Description
本出願は、引用により本明細書に組み込まれている2006年2月21日出願の米国特許出願第60/775,132号の優先権を主張するものである。
技術分野は、いくつかの態様において、免疫系を活性化するための表面化学作用を有するナノ粒子に関する。
抗原に対する耐性を発達させること、又はそれを拒絶することを習得すること等による所望の方法で抗原に応答するように免疫系を訓練することができれば、多くの医学的メリットを実現することが可能である。この課題に対処するために、全身薬物治療、抗原の注入及び抗体治療を含む様々な手法が試みられてきた。
本明細書に開示されている手法のなかで、1つの新しい手法は、治療物質を体内の特定箇所の抗原提示細胞(antigen-presenting cell)(APC)に特異的に標的化する。APCは、典型的には樹状細胞及びマクロファージであり、場合によってはB細胞である。本明細書において、APCは、樹状細胞及びマクロファージのみを表すのに用いられる用語であり、B細胞は除外される。APCは、体全体に散在するが、この手法は、作用物質を特定箇所、すなわちリンパ節のAPCに標的化する。APCは、リンパ節における作用物質の吸収が有利になるように、リンパ節では他の箇所とは異なる挙動を示す。さらに、媒体は、その効果を果たすことができるように数時間又は数日間にわたって残留するとともに、生分解可能である。APCは、リンパ節において特異的に標的化されるだけでなく、治療薬の送達媒体が特定の方法で、すなわち補体系を活性化することによってAPCを活性化する。補体系を活性化することは、適切な免疫治療薬を選択できるように既知の応答経路を含む。さらに、補体系は、生物剤を含まない送達系における合成物質によって活性化される。これらの特異的に標的化される特徴のすべての結果は、所望の免疫治療を達成するようにAPCを活性化する時間と場所において治療薬をAPCに遍く送達する媒体である。媒体そのものは、コンフリクト、交叉反応又は免疫系の望ましくないアンタゴニズムを生じることなく任意の作用物質を容易に受け入れるような生体分子又はポリペプチドを含まない。
(発明の概要)
好適な特性を有するナノ粒子を使用して、治療物質をリンパ節の樹状細胞(dendritic cell)(DC)を含む抗原提示細胞(antigen-presenting cell)(APC)に特異的に標的化することができる。ナノ粒子粒径のリンパ吸収、リンパ節滞留及びリンパ節APC及びDCによる内在化に対する影響を、ここでは、マウス(ヒトと同様の大きさのリンパ毛細血管(10〜80μm)を有するが、これらは両種において大きく変動する[20、42])における皮内注入に対応して実証する。様々なナノ粒子粒径を用いることができるが、約20nmの直径のナノ粒子が最も容易に吸収され、さらに、他の粒子について既に報告されたものより長い時間(120時間まで)にわたってリンパ節に滞留する[31〜34、36]。驚いたことには、特定のナノ粒子表面化学作用は、補体を活性化することが実証される。リンパ節内部で、補体活性ナノ粒子は、標的化リガンドがなくても、常在APC(DC及びいくつかのマクロファージを含むMHCII+細胞)及び他の非抗原提示マクロファージ(MHCII-)によって効率的に内在化されることが示された。リンパ節常在DCの大部分(50%まで)が20nmの補体活性ナノ粒子を内在化させ、その数が時間とともに増加した。補体活性ナノ粒子内在化の後にDC成熟が生じることが見いだされた。
抗原提示細胞(antigen-presenting cell)(APC)は、MHCクラスI、II及び他の共刺激性分子(すなわちCD86及びCD80)を利用して原性T細胞を刺激し、細胞媒介免疫を誘発する極めて効率的な食細胞である。いくつかのマクロファージ及びより強力な樹状細胞(dendritic cell)(DC)を含むAPCは、末梢組織に存在し、外来抗原の内在化及び処理に続いて、T細胞に対する抗原提示を目的として、成熟し、リンパ節に移動するセンチネルとして作用する[1〜3]。APC及びDCが順応免疫において果たす重要な役割について、これらの細胞にDNA、タンパク質及びポリペプチドのような免疫調節剤を標的化するための様々な実験が行われている[4〜14]。ポリペプチドは、互いに結合した2つ以上のアミノ酸を指す用語であり、タンパク質を含む。
