Abstract

Description
本願は、米国特許仮出願第60/238,232号(2000年10月4日出願)、米国特許仮出願第60/241,991号(2000年10月20日出願)および米国特許仮出願第60/242,037号(2000年10月20日出願)(これらの開示は、参考として本明細書中に援用される)からの優先権を主張する。

レラキシン核酸、ポリペプチド、アナログおよびフラグメント、ならびにレラキシンレセプター核酸、ポリペプチド、アナログおよびフラグメントおよびアナログ、はまた、レラキシンポリペプチドまたはレラキシンレセプターに特異的に結合する候補化合物を検出するためのスクリーニングアッセイにおいて使用され、アポトーシスを変調する。そのような候補化合物は、典型的に低分子エフェクター(アゴニスト)またはアンタゴニストであり、そしてインビトロおよび/またはインビボアッセイによって同定され得る。そのようなアッセイは、レラキシンアゴニストまたはアンタゴニストとして、あるいは薬剤開発のためのリード化合物として治療的に有効である、低分子エフェクターまたはアンタゴニストを同定するために使用され得る。従って、本発明は、化合物を検出するためのアッセイを提供する。その化合物は、レラキシン核酸、レラキシンポリペプチド、レラキシンレセプター核酸、レラキシンレセプターなどの活性または発現に特異的に影響を与える。
ファージディスプレイライブラリーの例は、ScottおよびSmith(Science 249:386−90(1990))、Devlinら(Science 249:404−06(1990))、Christianら(J.Mol.Biol.227:711−18(1992))、Lenstra(J.Immunol.Meth.152:149−57(1992))、Kayら(Gene 128:59−65(1993))および国際特許公開WO94/18318(これらの開示は、本明細書中に参考として援用される)に記載される。
レラキシン核酸(センスおよびアンチセンスの両方)、ならびにそれらのフラグメントおよびアナログ、ならびに抗レラキシン抗体はまた、レラキシン関連異常を有する被験体を同定するための有用性を有する。このような分子を、アッセイ(ハイブリダイゼーションまたは免疫アッセイ)において使用して、種々の異常を検出、予後判定、診断またはモニターし得るか、レラキシン発現、あるいはレラキシンまたはレラキシンアナログに対する応答が影響されるか否かを決定し得る。同様に、このような分子は、細胞異常または組織異常の処置をモニターするための有用性を有する。詳細には、免疫アッセイのような方法は、被験体由来のサンプルに、抗レラキシン抗体を、免疫特異的結合を導く条件下で接触させること、およびこの抗体によるタンパク質の任意の免疫特異的結合の量を検出または測定することを含む工程によって行われ得る。組織切片中または精液由来のレラキシンまたはレラキシンレセプターポリペプチドに対する抗体の結合を使用して、レラキシンおよび/またはレラキシンレセプターポリペプチドの異常なレベル(例えば、レベルの低下、非存在または上昇)を検出し得る。特定の実施形態において、レラキシンまたはレラキシンレセプターポリペプチドに対する抗体を使用して、レラキシンまたはレラキシンレセプターポリペプチドの存在について、被験体の組織または精液をアッセイし得、ここで、レラキシンの異常なレベルは、レラキシン関連異常の指標である。「異常なレベル」によって、異常を有さない身体の一部分由来または被験体由来の類似のサンプル中に存在するレベル、またはそれらの類似のサンプル中の存在を示す標準レベルと比較して、上昇または低下したレベルを意味する。
マウスRLX遺伝子の1つの対立遺伝子または両方の対立遺伝子の不活性化の効果を試験した。以下の方法および材料に従った。
