Abstract
Description
チオネインは、約20個のシステインアミノ酸を含む60個超のアミノ酸のタンパク質である。チオネインは芳香族残基もヒスチジン残基も含まない。メタロチオネインは1957年に発見された(Margoshes及びVallee、J.Am.Chem.Soc.79:4813−4814頁、1957年)。2つの極めて類似したMT−1型とMT−2型とが同定されたが、最近になって、第3のMT−3型が成長阻害因子としてアルツハイマー患者の脳において同定された(Uchidaら、Neuron 7:337−347頁、1991年)。第4の変異体MT−4もまた、重層扁平上皮においてのみ発現することが分かった(Quaifeら、Biochemistry 33:7250−9頁、1994年)。さらなる(全部で17個もの数の)MTアイソフォームをコードする遺伝子もまた同定されている。
メタロチオネイン及びチオネインに依拠する機能の1つは、細胞内亜鉛レベルの調節である(Jacobら、Proc.Natl.Acad.Sci.USA 95:3489−3494頁、1998年)。亜鉛は、数多くの酵素における重要な補因子である。MT/Tとの亜鉛結合は強力だが(pH7.4での結合定数が3.2×10−13M)、MT/Tは大腸菌(Esherichia coli)アルカリホスファターゼ及びウシカルボキシペプチダーゼAなどの亜鉛酵素に対し亜鉛を提供できることが示されている。
先行研究(Maret及びVallee、Proc.Natl.Acad.Sci.USA 95:3478−3482頁、1998年)から、亜鉛それ自体は酸化還元不活性だが、メタロチオネイン及びチオネインには酸化還元活性があることが示されている。酸化剤がMT/T中でシステインを還元し、それによりタンパク質からの金属の同時放出が生じることが示されている。このように、MT/Tは細胞における酸化状態の維持に関与しているものと思われる。
ヌクレオチド三リン酸は、アデノシン5’[γ−チオ]三リン酸及びAMP−PNPなどのATP、GTP、及びATP類似体を含め、MT/Tと結合するとMT/Tからの金属の放出を生じさせる(Jiangら、Proc.Natl.Acad.Sci.USA 95:9146−9149頁、1998年)。この結合は、哺乳動物のメタロチオネイン及びチオネイン中に見られる8個の保存されたリシン残基により媒介される。
メタロチオネインは一般に金属と結合した形態で観察されるが、チオネインは生物学的材料中に検出され、およびそこから単離されており(Maretら、Proc.Natl.Acad.Sci.USA.96:1936−1940頁、1999年)、内因性キレート剤として作用する。
メタロチオネイン又はチオネインは、チオネインを自然に産生する任意の生物体又はチオネインを産生するよう改変された生物体から単離、精製又は分画できる。ある実施形態においては、ヒト、ウシ、又はウマチオネインを単離、精製又は分画できる。メタロチオネインはまた、その免疫学的特性、又はその反応特性(例えば、本明細書に記載される任意の金属、任意のヌクレオチド、ヌクレオシド、又は任意の他の化合物若しくはポリペプチドとの)について評価できる。ある実施形態において、MT/T又は本明細書に記載される任意の変異体の、AMP、ADP、ATP、GSH、GSSG、又はこれらの任意の組み合わせとの相互作用が研究される。
一態様において、本発明は、1個又は複数の硫黄原子がセレンと置換されたメタロチオネイン又はチオネインを特徴とする。置換メタロチオネイン又はチオネインは、0、1、2、3、4、5、6、7個又はそれ以上の金属原子に結合し得る。ある実施形態において、システインがセレノシステインと置換される(例えば、本明細書に記載されるとおり、任意の、又は全てのシステインにおいて)。セレノシステインは既知の生理的分布を有し、非毒性で、耐容性が十分に高い。従って、セレノシステインを含有するタンパク質は、治療剤として(例えば、本明細書に記載されるものなどの酸化ストレスに関連する疾患の治療において)有用であり得る。
