Synthesis 11:1592−1596.8:2:1のDMF/DIEA/Ac2Oを用いた処理によって、各ペプチドのN末端にキャップ状の構造を付加した。樹脂を徹底的に洗浄し(3xDMF、3xCH2Cl2、3xMeOH)、次に、真空下で乾燥させた。全てのペプチドを、94:2.5:2.5:1のTFA/H2O/エタンジチオール/トリイソプロピルシランを用いた処理によって樹脂から切断した。前記樹脂をろ過し、さらなるTFAで洗浄して、乾燥窒素流の下で混合ろ液を2mLまで濃縮させた。粗ペプチドを、冷たいエーテル(45mL)の添加によって切断混合物から沈殿させた。前記混合物を遠心分離してデカントし、さらに、残りの固体を窒素流の下で乾燥させた。逆相HPLCによってprep−C18カラムで、0.1%TFA水溶液と0.1%TFAアセトニトリル溶液との勾配を使用してペプチドを精製した。最終生成物の正体及び純度を、それぞれMALDI−TOF−MS及び分析用HPLCによって確認した。原液の濃度を、UV吸光度によって決定した。Gill,S.C.;Vonhippel,P.H.Anal.Biochem.1989,182,319−326.MALDI−TOF−MS(モノアイソトピック〔M+H〕+,m/z): 1:obsd.=4162.6,calc.=4162.4; 2:obsd.=4288.7,calc.=4288.0; 3:obsd.=4455.0,calc.=4455.3; 4:obsd.=4609.9,calc.=4609.5; 5:obsd.=4526.1,calc.=4525.4; 6:obsd.=4552.7,calc.=4553.4; 7:obsd.=4631.6,calc.=4631.4; 8:obsd.=4516.5,calc.=4515.3; 9:obsd.=4713.0,calc.=4713.5; 10:obsd.=4539.9,calc.=4539.4; 11:obsd.=3299.4,calc.=3299.8。
遊離なN末端を有した全長C38ペプチド(WMEWDREINNYTSLIHSLIEESQNQQEKNEQELLELDK;SEQ ID番号:23)を有する“NovaSyn TGR”ブランドの樹脂を、Symphony Multiple Peptide Synthesizer(Protein Technologies,Inc.社)で標準的なFmoc固体相ペプチド合成方法によって25μmolスケールで調製した。合成に続き、樹脂をガラス注射器まで移した。5−カルボキシフルオレスセイン(28mg、0.075mmol)及びHOBT・H2O(11mg、0.075mmol)を、N−メチル−2−ピロリジノン(0.75nL)に溶解した。ジイソプロピルカルボジイミド(12μL、0.075mmol)を添加した。結果として生じる溶液を、ペプチドを有する樹脂まで移した。反応容器をホイルで覆い、振盪機上に一晩置いた。前記樹脂を、DMF(3x)で洗浄し、新たな試薬を用いて結合反応を繰り返した。次に、前記樹脂をDMF(3x)、20%ピペリジン(2x)、DMF(3x)、CH2Cl2(3x)、及び、MeOH(3x)で洗浄した。Fischer,R.;Mader,O.;Jung,G.;Brock,R.Bioconjugate Chem.2003,14,653−660.上記のように、粗ペプチドを切断及び精製した。水溶液中で調製した原液を、可視吸光度によって定量化した(pH8にてε494=68,000M−1cm−1)。MALDI−TOF−MS(モノアイソトピック〔M+H〕+,m/z):obsd.=5089.3,calc.=5089.3。