野生型、ヘテロ接合性およびホモ接合性のレラキシンノックアウトマウスを、Howard Florey Institute of Experimental Physiology and Medicine(Parkville、Victoria、3052、Australia)から得、そしてこれらを使用して、育種プログラムを確立した。RLX+/+(野生型)、RLX+/−(ヘテロ接合型)およびrlx−/−(ヌル変異体)マウスのその後の世代を、RLX+/−親から作製した。全ての動物を、制御された環境に収容し、そしてLabdietげっ歯類実験試料(Deans Animal Feed、San Bruno、CA)および水へのアクセスを与えながら、14時間の明所、10時間の暗所スケジュールで維持した。これらの実験は、実験動物の世話および使用についてのNIH Code of Practiceを順守する、Institute’s Animal Experimental Ethics Committeeによって承認された。
マウスDNAを、400μlのPCR溶解緩衝液(50mM Tris−HCl、pH8.0、0.5% SDS、0.1M EDTAおよび1mg/mlのプロテイナーゼK(Gibco BRL、Gaithersburg、MD)を含む)中で、50〜55℃にて一晩、尾組織(5〜7mm)を溶解することによって単離した。次いで、消化したサンプルを、血清容器(vaccutainer)チューブ中で3M 酢酸ナトリウム(40μl)、緩衝液飽和フェノール(200μl)およびクロロホルム(200μl)と混合し、その後、サンプルを、3000rpmで10分間、遠心分離した。DNA(上側の水相に含まれる)を、イソプロピルアルコール(240μl)を含む別の微量遠心チューブにデカントして、DNAを沈殿させ、その後、サンプルをボルテックスし、遠心し、そして上清を除去した。残ったDNAペレットを、40〜50μlの滅菌水中に溶解した。PCRのために、各DNAテンプレート(1μl)を、以下を含む30μl反応混合物中で使用した:PCR緩衝液(10mM Tris−HCl、pH8.3、50mM KCl、1.5mM MgCl2)、2.5mM dNTP、2.5U Taqポリメラーゼ(PGC Scientific,Gaithersburg,MD)、ならびに各150ngのRLX+/+プライマーおよびrlx−/−プライマー(Zhaoおよび共同研究者(Zhao Endocrinology 140:445−53(1999)によって設計)。増幅プロトコルは、94℃(3分間)での開始変性工程、それに続く、94℃(60秒)、55℃(60秒)および72℃(90秒)の35連続サイクルからなり、そして72℃でのさらに10分間の伸長で終了した。次いで、15μlの各サンプルを、2%(w/v)アガロースゲルでの電気泳動によって分析し、そしてエチジウムブロミドで染色した。235bpの産物が、野生型対立遺伝子から設計したプライマーによって生成され、170bpの産物が、変異体対立遺伝子プライマー(Zhao(1999)、前出)から生成された。
レラキシンの野生型、ヘテロ接合体およびホモ接合体の雄性および雌性(6個の群の各々において、nは20以上)を、1週齢、1月齢、2月齢および3月齢で得て、計量した。次いで、RLX+/+およびrlx−/−の雄性マウスを、組織収集のために、二酸化炭素での麻酔下で屠殺した。雄性生殖管(精巣、精巣上体、前立腺、精嚢および付随する脂肪を含む)を、各動物から、1週齢(n=10 RLX+/+雄性、n=11 −/−雄性);1月齢(n=10 RLX+/+雄性、n=10 −/−雄性);および3月齢(n=10 RLX+/+雄性、n=10 rlx −/−雄性)で収集した。各組織を計量した後、これらを、組織学的分析のために、10%ホルマリン中に配置した。

1月齢および3月齢の雄性マウスの生殖管由来の組織を、予めコーティングしたスライド上に載せ、そして58℃で(約30分間)加熱してパラフィンを除き、次いで、キシレンで3回洗浄し、無水エタノールで2回洗浄し、そして95%エタノールで2回洗浄し、その後短時間水に浸漬した。次いで、これらのサンプルを、加湿環境において、西洋ワサビペルオキシダーゼ(HRP)−ストレプトアビジン複合体(Santa Cruz Biotechnology Inc、Santa Cruz、CA)を利用するImmunocruz染色システムを使用して染色した。組織切片を、(内因性ペルオキシダーゼ活性をクエンチするため)最初にペルオキシダーゼブロッカーで処理し(5分間)、その後ヤギ血清中で予めブロッキングした(20分間)。次いで、各RLX+/+組織サンプルおよびrlx−/−組織サンプル由来の連続切片を、BaxモノクローナルIgG一次抗体(4μg/ml)(Santa Cruz Biotechnology Inc)、カスパーゼ−9ポリクローナルIgG抗体(4.5μg/ml)(Santa Cruz Biotech.)