本明細書に記載されているように、粒径は、リンパ節におけるナノ粒子吸収及び滞留に関連する。ナノ粒子のリンパ吸収、リンパ節滞留並びにリンパ節内及び細胞集団間における局在化は、本明細書に記載されている。粒径及び表面特性のようなナノ粒子の特性は、矛盾する効果を有し得るため、従来の手法を用いる1つの課題は、効率的なリンパ吸収及びリンパ節滞留の双方を得ることである。概して、小さい粒子は、大きい粒子よりリンパ吸収が良好であるが、リンパ節滞留が劣る。直径約5nmから約100nmのナノ粒子が好ましい。すべての範囲及び明記された範囲内の値、例えば、20、25、30、35、40、45、50、55、60、65、70、75又は80nmが考えられることを当業者は直ぐに理解するであろう。約5から約100nmの平均直径を有する粒子の集合体でナノ粒子を製造することができる。すべての範囲及び明記された範囲内の値、例えば、約10から約70nmが考えられることを当業者は直ぐに理解するであろう。粒子集合体の平均直径付近の変動係数(標準偏差を平均粒径で割った値)を約50、約35、約20、約10又は約5nm未満にできるように、当該粒子の集合体の粒径分布を調節することが可能である[39]。すべての範囲及び明記された範囲内の値が考えられることを当業者は直ぐに理解するであろう。
使用に際して、当該ナノ粒子は、例えば、リンパ節のAPC、特にDCに標的化するための抗原及び薬物媒体として有用である。抗原及び/又は薬物及び/又は危険信号をリンパ節のDCに送達する能力は、免疫治療の有用な手法である。リンパ節DC及び他のAPCにナノ粒子を効果的に標的化する能力は、抗原タンパク質及びポリペプチド並びに抗原コード核酸を送達するための方法を提供する。DCは、T細胞に対する抗原提示による細胞媒介免疫の開始に関与するため、この送達手法をいくつかのワクチン及び免疫治療用途に利用することができる。さらに、ナノ粒子は、診断ツール(例えば画像診断)、研究ツール(例えば、ALDRICHカタログで販売、又は顕微鏡を使用する視覚化に対応)又はインビトロ薬物送達又は視覚化(薬物又は画像化剤のインビトロのAPC及び/又はDC及び/又はマクロファージ吸収)として有用である。
免疫抑制は、臨床移植(例えば同種移植)の状況及び自己免疫疾患(例えば多発性硬化症)において大いに必要とされる免疫治療の重要な形態である。コルチコステロイド(例えばシクロスポリンA)及びラパマイシンのような免疫抑制薬の使用は、これらの免疫障害の治療に大きな進歩をもたらした[122]。T細胞は、一般に、炎症性サイトカイン、主としてIL-2及びTNF-γに対する遺伝子を阻害することによって作用するため、T細胞集団を減少させる免疫抑制薬の重要な標的であると考えられている。免疫抑制を生じさせるための別の手法は、T細胞受容体CD28及びCD40をブロックするための抗体の使用である[123]。これらの受容体をブロックすると、DC上に位置する共刺激性分子による活性が不十分になるため、T細胞増殖を効果的に防ぐ耐性効果が生じる。しかし、コルチコステロイド又はブロッキング抗体による治療は、非常に非特異的であり、他の感染を退治する免疫系の能力を低下させるなどの副作用をもたらし得る。したがって、より特異的な免疫抑制のための方法、すなわち抗原特異的な耐性を誘発する方法は、免疫治療における格別の進歩である。
タンパク質抗原を内在化させるAPCは、抗原ペプチドエピトープを処理し、MHC-I及びII経路を通じて提示することが可能である。1つの免疫治療手法は、1つ以上の抗原をナノ粒子に共有結合させるか、或いはそれらを会合させることを含む。抗原は、免疫系によって認識され得るグリコシル化を伴わない、又は伴うポリペプチドであり、一般には、少なくとも約3つのアミノ酸の長さを有する。抗原をDNA又はRNAなどの核酸配列を介してコードすることもできる。例えば、DNAは、病原に存在するポリペプチド抗原をコードする場合は抗原をコードすることが可能である。DNAのAPCの核への送達に続いて、抗原ポリペプチド抗原の発現が生じMHCI上にそれが提示される。全タンパク質を使用できるが、抗原断片も使用することができるため、約3から約20の残基を有するポリペプチドを使用することができる。すべての範囲及び明記された範囲内の値、例えば、約10未満の残基が考えられることを当業者は直ちに理解するであろう。ポリペプチドを必要とせずに補体系を直接活性化するナノ粒子を使用できるため、補体系を活性化しない抗原を使用することができる。
