Description
(1)NB-206およびその類似体が、HIV介在の細胞融合を阻害した。
(2)ウイルスを添加してから2時間より前にNB-206およびその類似体を添加した時にのみHIV複製を阻害した。
(3) NB-206およびその類似体が、gp41コアの形成を遮断した。これは、立体構造特異的MAb NC-1を用いたサンドイッチ酵素免疫測定法(ELISA)で検出されたものである。
(4) NB-206およびその類似体が、gp41の 6へリックス束の形成を阻害した。これは、蛍光ネイティブポリアクリルアミドゲル電気泳動法(FN-PAGE)で明らかにされたものである。
これらの結果は、NB-206とその類似体が疎水性空洞に作用して、融合活性を有するgp41のコイルドコイル領域の形成を阻害することにより、HIV-1が介在する膜融合とウイルスの進入を阻害することを示唆している。
a) マウス以外の動物から、HIV-1 gp41が有する3個のN-ペプチドおよびHIV-1 gp41が有する3個のC-ペプチドを含むHIV-1 gp41の3量体構造に対するポリクローナル抗体を獲得して固相上におくことにより、ポリクローナル抗体がコーティングされた固相を形成し、
b) HIV-1 gp41のN-ペプチドと試験化合物の混合物を調製し、次にHIV-1 gp41のC-ペプチドを添加し、
c) 段階(b)で得られた混合物を段階(a)で得られたポリクローナル抗体がコーティングされた固相に加え、非結合ペプチドと非結合化合物を除去し、HIV-1 gp41およびHIV-1 gp41の3個のC-ペプチドと特異的に反応するがHIV-1 gp41の個々のN-ペプチドとは反応せず、HIV-1 gp41の個々のC-ペプチドとも反応しないモノクローナル抗体を添加し、
d) 前記モノクローナル抗体の結合を測定する。
前記R1-R6は、それぞれ、H、アルキル、シクロアルキル、アルケニル、アルキニル、アリール、アリールアルキル、アルキルアリール、ヘテロシクリル、テトラゾイル、アダマンチル、ハロゲン、トリフルオロメチル、OH、CN、NO2およびOR7(該R7はアルキル、アリールまたはアリールアルキル)、COOR8(該R8はHおよびアルキル)、SO3R9(該R9はHおよびアルキル)、SO2NHR10(該R10はHもしくはアルキル)、CONHR11(該R11はHまたはアルキル)からなる群のうちいずれであってもよいが、これらに限定されない。
(試薬)MT-2細胞、HIV-1IIIB感染H9細胞(H9/HIV-1IIIB)、U87-T4-CXCR4およびU87-T4-CCR5細胞、実験室適応HIV-1株および一次HIV-1株、ならびに抗p24mAb (183-12H-5C)を、NIH AIDS Research and Reference Reagent Programから入手した。リンパ細胞系CEMx174 5.25M7は、C. Cheng-Mayerにより親切にも提供されたものであり、これは、HIV-1の長い末端反復配列(LTR)グリーン蛍光タンパク質(GFP)リポーターおよびルシフェラーゼリポーターコンストラクトが安定に形質導入されている。この細胞は、CD4と両方のコレセプターCXCR4およびCCR5を発現する(文献30)。これらの細胞を10%ウシ胎仔血清(FBS)、1μg/mlピューロマイシン、200μg/ml G418が添加されたRPMI-1640培地で維持した。組み換え可溶性CD4(sCD4)は、ジェネンテック社(Genentech Inc. (South San Francisco, CA))から入手した。ペプチドN36、C34(文献7,17)、IQN17(文献31)およびT22(文献32,33)は、ニューヨーク血液センター(New York Blood Center)の微量化学研究所(MicroChemistry Laboratory)で標準の固相FMOC法を用いて合成した。ビオチン化D-ペプチドであるD10-p5-2K(文献31)についても、前述のように(文献31)、D-アミノ酸を使って施設内で合成し、酸化させた。ペプチドは、高速液体クロマトグラフィー(HPLC)で、均質になるまで精製された。