または増殖細胞核抗原(PCNA)(Santa Cruz Biotech.)モノクローナルIgG抗体(4μg/ml)のいずれかと共にインキュベートした(2時間)。使用される抗体の型に依存して、組織のマウスIgGコントロールまたはウサギIgGコントロール(Santa Cruz Biotech.)のいずれかの染色(2時間)をまた、実施された全ての実験において使用した。サンプルを、PBSで洗浄し(2分間)、適切な二次抗体(ヤギ抗マウスIgGまたはヤギ抗ウサギIgG)に供し(30分間)、上記のように洗浄し(2分間)、HRP−ストレプトアビジン複合体で処理し(30〜45分間)、そして製造業者の指示に従って調製したジアミノベンジジン色素原基質と共にインキュベートした(2〜10分間)。次いで、このスライドを、蒸留水で洗浄し(2分間)、その後95%アルコールからキシレンまで脱水し、そしてマウントし、次いで上記のように写真撮影した。
結果を、one−ANOVA試験を使用して分析した。この文書における全てのデータは、平均±SEMとして示され、p<0.05が、統計的に有意であるとみなされる。
(マウスの成長に対する、レラキシン遺伝子ノックアウトの影響)
雄性および雌性のレラキシン野生型(+/+)マウス、ヘテロ接合体(+/−)マウスおよびヌル変異体(−/−)マウスの体重を、1週齢、1月齢、2月齢および3月齢で測定した(各群について、n=20〜21)。1週齢では、雄性マウスにおいても(RLX+/+:3.87±0.09g;rlx−/−:3.6±0.13g)、雌性マウスにおいても(RLX+/+:3.52±0.09g;rlx−/−:3.37±0.08g)、平均体重の有意な差異は、注目されなかった。しかし、1月齢時点のマウスでは、rlx−/−の雄性(17.05±0.65g)および雌性(14.77±0.42g)の両方の平均体重は、それぞれのRLX野生型対応マウスよりも、有意に少なかった(p<0.05)(RLX+/+M:18.92±0.61g;RLX+/+F:16.34±0.51g)。雄性および雌性のヌル変異体マウスは、2月齢において、RLX野生型動物よりも有意に小さい(p<0.05)ままであった(RLX+/+M:25.42±0.41g;rlx−/−M:24.23±0.42g;RLX+/+F:20.96±0.32g;rlx−/−F:19.94±0.22g);しかし、2つの群間のサイズの差異は、1月齢で観察された差異よりも小さかった。成体(3月齢)までに、rlx−/−マウスの平均体重(rlx−/−M:26.57±0.47g;rlx−/−F:22.54±0.31g)は、成体のRLX+/+マウスの平均体重(RLX+/+M:27.30±0.32g;RLX+/+F:23.03±0.71g)よりもなお僅かに少なかったが、この差異は、もはや有意ではなかった。RLX+/−マウスの平均体重は、RLX+/+の平均体重とrlx−/−マウスの平均体重との間であった。RLX+/+動物の平均体重とRLX+/−動物の平均体重との間にも、RLX+/−マウスとrlx−/−マウスとの間にも、有意な差異は観察されなかった。
1週齢では、雄性の生殖管の重量においてもサイズにおいても、有意な差異は観察されなかった。これは、主に精巣によって示される(表1)。1月齢までには、収集された雄性生殖管は、精巣、精巣上体、前立腺、精嚢、精管および付随する脂肪から構成された。1月齢では、生殖管全体の重量において差異は観察されなかったが、1月齢のヌル変異マウスに由来する精巣および前立腺のサイズにおける差異は、RLX野生型動物の対応する組織よりも、それぞれ、約20%および約30%小さかった。一方、rlx−/−マウスに由来する精巣上体のサイズは、RLX+/+動物から得られた精巣上体よりも、有意に小さかった(p<0.05)。しかし、RLX+/+マウスから収集された組織サンプルとrlx−/−マウスから収集された組織サンプルとの間の、精嚢のサイズにおいては、差異は注目されなかった。
1精巣および3前立腺の各々のサイズを、組織の長さを幅で乗じて、その面積によって測定した。精巣上体および精嚢のサイズ(面積)は、正確には測定できなかったので、各組織の緊密な近似を、以下のように計算した:精巣上体(組織の長さに、精巣上体頭部の幅を乗じて測定した2);精嚢(各器官の長さに、組織の平均幅を乗じて測定した4)。*は、p<0.05を示す。