いくつかのナノ粒子集合体は、ナノ粒子及び核酸、例えば、抗原をコードするDNA又はRNAを含むことができる。さらに、これらは、発現カセットを含み、プロモータ又はエンハンサを含み、ベクターの一部となり、或いはこれらの技術分野で知られている遺伝子送達モチーフを取り入れることもできる(例えば、明確に開示されていることに矛盾しない範囲で引用により本明細書に組み込まれている米国特許第7,16,0695号、同第7,157,089号、同第7,122,354号、同第7,052,694号、同第7,026,162号、同第6,869,935号参照)。さらに、該配列は、他の治療用生体分子をコードすることができる。DNAなどの核酸抗原を抗原及び他の作用物質について本明細書に記載されているナノ粒子(例えば、PLURONIC安定化PPSナノ粒子の表面)に結合させることができる。そして、例えば、静電吸着、ポリマー-ビオチン化を通じて、DNAポリマー結合を行うことができる[139]。本明細書における使用に合わせて構成できる、ポリマーをDNAに結合するためのいくつかの他の化学結合方法が存在する[140]。本明細書に記載されているナノ粒子を使用することによって、リンパ節DCを含むAPCは、抗原遺伝子発現のために標的化されるとともに、活性化されて、適切なT細胞刺激を確保することができる。
抗原を求核又は求電子官能基に結合させるための様々なスキームが利用可能である。概して、当該スキームを薬物又は危険信号の結合に適宜適応することができる。
例示を目的として、実施例のPPSナノ粒子におけるPLURONICに対する抗原の結合を示す。PLURONIC(PEG及びPPGのブロック共重合体)表面をタンパク質又はペプチドで官能化することによって、PPSナノ粒子に対する抗原結合を達成することができる。PLURONIC F127(Sigma)、ジビニルスルホン(Fluka)、水素化ナトリウム(Aldrich)、トルエン(VWR)、酢酸(Fluka)、ジエチルエーテル(Fisher)、ジクロロメタン(Fisher)及びセライト(Macherey Nagel)を受け取った状態で使用した。反応をアルゴン(Messer)下で実施した。1H NMRを重水素化クロロホルム(Armar)中で測定し、化学シフト(δ)を0.0ppmにおける内部標準テトラメチルシラン(Armar)信号に対して相対的にppmで示す。15g(1.18mmol)のPLURONIC F-127を400mlトルエンに溶解させた溶液を、Dean-Starkトラップを使用して4時間にわたって共沸蒸留によって乾燥した。溶液を氷浴で冷却し、0.283g(11.8mmol)水素化ナトリウムを添加した。反応混合物を15分間にわたって撹拌し、3.55ml(35.4mmol)ジビニルスルホン(Sigma-Aldroch)を迅速に添加した。室温にて暗所で5日間撹拌した後に、1.35ml(23.6mmol)の酢酸を添加することによって反応をクエンチした。セライトで濾過し、濾液を減圧下で小容量まで濃縮した後に、生成物を1リットルの氷冷ジエチルエーテル中に析出させた。固体を濾別し、最小量のジクロロメタンに溶解させ、氷冷ジエチルエーテルに全部で4回析出させた。ポリマーを真空下で乾燥させて、6.0gとし、OVA連結前に-20℃にてアルゴン下で保存した。NMRは、ビニルスルホンが存在し、官能化度が88%であることを示した。δ = 1.1 (m、CH3、PPG)、3.4 (m、CH、PPG)、3.5 (m、CH2、PPG)、3.6 (PEG)、6.1 (d、CHcis=CH-SO2)及び6.4 (d、CHtrans=CH-SO2)、6.85(dd、CH2=CHSO2-)。