本発明はまた、1つ又は複数の非システイン残基の種々のアミノ酸(例えば、天然に存在する、又は天然に存在しないアミノ酸)との置換を伴うMT又はT変異体も特徴とする。さらに、本発明はまた、1個又は複数の硫黄原子のセレン(例えば、システイン残基のセレノシステインとの)置換を伴うMT又はTも特徴とする。MT/T変異体は、メタロチオネインのα又はβドメインの一次配列の1つ又は複数の反復(例えば、1つ又は複数のアミノ酸のスペーサー配列により分離される)を含む。MT/T変異体は、任意の順序で連結されたαドメイン(例えば、1、2、3、4、5、8、10、12個、又はそれ以上)又はβドメイン(例えば、1、2、3、4、5、8、10、12個、又はそれ以上)の任意の組み合わせを含み、場合によりドメイン間に1つ又は複数のスペーサーを伴い得る。ドメインはさらに、任意の非システインアミノ酸又はシステインの、セレノシステインなどのセレンを含む残基との置換を含み得る。本発明に包含される変更を含むドメインは、参照により本明細書に援用される国際公開第00/50448号パンフレットの12頁4行目から14頁5行目にかけて記載される。国際公開第00/50448号パンフレットに記載される変更はまた同様に、完全長メタロチオネインタンパク質、その任意の断片、又は本明細書に記載される任意の他の変異体に導入されてもよい。
ここで本出願人は、メタロチオネイン及びチオネインの亜鉛結合及び調節タンパク質としての同定、並びにMT/T亜鉛結合を伴う細胞酸化におけるその役割に基づき、MT/Tの亜鉛結合活性の、MT/Tの酸化との分離を探る。従って、本発明は、(i)金属のMT/Tに対する結合性を低下させ、および(ii)MT/T又は第2のポリペプチドの酸化を実質的に増進しない、化合物を同定するためのスクリーニング方法を特徴とする。本発明の方法により同定された化合物により、メチオニンスルホキシド残基を含むメタロチオネイン又はチオネインなどの害を及ぼす可能性のある酸化種を還元するために利用可能なチオネインの利用能が増加し得る。酸化アミノ酸が疾患進行において役割を果たす疾患の具体例としてはメチオニンスルホキシドを含む酸化されたα−シヌクレインが同定されたパーキンソン病(Glaserら、Biochim.Biophys.Acta.1703:157−69頁、2005年)、及びメチオニンスルホキシド残基を含む酸化されたβ−アミロイドタンパク質が同定されたアルツハイマー病(Schoneich、Biochim.Biophys.Acta.1703:111−9頁、2005年)を含む。酸化ストレスに関連する疾患の他の例は、本明細書に記載される。このように、本発明のスクリーニング方法により同定される化合物は、酸化ストレスに関連する疾患の治療において有用であり得る。

本発明のスクリーニング方法は、MT/T又は本明細書に記載されるその任意の断片若しくは変異体からの金属の放出を判定する工程を含む。金属放出を判定するための当該技術分野において周知の任意の方法を使用できる。
本発明のスクリーニング方法はまた、第2のポリペプチド(例えば、MT、T、又は本明細書に記載される任意の断片若しくは変異体)のアミノ酸の酸化を計測する工程、又は試験化合物の細胞の酸化状態(例えば、酸化アミノ酸又は活性酸素種の形成)が実質的に増進したかどうかを判定する工程も含む。
チオネイン(例えば、MT−3)発現を増加させる化合物を同定するためのスクリーニングアッセイの実施にあたっては、あらゆる方法が利用可能である。一手法に従えば、メタロチオネインをコードするポリヌクレオチドを発現する細胞の培養培地に対し、候補化合物が様々な濃度で添加される。次に遺伝子発現が、例えば、標準的なノーザンブロット解析(Ausubelら、「Current Protocols in Molecular Biology」、Wiley Interscience、ニューヨーク(New York)、1997年)により、ポリヌクレオチド分子からハイブリダイゼーションプローブとして調製された任意の然るべき断片を使用して計測される。候補化合物の存在下での遺伝子発現レベルが、候補分子のない対照培養培地において計測されるレベルと比較される。チオネイン発現(例えば、MT−3)の増加を促進する化合物が、本発明においては有用と見なされる。かかる分子は、例えば、酸化ストレスに関連する疾患(例えば、本明細書に記載されるもの)用の治療剤として使用できる。