1μLの結晶化原料(crystallization stock)と1μLの貯蔵緩衝液(reservoir buffer)を混ぜ合わせた後に、0.7mL緩衝液で室温にて平衡化することによって懸滴を調製した。1+3の複合体及び1+8の複合体の原液を、水中全ペプチドが2.2mMという最終濃度まで、それぞれのペプチドの濃縮された原料を1:1の比で混ぜ合わせることによって調製した。1+3の結晶を、0.1M Tris pH8.5、1M(NH4)H2PO4を含む貯蔵緩衝液から得た。1+8の複合体の結晶を、0.4M Li2SO4・H2O、12%v/vPEG8000、20%v/vグリセロールを含む貯蔵緩衝液で成長させた。1+10の複合体を結晶化する初期の試みにおいて、水中全ペプチドが0.76mMという最終濃度までそれぞれのペプチドの濃縮された原料を1:1の比で混ぜ合わせることによって原液を調製した。この方法で調製した1+10の原料は、完全に可溶性であるというわけではなかった。しかし、結果として生じる粘性の懸濁液は、0.5M 硫酸アンモニウム、0.1M HEPES−Na、pH7.5、30%v/v2−メチル−2,4−ペンタンジオールから構成される望ましい緩衝液から、α/βペプチド10の結晶のみをもたらした。続く1+10の結晶化の試みに対して、水中全ペプチドが130μMで1:1のペプチド混合物を再び折り畳むことによって、複合体の原液を調製し、続いて、10kDa分子量のカットオフ膜を介して、4℃での遠心分離によって1.1mMまで濃縮した。1+10の結晶を、この方法で調製した原料、及び、0.2M NaCl、0.1M Tris pH8.5、25%w/vPEG3350から構成される貯蔵緩衝液から得た。
全ての結晶を液体窒素において急速冷凍した。冷凍に先立ち、1+3の複合体の結晶を、0.08M Tris pH8.5、1.6M(NH4)H2PO4、20%v/vグリセロールの中に簡単に浸した。10の結晶及び1+8の結晶を、結晶化ドロップから直接冷凍した。冷凍に先立ち、1+10の複合体の結晶は、0.2M NaCl、0.1M Tris pH8.5、25%w/vPEG3350、20%v/vグリセロール内に簡単に浸した。1+3の複合体及び1+8の複合体に対する回折データを、Cu Kα放射線を使用してBruker X8 Proteum Diffractometer(Bruker AXS,Inc.社、Madison,Wisconsin USA)で収集し、Bruker Proteum2ソフトウェアパッケージを用いて処理した。10の結晶及び1+10の複合体の結晶に対する回折データを、Advanced Photon Source,Argonne National LaboratoryのLife Sciences Collaborative Access Team beamline 21−ID−Gにて収集し、HKL−2000ブランドのソフトウェア(HKL Research,Inc.社、Charlottesville,Virginia,USA)を用いて処理した。CCP4ソフトウェアスイートを使用して構造決定を実行した。Collaborative Computational Project Number4(1994)The CCP4 Suite−Programs for Protein Crystallography.Acta Crystallogr,Sect D 50:760−763.分子置換を、Phaserソフトウェア(McCoy AJ,Grosse−Kunstleve RW,Storoni LC,&Read RJ(2005)Likelihood−enhanced fast translation functions.Acta Crystallogr,Sect D61:458−464)、又は、Molrepソフトウェア(Vagin A&Teplyakov A(1997)MOLREP:An automated program for molecular replacement.J Appl Crystallogr 30(6):1022−1025)を用いて実行した。自動化された精製に対するRefmac(Murshudov GN,Vagin AA,&Dodson EJ(1997)Refinement of macromolecular structures by the maximum−likelihood method.Acta Crystallogr,Sect D 53:240−255)、手動のモデル構成に対するCoot(Emsley P&Cowtan K(2004)Coot:Model−building tools for molecular graphics.Acta Crystallogr,Sect D 60:2126−2132)、及び、自動化された水構成及び自由原子密度改変に対するARP/wARPの組み合わせによって精製を成し遂げた。