天然ポリペプチド抗原に対して外来性のアミノ酸との共有結合、天然抗原に対して内性のアミノ酸との共有結合、及び生理化学的吸着等を含む他の手段によって、抗原をナノ粒子に連結させることができる。
他の免疫治療手法は、危険信号の適用を含む。本明細書における結果は、PLURONIC安定化補体活性(つまりヒドロキシル化)PPSナノ粒子が、共刺激性分子CD86及びCD80の発現の増強によってわかるように、DCを成熟させる危険信号又は刺激の機能を与えることを示している。ナノ粒子は、非補体危険信号又は成熟信号を必要としないが、場合によっては、当該信号を追加することが、免疫応答の発達にさらに役立つこともある。抗原吸収後、炎症性サイトカイン(すなわち、CD40リガンド)及び/又はToll様受容体(例えば、LPS及びCpG DNA)の活性体などの危険信号は、DCを成熟させ、続いて細胞媒介免疫を誘発する必要があることを他のDC標的化についての研究によって示唆された[113〜115]。危険信号は、遺伝子NF-κBの上方制御をもたらし、それが次にAPCの成熟及び炎症性サイトカインの放出をもたらす生体分子として識別される。そのような場合、例えば、タンパク質又はペプチド結合について既に記載されたプロトコルに従って、CD40リガンド、GM-CSF又はToll様受容体活性体などの危険信号サイトカインを(例えば、コア若しくは表面層又は親水性ポリマー層において)ナノ粒子に結合させることが可能である[109〜111]。さらに、ナノ粒子を、その表面に結合された抗原及び/又は非補体危険信号を用いて合成することができる。
いくつかの実施態様は、抗原を患者に導入して、患者の中で抗原に対する抗体を産生することに向けられる。例えば、このようにして、予防接種又は抗腫瘍治療を遂行することができる。或いは、当該手法を用いて、例えば動物において科学的試薬として使用される抗体を産生することができる。
免疫治療のための用途を含めて、抗原提示細胞(APC)、特に樹状細胞(DC)への生分解性ナノ粒子の送達について説明する。本明細書の詳細な実施例では、リンパ節濃度におけるDCへの直径20、25、45及び100nmのポリ(エチレングリコール)安定化ポリ(プロピレンスルフィド)(PPS)ナノ粒子の送達について説明する。詳細な実施例におけるナノ粒子は、ポリ(エチレングリコール)の親水性コロナに囲まれたPPSの架橋ゴム状コアを含む。PPS領域は、疎水性薬物を担持することが可能であり、酸化環境内で可溶性ポリマーに分解する。グリコペプチド(本明細書ではグリコシル化ポリペプチドと定義される)抗原を含むペプチド又はタンパク質抗原及び核酸コード化抗原をナノ粒子表面に結合させることができる。20nmの粒子は、間質注入後に最も容易にリンパに吸収されたが、20及び45nmの粒子は、リンパ節における有意な残留性を示し、注入後24、72、96及び120時間目に一貫した強い存在を示した。ナノ粒子は、外来性標的化リガンドを使用することなく、40〜50%までのリンパ節DC(及びAPC)によって内在化され、内在化の部位は、注入部位でなく、リンパ節に存在していた。24時間目に対して96時間目にナノ粒子含有DC(及び他のAPC)に増加が見られ、これらの細胞のリンパ節への浸透が証明された。ナノ粒子粒径及び表面化学作用の双方が、インビボ注入後のDC成熟に影響を与えることが判明した。
他の箇所に記載されているように、直径が20、45及び100nmのPPSナノ粒子を逆エマルジョン重合によって合成した。本明細書に用いられている「エマルジョン重合」という用語には、逆エマルジョン重合が含まれ、「エマルジョン」という用語には、逆エマルジョンが含まれる[39]。手短に述べると、PEGブロック共重合体乳化剤、PLURONIC F-127(Sigma-Aldrich(スイスBuchs))及びプロピレンスルフィドモノマーを一定撹拌しながら超純粋ミリQ水に添加することによって、エマルジョンを作製した。他の箇所に記載されているように保護された開始剤ペンタエリスリトールテトラチオエステルを合成し[39]、個別のフラスコ内で、0.20mLの0.5Mナトリウムメチレート溶液と撹拌下で10分間混合することによって脱保護した。脱保護に続いて、開始剤をモノマーエマルジョンに添加し、5分後に、60μlの塩基ジアゾ[5.4.0]ビシクロウンデカ-7-エン(DBU)を反応物に添加し、不活性雰囲気下で24時間連続的に撹拌させた。次いで、ジスルフィド架橋を行うために、ナノ粒子を空気に接触させた。
