本出願は、2007年7月16日出願の米国仮出願第60/959,728号に対して優先権の特典を主張し、その内容の全体は、参照により本明細書に援用したものとする。
「タンパク質」という用語は、本明細書で「ペプチド」および「ポリペプチド」という用語と互換的に使用される。
本明細書に記載するナノ粒子には、多くのタイプの化合物、例えば治療用薬剤または薬剤を含めることができる。そのような治療薬は、それだけに限定されないが、ステロイド、鎮痛薬、局所麻酔薬、抗生剤、化学療法剤、免疫抑制剤、抗炎症剤、増殖阻害剤、有糸分裂阻害剤、血管形成剤、抗精神病薬、中枢神経系(CNS)薬;抗凝血剤、線維素溶解剤、増殖因子、抗体、眼薬、および代謝産物、類似体、誘導体、フラグメント、およびこれらの化学種の精製、単離、組換え、および化学合成バージョン、ならびにそれらの組合せであり得る。

本明細書に記載したナノ粒子を使用して、疾患細胞および組織を治療する(例えば、疾患細胞および組織への薬剤の移行を仲介する)ことができる。この点で、様々な疾患が、本明細書に記載したナノ粒子および方法を用いる治療に適している。対象ナノ粒子によって治療できる疾患の例示的で、非制限的リストには、乳癌、前立腺癌、肺癌、リンパ腫、皮膚癌、膵癌、大腸癌、黒色腫、卵巣癌、脳腫瘍、頭頚部癌、肝臓癌、膀胱癌、非小細胞肺癌(non-small lung cancer)、子宮頚癌、白血病、非ホジキンリンパ腫、多発性硬化症、神経芽細胞腫および神経膠芽腫、TおよびB細胞介在自己免疫疾患、炎症性疾患、感染、過剰増殖性疾患、AIDS、変性疾患、心血管疾患、移植拒絶などが含まれる。場合によっては、治療する癌細胞は転移性である。
本明細書に記載したナノ粒子は、キットで提供することができる。場合によっては、キットには、(a)ナノ粒子を含む容器、および任意により(b)資料を含める。資料は、本明細書記載の方法および/またはナノ粒子の使用に関する説明書、指示書、販売書、または他の材料、例えば治療上の便益を目的したものであってよい。
難溶性薬剤の可溶化および生物学的利用能を増大するために、安定した難溶性薬剤コロイドを調製した。このハイパワー超音波処理を行うために、難溶性薬剤の水性分散剤を同時LbL−ナノコーティングと共に使用する。そのようなコーティングによって粒子表面電荷は逆転し増強され、それによって薬剤の再凝集は防止され、薬剤コロイドは(超音波処理時間に比例して)ますます小さくなる。

この実験では、難溶性かつ強力な抗癌薬タモキシフェン(TMF)およびパクリタキセル(PCT)を使用した(溶解度1μg/mL未満)。LbL組立法に使用した高分子電解質は全て、濃度2mg/mLで使用した。正電荷高分子電解質として、ポリ(アリルアミン塩酸塩)(PAH)、FITC標識PAH、およびポリ(塩化ジメチルジアリルアミドアンモニウム)(PDDA)を使用した。負電荷高分子電解質として、ポリスチレンスルホン酸ナトリウム(PSS)を使用した。溶媒として、脱イオン水およびpH7.4のPBSを使用した。ウルトラソニケーター(超音波破砕装置)3000(Misonix Inc, Farmingdale, NY)を3〜18Wtで10〜30分使用し、薬剤結晶の崩壊を行った。超音波処理中の試料の過熱を防止し、20〜30℃の範囲に温度を保つために、液体窒素を使用しサンプルチューブを冷却した。高分子電解質多層の厚みは、Quartz Crystal Microbalance(9 MHz QCM, USI-System, 日本)を使用し測定した。ZetaPlus Microelectrophoresis(Brookhaven Instruments)を使用し、表面電位(ゼータ電位)および粒径測定を実施した。粒子イメージングには、電界放射型走査型電子顕微鏡(日立製作所,2006年)を使用した。シェル形成を制御しコロイドの安定性を追跡するために、レーザー走査型共焦点顕微鏡(Leica Microsystems社製Leica TCS SP2)も使用した。