(Lamzin VS&Wilson KS(1993)Automated refinement of protein models.Acta Crystallogr,Sect D 49:129−147)1+3の複合体の構造を、発表されたgp41六量体構造(PDB ID:1AIK)から得たサーチモデルを用いて解析した。Chan DC,Fass D,Berger JM,&Kim PS(1997)Core structure of gp41 from the HIV envelope glycoprotein.Cell 89(2):263−273.α/βペプチド10の構造を、1+3の複合体からのCHRヘリックスをサーチモデルとして用いて解析した。2つのサーチモデルである、1+3の複合体からのNHRヘリックス及びα/βペプチド10のみの構造からのCHRヘリックスを用いて、1+10の複合体の構造を解析した。2つのサーチモデルである、1+3の複合体からのNHRヘリックス、並びに、1+3及び1+10の構造体から調製したキメラCHRヘリックスを用いて1+8の複合体の構造を解析した。PyMOL(DeLano Scientific、Palo Alto,California,USA)を使用して、分子グラフィックスを調製した。
ペプチドの原液を、TBS中(UV吸光度に基づく)25μMの濃度で調製した。プロテイナーゼKの溶液を、TBS中(重量対容積に基づく)50μg/mLの濃度で調製した。各蛋白質分解反応に対して、40μLのペプチド原料を、10μLのプロテイナーゼK原料と混ぜ合わせた。室温にて反応を進めさせ、100μLの1%TFA水溶液を添加することによって所望の時点にて反応を停止させた。125μLの結果として生じる反応停止された反応を、分析用逆相HPLC上に注入し、0.1%TFA水溶液と0.1%TFAアセトニトリル溶液との勾配で作動させた。存在する開始ペプチドの量は、220nmでのピークの積分によって定量化した。各時点に対して2度反応を行った。GraphPad Prismブランドのソフトウェア(GraphPad Software,Inc.社、La Jolla,California,USA)を使用して時間依存的なペプチド濃度を指数関数型減衰に適合させることによって、半減期を決定した。一部の時点に対する未精製の試料を、MALDI−MSによって分析し、観察した生成物を使用して、反応の間に切断されたアミド結合を同定した。
本明細書に使用されるgp41−5コンストラクトの配列は以下である。
蛍光偏光アッセイを、室温にて黒色のポリスチレンプレートにおいて行った。全ての測定を2ウェルで行った。アッセイ緩衝液を、20nM ホスフェート、pH7.4、1mM EDTA、50mM NaCl、0.2mM NaN3、0.5mg/mL “Pluronic F−68”ブランドのポリオキシアルキレンエーテル界面活性剤から構成した。gp41−5に対するFlu−C38の結合親和性を、384−ウェルプレートにおいて、(競合FP実験の条件を模倣するよう添加された)1%v/v DMSOを有したアッセイ緩衝液における1つのウェルあたりの最終容積が50μLになるまで、増加する濃度の蛋白質により固定された濃度のラベルされたペプチド(0.2nM)を滴定することによって測定した。全てのウェルにおいて二度行った。そのプレートを30分間平衡化させ、Envision 2100プレートリーダーで分析した。Graphpad Prism software(Graphpad Software Inc.社、La Jolla,California)を使用して、データをFP直接結合モデルに適合させた。Roehrl,M.H.A.;Wang,J.Y.;Wagner,G.Biochemistry 2004,43,16056−16066.トレーサーのKdを、0.4±0.1nMであるように決定した。測定した結合親和性は、gp41−5/Flu−CHR相互作用に対して以前に報告されたものよりもいくらかきつかった(Kd=3nM)が、以前の研究は、はるかに高い濃度のトレーサー(5nM)を直接結合実験において利用していた。直接結合FP実験において正確に決定することができるKd値の下限は、利用されるトレーサーの濃度にほぼ等しい。Roehrl,Wang&Wagner,supra。