一般に、酸化ストレスに関連する疾患の治療能を有する化合物は、当該技術分野において周知の方法に従い、天然産物又は合成(又は半合成)抽出物の双方の大型ライブラリ又は化学ライブラリから同定される。創薬及び新薬開発分野の当業者は、試験抽出物又は化合物の正確な供給源が本発明のスクリーニング方法にとって決定的に重要なものではないことは理解するであろう。従って、事実上、あらゆる化学的抽出物又は化合物が、本明細書に記載される方法を使用してスクリーニングされ得る。かかる抽出物又は化合物の例としては、植物、真菌、原核生物又は動物ベースの抽出物、発酵ブロス、及び合成化合物、並びに既存化合物の修飾物を含むが、これらに限定はされない。サッカリド、脂質、ペプチド、及びポリヌクレオチドベース(例えば、siRNA又はマイクロRNA)化合物を含むが、これらに限定はされないあらゆる化学的化合物のランダムな、又は特異的な合成(例えば、半合成又は全合成)を生じさせるためにもまた、数多くの方法が利用可能である。本発明のスクリーニング方法に使用され得るさらなる化合物としては、本明細書に記載される任意の化合物(例えば、キレート剤、修飾チオネイン、並びにセレン化合物(例えば、セレノシスタミン、塩化ベンゼンセレネニル、及びベンゼンセレニン酸))が含まれる。これらの化合物のいずれも、当該技術分野で標準的な方法を使用して化学的に修飾され得る。
本発明はまた、金属(例えば、亜鉛)結合性の低減した修飾チオネイン(T)を生成するための方法も特徴とする。ある実施形態において、修飾チオネインは、野生型チオネインと比較した場合、酸化還元反応に関与する能力をさらに保持し得る。かかる修飾チオネイン分子は、酸化ストレスに関連する疾患の治療において有用であり得る。修飾は、当該技術分野において周知の任意の手段により実施され得る。配列の改変(例えば、点突然変異、挿入、欠失、又はこれらの任意の組み合わせ)を導入するための方法は、当業者に周知である。
チオネインは、任意の残基において修飾されてもよく(例えば、化学的誘導体化又は点突然変異により)、1個又は複数の(例えば、2、3、4、5、6、7、8、10、15、又は20個の)アミノ酸の挿入又は欠失により修飾されてもよい。本明細書に記載されるチオネインへの任意の修飾が、本発明の方法において使用され得る。こうした修飾は、例えば、標準的な分子生物学的技術を使用することにより実施され得る。ある実施形態においては、リシン残基が改変されることにより金属(例えば、亜鉛)結合性の低下が生じる。かかる改変としては、チオール、スルフェン酸、スルフィン酸、スルホン酸、スルホン酸エステル、スルホキシド、又はスルホン部分の付加又は置換を含むことができる。これらのリシン残基としては、今のところは未知の機序でATP結合(Jiangら、上記を参照)に関与しているMT1又はMT2(配列番号1及び2)の20、22、25、30、31、43、51、及び56位における8個のリシン残基、MT3(配列番号3)の21、26、31、32、44、47、52、及び63位におけるリシン残基、又はMT4(配列番号4)の21、28、32、44、52、又は57位におけるリシン残基を含む。上述のとおり、ATP結合は、MTの金属に対する親和性を低下させる。従って、チオネイン中のリシン残基(例えば、上述されるもの)若しくはリシン残基に隣接する残基、又はチオネインの三次元構造に基づいたリシン残基に近接している残基の修飾が、AMP、ADP、ATP、GSH、又はGSSG結合性、又はそれらの任意の組み合わせの亢進について修飾及び試験され得る。ATP結合性、又は他のヌクレオチド三リン酸若しくはその類似体の結合性を増進する修飾は、チオネインの金属(例えば、亜鉛)に対する親和性を低下させ得るとともに、従って特に本発明の方法に有用であり得る。
金属親和性及び結合性についての測定は、本明細書に記載されるか、又は当該技術分野において周知のとおり行うことができる。かかる測定を使用して、非修飾チオネインと比較したとき、どのチオネイン変異体が、亜鉛、銅、カドミウム、鉛、銀、ガドリニウム、コバルト、カルシウム、金、セレン、ヒ素、タングステン、アルミニウム、マンガン、鉄、クロム、ニッケル、モリブデン、バリウム、ストロンチウム、ビスマス、ハフニウム、テクネチウム、又はランタンなどの金属の結合性の低減を呈するかを判定できる。
