モノクローナル抗体のクローン化は、様々な理由から有用である。単一の抗体を産生するハイブリドーマ細胞によって、同一であり、同一な効果を発揮する多数の抗体を生成する方法が提供される。これは、治療的使用の対象とされる抗体に有用である。モノクローナル抗体、とりわけ軽鎖及び重鎖の可変領域の配列を決定することは、それによって、例えば一本鎖抗体(例えばscFv)又は組み換え一価抗体(例えばFab)のような抗体変異体の生成を可能とする多くの形態のタンパク質工学が可能となるため、特に有用である。モノクローナル抗体(monoclonals)、特に可変領域の配列を決定することは、重要なことには、ヒト様に見えることによってより良く宿主免疫系を回避することができるためにヒトに対する療法に好ましい場合がある、「ヒト化されている」又は部分的にヒト化されている(キメラ)抗体の生成を可能とする。
当業者によく知られている標準的なハイブリドーマ生成及びその後のスクリーニングを用いて、MUC1*ペプチドに対するモノクローナル抗体を生成した。MUC1*ペプチド(GTINVHDVETQFNQYKTEAASRYNLTISDVSVSDVPFPFSAQSGA、配列番号1)、又は単一のアミノ酸置換を保有するペプチド変異体(GTINVHDVETQFNQYKTEAASPYNLTISDVSVSDVPFPFSAQSGA、配列番号2)を、KLHへの結合を可能にするためにカルボキシ末端にシステイン残基を追加して合成した。この結合ペプチドを使用して、マウスを免疫化した。融合細胞を含有するウェルからの上清を、MUC1*ペプチドの認識についてELISAによってスクリーニングした。これらのうち幾つかのクローンが、ELISA結合アッセイにおいてMUC1*ペプチドを認識する能力に対するその高い力価に基づいて選択された。6つのハイブリドーマクローンからの上清のかかる結合の結果を図1に示す。次いで、クローンを無傷細胞上のMUC1*と結合するその能力について、FACS(蛍光活性化細胞分類)によって試験した(図2及び図3)。これらうちで最良のものをハイブリドーマとして維持して、シークエンシングした。MIN−C2及びMIN−E6という2つのクローンが、MUC1を発現する乳癌細胞株(ZR−75−1)、及びMUC1*をコードするプラスミドをトランスフェクトしたHCT−116結腸癌細胞の細胞表面上のMUC1*に対するそれらの結合に基づくと、とりわけ好ましかった(図2及び図3)。このハイブリドーマ生成プロセスを複数回繰り返し、MUC1*の細胞外ドメイン(本質的にPSMGFR配列(GTINVHDVETQFNQYKTEAASRYNLTISDVSVSDVPFPFSAQSGA、配列番号1)からなるMGFRとも呼ばれる)に選択的に結合する、幾つかのモノクローナル抗体、IgG及びIgMを産生した。選択されるモノクローナル抗MUC1*抗体の可変領域の配列を図11〜図14に示す。
細胞表面上のMUC1*の認識を、FACSを用いた細胞の表面染色によって分析した。MIN−C2抗体については、精製抗体の10μg/ml溶液50μlを、空のベクターをトランスフェクトしたMUC1陰性HCT116細胞(HCT116−VEC8)、又はMUC1*発現ベクターをトランスフェクトしたHCT116細胞(HCT−MUC1*−10)及びMUC1陽性ZR−75−1細胞の個々に対して4℃で30分間結合させた。細胞を2回洗浄し、10μg/mlの抗マウスPEで4℃で30分間処理した。細胞を2回洗浄し、PBS中2%ホルムアルデヒドで固定し、BDのFACS CantoIIフローサイトメーターを用いて分析した(図2)。MIN−E6抗体についても、精製抗体の代わりにMIN−E6ハイブリドーマからの不希釈の上清50μlを使用したことのみを変えて、同様の手順に従った(図3)。
抗MUC1*モノクローナル抗体の能力を測定するために、MUC1を発現する乳癌細胞株T47Dを使用した。96ウェルプレートの1ウェル当たり7500個の細胞を、10%血清を含有する培地中にプレーティングした。翌日、3つのウェル中の細胞をトリプシン処理し、培地中に再懸濁し、血球計算盤を用いて計数して、0日目のカウント(zero day count)を得た。細胞の培地を、3%FBSを含有するものに交換し、抗MUC1*抗体を様々な濃度で添加した。48時間後、培地を交換した後、抗体の2回目の添加を行った。さらに48時間後、細胞を計数し、細胞成長の刺激を推定した(図4及び図5)。