円二色性測定を、Aviv 202SF Circular Dichroism Spectrophotometerで実行した。各ペプチドの試料を、PBS中20μMの濃度で調製した。1:1ペプチド混合物の溶液を、それぞれのペプチド測定に使用する同じ20μMの原料から等しい容積を混ぜ合わせることによって調製した。スペクトルを、1mmセルにおいて、1nmのステップサイズ及び5秒の平均時間で記録した。全てのスペクトルは、同じセルにおいて測定される緩衝液に対してバックグラウンド補正される。熱融解(thermal melt)を、各温度変化の間に10分の平衡時間を設けて5度の上昇で実行した。GraphPad Prismを使用して、熱変性データを、単純な二状態の折り畳みモデル(Shortle,D.;Meeker,A.K.;Freire,E.Biochemistry 1988,27,4761−4768)に適合させた。
ペプチドの原液を、TBS中(UV吸光度に基づき)25uMの濃度で調製した。プロテイナーゼKの溶液を、TBS中(重量対容積に基づき)50μg/mLの濃度で調製した。各蛋白質分解反応に対して、40μLのペプチド原料を、10μLのプロテイナーゼK原料と混ぜ合わせた。室温にて反応を進めさせ、100μLの1%TFA水溶液を添加することによって所望の時点にて反応を停止させた。125μLの結果として生じる反応停止された反応を、分析用逆相HPLC上に注入し、存在する開始ペプチドの量を、220nmでのピークの積分によって定量化した。各時点に対して2度反応を行った。GraphPad Prismを使用して時間依存的なペプチド濃度を指数関数型減衰に適合させることによって、半減期を決定した。一部の時点に対する未精製の試料を、MALDI−MSによって分析し、観察した生成物を使用して、反応の間に切断されたアミド結合を同定した。
CHO細胞において発現されたHIV−1クローンHXB2の外被糖蛋白質に基づき、U373−MAGI細胞を標的として用いる細胞間融合アッセイを、以前に記載されたように実行した。(Deng YQ,Zheng Q,Ketas TJ,Moore JP,&Lu M(2007)Protein design of a bacterially expressed HIV−1 gp41 fusion inhibitor.Biochemistry 46(14):4360−4369)。U373−MAGI標的細胞におけるβ−ガラクトシダーゼ発現の基礎レベルを測定することによって判定されるように、α/βペプチド全てが5μMにて細胞毒性を示さなかった。HIV−1感染性の阻害を、HIV−1 LTR(末端反復配列)の制御下でCD4、CXCR4、CCR5、及び、ルシフェラーゼ遺伝子を発現するTZM−b1(JC53BL)細胞で測定した。(Wei XP,et al.(2002)Emergence of resistant human immunodeficiency virus type 1 in patients receiving fusion inhibitor(T−20)monotherapy.Antimicrob Agents Chemother 46(6):1896−1905)。4つのHIV−1株:クレイドBのX4−向性T細胞系統適応分離株IIIB由来のクローンであるNL4−3;クレイドBのX4一次分離株であるHC4(Trkola A,et al.(1998)Neutralization sensitivity of human immunodeficiency virus type 1 primary isolates to antibodies and CD4−based reagent is independent of coreceptor usage.J Virol 72(3):1876−1885);R5一次分離株、CC1/85(クレイドB)(Connor RI,Sheridan KE,Ceradini D,Choe S,&Landau NR(1997)Change in coreceptor use correlates with disease progression in HIV−1−infected individuals.J Exp Med 185(4):621−628);及び、別のR5一次分離株、DJ258(クレイドA)(Louwagie J,et al.