まず、いかなる高分子電解質を加える前に、薬剤試料は全て、冷却しながら、1mL容量中、18Wで最長30分間の超音波処理を使用し崩壊させた。形成された薬剤粒子径を定期的に測定した。高分子電解質の第一層を加える前に、ゼータ電位読取り値も読み取った。両薬剤の薬剤ナノ粒子は、固有の負電荷を有することが判明したので、ポリカチオンを使用して第一表面層を形成した。次いで、薬剤試料を14,000rpmで7分間遠心分離し、洗浄し、水またはPBSに再懸濁して、余分な高分子電解質を除去した。次いで、ゼータ電位の読み取りを行った。ポリアニオンポリマーを使用して、コーティング過程を繰り返したが、超音波処理はしなかった。ゼータ電位の測定は、各層を加えた後に行った。
LbL組立法を使用し調製したコロイド粒子からの様々な薬剤の放出速度を測定するために、1mLの酢酸セルロース膜製水平拡散チャンバー内に、異なる数のコーティングサイクルを使用し調製した試料を置いた。次いで、大容量のPBS(pH7.2)に試料を攪拌して、インビボで予想される浸漬条件を模倣した。放出される薬剤の濃度をHPLCにより測定した。
様々なリガンドの共有結合に好適な「反応性」表面を有するナノコロイドを調製するために、遊離アミノ基を含むPAHを使用して薬剤粒子に外層を形成した。この一連の実験でパクリタキセルを薬剤として使用した。モノクローナルヌクレオソーム特異的2C5抗体(mAb 2C5)をLbLパクリタキセルナノ粒子に抱合させた。癌細胞表面結合ヌクレオソームを介して、この抗体は多様な癌細胞を認識する(Iakoubovら, Oncol. Res. 9 (1997) 439-446、およびIakoubovら, Cancer Detect. Prev. 22 (1998) 470-475を参照)。この抗体を2段階で抱合させた(図1B)。第1段階では、1−エチル−3−カルボジイミド塩酸塩(EDC)とN−ヒドロキシスルホスクシンイミド(スルホ−NHS)とを用いて、mAb 2C5のカルボキシル基を活性化し、抗体をアミン反応性にした。第2段階では、ポリアミノ含有PAHポリマーでコーティングしたLbLパクリタキセルナノ粒子に、活性化した抗体を加えた。全ての反応は、アルゴンガスの存在下、4℃、pH7.4のHBS中で攪拌を継続しながら実施した。改変された粒子を12rpmで10分間遠心分離し、PBSを使用し2度再懸濁させて、抱合していない抗体を除去した。
LbL−パクリタキセルナノ粒子と抱合後のmAb 2C5比活性の保存を検証するために、標準的ELISAを実施した。要するに、40μg/mlポリリジン溶液を含むTBS(pH7.4)で予め処理したELISAプレートを40μg/mLヌクレオソーム(仔ウシ胸腺核ヒストンの水溶性画分、Worthington Biochemical, Lakewood, NJ)50μLでコーティングし、室温で1時間インキュベートした。次いで、0.2%カゼイン、0.05%Tween 20を含むTBS(カゼイン/TBS)(pH7.4)でプレートを水洗いした。これらのプレートに、mAb 2C5含有試料の段階希釈液を加え、室温で1時間インキュベートした。プレートをカゼイン/TBSで徹底的に洗浄し、西洋ワサビペルオキシダーゼヤギ抗マウスIgG抱合体(ICN Biomedical, Aurora, OH)でコーティングし、製造業者の指示に従って希釈した。室温で1時間インキュベーション後、プレートをカゼイン/TBSで洗浄した。結合したペルオキシダーゼをその基質であるジアミノベンジジン分解によって数量化したが、この基質は、増感型K-Blue TMB基質(Neogen, Lexington, KY)という使用可能な溶液として提供された。現像した色彩強度は、Labsystems Multiscan MCC/340 ELISAリーダーを使用し492nmで分析した(Labsystems and Life Sciences International, UK)。