MUC1*抗体の一価Fabフラグメントを調製するために、パパイン消化法(Pierce)を採用した。MIN−C2抗体及びMIN−E6抗体はパパインによって効果的に消化され、これをプロテインA−アガロースアフィニティクロマトグラフィーによって精製した(図6)。
MUC1*を発現する乳癌細胞株T47Dを、96ウェルプレートで完全培地(10%FBSを含有するRPMI)中にプレーティングし(5000細胞/ウェル)、一晩付着させた。翌日、培地を、3%FBSを含有するものに交換した。3つのウェル中の細胞をトリプシン処理及び再懸濁した後、血球計算盤を用いて計数し、0日目のカウントを得た。次に、MIN−C2又はMIN−E6 Fabタンパク質を、個々のプレートに様々な濃度で添加した。48時間後、培地を取り替え、Fabタンパク質の2度目の添加を行った。さらに48時間後、全てのウェル中の細胞を計数した。細胞成長の阻害を、Fabタンパク質を含有しない細胞の成長に対して算出した。MIN−E6の場合には、タンパク質のペグ化形態も試験したが(図8、図9)、非ペグ化Fabと同等に機能することが示された。
MIN−C2(アミノ酸1110から1155までのMUC1の45アミノ酸膜近位の細胞外領域:PSMGFRを認識するモノクローナル抗体)のscFvフラグメントを設計し、構築して、大腸菌において発現させた。この構築物は、いかなる分泌シグナルも付加せずに、ベクターpET21b(Novagen)を用いて設計したが、これにより6個のヒスチジン残基を含有するカルボキシ末端タグを有するタンパク質が得られる。MIN−C2及びMIN−E6 scFvの構築物の設計を図10A及び図15及び図16に示す。カルボキシ末端ヒスチジンタグによって、Ni−NTAアフィニティクロマトグラフィーによるscFvタンパク質の精製が可能となる(図17)。
MUC1*ペプチドを、96ウェルELISAプレートのウェルに一晩インキュベートすることによって結合させた。その後、superblock(Pierce)を用いて同様に一晩インキュベートすることによってブロッキングした。次に、superblockを取り除き、3%FBSを含有するDMEM培地を用いてウェルを30分間ブロッキングした。scFvタンパク質を、8回の段階希釈(最高濃度は50ug/ml)を用いて様々な濃度で添加した。scFvタンパク質の非特異的結合を決定するために、タンパク質を結合したMUC1*ペプチドを含有しない別のウェルセットに同じ希釈率で添加した。インキュベーションを1時間継続した。次に、0.3mlのPBST(0.02%Tween 20を含有するリン酸緩衝食塩水)でウェルを3回洗浄した。その後、ウサギ抗His抗体(AbCam)を、3%FBSを含有するDMEMを100倍に希釈することによって調製した培地で40000倍に希釈して添加した。これを1時間インキュベートした後、0.3mlのpf PBSTで3回洗浄した。次に、0.1mlの基質テトラメチルベンジジンをウェルに添加し、暗所で15分間発色させ、その後(following wihich)2N HClを0.1ml添加した。これらの実験によって、組み換えscFvが、PSMGFR(MUC1*細胞外ドメイン)ペプチドとの結合について無傷親抗体と効果的に競合することが確認された。
乳癌細胞株ZR−75−1を、MIN−C2 scFvの細胞表面上のMUC1*を認識する能力を試験するために使用した。細胞を、2倍又は10倍に希釈した1.5ug/mlのscFv原液(stock)と共に、又は対照としていかなるscFvタンパク質も添加せずに4℃で30分間インキュベートした。2回の洗浄の後、細胞を、Alexa 488(Qiagen)と結合させた二次抗体の抗penta−Hisと共に200倍、50倍又は10倍の希釈率でインキュベートし、scFv上の6×ヒスチジンタグを検出した。フローサイトメトリー分析によって、細胞サブセットの濃度依存的シフトが明らかとなり、MIN−C2 scFvの非存在下は見られない特異的結合が示唆された(図19)。
