Abstract

Description
[発明の分野]
本発明は、一般にアルツハイマー病または関連する神経学的な病状の予防または治療に関する;特に、アルツハイマー型の疾患を治療するためのβアドレナリン作動性またはオピオイドレセプターアンタゴニストの使用に関する。
進行性の痴呆および人格障害で特徴付けられるアルツハイマー病 (AD)は、老化に関連する最も一般的な神経変性疾患である。ADは、65齢をこえる集団の5-11%に及び85齢をこえる集団の30%に影響する。ADは、変性しているニューロンの及び反応性アストロサイトの近傍のアミロイドプラークの異常な蓄積によって生じる。 D. J. Selkoe, Annu. Rev. Neurosci. 17, 489 (1994).
アミロイドプラーク〔殆どアミロイド β (Aβ)から構成される〕は、AD 神経病理学の特徴である。そして、アミロイドプラークの形成は、ADの主な原因であると考えられる。加えて、最近の研究は、拡散性のオリゴマーのAβも神経毒性で潜在的にADに関連しえることを明らかとした(Walsh, D.M. et al., "Naturally Secreted Oligomers of Amyloid Beta Protein Potently Inhibit Hippocampal Long-Term Potentiation in vivo," Nature 416, 535-9 (2002))。
本発明の課題には、アルツハイマー病または関連する神経学的な病状を治療する又は予防するための試薬をスクリーニングするための方法の提供が含まれ、これによってアルツハイマー病または関連する神経学的な病状を治療する又は予防するための試薬が提供される。

Aβ産生におけるβ2AR活性化の効果は、最初に機能的な GPCR シグナル伝達経路を所有し、正常なAβ分泌を示すHEK293細胞で評価された。本実験に使用されたHEK293細胞は、β2ARおよび変異体 APP(APPswe)(コドン670 および 671にFAD関連「スウェーデン」変異を保持する)でトランスフェクトされた。図 5aに示したとおり、β2ARのアゴニスト〔イソプラノロール(Iso;isopranolol)〕での刺激によって、二つの分泌されたAβ 種 (Aβ40 および Aβ42)のレベルが増加した。他方で、それ自身では効果を有さないβ2ARアンタゴニスト〔プロプラノロール(Pro)〕の添加によって、分泌されたAβレベルを増加させるIsoの能力が無効にされる。特異的なγ-セクレターゼインヒビターL685,458での前処理によって分泌されたAβの産生の増加が無効になるとの事実によって証明されているとおり、Aβ 産生の増加はγ-セクレターゼを必要とする。
上記のβ-ARまたはDORの活性化に際したAβの産生の増加は、γ-セクレターゼ発現の増加またはγ-セクレターゼ活性の増強から生じえる。この質問に答えるために、γ-セクレターゼの発現 および 活性化におけるβ2AR 活性化の効果を評価した。図 6aにウエスタンブロット分析によって示されたとおり、C60(C99のγ-セクレターゼ媒介性の切断から産生される)産生は、C99トランスフェクションしたHEK293細胞のIso処理後に増加した。しかしながら、同じ処理は、γ-セクレターゼの活性部位コンポーネントであり、アミノ- およびカルボキシル-末端の断片(PS1-NTFおよびPS1-CTF)のヘテロ二量体として存在する、PS1の発現レベルの変化を生じることに失敗した。この結果は、β2AR活性化によって、γ-セクレターゼ発現が変化することなくγ-セクレターゼ活性が増加したことを示唆している。
上記で議論したとおり、一度活性化したら、GPCR (β2ARを含む)は、Gs タンパク質依存性アデニリルシクラーゼの活性化を誘発し、細胞内cAMPレベルの上昇へと導くだろう。β2AR 活性化によるγ-セクレターゼ活性増強の原因となる分子機構を明らかにするために、Gs タンパク質活性化の能力を有していない新規のβ2AR変異体(β2AR T68F, Y132G, Y219A またはβ2AR TYY)を更なる研究に使用した。β2ARのこれらの変異体がγ-セクレターゼ活性の増強の無効化に失敗したことが見出された(図 8a)。この結果は、γ-セクレターゼにおけるβ2AR効果におけるGs タンパク質シグナル伝達の関与を除外している。さらにまた、例えば、コレラ毒素(CTX), フォルスコリン(Fsk), およびジブチル-cAMP(db-cAMP)などの試薬(Gタンパク質活性化およびcAMPレベル上昇を模倣する)で処理された細胞は、γ-セクレターゼ活性の増加を生じなかった(図 8b)。従って、β-アドレナリンレセプター活性化から生じるγ-セクレターゼ活性の増強にcAMPシグナル伝達は関与しない。