H&E&Massonトリクロム染色:RLX+/+マウスおよびrlx−/−マウス由来の雄性生殖管組織切片を、精子成熟、管サイズ/コンパクトさ(精巣、精巣上体)およびコラーゲンにおける差異について最初に観察した。
表2:組織切片を、H&Eで染色し、以下のように等級に分けた:
a細管サイズ−1=全ての細管が小さく、環状の形態である;2=ほとんどの細管が1と比較してわずかに伸張しているが、いくつかのより小さい環状細管が観察される;3=細管は1と比較して著しく伸張しているが、いくつかのより小さい環状細管および中サイズの細管が観察される;4=全ての細管が1および2と比較して著しく伸張している。
b細管のコンパクトさ−1=全ての細管の形が緩んでいる;2=形が緩んでいる細管の割合のほうが、コンパクトな細管の割合よりも多い;3=コンパクトな細管の割合のほうが、形が緩んでいる細管の割合よりも多い;4=全ての細管がコンパクト。
c精子の成熟度−0=精子は検出されず;1=未成熟な精子のみが検出された;2=未成熟な精子の割合のほうが成熟な精子の割合よりも多い;3=成熟な精子の割合のほうが未成熟な精子の割合よりも多い;4=成熟な精子のみが検出された。
示した数値は、測定した各パラメーターについて使用した等級分けスケールの平均±SE(n)である。*はp<0.05を示す。
表3:組織切片を、Massonトリクロム染色によって染色し、以下のように等級に分けた:
aコラーゲン−1=構造の外部層を取り囲むコラーゲンの薄い内層のみ;2=組織の内部構成成分を取り囲むコラーゲンのさらなる内層;3=組織の外部層および内部構成成分を取り囲むより厚いコラーゲンの内層;4=3+コラーゲンで充填された細管/構成成分との間の空間。
H&E染色からなされた観察(これによるとrlx−/−マウス由来の組織が低下した精子成熟および増大した細胞死(精巣)を引き起こし、そして減少した上皮(細胞)層(前立腺)を含む)に基づいて、観察された増大した細胞死がアポトーシス経路の結果であるか否かを調査することを決定した。
PCNA染色:増殖細胞核抗原(PCNA)抗体を使用して、DNA合成が生じる核領域と関連する能力に基づいて、雄性生殖管における細胞増殖を検出した。精巣:PCNAを未成熟な精細管および成熟な精細管において検出したが、PCNA染色における有意な差異は、1ヶ月齢および3ヶ月齢のRLX+/+マウスおよびrlx−/−マウス由来の精巣管において検出されなかった。未成熟組織において、PCNA染色は、通常、有糸分裂活性な精原細胞で高度に発現され、時に、いくつかのセルトーリ細胞で高度に発現され、これらの染色は、増殖活性に対応する。この研究において、これらの細胞は、マウス成体においても良好に、より均一かつ絶え間なくPCNAについて標識され、これはおそらく、これらの増殖細胞の不断の活性化を介して、再生を開始し得る両方の群の能力を反映する。精巣上体:PCNAについての染色は、1ヶ月齢および3ヶ月齢のRLX+/+マウスまたはrlx−/−マウスのいずれか由来の精巣上体においても検出されなかった。これらの知見は、成熟精子のレザバーとして作用する精巣上体の役割と一致する。前立腺:PCNAについての弱い染色が1ヶ月のRLX+/+マウスおよびrlx−/−マウス由来の前立腺管の上皮細胞と関連していた。しかし、3ヶ月齢の群からはいずれも、PCNAについての染色は検出されなかった。白血球増殖研究は、ある程度の生殖組織に限定され、この蓄積された知見によって、おそらく、レラキシンはおそらく、細胞増殖に対してよりも細胞アポトーシスの調節において、影響のある役割を果たすということが、確認された。
RLX野生型(RLX+/+)の雄および雌ならびにRLX遺伝子ノックアウト(rlx−/−)の雄および雌を、上記に記載されるように、RLXヘテロ接合体(RLX+/−)の親から発生させた。マウスを、組織を回収するために、1週間、1ヶ月および/または3ヶ月で重量し、屠殺した。以下の組織(脳、心臓、肝臓、腎臓、胸腺、脾臓および雄性生殖管を含む)を回収し、重量を量り、詳細な組織学的分析のために10%ホルマリン中においた。これらの組織重量のまとめを表4〜8に示す。