精製したペプチドの同定は、レーザー脱離質量分析(PerSeptive Biosystems)を用いて確認した。N36/C34の混合物およびIQN17に対するウサギ抗血清は、前述の方法(文献29)で調製した。gp41の6へリックス束に特異的なマウスのmAb NC-1を調製し、前述のように(文献29)特性付けた。ウサギおよびマウスの免疫グロブリン(IgG)は、プロテインA/Gビーズ(Protein A/G beads)(Pierce, Rockford, IL)を用いて精製した。CXCR4に特異的なマウスmAb 12G5は、R&D Systems (Minneapolis, MN)から購入した。スクリーニングのための化学物質ライブラリーは、Nanosyn (Menlo Park, CA)から購入した。NB-206およびその類似体はChemBridge Corporation (San Diego, CA) から購入した。クロロペプチンは、北里研究所(日本 東京)のSatoshi OmuraおよびHaruo Tanakaからの有り難い提供によるものである。
2×105/mlのHIV-1IIIB感染H9細胞(H9/HIV-1IIIB)を、スクリーニング対象である化合物の存在下(化合物の最終濃度25μg/ml)で、MT-2細胞(2×106/ml)とともに共培養した。この培養は96ウェルプレート中で37℃2日間行った。HIV-1誘導の合胞体形成を倒立顕微鏡下で観察し、「−」(合胞体が観察されなかった)、「±」(50%の合胞体が阻害された)、「+」(合胞体形成が阻害されなかった)の評価をつけた。「−」と「±」の評価を受けた前記化合物を選択し、さらにgp41の6へリックス束形成に対する阻害物質についてELISA法を用いてスクリーニングを行った。
前述のサンドイッチELISA法(文献27)を用いて、gp41の6へリックス束形成を阻害する化合物をスクリーニングした。簡潔に述べると、ペプチドN36 (2μM)を試験化合物と一緒に指定濃度で37℃30分間プレインキュベートし、その後C34 (2μM)を加えた。対照実験では、N36はC34とともに37℃で30分間プレインキュベートし、その後試験化合物を添加した。37℃で30分間インキュベートした後、混合物を96ウェルポリスチレンプレート(Costar, Corning Inc., Corning, NY)のウェルに加えた。該ウェルは、N36/C34混合物に対するウサギ抗血清から精製したIgG (2μg/ml)を予めコーティングしておいたものである。その後、該mAb NC-1、ビオチン標識ヤギ抗マウスIgG (Sigma Chemical Co., St. Louis, MO)、ストレプトアビジン標識ホールラディッシュペルオキシダーゼ(SA-HRP)(Zymed, S. San Francisco, CA)、および基質3,3 ',5,5'-テトラメチルベンジジン(TMB) (Sigma)を連続的に加えた。ELISAリーダー(Ultra 384, Tecan, Research Triangle Park, NC) を用いて、450 nmの吸光度を測定した。該化合物による阻害率を前述の方法(文献34)で計算し、50%阻害濃度(IC50)を指定ソフトウェアCalcusyn(文献35)を用いて算出した。このソフトウェアは、Dr. T. C. Chou (Sloan-Kettering Cancer Center, New York, New York)により、親切にも提供されたものである。
