Abstract
Description
本願は、2006年7月11日に出願された米国仮特許出願第60/819,964号の利益および優先権を主張する。米国仮特許出願第60/819,964号の全開示は、本明細書中に参考として援用される。
本開示は、合成ポリマーから作製される架橋組成物、および生体接着剤および/またはシーラントとしてのそのような組成物の使用に関する。
近年、縫合を接着結合と代替する、または接着結合で増強することにおける興味が増大してきている。この興味の増大の理由として、(1)接着結合で修復が達成されると思われる潜在的な速度;(2)結合物質の、完全な閉鎖をもたらし、したがって体液の漏出を防止する能力;および(3)組織の過剰な変形を伴うことなく、結合を形成する可能性が挙げられる。
本開示は、組織中の創傷または欠損部を密封するための接着剤またはシーラントとして利用することのできる、生体適合性マクロマー組成物を提供する。実施形態では、生体適合性マクロマー組成物は、第1のヌクレオシドを含む第1のポリマーおよび第2のヌクレオシドを含む第2のポリマーを含み、この第1のヌクレオシドと第2のヌクレオシドは塩基対形成し、それによって本開示の生体適合性マクロマー組成物を形成する。
本明細書に記載される生体適合性マクロマー組成物は、動物組織を接着させるための接着剤として、または動物組織中の空隙を密封するためのシーラントとして有用となり得る。本開示の組成物は、ヌクレオシドまたはオリゴヌクレオチドもしくはポリヌクレオチドを含めたヌクレオチドで末端処理したポリマー鎖を含む、少なくとも2つの成分を使用して作製することができる。
Claims (23)
- 第1のヌクレオシドを含む第1のポリマーと、
第2のヌクレオシドを含む第2のポリマーと
を含む生体適合性マクロマー組成物であって、前記第1のヌクレオシドおよび前記第2のヌクレオシドは塩基対形成し、前記組成物は、接着剤、シーラント、または遺伝子送達デバイスとして作用する生体適合性マクロマー組成物。 - 前記第1のポリマーがオリゴヌクレオチドを含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第2のポリマーがオリゴヌクレオチドを含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマーがポリヌクレオチドを含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第2のポリマーがポリヌクレオチドを含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマー、および場合によって前記第2のポリマーの分子量が、約1×103g/molから約1×106g/molである、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマー、および場合によって前記第2のポリマーの分子量が、約1×104g/molから約1×105g/molである、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマーが、ポリアルキレンオキシド、ポリサッカライド、エステル、脂肪族エステル、オルトエステル、リン酸エステル、カーボネート、ウレタン、エーテル、アミド、フェノール類、およびその組合せからなる群から選択されるポリマーをさらに含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第2のポリマーが、ポリアルキレンオキシド、ポリサッカライド、エステル、脂肪族エステル、オルトエステル、リン酸エステル、カーボネート、ウレタン、エーテル、アミド、フェノール類、およびその組合せからなる群から選択されるポリマーをさらに含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマーが、酸、アミン、スルホン、イソシアネート、ヒドロキシル基、ビニル基、エステル、およびその組合せからなる群から選択される第1の官能基をさらに含み、前記第2のポリマーが、前記第1の官能基と架橋された第2の官能基をさらに含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマー、および場合によって前記第2のポリマーが、α−ヒドロキシ酸、エステル、ラクトン、カーボネート、エステルエーテル、二酸、およびその組合せからなる群から選択される生体吸収性基をさらに含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマー、および場合によって前記第2のポリマーが、乳酸、グリコール酸、ラクチド、グリコリド、ε−カプロラクトン、トリメチレンカーボネート、1,4−ジオキサン−2−オン、1,3−ジオキサン−2−オン、コハク酸、アジピン酸、セバシン酸、マロン酸、グルタル酸、およびその組合せからなる群から選択される生体吸収性基をさらに含む、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマーが、合成マクロマー組成物の約10重量%から約90重量%の量で存在する、請求項1に記載の生体適合性マクロマー組成物。
- 前記第2のポリマーが、合成マクロマー組成物の約90重量%から約10重量%の量で存在する、請求項1に記載の生体適合性マクロマー組成物。
- 前記第1のポリマー、および場合によって前記第2のポリマーが、酵素的に分解可能な成分と混合される、請求項1に記載の生体適合性マクロマー組成物。
- 請求項1に記載の生体適合性マクロマー組成物を含む、創傷閉鎖用の接着剤。
- 請求項1に記載の生体適合性マクロマー組成物を含む、医療用途で使用するためのシーラント。
- 創傷を閉鎖するための方法であって、
前記創傷に請求項1に記載の生体適合性マクロマー組成物を適用するステップと、
前記生体適合性マクロマー組成物を硬化させ、それによって前記創傷を閉鎖するステップと
を含む方法。 - 前記創傷が外科的切開部である、請求項18に記載の方法。
- 動物組織中の空隙を満たすための方法であって、
請求項1に記載の生体適合性マクロマー組成物を前記空隙に適用するステップと、
前記生体適合性マクロマー組成物を硬化させ、それによって前記空隙を満たすステップと
を含む方法。 - 動物組織の表面に医療デバイスを接着させるための方法であって、
請求項1に記載の生体適合性マクロマー組成物を、前記デバイス、前記表面、または両方に適用するステップと、
前記デバイス、生体適合性マクロマー組成物および表面を互いに接触させるステップと、
前記生体適合性マクロマー組成物を硬化させ、それによって前記デバイスおよび表面を互いに接着させるステップと
を含む方法。 - 前記医療デバイスがインプラントである、請求項21に記載の方法。
- 遺伝子を送達するための方法であって、
請求項1に記載の生体適合性マクロマー組成物を動物に投与するステップと、
前記生体適合性マクロマー組成物を硬化させ、それによってゲルを形成するステップと、
前記生体適合性マクロマー組成物をin vivoで分解させるステップであり、前記生体適合性マクロマー組成物の分解により、前記動物に遺伝子を送達するステップと
を含む方法。
