发明背景

尽管临床上和病理学上偶发和家族性PD可以在几个重要方面不同,但是它们共有相同的生化脑部异常,即脑多巴胺的显著减少(Dauer andPrzedborski,2003)。PD患者显示低水平脑多巴胺的原因源于黑质纹状体多巴胺能途径的退化,所述多巴胺能途径由多巴胺能神经元构成,其细胞体位于黑质中,伸出的轴突和神经末梢可见于纹状体中(Vila andPrzedborski,2004)。
PD的最佳表征模型已经通过使用神经毒素MPTP而开发出来(Bloemet al.,1990,Flint Beal,2001)。对MPTP的发现发生在1982年,当时加利福尼亚的一群吸毒者表现出严重帕金森综合征的急性发作。研究发现该综合征是由于自身注射一种合成的海洛因类似物而引起,而这种海洛因类似物在制造过程中被副产物MPTP污染。随后,MPTP给药显示在小鼠和灵长类动物中模拟PD。MPTP为高度亲脂的,其可容易地穿过血脑屏障。随后,其被B型单胺氧化酶转化为其活性代谢物-1-甲基-4-苯基吡啶鎓(MPP+),然后该活性代谢物被高亲和性多巴胺和去甲肾上腺素摄取系统摄取,并由此在黑质纹状体多巴胺能细胞的线粒体内聚集。这可导致对细胞功能的多种有害作用,引起神经细胞死亡(Tatton and Kish,1997,Tanji et al.,1999)。在小鼠中,2′-CH3-MPTP是比常规MPTP更为有效的神经毒素,显示严重的组织病理学变化,包括细胞质膨胀、间质水肿、多巴胺能神经元减少以及反应性小神经胶质细胞增殖和神经胶质增生(Abdel-Wahab,2005)。
尽管对PD的大部分研究集中于成体进行,一些报告令人信服地证实对新生小鼠的全身MPTP注射导致永久的脑损伤,在成年期也还有痕迹。发育中的脑对MPTP损伤敏感的事实还值得进一步研究。
除了MPTP,也已报道了多种其它的化学品在成体小鼠脑的不同区域引起神经损伤。这些化学品包括一系列结构和功能上不同的化合物,从工业上有毒的化合物、农业杀虫剂到食品添加剂。同样,迄今为止进行的大部分神经毒性研究基于成体模型。但是,值得注意的是,具有较不完整血脑屏障的发育中的脑更易于受到已知神经毒素的损伤,更关键的是,该施加的损伤不能由发育中的脑完全和正确地代偿。因此,对于测试化合物在发育中的脑中的潜在神经毒性而言,其重要性无论如何强调也不过分,尤其是在处理沉默神经毒性(即,早期暴露于神经毒剂,但是直至成年期才有临床症状)的问题时。美国环保局的发育神经毒性测试指南(Developmental Neurotoxicity Testing Guideline,DNTG)进一步强调了这点。最近,欧委会采纳了用新的欧盟管理框架来通过严格制度测试所有的化学品的建议,据估计新措施耗资高至70亿欧元并需要至少10年来实施。
O′Callaghan(O′Callaghan,1988)已提出,在中枢神经系统(CNS)的星形细胞中优势表达的中间纤维蛋白-胶质纤维酸性蛋白(GFAP),为监测成体啮齿类动物脑中由各种神经毒剂(包括1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP))引起的神经损伤的早期和灵敏的生物标记(Reinhard et al.,1988,Araki et al.,2001,Chen et al.,2002,Fields and tevens-Graham,2002,Kurosaki et al.,2004)。GFAP表达上调与神经学损伤增加相关。用于分析内源GFAP表达的常规方法主要包括免疫细胞化学、Northern印迹、Western印迹和ELISA(O′Callaghan,1991,Eng et al.,2000)。
GFAP基础启动子包括TATA和CAAT盒。增强子和沉默子序列可见于-250和-80bp之间以及-1980和-1500bp之间。这些正调节区含有很多转录因子的共有序列,包括cAMP应答元件和Sp-1、NF-1、AP-1以及AP-2转录因子的结合位点。组织特异性由位于-1980至-1500bp区域的人GFAP共有序列所赋予。
反应性神经胶质增生(星形胶质增生)在应答对中枢神经系统(CNS)的几乎任何物理或化学损伤中产生,以星形细胞体及其细胞突的肥大为特征,伴随有GFAP表达的增加。反应性神经胶质增生伴随有GFAP的上调。在外周神经系统创伤和毒性损伤后发生类似的GFAP增加。
随着小鼠转基因学的进展和新的报告基因的获得,包括β-半乳糖苷酶(lacZ)、绿色荧光蛋白(GFP)和荧光酶(LUC)在内的几种报告基因被引入转基因小鼠中在GFAP启动子控制下,并被证实是体内研究神经胶质增生过程中GFAP转录活性的有用替代手段(Brenner et al.,1994,Zhuo et al.,1997,Zhu et al.,2004)。
WO 00/02997和US 6,501,003描述了在神经胶质细胞中表达绿色荧光蛋白的转基因小鼠的产生。作者描述了在全包埋组织或者其切片中体外检测视神经、脑、视网膜、坐骨神经和角膜中的荧光。
发明内容
出于有助于研究PD的发育方面和神经毒性的目的,本发明人建立了非侵入性系统,其包括GFAP-GFP转基因小鼠模型和可购得的成像系统,其使得能够在活体动物中研究神经学疾病、损伤和毒性的各个方面。该系统还使以有效方式筛选大量化学品对发育中神经系统的神经毒性危险成为可能。
通过采用表达荧光蛋白的转基因动物(其中该荧光蛋白位于来自通常在动物神经系统中表达的蛋白的启动子控制下),本发明人已证实在整体活体转基因动物中的荧光检测可用于监测刺激物和测试物对来自该启动子的荧光蛋白表达的影响。这相对于现有技术有显著改进,其使得能在体内实时或接近实时地监测测试物对完整动物神经系统的影响。
最概括而言,本发明涉及在转基因动物中非侵入性检测来自荧光蛋白的荧光的方法。
根据本发明的第一方面,提供了一种涉及检测动物中目的荧光蛋白表达的方法,其中所述动物为转基因动物,在其基因组中具有编码所述荧光蛋白的核酸,所述编码所述荧光蛋白的核酸可操作地连接来自通常在所述动物神经系统中表达的蛋白的启动子核酸,所述方法包括非侵入性检测在所述动物中表达的所述蛋白的荧光的步骤。
