Description
a)前記組み換えコラーゲン類似タンパク質をコードする第一の発現ベクター;および、
b)プロリル−4−ヒドロキシラーゼ(P4H)をコードする核酸を含む、第二の発現ベクター。
a)ここで定義した第一の発現ベクター;そして
b)上記で定義した第二の発現ベクター。
a)上記で定義された酵母細胞を提供する工程;
b)前記酵母細胞を発現ベクターまたは上記で説明した共発現システムで形質転換する工程;
c)前記宿主細胞から適切な条件下で組み換えタンパク質を発現する工程;および
d)前記タンパク質を回収する工程。

一般に、コラーゲンの合成は複雑な生化学的過程を反映する。その過程は、例えば、小胞体内で、プロリル−4−ヒドロキシラーゼ(P4H)により、それぞれのコラーゲンの特定のプロリンを4−ヒドロキシプロリンにする翻訳後修飾を必要とする。脊椎動物ではα2β2四量体であるP4Hは、コラーゲンの合成において中心的な役割を果たす。P4Hによって生じる4−ヒドロキシプロリン残基は、新しく合成されたコラーゲンポリペプチド鎖が三重らせんのコラーゲン分子となるための折りたたみに必須である[13]。
P4Hのコンストラクトのためには、P4Hの2つのサブユニットであるα−PHとPDIのための遺伝子に隣接する酵母ベクターの内部にシグナル配列をクローニングする必要がある。両遺伝子は、ガラクトースの存在下で誘導される両方向性のプロモーター(Gal1/10)の制御のもとに置かれている(図6参照)。シグナル配列は、P4Hサブユニットの小胞体への移動のために必要である。小胞体でP4Hは集合して元来の四量体になることができる。最高効率での局在化は、ヒトのシグナル配列がmating factor α−MFの酵母自身のシグナル配列に置換されて達成される[12]。これに関連して図6を参照のこと。
イガイ類足糸の2つの主要なタンパク質因子であるColD前駆体とColP前駆体の組み換えでの合成は本発明の一例である。E. Coliクローニングベクター中のColP前駆体とColD前駆体のcDNAは、Waite教授(アメリカ合衆国、UCSB)から入手した。cDNAはPCRで増幅し、異なる酵母発現ベクターにクローニングする。ベクターは、アクティベーター(恒常的プロモーターのGPDか誘導性プロモーターのGAL4のどちらか)の選択だけでなく、細胞ごとのコピー数が異なる。また、元々のシグナル配列も、最高効率での局在化のために、酵母のαMFのシグナル配列に置換する。
イガイ類コラーゲンの組み換えによる効率の良い合成を試験するには、イガイ類コラーゲンに対するポリクローナル抗体が利用できることが必要である。ヒトコラーゲンタイプI〜IIIに対するポリクローナル抗体を使った予備試験によると、イガイ類足糸の化学的に変性したコラーゲンに対してはごく弱い交差活性しか持っていないことが示された。この交差活性は、組み換えによる合成の間に存在するレベルのコラーゲンを検出するのに十分ではない。したがって、イガイ類コラーゲンに対する抗体を作製する必要がある。抗体を作製することができるようになるために、精製した天然のイガイ類コラーゲンが必要である。新鮮なイガイから足糸を抽出して、いくつかのクロマトグラフィー法(とりわけ逆相クロマトグラフィー)を用いて精製する。
ColP前駆体/ColD前駆体の個々のタンパク質をキャラクタライズし、自己重合して繊維を形成する効率を評価するために多様な物理的方法を用いることができる。前記方法には、遠紫外線および近紫外線による円偏光二色性分光分析(CD)、静的・動的光散乱、フーリエ変換赤外分光法(FTIR)、電子顕微鏡(EM)、原子間力顕微鏡(AFM)、ならびに流動場分画法(FFF)が含まれる。
CDやFTIRは、ColP前駆体とColD前駆体の二次構造、三次構造を決定するために用いられる。前記タンパク質の化学的、温度的な安定性もまた多様な条件下で試験することができる。コラーゲン形成に関与するタンパク質の形に関するデータは、光散乱、AFM、TEMによって提供される。