他に指定がなければ、生後6〜9週間で体重20〜30gのBALB/cマウスをこの研究に使用した。すべてのプロトコルは、スイスの法律に従うVeterinary Authorities of the Canton Vaudによって承認されたものである。10mg/kgの塩酸ケタミン及び1mg/kgのキシラジンの皮下注入によって麻酔を送達した。CO2窒息によってマウスを安楽死させた。
皮膚への間質注入の後にナノ粒子の相対的な吸収特性を測定するために、既に記載したように、尾の先端への一定圧力注入によって蛍光顕微リンパ管造影を実施した[43〜45]。マウスの尾を除毛し、37℃の体温を維持するための加熱パッドを備えた顕微鏡ステージ(Axiovert 200M、Zeiss)にマウスを配置した。蛍光PPSナノ粒子(直径20、45又は100nm)を無菌リン酸緩衝生理食塩水(PBS)に溶解させた20mg/ml溶液をカテーテルに導入した;カテーテルに取りつけられた30ゲージニードルを尾の先端から約1mmの箇所に挿入し、活栓を開いて、40mmHgの一定圧力で流動を開始させた。ナノ粒子溶液の流量(管における遠上流の泡によって監視した)は、およそ20mm3の組織に対して平均0.1μl/分(注入箇所から見える容量から概算した)或いは約5μl/g/分とした。目に見える膨張は、観察されなかった。尾に沿う連続的な画像を一定の接触時間に収集した。ナノ粒子の粒径毎に実験を3回繰り返した。

リンパ節滞留の評価を行うために、20μlの20mg/ml蛍光PPSナノ粒子(直径20、45及び100nm)を、30ゲージニードルを通じてマウスの尾の先端又は前足蹠にボーラスとして注入した。20μlのPBSを注入することによって対照を実施した。注入部位で炎症は観察されなかった。24、72、96又は120時間目に、マウスをCO2窒息によって安楽死させた。尾及び脚リンパの排液を行う仙骨及び腰部リンパ節、前足蹠部リンパの排液を行う鰓及び腋窩リンパ節を除去し、フラッシュ凍結し、10μmの切片に凍結裁断し、マウスに対する抗体CD3e(Pharmingen(カリフォルニア州San Diego))、CD45R(Caltag(カリフォルニア州Burlinhgame))、CD68(Serotec(ドイツDusseldorf))、Dec-205(Serotec)及びCD31(Pharmingen)で免疫染色し、T細胞、B細胞、マクロファージ/DC、DC及び内皮細胞をそれぞれ標識した。Alexa Flour 594nm(Molecular Probes)抗体を用いて二次的な検出を実施した。リンパ節切片を蛍光(Axiovert 200M、Zeiss)及び共焦点レーザ走査顕微鏡(LSM 510 Meta、Zeiss)で画像化した。
既に記載したプロトコルに従ってリンパ節細胞を単離した[46]。手短に述べると、先に記載した蛍光ナノ粒子又はPBSの注入に続いて、リンパ節を除去し、26ゲージニードルで裂き、37℃で25分間にわたってコラゲナーゼD(Roche(スイス、Basel))で消化させた。次いで、組織を70μm細胞染色器(BD(スイス、Basel))に通して、細胞懸濁物を回収した。リンパ節細胞懸濁物について、APCを抗MHCクラスII-(I-A)-R-PE(Chemicon(カリフォルニア州Temecula))で染色し、DCを抗CD11c-アロフィコシアニン(Pharmingen)で染色した。抗CD86-R-PE及び抗CD80-R-PE(Pharmingen)で染色することによってDC成熟を測定した。
リンパ節細胞の単離に続いて、細胞を約500000個/mlでRPMI(5%FBS)に接種した。次いで、細胞に20μlの20mg/mlの蛍光ナノ粒子を適用し、24時間インキュベートした。次いで、細胞をHBSSで2回洗浄し、先に述べたようにAPC及びDCに対して染色した。