相应地,还提供了转基因动物在非侵入性检测在所述动物中表达的所述荧光蛋白的荧光的方法中的用途,其中所述转基因动物在其基因组中具有编码所述荧光蛋白的核酸,所述编码所述荧光蛋白的核酸可操作地连接来自通常在该动物神经系统中表达的蛋白的启动子核酸。
所述方法优选用于研究向所述动物给予测试物或施用测试刺激物的作用,其中所述方法包括在非侵入性检测荧光的步骤之前向所述动物给予所述测试物或刺激物的步骤。优选地,所述方法用于研究测试物或刺激物对动物的神经学状况的影响。因此,本发明的方法可以提供用于筛选神经学调节剂的方法。
该方法优选包括将从不给予所述测试物的对照动物检测的荧光和给予所述测试物至所述转基因动物后从所述转基因动物检测的荧光进行比较的步骤。
对不给予测试物情况下的荧光的检测使得能提供对照值或基础读数(例如自体荧光),其可以与给予测试物后的荧光检测一起使用,出于标准化的目的计算相对荧光值。所述对照值可以在转基因动物例如其中待测试测试物的相同类型转基因动物中检测,或者在非转基因动物中检测。所述动物优选为相同类型,例如小鼠,最优选相同品系。为了获得对照值,可以向对照动物给予安慰剂,例如盐水。
因此,在一个优选的实施方案中,所述方法包括以下步骤:
(i)在给予测试物之前非侵入性检测所述转基因动物中的荧光以提供对照;
(ii)在给予测试物之后非侵入性检测所述相同动物中的荧光;以及
(iii)将在(i)中检测的荧光与在(ii)中检测的荧光进行比较。
在另一可选的优选实施方案中,所述方法包括以下步骤:
(i)非侵入性检测第一动物中的荧光以提供对照;
(ii)向第二转基因动物给予测试物,接着非侵入性检测所述动物中的荧光;以及
(iii)将在(i)中检测的荧光与在(ii)中检测的荧光进行比较。
所述第一动物可以为转基因的或非转基因的。
在一个优选的实施方案中,提供了本发明的方法用于确定测试物的神经毒性。
在另一优选的实施方案中,提供了本发明的方法用于测试测试物促进治疗或调节神经学损伤。
相应地,所述方法包括以下步骤:
(i)在所述转基因动物中诱导神经学损伤;
(ii)在给予测试物之前非侵入性检测来自(i)的所述神经学损伤动物中的荧光,以提供对照;

(ii)在第二动物中诱导神经学损伤,并且向所述动物给予测试物,接着非侵入性检测所述动物中的荧光;以及
(iii)将在(i)中检测的荧光与在(ii)中检测的荧光进行比较。
所述第一动物可以是转基因的或者非转基因的。
所述神经学损伤可以是对神经系统的任何损伤或伤害,无论是如何产生的。优选地,所述损伤是针对中枢神经系统,更优选针对脑。
所述神经学损伤可以是反应性神经胶质增生。
所述神经学损伤可以是通过向动物给予选择的化学品而化学诱导的,所述化学品例如神经毒素,如MPTP、2′-CH3-MPTP、6-羟基多巴胺(6-OHDA)、红藻氨酸(Kainic acid,KA)、三甲基锡(Trimethytin),农药(pesticides),如毒死蜱(CPF),杀真菌剂,如亚乙基双二硫代氨基甲酸锰(代森锰或MB),杀虫剂(insecticides),如鱼藤酮,除草剂,如百草枯(N,N′,-二甲基-4-4′-联吡啶鎓),有机氯,如聚氯联苯(PCBs),食品添加剂,如谷氨酸单钠盐(MSG)、工业化学品,如3,3′-亚氨基二丙腈(IDPN)、甲苯(甲基苯)。本领域技术人员能够选择合适量的化学品用于向动物给药并且能够设计适合的给药方案,以便诱导神经学损伤。举例而言,可以给予单一或多(如2、3、4、5或更多)剂量的所选化学品。多剂量可以以预定的时间间隔给药,例如1、2、3、4、5、6或更多个小时。还是通过举例,每一剂量可以选自2、4、6、7、8、12或14mg/kg。
或者,可以通过对动物的头和/或颈和/或背部施以物理力来诱导神经学损伤,例如挤压伤。可以通过减少对动物神经系统的供氧导致中风和/或大脑缺血来诱导神经学损伤。其它的诱导神经学损伤的手段可以包括环境刺激、机械力、暴露于病毒粒子或遗传因子。
在优选的实施方案中,所述神经学损伤会提供所选疾病的动物模型。例如,所述疾病可以选自帕金森病、阿耳茨海默病或亨廷顿病。帕金森病的动物模型可以通过给予MPTP(1-甲基-4-苯基-1,2,3,6-四氢吡啶)或2′-CH3-MPTP来化学诱导。
在其它的实施方案中,提供了本发明的方法用于测试测试物促进神经系统疾病治疗的能力。
相应地,在一个优选的实施方案中,所述方法包括以下步骤:
(i)提供所述转基因动物,其中所述动物也是神经系统疾病的动物模型;
(ii)在给予测试物之前非侵入性检测来自(i)的所述动物中的荧光;

优选地,所述神经系统疾病为神经系统肿瘤,其可以选自神经胶质瘤、成神经管细胞瘤、脑膜瘤、神经纤维瘤、室管膜瘤、神经鞘瘤、神经纤维肉瘤、星形细胞瘤、少突神经胶质瘤。神经系统肿瘤的动物模型可以通过本领域技术人员熟知的肿瘤异种移植技术产生。
适合地,所进行的治疗可以包括应用放射治疗,例如X射线或γ射线,或者化疗药物。
当本发明的方法包括使用具有神经学损伤或神经系统疾病的动物时,本发明可以任选地省略在所述动物中诱导所述疾病和/或损伤的步骤,这种情况下,所述方法不涉及这类动物的产生,而涉及其用途。
所述动物(转基因的和非转基因的)优选为非人哺乳动物,例如兔、豚鼠、大鼠、小鼠或其它啮齿类动物(包括啮齿目中的任何动物)、猫、狗、猪、绵羊、山羊、牛、马、非人灵长类动物;或任何其它非人脊椎动物。最优选地,所述动物(转基因的和非转基因的)为小鼠。在任何给定的方法中,相同类型的动物如小鼠优选用作对照和测试动物。这些动物的区别可以在于,一个可以为转基因的,另一个为非转基因的,但测试动物总是转基因的。对照和测试动物都可以是转基因的。
启动子核酸来自通常在动物神经系统中表达的蛋白。尽管启动子的核酸序列可以相对于野生型被加以改变,例如通过核苷酸的置换、添加、删除(截短)、插入或替换,但是启动子核酸的特征在于当可操作地连接编码蛋白的核酸时能够调节所述蛋白的神经系统表达。最优选地,与野生型启动子相比,所述启动子保留了调节神经系统组织中转录(以及因而蛋白表达)的相应能力。
