Abstract
Description
The present invention relates to an external patch containing loxoprofen or an alkali addition salt thereof known as a non-steroidal anti-inflammatory agent as a medicinal component.
Loxoprofen sodium hydrate (trade name: Loxonin), which is a medicinal ingredient used in the present invention, is a non-steroidal anti-inflammatory analgesic having excellent anti-inflammatory and analgesic effects, and is a tablet, fine granule, shipping agent and tape. Rheumatoid arthritis, osteoarthritis, low back pain, myalgia, shoulder periarthritis, toothache, swelling / pain after surgery / traumatic injury, and other inflammatory diseases in each surgical / internal medicine area, etc. It is a drug that is widely used in the treatment of cancer.
Until now, side effects such as gastrointestinal and renal disorders have been reported for oral preparations, and patients have to be careful when taking them. Therefore, research and development of external preparations for loxoprofen have been actively promoted for the purpose of reducing side effects, and many shipping agents and tape products have been developed.
Examples of the prior art include anti-inflammatory analgesic patches (see Patent Document 1) containing styrene-isoprene-styrene block copolymer, crotamiton and loxoprofen sodium, and anti-inflammatory analgesia containing loxoprofen or a medically acceptable salt thereof as an active ingredient. A patch, ointment, cream, lotion, or aerosol is disclosed as an external preparation (see Patent Document 2).
When a rubber-based elastomer or the like which is a thermoplastic elastic body is used, it is possible to obtain an anti-inflammatory analgesic patch having a sufficient utilization rate through percutaneous absorption of loxoprofen or an addition salt thereof. However, the solubility of loxoprofen or its addition salt in the adhesive base is very poor, and furthermore, it is very difficult to transdermally absorb a drug in the form of a salt such as loxoprofen sodium from the skin. In order to solve this problem, non-steroidal anti-inflammatory analgesics in the form of salts include oxyacids such as citric acid, tartaric acid and salicylic acid, succinic acid and acetic acid, and dibasic acids such as maleic and fumaric
Alkaline addition of loxoprofen by containing the above pH adjuster (see
However, depending on the type and blending amount of the pH regulator used, the percutaneous absorption of loxoprofen or its addition salt is insufficient, and satisfactory anti-inflammatory analgesic action cannot be obtained, or loxoprofen or its addition in the adhesive base. There was a problem that the stability of the salt was impaired. In the prior art (see Patent Document 4), the use of an alicyclic saturated hydrocarbon resin as a tackifier results in a decrease in the content of acidic drug or alkali addition salt over time, indicating a negative view. Has been.
In addition, it has been reported that an external patch containing loxoprofen sodium produces several types of degradation products under severe conditions (stored at 80 ° C. for 3 weeks) (see Non-Patent Document 1).

It is very useful for diseases aimed at anti-inflammatory analgesic action.
Hereinafter, the medicinal component and each base component in the present invention will be specifically described.
The medicinal component is loxoprofen or an alkali addition salt thereof having anti-inflammatory analgesic action known as a nonsteroidal anti-inflammatory agent. Alkali addition salts mean medically acceptable inorganic salts such as sodium, potassium, calcium and ammonium salts. When used as a medicinal ingredient, it is substantially an alkali addition salt of loxoprofen, and more specifically, loxoprofen sodium hydrate (hereinafter abbreviated as loxoprofen sodium) is preferred. The content of loxoprofen or an alkali addition salt thereof is blended and formulated within the range of 0.3 to 8% by mass, preferably 0.5 to 6% by mass. When the content of loxoprofen or its alkali addition salt is less than 0.5% by mass, the medicinal component has a low percutaneous absorbability, and it tends to be difficult to obtain a sufficient anti-inflammatory analgesic effect. . Further, when the amount exceeds 6% by mass, it tends to be difficult to dissolve the medicinal component in the adhesive base.
Next, each base component of aliphatic hydroxy acid and higher fatty acid will be described.
The effects of the present invention can be exhibited by using aliphatic hydroxy acids and higher fatty acids in combination, not individually. Examples of the aliphatic hydroxy acid include glycolic acid, lactic acid, malic acid, citric acid, isocitric acid, tartaric acid, and leucine acid. Among these, lactic acid, malic acid, and citric acid having 3 to 6 carbon atoms are particularly preferable. Even more preferred is a combination of citric acid or malic acid. These aliphatic hydroxy acids include optical isomers of D-form, L-form and DL-form. The blending amount of these aliphatic hydroxy acids is 0.1 to 3% by mass, preferably 0.2 to 3% by mass, and more preferably 0.5 to 2% by mass. As the amount of the aliphatic hydroxy acid is less than 0.5% by mass, the solubility of the medicinal component in the adhesive base and the percutaneous absorbability from the adhesive base tend to be insufficient, less than 0.1% by mass In this case, the tendency is remarkable, which is not preferable. Further, as the amount exceeds 2% by mass, the stability of the medicinal component tends to decrease, and when it exceeds 3% by mass, it is not preferable because it is remarkable.
Examples of higher fatty acids include capric acid, myristic acid, palmitic acid, stearic acid, isostearic acid, oleic acid, linoleic acid, and linolenic acid. Of these, saturated fatty acids such as myristic acid, palmitic acid, stearic acid, and isostearic acid having 14 to 18 carbon atoms are preferable. Even more preferred among these is the blending of isostearic acid.
The amount of these higher fatty acids is 0.1 to 5% by mass, preferably 0.2 to 4% by mass, and more preferably 0.5 to 3% by mass. As the amount of the higher fatty acid is less than 0.5% by mass, the solubility of the medicinal component in the adhesive base and the transdermal absorbability from the adhesive base tend to be insufficient, 0.2% by mass If it is less, it is not satisfactory in terms of the transdermal absorbability of the drug. Less than 0.1% by mass is not preferable because the tendency is remarkable. On the other hand, as the amount exceeds 3% by mass, the stability of the medicinal component tends to decrease, and when it exceeds 5% by mass, it is not preferable because it is remarkable.
As the pH regulator and / or skin permeation enhancer for the adhesive base containing an alkali addition salt of loxoprofen, it is essential to use the above-mentioned aliphatic hydroxy acid and higher fatty acid in combination. In order to adjust the pH in the adhesive base containing the loxoprofen alkali addition salt in a well-balanced manner, it is preferable to use an aliphatic hydroxy acid and a higher fatty acid in combination.
The mixing ratio (mass ratio) (α: β) of both the aliphatic oxyacid (α) and the higher fatty acid (β) is 1: 4 to 2: 1 in order to control the transdermal absorbability of the drug. It is desirable to specify the ratio.
In addition, by using together aliphatic hydroxy acid and higher fatty acid in a specific ratio, it has the effect of stabilizing the medicinal component in the adhesive base and suppressing the decomposition of the medicinal component, as well as the stability of the patch base. Moreover, it is excellent in transdermal absorbability of medicinal ingredients and can reduce skin irritation. Regarding the formulation related to the above-mentioned blending ratio, there is no fact of the prior art as well as the fact that it has been reported, and there has been no report on the inhibitory effect on the formation of degradation products of medicinal ingredients, and it can be used as a stabilizer for medicinal ingredients. Useful. In particular, the combination of malic acid and isostearic acid is a finding that has been found for the first time as a result of accumulating diligent research by the present inventors.