実験室適応HIV-1株による感染に対する化合物の阻害活性を前述の方法(文献34)により決定した。簡潔に述べると、1×104個のMT-2細胞に、100 TCID50(50%組織培養感染量)のHIV-1を、10%FBSを含むRPMI 1640培地200μl中で感染させた。これは、一晩中、段階的に化合物の濃度の変える条件下、またはその非条件下で行われた。添加時期検定(time- of addition assay)では、感染後の様々な時点で化合物を添加した。その後、培養上清を除去して新鮮培地を加えた。感染4日後に100μlの培養上清をそれぞれのウェルから採取し、等量の5% Triton X-100と混合してp24抗原を分析した。この分析は、ELISA法で定量化した(文献23)。簡潔に述べると、ポリスチレンプレート(Immulon IB, Dynex Technology, Chantilly, VA)のウェルをHIV免疫グロブリン(HIVIG)でコーティングした。これは、前述のように(文献36)、HIV-1IIIBに対して高い中和力価を有するHIV血清反応陽性ドナーの血漿を、0.085 M炭酸塩−重炭酸イオン緩衝液(pH 9.6)中4℃で一晩おいて調製された。コーティング後、PBS-T緩衝液(0.05% Tween-20を含む0.01M PBS)で洗浄し、1%乾燥脱脂粉乳(Bio-Rad Inc., Hercules, CA)を含むPBSで遮断した。ウイルス溶解物をウェルに添加し、37℃で1時間インキュベートした。全面を洗浄した後、抗p24 mAb(183-12H-5C)、ビオチン標識抗マウスIgG1 (Santa Cruz Biotech., Santa Cruz, CA)、SA-HRPおよびTMBを連続的に加えた。1N H2SO4を加えることにより反応を停止させた。ELISAリーダー(Ultra 384, Tecan)を用いて、450nmの吸光度を記録した。標準用量応答曲線を確立するにあたり、組み換えタンパク質p24 (US Biological, Swampscott, MA)を含めた。
色素移動分析(dye transfer assay)を用いて、前述のように(文献11,25,26)、HIV-1介在の細胞融合を検出した。H9/HIV-1IIIB細胞を蛍光試薬であるCalcein-AM (Molecular Probes, Inc., Eugene, OR)で標識し、MT-2細胞とともに(比= 1:5)、96ウェルプレート中で37℃2時間、試験化合物の存在下または非存在下でインキュベートした。倒立蛍光顕微鏡(Zeiss, Germany)に測微接眼レンズディスクを装着して、融合および非融合のCacein標識HIV-1感染細胞を数えた。細胞融合の阻害率とIC50値については、前述の方法(文献11)で計算した。
MT-2細胞に対する化合物のインビトロ細胞毒性を、前述のように(文献21)、XTTテトラゾリウム色素を用いる比色法で測定した。簡潔に述べると、細胞数(5×105/ml)と等量の段階的に濃度を変えた化合物を、96ウェルプレート中の同じウェルに加えた。37℃で4日間インキュベートした後、XTT (1 mg/ml; 50 ml/ウェル; PolySciences, Inc., Warrington, PA)を添加した。4時間後に、可溶性の細胞内ホルマザンを、570nmを基準とした450 nmで比色分析をして定量化した。細胞毒性率(文献37)およびCC50(50%の細胞毒性を示す濃度)の値を、ソフトウェアCalcusyn(文献35)を用いて計算した。
ポリスチレンプレートのウェルは、2μg/mlのヒツジ抗gp120抗体D7324 (Cliniqa, Fallbrook, CA) 100μlでコーティングした。該コーティングは、炭酸塩緩衝液(pH9.6)中4℃で一晩行ったものである。その後、1%乾燥脱脂粉乳を含むPBS溶液で37℃1時間かけて遮断した。PBS中0.5μg/mlの組み換えgp120分子(Immunodiagnostics, Woburn, MA)を100マイクロリットル添加し、37℃で1時間インキュベートし、その後、PBS-Tで3回洗浄した。可溶性CD4(sCD4)0.25μg/mlを、化合物(25μM)の存在下で添加し、37℃で1時間インキュベートした。3回洗浄した後、ウサギ抗sCD4 IgG (PBS中0.25μg/ml、100μl/ウェル)を加え、37℃で1時間インキュベートした。ウサギ抗sCD4 IgGの結合は、ビオチン化ヤギ抗ウサギIgG、SA-HRP、およびTMBを連続的に添加して決定した。その反応が終了した後、450 nmの吸光度をELISAリーダー(Tecan)で記録した。