在本说明书中,术语“可操作地连接”可以包括这样的情况,其中所选择的核苷酸序列(如蛋白编码序列)和调节核苷酸序列(如启动子)如此共价连接,以使得将所选的核苷酸序列的表达置于调节序列的影响或控制下。因而,当调节序列能够影响形成所选核苷酸序列的一部分或全部的蛋白编码序列的转录时,所述调节序列可操作地连接所选的核苷酸序列。如果需要,得到的转录本可以被翻译成需要的蛋白或多肽。
最优选地,所述启动子为GFAP启动子。GFAP启动子序列为本领域技术人员所熟知。Brenner和Messing,Methods:A Companion to Methods inEnzymology 10,351-364(1996)中描述了GFAP启动子的实例和它们在构建GFAP转基因中的用途,其中蛋白的表达被置于GFAP转录控制区域的控制下,通过引用将该文献并入本文。
所述荧光蛋白可以是任何荧光蛋白。很多适合的荧光蛋白为本领域技术人员所知,如绿色、蓝色、青蓝色、黄色、橘色和红色荧光蛋白(例如,参见http://www.microscopyu.com/articles/livecellimaging/fpintro. html)。最优选地,所述荧光蛋白为绿色荧光蛋白(GFP),如hGFP-S65T绿色荧光蛋白基因、EGFP-1荧光蛋白基因、或EYFP-1绿色荧光蛋白基因,或具有哺乳动物相容性或人源化的序列(例如,密码子改变,其使得构建体与哺乳动物核糖体翻译更具有相容性)并具有增加其光发射系数的突变的任何其变体。
在本说明书中,“转基因动物”包括其中编码荧光蛋白的核酸通过使用重组核酸技术被引入生物体基因组的动物,所述编码荧光蛋白的核酸可操作地连接来自通常在所述动物神经系统中表达的蛋白的启动子核酸。产生非人转基因哺乳动物的技术为本领域技术人员所熟知。在本发明的方法中,所述动物优选为新生的,即从出生起不超过四周龄。更优选地,所述新生动物从出生起不超过三周龄,更优选从出生起不超过14天龄。任选地,所述新生动物可以为从出生起1至14天龄,优选从出生起1至7天龄,或者从出生起7至14天龄。新生动物可以为从出生起1、2、3、4、5、6、7、8、9、10、11,12、13、14、15、16、17、18、19、20、21、22、23、24、25、26、27或28天龄。
通过使用新生动物,可以监测神经系统的发育,尤其是测试物对发育中的神经系统的影响。
在本发明的方法中,通过非侵入性成像检测荧光。所述成像技术为非侵入性的,这是因为所述动物从所述动物的外部成像,在荧光的检测中不涉及任何手术介入。因此所述动物在成像期间保持完整和完好。尽管为了检测荧光所述动物可任选地被麻醉,但是所述动物优选在荧光检测中为可活动的。用于非侵入性荧光成像的适合仪器和技术为本领域技术人员已知,例如成像系统100系列(Xenogen Corp.,Alameda,USA)。
可以在给予测试物之前或之后的任何方便时间点进行荧光检测,以得到对照或测试值。通常,优选进行一系列的给药后检测,以便监测荧光随时间的变化。任选地,可以例如从给予了安慰剂而不是测试物的动物在每一对应时间点进行对照读数。例如,可以在给予测试物之前立即或不久例如最多至1小时进行检测,接着进行定期检测,例如每隔1小时。或者,给予测试物之后的检测可以每天一次、每天两次、每天三次或每天四次,且持续研究者所希望的天数,例如1、2、3、4、5或更多天。
优选对来自动物的目的区域(ROI)进行荧光检测。所述ROI优选为研究者感兴趣的区域。优选的ROI为中枢神经系统或其部分,最优选脑或其部分。任选地,ROI可以不包括视神经、视网膜、坐骨神经、角膜或哺乳动物眼的任何部分中的一种或多种。对应的ROI优选用于比较在对照和测试动物中的荧光检测的目的。
检测对照动物中的荧光提供了动物自体荧光的基础读数。可以进行基础读数与测试读数之间的直接比较。但是,在一个尤其优选的实施方案中,可以计算相对荧光(RF)值,并用于比较目的。RF值优选作为在给予测试物后的时间点在转基因小鼠中从ROI检测的总荧光与在对照小鼠中检测的组织自体荧光(任选地,在相同时间点,例如在给予安慰剂的情况下)的比值来计算,所述对照小鼠可以为转基因的或非转基因的。
在优选的实施方案中,所检测的荧光的增加表明神经学损伤的增加,神经毒性或神经系统疾病的进展。在其它实施方案中,荧光的减少可以表明神经学损伤的增加。
荧光的检测可以为定量和/或定性的。
在本说明书中,对动物神经系统的提及可以包括中枢神经系统或外周神经系统。更优选地,其为中枢神经系统,更优选脑和/或脊髓。
测试物可以为任何物质、材料、组合物、化合物、药物、药剂、蛋白、肽、抗体、核酸、小分子、工业化学品、杀虫剂、食品添加剂或防腐剂。测试物可以为手术植入测试动物的材料或组织,例如人造生物材料。
本发明的方法可以用于监测测试物对动物的影响,并且可以用于:
-设计疾病的诊断;
-设计疾病的预后;
-监测和/或定量脑内化学诱导的神经胶质增生和/或损伤的进展;

优选实施方案的详细说明
在本发明优选的实施方案中,所述转基因动物的基因组具有可操作地连接GFAP启动子的编码绿色荧光蛋白的核酸。这种类型的转基因小鼠在Zhou et al 1997 Developmental Biology 187,36-42,和WO 00/02997和US 6,501,003中有描述,通过引用将它们都整体并入本文。所述GFAP启动子驱动该荧光蛋白在诸如星形细胞、Schwann细胞和Mueller细胞的神经胶质细胞中的表达。
转基因小鼠被改造为表达编码人源化绿色荧光蛋白的转基因,所述转基因可操作地连接胶质纤维酸性蛋白启动子。在所述转基因小鼠中,在应答神经学损伤时,绿色荧光蛋白特别在诸如星形细胞、Schwann细胞和Mueller细胞的神经胶质细胞中上调。
通过将遗传构建体插入哺乳动物受精卵的前核并使稳定的基因组整合天然发生来构建转基因小鼠。然后将受精卵转移至接纳子宫,并使其发育成熟。
所述遗传构建体包括提供神经胶质细胞特异表达的全长胶质纤维酸性蛋白启动子。所述启动子位于编码绿色荧光蛋白的突变基因的5′,并与其可操作地连接,位于所述绿色荧光蛋白编码基因3′的DNA片段含有用于正确RNA剪接和多腺苷酸化的信号序列。
进行动物的非侵入性成像以获得针对自体荧光的对照值和在给予测试物后的测试值-例如参见实施例1的材料和方法。
