Abstract

水、又は生理学的に許容される緩衝液のような緩衝液において、濃縮又は希釈のいずれかが行なわれるDIPEである。1つの溶媒の組み合わせはアルコール及びエーテルを含む。別の好ましい溶媒は、非対称エーテル又はハロゲン化エーテルの形態のいずれかで、エーテル又はエーテルの組み合わせを含む。
タノールの有用な比率は、約99.99部のDEEに対する約0.01部のブタノール〜40部のDEEに対する60部のブタノールであり、具体的には、約90部のDEEに対する約10部のブタノール〜95部のDEEに対する5部のブタノールの比率範囲で、約90部のDEEに対する約10部のブタノール〜50部のDEEに対する50部のブタノールの比率範囲で、約80部のDEEに対する約20部のブタノール〜55部のDEEに対する45部のブタノールの比率範囲で、約60部のDEEに対する約40部のブタノールの範囲である。
上述のように、癌細胞の癌細胞関連抗原を露出するために、様々な癌が本発明の方法で処理されてもよい。好ましい実施形態において、動物又はヒトから得られた癌サンプルは、脂質の除去及び癌細胞関連抗原の露出又は改変のために本発明の方法で処理される。この実施形態において、生検のような癌サンプルは、様々な外科技法を含む従来の任意の手段によって動物又はヒト患者から得ることができ、そして癌細胞を単離するためにそのサンプルを処理する。いくつかの癌細胞は、血漿、腹水、胸膜水、心嚢液及び脳脊髄液のような流体から得てもよい。癌細胞を切除し、単離させるそのような方法は、当業者に既知である。

第1の溶媒を培地と混合した後に、溶媒は処理されている培地から分離される。有機相及び水相は、当業者に既知の任意の適切な様式によって分離されてもよい。第1の溶媒が流体の中で典型的には不混和性であるので、2つの層は分離が可能であり、不要な層が除去される。不要な層は溶解された脂質を含む溶媒層であり、その同定は、当業者に既知であるように、溶媒の密度が水相よりも大きいか又は小さいかに依存する。この様式の分離の利点は、溶媒層中に溶解される脂質が除去されてもよいことである。
好ましい解乳化剤はエーテルであり、より好ましい解乳化剤はジエチルエーテルである。解乳化剤は流体に加えられてもよく、又は代替的に、流体は解乳化剤で分散してもよい。ワクチン調製物において、約1部のエマルジョンに対して約0.5〜4.0の比率のアルカン(体積:体積)は、解乳化剤として利用され、その後洗浄することによって、ワクチンの調製に使用される残った脱脂された癌細胞から、残存アルカンを除去することができる。好ましいアルカンは、ペンタン、ヘキサン、高次直鎖アルカン及び分枝鎖アルカンを含むが、これらに限定されない。
本発明の方法が、連続的又は不連続的な様式で利用されることが理解される。不連続的な操作又はバッチモードの操作において、本発明は、ヒト又は動物からこれまでに得られた癌組織サンプル又は分散細胞を利用する。サンプルは、少なくとも部分的に若しくは完全に脱脂された癌細胞、又は改変された癌細胞を含む新しいサンプルを産生するために、本発明の方法により処理される。本発明のこの様式の1つの実施形態は、動物又はヒトからこれまでに得られ、続いてDC−癌細胞ハイブリッドの作製に使用するために、細胞バンク中に保存された癌細胞サンプルを処理することである。これらのサンプルは、癌を除去するために、又は癌の増殖を最小限にするために、本発明の方法により投与されてもよい。

樹状細胞は患者内の抗腫瘍応答を誘導するのに使用することができる。樹状細胞は、免疫監視、抗原捕捉、及び抗原提示に関与する細胞のネットワークを形成する、造血に由来する白血球である。当業者に既知のin vitroでDCを単離及び増殖するための多数の技術があり(M.B. Lutz et al. J. Imm. Methods 223(1999) 9277-9279を参照)、したがってそれらを免疫戦略に使用することができる。現在まで、既知の癌抗原を有する合成ペプチド、クラスI分子に由来する剥がれたペプチド(stripped peptides)、腫瘍RNA、又は腫瘍溶解物と共に、DCを癌に対する患者の免疫原性応答を改善するのに使用してきた。