(ハイスループットスクリーニング(HTS)によるNB-145の同定)
合胞体形成分析、およびサンドイッチELISA法を基礎にしたHTS技術を用いて、単一用量(25μg/ml)で46,640種の化合物からなるNanosyn Corporationの化学物質ライブラリーのスクリーニングを行った。これらの化合物は「薬らしい(drug-like)」分子であり、「ユニバーサル」ライブラリーとなるように合理的に選定されたものである。このライブラリーは、最小数の化合物で最大限の多様な薬理作用性に及ぶものである。ある濃度のNB-145と称される化合物が、HIV-1介在の合胞体形成とgp41 N-ペプチドN36およびC-ペプチドC34間の6へリックス束形成を完全に阻害した。これは、該化合物が、gp41介在の膜融合を阻害することによりHIV-1感染を阻害する可能性があることを示唆している。したがって、この化合物は、より強力なHIV-1融合阻害剤を同定するためのリード化合物として使用できると考えられる。
NB-145の化学構造に基づいて、我々は、Chembridge Corporationの化学データベースを検索し、NB-145に類似した構造を有する73種の化合物を発見した。そこで、我々は、これらの化合物を購入し、それらの阻害活性について、以下(1)HIV-1複製(p24産生)、(2)HIV-1介在の細胞変性効果(CPE)、(3)HIV-1 Env-誘発の細胞間融合、およびMT-2細胞に対する細胞毒性について試験した。CC50(50%細胞毒性濃度)およびIC50(50%阻害濃度)の値に基づき、選択指数(SI)を計算した。表2(1〜2)に示すように、化合物の1つであるNB-206と称される化合物は、HIV-1の複製阻害(IC50=19nM)およびHIV-1介在の細胞間融合阻害(IC50≦0.667μM)に最も強力であり、SIは981であった。NB-206以外に、NB-206と同一の親構造を有する他の20種の化合物もHIV-1感染に対して強力な阻害活性があり、IC50値は87〜943nM、SI値は48〜>1778であった。これらの活性抗HIV-1化合物のほとんどは、細胞毒性が低かった。
添加時期検定を実施し、NB-206およびその類似体がHIV-1進入阻害剤であるかどうかを決定した。MT-2細胞をHIV-1IIIBとともに、37℃で0、1、 2、3、4、6および8時間インキュベートした後、それぞれにNB-206およびNB-231を2.5μMの濃度で添加した。対照としてアジドチミジン(AZT)(0.1μM)を用いた。さらに2時間培養した後、細胞を洗浄して遊離ウイルスと化合物を除去した。感染から4日後に上澄みを採取して、p24産生量を測定した。NB-206およびその類似体は、ウイルスと一緒に細胞に添加した場合にのみHIV-1複製を阻害したが、ウイルスを細胞に加えて1時間以上経過してから添加すると阻害活性を示さなかった。しかしながら、AZTは、感染から8時間後に添加した場合であってもHIV-1複製を有効に阻害した(図1)。
NB-206およびその類似体の阻害活性を、実験室適応HIV-1株によるMT-2細胞への感染、および一次HIV-1株によるCEMx174 5.25 M7細胞への感染に対して、前述の方法(文献23, 38)で決定した。HIV-1IIIBに加えて、NB-206およびその類似体(NB-231, NB-154, NB-179)は、他の実験室適応HIV-1株(RF、SF2、MN、AZT-R、AZT耐性株を含む)による感染をも用量依存的に阻害し、IC50値はナノモル域であった(表3)。