イガイ類足糸コラーゲンの二次、三次構造は、CDやFTIRで解析される。AFMやEMは集合した凝集体や繊維の四次構造や形態に関して情報をもたらす。マトリックスの無い一相クロマトグラフィーであるFFFは、コラーゲン集合の間に形成される異なる種類の種を評価するのに用いられる。FFFは、マトリックスの無いクロマトグラフィー技術なので、溶解した異なる巨大分子、特に他の古典的なクロマトグラフィー技術では分離できない繊維を分離することができる。
(DOPAとチロシンのクロスリンクにおけるGGHの役割)
GGHのアミノ酸配列は、ColP前駆体とColD前駆体の両方のカルボキシ末端で観察されている。トリペプチドである、NH2−Gly−Gly−His−COOH(GGH)は、酢酸ニッケル[Ni(OAc)2]と酸化モノペルオキシフタル酸マグネシウム[MMPP]の存在下で、溶液中の結合したタンパク質のクロスリンクを媒介する[18,19]。さらに、ペプチドは、ニッケル中心にとって好ましい調和的環境を提供し、推定上のNi(III)中間体は接近可能なチロシンの芳香環から電子を引き抜き、プロトンを失った後にチロシンのラジカルが生じる(図5参照)、と考えられている。前記の強く活性化されたラジカル中間体は、近傍のチロシンに結合し、クロスリンクされた付加化合物が生じる。
チロシンクロスリンクの機構
ルテニウム(III)トリス(ビピリジル)ジカチオン[Ru(bpy)3 2+]と過硫酸アンモニウム(APS)のような電子受容体の存在下で[18,20,21]可視光を照射すると、接触するタンパク質間で非常に効率の良いクロスリンクが誘導される。この過程はたいへん効率が良く、その機構はNi/GGH/MMPPの場合と似ていると考えられている。ColP前駆体および/またはColD前駆体の自己集合で形成された繊維は、APSの存在下で[Ru(bpy)3 2+]とともに光照射に供する。このことにより、足糸コラーゲン中のチロシン/DOPAのクロスリンクが増加し、機械的特性の異なった繊維が形成され、前記特性は上述した物理化学的キャラクタリゼーションによって評価されることになる。
イガイ類ColP前駆体とColD前駆体のDNA配列を下記に示す。両方のCol前駆体(PとD)のcDNAをpGEM−Tクローニングベクターに組み込んだ。出発材料を確認するために、両方のcDNAを完全にシーケンスし、スタンダードなプライマーとしてT7とSP6を用い、内部プライマーとしてpreCol(PまたはD)−T7/1とSP6/1をそれぞれ用いた。得られたDNA配列は、出版されたヴァージョンにおける両方のCol前駆体の配列とは異なっており、それに応じて比較を行った。

a)前記コラーゲン類似組み換えタンパク質をコードする、第一の発現ベクター;および、
b)プロリル−4−ヒドロキシラーゼ(P4H)をコードする核酸を含む、第二の発現ベクター。 - 請求項1に記載の酵母細胞であって、前記P4Hの配列にシグナル配列が連結されており、前記酵母細胞の小胞体へ、前記配列が効率良く輸送される酵母細胞。
- 請求項2に記載の酵母であって、前記シグナル配列がS. cerevisiae(配列番号10)のmating factor alpha 1(MFa)である酵母。
- 請求項1〜3のいずれか1項以上に記載の酵母細胞であって、好ましくはS. cerevisiae、Schizosaccharomyces Pombe、Pichia pastoris、Candida albicans、またはHansenula polymorphaである酵母細胞。
- 請求項1〜4のいずれか1項以上に記載の酵母細胞であって、前記第一の発現ベクターが、1つ以上の制御因子をさらに含む酵母細胞。
- 請求項5に記載の酵母細胞であって、前記制御因子が、恒常性または誘導性のプロモーターから選択されたプロモーターを含む酵母細胞。