ある実施形態において、修飾チオネインはさらに、酸化還元活性について測定される。用いられる正確な方法は本発明にとって決定的に重要なものではなく、かかる計測は当該技術分野において周知の任意の方法を使用して行われ得る。N末端メチオニンの酸化還元状態は、例えば、修飾GFP又は例えば本明細書に記載されるとおりの市販のキットを使用した酸化還元電位計測を含む上記の方法により判定され得る。他の実施形態においては、溶液中のタンパク質の酸化還元電位が、例えば、Broadley James Corporation、カリフォルニア州アーバイン(Irvine,Calif.)から市販されている電極を使用して計測され得る。
修飾チオネインポリペプチドは、好適な発現媒体中のチオネインをコードするポリヌクレオチド分子の全部又は一部若しくはその断片による好適な宿主細胞の形質転換により産生できる。分子生物学分野の当業者は、多種多様な発現系のいずれを使用しても、チオネインポリペプチドを提供し得ることを理解するであろう。使用される正確な宿主細胞は本発明にとって決定的に重要なものではない。修飾チオネインは、原核生物宿主(例えば、大腸菌(E.coli))又は真核生物宿主(例えば、出芽酵母(Saccharomyces cerevisiae)、昆虫細胞、例えば、Sf21細胞、又は哺乳動物細胞、例えば、NIH 3T3、HeLa、又は好ましくはCOS細胞)において産生できる。かかる細胞は広範な供給源から入手可能である(例えば、American Type Culture Collection、メリーランド州ロックランド(Rockland,Md.);また、例えば、Ausubelら、上記も参照のこと)。形質転換又はトランスフェクションの方法及び発現媒体の選択は、選択される宿主系に依存し得る。形質転換及びトランスフェクション方法は、例えば、Ausubelら(上記)に記載され、発現媒体は、例えば、「Cloning Vectors:A Laboratory Manual」(Pouwels,P.H.ら、1985年、補遺1987年)に提供されるものから選択されてもよい。
本発明は、酸化ストレスに関連する疾患を有する対象者を治療するための方法を特徴とする。例えば、本発明の方法の治療において使用される化合物は、本明細書に記載されるスクリーニング方法を使用して同定される化合物、(例えば、本明細書に記載される)修飾チオネイン、又はキレート剤であってもよい。
酸化ストレスに関連する疾患としては、アルツハイマー病、パーキンソン病、クロイツフェルトヤコブ病、筋萎縮性側索硬化症、呼吸窮迫症候群、筋ジストロフィー、白内障形成、関節リウマチ、早老症、ウェルナー症候群、アテローム性動脈硬化症、糖尿病、本態性高血圧症、嚢胞性線維症、限局性回腸炎(クローン病)、黄斑変性症、卒中、虚血、及び潰瘍性大腸炎が挙げられる。
修飾チオネインタンパク質(例えば、本発明の、又は本明細書に記載される方法により同定されるもの)が、酸化ストレスに関連する疾患を治療するため対象者に投与され得る。修飾チオネインを対象者に提供することにより、酸化ストレスの作用が低減して、酸化ストレスに関連するかかる疾患を治療できる。
修飾チオネインタンパク質の投与に加え、遺伝子ベクターを対象者に導入することによりチオネインをコードするポリヌクレオチド(例えば、本明細書に記載される修飾チオネイン)の発現を誘導してもまた、酸化ストレスに関連する疾患を治療できる。かかる投与には、任意の標準的な遺伝子治療ベクター及び方法を用いることができる。
亜鉛などの金属をMTから除去する能力を有するキレート剤もまた、クロイツフェルトヤコブ病、呼吸窮迫症候群、ジストロフィー、白内障形成、関節リウマチ、早老症、ウェルナー症候群、アテローム性動脈硬化症、糖尿病、本態性高血圧症、嚢胞性線維症、限局性回腸炎(クローン病)、黄斑変性症、卒中、虚血、及び潰瘍性大腸炎などの酸化ストレスに関連する疾患の治療において使用され得る。かかる薬剤は、MTから金属を除去し、それによりアポタンパク質Tを酸化還元反応に関与させて酸化ストレスを軽減し得る。