続いて、gp41の6へリックス束形成に対するNB-206およびその類似体の作用について決定した。該6へリックス束形成は、HIV-1が標的細胞と融合する際の重要な立体構造の変化である。NペプチドとCペプチドを等しいモル濃度で混合することにより(文献16)、gp41の6へリックス束のモデル系を確立した。このモデルgp41コア構造は、サンドイッチELISA法で立体構造特異的なmAbのNC-1を使って検出できる(文献27,29)。この系を用いて、gp41の6へリックス束形成に対するNB-206およびその類似体の阻害活性を試験した。図3に示すように、NB-206およびその類似体 (NB-231, NB-154,およびNB-179)は、用量依存的にN36とC34間の6へリックス束形成を著しく阻害した。NB-206、NB-231、NB-154、NB-179のIC50値(μM)は、それぞれ0.83±0.03、0.93±0.33、1.56±0.12、2.51±0.27であった。これらの結果は、NB-206およびその類似体が、gp41のコイルドコイル領域のコンポーネントと結合し、gp41のNHR領域とCHR領域間の結合を干渉することを示唆するものである。
過去20年間、HIV/AIDS研究における最大の進展の一つは、抗HIV治療薬が開発されたことである(文献39)。現在まで、20種の抗HIV治療薬が米国FDAの承認を受けており、さらなる新薬候補が控えている(文献40)。これらの薬剤のほとんどは、HIV-1の逆転写酵素とプロテアーゼを標的にしている。その中の1つであるFuzeon(T-20)だけが、ウイルスエンベロープ糖タンパク質gp41を標的にしている(文献14,38,41,42)。T-20(文献41)は、SJ-2176 (文献11,43)やC34(文献17)のようなHIV-1 gp41のCHR領域に由来する他のペプチドと同様に、HIV-1の融合と進入を阻害する。臨床試験では、これがHIVの複製に対して極めて有望であることが示されている(文献14,44)。しかしながら、T-20は2つの大きな制限がある。それらは、経口アベイラビリティーがないこと(皮下注射される)、および製造原価が高いことである(文献40)。したがって、低分子のHIV-1融合阻害剤の開発が至急必要である。
Claims (26)
- 下式(化1)により表される構造式(I)を有する抗ウイルス化合物またはその薬学的に許容可能な塩。
R1、R2、R3、R4およびR5は、それぞれ、H、ハロゲン、(CH2)nCOOR、アルキル、シクロアルキル、アルケニル、アルキニル、アリール、アリールアルキル、ヘテロシクリル、アダマンチル、トリフルオロメチル、OH、CN、ニトロ、OR7、SO3R、SO2NHRおよびCONHRからなる群のうちいずれであってもよく、該nは、0乃至10の整数であり、該ヘテロシクリルは、ピリジル、フリルおよびテトラゾイルであるがこれらに限定されず、該R7は、アルキル、アリールまたはアリールアルキルであり、該RはHまたはアルキルであって、
R6は、H、アリール、アルキル、シクロアルキル、アリールアルキル、アルケニル、アルキニル、アルコキシアルキル、CH2CONHアリール、CH2CON(CH2CH2) 2O、CH2COOR、ヘテロシクリル-CH2およびCH(CH3)CO2Rからなる群から選択され、該RはHまたはアルキルであり、該アルコキシは、メトキシ、エトキシ、プロポキシ、ブトキシ、ペントキシまたはヘキソキシであり、該ヘテロシクリルは、ピリジル、フリルまたはテトラゾイルであるがこれらに限定されず、該アリールは、H、ハロゲン、アルキル、アルコキシ、ニトロ、トリフルオロメチルおよび(CH2)nCOORからなる群の1種以上で任意に置換されたフェニルまたはナフチルであり、該nは、0乃至10の整数であり、該RはHまたはアルキルであって、

前記R2がH、F、Cl、BrまたはIであって、
前記R3、R4およびR5がHであって、
前記R6が置換または非置換のアルキル、アルケニル、フェニルまたはアリールアルキル基であることを特徴とする請求項1記載の抗ウイルス化合物またはその薬学的に許容可能な塩。 - 前記ZがSであって、
前記R1が(CH2)nCOOHであり、該nは0乃至3の整数であって、
前記R2がH、FまたはClであって、
前記R3、R4およびR5がHであって、
前記R6がアルキルもしくは1以上の炭素原子を有する置換のアルキル基、または置換のフェニルもしくはベンジル基であることを特徴とする請求項1記載の抗ウイルス化合物またはその薬学的に許容可能な塩。 - 前記R2がHまたはClであって、
前記R3、R4およびR5がHであって、