(皮膚の組織学に対するレラキシン遺伝子ノックアウトの影響)
組織リモデリングの調節におけるレラキシンの役割を、rlx−ヌルマウス(rlx−/−)における皮膚組織学を研究することによって調べた。これらのマウスは、rlx−/−のオス親およびメス親の子孫であった。少なくとも5匹のオスマウスおよび5匹のメスマウスの、耳および背中からの連続した皮膚サンプルを、H&E染色およびMassonトリクロム染色によって染色し、各時点で調べた。同じ時点の類似のサンプルを、同じ系統のRlx+/+マウスから得た。rlxヌルマウスの組織学的試験の際に、真皮が、時間の進行につれ厚くなること、および真皮一帯の増大した線維症を有することを見出した。rlxヌルマウスの皮膚サンプルは、1週齢において正常であった;しかし、1ヶ月までに初期の皮膚線維症が明らかとなった。3ヶ月齢までに、皮膚線維症の顕著な増大が存在し、これは、6ヶ月齢までに密度が増大した。これらの皮膚に関する知見は、雄性および雌性のrlxヌルマウスにおいて類似していた。
Claims (22)
- レラキシンレセプターを発現する細胞の集団においてアポトーシスを調節するための組成物であって、以下:
該レラキシンレセプターを発現する細胞の集団においてアポトーシスを増加するのに十分な有効量のレラキシンのアンタゴニストを含み、該レラキシンアンタゴニストが、レラキシンアンチセンス核酸、抗レラキシン抗体、可溶性レラキシンレセプター、可溶性レラキシンレセプターをコードする核酸、および抗レラキシンレセプター抗体である、組成物。 - 前記レラキシンアンタゴニストが、レラキシンレセプターに対するレラキシンの結合を阻害する、請求項1に記載の組成物。
- 前記細胞の集団は、脳、心臓、肝臓、腎臓、脾臓、胸腺、皮膚、雌性生殖管組織または雄性生殖管組織由来である、請求項2に記載の組成物。
- 前記雄性生殖管組織が、前立腺、精巣上体、精嚢または精巣である、請求項1に記載の組成物。
- 前記組織が、前立腺組織である、請求項4に記載の組成物。
- 前記雄性生殖管組織が、成熟している、請求項4に記載の組成物。
- 前記抗体が、モノクローナル抗体、ポリクローナル抗体、単鎖抗体、Fab、Fab’、F(ab’) 2 、Fv、単重鎖、またはキメラ抗体である、請求項1に記載の組成物。
- 前記組成物が、注入、注射、経口送達、鼻内送達、肺内送達、直腸送達、経皮送達、組織内送達または皮下送達による投与に適したものである、請求項1に記載の組成物。
- 前記投与が、遅延放出送達によるものである、請求項8に記載の組成物。
- 前記皮下送達が、注入または注射によるものである、請求項8に記載の組成物。
- 前記投与が、肺内送達、皮下送達または経皮送達によるものである、請求項8に記載の組成物。
- 増加されたアポトーシスが、所望でない細胞蓄積を減少させる、請求項1に記載の組成物。
- 前記所望でない細胞蓄積が、過形成、肥大、癌または新形成である、請求項12に記載の組成物。
- 被験体の組織におけるアポトーシスを増加させるための医薬の製造のための、治療有効量のレラキシンアンタゴニストの使用であって、該レラキシンアンタゴニストが、レラキシンアンチセンス核酸、抗レラキシン抗体、可溶性レラキシンレセプター、可溶性レラキシンレセプターをコードする核酸、および抗レラキシンレセプター抗体である、使用。
- 前記抗体が、モノクローナル抗体、ポリクローナル抗体、単鎖抗体、Fab、F(ab’) 2 、Fv、単重鎖、またはキメラ抗体である、請求項14に記載の使用。
- 前記レラキシンアンタゴニストが、レラキシンレセプターに対するレラキシンの結合を阻害する、請求項14に記載の使用。
- 前記医薬が、注入、注射、経口送達、鼻内送達、肺内送達、直腸送達、経皮送達、組織内送達または皮下送達のために処方される、請求項14に記載の使用。
- 前記医薬が、遅延放出送達のために処方される、請求項17に記載の使用。
- 前記組織が、前立腺組織、精巣上体組織、輸精組織または精巣組織である、請求項14に記載の使用。
- 前記被験体が、思春期後の雄性である、請求項14に記載の使用。
- 前記被験体が、思春期前の雄性である、請求項14に記載の使用。
- レラキシンレセプターを発現する前記細胞が、骨芽細胞、単球、上皮細胞または内皮細胞である、請求項1に記載の組成物。