染色に続いて、リンパ節細胞懸濁物を流動細胞計測(CyAn ADP、Dako(デンマーク、Glostrup))によって分析した。FloeJoソフトウェア(TreeStar(オレゴン州Ashland))を使用してさらなる分析を行った。蛍光ナノ粒子を内在化させたAPC及びDCは、FITCがナノ粒子の標識を表すMHCII+FITC+及びCD11c+FITC+であると判断された。CD86及びCD80を発現した細胞の割合を計算することによって、ナノ粒子内在化に続くDC成熟の評価を行った。
96ウェルプレート(Becton Dickinson)に、1:4000の希釈率、100μl/ウェルでPBSに含めたC3a/C3a(desArg)マウス抗ヒトモノクローナル抗原(AntibodyShop、Grusbakken、Denmark)を塗布した。プレートを室温(RT)で一晩放置した。未結合抗体を払い落とし(すなわち、突発的な機械的運動によって除去し)、プレートを200μl/ウェルのDI水で3回洗浄した。次いで、プレートを200μl/ウェルのブロッキング緩衝液(PBS+0.05%Tween 20+0.5%ウシ血清アルブミン)で室温にて1.5時間ブロックした。
25nmのPLURONIC安定化ナノ粒子(すなわちヒドロキシル化ナノ粒子;本明細書に記載されているように製造された)、メトキシ末端PLURONICで安定化された25nmのナノ粒子(すなわち、ヒドロキシル化されていないナノ粒子)、20nmのカルボキシル化ポリスチレンナノ球体(COOH-NS)(Invitrogen)、PBS及びLPS(30μg)を既に記載したようにマウスに皮内注入した。次いで、リンパ節を収穫し、細胞を単離し、既に記載したようにCD11c、CD86、CD80及びCD40に対して染色した。流動細胞計測を行って、リンパ節DCの成熟プロファイルを測定した。
25nmのPLURONIC安定化(すなわちヒドロキシル化)ナノ粒子及び100nmのPLURONIC安定化(すなわち、ヒドロキシル化)ナノ粒子、及びPBSを既に記載したようにマウスに皮内注入した。次いで、リンパ節を収穫し、細胞を単離し、本明細書に記載されているようにCD11c、CD86、CD80及びCD40に対して染色した。流動細胞計測を行って、リンパ節DCの成熟プロファイルを測定した。
タンパク質、又はグリコペプチドを含むペプチドでPLURONIC(PEGとPPGのブロック共重合体)表面を官能化することによって、PPSナノ粒子に対する抗原結合を遂行することができる。化学結合のための遊離システイン残基を含むタンパク質抗原についての結合スキームを本実施例で提示する。N末端又はリシン残基におけるアミンのような他の官能基を関連スキームで使用することができる。抗原をナノ粒子表面に吸着させることもできる。
本明細書に記載したように、二重標識OVA連結ナノ粒子をマウスに皮下注入した。次いで、リンパ節を注入から24及び48時間後に除去し、次いでそれらを凍結し、凍結裁断した。次いで、リンパ節切片を蛍光顕微鏡で観察した。
25nmのOVA連結PLURONIC安定化ナノ粒子及びリポ多糖(LPS)と混合されたOVAを、本明細書に記載したようにマウスに皮内注入した。次いで、本明細書に記載したように、リンパ節を収穫し、細胞を単離し、CD11c、CD86、CD80及びCD40に対して染色した。流動細胞計測を実施して、リンパ節DCの成熟プロファイルを測定した。
T細胞養子移入として知られる方法を用いて、T細胞増殖の測定を行った。OT-II Tg(Jackson Immunoresearch)マウスは、CD4 T細胞に上方制御レベルのOVA T受容体を有するという点において形質転換性である。OT-II Tgマウスの脾臓及びリンパ節を単離して、細胞懸濁物を作製した。脾臓細胞懸濁物については、赤血球を1.667%のNH4Clで溶解させた。脾臓及びリンパ節の細胞を蓄積し、計数した。合計400×106個の細胞を回収した。