荧光的增加表明神经毒性或神经学损伤的增加。

发明详细说明
以下将通过举例来说明本发明人认为的用于实施本发明的最佳模式的具体细节。对本领域技术人员来说显而易见的是,本发明的实施不限于这些具体细节。
实施例1
为了开发用于上述筛选目的的非侵入性荧光成像系统,我们将2′-CH3-MPTP用作转基因GFAP-GFP小鼠新生脑中的模型神经毒剂,这种小鼠先前由Zhuo et al.,1997建立。这里,我们证实,当与适合的体内光学成像系统联合时,转基因GFAP-GFP小鼠模型可可靠地检测转基因小鼠中的GFAP-GFP标记,并且可对其应答MPTP诱导时以剂量和时间依赖方式的上调定量。这种具有适度处理量的非侵入性光学荧光系统会在研究帕金森病和其它神经变性疾病、发育神经毒理学以及在临床前化合物筛选方面有广泛应用。
材料和方法
转基因GFAP-GFP小鼠
按先前所描述的(Zhuo et al.,1997)进行转基因GFAP-GFP小鼠的建立和基因分型。在本项研究中使用4天龄、重2.0g-2.5g的FVB/N背景的新生小鼠。动物饲养由新加坡国立大学实验动物维持单位动物房(animal holding unit facility)提供。涵盖本研究的实验方案获得实验动物管理及使用委员会(Institutional Animal Care and Use Committee)的许可。神经毒剂和给药
从(Sigma-Aldrich,M-103)购得测试化合物2′-CH3-MPTP-一种比常规MPTP(Abdel-Wahab,2005)更有效的类似物。新生小鼠在0小时接受2′-CH3-MPTP(12mg/kg,盐水中)单次皮下(sc)注射。将盐水用作对照组的载体。随后在处理后2小时、4小时、6小时和8小时进行神经成像。
体内神经成像
采用IVIS成像系统100系列(Xenogen Corp.,Alameda,CA,U.S.A.)进行非侵入性成像。包括转基因和非转基因新生小鼠的2′-CH3-MPTP处理对并排放置并通过使用带子物理限制小鼠下背来使其保持在适当位置,不麻醉。这是根据一项最近的研究,其报道广泛用于儿科麻醉的氯胺酮(ketamine)通过增加神经元凋亡的速率而显示在新生大鼠中的发育神经毒性(Scallet et al.,2004)。然后,该对新生小鼠用配备有IVIS系统的GFP滤境组荧光成像。图像采集时间为10秒。相同的操作也应用于注射有盐水的对照对。2′-CH3-MPTP对小鼠的神经毒性作用结果表示为相对荧光(RF),即从转基因(Tg)小鼠中目的区域(ROI)检测的总GFP荧光信号与从非转基因(nTg)小鼠中ROI检测的组织自体荧光信号之间的比例(TgROI/nTgROI)。两种小鼠的ROI区域完全相同并且在定量辐射的研究中所用的所有ROI始终相同。为了使成像系统引起的任何波动最小化,对每对处理的小鼠和对照小鼠获取三个图像,所得到的均值用作该特定对的定量读数。对照组(n=5对小鼠)和处理组(n=5对小鼠)的值表示为均值±标准误,采用斯氏双尾配对t检验来评估统计学显著性。
免疫组织化学
为了确定光子是否是从CNS中星形细胞发出的,收集脑并在4℃使用于0.1M磷酸盐缓冲盐水(PBS,pH 7.4)中的4%多聚甲醛固定4小时。然后将脑在PBS中洗涤三次,每次孵育时间是15min,接着于4℃在30%蔗糖中浸泡过夜。在封固和采用恒冷切片机(Leica Microsystems,NusslochGmbH;CM-3050S)将它们切片之前,将组织浸入液氮后,所有的脑被包埋在冷冻介质中。根据Atlas of Mouse brain(Franklin and Paxinos,2001),将黑质(前囱-3.16mm,耳间0.64mm)的20μm厚度的冠状冷冻切片用于免疫组织化学。对照组和处理组各含有4个动物(n=3)。
对于酪氨酸羟化酶(TH)和GFAP免疫染色,采用了兔抗TH多克隆抗体(Chemicon International,Temecula,CA,USA;Ab-152)和兔抗GFAP多克隆抗体(DakoCytomation,Denmark;Z-0334)。将冷冻切片在0.15M 1xPBS中洗涤5min,接着于4℃在含有0.1%(v/v)Triton X-100和10%未免疫山羊血清的PBS封闭液中孵育4小时。然后在0.15M 1x PBS中洗涤冷冻切片三次,每次15min。接着,于4℃将脑切片与抗TH抗体(1∶200)和抗GFAP抗体(1∶200)一起在含有0.01%(v/v)Triton X-100和1%未免疫山羊血清的1x PBS中孵育过夜。在1x PBS中洗涤三次(每次15min)后,在室温下,将切片与1∶100稀释度的德克萨斯红偶联的山羊抗兔多克隆第二IgG抗体一起在含有0.01%(v/v)Triton X-100和1%未免疫山羊血清的PBS中孵育2小时。再次在1x PBS中洗涤三次(每次15min)后,用盖玻片使切片位于10μl的荧光介质中,并在共聚焦激光扫描显微镜下观察(Olympus Optical Co.Ltd.Tokyo,Japan;IX-71)。
结果
2′-CH3-MPTP对GFP荧光水平的影响
从小鼠脑中ROI检测固有GFP荧光,并采用Living软件(Xenogen Corp.)将来自神经区域的辐射以光子/秒/cm2/球面度(sr)定量,如图1A所示。为了确定光子是否是从CNS中星形细胞发出的,在最后的体内成像时间点后取出脑并离体成像(图1B)。采用这个平台,使对照和经处理的转基因小鼠的GFP荧光间的定性和定量比较成为可能。
如图2中的体内成像显示的,在处理后6小时,接受了单次剂量2′-CH3-MPTP(12mg/kg sc)的转基因新生小鼠相对于其非转基因对应小鼠以及转基因对照小鼠显示了ROI中GFP荧光(光子/秒/cm2/球面度)的最显著增加。图3中显示的GFP荧光定量进一步加强了在之前图中得到的观察结果,其中在处理后4小时、6小时和8小时时记录了2′-CH3-MPTP处理组和对照组之间相对荧光(RF)的显著差异,处理组在处理后6小时时显示了相对于对照组的22%均值的最显著增加(P<0.01)。类似地,在2′-CH3-MPTP组中,与0小时相比,在处理后4小时、6小时和8小时,出现了RF的显著增加,其中在处理后6小时时出现了17%均值的最显著增加(P<0.0001)。在对照组中,不同时间点之间未出现RF的显著差异(图3)。