γ-セクレターゼ活性の増強にG-タンパク質シグナル伝達もcAMP経路も関与しないならば、どのような機構が関与しているのか?上記で図 3を参照して議論したとおり、GPCR(βAR および オピオイド レセプターを含む)活性化は頻繁にレセプターエンドサイトーシスを伴う(これも特異的なシグナル伝達を惹起できる)。レセプターエンドサイトーシスおよび関連するシグナル伝達がγ-セクレターゼ活性の増強に関与しているかどうかは、エンドサイトーシス経路の多様なインヒビターを用いることによって証明できる。
図 3に示したとおり、一旦細胞の内側にはいれば、エンドサイトーシスのベシクルは特異的なエンドサイトーシス経路を経由して彼等の目的地へと輸送される。エンドサイトーシス経路には、Rabグアノシントリホスファターゼ(Rab GTP加水分解酵素)によって制御される細胞内コンパートメントの輸送が関与する。原形質膜から早期エンドソーム、そしてLELへのエンドサイトーシス輸送が、それぞれ早期のエンドソームマーカーRab5またはLELマーカーRab7のドミナントネガティブ変異体であるRab5 S34NまたはRab7 T22Nによってブロックできることが知られている。図 11aおよび図 11bに示したとおり、HEK293細胞におけるRab5 S34NまたはRab7 T22Nの発現によって、γ-セクレターゼ活性 (図 11a) および Aβ産生 (図 11b)のβ2AR-刺激性の増強が無効化された。というのも、Rab7 T22Nトランスフェクト細胞は、早期エンドソームに輸送されるエンドサイトーシスベシクルをなおも有しえるからである。これらの結果は、次の事項を示唆している。その事項とは、増強したγ-セクレターゼ活性 および Aβ産生がLELへ輸送されるエンドサイトーシスベシクルを必要とし、LELがγ-セクレターゼ活性 および Aβ産生におけるβ2ARの効果に決定的に関与していること指摘していることである。
Dyn K44AまたはRab5 S34Nの共発現によってβ2AR刺激に続くLELにおけるPS1の局在化の上昇が効率的に阻止される(図 13a)との所見は、PS1が原形質膜からLELへと輸送されるだろうことを示唆している。クラスリン被覆ピットおよびベシクルを特異的にマークするβ-アダプチン(β-adaptin)を用いることで、HEK293細胞においてDORの3-min刺激後にPS1とβ-アダプチンおよびエンドサイトーシスしたレセプターとの共局在化が見出された(図 13b)。これらの所見は、β2ARまたはDORのアゴニスト刺激後のPS1および活性化レセプターの共エンドサイトーシスを暗示する。この事項は、驚くべきことではない。というのも、PS1は、構成的に膜タンパク質(例えば、APPおよびNotch)と相互作用し、β2ARは他の膜貫通タンパク質のエンドサイトーシスを後者とヘテロダイマーを形成することによって仲介できるからである。このことが行われていることを示すために、PS1およびβ2ARまたはDORの間の会合を共免疫沈降アッセイで検討した。
PS1 および レセプターの間の会合を崩壊させる又は弱めることができる試薬は、FRET(蛍光共鳴エネルギー移動)などの任意の適切な方法でスクリーニングしえる。FRETは、エネルギーが励起したドナーフルオロフォアからアクセプターフルオロフォアへと短距離(< 10 nm)の双極子間相互作用を介して移動するプロセスである。従って、FRETを使用して二つの結合タンパク質の間の物理的な相互作用を検出できる。FRETでは、エネルギーの非放射性の伝達によって、前記ドナーからの光の放射を減弱する。従って、FRETは、例えば、アクセプター上の蛍光部分を光退色などの適切な方法で破壊する前後で、同じサンプルにおける前記ドナーからの光放射の強度を比較することにより検出しえる。FRETが現在する場合、ドナーの発光はアクセプターの光退色後により強い。
