Abstract

上述したスクリーニング目的の非侵襲的蛍光イメージングシステムを構築する際に、トランスジェニックGFAP−GFPマウス(Zhuo et al., 1997によって過去に作製)の新生児脳における神経毒物のモデルとして、2’−CH3−MPTPを使用した。以下に示されるように、トランスジェニックGFAP−GFPマウスモデルを適切なインビボ光学的イメージングシステムと組み合わせることで、トランスジェニックマウスの体内におけるGFAP−GFPの特徴を確実に検出することができ、MTPTの導入に応じた上方制御を量依存的かつ時間依存的に定量化することができる。このような合理的なスループットを示す非侵襲的光学的蛍光システムは、パーキンソニズム等の神経変性疾患や発育神経毒性の研究、或いは前臨床化合物スクリーニングにおいて、幅広い応用が可能である。
(トランスジェニックGFAP−GFPマウス)
トランスジェニックGFAP−GFPマウスの作製及びジェノタイピングは、過去の報告(Zhuo et al., 1997)に従って行った。4日齢、体重2.0g〜2.5gのFVB/N系統の新生児マウスを実験に用いた。動物の管理は、国立シンガポール大学の動物飼育施設で行った。本研究の実験プロトコルは、研究機関内の動物の管理及び使用に関する委員会(Institutional Animal Care and Use Committee)によって承認されている。
被検物質である2’−CH3−MPTPは、従来からのMPTPよりも効果の強い類縁体であり(Abdel−Wahab, 2005)、Sigma−Aldrichから購入した(Sigma−Aldrich, M−103)。新生児マウスには、0時間の時点で2’−CH3−MPTP(12mg/kgの生理食塩水溶液)を単回皮下注射(sc)した。対照群には、担体として生理食塩水を用いた。そして、投与後2時間,4時間,6時間,8時間の時点で神経イメージングを行った。
IVIS(登録商標)イメージングシステム100シリーズ(Xenogen, Alameda, CA, USA)を用いて、非侵襲的イメージングを行った。先ず、2’−CH3−MPTPで処理されたトランスジェニック及び非トランスジェニックのペアのマウスを隣同士に配置し、麻酔を施さず、マウスの下背部をテープによって固定した。これは、小児麻酔に広く用いられているケタミンが、新生児ラットにおいては神経のアポトーシスを増加させることにより発育神経毒性を示すという最近の研究(Scallet et al., 2004)に基づくものである。その後、一対の新生児マウスについて、IVIS(登録商標)システムに備えられたGFPフィルターセットで蛍光イメージングを行った。画像収集時間は10秒間であった。さらに、生理食塩水が注射された対照のペアについても同様の操作を行った。そして、マウスに対する2’−CH3−MPTPの神経毒性作用を相対蛍光(RF)、すなわちトランスジェニック(Tg)マウスにおける所望の領域(ROI)から検出された全蛍光と、非トランスジェニック(nTg)マウスのROIから検出された組織自己蛍光との比(TgROI/nTgROI)として表現した。両者のマウスのROIは完全に同一であり、このことは本研究で輝度を定量化するために使用した全てのROIについて共通である。イメージングシステムに起因するバラつきを最小化するため、処理マウス及び対照マウスの各ペアについてそれぞれ3枚の画像を取得し、その平均値をその特定のペアにおける定量値とした。対照群(n=5ペアのマウス)の値と処理群(n=5ペアのマウス)の値とは平均値±SEMとして表され、統計的有意性は、スチューデントの2標本両側t検定によって評価した。
CNSのアストロサイトからフォトンが放出されているか否かを確認するため、脳を摘出し、4%パラホルムアルデヒドを含有する0.1Mリン酸緩衝生理食塩水(PBS, pH7.4)によって4℃、4時間の条件で固定化した。PBS中で15分間ずつ3回インキュベーションすることにより脳を洗浄し、その後、30%ショ糖溶液中に4℃で一晩浸漬した。全ての脳を液体窒素中に浸漬することにより凍結溶媒中に埋め込み、クリオスタット(Leica Microsystems, Nussloch GmbH; CM−3050S)を用いてそれらを切片化した。得られた冠状凍結切片(20μm厚の黒質;ブレグマ −3.16mm、両耳間 0.64mm)を、Atlas of Mouse brain(Franklin and Paxinos,2001)に従って免疫組織化学に用いた。対照群及び処理群のそれぞれは4匹の動物を含む(n=3)。