GFAP免疫组织化学
海马中GFAP免疫染色的代表性图像显示在图4中。在一次2′-CH3-MPTP(12mg/kg sc)处理后6小时时,在脑室和海马中星形细胞的GFP表达显著增加。在非转基因小鼠中不存在GFP表达(图4A)。为了证实GFP表达细胞确实是星形细胞,我们在相同切片上进行了GFAP免疫染色,发现GFAP免疫阳性的海马沟处血管周围星形细胞与内源GFP标记的星形细胞之间的共定位主要发生在细胞突而不是细胞体中(在图4D和4E中双标记细胞突的实例用箭头指出)。
TH免疫组织化学染色
黑质致密区(SNC)中TH免疫染色的代表性图像显示在图5中。带有TH抗体的多巴胺能神经元(约20μm)在尺寸上比非IHC染色的内源GFP标记星形细胞(约10μm)大,在经2′-CH3-MPTP处理的新生小鼠SNC中可容易地检测到。多巴胺能细胞的细胞体或细胞纤维与SNC中明显的免疫阳性细胞突一起被密集染色。在2′-CH3-MPTP(12mg/kg sc)处理后6小时未观察到TH免疫阳性纤维和细胞体中的可见减少。但是,与0小时相比,处理后6小时,可明显看到星形细胞中GFAP-GFP转基因表达的上调。如图5A中所显示的,非转基因小鼠的SNC中多巴胺能神经元对TH是免疫阳性染色,但是神经胶质细胞不产生转基因的表达。TH免疫阳性多巴胺能神经元和非IHC染色的内源GFP标记星形细胞之间没有共定位。
实施例2-转基因GFAP-GFP成体小鼠的成像
皮瓣体内成像
在麻醉的转基因GFAP-GFP成体小鼠上制做皮瓣(从身体撕下的一块皮肤,该皮肤的一侧仍保持连接)。
采用IVIS成像系统100系列(Xenogen Corp.,Alameda,CA,U.S.A.),在脑和肝的皮瓣区成功进行了体内成像。将低荧光的黑色纸用于阻挡来自小鼠其它部分的组织自体荧光,仅暴露脑(图6A)和肝(图6B)的皮瓣区。离体成像
如在图2A-2D的图像中所显示的,取出转基因(Tg)和非转基因(nTg)成体小鼠的脑、肝、肾和坐骨神经并采用IVIS成像系统离体成像。从对应于发射的光子数的数值的假彩色图像,可观察到Tg和nTg小鼠间的显著差异。目前,正在进行成体小鼠替代性的成像方式的可行性研究。
实施例3-在新生转基因小鼠中研究帕金森病和神经毒性的非侵入性脑
成像方法
实验操作
动物
本项研究在4天龄新生小鼠上进行(实验时3-5g体重)。动物自由进食和饮水,尽量使所使用的动物数量和它们的痛苦减至最小。
毒素注射
所用的神经毒素为2′-CH3-MPTP。每隔2小时进行8mg/kg MPTP的皮下给药,进行4次,在处理后24小时、48小时和72小时获得体内神经图像。
BCATM蛋白测定和Western印迹
对于进行BCATM蛋白测定和Western印迹,将小鼠在处理后24小时处死。取出脑,然后在含有磷酸盐缓冲盐水(PBS)和蛋白酶抑制剂的样品缓冲液中裂解。采用牛血清白蛋白(BSA)作为标准品,通过二辛可宁酸(BCA)法测定总蛋白。然后采用NuPAGE Novex Bis-Tris凝胶(4%-12%)对样品进行NuPAGE Bis-Tris电泳。每个样品含有15μg蛋白。电泳后,将蛋白转移至硝酸纤维素膜上。将膜在4℃下用于PBS中的5%脱脂奶粉和0.1%Tween 20(PBS-T)封闭过夜。然后将膜在室温下于1∶1000稀释度的多克隆兔抗GFAP抗体(Dako)/多克隆兔抗GFP抗体(abcam)孵育1小时,然后洗涤并与PBS-T一起孵育3×15min。在室温下与1∶2000稀释度的山羊抗兔IgG-HRP第二抗体(Santa Cruz)一起孵育1小时后,重复5分钟的孵育和洗涤过程3次。通过使用化学发光检测试剂(ECL Plus,Amersham)和放射自显影膜观察第二抗体。进行Western印迹后,将硝酸纤维素膜置于IVIS仪器下成像。当采用ECL Plus检测试剂时,组合的HRP和过氧化物催化的Lumigen PS-3 Acridan底物的氧化每分钟产生数千的吖啶鎓酯中间体。这些中间体在轻度碱性条件下与过氧化物反应,产生持续的高密度化学发光,最大发射在430nm。这种化学发光信号可采用IVIS成像系统采集。获得了具有描绘发光密度的颜色的膜图像。
实时RT-PCR
对于实时RT-PCR,在处理后24小时处死小鼠。取出脑,然后在缓冲液RA1和β-巯基乙醇中裂解。采用RNA II试剂盒分离来自脑的总RNA。用随机六聚物进行总RNA逆转录,对cDNA进行实时RT-PCR以定量基因表达。
实验结果
MPTP(4×8mg/kg)
体内神经图像
如图8所示,检测来自小鼠脑中目的区域(ROI)的固有GFP荧光,并采用Living软件(Xenogen Corp.)对来自神经区域的辐射以光子/秒/cm2/球面度(sr)定量。如在体内图像中显示的,接受了4剂MPTP(8mg/kgsc)的转基因新生小鼠相对于对照小鼠在处理后24小时显示了ROI中GFP荧光(光子/秒/cm2/球面度)的最显著增加。对显示在图9中的GFP荧光的定量显示,在处理后24小时记录到MPTP处理组和对照组之间的相对荧光(RF)的显著差异,与对照组相比有15%均值的统计学显著性增加(p<0.01)。对于接受了单剂量MPTP(12mg/kg sc)的转基因新生小鼠以及对照组中,不同时间点之间未出现RF显著差异(图9)。
Western印迹结果
从显示在图10A中的western印迹结果可见,在约51kDa处检测到带,其与采用Dako抗兔GFAP抗体报告的小鼠GFAP分子大小相对应。在约27kDa处也检测到带,其与采用Abcam抗兔GFP抗体报告的小鼠GFP分子大小一致。从带的浓度可看出在转基因小鼠中GFAP和GFP蛋白的量从0至24小时是增加的(比较道a和b)。在非转基因小鼠中,GFAP蛋白的量从0至24小时也是增加的(比较GFAP western印迹中道c和d)。由于没有转基因,在非转基因小鼠中没有检测到GFP蛋白(GFP western印迹中道c和d)。当采用IVIS对western印迹膜成像时,彩色描绘的蛋白存在量给出了相同结果(参考图10B)。这支持了这样的事实,即在给予MPTP后GFAP被上调,其又导致GFP的上调。