<Test Example 1 (dissolution test)>
A part of the adhesive base immediately after production of Examples 1 to 5 and Comparative Examples 1 to 3 was collected on a glass plate, and the drug solubility in the adhesive base was observed with the naked eye and a microscope according to the following criteria. Further, the same observation was made after storage in a dryer at a constant temperature of 60 ° C. for 4 weeks. The results are shown in Table 1.
○: No crystals (complete dissolution)
×: Crystal
As shown in (Table 1), in the patches for external use of Examples 1 to 5 formulated with an aliphatic hydroxy acid and a higher fatty acid at a specific blending ratio, the drug is completely dissolved immediately after production, and the condition is 60 ° C. No drug crystals were observed after storage for 4 weeks under. That is, in the external patches of Examples 1 to 5, no drug crystals were observed, and they were completely dissolved. On the other hand, in the external patch of Comparative Example 1, the drug was not completely dissolved, and in the external patches of Comparative Examples 2 and 3, the drug was dissolved immediately after production, but stored for 4 weeks at 60 ° C. After that, crystals were precipitated in the adhesive base. From these results, it was confirmed that the external patch of the present invention was an anti-inflammatory analgesic patch excellent in solubility of a medicinal component (loxoprofen or an addition salt thereof) in an adhesive base.

<Test Example 2 (Stability Test)>
The patches for external use of Examples 1, 2, 3, 5 and 6 and Comparative Examples 4 and 5 were put in an aluminum bag and sealed using a heat sealer. After storage for 4 weeks in a drier at a constant temperature of 60 ° C., the content of sodium loxoprofen and the amount of degradation products in the adhesive base were measured using high performance liquid chromatography (HPLC), and the stability of loxoprofen sodium in the adhesive base was measured. Sex was evaluated.
The loxoprofen content measured immediately after preparation of the preparation was taken as the initial value, the ratio of the loxoprofen content after storage to this initial value was calculated, and the content ratio (%) was determined. Further, the ratio of the total amount of the peak areas of the plurality of decomposition products to the peak area of loxoprofen after storage was defined as the decomposition product generation amount (%). The results are shown in Table 2.
As shown in (Table 1), the patches for external use of Examples 1, 2, 3, 5 and 6 showed little change in the content of loxoprofen sodium even after storage for 4 weeks under the condition of 60 ° C. The amount of decomposition products was also small. Above all, the stability of loxoprofen sodium of Examples 1, 2 and 5 was very good. It was also found that the content of loxoprofen sodium in the external patch of Example 6 was slightly decreasing. On the other hand, in the patches for external use of Comparative Examples 4 and 5, the amount of decomposition products was large, and a marked decrease in the content of loxoprofen sodium was observed. From these results, it was confirmed that the external patch of the present invention was an external patch excellent in retention stability of loxoprofen sodium, which is a medicinal component, without lowering the content of loxoprofen sodium in the adhesive base.