(1995)Genetic diversity of the envelope glycoprotein from human immunodeficiency virus type−1 isolates of African origin.J Virol 69(1):263−271);のPBMCにおいて生成されるウイルスストックを使用した。
上記のように、蛋白質上の特異的部位に堅固及び選択的に結合する分子を設計することは、分子認識において重要な挑戦であると考えられる。従って、適切な分子を同定するための系統的アプローチは、紛れもない利点である。この実施例は、天然の蛋白質結合領域にわたった系統的な主鎖の改変(すなわち、配列ベースの設計)を使用して、標的蛋白質の表面に堅固及び選択的に結合するα/βペプチドフォルダマーを迅速に生じることができるということを示すよう提供されている。この実施例においては、配列ベースの設計アプローチを使用して、蛋白質Bcl−xLのBH3認識クレフトに対するα/βペプチドフォルダマーリガンドを開発した。Bcl−xLは、プログラムされた細胞死経路を制御する、及び、抗アポートシスメンバー(例えば、Bcl−2、Bcl−xL、Mcl−1)もアポトーシス促進メンバー(例えば、Bak、Bad、Puma)も含むBcl−2ファミリーのメンバーである。Adams&Cory(2007)Oncogene 26:1324−1337を参照されたい。
以下のペプチド12、13、及び、8は、リードα/βフォルダマー10のキメラα+α/βフォルダマーである。これらのペプチドを、N末端付近の領域におけるベータ置換の効果を決定するために作製した。ベータ残基を、7価の原子に沿って「f」及び「c」位置において順次差し引いた。αからβへの置換の効果を、以前に報告した蛍光偏光(FP)競合アッセイを用いてモニターした。(Frey,G.;Rits−Volloch,S.;Zhang,X.Q.;Schooley,R.T.;Chen,B.;Harrison,S.C.Small molecules that bind the inner core of gp41 and inhibit HIV envelope−mediated fusion.Proc.Natl.Acad.Sci.,2006,103,13938−43)。ゆるやかに増加する、結合減少の効果をβ置換が有していることを結果は示唆している。

Claims (13)
- 非天然型生理活性ポリペプチドを作製する方法であって:
(a)α−アミノ酸残基を含むアミノ酸配列を有した生理活性ポリペプチド又はその生理活性断片を選択するステップ;及び
(b)ステップ(a)の前記生理活性ポリペプチド又は断片の配列に一致するアミノ酸配列を有する合成ポリペプチドを作製するステップであり、
(i)前記合成ポリペプチドにおいて、ステップ(a)の前記生理活性ポリペプチド又は断片に存在する前記α−アミノ酸残基のうち約14%から約50%が、β−アミノ酸残基と置換され、
(ii)前記合成ポリペプチドにおいて、前記β−アミノ酸残基及び前記α−アミノ酸残基は、繰り返しパターンで分配され、さらに、
(iii)前記合成ポリペプチドは、約10残基から約100残基までの長さを有し、少なくとも2つのβ−アミノ酸残基を含む、ステップ;
を含む方法。 - ステップ(b)の(i)は、前記α−アミノ酸残基のうち少なくとも1つを、β2及びβ3炭素原子を包含する環を介して環状にされた少なくとも1つのβ−アミノ酸残基と置換するステップを含む、請求項1に記載の方法。
- ステップ(b)の(i)は、ステップ(a)の前記生理活性ポリペプチド又は断片に存在する前記α−アミノ酸残基のうち約14%から約50%を、β2及びβ3炭素原子を包含する環を介して環状にされたβ−アミノ酸残基と置換するステップを含む、請求項1に記載の方法。