实时RT-PCR结果
mRNA的研究与BCA和western印迹中的蛋白研究结果是一致的,就更长的研究周期而言,与体内神经成像也是一致的。如图11A所示,发现MPTP处理后24小时GFAP基因表达的增强对于转基因和非转基因小鼠都具有统计学显著性,相对于未经处理的小鼠分别为约91%(p<0.01)和88%(p<0.01)。但是,发现MPTP处理后24小时GFP的基因表达与未处理小鼠相比没有统计学显著的增强(图11B)。这解释了MPTP处理后48小时和72小时神经图像的相对荧光的减弱(参见图8和9)。

实施例4-在新生转基因小鼠中研究帕金森病和神经毒性的非侵入性脑
成像方法
实验操作
动物
本研究在4天龄新生小鼠上进行(实验时3-5g体重)。动物自由进食和饮水,尽量使所使用的动物数量和它们的痛苦减至最小。
毒素注射

BCATM蛋白测定和Western印迹

实时RT-PCR结果
mRNA的研究与BCA和western印迹中的蛋白研究结果是一致的。当GFAP和GFP蛋白上调在6小时达到峰值时,发现GFAP和GFP mRNA的上调在早至MPTP处理后2小时就出现了。这个时间差异说明了mRNA被翻译成蛋白所需的时间。如图15所显示的,发现在MPTP处理后2小时GFAP和GFP基因表达的增加均具有统计学显著性,分别为约47%(p<0.001)和20%(p<0.01)。
KA(1×2mg/kg)
体内神经成像
如图16所示,检测来自小鼠脑中ROI的固有GFP荧光,并采用Living软件(Xenogen Corp.)对来自神经区域的辐射以光子/秒/cm2/球面度(sr)定量。如在体内图像中显示的,接受了单次剂量KA(2mg/kg sc)的转基因新生小鼠相对于其非转基因对应小鼠以及转基因对照小鼠在处理后6小时显示了ROI中GFP荧光(光子/秒/cm2/球面度)的最显著增加。对显示在图17中的GFP荧光的定量显示,在处理后4小时和6小时记录到KA处理组和对照组之间的相对荧光(RF)的显著差异,与对照组相比,在处理后6小时处理组有25%均值的最显著增加(p<0.01)。类似地,KA组中与0小时相比时,处理后6小时出现了RF的显著增加,在处理后6小时出现最显著的22%均值增加(P<0.05)。对照组内,不同时间点之间未出现RF的显著差异(图17)。
Western印迹结果

mRNA的研究与BCA和western印迹中的蛋白研究结果是一致的。当GFAP和GFP蛋白的上调在6小时达到峰值时,发现GFAP和GFPmRNA的上调在早至KA处理后2小时就出现了。这个时间差异说明了mRNA被翻译成蛋白所需的时间。如图19所显示的,发现在KA处理后2小时GFAP和GFP基因表达的增加具有统计学显著性,分别为约21%(p<0.01)和22%(p<0.05)。
GFAP免疫组织化学
海马CA1、CA2和CA3亚区域中GFAP免疫染色的代表性图像显示在图20A-D中。在一次KA(2mg/kg sc)处理后6小时,海马各个区域星形细胞中GFP表达明显增加。为了证实GFP表达细胞确实是星形细胞,我们对相同切片进行了GFAP免疫染色,发现海马CA1区域GFAP免疫阳性细胞和内源GFP标记星形细胞之间的共定位主要出现在细胞突而不是细胞体中(如图20B中,双标记的细胞突的实例以箭头指出)。
实施例讨论
在转基因小鼠模型中,胶质纤维酸性蛋白(GFAP)启动子驱动绿色荧光蛋白(GFP)的表达。GFAP是主要在中枢神经系统(CNS)星形细胞中表达的中间纤维蛋白。这些神经胶质细胞的变化可用于监测神经元活性。对CNS中神经元的损伤是GFAP的有效诱导者。但是,通过体外测定来监测GFAP需要处死动物。我们已经证实GFAP-GFP表达的非侵入性体内神经成像使得能够长期监测GFAP,并且减少了该过程中所处死动物的数量。
2′-CH3-MPTP是引起黑质致密区中多巴胺能神经元变性的神经毒素。卡因酸是引起兴奋性脑中毒损伤的神经毒素。已知这两种神经毒素均使GFAP表达升高。这可被开发成非侵入性体内成像模型,用于诊断和筛选治疗,即候选药物和疗法;以及用于研究发育神经毒性。向新生小鼠注射这些神经毒素,并采用IVIS成像系统研究随后由于神经毒素的作用新生脑中GFAP-GFP转基因的荧光增加。为了支持非侵入性体内神经成像模型在研究神经发育方面是可靠的这个结论,还进行了体外测定。进行了BCATM蛋白测定(GFP)和western印迹以定量蛋白表达,而实时RT-PCR用于定量基因表达。这些测定可用于跟踪在新生小鼠中由于给予神经毒素而发生的变化,其结果可与体内神经成像的结果进行比较。
在过去的半个世纪,细胞生物学和分子生物学分析是针对在培养皿中生长的细胞并通过细胞外分析包括基因和蛋白在内的细胞组分而进行的。使用光学探针来体内跟踪和报告分子、蛋白和细胞的功能信息是新的和快速发展的技术。这项技术有广泛应用,包括研究传染性疾病、肿瘤学、药物动力学、药效学、毒理学以及转基因动物中生物发光或荧光报告子的基因表达(Contag et al.,2000,Rehemtulla et al.,2000,Yang et al.,2000,Zhang et al.,2001,Bhaumik and Gambhir,2002,Bouvet et al.,2002,Ntziachristos et al.,2002)。如Hoffman(Hoffman,2004)所阐述的,GFP作为报告基因的来临使细胞和分子生物学发生范式转变成为可能。引入快速和可进入临床前研究的成像技术将使每次实验设计产生更多和更高质量的实验数据,这通过增加可收集定量数据的次数来实现。通过在多个时间点对整体完好的活体动物成像,研究者可在完好器官背景下分析生物分子过程。额外的优点是,采用这些方法,较少的动物就可产生具有更高统计学显著性和更低压力(lower stress)的数据。非侵入性方法可用于产生更具有预测性的动物模型,它们共有纵向研究设计、内部实验性对照、分子信息和定量数据的特征,这些方法有益于科学调查和人道的动物使用。此外,成像可通过指导用于组织学或生化分析的合适的终点组织取样而进一步改进这些方法。