2日目に、20μlの抗原(10ugのOVA+5ugのLPS又は10ugの25nmのOVA連結PLURONIC安定化ナノ粒子)を受容マウスの前足蹠に注入した。5日目に、マウスを殺し、各マウスから鰓及び腋窩リンパ節を除去し、蓄積して、細胞懸濁物を作製した。細胞をCD45.2 PE、ヨウ化プロピジウム(死細胞)及びCD4アロフィコシアニンに対して染色した。流動細胞計測を実施して、T細胞増殖の測定を行った。
OTIIマウスは、OVAに対するT細胞受容体を上方制御するCD4 T細胞を有するという点において形質転換性である。したがって、これらのT細胞は、OVA抗原に遭遇すると極めて敏感になる。したがって、CFSE標識OT-II T細胞へのWTマウスへの養子移入は、インビボのT細胞増殖を測定するための優れたモデルである。
本明細書に記載したように、25nmのOVA連結PLURONIC安定化ナノ粒子、PBSにおけるOVA及びLPSによるOVAをC57/BL6マウスに注入した。次いで、7日目にマウスにブースタ注入を行った。次いで、21日目に、本明細書に記載したように、マウスを殺し、リンパ節を除去し、細胞を単離した。血球計算器を使用して細胞懸濁物を計数した。
25nmのOVA連結PLURONIC安定化(すなわちヒドロキシル化)ナノ粒子、100nmのOVA連結PLURONIC安定化(すなわちヒドロキシル化)ナノ粒子、25nmのOVA連結メトキシ末端PLURONIC安定化(すなわちヒドロキシル化されていない)ナノ粒子、PBS中のOVA、及びLPS含有OVAを本明細書に記載したようにC57/BL6マウスに注入した。25nmのOVA連結PLURONIC安定化(すなわちヒドロキシル化)ナノ粒子、及びLPS含有OVAをC3-/-マウスに注入した。注入前及び注入後21日目にマウスから採取した血液から血清を単離し、使用するまで-20℃で保管した。ブースタ注入は行わなかった。
プレートのブロッキングが完了すると、プレートを150μl/ウェルの洗浄緩衝液で3回洗浄した。次いで、血清サンプルを室温にて2時間にわたって50μl/ウェルでウェルに添加した。注入前血清サンプルを3通り添加した。次いで、サンプルを払い落とし、次いでプレートを150μl/ウェルにて洗浄緩衝液で5回洗浄した。マウス抗IgG-HRPをブロッキング緩衝液で1:3000に希釈し、室温にて2時間にわたって50μl/ウェルで添加した。次いで、HRP抗体を払い落とし、プレートを150μl/ウェルで洗浄緩衝液により5回洗浄した。次に、HRP基質試薬(R&D systems)を室温にて45分間にわたって暗所において100μl/ウェルで添加した。50μl/ウェルの2NのH2SO4を添加することによって反応を停止させた。次いで、450nm及び540nm波長におけるプレート吸収をTecanプレートリーダで測定した。540nmのバックグラウンド値を450nmから引いて、最終値を得た。注入後血清値がカットオフ値より大きい場合に陽性のサンプルを測定した。注入前の3つの値の平均値+標準偏差×3からカットオフ値を計算した。正の値を有する最大希釈率を抗体(Ab)力価値と見なす。
疎水性薬物、例えばデキサメタソンの充填を適応溶媒蒸発法[132、133]によって達成した。手短に述べると、例示を目的として、薬物を溶媒ジクロロメタン(1mg/ml)に添加した。次いで、1mlの薬物-溶媒懸濁物を1mlのPPSナノ粒子溶液に20mg/mlで添加した。溶媒を蒸発させるために、エマルジョンを室温の暗所で連続的に撹拌した。薬物充填効率をGPCで測定した。
両側スチューデントt検定を行うことによって実施例における統計的有効性を測定した。結果は、平均値±SDを示し、条件毎に3〜8の実験を行った。

Claims (29)
- 補体を活性化する合成ポリマーを含む合成粒子の孤立集合体を含むナノ粒子組成物であって、該集合体は、約10nmから約100nmの平均粒径を有し、該粒子は、抗原とさらに結合されている、前記ナノ粒子組成物。
- 前記抗原が、前記粒子と共有結合している、請求項1記載の組成物。
- 前記合成ポリマーがヒドロキシル化されている、請求項1記載の組成物。
- 前記合成ポリマーが、補体を活性化するアミノ酸の配列又は糖類の配列を含まない、請求項1記載の組成物。