- ステップ(b)の(i)は、ステップ(a)の前記生理活性ポリペプチド又は断片に存在する前記α−アミノ酸残基のうち約14%から約50%をβ−アミノ酸残基と置換するステップを含み、前記β−アミノ酸残基のうち少なくとも1つが、そのβ2及びβ3炭素原子にて未置換である、請求項1に記載の方法。
- ステップ(b)の(i)は、ステップ(a)の前記生理活性ポリペプチド又は断片に存在する前記α−アミノ酸残基のうち約14%から約50%をβ−アミノ酸残基と置換するステップを含み、前記β−アミノ酸残基のうち全てが、そのβ2及びβ3炭素原子にて置換される、請求項1に記載の方法。
- ステップ(b)の(i)は、ステップ(a)の前記生理活性ポリペプチド又は断片に存在する前記α−アミノ酸残基のうち約14%から約50%をβ−アミノ酸残基と置換するステップを含み、各β−アミノ酸残基が、置換する前記α−アミノ酸残基と同じ側鎖を少なくとも1つ有する、請求項1に記載の方法。
- 蛋白質分解耐性の非天然型生理活性ポリペプチドを作製する方法であって:
(a)α−アミノ酸残基を含むアミノ酸配列を有した生理活性ポリペプチド又はその生理活性断片を選択するステップ;及び
(b)ステップ(a)の前記生理活性ポリペプチド又は断片の配列に一致するアミノ酸配列を有する合成ポリペプチドを作製するステップであり、
(i)前記合成ポリペプチドにおいて、ステップ(a)の前記生理活性ポリペプチド又は断片に存在する前記α−アミノ酸残基のうち約14%から約50%が、類似性のβ−アミノ酸残基と置換され、
(ii)各類似性のβ−アミノ酸残基は、置換する前記α−アミノ酸残基と同じ側鎖を少なくとも1つ有し、
(iii)前記合成ポリペプチドにおいて、前記β−アミノ酸残基及び前記α−アミノ酸残基は、繰り返しパターンで分配され、さらに、
(iv)前記合成ポリペプチドは、約10残基から約100残基までの長さを有し、少なくとも2つのβ−アミノ酸残基を含む、ステップ;
を含む方法。 - 前記ポリペプチドによってとられる折り畳み構造において、前記繰り返しパターンは、前記非天然型ポリペプチドがらせん形構造をとる場合に、前記β−アミノ酸残基を前記折り畳み分子構造の一側面に沿って一直線をなすように配置する、請求項1乃至7のいずれか一項に記載の非天然型生理活性ポリペプチドを作製する方法。
- β−アミノ酸残基及びα−アミノ酸残基の前記繰り返しのパターンは、(ααααααβ)、(αααααβ)、(ααααβ)、(αααβ)、(ααβ)、(ααβαααβ)、(ααβαβαβ)、及び、(αβ)からなる群から選択される、請求項1乃至7のいずれか一項に記載の非天然型生理活性ポリペプチドを作製する方法。
- ステップ(b)は、約20残基から約50残基を有する合成ポリペプチドを作製するステップを含む、請求項1乃至7のいずれか一項に記載の非天然型生理活性ポリペプチドを作製する方法。
- SEQ ID番号:4〜11及び25〜30に示された一次アミノ酸配列を含む単離された非天然型ポリペプチド。
- ヒト細胞に対するヒト免疫不全ウイルスの融合を阻害する方法であって、SEQ ID番号:4〜11及び25〜30に示された一次アミノ酸配列を含む単離された非天然型ポリペプチドにヒト細胞を接触させるステップを含む方法。
- ヒト細胞に対するヒト免疫不全ウイルス(HIV)の融合を阻害する方法であって:
(a)α−アミノ酸残基を含むアミノ酸配列を有し、in vivoでのHIV融合に必要な天然の生理活性ポリペプチド又はその生理活性断片を選択するステップ;
(b)ステップ(a)の前記生理活性ポリペプチド又は断片の配列に一致するアミノ酸配列を有する合成ポリペプチドを作製するステップであり、
(i)前記合成ポリペプチドにおいて、ステップ(a)の前記生理活性ポリペプチド又は断片に存在する前記α−アミノ酸残基のうち約14%から約50%が、β−アミノ酸残基と置換され、
(ii)前記合成ポリペプチドにおいて、前記β−アミノ酸残基及び前記α−アミノ酸残基は、繰り返しパターンで分配され、さらに、
(iii)前記合成ポリペプチドは、約10残基から約100残基までの長さを有し、少なくとも2つのβ−アミノ酸残基を含む、ステップ;並びに、
(c)ステップ(b)の前記合成ポリペプチドにヒト細胞を接触させるステップ;
を含む方法。