(GFP蛍光レベルに対する2’−CH3−MPTPの影響)
先ず、脳のROIから内在的GFP蛍光を検出し、Living Image(登録商標)ソフトウェア(Xenogen Corp.)を用いて、図1(A)に示すように神経領域からの発光をフォトン/秒/cm2/ステラジアン(sr)の単位で定量化した。CNSのアストロサイトからフォトンが放出されているか否かを確認するため、インビボでの最後のイメージングが終わった時点で、脳を摘出し、エクスビボでのイメージングを行った(図1(B))。この方式により、対照トランスジェニックマウス及び処理トランスジェニックマウスについて、GFP蛍光を定性的かつ定量的に比較することが可能になる。
海馬におけるGFAP免疫染色の代表画像を図4に示す。アストロサイトにおけるGFP発現は、2’−CH3−MPTP(12mg/kg sc)の単回投与から6時間の時点で、脳室領域及び海馬領域において明らかに増加していた。非トランスジェニックマウスでは、GFP発現は観察されなかった(図4のA)。GFP発現細胞がアストロサイトであることを確かめるため、同じ切片についてGFAPの免疫染色を行ったところ、海馬溝におけるGFAP免疫陽性の血管周囲アストロサイトと内在性GFPの蛍光が観察されたアストロサイトとの共局在化は、殆どが突起部分で生じており、細胞体では生じていなかった(2重ラベルされた細胞突起を図4のD,Eにおいて矢印で示す)。
黒質緻密部(SNC)におけるTH免疫染色の代表画像を図5に示す。TH抗体が結合したドーパミン作動性ニューロンのサイズ(〜20μm)は、免疫組織化学染色を行っていない、内在性GFPの蛍光が観察されたアストロサイトのサイズ(〜10μm)よりも大きく、2’−CH3−MPTPで処理した新生児マウスのSNCにおける検出が容易であった。ドーパミン作動性細胞の細胞体や繊維は強く染まっており、SNCには明らかに免疫陽性突起が存在した。2’−CH3−MPTP(12mg/kg sc)の処理から6時間の時点で、TH免疫陽性の繊維及び細胞体の減少は観察されなかった。しかしながら、アストロサイト内におけるGFAP−GFP導入遺伝子の発現は、0時間の時点と比べて6時間の時点では、明らかに上方制御されていた。図5のAに示すように、非トランスジェニックマウスのSNCにおけるドーパミン作動性ニューロンは、TH免疫陽性であったが、グリア細胞は導入遺伝子を発現しなかった。TH免疫陽性のドーパミン作動性ニューロンと、免疫組織化学染色を行っていない、内在性GFPの蛍光が観察されたアストロサイトとの共局在化は観察されなかった。
(皮弁インビボイメージング)
麻酔が施されたトランスジェニックGFAP−GFP成体マウスに皮弁(片側を残したまま皮膚を体から切り剥がしたもの)を形成した。脳及び肝臓の皮弁が形成された部分について、IVIS(登録商標)イメージングシステム100シリーズ(Xenogen, Alameda, CA, USA)を用いて、インビボイメージングを行った。マウスの体の他の部分からの組織自己蛍光を遮蔽するために低蛍光の黒紙を用い、脳(図6(A))及び肝臓(図6(B))の皮弁が形成された部分のみを露出させた。
図7(A)〜(D)に示すように、トランスジェニック(Tg)マウス及び非トランスジェニック(nTg)マウスの両者から脳、肝臓、腎臓、及び坐骨神経を摘出し、IVIS(登録商標)イメージングシステムを用いてエクスビボでイメージングを行った。放出されたフォトンの数に対応した擬似カラー画像において、TgマウスとnTgマウスとで有意差が観察された。現在、成体マウスに対する代替的な画像診断法の実現可能性調査を行っているところである。
[実験手順]
(動物)
この研究は、4日齢の新生児マウス(実験時点で体重3〜5g)を用いて行った。動物は自由に摂食させ、使用する動物数及び動物の苦痛を最小限とするように努めた。
使用した神経毒は2’−CH3−MPTPである。8mg/kgのMPTPを2時間毎に4回皮下注射し、投与後24時間、48時間、72時間の時点でインビボ神経画像を取得した。