当業者に既知の様々な懸濁する流体又はキャリアは、ワクチン組成物を懸濁するのに利用されてもよい。そのような流体は、滅菌水、生理食塩水、緩衝液、増殖培地に由来した複雑な流体又は他の体液を含むが、これらに限定されない。当業者に既知の防腐剤、安定剤及び抗生物質は、ワクチン組成物中で利用されてもよい。

癌細胞の脱脂は以下のように達成することができる。表2に示される癌細胞、例えばリン酸緩衝生理食塩水(PBS)中の約8×105個の細胞に、最終的な容量で3%ジイソプロピルエーテル(DIPE)及び0.05%トリトンX−100を加えた。癌細胞を1mlの生理食塩水中で再懸濁した。0.05%トリトンX−100及びDIPEは、最終的な濃度の3%まで加えられた。DIPE(30μlの希釈していない(neat)DIPE)を100%として癌細胞懸濁液に加え、5μlの希釈していない(100%)トリトンも加えて、1mLの癌細胞懸濁液の最終的な容量を作製した。溶媒混合物もまた独立して作製し、次に癌細胞懸濁液に加えることができる。癌細胞懸濁液を、室温で20分間転倒により混合した。サンプルを、続いて1000rpmで1分間遠心分離した。残存溶媒は、炭カラムを通して除去した。溶媒除去工程後に、細胞懸濁液を2.5mlの最終的な容量に希釈した。再懸濁されたペレットのアリコートを、下記に述べられるように、癌細胞の脱脂を確認するための総コレステロール含有量又は総タンパク質含有量を検出するのに使用した。
実施例1からの再懸濁液溶液のアリコートを総コレステロール分析に使用した。実施例1からの再懸濁液溶液のさらなるアリコートを、市販のキットを使用して、総タンパク質測定に使用した。例えば、実施例1からの50μlの再懸濁液溶液を、アンプレックスレッド総コレステロール分析(Amplex Red Total Cholesterol Assay)(Molecular Probes, Eugene, Oregon)に使用した。別の50μlの再懸濁液溶液は、バイオラッド総タンパク質測定(BioRad Total Protein Assay)(Bio-Rad Laboratories, Hercules, CA)を使用して、総タンパク質測定に使用した。これらの研究の結果は表3に示され、コレステロール濃度の減少によって観察されるように、ソースにかかわらず脱脂されたすべての癌細胞株が首尾よく脱脂されたことを実証する。明らかに、実施例1に記述される3%DIPE−0.05%トリトンX−100のプロトコルは、マウス癌細胞株B16−F10を含む様々な癌細胞から脂質を効率的に除去する。表3により、50%超のタンパク質が保持されている一方、コレステロールの99%超が除去されていることが実証される。明らかに、B16−F10及びTF−1細胞株は、非常に効率的に脱脂された。さらに、良好なタンパク質回収率が達成されることから、細胞構造が維持されていると考えられる。
未熟な樹状細胞(DC)は古典的な抗原提示細胞(APC)である。これらのDCは、効率的に病原体全体を取り込み、癌細胞/抗原を処理し、且つ抗体産生のためにB細胞に及び細胞介在性免疫応答のためにT細胞にエピトープを提示する。この実験において、未熟なDCを、蛍光活性化細胞分別(FACS)装置のフルオレセインイソチオシアネート(FITC)チャンネルで読み取る色素(図1の「A」)により標識した。脱脂されたB16−F10黒色腫を、FACS装置のフィコエリトリン(PE)チャンネルで読み取る別個の色素(図1の「C」)により標識した。したがって、脱脂されたB16−F10細胞がDCによって取り込まれる場合、そのときB16−F10細胞の蛍光は低減するが、DCの蛍光は損なわれないままである。
1. 接着細胞又は結合細胞は、最初に、タンパク質分解酵素(すなわちトリプシン/EDTA)を使用して取り出し、単細胞浮遊液を形成した。
2. 工程はすべて25℃で実行された。合計約2×107個の単細胞を円錐形の底のポリプロピレンチューブに入れ、無血清培地を使用して1回洗浄した。