本研究所面对的一个问题是自体荧光源。通常,由动物组织中包括弹性蛋白、胶原、色氨酸、烟酰胺腺嘌呤二核苷酸(NADH)、卟啉和黄素在内的内源生色团引起的组织自体荧光比仪器的自体荧光高得多,尤其是在可见光范围内(Troy et al.,2004)。对于体内成像,组织自体荧光是主要问题,这是由于其影响了检测目的荧光探针的能力。如在这项研究中所看到的,来自新生小鼠的自体荧光随着小鼠从出生发育至成体而增加。于是,本发明人设计了安全防护系统来监测信噪(s/n)比,其通过使用来自非转基因小鼠的自体荧光将来自转基因小鼠的总GFP荧光标准化而产生相对荧光(RF)来实现。如果RF突破了从转基因小鼠检测的GFP信号的阈值以下1.3或30%,则该特定数据可被认为是具有过低的s/n比,因而过于不可靠而不能用于产生精确的定量结果。
采用2′-CH3-MPTP作为模型化合物,通过非侵入性体内神经成像模型获得的数据与组织学测定的存在上调的星形细胞一致。有大量文献报道,MPTP在体内可特异地诱导中脑多巴胺神经元的神经变性死亡,导致动物的帕金森病或人类帕金森病(Damier et al.,1999,German et al.,1999,Olanow and Tatton,1999)。之前的证据提示了星形细胞在MPTP诱导的帕金森病病理过程中的可能作用。例如,全身注射了MPTP的成体小鼠显示了星形细胞中显著的GFAP升高,同时脑纹状体区内多巴胺能神经元中多巴胺减少(Reinhard et al.,1988,Chen et al.,2002,Dervan et al.,2004,Kurosaki et al.,2004)。值得注意的是,在相同遗传背景的较老小鼠中观察到脑更易受到MPTP伤害(Ali et al.,1993)。与成体小鼠脑中MPTP作用的研究相比,只有有限量的关于新生脑中星形细胞如何对MPTP作出反应的文献,推测是由于缺乏允许在脑发育过程中反复非侵入性测量GFAP转录活性的合适方法。但是,少数报道确实证实对新生小鼠全身MPTP注射导致在成年期的永久脑损伤,如所测量的(Ali et al.,1993,Fredrikssonet al.,1993,Schwartz and Nishiyama,1994)。尽管如此,这些早期的报道产生了这样的观点,即发育中的脑也易受到MPTP伤害,值得仔细研究。因此,本文描述的模型提供了在活体动物中研究神经元变性的新的可能性。在本发明中,现在可能监测各种药物的作用,以防止由于活体动物中中风和挤压伤导致的神经损伤。此外,由于GFAP上调出现在阿尔茨海默病(Vanzani et al.,2005)和亨廷顿病(Ishiguro et al.,2001)的各个模型中,可使用这种GFAP-GFP转基因新生小鼠来研究GFAP上调,并且非侵入性监测各种药物治疗的神经保护作用。除了神经变性,这些小鼠还可以用于研究脑肿瘤的治疗。Kalamarides等人最近的文章(Kalamarides etal.,2002)描述了小鼠脑膜瘤模型,其中Nf2基因在蛛网膜细胞中是失活的。当GFAP侵入脑组织并生长时,其表达在围绕该肿瘤的星形细胞中被上调(Pekny and Nilsson,2005)。本发明的其它优点是没有麻醉剂被引入到新生小动物,因而避免了对小鼠任何可能的神经毒性作用。在2′-CH3-MPTP处理小鼠的SNC和腹侧被盖区中没有检测到TH样免疫反应性的明显变化,提示黑质纹状体多巴胺神经元的变性在2′-CH3-MPTP注射后6小时仍不明显。这与Araki等人的观察(在MPTP处理后5天出现多巴胺能神经元的减少)和Kurosaki等人的发现(仅在MPTP处理后1天纹状体和黑质中TH免疫阳性纤维和细胞体减少)很好地相符。集中于2′-CH3-MPTP的全身给药以及处理后24、48和72小时期间对GFAP和TH免疫染色的影响的其它研究将扩展我们对该领域的认识。
除了MPTP,还报道了导致成体小鼠脑各个区域中神经元损伤的多种其它化学品,如通过GFAP升高所证实的。这些化学品包括一系列结构和功能上不同的化合物,从工业上有毒的化合物如三甲基锡(O′Callaghan,1988)和甲基汞(O′Callaghan,1988,Barone Jr.et al.,1998,Garcia et al.,2002)、农业杀虫剂(Garcia et al.,2002)到食品添加剂(Ostergren et al.,2005)。与成体中经发育的脑相比,具有较不完整血脑屏障的发育中脑更易于受到已知神经毒素的伤害,更关键的是受多种化合物的伤害,这些化合物可进入人类食物链和环境而未经首先适当地检测其神经毒性。对于测试化合物在发育中的脑中的潜在神经毒性而言,其重要性无论如何强调也不过分(Olney,2002),尤其是在处理沉默神经毒性(Costa et al.,2004))的问题时。(Tilson,2000)评述过的美国环保局的发育神经毒性测试指南(Developmental Neurotoxicity Testing Guideline,DNTG)进一步强调了这点。最近,欧委会采纳了用新的欧盟管理框架来通过严格制度测试所有的化学试剂的建议,据估计新措施耗资高至70亿欧元并需要至少10年来实施(European,2003)。但是,对于使用常规的GFAP技术试图评估大量化合物在体内对发育中CNS的神经毒性风险来说,是个难以克服的挑战,尤其是考虑到该测定方法必须以适当的处理量以及工业(Kaufmann,2003)和管理机构(Hass,2003)都可接受的成本提供合理的科学依据时。由于在脑的所有主要区域中显示独特的神经胶质细胞表达模式(Su et al.,2004)的经证实的GFAP-GFP转基因小鼠模型(Zhuo et al.,1997)与合适的体内光学生物成像系统的整合,有可能非侵入性研究神经变性疾病、发育神经毒理学,实时筛选化学化合物,以及对诸如创伤后伤口愈合、中风和肿瘤生长的其它CNS病理学的潜在治疗介入。