先ず、BCATMタンパク質アッセイ及びウエスタンブロットのため、投与後24時間の時点でマウスを犠牲死させた。そして、脳を摘出し、リン酸緩衝生理食塩水(PBS)及びポトテイナーゼ阻害剤を含むサンプルバッファで溶解した。全タンパク質は、ウシ血清アルブミン(BSA)を標準として、ビシンコニン酸(BCA)法で確認した。次に、そのサンプルを、NuPAGE Novex ビス−トリスゲル(4%〜12%)を用いて、NuPAGE Novex ビス−トリス電気泳動にかけた。各サンプルは15μgのタンパク質を含む。電気泳動後、タンパク質をニトロセルロース膜に転写した。その後、5%無脂肪ドライミルク及び0.1%Tween 20を含有するPBS(PBS−T)を用いて、4℃で膜のブロッキングを行った。続いて、その膜を1:1000に希釈したポリクローナルウサギ抗ヤギGFAP抗体(Dako)/ウサギポリクローナル抗GFP抗体(abcam)とともに室温で1時間インキュベートした後、PBS−Tで15分間ずつ3回洗浄した。さらに、その膜を1:2000に希釈したヤギ抗ウサギIgG−HRP2次抗体(Santa Cruz)とともに室温で1時間インキュベートした後、PBS−Tで5分間ずつ3回洗浄した。そして、化学発光検出溶液(ECL Plus, Amersham)及びオートラジオグラフィーフィルムを用いて、2次抗体を可視化した。ウエスタンブロットの終了後、そのニトロセルロース膜をIVIS(登録商標)装置でイメージングした。ECL Plus検出溶液を用いた場合、結合したHRP、及びLumigen PS−3 アクリダン基質の過酸化物によって触媒された酸化反応により、毎分何千ものアクリジニウムエステル中間体が生成される。この中間体は弱アルカリ性条件下で過酸化物と反応し、430nmが極大となる、長時間かつ高強度の化学発光を生じる。この化学発光シグナルは、IVIS(登録商標)イメージングシステムを用いて検出することができる。発光強度を示すカラーの膜の画像が得られる。
先ず、リアルタイムRT−PCRのため、投与後24時間の時点でマウスを犠牲死させた。そして、脳を摘出し、RA1バッファ及びβ−メルカプトエタノールで溶解した。次に、NucleoSpin(登録商標) RNA II キットを用いて、脳から全RNAを抽出した。そして、ランダムヘキサマーを用いてその全RNAを逆転写し、遺伝子発現を定量化するために、得られたcDNAについてリアルタイムRT−PCRを行った。
MPTP(4×8mg/kg)
(インビボ神経画像)
図8に示すように、脳における所望の領域(ROI)から内在的GFP蛍光を検出し、Living Image(登録商標)ソフトウェア(Xenogen Corp.)を用いて、神経領域からの発光をフォトン/秒/cm2/ステラジアン(sr)の単位で定量化した。インビボ画像に示されるように、MPTP(8mg/kg sc)が4回投与されたトランスジェニック新生児マウスは、投与後24時間の時点の対照マウスと比較したとき、ROIにおけるGFP蛍光(フォトン/秒/cm2/sr)が最も有意に増大していた。図9に示すGFP蛍光の定量値によると、MPTP処理群と対照群とでは、投与後24時間の時点で相対蛍光(RF)に有意差が観察され、対照群に対して平均15%と有意(p<0.01)に増大した。MPTP(12mg/kg sc)が単回投与されたトランスジェニック新生児マウスや、対照群内では、経時的なRFの有意差は観察されなかった(図9)。
図10(A)に示すウエスタンブロット結果から分かるように、Dakoの抗ウサギGFAP抗体を用いることで、マウスGFAPについて報告された分子量と一致した約51kDaの位置にバンドが検出された。また、Abcamの抗ウサギGFP抗体を用いることで、マウスGFPについて報告された分子量と一致した約27kDaの位置にバンドが検出された。バンドの太さから、トランスジェニックマウスにおいては、0時間から24時間までの時点で、GFAPタンパク質及びGFPタンパク質の量が増加していることが分かる(レーンaとレーンbとの比較)。非トランスジェニックマウスにおいても、0時間から24時間までの時点で、GFAPタンパク質の量が増加している(GFAPウエスタンブロットにおけるレーンcとレーンdとの比較)。非トランスジェニックマウス(GFPウエスタンブロットにおけるレーンc,d)においては、導入遺伝子が存在しないため、GFPタンパク質は検出されなかった。IVIS(登録商標)を用いてウエスタンブロット膜をイメージングしたところ、タンパク質量を表すカラー画像は、同様の結果を示していた(図10(B)を参照)。このことは、MPTPの投与によってGFAPが上方制御され、これによりGFPも上方制御されるという事実を補強するものである。