3. 細胞を5分間遠心分離し(400×g)、ゆるい沈殿とした。
4. 遠心分離後に注意深く上清を吸引し、ペレットにわずか25μlの上清を残した。5. 1mlの希釈剤C(Diluent C)(染色キットと共に供給される)を加え、溶液(PKH67−GL(緑色蛍光性細胞結合色素)、及びPKH−GL(赤色蛍光性細胞結合色素)、Sigma, St. Louis, MO)を、完全な分散を確実にするためにピペッティングによって再懸濁した。
6. 染色の直前に、4×10-6MのPKH26色素を希釈剤Cを使用して、ポリプロピレンチューブ中に調製した(2×ストックとして)。エタノール作用を最小限にするために、加えられた色素の量は個別のサンプル量の1%未満である。色素ストックのさらなる希釈が必要である場合、中間ストックを100%エタノールでの希釈によって作製する。その調製物は室温(25℃)で保存する。
7. 1mlの細胞(2×107個)を、1mlのPKH26色素(1×107細胞数/ml)に迅速に加えた。サンプルをピペッティングによって混合した。
8. その溶液を25℃で2〜5分間インキュベートした。25℃でのこの染色期間に、混合を確実にするためにチューブを周期的にゆっくりと転倒した。
9. 染色反応を停止するために、等量の血清又は適合するタンパク質溶液(すなわち1%のBSA)を加え、1分間インキュベートした。
10. 血清で停止したサンプルを、等量の完全培地で希釈した。
11. 染色溶液から細胞を取り出すために、細胞を25℃で10分間400×gで遠心分離した。
12. 上清を除去し、細胞ペレットをさらなる洗浄のために新しいチューブに移した。13. 細胞を洗浄するために10mlの完全培地を加え、次に遠心分離して、所望の濃度に再懸濁した。
実施例3において観察されたDCによるB16−F10細胞の効率的な取り込みが任意の治療的ワクチン接種に利益をもたらしたか否かを調べるために、B16癌を有するマウスを、脱脂されたB16−F10細胞をパルスした自己DCで1回ワクチン接種した。
D.脱脂されたB16−F10細胞をパルスしたBMDDCで免疫付与したマウスの試験群(1群当たり5匹のマウス)を使用して実行した。実験計画を図11に示す。
脱脂されたB16−F10細胞をパルスしたDCが、マウスでB16癌が発症するのを予防するという点で何らかの利益を有するか否かを調べるために、以下に述べるようにマウスを1回ワクチン接種し、B16−F10癌細胞で免疫性を試験した。
a)本発明の方法、例えば3%DIPE及び0.05%トリトンX−100の溶媒混合物を使用して脱脂された細胞、又は
b)複数の凍結融解サイクルを使用して溶解されたB16−F10細胞
から成っていた。
脱脂されたB16−F10細胞、又は脱脂されたB16−F10細胞をパルスしたDCのいずれかによるワクチン接種が腫瘍抗原特異的免疫応答を生じさせるか否かを判定する
ために、B16特異的な腫瘍抗原GP100及びTRP−2に対する抗体価を測定した。
以下は、骨髄由来樹状細胞(BMDDC)のin vivoの回収及び増殖のための方法を記述する。この実験において、使用されるマウス種はC57BL/6(約8〜16週齢)であった。5匹のマウスが各々の群に割り当てられた。この実験における癌モデルは、免疫原性の低い腫瘍モデルであると知られているB16−F10黒色腫(C57BL/6マウス中)であった。骨髄を、大腿骨及び脛骨からFalconの100μmナイロン細胞ストレーナーを通して回収した。
4〜12週齢のC57BL/6のメスマウスの大腿骨及び脛骨を、Kleenexのティッシュペーパーでこすることによって周囲の筋組織から取り出し精製した。無傷の骨を、消毒のために70%エタノール中に2〜5分間置き、PBSで洗浄した。骨の両端をはさみで切断し、直径0.45mmの針を備えた注射器を使用して、骨髄をPBSで洗い流した。骨髄懸濁液中の塊を崩すために、激しいピペッティングを使用した。次にその懸濁液をPBSで1回洗浄した。細胞を、ペニシリン(100U/mL、Sigma)、ストレプトマイシン(100U/mL、Sigma)、L−グルタミン(2mM、Sigma)、2−メルカプトエタノール(50μM、Sigma)、10%の熱不活性化した濾過したウシ胎仔血清を補充した(R10)RPMI−1640(GIBCO BRL)(Millipore又はCorningの0.22μmフィルターですべてろ過した)に再懸濁した。