- 前記合成ポリマーが、前記ナノ粒子のコアを形成する第2の生分解性ポリマーの疎水性部分に吸着する疎水性部分を含み、それによって前記合成ポリマーが該コアに結合する、請求項1記載の組成物。
- 前記合成ポリマーがポリアルキレンオキシドを含む、請求項5記載の組成物。
- 前記合成ポリマーがポリエチレングリコールを含む、請求項5記載の組成物。
- 前記合成ポリマーがPLURONICである、請求項5記載の組成物。
- 前記合成ポリマーがPLURONIC-F127である、請求項5記載の組成物。
- 前記第2のポリマーがポリプロピレンスルフィドである、請求項5記載の組成物。
- 前記粒子が、細胞に特異的に結合する標的化リガンドを含まない、請求項1記載の組成物。
- 前記抗原が、腫瘍免疫治療のための抗原又は感染性疾患の抗原である、請求項1記載の組成物。
- 前記粒子が、炎症性サイトカイン及びToll様受容体に対するリガンドからなる群から選択される危険信号をさらに含む、請求項1記載の組成物。
- 前記会合が混合又は吸着を含む、請求項1記載の組成物。
- 前記抗原が、ポリペプチド又はグリコペプチド抗原である、請求項1記載の組成物。
- 前記集合体が、約10nmから約50nmの平均粒径を有する、請求項1記載の組成物。
- 前記抗原が前記粒子と共有結合し、前記合成ポリマーがヒドロキシル化され、前記合成ポリマーが、補体を活性化するアミノ酸の配列又は糖類の配列を含まず、前記合成ポリマーが、前記ナノ粒子のコアを形成する第2の生分解性ポリマーと共有結合し、前記第2のポリマーが、ポリプロピレンスルフィドであり、前記合成ポリマーが、ポリアルキレンオキシド、ポリエチレングリコール、PLURONIC及びPLURONIC-F127からなる群の要素を含む、請求項16記載の組成物。
- 前記粒子が、細胞に特異的に結合する標的化リガンドを含まず、前記抗原が、腫瘍免疫治療又は感染性疾患に対する抗原であり、前記組成物が、炎症性サイトカイン及びToll様受容体に対するリガンドからなる群から選択される危険信号をさらに含む、請求項16記載の組成物。
- 前記合成ポリマーが、少なくとも1つの疎水性ブロックと少なくとも1つの親水性ブロックとの両親媒性ブロック共重合体を含み、該ブロック共重合体が水溶液中で自己集合して前記粒子を形成し、該親水性ブロック上の官能基が補体を活性化する、請求項16記載の組成物。
- 前記官能基がヒドロキシルである、請求項19記載の組成物。
- 前記親水性ブロックがヒドロキシル末端ポリエチレングリコールである、請求項19記載の組成物。
- 第1のポリマーと、重合時に使用されるエマルジョンである第2のポリマーとをエマルジョン重合して、20から50nmの平均直径を有する粒子の集合体を製造すること、補体を活性化するヒドロキシル官能基を含むように該第2のポリマーを選択すること、及び免疫治療薬を該粒子と会合させることを含む、ナノ粒子の免疫治療組成物の製造方法。
- 前記第1のポリマーが生分解性である、請求項22記載の方法。
- 患者の免疫系を刺激する免疫治療薬を送達する方法であって、該方法は、合成粒子の集合体を該患者に導入することを含み、該粒子は、補体を活性化する第1のポリマーを含み、該集合体は、約10nmから約50nmの平均直径を有し、該第1のポリマーは、補体を活性化するアミノ酸の配列又は糖類の配列を含まず、該粒子は、第1のポリマーに結合する第2のポリマーを含む、前記方法。
- 前記第2のポリマーが生分解性である、請求項24記載の方法。
- 前記粒子がリンパ節の抗原提示細胞に特異的に標的化される、請求項24記載の方法。
- 合成粒子の孤立集合体を含むナノ粒子組成物であって、該集合体は、約10nmから約100nmの平均直径を有し、該粒子は、免疫抑制薬を含み、該粒子は、抗原とさらに会合されている、前記ナノ粒子組成物。
- 前記粒子が、少なくとも1つの疎水性ブロックと少なくとも1つの親水性ブロックとの両親媒性ブロック共重合体を含み、該ブロック共重合体が、水溶液中で自己集合して、前記粒子を形成する、請求項27記載の組成物。
- 前記集合体が、約10nmから約50nmの平均粒径を有する、請求項27記載の組成物。