これらのAD-関連分子におけるβ2ARの効果を、さらに動物において調査した。インビボ実験は、次の事項を示した。その事項とは、ラット海馬におけるγ-セクレターゼ活性(図 14a)およびAβレベル(図 14b)の両方が、アドレナリン作動性レセプター(ノルエピネフリン, NE)に対する内在性リガンドまたは公知のβ2AR-選択的アゴニスト クレンブテロール(Cle)の急性注射によって有意に増強したことである。これらの結果に基づいて、これらのレセプターのアゴニストへの慢性的な暴露によって動物モデルにおけるAD-関連の病態が悪化することが予測できる。ADのマウスモデル(APPswe/PS1ΔE9 二重トランスジェニックマウス)での実験によって、このアイデアが支持される。というのも、これらのマウスは、30日間のIsoまたはCleの長期投与後に大脳のアミロイドプラークの増加を呈したからである(図14c - 14e)。この観察によって、β2ARの活性化がγ-セクレターゼ活性, Aβ産生, およびアミロイドプラーク形成を増強できることが指摘された。従って、これらのレセプターのアンタゴニストは、Aβまたはアミロイドプラークの形成の減少に有用である。図14fおよび14gは、このことが実際に事実であることを示しており、ICI 118,551(β2AR特異的なアンタゴニスト)は有意にアミロイドプラークの量を減少させた。
β-アドレナリンレセプターアンタゴニストのインビボ有効性を評価するために、係る化合物をアルツハイマー病のトランスジェニックマウスモデル(APPswe/PS1ΔE9)に投与した。約 6月齢で、これらのマウスは、大脳のAβレベルの上昇および大脳のアミロイドプラーク数の増加を伴う進行性の空間的な記憶障害を呈した。
一つの実験において、プロプラノロールおよびナルドロール(naldolol)をAPPswe/PS1ΔE9 トランスジェニックおよび非-トランスジェニック(NTg)マウス投与して、アミロイドプラーク形成におけるβ-アドレナリンレセプターアンタゴニストの効果を評価した。APPswe/PS1ΔE9 マウス および 非-トランスジェニック (NTg)同腹仔を、性および齢が適合したコホートに割当てた。化合物を、最初の4月齢および引き続き6月齢まで投与した。プロプラノロールは、容易に血液脳関門(BBB)を通過できるので、中枢神経系のβ-アドレナリンレセプターと拮抗できる。他方、ナドロール(同様にβ-アドレナリン作動性のレセプターアンタゴニストである)は、BBBを通過できない。マウスを、モーリス水迷路を用いて空間学習および記憶で試験した。
更なる動物実験において、四月齢のAPPswe/PS1ΔE9二重トランスジェニックマウスは、一日に生理食塩水 (Sal)、2 mg/kgのクレンブテロール(Cle, β2AR選択的なアゴニスト)または1 mg/kgのICI 118,551(ICI, β2AR選択的なアンタゴニスト)を30日間経口的に投与された。マウスを、次に実質的に上記のように従来のモーリス水迷路のバージョンを用いて空間的な学習および記憶で試験した。
以下は、上記で議論した実験および例で使用した幾つかの特定の処置を記載する。明確性のために、当業者に周知である使用された一般的な生化学的および分子生物学技術は、記載されない。
全ての試薬は、他に断らない限りSigmaまたは他の市販の供給源から得た。ヒト FL-APPを、pcDNA3 ベクターにクローン化し、PCRでAPPsweに変異させた。APPのシグナルペプチドを有しているC99を、pcDNA3 ベクターにサブクローン化した。DNA配列の信憑性を、シークエンシングにより確認した。ヒトのクラスリン重鎖に関するRNAiプラスミドを、設計して5'-GCTGGGAAAACTCTTCAGATT-3'の配列を標的とした。NS RNAiは、5'-GGCCGCAAAGACCTTGTCCTTA-3'であった。
全ての動物実験は、実験動物の取扱い及び使用に関する国立衛生研究所ガイドラインに従って行った。急性実験において、Sprague-Dawlyラット(Shanghai SLAC Laboratory Animal Companyからえた)は、2 μgのノルエピネフリンを脳室内に注射された。標準脳座標は:前側-後側, ~ 0.9 mm; 左-右, ~ 1.5 mm; 背側-腹側, ~ 3.8 mmであった。ラットの急性腹腔内注射を、0.5 mg/kgのクレンブテロールで行った。30-dの長期実験において、5月齢までのAPPswe/PS1ΔE9 マウス (Jackson Laboratory)は、生理食塩水(n = 4, 二個体のメスおよび二個体のオス) および 3 nMのイソプロテレノール (n = 6, 四個体のメス および 二個体のオス)の日々の注射のためにカニューレを備えつけられた(前側-後側, ~ 0.6 mm; 左-右, ~ 1.2 mm; 背側-腹側, ~ 1.8 mm)。APPswe/PS1ΔE9 マウスは、一日に生理食塩水 (n = 6, 三個体のメスおよび三個体のオス), 2 mg/kg/d クレンブテロール(n = 7, 四個体のメスおよび三個体のオス), および1 mg/kg/d ICI 118,551(n = 6, 三個体のメスおよび三個体のオス)を経口投与された。