mRNAの研究は、BCAのタンパク質における研究結果や、長い研究過程ではインビボ神経画像と一致するものである。図11(A)に示すように、MTPT処理後24時間の時点におけるGFAPの遺伝子発現は、トランスジェニックマウス及び非トランスジェニックマウスの両者とも、未処理のマウスと比較してそれぞれ91%(p<0.01)、88%(p<0.01)と有意に誘導されていた。しかしながら、MTPT処理後24時間の時点におけるGFPの遺伝子発現は、未処理のマウスと比較して有意に誘導されていなかった(図11(B))。これにより、MPTP処理後48時間、72時間の時点で神経画像における相対蛍光が減少していることが説明できる(図8,図9を参照)。
結果を図12,図13に示す。
[実験手順]
(動物)

使用した神経毒は2’−CH3−MPTP及びカイニン酸(KA)である。MPTPの投与は、12mg/kgの濃度の単回注射により行われた。KAの投与は、2mg/kgの濃度の単回注射により行われた。注射は全て皮下である。

図14(A)に示すウエスタンブロット結果から分かるように、Dakoの抗ウサギGFAP抗体を用いることで、マウスGFAPについて報告された分子量と一致した約51kDaの位置にバンドが検出された。また、Abcamの抗ウサギGFP抗体を用いることで、マウスGFPについて報告された分子量と一致した約27kDaの位置にバンドが検出された。バンドの太さから、トランスジェニックマウスにおいては、0時間から6時間までの時点で、GFAPタンパク質及びGFPタンパク質の量が増加していることが分かる(レーンaとレーンbとの比較)。非トランスジェニックマウスにおいても、0時間から6時間までの時点で、GFAPタンパク質の量が増加している(GFAPウエスタンブロットにおけるレーンcとレーンdとの比較)。非トランスジェニックマウス(GFPウエスタンブロットにおけるレーンc,d)においては、導入遺伝子が存在しないため、GFPタンパク質は検出されなかった。IVIS(登録商標)を用いてウエスタンブロット膜をイメージングしたところ、タンパク質量を表すカラー画像は、同様の結果を示していた(図14(B)を参照)。このことは、MPTPの投与によってGFAPが上方制御され、これによりGFPも上方制御されるという事実を補強するものである。
mRNAの研究は、BCAのタンパク質における研究結果やウエスタンブロットと一致するものである。GFAPタンパク質及びGFPタンパク質の上方制御は6時間の時点でピークとなるので、GFAP及びGFPのmRNAの上方制御は、MPTP処理の2時間後には生じていた。この時間差は、mRNAからタンパク質に翻訳する時間を表している。図15に示すように、MTPT処理後2時間の時点におけるGFAP及びGFPの遺伝子発現は、それぞれ47%(p<0.001)、20%(p<0.01)と有意に誘導されていた。
(インビボ神経画像)
図16に示すように、脳における所望の領域(ROI)から内在的GFP蛍光を検出し、Living Image(登録商標)ソフトウェア(Xenogen Corp.)を用いて、神経領域からの発光をフォトン/秒/cm2/ステラジアン(sr)の単位で定量化した。インビボ画像に示されるように、KA(2mg/kg sc)が単回投与されたトランスジェニック新生児マウスは、投与後6時間の時点の対照マウスと比較したとき、ROIにおけるGFP蛍光(フォトン/秒/cm2/sr)が最も有意に増大していた。図17に示すGFP蛍光の定量値によると、KA処理群と対照群とでは、投与後4時間、6時間の時点で相対蛍光(RF)に有意差が観察され、処理群については、投与後6時間の時点で対照群に対して平均25%と最も有意(p<0.01)に増大した。KA処理群内では同様であり、0時間の時点と比較すると、処理後6時間の時点で平均22%と最も有意(p<0.05)に増大した。対照群内では、経時的なRFの有意差は観察されなかった(図17)。