本明細書に記載されるか、又は本発明のスクリーニング方法を使用して同定される任意の化合物の投与は、酸化ストレスに関連する疾患を治療するための化合物の濃度をもたらす任意の好適な手段により得る。化合物は、任意の然るべき量で任意の好適な担体物質中に含まれてもよく、一般的には、組成物の総重量の1〜95重量%の量で存在する。組成物は、経口、非経口(例えば、静脈内、筋肉内、頭蓋内、髄腔内)、経直腸、経皮、経鼻、経膣、吸入、皮膚(パッチ)、眼内、又は頭蓋内の投与経路に好適な剤形で提供され得る。医薬組成物は、従来の薬務に従い調合できる(例えば、Remington:「The Science and Practice of Pharmacy」、第20版、2000年、A.R.Gennaro編、Lippincott Williams & Wilkins、Philadelphia、及び「Encyclopedia of Pharmaceutical Technology」、J.Swarbrick及びJ.C.Boylan編、1988−1999年、Marcel Dekker、ニューヨーク(New York)を参照)。
本明細書に記載された、又は本発明の方法を使用して同定される化合物を含む組成物は、注射、注入、又は埋め込み(皮下、静脈内、筋肉内、腹腔内、頭蓋内、髄腔内など)により、投薬形態で、製剤で、もしくは好適な送達器具又は従来の非毒性の薬学的に許容可能な担体及びアジュバントを含むインプラントを介して非経口的に投与できる。かかる組成物の調合及び調製は、医薬製剤分野の当業者に周知である。
多くの場合、投与される化合物は、特定の疾患に侵されている対象者の1つ又は複数の組織に限定されることが望ましい。アルツハイマー病又はパーキンソン病などの神経系を侵す疾患の場合、神経系の罹患領域への送達は、例えば、以下に概略が述べられる方法により実現できる。
本明細書に記載されるか、又は本明細書に記載される方法を使用して同定される任意の化合物の投薬量は、投与方法、治療される疾患、障害又は疾患の重症度、障害又は疾患が治療されるべきなのかまたは予防されるべきなのか、並びに治療対象者の年齢、体重、及び健康状態を含む、いくつかの要因に依存する。
一例において、セレノシステイン置換メタロチオネインは、基本的に国際公開第00/50448号パンフレットに記載されるとおり合成される。簡潔に言えば、この方法は、(a)固形担体と、脂肪族基を含有する側鎖(例えば、水素又はアルキル)を含むアミノ酸、芳香族基を含有する側鎖を含むアミノ酸、硫黄族を含有する側鎖(例えば、チオール又はチオエーテル)を含むアミノ酸、ヒドロキシル基を含有する側鎖を含むアミノ酸、アミン基を含有する側鎖を含むアミノ酸、グアニジニウム基を含有する側鎖を含むアミノ酸、カルボキシレート基を含有する側鎖を含むアミノ酸、及びアミド基を含有する側鎖を含むアミノ酸からなる群から選択されるαアミノ基を有する少なくとも2個のαアミノ酸とを使用して、MT又はTを合成する工程を含む。αアミノ基は、Fmoc、t−Boc、又はCBZで保護される。カルボキシレート基は、t−ブチルエステル又はベンジルエステルで保護される。ヒドロキシル基は、t−ブチルエーテル又はジメチルホスフェートエステルで保護される。アミン基は、t−Boc又はCBZで保護される。チオール基は、アセトイミドメチル基で保護される。工程(a)に続くこの方法の工程(b)は、工程(a)で固形担体から合成されたペプチドを切断するとともに非アセトイミドメチル保護基を除去する工程を含む。工程(c)は工程(b)から得られたペプチドを精製する工程を含み、及び工程(d)は工程(c)から得られたペプチドを沈殿させる工程を含む。工程(e)はアセトイミドメチル保護基を銀(I)塩を含む溶液で除去する工程を含む。MT又はTの一次アミノ酸配列は、野生型配列と異なり得る。例えば、このアミノ酸配列は、1つ又は複数の非システイン残基の、任意の天然に存在する、又は天然に存在しないアミノ酸との置換を含み得る。ある実施形態において、1つ又は複数のシステイン残基がセレノシステインと置換される。他の修飾としては、MTのα又はβドメインの一次配列の1つ又は複数の反復の付加を含む。これらの反復は、α及びβドメインが順番に、又は任意に並んだ状態で共に融合され得る。ドメインは、1つ又は複数のアミノ酸のスペーサー配列により分離され得る。反復されたドメインにおいて、1つ又は複数のシステイン残基がセレノシステインと置換され得る。