マウスを、麻酔し、心臓への生理食塩水灌流によって屠殺した。脳を単離し、中部矢状縫合で二分し、4% パラホルムアルデヒドをリン酸緩衝液(pH 7.6)中に有する溶液に5 h、4°Cで配置した。次に、固定後の半球を、10 μmの切片に環状に凍結切片を作製した。切片を、抗-Aβ 6E10 抗体 (Chemicon)でインキュベーションし、TRITC-結合型の 抗-マウス抗体でインキュベーションした。切片を、次にレーザー共焦点蛍光顕微鏡(Leica TCS SP2)を用いて視覚化し、イメージを作製した。アミロイドプラークの領域を、Image-Pro Plus 5.1ソフトウェア(Media Cybernetic)で定量した。イメージを、閾値化によってグレイスケールに転換し、プラーク領域を推定した。
初代の海馬培養を、Amaxa Nucleofectorシステムを用いてエレクトロポレーションした出生後1日のSDラットから調製し、B27/neurobasal培地(インビトロゲン)で二週間維持し、それからアゴニスト処理に使用した。出生後8週のSDラットからの急性海馬スライスを、氷冷の人工的なCSF中でビブラトームを用いて調製した。
細胞を、Iso (10 μM)またはDADLE (1 μM)に1 h暴露し、さらに馴化培地中で別に6 hインキュベーションした。その馴化培地を、サンドイッチELISAキット(Biosource)でAβ40 および Aβ42に関して検出した。ラット海馬を、ホモジナイズし、100,000 X g で1 h遠心分離した。上清を、以前の報告52にしたがって、BNT77/BA27 および BNT77/BC05サンドイッチ ELISAキット(Wako)でラット Aβ40 および Aβ42に関して検出した。全ての測定は、二重に行われた。
HEK293細胞またはラット海馬スライスを、RIPA緩衝剤で溶解した。Flagタグをつけたレセプターおよび内在性DORを、Flag 抗体結合ビーズまたはDOR 抗体(Santa Cruz)で免疫沈降した。免疫沈降した複合体を、SDS-PAGEで分離し、Flag, PS1-NTF, PS1-CTF, ニカストリン, APH-1a およびPEN-2抗体(Calbiochem)でブロットした。HEK293
発現基質アッセイ
実験を、以前27に記載と同様に行った。HEK293細胞を溶解し、アリクウォット(50 μg タンパク質を含有している)を13, 000 X g で15 min遠心分離した。細胞膜分画を、再懸濁し、50 μlの1, 10-フェナントロリン, アプロチニン, およびロイペプチンを含有している緩衝液(pH 6.5)中で37 °Cで2 hインキュベーションした。インキュベーションした膜分画から生じたC60を、HA抗体12CA5を用いるウエスタンブロットで測定した。
アッセイを、報告28,29のとおり行った。細胞または海馬組織を、溶解し、ホモジナイズした。アリクウォット(50 μgの細胞溶解産物または海馬のホモジネートを含有している)を13, 000 X g で15 min遠心分離した。膜のペレットを、再懸濁し、12 μlの蛍光原基質(Calbiochem)を含有している50 μlの緩衝液(pH 6.5)中で37 °Cで2 hインキュベーションした。インキュベーション後、蛍光を励起波長355 nm および発光波長440 nmで分光計を用いて測定した。
HA-C99 および DORでコトランスフェクションしたHEK293細胞を、メチオニンおよび血清がない培地(Invitrogen)中で2 h飢餓させ、引き続いてDADLEの非存在下または存在下で、1 hで500μCi[35S]メチオニン(Amersham Pharmacia)でパルス標識した。細胞を、次に過剰量の非標識メチオニンを含有している培地中で3 h追跡した。細胞溶解産物中のC99を、12CA5で免疫沈降し、オートラジオグラフィーで分析した。
