Abstract
Description
第2の領域よりも第1の領域において大きい寸法を有するゲルを提供するステップと、
前記第1及び第2の領域のゲルの寸法の差異が減少するようにゲルを可塑的に圧縮し、それにより生体材料を製造するステップと
を含み、前記生体材料が第2の領域と比較して第1の領域において増大した剛性を有する、可変剛性を有する生体材料を製造する方法を提供する。
第1の領域から第2の領域に向けて徐々に増大する寸法を有するゲルを提供するステップと、
前記第1及び第2の領域のゲルの寸法の差異が減少するようにゲルを可塑的に圧縮し、それにより生体材料を製造するステップと
を含むことができ、ここで、前記生体材料の剛性は第1の領域から第2の領域に向けて徐々に増大する。
本明細書に記載の方法を用いて組織等価インプラントを製造するステップと、
前記インプラントを前記損傷組織に固定して該組織を修復および/または置換するステップと
を含む、個体内の前記損傷組織の治療方法を提供する。
細胞は、先の文献の記載に沿って投入及び播種した(WO2006/003442;WO2007/060459;Eastwoodら、Biochem Biophys Acta 1201(1994)186〜192頁)。
Claims (24)
- 第2の領域よりも第1の領域において大きい寸法を有するゲルを提供するステップと、
前記第1及び第2の領域のゲルの寸法の差異が減少するようにゲルを可塑的に圧縮し、それにより生体材料を製造するステップと
を含み、前記生体材料が第2の領域と比較して第1の領域において増大した剛性を有する、可変剛性を有する生体材料を製造する方法。 - 可塑的圧縮後に、ゲルの寸法が第1と第2の領域において同一である、請求項1に記載の方法。
- 生体材料の剛性が第1の領域から第2の領域にかけて徐々に増大するように、ゲルの寸法が、ゲルの第1の領域から第2の領域にかけて徐々に増大する、請求項1または2に記載の方法。
- 生体材料の剛性が第1の領域から第2の領域にかけて連続的に増大するように、ゲルの寸法が、ゲルの第1の領域から第2の領域にかけて連続的に増大する、請求項3に記載の方法。
- 生体材料の剛性が第1の領域から第2の領域にかけて直線的に増大するように、ゲルの寸法が、ゲルの第1の領域から第2の領域にかけて直線的に増大する、請求項4に記載の方法。
- ゲルがコラーゲンゲルである、請求項1〜5のいずれか一項に記載の方法。
- ゲルが、カーボンナノチューブのコファイバー(co-fibres)を含む、請求項6に記載の方法。
- 可塑的圧縮前にゲルに生存細胞を播種する、請求項1〜7のいずれか一項に記載の方法。
- 生存細胞が、筋肉細胞、肝細胞、腎細胞、心臓細胞、肺細胞、腸細胞、気管支細胞、眼細胞、生殖細胞、血管細胞、神経細胞、分泌細胞、幹細胞、線維芽細胞、シュワン細胞、平滑筋細胞、内皮細胞、尿路上皮細胞、骨細胞、軟骨細胞、および腱細胞からなる群より選択される、請求項8に記載の方法。
- ゲルの可塑的圧縮により、第2の領域に比較して第1の領域で細胞密度が高くなる、請求項8または9に記載の方法。
- 可塑的圧縮前に、ゲルの第2の領域に向けて細胞を沈降させる、請求項8または9に記載の方法。
- 可塑的圧縮により、第1の領域に比較して第2の領域で細胞密度が高くなる、請求項11に記載の方法。
- 一軸張力をゲルに負荷することによりゲルのフィブリルを整列させる、請求項1〜12のいずれか一項に記載の方法。
- 一軸張力が可塑的圧縮前に負荷される、請求項13に記載の方法。
- 損傷組織を修復または置換するために、ヒトまたは動物の体内に前記生体材料を埋め込むステップを含む、請求項1から14のいずれか一項に記載の方法。
- 前記生体材料をモールディングまたは成形して組織等価インプラントを製造するステップを含む、請求項1から14のいずれか一項に記載の方法。
- 前記生体材料を折りたたむか、または丸めて前記インプラントを製造するステップを含む、請求項16に記載の方法。

- 請求項1から14のいずれか一項に記載の方法により製造される生体材料。
- 請求項19に記載の生体材料を含む組織等価インプラント。
- 個体内の損傷組織を治療する方法であって、
請求項20に記載の組織等価インプラントを前記損傷組織に固定して、前記組織を修復および/または置換するステップ
を含む、方法。 - 個体内の損傷組織を治療する方法に用いるための、請求項20に記載の組織等価インプラント。
- 損傷組織の治療に用いられる薬物の製造における、請求項20に記載の組織等価インプラントの使用。
- 前記損傷組織が、関節炎疹、神経−筋損傷/変性、筋腱損傷、および加齢変性、外傷後の再生不良、組織壊死、または外科切除に起因する、請求項21に記載の方法、請求項22に記載のインプラント、または請求項23に記載の使用。