Claims (33)
- メタロチオネイン又はチオネインと実質的に同一のアミノ酸配列を含むポリペプチドであって、少なくとも1個の硫黄原子がセレンと置換されている、ポリペプチド。
- 前記硫黄原子が前記ポリペプチドのシステイン残基中にある、請求項1に記載のポリペプチド。
- 前記ポリペプチドの10個のシステイン残基がセレノシステインと置換されている、請求項2に記載のポリペプチド。
- 前記ポリペプチドの全てのシステイン残基がセレノシステインと置換されている、請求項2に記載のポリペプチド。
- メタロチオネイン又はチオネインの断片を含むポリペプチドであって、少なくとも1個の硫黄原子がセレンと置換され、前記断片が金属との結合能を有する、ポリペプチド。
- 前記硫黄が前記ポリペプチドのシステイン残基中にある、請求項5に記載のポリペプチド。
- 前記ポリペプチドの全てのシステイン残基がセレノシステインと置換されている、請求項5に記載のポリペプチド。
- 前記断片がメタロチオネイン又はチオネインのα−ドメイン又はβ−ドメインを含む、請求項5に記載のポリペプチド。
- 前記金属が亜鉛である、請求項5に記載のポリペプチド。
- 酸化ストレスに関連する疾患の治療用候補化合物を同定するための方法であって、
(a)化合物をメタロチオネイン及び被酸化能を有するアミノ酸を含む第2のポリペプチドと接触させる工程と、
(b)前記化合物の存在下における、前記メタロチオネインから放出された金属の量及び前記第2のポリペプチド上の酸化アミノ酸の形成を計測する工程と、
を含み、前記化合物の不在下と比較したとき(i)メタロチオネインからの金属の放出を増加させ、および(ii)前記第2のポリペプチド中の前記酸化アミノ酸の量を実質的に増加させない化合物により、前記化合物が酸化ストレスに関連する疾患の治療用候補化合物であることが示される、方法。 - 前記化合物が化学ライブラリから選択される、請求項10に記載の方法。
- 前記酸化アミノ酸がメチオニンスルホキシドである、請求項10に記載の方法。
- 前記金属が、亜鉛、銅、カドミウム、鉛、銀、ガドリニウム、コバルト、カルシウム、金、セレン、ヒ素、タングステン、アルミニウム、マンガン、鉄、クロム、ニッケル、モリブデン、バリウム、ストロンチウム、ビスマス、ハフニウム、テクネチウム、又はランタンである、請求項10に記載の方法。

(a)細胞又は細胞抽出物を化合物と接触させる工程と、
(b)前記細胞又は細胞抽出物についてメタロチオネイン又はチオネイン量及び酸化状態を計測する工程と、
を含み、前記化合物と接触していない細胞又は細胞抽出物と比較したとき(i)チオネインの量を増加させるか、又はメタロチオネインの量を低下させるとともに(ii)前記細胞又は細胞抽出物の酸化状態を実質的に増進しない化合物により、前記化合物が酸化ストレスに関連する疾患の治療用候補化合物であることが示される、方法。 - 前記化合物が化学ライブラリから選択される、請求項18に記載の方法。
- 前記酸化状態の計測が、酸化アミノ酸の存在の検出を含む、請求項18に記載の方法。

を含み、前記化合物の不在下と比較したときの存在下における発現の増加により、前記化合物が酸化ストレスに関連する疾患の治療用候補化合物であることが示される、方法。 - 前記化合物が化学ライブラリから選択される、請求項23に記載の方法。

- 金属に対する親和性の低減したチオネイン変異体を同定するための方法であって、
(a)点突然変異、挿入、又は欠失をチオネインに導入するか、又はチオネインを化学的に変性させることにより修飾チオネインを作成する工程と、
(b)前記金属の前記修飾チオネインとの親和性を判定する工程と、
を含み、前記金属に対する親和性の低下により、前記修飾チオネインが金属に対する親和性の低減したチオネイン変異体であることが示される、方法。 - 前記判定工程(b)が前記修飾チオネインの活性の低減を計測する工程をさらに含み、前記修飾チオネインの活性の低減について実質的な低下がないことにより、前記修飾チオネインが金属に対する親和性の低減した酸化還元活性のあるチオネイン変異体であることが示される、請求項26に記載の方法。
- 前記点突然変異がシステインのセレノシステインへの点突然変異を含む、請求項26に記載の方法。