0日目:骨髄の播種。白血球を、200U/ml(=20ng/ml)のrmGM−CSFを含む10mlのR10培地中で100mmのディッシュ当たり2×106個で播種した。
3日目:200U/mlのrmGM−CSFを含む10mlのR10培地をさらに加えた。
6日目:培養上清の半分を回収し、取り出した培養上清を遠心分離し、ペレットを200U/mlのrmGM−CSFを含む10mlの新しいR10培地中で再懸濁した。その懸濁液をもとのプレートに再び戻した(re-plated back)。
7日目:未熟なDCに、溶解又は脱脂されたB16−F10細胞を与えた。1×106個のDC及び1×106個の脱脂されたB16−F10細胞を24時間加えた。
8日目:DC及び脱脂されたB16−F10細胞を、100ng/mlのリピドA又は1μg/mlのリポ多糖類(LPS)+30〜100U/mlのrm(組換型マウス)GM−CSFを使用して成熟させた。注射に先立って、細胞をさらに24時間インキュベートした。
培養された細胞を1回洗浄した。細胞のアリコートを、トリパンブルー溶液(Sigma)と共に1:1(体積:体積)で混合した。トリパンブルー陰性の大きな白血球(赤血球は大きさ及び形によって除外された)を生細胞として、顕微鏡下で計測した。
1×105個の細胞を、0.1%アジ化ナトリウムを含む50μlハイブリドーマ培養上清、又は精製された一次抗体及び二次抗体により染色した(M.B. Lutz et al. Journal
of Immunological Methods 223(1999)77-92 79を参照)。細胞並びに一次抗体及び二次抗体(5〜20μg/ml)を、氷上で30分間インキュベートした。一次試薬及び二次試薬を、5%ウシ胎仔血清(FCS)及び0.1%アジ化ナトリウムを含むPBS中で希釈し、このPBSは洗浄培地としてもまた有用だった。サンプルをFACScan(Becton Dickinson, Heidelberg, Germany)により分析した。
Claims (16)
- 脂質含有量が減少し、少なくとも1つの癌細胞抗原を有する癌細胞と、
薬学的に許容可能なキャリアと
を含むワクチン組成物。 - 樹状細胞をさらに含む、請求項1に記載のワクチン組成物。
- 免疫賦活剤をさらに含む、請求項1又は2に記載のワクチン組成物。
- 脂質含有量が減少し、少なくとも1つの癌細胞抗原を有する前記癌細胞が、動物又はヒトに投与された場合に免疫原性である、請求項1又は2に記載のワクチン組成物。
- 動物又はヒトの免疫原性応答を誘導するのに有用な医薬品の調製における、脂質含有量が減少し、少なくとも1つの癌細胞抗原を有する癌細胞の使用。
- 動物又はヒトの免疫原性応答を誘導するのに有用な医薬品の調製における、脂質含有量が減少し、少なくとも1つの癌細胞抗原を有する癌細胞の使用であって、該免疫原性応答が、該動物又はヒトにおいて、癌を治療し、癌を予防し、又は癌の進行を遅らせる、使用。
- 前記医薬品がさらに樹状細胞を含む、請求項5又は6のいずれかに記載の使用。
- 医薬品がさらに免疫賦活剤を含む、請求項5〜7のいずれかに記載の使用。
- 癌細胞において脂質のレベルを低減する方法であって、
流体中の癌細胞を第1の抽出溶媒と接触させること、
前記癌細胞から脂質を抽出するのに十分な時間、前記流体と前記第1の抽出溶媒を混合し、それにより脂質含有量が減少した癌細胞を産生すること、
前記流体から前記溶媒層を取り去ること、及び
前記脂質含有量が減少した癌細胞を含む前記流体を回収すること
を含む、方法。 - ワクチン組成物を作製する方法であって、
流体中の癌細胞を第1の抽出溶媒と接触させること、
前記癌細胞から脂質を抽出するのに十分な時間、前記流体と前記第1の抽出溶媒を混合し、それにより脂質含有量が減少した癌細胞を産生すること、