Claims (25)
1.涉及检测动物中目的荧光蛋白表达的方法,其中所述动物为转基因动物,在其基因组中具有编码所述荧光蛋白的核酸,所述编码所述荧光蛋白的核酸可操作地连接来自通常在所述动物神经系统中表达的蛋白的启动子核酸,所述方法包括非侵入性检测在所述动物中表达的所述蛋白的荧光的步骤。
2.如权利要求1所述的方法,其中所述启动子为GFAP启动子。
3.如权利要求1或2所述的方法,其中所述荧光蛋白为绿色荧光蛋白(GFP)。
4.如权利要求1至3中任一项所述的方法,其中所述方法用于研究向所述动物给予测试物的影响,所述方法包括在非侵入性检测荧光的步骤之前向所述动物给予所述测试物的步骤。
5.如权利要求4所述的方法,其中所述方法包括以下步骤:将来自未给予所述测试物的对照动物的荧光与向所述转基因动物给予所述测试物后来自所述转基因动物的荧光进行比较。
6.如权利要求4或5所述的方法,其中所述方法包括以下步骤:
(i)在给予所述测试物之前非侵入性检测所述转基因动物中的荧光,以提供对照;
(ii)给予所述测试物之后非侵入性检测所述相同动物中的荧光;以及
(iii)将在(i)中检测的荧光与在(ii)中检测的荧光进行比较。
7.如权利要求4或5所述的方法,其中所述方法包括以下步骤:
(i)非侵入性检测第一动物中的荧光,以提供对照;
(ii)向第二转基因动物给予测试物,接着非侵入性检测所述动物中的荧光;以及
(iii)将在(i)中检测的荧光与在(ii)中检测的荧光进行比较。
8.如权利要求4至7中任一项所述的方法,其中所述方法用于确定所述测试物的神经毒性。
9.如权利要求1至5中任一项所述的方法,其中所述方法用于测试测试物促进神经学损伤的治疗或调节神经学损伤的能力,其中所述方法包括以下步骤:
(i)在所述转基因动物中诱导神经学损伤;
(ii)在给予所述测试物之前非侵入性检测来自(i)的所述神经学损伤动物中的荧光,以提供对照;
(iii)在给予所述测试物之后非侵入性检测来自(ii)的该相同动物中的荧光;以及
(iv)将在(ii)中检测的荧光与在(iii)中检测的荧光进行比较。
10.如权利要求1至5中任一项所述的方法,其中所述方法用于测试测试物促进神经学损伤的治疗或调节神经学损伤的能力,其中所述方法包括以下步骤:
(i)在第一动物中诱导神经学损伤,且非侵入性检测所述神经学损伤动物中的荧光,以提供对照;
(ii)在第二转基因动物中诱导神经学损伤,并向所述动物给予所述测试物,接着非侵入性检测所述动物中的荧光;以及
(iii)将在(i)中检测的荧光与在(ii)中检测的荧光进行比较。
11.如权利要求9或10所述的方法,其中所述神经学损伤是通过向所述动物给予选择的化学品而诱导的。
12.如权利要求11所述的方法,其中给予所述化学品以便提供所选疾病的化学诱导动物模型。
13.如权利要求12所述的方法,其中所述疾病选自帕金森病、阿尔茨海默病或亨廷顿病。
14.如权利要求11至13中任一项所述的方法,其中所述化学品选自MPTP、2′-CH3-MPTP、6-羟基多巴胺(6-OHDA)、红藻氨酸、三甲基锡、毒死蜱(CPF)、乙烯双二硫代氨基甲酸锰(代森锰或MB)、鱼藤酮、百草枯(N,N′-二甲基-4-4′-联吡啶鎓)、聚氯联苯(PCBs)、3,3′-亚氨基二丙腈(IDPN)、甲苯(甲基苯)。
15.如权利要求9或10所述的方法,其中所述神经学损伤是通过对所述动物的头和/或颈和/或背施加物理力而诱导的。
16.如权利要求9或10所述的方法,其中所述神经学损伤是通过减少对所述动物神经系统的供氧导致脑缺血而诱导的。
17.如权利要求1至5中任一项所述的方法,其中所述方法用于测试测试物促进神经系统疾病治疗的能力,其中所述方法包括以下步骤:
(i)提供所述转基因动物,其中所述动物也是所述神经系统疾病的动物模型;
(ii)在给予所述测试物之前非侵入性检测来自(i)的所述动物中的荧光,以提供对照;
(iii)在给予所述测试物之后非侵入性检测来自(ii)的该相同动物中的荧光;以及
(iv)将在(ii)中检测的荧光与在(iii)中检测的荧光进行比较。
18.如权利要求1至5中任一项所述的方法,其中所述方法用于测试测试物促进神经系统疾病治疗的能力,其中所述方法包括以下步骤:
(i)提供第一和第二动物,其中这两动物均为所述神经系统疾病的动物模型;
(ii)在来自(i)的第一动物中,非侵入性检测所述动物中的荧光,以提供对照;
(iii)在来自(i)的第二转基因动物中,向所述动物给予所述测试物,接着非侵入性检测所述动物中的荧光;以及
(iv)将在(ii)中检测的荧光与在(iii)中检测的荧光进行比较。
19.如权利要求1至5中任一项所述的方法,其中所述方法用于测试对神经系统疾病的治疗,其中所述方法包括以下步骤:
(i)提供所述转基因动物,其中所述动物也是所述神经系统疾病的动物模型;
(ii)在进行所述治疗之前非侵入性检测来自(i)的所述动物中的荧光,以提供对照;
(iii)在进行所述治疗后非侵入性检测来自(ii)的所述相同动物中的荧光;以及
(iv)将在(ii)中检测的荧光与在(iii)中检测的荧光进行比较。
20.如权利要求4或5所述的方法,其中所述方法用于测试对神经系统疾病的治疗,其中所述方法包括以下步骤:
(i)提供第一和第二动物,其中这两动物均为所述神经系统疾病的动物模型;
(ii)在来自(i)的第一动物中,非侵入性检测所述动物中的荧光,以提供对照;
(iii)在来自(i)的第二转基因动物中,对所述动物进行治疗,接着非侵入性检测所述动物中的荧光;以及
(iv)将在(ii)中检测的荧光与在(iii)中检测的荧光进行比较。
21.如权利要求19或20所述的方法,其中所述治疗包括应用X射线或γ射线。
22.如权利要求17至21中任一项所述的方法,其中所述神经系统疾病是神经系统肿瘤。
23.如权利要求22所述的方法,其中所述肿瘤选自神经胶质瘤、成神经管细胞瘤、脑膜瘤、神经纤维瘤、室管膜瘤、神经鞘瘤、神经纤维肉瘤、星形细胞瘤、少突神经胶质瘤。
24.如前述权利要求中任一项所述的方法,其中所述动物为哺乳动物。
25.如前述权利要求中任一项所述的方法,其中所述动物为新生动物。