MUC1*受容体は、その活性化リガンドによって二量化された場合に細胞成長を刺激するため、単量体抗体及び抗体フラグメントがMUC1*陽性細胞の成長を阻害することが期待される。MUC1*を発現する乳癌細胞株ZR−75−1(別称1500)を、96ウェルプレートで完全培地(10%FBSを含有するRPMI)中にプレーティングし(10000細胞/ウェル)、一晩付着させた。翌日、培地を、3%FBSを含有するものに交換した。対照として、別の細胞株HCT116(結腸癌由来の、MUC1*を発現しない)を、同様に完全培地(10%FBSを含有するDMEM)中にプレーティングし(2000細胞/ウェル)、一晩付着させ、培地を3%FBSを含有するものに交換した。各々の細胞株について、3つのウェル中の細胞をトリプシン処理及び再懸濁した後、血球計算盤を用いて計数し、0日目のカウントを得た。次に、MIN−C2 scFv抗体変異体を様々な濃度で添加した。48時間後、培地を取り替え、scFvタンパク質の2度目の添加を行った。さらに48時間後、全てのウェル中の細胞を計数する。細胞成長の阻害を、scFvタンパク質を含有しない細胞の成長に対して算出した。scFvタンパク質が、MUC1*を発現するZR−75−1細胞の成長を阻害する一方で、MUC1*を発現しない対照細胞株HCT 116に対してはこの阻害効果を有しないことが観察された(図20)。
組み換えFabを同様に生成した。状況によっては、FabはscFv設計よりも好ましい。scFvと比べると、組み換えFabは血清中での半減期がより長く、その結合親和性は通常は完全な親抗体と同程度である。哺乳動物系における発現は、適切にフォールディングされたタンパク質の量を増大させることで活性の増強をもたらす。哺乳動物細胞から産生された抗体変異体は、大規模に生成される。哺乳動物発現のための組み換え抗体を作製するために、モノクローナル抗体を産生するハイブリドーマ細胞から可変領域及び定常領域をクローン化した。組み換えFabは多数の抗体設計を用いて作製される。組み換えFabは、本質的にVH領域、VL領域、CH領域及びCL領域を含む。場合によっては、重鎖と軽鎖とを結合するためにリンカーが使用される。他の場合では、重鎖及び軽鎖を別個のベクターに載せ、同じ細胞中で発現させるが、そこでそれらは自己会合して機能的Fabを形成する。両方の設計のFabを作製した。
リフォールディングの効率をさらに増大させ、最終的に抗体変異体の特異的活性を増大させるために、一本鎖Fab(scFab)を生成した。抗MUC1*抗体であるMIN−C2及びMIN−E6に対応する組み換え一本鎖Fab構築物を、図10B及び図22に示す設計を用いて生成する。scFab構築物は、N末端から始めて、軽鎖の可変領域(VL)、軽鎖の定常領域(CL)、34アミノ酸可動性リンカー、重鎖の可変領域(VH)、及び重鎖の定常領域のCH1部分から構成される。重鎖及び軽鎖は、以下の配列を有する可動性リンカーによって結合する:SGGGSGGGSEGGGSEGGGSEGGGSEGGGSGGGSG(配列番号361)。軽鎖(VL+CL)及び重鎖フラグメント(VH+CH1)は、特異的ハイブリドーマクローンから単離したmRNAを用い、特異的プライマーを用いて逆転写PCRを行うことによって得られる。
本発明の抗MUC1*モノクローナル抗体はヒト化モノクローナル抗体であっても、又はヒトモノクローナル抗体であってもよい。マウス抗体のごく一部をヒト免疫グロブリン分子に移植して、免疫グロブリン分子のFc部分のみがヒトであるキメラ抗体、又は免疫グロブリンの相補性決定領域(CDR)のみがマウスであり、分子の90%〜95%がヒトであるヒト化抗体を作り出すことによって、完全に抗原性のマウスmAbがヒトに負担をかけない(friendly)ものとなり得る。一面では、HuMAb−Mouse(GenPharm-Medarex)技術又はXenoMouse(Abgenix, Inc.)技術等の従来の方法を利用することによって、完全にヒトのモノクローナル抗体をトランスジェニックマウスにおいて生成することができる。ヒト化抗体には、レシピエントのCDRの残基を、所望の特異性、親和性及び生物学的機能を有するマウス、ラット又はウサギ等の非ヒト種のCDRの残基と置き換えたヒト免疫グロブリンが含まれる。
ウサギポリクローナル抗MUC1*抗体(配列番号1のペプチドによる免疫化によって生成した)を、モノクローナル抗MUC1*抗体MIN−C2と、bFGF又はフィーダー細胞抽出物の非存在下での幹細胞の成長を、それらの多分化能を維持した上で可能とするそれらの能力について比較した。この能力は、本質的にPSMGFR配列(配列番号1)から構成されるMUC1*の細胞外ドメインを二量化することによって達成される。