前記R6が2乃至6の炭素原子を有するアルキル基、または置換のフェニルもしくはベンジル基であることを特徴とする請求項4記載の抗ウイルス化合物またはその薬学的に許容可能な塩。 - 前記R6がエチルであることを特徴とする請求項6記載の抗ウイルス化合物またはその薬学的に許容可能な塩。
- 前記ZがSであって、
前記R1が(CH2)nCOOHであり、該nは0乃至10の整数であって、
前記R2がH、F、ClまたはIであって、
前記R3、R4およびR5がHであって、
前記R6が置換または非置換のアルキル、アルケニル、フェニルもしくはアリールアルキル基であって、該アルキル、アルケニル、フェニルもしくはアリールアルキル基がそれぞれ12までの炭素原子を有することを特徴とする請求項1記載の抗ウイルス化合物またはその薬学的に許容可能な塩。 - 前記R6が置換もしくは非置換の1乃至3の炭素原子を有するアルキル基、置換もしくは非置換の6までの炭素原子を有するアルキル部分を備えるアリールアルキル基、または置換もしくは非置換のフェニルであることを特徴とする請求項9記載の抗ウイルス化合物またはその薬学的に許容可能な塩。
- 前記ZがSであって、
前記R1が(CH2)nCOOHであり、該nは0であって、
前記R2がClであって、
前記R3、R4およびR5がHであって、
前記R6がフェニルエチルであることを特徴とする請求項9記載の抗ウイルス化合物またはその薬学的に許容可能な塩。 - 下式(化6)により表される構造式(I)を有する抗ウイルス化合物またはその薬学的に許容可能な塩、ならびに薬学的に許容可能な担体を含む医薬組成物。
R1、R2、R3、R4およびR5は、それぞれ、H、ハロゲン、(CH2)nCOOR、アルキル、シクロアルキル、アルケニル、アルキニル、アリール、アリールアルキル、ヘテロシクリル、アダマンチル、トリフルオロメチル、OH、CN、ニトロ、OR7、SO3R、SO2NHRおよびCONHRからなる群のうちいずれであってもよく、該nは、0乃至10の整数であり、該ヘテロシクリルは、ピリジル、フリルおよびテトラゾイルであるがこれらに限定されず、該R7は、アルキル、アリールまたはアリールアルキルであり、該RはHまたはアルキルであって、

- 適切な宿主細胞へのヒト免疫不全ウイルス(HIV)感染を阻害または予防するための使用に適切であることを特徴とする請求項13記載の医薬組成物。
- 前記感染が宿主細胞へのウイルスの進入を妨げることにより阻害または予防されることを特徴とする請求項18記載の医薬組成物。
- 適切な宿主細胞におけるヒト免疫不全ウイルス(HIV)の複製を阻害するための方法であって、該方法は、
(a)適切な宿主細胞を、HIVの複製を阻害するために有効な量の請求項1記載の化合物または請求項14記載の組成物と接触させる段階と、
(b)適切な条件下において、HIVを(a)段階の宿主細胞に感染させる量のHIVを、(a)段階の宿主細胞に接触させる段階と、
(c)該宿主細胞の感染レベルと、請求項1記載の化合物または請求項13記載の医薬組成物と接触しなかった適切な宿主細胞の感染レベルとを比較する段階を備えることを特徴とする方法。 - 適切な宿主細胞がヒト細胞であることを特徴とする請求項20記載の方法。
- ヒト免疫不全ウイルスに感染した対象を治療する方法であって、
前記方法が、有効量の請求項1記載の化合物またはその薬学的に許容可能な塩、ならびに薬学的に許容可能な担体を含む医薬組成物を該対象に投与することを特徴とする方法。 - 抗HIV薬、抗感染薬および免疫賦活剤からなる群から選択される後天性免疫不全症候群(AIDS)治療薬の有効量をさらに含むことを特徴とする請求項22記載の方法。
- 対象の後天性免疫不全症候群(AIDS)の徴候を予防する方法であって、
該対象の症候群を予防するために有効な量の請求項1記載の化合物を含む医薬組成物を該対象に投与する方法。 - 抗HIV薬、抗感染薬および免疫賦活剤からなる群から選択される後天性免疫不全症候群(AIDS)治療薬の有効量をさらに含むことを特徴とする請求項24記載の方法。
- 前記対象がヒトであることを特徴とする請求項22、23、24または25記載の方法。
