[実施例2]
<抗老化効果(ヒト真皮繊維芽細胞タイプIコラーゲン産生促進作用の評価)>
正常ヒト真皮繊維芽細胞を1ウェル当り2.0×10個となるように96ウェルマイクロプレートに播種した。播種培地には5質量%のウシ胎児血清(FBS)を添加したダルベッコ改変イーグル培地(DMEM)を用いた。24時間後、0.5質量%FBS添加DMEM培地にて表2に示す各濃度となるようにアミノ酸及び/又は抽出物1を添加した培養液に交換し、さらに24時間培養した。培養上清中に分泌されたタイプIコラーゲンの定量にはELISA法を用い、最後は標識されたペルオキシダーゼに対し2,2’−アジノビス(3−エチルベンゾチアゾリン−6−スルホン酸)ジアンモニウム塩(ABTS)及び過酸化水素を添加し反応させた後、405nmの吸光度を測定した。PIERCE社製BCAプロテインアッセイキットにて各ウェルのタンパク量を測定し、単位タンパク量当りのタイプIコラーゲン産生量を求めた。
評価結果を試料無添加のコントロールにおける単位タンパク量当りのタイプIコラーゲン産生量を100とした時の相対値にて表2に示す。
[Example 2]
<Anti-aging effect (Evaluation of human dermal fibroblast type I collagen production promoting effect)>
Normal human dermal fibroblasts were seeded in a 96-well microplate at 2.0 × 10 4 cells per well. Dulbecco's modified Eagle medium (DMEM) supplemented with 5% by weight fetal bovine serum (FBS) was used as the seeding medium. After 24 hours, the medium was replaced with a culture solution to which amino acids and / or extract 1 were added so as to achieve the respective concentrations shown in Table 2 in a DMEM medium supplemented with 0.5% by mass FBS, and further cultured for 24 hours. The ELISA method was used for the quantification of type I collagen secreted into the culture supernatant. Finally, 2,2′-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt ( ABTS) and hydrogen peroxide were added and reacted, and then the absorbance at 405 nm was measured. The amount of protein in each well was measured with a BCA protein assay kit manufactured by PIERCE, and the amount of type I collagen produced per unit protein amount was determined.
The evaluation results are shown in Table 2 as relative values when the type I collagen production amount per unit protein amount in the control with no sample added is defined as 100.

[実施例3]
1.ヒト皮膚での弾力測定
吸引法を用いた測定機器Cutometer SEM575(Courage-Khazaaka slectronic GmbH, Cologene社, ドイツ製)を使用した。これは平滑な皮膚表面に2mmの円形の開口部を有するプローブを軽く密着させた後、プローブ内を一定の陰圧状態にすることで皮膚がプローブ内に吸引され、盛り上がり変形する際の変位した量や吸引終了までの変位量を測定することで皮膚の弾性を計測する。
試験は前腕部の3×4cmの範囲で使用前の皮膚の弾性をCutometer SEM575にて測定し、その後、各サンプル水溶液を24μLずつ範囲内に塗布した。塗布後15、60、120分にハリ値(R7)値を測定した。
評価結果を表3に示す。

The evaluation results are shown in Table 3.