図18(A)に示すウエスタンブロット結果から分かるように、Dakoの抗ウサギGFAP抗体を用いることで、マウスGFAPについて報告された分子量と一致した約51kDaの位置にバンドが検出された。また、Abcamの抗ウサギGFP抗体を用いることで、マウスGFPについて報告された分子量と一致した約27kDaの位置にバンドが検出された。バンドの太さから、トランスジェニックマウスにおいては、0時間から6時間までの時点で、GFAPタンパク質及びGFPタンパク質の量が増加していることが分かる(レーンaとレーンbとの比較)。非トランスジェニックマウスにおいても、0時間から6時間までの時点で、GFAPタンパク質の量が増加している(GFAPウエスタンブロットにおけるレーンcとレーンdとの比較)。非トランスジェニックマウス(GFPウエスタンブロットにおけるレーンc,d)においては、導入遺伝子が存在しないため、GFPタンパク質は検出されなかった。IVIS(登録商標)を用いてウエスタンブロット膜をイメージングしたところ、タンパク質量を表すカラー画像は、同様の結果を示していた(図18(B)を参照)。このことは、KAの投与によってGFAPが上方制御され、これによりGFPも上方制御されるという事実を補強するものである。
mRNAの研究は、BCAのタンパク質における研究結果やウエスタンブロットと一致するものである。GFAPタンパク質及びGFPタンパク質の上方制御は6時間の時点でピークとなるので、GFAP及びGFPのmRNAの上方制御は、MPTP処理の2時間後には生じていた。この時間差は、mRNAからタンパク質に翻訳する時間を表している。図19に示すように、KA処理後2時間の時点におけるGFAP及びGFPの遺伝子発現は、それぞれ21%(p<0.01)、22%(p<0.05)と有意に誘導されていた。
海馬のCA1,CA2,及びCA3サブエリアにおけるGFAP免疫染色の代表画像を図20のA〜Dに示す。アストロサイトにおけるGFP発現は、KA(2mg/kg sc)の単回投与から6時間の時点で、様々な領域において明らかに増加していた。GFP発現細胞がアストロサイトであることを確かめるため、同じ切片についてGFAPの免疫染色を行ったところ、海馬のCA1領域におけるGFAP免疫陽性のアストロサイトと内在性GFPの蛍光が観察されたアストロサイトとの共局在化は、殆どが突起部分で生じており、細胞体では生じていなかった(2重ラベルされた細胞突起を図20のBにおいて矢印で示す)。
トランスジェニックマウスにおいて、グリア細胞線維性酸性タンパク質(GFAP)プロモーターは緑色蛍光タンパク質(GFP)の発現を誘導する。GFAPは、主として中枢神経系(CNS)のアストロサイトに発現している中間フィラメントタンパク質である。これらのグリア細胞における変化は、神経活性のモニターに利用することができる。CNSのニューロンに対する傷害により、GFAPが強く誘導される。しかしながら、インビトロアッセイでGFAPをモニタリングするには、動物を犠牲死させる必要がある。本件発明者らは、GFAP−GFP発現の非侵襲的インビボ神経イメージングにより、長期間に亘りGFAPをモニターすることができ、この過程で犠牲となる動物の数を減らせることを示した。

- 動物の体内における目的とする蛍光タンパク質の発現を検出することを含む方法であって、
前記動物は、前記蛍光タンパク質をコードする核酸が該動物の神経系で通常発現しているタンパク質のプロモーター核酸に作動可能に連結されたゲノムを有するトランスジェニック動物であり、
前記動物の体内で発現された前記タンパク質からの蛍光を非侵襲的に検出する工程を含む、前記方法。 - 前記プロモーターがGFAPプロモーターである、請求項1記載の方法。
- 前記蛍光タンパク質が緑色蛍光タンパク質(GFP)である、請求項1又は2記載の方法。
- 前記動物に対する被検物質の投与による影響を検査するための方法であり、

(iii)前記被検物質の投与後に、前記(ii)と同じ動物における蛍光を非侵襲的に検出する工程、及び
(iv)前記(ii)で検出された蛍光と前記(iii)で検出された蛍光とを比較する工程を含む、請求項1から5のいずれか1項記載の方法。 - 神経障害の治療促進又は調節に関する被検物質の効果を検査する方法であり、