実験は、ベシクル単離プロトコール53を修飾したものである。β2AR, C99 およびFlag-Rab7でコトランスフェクションしたHEK293細胞からのホモジネートを、500 x g で10 min遠心分離した。生じた上清を、4 °CでM2抗体結合ビーズで8 hインキュベーションした。単離したLELを、次にLAMP-1およびEEA1に対する抗体(BD Biosciences)でのウエスタンブロットに供試した。
HA-タグをつけたレセプターとGFP-Rab7またはGFP-Rab7 T22Nの有り無しの条件でトランスフェクションしたHEK293細胞における実験に関して、細胞は抗体 12CA5 を30 min与えられ、アゴニストで処理され、固定された。Flag-Rab7およびHA-Dyn K44AまたはGFP-Rab5 S34Nでトランスフェクションした細胞における実験に関して、細胞をアゴニストで処理し、次に固定した。急性海馬スライスにおける実験に関して、IsoまたはDADLEに暴露されたスライスを固定し、凍結切片を作製した。引き続く免疫染色には、一次抗体(PS1-NTF, Flag, LAMP-1, またはβ-アダプチン抗体またはFITC結合HA抗体を含む)および二次抗体(Cy3結合抗ウサギおよび FITC結合抗マウス抗体, Jackson ImmunoResearch)が関与した。イメージを、レーザー共焦点蛍光顕微鏡(Leica TCS SP2)を用いて獲得した。
FRET分析に関して光退色有り無しのイメージを、Leica TCS SP2共焦点顕微鏡で得て、ソフトウェアで分析した。要約すると、HEK293細胞を、GFP-DOR および HA-ps1でコトランスフェクションした。HA-PS1発現は、HAに対する一次抗体およびCy3(Jackson ImmunoResearch)を結合させた二次抗体でインキュベーションした後のCy3蛍光をモニタリングすることによって検出された。他方、GFP-DOR 発現はGFP蛍光検出で同定された。GFP-DORまたはPS1-Cy3を発現している細胞からの発光スペクトルを、488 nm レーザーを用いてλモードで取得した。この方法によるFRET効率の測定に関して、アクセプター光退色に関するLeicaソフトウェアアプリケーションを適用した。選択した細胞表面領域を、Cy3フルオロフォアを退色させる561 nm レーザーで光退色させた。GFP-DORおよびHA-ps1でコトランスフェクションしたHEK293細胞における光退色後のCy3シグナルの減少は、平均して84 ± 5.3% (n = 50)であった。FRETは、ps1-Cy3(アクセプター)の光退色後のGFP-DOR(ドナー)シグナルにおける増加として解釈された。相対的なFRET効率を、(1-[Cy3 I退色前/Cy3 I退色後]) X 100%として計算された。コントロールの目的で、光退色のない細胞表面の領域も、FRETで分析した。
細胞実験からのデータを、共分散の蓄積推定値での独立平均の比較に関するスチューデントのt検定で分析した。マウスのデータの統計学的な有意性を、ANOVAとスチューデントのt検定で決定した。
Claims (15)
- アルツハイマー病または関連する神経学的な病状を治療する又は予防するための試薬を調製するための方法であって、以下の工程を含むことを特徴とする方法:
(a) レセプターを活性化すること及び前記レセプターのエンドサイトーシスの第一の程度を決定すること(ここで、前記レセプターは、プレセニリン-1と会合するGタンパク質共役レセプターである);
(b) 前記レセプターを工程(a)と同じ条件下で候補試薬の存在下で活性化すること、及び前記レセプターのエンドサイトーシスの第二の程度を決定すること;
(c) エンドサイトーシスの第一の程度とエンドサイトーシスの第二の程度との間の差を決定すること;
(d) 前記差が閾値未満である場合、工程(a) - (c)を繰り返すこと;および
(e) アルツハイマー病または関連する神経学的な病状を治療する又は予防するための医薬として候補試薬を調製すること及び/又は精製すること。 - アルツハイマー病または関連する神経学的な病状を治療する又は予防するための試薬をスクリーニングするための方法であって、以下の工程を含むことを特徴とする方法:
(a) レセプターを活性化すること及び前記レセプターのエンドサイトーシスの第一の程度を決定すること(ここで、前記レセプターは、プレセニリン-1と会合するGタンパク質共役レセプターである);