- 請求項6に記載の酵母細胞であって、前記プロモーターがGPD、GAL4、CUP1、MET25、GAL1、またはGAL1−10から選択される酵母細胞。
- 前記請求項のいずれか1項以上に記載の酵母細胞であって、前述の発現ベクターがプラスミドである酵母細胞。
- 前記請求項のいずれか1項以上に記載の酵母細胞であって、前記コラーゲン類似組み換えタンパク質が、エラスチン若しくはシルクフィブロインに隣接した1つ以上のコラーゲンドメイン断片から成る、またはそれを含む、イガイ類足糸組み換えタンパク質である酵母細胞。
- 前記請求項のいずれか1項以上に記載の酵母細胞であって、前記コラーゲンドメイン断片がイガイ類に由来するもので、好ましくはM. edulis、M. galloprovincialis、M. californians、またはGeukeria demissaである酵母細胞。
- 前記請求項のいずれか1項以上に記載の酵母細胞であって、前記イガイ類足糸組み換えタンパク質が、ColP前駆体および/若しくはColD前駆体またはそれらの変異体の断片1つ以上から成る、またはそれを含む、酵母細胞。
- 前記請求項のいずれか1項以上に記載の酵母細胞であって、前記組み換えタンパク質が、配列番号3および/若しくは4、またはそれらの変異体のアミノ酸配列から成る、またはそれを含む、酵母細胞。
- 前記請求項のいずれか1項以上に記載の酵母細胞であって、前記組み換えタンパク質において、それぞれのアミノ酸配列のシグナル配列が、酵母特異的なシグナル配列、好ましくはS. cerevisiaeのmating factor alpha 1(MFa)に置換された、酵母細胞。
- 前記請求項のいずれか1項以上に記載の酵母細胞であって、P4Hがヒトまたはイガイ類のP4Hである、酵母細胞。
- 次に示す構成要素を含むタンパク質を含む組み換えコラーゲンの製造に用いる、部品のキットまたは共発現システム。
a)請求項1〜14のいずれか一つ以上により定義される第一の発現ベクター;および
b)請求項1〜14のいずれか一つ以上により定義される第二の発現ベクター; - コラーゲン類似組み換えタンパク質、好ましくはイガイ類足糸タンパク質を製造する方法であって、次の工程を含む方法。
a)酵母細胞を提供する工程;
b)前記酵母細胞を、請求項1〜14のいずれか1項以上で定義される第一および第二の発現ベクター若しくは請求項15に記載の共発現システムで形質転換する工程;
c)適切な条件下で、前記酵母細胞から前記コラーゲン類似組み換えタンパク質、好ましくはイガイ類足糸組み換えタンパク質を発現させる工程;および、
d)前記組み換えタンパク質を回収する工程。 - イガイ類足糸組み換えタンパク質から糸を製造する方法であって、次に示す工程を含む方法。
a)請求項16で製造された組み換えタンパク質の提供;および、
b)適切な方法によって前記タンパク質を(電気的に)紡績、または型どりをする工程。 - 請求項16の方法で製造可能なタンパク質または請求項17に記載の方法で製造可能な糸。
- バイオテクノロジーおよび/または医学の分野での、請求項18に記載のタンパク質・糸の使用。
- 創傷治癒または保護システムの製造のための、請求項18に記載のタンパク質/糸の使用。
- 縫合材料を製造するための請求項20の使用。
- 前記縫合材料が神経外科または眼科の外科術での使用を意図された、請求項21に記載の使用。
- 代替材料、好ましくは人工の軟骨または腱の材料を製造するための請求項18に記載のタンパク質/糸の使用。
- 請求項18に記載のタンパク質/糸を使って製造することができる、創傷治癒または保護システム、縫合材料、代替材料、好ましくは人工の軟骨若しくは腱の材料。
- 請求項18に記載のタンパク質/糸を含む、化粧品、医薬品送達媒体、布地、織物、紙製品、皮革製品、自動車の部品、または航空機の部品。