(i)対照とするため、第1の動物に神経障害を誘導し、得られた神経障害動物における蛍光を非侵襲的に検出する工程、
(ii)第2のトランスジェニック動物に神経障害を誘導し、前記被検物質を該動物に投与した後、該動物における蛍光を非侵襲的に検出する工程、及び
(iii)前記(i)で検出された蛍光と前記(ii)で検出された蛍光とを比較する工程を含む、請求項1から5のいずれか1項記載の方法。 - 前記神経障害は、所定の化学物質を前記動物に投与することにより誘導される、請求項9又は10記載の方法。
- 前記化学物質は、所定の疾患についての化学誘導動物モデルを得るために投与される、請求項11記載の方法。
- 前記疾患が、パーキンソン病、アルツハイマー病、及びハンチントン病の中から選択される、請求項12記載の方法。
- 前記化学物質が、MPTP、2’−CH3−MPTP、6−ヒドロキシドーパミン(6−OHDA)、カイニン酸、トリメチル錫、クロルピリホス(CPF)、エチレンビス(ジチオカルバミン酸)マンガン(Maneb又はMB)、ロテノン、パラコート(N,N’−ジメチル−4,4’−ビピリジニウム)、ポリ塩化ビフェニル(PCB)、3,3’−イミノジプロピオニトリル(IDPN)、及びトルエン(メチルベンゼン)の中から選択される、請求項11から13のいずれか1項記載の方法。
- 前記神経障害は、前記動物の頭部及び/又は首及び/又は背中に対する物理的な力によって誘導される、請求項9又は10記載の方法。
- 前記神経障害は、前記動物の神経系への酸素供給を減らし、脳虚血を引き起こすことにより誘導される、請求項9又は10記載の方法。
- 神経系疾患の治療促進に関する被検物質の効果を検査する方法であり、
(i)前記神経系疾患の動物モデルでもある前記トランスジェニック動物を準備する工程、
(ii)対照とするため、前記被検物質の投与前に、前記(i)の動物における蛍光を非侵襲的に検出する工程、
(iii)前記被検物質の投与後に、前記(ii)と同じ動物における蛍光を非侵襲的に検出する工程、及び
(iv)前記(ii)で検出された蛍光と前記(iii)で検出された蛍光とを比較する工程を含む、請求項1から5のいずれか1項記載の方法。 - 神経系疾患の治療促進に関する被検物質の効果を検査する方法であり、
(i)前記神経系疾患の動物モデルでもある第1及び第2の動物を準備する工程、
(ii)対照とするために、前記(i)の第1の動物における蛍光を非侵襲的に検出する工程、
(iii)前記被検物質を前記(ii)の第2のトランスジェニック動物に投与した後、該動物における蛍光を非侵襲的に検出する工程、及び
(iv)前記(ii)で検出された蛍光と前記(iii)で検出された蛍光とを比較する工程を含む、請求項1から5のいずれか1項記載の方法。 - 神経系疾患の治療効果を検査する方法であり、
(i)前記神経系疾患の動物モデルでもある前記トランスジェニック動物を準備する工程、
(ii)対照とするため、治療を施す前に、前記(i)の動物における蛍光を非侵襲的に検出する工程、
(iii)治療を施した後に、前記(ii)と同じ動物における蛍光を非侵襲的に検出する工程、及び
(iv)前記(ii)で検出された蛍光と前記(iii)で検出された蛍光とを比較する工程を含む、請求項1から5のいずれか1項記載の方法。 - 神経系疾患の治療効果を検査する方法であり、
(i)前記神経系疾患の動物モデルでもある第1及び第2の動物を準備する工程、
(ii)対照とするために、前記(i)の第1の動物における蛍光を非侵襲的に検出する工程、
(iii)前記(ii)の第2のトランスジェニック動物に治療を施した後、該動物における蛍光を非侵襲的に検出する工程、及び
(iv)前記(ii)で検出された蛍光と前記(iii)で検出された蛍光とを比較する工程を含む、請求項4又は5記載の方法。 - 前記治療がX線又はγ線の照射を含む、請求項19又は20記載の方法。
- 前記神経系疾患が神経系腫瘍である、請求項17から21のいずれか1項記載の方法。
- 前記腫瘍が、神経膠腫、髄芽腫、髄膜腫、神経繊維腫、上衣腫、神経鞘腫、神経線維肉腫、星状細胞腫、及び乏突起膠腫の中から選択される、請求項22記載の方法。
- 前記動物が哺乳動物である、請求項1から23のいずれか1項記載の方法。
- 前記動物が新生児である、請求項1から24のいずれか1項記載の方法。