(b) 前記レセプターを工程(a)と同じ条件下で候補試薬の存在下で活性化すること、及び前記レセプターのエンドサイトーシスの第二の程度を決定すること;
(c) エンドサイトーシスの第一の程度とエンドサイトーシスの第二の程度との間の差を決定すること;および
(d) 前記差が閾値未満である場合、工程(a) - (c)を繰り返すこと。 - アルツハイマー病または関連する神経学的な病状を治療する又は予防するための試薬を調製するための方法であって、以下の工程を含むことを特徴とする方法:
(a) レセプターおよびプレセニリン-1またはγ-セクレターゼの間の会合の第一の程度を測定すること(ここで、前記レセプターは、プレセニリン-1と会合するGタンパク質共役レセプターである);
(b) 前記レセプターおよびプレセニリン-1またはγ-セクレターゼの間の会合の第二の程度を、工程(a)と同じ条件下で候補試薬の存在下で測定すること;
(c) 会合の第一の程度と会合の第二の程度との間の差を決定すること;
(d) 前記差が閾値未満である場合、工程(a) - (c)を繰り返すこと;および
(e) アルツハイマー病または関連する神経学的な病状を治療する又は予防するための医薬として候補試薬を調製すること及び/又は精製すること。 - アルツハイマー病または関連する神経学的な病状を治療する又は予防するための試薬をスクリーニングするための方法であって、以下の工程を含むことを特徴とする方法:
(a) レセプターおよびプレセニリン-1またはγ-セクレターゼの間の会合の第一の程度を測定すること(ここで、前記レセプターは、プレセニリン-1と会合するGタンパク質共役レセプターである);
(b) 前記レセプターおよびプレセニリン-1またはγ-セクレターゼの間の会合の第二の程度を、工程(a)と同じ条件下で候補試薬の存在下で測定すること;
(c) 会合の第一の程度と会合の第二の程度との間の差を決定すること;および
(d) 前記差が閾値未満である場合、工程(a) - (c)を繰り返すこと。 - 請求項1〜4の何れか1項に記載の方法であって、前記レセプターがβ-アドレナリンレセプターおよびδ-オピオイドレセプターからなる群から選択される少なくとも一つである方法。
- 請求項5に記載の方法であって、前記β-アドレナリンレセプターが2型β-アドレナリンレセプター(β2AR)である方法。
- 請求項1〜6の何れか1項に記載の方法であって、前記レセプターが、前記レセプターをコード化している遺伝子でトランスフェクションされている細胞で発現される方法。
- 請求項1, 2, 5, 6, および7の何れか1項に記載の方法であって、前記エンドサイトーシスの第一の程度を決定すること及び前記エンドサイトーシスの第二の程度を決定することが、エンドサイトーシスしたベシクル, エンドサイトーシスしたベシクル中のプレセニリン-1, 後期エンドソームまたはリソソーム(LEL)中のγ-セクレターゼ活性, またはアミロイド-β(Aβ)形成の量をモニターすることによって行われる方法。
- 請求項3〜7の何れか1項に記載の方法であって、会合の第一の程度を決定すること及び会合の第二の程度を決定することが、蛍光共鳴エネルギー移動をモニターすることによって行われる方法。
- アルツハイマー病または関連する神経学的な病状を治療する又は予防するための医薬の製造におけるレセプターアンタゴニストの使用であって、前記レセプターアンタゴニストはエンドサイトーシスの間にプレセニリン-1と会合するGタンパク質共役レセプターのエンドサイトーシスを阻害する使用。
- アルツハイマー病または関連する神経学的な病状を治療する又は予防するための医薬の製造における試薬の使用であって、前記試薬はGタンパク質共役レセプターおよびプレセニリン-1またはγ-セクレターゼの会合に干渉する使用。
- 請求項10または11に記載の使用であって、前記Gタンパク質共役レセプターがβ-アドレナリンレセプターおよびδ-オピオイドレセプターからなる群から選択される少なくとも一つである使用。
- 請求項12に記載の使用であって、前記β-アドレナリンレセプターが2型β-アドレナリンレセプター(β2AR)である使用。
- 請求項10, 12 および13の何れか1項に記載の使用であって、前記アンタゴニストがICI 118,551, プロプラノロール, ブトキサミン, およびナルトリンドールから選択される少なくとも一つである使用。
- 請求項10, 12 および13の何れか1項に記載の使用であって、前記アンタゴニストがICI 118,551またはブトキサミンである使用。