MCF−7およびBT−20細胞に対する様々な濃度のLbL−パクリタキセルナノ粒子の細胞毒性を、MTTテストを用いて研究した。製造業者のプロトコルに従って、MTT(Promega, Madison, WI)の使用可能なCellTiter 96(登録商標)Aqueous One solutionを使用した。Hank緩衝液に分散させたパクリタキセルの濃度が最高200ng/mLまでの製剤を96ウェルプレートで約40%集密に増殖させた細胞に加えた。37℃、5%CO2で48時間または72時間インキュベーション後、プレートをHank緩衝液で3回洗浄した。次に、培養液100μlおよびCellTiter 96(登録商標)Aqueous One solution20μlをプレートに加え、プレートを37℃、5%CO2で1時間インキュベートした。次いで、ELISAプレートリーダーを使用し、492nmでのMTT分解生成物の色彩強度を測定することによって細胞生存率を推定した。未処理細胞を100%増殖と考慮した。
B.結果
最適の超音波処理条件を見つけるために、初期実験は、懸濁液中のタモキシフェン結晶の濃度2mg/mLで実施した。図2に示すように、粒径は、超音波処理時間により制御でき、超音波処理時間を長くして減少させることができた。18Wtで30分の超音波処理後、約100nmの粒子を得た(粒子の再凝集を防ぐため、サイズ測定前にポリカチオンのPDDAを加えた)。類似の超音波処理条件をパクリタキセルの結晶にかけた場合、約100nmの粒径も得られた。超音波処理時間をさらに延ばしても、薬剤粒径で著しい減少は見られなかった。
走査型電子顕微鏡(SEM)および共焦点蛍光顕微鏡を使用して、本明細書に記載したLbL技術により形成したナノ粒子のサイズを確認した。タモキシフェンを2mg/mLのPAHの存在下で20分間超音波処理した後、主として球形であり、直径が120±30nmであるナノ粒子が得られた(図7Bおよび7C)。48時間後に撮影したSEMイメージでは、個々に集塊していないナノサイズ粒子が依然として見られた。従ってナノコロイドは水中で安定していた(図7Bおよび7C)。図8は、ポリアニオンPSSの第一層を加えても、超音波処理の20分後でさえタモキシフェンのサイズが減少しないことを示している。
LbL技術は、ナノ粒子の厚みまたは組成を変えることによって、ポリマーにより安定化したコロイドナノ粒子からの薬剤放出速度を制御するために使用することができる。従って、2mg/mLタモキシフェンを含み、単一PDDAコーティングまたは(PDDA/PSS)3コーティング組成を有するLbLナノコロイド粒子からのタモキシフェンの放出を標準的浸漬条件(pH7.4のPBS緩衝液)で測定した。指数関数近似のPeppasモデルを使用し、実験データから曲線を生成した(Peppas, Pharm. Acta Helv. (1985) 60: 110-112を参照)。図12に示すように、シェルの高分子電解質層数が増加するにつれ、放出速度が遅くなることが観察された。浸漬条件(pH7.4のPBS緩衝液)で、未コートタモキシフェン結晶(超音波処理有りおよび無し)を約2時間以内に可溶化した。PDDAでコーティングしたナノ粒子および(PDDA/PSS)3でコーティングしたナノ粒子は、およそ10時間で可溶化すると推測された。パクリタキセルでも同様の結果が得られた。異なるポリカチオンとポリアニオンを含むLbLコーティングを使用し、シェル数を変えると、遅い放出速度が得られた。パクリタキセルナノ粒子でも同様の結果が見られた(図13)。
LbLでコーティングした薬剤ナノ粒子を誘導体化する能力を実証するために、上記のように、PAH一層を有するパクリタキセル含有ナノ粒子を作製した。次いで、PAH表面層の遊離アミノ基を介して、腫瘍特異的mAb 2C5を、PAHでコーティングしたパクリタキセルナノ粒子に結合させた。図14に示すように、2C5で改変したLbLコートパクリタキセルナノ粒子は、標的抗原(すなわちヌクレオソーム)を特異的に認識した。
実施例1に記載したように、メソ-テトラフェニルポルフィリンおよびカンプトテシンのLbLナノ粒子を調製した。図15に示すように、メソ-テトラフェニルポルフィリンナノ粒子は、表面電荷を負から正に逆転させるFITC標識PAHコーティングを使用し生成した。SEMにより、粒径は約83nm〜約194nmであることが示された(図15B)。
実施例1に記載したように、パクリタキセルのLbL薬剤ナノ粒子を調製したが、コーティングに生体適合性材料を使用した。第一層をプロタミン硫酸塩(PS)に、続いて次のコーティングをヒト血清アルブミン(HSA)にして、パクリタキセル含有ナノ粒子を調製した。30分間の超音波処理+プロタミン硫酸塩でのLbLコーティングにより、より小さいナノ粒子が得られた。
本発明は、その詳細な説明に関連して記載されているが、当然ことながら、先の記載は、添付の請求項の範囲によって定義される本発明を説明することを目的とするもので、本発明の範囲を制限するものではない。他の態様、利点、および変更形態も添付の請求項の範囲内にある。
Claims (25)
- (a)化合物、
(b)該化合物を取り巻く、第一ポリマーを含む第一所定固体ポリマー層、および
(c)該第一層を取り巻く、第二ポリマーを含む第二所定固体ポリマー層
を含む安定したナノ粒子であって、該第一ポリマーと該第二ポリマーが反対電荷を有し、かつ直径が約100nm〜約500nmであるナノ粒子。 - 該化合物が、約5重量%〜約95重量%存在する、請求項1に記載のナノ粒子。
- 該第一ポリマー層と該第二ポリマー層とを併せた厚みが、約5nm〜約30nmである、請求項1に記載のナノ粒子。
- 該第一ポリマーが正電荷であり、かつ該第二ポリマーが負電荷である、請求項1に記載のナノ粒子。
- 該第一ポリマーが負電荷であり、かつ該第二ポリマーが正電荷である、請求項1に記載のナノ粒子。
- 3層以上の所定固体ポリマー層を含む、請求項1に記載のナノ粒子。
- さらに、該第二ポリマー層を取り巻く第三ポリマー層であって、該第二ポリマーの反対電荷を有する第三ポリマーを含む第三ポリマー層を含む、請求項1に記載のナノ粒子。
- 該第一ポリマーと該第三ポリマーが同一である、請求項7に記載のナノ粒子。
- さらに、該第三ポリマー層を取り巻く第四ポリマー層であって、該第三ポリマーの反対電荷を有するポリマーを含む第四ポリマー層を含む、請求項7に記載のナノ粒子。
- 該第二ポリマー層が、標的薬によって改変されている、請求項1に記載のナノ粒子。
- 該第三ポリマー層が、標的薬によって改変されている、請求項7に記載のナノ粒子。
- 該第四ポリマー層が、標的薬によって改変されている、請求項9に記載のナノ粒子。
- 該標的薬が抗体である、請求項10に記載のナノ粒子。
- 洗浄剤または界面活性剤を含まない、請求項1に記載のナノ粒子。
- 該化合物が、約2時間以内に7%の速度で該ナノ粒子から放出される、請求項1に記載のナノ粒子。
- 該化合物が、約2時間以内に約3%の速度で該ナノ粒子から放出される、請求項12に記載のナノ粒子。
- (a)化合物、および
(b)逆帯電したポリマー(複数)の交互ポリマー層を含むポリマーコーティング
を含み、直径が約100nm〜約500nmであるナノ粒子。 - 2層以上の逆帯電したポリマー層を含む、請求項17に記載のナノ粒子。
- 該化合物が、約5重量%〜約95重量%存在する、請求項17に記載のナノ粒子。

- 超音波処理後、該非水溶性化合物が、該ポリマーの非存在下では負電荷を有する、請求項21に記載の方法。
- 該化合物に加える該ポリマーが正電荷を有する、請求項21に記載の方法。
